Electron shuttle of carboxyl boosted Fe(III)-anchored carboxylated cellulose activating sodium perborate for organic pollutant abatement

The traditional Fenton application is limited by the disproportionation of hydrogen peroxide (H2O2) and the tardy reduction of Fe(III). To conquer these issues, we construct an enhanced Fenton-like process using a composite of citric acid-carboxylated microcrystalline cellulose complexing with Fe(III) (CA-Fe(III)@MCC) to activate sodium perborate (SPB), which can effectively eliminate antibiotic sulfamethoxazole (SMX, 89.2%, 0.035 min-1). The CA-Fe(III)@MCC composite significantly enhances the circulation of Fe(III)/Fe(II), leading to the generation of hydroxyl radicals and perboric radicals with excellent structural stability and minimal Fe leaching during CA-Fe(III)@MCC/SPB process. Systematic investigations reveal that the introduced carboxyl groups not only anchor Fe(III) to prevent its deactivation and leaching, but also regulate the coordination environment and serve as electron shuttles, driving the formation of reactive species and reduction of Fe(III). This process presents a novel, environmentally friendly, and sustainable strategy for utilizing natural cellulose and its industrial biomass product in advanced oxidation processes.

Cerium Oxide Catalyzed Disproportionation of Hydrogen Peroxide: A Closer Look at the Reaction Intermediate.

Cerium oxide nanoparticles (CNPs) have recently gained increasing interest as redox enzyme-mimetics to scavenge the intracellular excess of reactive oxygen species, including hydrogen peroxide (H2 O2 ). Despite the extensive exploration, there remains a notable discrepancy regarding the interpretation of observed redshift of UV-Visible spectroscopy due to H2 O2 addition and the catalase-mimicking mechanism of CNPs. To address this question, we investigated the reaction mechanism by taking a closer look at the reaction intermediate during the catalase mimicking reaction. In this study, we present evidence demonstrating that in aqueous solutions, H2 O2 adsorption at CNP surface triggers the formation of stable intermediates known as cerium-peroxo (Ce-O2 2- ) and/or cerium-hydroperoxo (Ce-OOH- ) complexes as resolved by Raman scattering and UV-Visible spectroscopy. Polymer coating presents steric hinderance for H2 O2 accessibility to the solid-liquid interface limiting further intermediate formation. We demonstrate in depth that the catalytic reactivity of CNPs in the H2 O2 disproportionation reaction increases with the Ce(III)-fraction and decreases in the presence of polymer coatings. The developed approach using UV-Visible spectroscopy for the characterization of the surface peroxide species can potentially serve as a foundation for determining the catalytic reactivity of CNPs in the disproportionation of H2 O2 .

The electrochemical reduction of hydrogen peroxide on a palladium-amorphous carbon composite in an alkaline medium

The study of hydrogen peroxide reduction in an alkaline aqueous medium on a composite of palladium particles dispersed in an amorphous carbon matrix (Pd@AC) was performed. Scanning Electron Microscopy (SEM), Energy-dispersive X-ray spectroscopy (EDX), Fourier-transform infrared spectroscopy (FTIR), and X-ray Powder Diffraction (XRD) were used to examine morphology, composition, and crystalline structure. The crystalline structure and size of palladium were analyzed using XRD. The reduction of hydrogen peroxide was studied through cyclic and square-wave voltammetry in an alkaline solution in the absence and presence of dissolved oxygen. The overall electrocatalytic effect of Pd@AC toward hydrogen peroxide reduction strongly depends on the presence of oxygen and the hydrogen peroxide disproportionation reaction in the three-phase system of amorphous carbon, palladium particles, and electrolyte solution.

Hydrogen Peroxide Disproportionation Activity Is Sensitive to Pyridine Substitutions on Manganese Catalysts Derived from 12-Membered Tetra-Aza Macrocyclic Ligands.

The abundance of manganese in nature and versatility to access different oxidation states have made manganese complexes attractive as catalysts for oxidation reactions in both biology and industry. Macrocyclic ligands offer the advantage of substantially controlling the reactivity of the manganese center through electronic tuning and steric constraint. Inspired by the manganese catalase enzyme, a biological catalyst for the disproportionation of H2O2 into water and O2, the work herein employs 12-membered tetra-aza macrocyclic ligands to study how the inclusion of and substitution to the pyridine ring on the macrocyclic ligand scaffold impacts the reactivity of the manganese complex as a H2O2 disproportionation catalyst. Synthesis and isolation of the manganese complexes was validated by characterization using UV-vis spectroscopy, SC-XRD, and cyclic voltammetry. Potentiometric titrations were used to study the ligand basicity as well as the thermodynamic equilibrium with Mn(II). Manganese complexes were also produced in situ and characterized using electrochemistry for comparison to the isolated species. Results from these studies and H2O2 reactivity showed a remarkable difference among the ligands studied, revealing instead a distinction in the reactivity regarding the number of pyridine rings within the scaffold. Moreover, electron-donating groups on the 4-position of the pyridine ring enhanced the reactivity of the manganese center for H2O2 disproportionation, demonstrating a handle for control of oxidation reactions using the pyridinophane macrocycle.

Effect of Redox Potential on Diiron-Mediated Disproportionation of Hydrogen Peroxide.

Heme and nonheme dimanganese catalases are widely distributed in living organisms to participate in antioxidant defenses that protect biological systems from oxidative stress. The key step in these processes is the disproportionation of H2O2 to O2 and water, which can be interpreted via two different mechanisms, namely via the formation of high-valent oxoiron(IV) and peroxodimanganese(III) or diiron(III) intermediates. In order to better understand the mechanism of this important process, we have chosen such synthetic model compounds that can be used to map the nature of the catalytically active species and the factors influencing their activities. Our previously reported μ-1,2-peroxo-diiron(III)-containing biomimics are good candidates, as both proposed reactive intermediates (FeIVO and FeIII2(μ-O2)) can be derived from them. Based on this, we have investigated and compared five heterobidentate-ligand-containing model systems including the previously reported and fully characterized [FeII(L1-4)3]2+ (L1 = 2-(2'-pyridyl)-1H-benzimidazole, L2 = 2-(2'-pyridyl)-N-methyl-benzimidazole, L3 = 2-(4-thiazolyl)-1H-benzimidazole and L4 = 2-(4'-methyl-2'-pyridyl)-1H-benzimidazole) and the novel [FeII(L5)3]2+ (L5 = 2-(1H-1,2,4-triazol-3-yl)-pyridine) precursor complexes with their spectroscopically characterized μ-1,2-peroxo-diiron(III) intermediates. Based on the reaction kinetic measurements and previous computational studies, it can be said that the disproportionation reaction of H2O2 can be interpreted through the formation of an electrophilic oxoiron(IV) intermediate that can be derived from the homolysis of the O-O bond of the forming μ-1,2-peroxo-diiron(III) complexes. We also found that the disproportionation rate of the H2O2 shows a linear correlation with the FeIII/FeII redox potential (in the range of 804 mV-1039 mV vs. SCE) of the catalysts controlled by the modification of the ligand environment. Furthermore, it is important to note that the two most active catalysts with L3 and L5 ligands have a high-spin electronic configuration.

Biomimetic Molybdenum Sulfide-Catalyzed Tumor Ferroptosis and Bioimaging.

The chemical generation of singlet oxygen (1 O2 ) by the MoO4 2- -catalyzed disproportionation of hydrogen peroxide (H2 O2 ) has been widely applied in numerous catalytic processes; however, such molybdate ions cannot be administered for redox-based cancer therapeutics. This work reports the albumin-mediated biomimetic synthesis of highly active molybdenum sulfide (denoted MoB) nanocatalysts that mediate the simultaneous generation of 1 O2 and superoxide anion (O2 •- ) from H2 O2 , which is relatively abundant in malignant tumors. The MoB-catalyzed reactive oxygen species (ROS) are able to activate the ferroptosis pathway and cause lipid peroxidation for efficient cancer therapy. Furthermore, for the first time, the catalytic activity of MoB is visualized in situ. Moreover, a catalytic imaging modality based on MoB is developed for specific imaging of inflammation diseases without background interference. Therefore, this study presents a biomimetic strategy toward Mo-based nanocatalysts for ROS-facilitated tumor ferroptosis and catalytic imaging.

Tetravalent Spiroselenurane Catalysts: Intramolecular Se···N Chalcogen Bond-Driven Catalytic Disproportionation of H2O2 to H2O and O2 and Activation of I2 and NBS.

Chalcogen-bonding interactions have recently gained considerable attention in the field of synthetic chemistry, structure, and bonding. Here, three organo-spiroselenuranes, having a Se(IV) center with a strong intramolecular Se···N chalcogen-bonded interaction, have been isolated by the oxidation of the respective bis(2-benzamide) selenides derived from an 8-aminoquinoline ligand. Further, the synthesized spiroselenuranes, when assayed for their antioxidant activity, show disproportionation of hydrogen peroxide into H2O and O2 with first-order kinetics with respect to H2O2 for the first time by any organoselenium molecules as monitored by 1H NMR spectroscopy. Electron-donating 5-methylthio-benzamide ring-substituted spiroselenurane disproportionates hydrogen peroxide at a high rate of 15.6 ± 0.4 × 103 μM min-1 with a rate constant of 8.57 ± 0.50 × 10-3 s-1, whereas 5-methoxy and unsubstituted-benzamide spiroselenuranes catalyzed the disproportionation of H2O2 at rates of 7.9 ± 0.3 × 103 and 2.9 ± 0.3 × 103 μM min-1 with rate constants of 1.16 ± 0.02 × 10-3 and 0.325 ± 0.025 × 10-3 s-1, respectively. The evolved oxygen gas from the spiroselenurane-catalyzed disproportion of H2O2 has also been confirmed by a gas chromatograph-thermal conductivity detector (GCTCD) and a portable digital polarographic dissolved O2 probe. Additionally, the synthesized spiroselenuranes exhibit thiol peroxidase antioxidant activities for the reduction of H2O2 by a benzenethiol co-reductant monitored by UV-visible spectroscopy. Next, the Se···N bonded spiroselenuranes have been explored as catalysts in synthetic oxidation iodolactonization and bromination of arenes. The synthesized spiroselenurane has activated I2 toward the iodolactonization of alkenoic acids under base-free conditions. Similarly, efficient chemo- and regioselective monobromination of various arenes with NBS catalyzed by chalcogen-bonded synthesized spiroselenuranes has been achieved. Mechanistic insight into the spiroselenuranes in oxidation reactions has been gained by 77Se NMR, mass spectrometry, UV-visible spectroscopy, single-crystal X-ray structure, and theoretical (DFT, NBO, and AIM) studies. It seems that the highly electrophilic nature of the selenium center is attributed to the presence of an intramolecular Se···N interaction and a vacant coordination site in spiroselenuranes is crucial for the activation of H2O2, I2, and NBS. The reaction of H2O2, I2, and NBS with tetravalent spiroselenurane would lead to an octahedral-Se(VI) intermediate, which is reduced back to Se(IV) due to thermodynamic instability of selenium in its highest oxidation state and the presence of a strong intramolecular N-donor atom.

Ultra‐Sensitive Iron‐Doped Palladium Nanocrystals with Enhanced Hydroxyl Radical Generation for Chemo‐/Chemodynamic Nanotherapy

AbstractChemodynamic therapy (CDT) is a novel therapeutic modality that employs Fenton or Fenton‐like reaction to destroy cancer cells via the disproportionation of intratumoral hydrogen peroxide (H2O2) to the highly toxic hydroxyl radical (·OH) without external energy input. A highly efficient chemodynamic catalyst in tumor microenvironment (TME) is key to effective anti‐tumor efficacy. Therefore, ultra‐sensitive FePd‐TPP/adriamycin (ADM) nanocrystals (NCs) are constructed via a simple thermal decomposition method with high catalytic efficiency for combined chemo‐/chemodynamic cancer nanotherapy. These FePd‐TPP/ADM NCs exhib high Fenton‐reaction efficiency with a Michaelis–Menten constant (Km) and maximum velocity (Vmax) of 3.65 mm and 1.69 × 10−9 m s−1, respectively. The ab‐initio molecular dynamics simulation based on density functional theory demonstrate that the repulsion effect between Pd and H atom contribute to the enhancement of the Fe2+ mediated Fenton oxidation. Moreover, the doping of Fe enables FePd‐TPP/ADM NCs to be an efficient contrast agent for T2‐weighted magnetic resonance imaging. Finally, upon intravenous injection, the FePd‐TPP/ADM NCs reveal synergistic tumor growth inhibition compared with chemotherapy and CDT alone. Considering the good biocompatibility and biosafety, this study may pave the way to design a theranostic nano‐platform based on palladium nanoparticles for various catalytic applications.

Mediatorless electrocatalytic oxygen reduction with catalase on mercury–gold amalgam microelectrodes

Catalase is known for its ability to catalyze disproportionation of hydrogen peroxide into oxygen and water. This work reports the oxygen reduction reaction (ORR) catalyzed by catalase, with a decreased activation energy manifested by a positive shift (+0.2 V) in onset potential. Direct bioelectrocatalysis with bovine liver catalase on a mercury–gold amalgam microelectrode is demonstrated. The background oxygen reduction reaction on the amalgam surface is inhibited by the protein, while the hydrogen evolution reaction (HER) is facilitated. Catalase molecules whose prosthetic groups are not wired to the electrode retain their intrinsic activity towards disproportionation of H2O2 generated at the electrode. The Hg–Au amalgam electrode may also be used to detect dissolved catalase in the absence of its substrate (H2O2) in the solution.

Hydrogen peroxide disproportionation: time-resolved optical measurements of spectra, scattering and imaging combined with correlation analysis and simulations

We study how time-dependent optical measurements of spectra, scattering, and imaging can be used to add to the understanding of heterogeneous reactions, compared to work performed using tools developed for homogeneous reactions. Using hydrogen peroxide disproportionation by potassium permanganate as a model reaction, we measure the entire spectrum over reaction time, enabling a clear and useful correlation analysis and assignment of chemical species in heterogeneous conditions. We measure time-dependent dynamic light scattering to study oxygen nanobubble product formation kinetics and equilibrium. We perform macroscopic video-rate reaction imaging and information-theoretic analysis to characterize reaction and transport contributions to the observed signal. To illustrate the differences arising from measuring sample subsets vs the entire system, we integrate stochastic and macroscopic numerical simulations of reaction-diffusion to study homogeneous and heterogeneous reaction conditions. We hope the tools presented here may help understanding other chemical reactions in similar conditions.

Palladium-graphene hybrid as an electrocatalyst for hydrogen peroxide reduction

The electrocatalytic reduction of hydrogen peroxide in an alkaline solution has been studied on a hybrid material based on hydrothermally obtained palladium particles supported on graphene (Pd@G). The morphology, structure, and chemical composition of the prepared hybrid material, including spatial distribution of palladium nanoparticles, were examined by SEM, XRD, EDX, elemental mapping, XPS, FTIR, and BET. The hydrogen peroxide reduction reaction (HPRR) was studied by cyclic and square wave voltammetry in an oxygen-free and air-saturated alkaline medium. The Pd@G exhibited pronounced electrocatalytic activity for HPRR, which proceeds via complex electrode mechanisms coupled with adsorption phenomena and hydrogen peroxide disproportionation reaction as a heterogeneous chemical process on the surface of palladium nanoparticles.

Sustainable amorphous Fenton nanosystem for visualization-guided synergistic tumor elimination

Chemodynamic therapy (CDT) shows great potential in tumor treatment, in which tumor cells are killed by hydroxyl radicals (·OH) generated by Fenton-catalysis-mediated hydrogen peroxide (H2O2) disproportionation. Fabricating a high-performance tumor microenvironment (TME)-responsive Fenton nanocatalyst and realizing sustainable ·OH production are essential for effective tumor elimination but still challenging. Herein, we reported a sustainable amorphous Fenton nanosystem (SAFN) for visualization-guided synergistic tumor therapy, highlighting the superiority of cascade catalysis and the sustainability of the Fenton nanosystem. The SAFN was fabricated by rational integration of a polydopamine (PDA) core, an amorphous ferric phosphate (FePi) payload, glucose oxidase (GOx) decoration, and a bull serum albumin coating. In an acidic TME, Fe(III) ions were released on demand from FePi and reduced to high-activity Fe(II) by PDA. The reducing capacity of PDA was stronger under acidic conditions, due to C=O exposure. Fe(II) and H2O2 are sustainably supplied by the PDA-reducing and GOx catalytic chemistry, achieving long-lasting ·OH production for enhanced CDT (ECDT). Localized hyperthermia can be generated by PDA under near-infrared laser irradiation for low-temperature photothermal therapy (LT-PTT) in a GOx-induced energy-shortage TME. In addition, a remarkable antitumor effect is achieved by this sustainable nanosystem through the synergistic effect of the starvation/LT-PTT/ECDT strategy.

Why is catalase so fast?

As catalases function, they provide coherent oxidative ‘pulses’, which rapidly spread throughout the H-bonded network, effectively ‘unzipping’ H2O2 molecules as far as they extend from the enzyme.
 Result/discussion: This 'memory-of-water'-like mechanism predicts catalase H2O2 disproportionation should occur outside the enzyme. An experimental protocol is proposed to test this prediction. If successful, it would suggest a) holistic re-appraisal of the conventional mechanistic framework of enzyme kinetics is required, and b) should encourage more research into understanding the biochemical effects of CAM therapies.

Stochastic light concentration from 3D to 2D reveals ultraweak chemi- and bioluminescence

For countless applications in science and technology, light must be concentrated, and concentration is classically achieved with reflective and refractive elements. However, there is so far no efficient way, with a 2D detector, to detect photons produced inside an extended volume with a broad or isotropic angular distribution. Here, with theory and experiment, we propose to stochastically transform and concentrate a volume into a smaller surface, using a high-albedo Ulbricht cavity and a small exit orifice through cavity walls. A 3D gas of photons produced inside the cavity is transformed with a 50% number efficiency into a 2D Lambertian emitting orifice with maximal radiance and a much smaller size. With high-albedo quartz-powder cavity walls (rho =99.94%), the orifice area is 1/(1-rho )approx 1600 times smaller than the walls’ area. When coupled to a detectivity-optimized photon-counter (mathcal{D}=0.015,{text{photon}}^{-1},{text{s}}^{1/2}text{ cm}) the detection limit is 110;{text{photon}};{text{s}}^{ - 1} ;{text{L}}^{ - 1}. Thanks to this unprecedented sensitivity, we could detect the luminescence produced by the non-catalytic disproportionation of hydrogen peroxide in pure water, which has not been observed so far. We could also detect the ultraweak bioluminescence produced by yeast cells at the onset of their growth. Our work opens new perspectives for studying ultraweak luminescence, and the concept of stochastic 3D/2D conjugation should help design novel light detection methods for large samples or diluted emitters.

Ferric ion-ascorbic acid complex catalyzed calcium peroxide for organic wastewater treatment: Optimized by response surface method

Hydrogen peroxide (H2O2) disproportionation, iron precipitation, and narrow pH range are the drawbacks of traditional Fenton process. To surmount these barriers, we proposed a ferric ion (Fe3+)-ascorbic acid (AA) complex catalyzed calcium peroxide (CaO2) Fenton-like system to remove organic dyes in water. This collaborative Fe3+/AA/CaO2 system presented an obvious improvement in the methyl orange (MO) decolorization, and also effectively eliminated other dyes. Response surface method was employed to optimize the running parameters for this coupling process. Under the optimized arguments (2.76 mmol/L Fe3+, 0.68 mmol/L AA, and 4 mmol/L CaO2), the MO removal achieved 98.90% after 15 min at pH 6.50, which was close to the computed outcome of 99.30%. Furthermore, this Fenton-like system could perform well in a wide range of pH (3–11), and enhance the H2O2 decomposition and Fe ions recycle. The scavenger experiment result indicated that hydroxyl radical, superoxide anion free radical, and singlet oxygen were acted on the dye elimination. Moreover, electron spin resonance analysis corroborated that the existences of these active species in the Fe3+/AA/CaO2 system. This study could advance the development of Fenton-like technique in organic effluent disposal.

Tuning the π-electron delocalization degree of mesoporous carbon for hydrogen peroxide electrochemical generation

Tuning the selectivity of oxygen reduction reaction to 2 e− pathway is of significant importance for hydrogen peroxide electrochemical generation. C-800 with superior 2 e− oxygen reduction reaction activity and inferior peroxide reduction reaction and peroxide disproportionation reaction activity is the most promising catalyst for hydrogen peroxide electrochemical generation. The thorough understanding of oxygen reduction reaction mechanism provides great insights into the catalyst design. Different π-electron delocalization degree of carbon can tune the electron transferring ability out of catalyst surface and the density of active sites. A plausible volcano relationship of oxygen reduction reaction activity vs. π-electron delocalization degree of carbon is established. High π-electron delocalization degree benefits the electron transfer from catalyst surface to hydrated O2 in outer Helmholtz plane but is unfavorable to the increase of active sites for the further adsorption and reduction of intermediate OOH. This work provides a new insight into the design of carbon materials for effective hydrogen peroxide electrochemical generation.

Interaction between the ins/IGF-1 and p38 MAPK signaling pathways in molecular compensation of sod genes and modulation related to intracellular ROS levels in C. elegans

Superoxide dismutases, which catalytically remove intracellular superoxide radicals by the disproportionation of molecular oxygen and hydrogen peroxide, are encoded by the sod-1 to -5 genes in the nematode C. elegans. Expression of the sod genes is mutually compensatory for the modulation of intracellular oxidative stress during aging. Interestingly, several-fold higher expression of the sod-1 to -4 was induced in a sod-5 deletion mutant, despite the low expression levels of sod-5 in wild-type animals. Consequently, this molecular compensation facilitated recovery of lifespan in the sod-5 mutant. In previous reports, two transcription factors DAF-16 and SKN-1 are associated with the compensatory expression of sod genes, which are downstream targets of the ins/IGF-1 and p38 MAPK signaling pathways activated under oxidative and heavy metal stresses, respectively. Here, we show that p38 MAPK signaling regulates induction of not only the direct expression of sod-1, -2 and -4 but also the indirect modulation of DAF-16 targets, such as sod-3 and -5 genes. Moreover, a SKN-1 target, the insulin peptide gene ins-5, partially mediates the expression of DAF-16 targets via p38 MAPK signaling. These findings suggest that the interaction of ins/IGF-1 and p38 MAPK signaling pathways plays an important role in the fine-tuning of molecular compensation among sod genes to protect against mitochondrial oxidative damage during aging.

Hydrogen Peroxide Disproportionation with Manganese Macrocyclic Complexes of Cyclen and Pyclen.

The catalase family of enzymes, which include a variety with a binuclear manganese active site, mitigate the risk from reactive oxygen species by facilitating the disproportionation of hydrogen peroxide into molecular oxygen and water. In this work, hydrogen peroxide disproportionation using complexes formed between manganese and cyclen or pyclen were investigated due to the spectroscopic similarities with the native MnCAT enzyme. Potentiometric titrations were used to construct speciation diagrams that identify the manganese complex compositions at different pH values. Each complex behaves as a functional mimic of catalase enzymes. UV-visible spectroscopic investigations of the H2O2 decomposition reaction yielded information about the structure of the initial catalyst and intermediates that include monomeric and dimeric species. The results indicate that rigidity imparted by the pyridine ring of pyclen is a key factor in increased TON and TOF values measured compared to cyclen.

urpose: The aim of this study was to investigate the cytotoxicity effect, iron chelator and antioxidant activities of iron (III) ions with curcumin ligand that may be used in the treatment of iron overload. Materials and Methods: The cytotoxic activities of the ligand and the complex were evaluated by the MTT assay. The SOD activity of the complex of curcumin was determined by using its ability to inhibit the reduction of NBT. The catalytic activity studies of Fe(III) complex in DMSO towards the disproportionation of hydrogen peroxide were also performed.Results: The IC50 values are found in 6.8 μM catalase activity was measured. Where at a concentration of 2.0 mM, the activity was equivalent to 183.30 U/L. The complex shows a catalase activity. The complex showed minimal toxicity. IC50 values found 5.3 mg/ml. The observed cytotoxicity could be pursued to obtain a potential drug. The iron chelator effects were determined by Ferrozine reagent. Curcumin, the most active extract interfered with the formation of ferrous and ferrozine complex. It demonstrated strong chelating activities. The result showed that the complexes possess considerable SOD activity. This finding indicates that the iron complex is capable of removing free radicals. Conclusion: The study results revealed that the iron(III) complex of curcumin with an appropriate potential drug may act as a protector against oxidative stress. Therefore, all results suggest that curcumin may represent a new approach in the treatment of iron overload.
