Dopaminergic Input Regulates the Sensitivity of Indirect Pathway Striatal Spiny Neurons to Brain-Derived Neurotrophic Factor.

Motor dysfunction in Parkinson's disease (PD) is closely linked to the dopaminergic depletion of striatal neurons and altered synaptic plasticity at corticostriatal synapses. Dopamine receptor D1 (DRD1) stimulation is a crucial step in the formation of long-term potentiation (LTP), whereas dopamine receptor D2 (DRD2) stimulation is needed for the formation of long-term depression (LTD) in striatal spiny projection neurons (SPNs). Tropomyosin receptor kinase B (TrkB) and its ligand brain-derived neurotrophic factor (BDNF) are centrally involved in plasticity regulation at the corticostriatal synapses. DRD1 activation enhances TrkB's sensitivity for BDNF in direct pathway spiny projection neurons (dSPNs). In this study, we showed that the activation of DRD2 in cultured striatal indirect pathway spiny projection neurons (iSPNs) and cholinergic interneurons causes the retraction of TrkB from the plasma membrane. This provides an explanation for the opposing synaptic plasticity changes observed upon DRD1 or DRD2 stimulation. In addition, TrkB was found within intracellular structures in dSPNs and iSPNs from Pitx3-/- mice, a genetic model of PD with early onset dopaminergic depletion in the dorsolateral striatum (DLS). This dysregulated BDNF/TrkB signaling might contribute to the pathophysiology of direct and indirect pathway striatal projection neurons in PD.

Receptor and Ionic Mechanism of Histamine on Mouse Dorsolateral Striatal Neurons.

The dorsolateral striatum (DLS) is the critical neural substrate that plays a role in motor control and motor learning. Our past study revealed a direct histaminergic projection from the tuberomammillary nucleus (TMN) of the hypothalamus to the rat striatum. However, the afferent of histaminergic fibers in the mouse DLS, the effect of histamine on DLS neurons, and the underlying receptor and ionic mechanisms remain unclear. Here, we demonstrated a direct histaminergic innervation from the TMN in the mouse DLS, and histamine excited both the direct-pathway spiny projection neurons (d-SPNs) and the indirect-pathway spiny projection neurons (i-SPNs) of DLS via activation of postsynaptic H1R and H2R, albeit activation of presynaptic H3R suppressed neuronal activity by inhibiting glutamatergic synaptic transmission on d-SPNs and i-SPNs in DLS. Moreover, sodium-calcium exchanger 3 (NCX3), potassium-leak channels linked to H1R, and hyperpolarization-activated cyclic nucleotide-gated channel 2 (HCN2) coupled to H2R co-mediated the excitatory effect induced by histamine on d-SPNs and i-SPNs in DLS. These results demonstrated the pre- and postsynaptic receptors and their downstream multiple ionic mechanisms underlying the inhibitory and excitatory effects of histamine on d-SPNs and i-SPNs in DLS, suggesting a potential modulatory effect of the central histaminergic system on the DLS as well as its related motor control and motor learning.

Pathway-Specific Remodeling of Thalamostriatal Synapses in a Mouse Model of Parkinson's Disease.

BackgroundThe network pathophysiology underlying the motor symptoms of Parkinson's disease (PD) is poorly understood. In models of late‐stage PD, there is significant cell‐specific remodeling of corticostriatal, axospinous glutamatergic synapses on principal spiny projection neurons (SPNs). Neurons in the centrolateral nucleus (CLN) of the thalamus that relay cerebellar activity to the striatum also make axospinous synapses on SPNs, but the extent to which they are affected in PD has not been definitively characterized.ObjectiveTo fill this gap, transgenic mice in which CLN neurons express Cre recombinase were used in conjunction with optogenetic and circuit mapping approaches to determine changes in the CLN projection to SPNs in a unilateral 6‐hydroxydopamine (6‐OHDA) model of late‐stage PD.MethodsAdeno‐associated virus vectors carrying Cre‐dependent opsin expression constructs were stereotaxically injected into the CLN of Grp‐KH288 mice in which CLN, but not parafascicular nucleus neurons, expressed Cre recombinase. The properties of this projection to identify direct pathway spiny projection neurons (dSPNs) and indirect pathway spiny projection neurons (iSPNs) were then studied in ex vivo brain slices of the dorsolateral striatum from control and 6‐OHDA lesioned mice using anatomic, optogenetic, and electrophysiological approaches.ResultsOptogenetically evoked excitatory synaptic currents in both iSPNs and dSPNs were reduced in lesioned mice; however, the reduction was significantly greater in dSPNs. In iSPNs, the reduction in evoked responses was attributable to synaptic pruning, because synaptic channelrhodopsin assisted circuit mapping (sCRACm) revealed fewer synapses per cell after lesioning. In contrast, sCRACm mapping of CLN inputs to dSPNs failed to detect any change in synapse abundance in lesioned mice. However, the ratio of currents through α‐amino‐3‐hydroxy‐5‐methyl‐4‐isoxazolepropionic acid receptors to those through N‐methyl‐D‐aspartate receptors was significantly reduced in dSPNs. Moreover, the distribution of currents evoked by optical stimulation of individual synapses shifted toward smaller amplitudes by lesioning, suggesting that they had undergone long‐term depression.ConclusionsTaken together, our results demonstrate that the CLN projection to the striatum undergoes a pathway‐specific remodeling that could contribute to the circuit imbalance thought to drive the hypokinetic features of PD. © 2022 The Authors. Enhanced GABAergic Inhibition of Cholinergic Interneurons in the zQ175+/- Mouse Model of Huntington's Disease.

Huntington's disease (HD) is an autosomal dominant neurodegenerative disorder that initially manifests itself in the striatum. How intrastriatal circuitry is altered by the disease is poorly understood. To help fill this gap, the circuitry linking spiny projection neurons (SPNs) to cholinergic interneurons (ChIs) was examined using electrophysiological and optogenetic approaches in ex vivo brain slices from wildtype mice and zQ175+/− models of HD. These studies revealed a severalfold enhancement of GABAergic inhibition of ChIs mediated by collaterals of indirect pathway SPNs (iSPNs), but not direct pathway SPNs (dSPNs). This cell-specific alteration in synaptic transmission appeared in parallel with the emergence of motor symptoms in the zQ175+/− model. The adaptation had a presynaptic locus, as it was accompanied by a reduction in paired-pulse ratio but not in the postsynaptic response to GABA. The alterations in striatal GABAergic signaling disrupted spontaneous ChI activity, potentially contributing to the network dysfunction underlying the hyperkinetic phase of HD.

Direct-Pathway Spiny Projection Neuron Inhibition Evokes Transient Circuit Imbalance Manifested as Rotational Behavior

The striatum collects and integrates information from many different areas of the brain and propels this forward to the basal ganglia (BG) output structures. In this way, the striatum is playing a pivotal role in control of voluntary movements and is implicated in debilitating movement disorders such as Parkinson’s disease. The functional backbone of the striatum is represented by direct pathway (dSPN) Drd1-expressing and indirect pathway (iSPN) Drd2-expressing spiny projection neurons (SPN), exerting opposite effects on movement. In rodent models of striatal function, unilateral dopamine deprivation is known to induce ipsilateral rotational behavior. To further study imbalance of the BG circuit and striatal domain influence on behavioral outcome, we employed a viral approach based on tetanus toxin light chain (TeLC) activity for permanent inhibition of dSPN activity in dorsomedial striatum (DMS). Cre-dependent TeLC injected unilaterally into the DMS of Drd1-Cre mice resulted in robust expression of TeLC in the dSPN cell populations as shown by immunohistochemistry. In the TeLC expressing mice, but not in control mice, we observed ipsilateral rotations that were enhanced upon administration of amphetamine to augment striatal dopamine levels. We argue that the observed single turns of ipsilateral rotations occur because of TeLC-mediated silencing of dSPN activity in one hemisphere, causing unresponsiveness to dopamine transients during movement initiation. This evokes a temporal BG circuit imbalance manifested as short bursts of rotations, particular evident during extrinsic dopaminergic modulation.

Optogenetic control of alcohol-seeking behavior via the dorsomedial striatal circuit

Alcohol consumption alters glutamatergic transmission in many brain regions, including the dorsomedial striatum (DMS); this aberrant plasticity is thought to be responsible for alcohol-seeking behavior. Recent studies reported that alcohol induced such plasticity specifically in direct pathway spiny projection neurons (dSPNs) of the DMS. However, it is unknown how this specific change contributes to alcohol-seeking behavior and relapse. Here, we first demonstrated that operant alcohol self-administration increased NMDA receptor activity in DMS dSPNs. Next, we found that optogenetic inhibition of dSPNs reversibly decreased operant lever presses for alcohol and alcohol intake. Furthermore, optogenetic stimulation of corticostriatal inputs at low and moderate frequencies induced reliable LTD in DMS slices. Surprisingly, in vivo delivery of the LTD-inducing protocol increased operant alcohol self-administration; this effect was blocked by a D2R antagonist. Importantly, LTD induction in the presence of both D1 and D2 receptor antagonists produced a long-lasting decrease in operant alcohol self-administration. Our results suggest that suppressing DMS dSPNs activity and their cortical inputs represents a novel treatment mechanism for alcohol use disorder.

Delayed Spine Pruning of Direct Pathway Spiny Projection Neurons in a Mouse Model of Parkinson's Disease.

In animal models of Parkinson’s disease (PD), principal striatal spiny projection neurons (SPNs) lose axospinous synapses. However, there has been a disagreement about whether this loss is restricted to a specific type of SPN or not, as some studies have reported pruning in both direct pathway SPNs and indirect pathway SPNs, while others have found this pruning to be restricted to indirect pathway SPNs. One possible explanation for the discrepancy is the period between the induction of the parkinsonian state and the assessment of spine loss. To test this hypothesis, transgenic mice were subjected to unilateral 6-hydroxydopamine (6-OHDA) lesions of nigrostriatal dopaminergic neurons and then direct pathway SPNs examined in ex vivo brain slices using two photon laser scanning microscopy either one or 2 months afterwards. These studies revealed that 1 month after the lesion, there was no loss of spines in direct pathway SPNs. However, 2 months after the lesion, spine loss was significant in direct pathway SPNs. In addition to reconciling the existing literature on the impact of the parkinsonian state on axospinous synapse elimination in SPNs, our results suggest that the delayed spine loss in direct pathway SPNs is not driven by homeostatic mechanisms [as posited for indirect pathway (iSPNs)], but rather by network pathophysiology.

Striatal ERK is Activated in Response to L‐DOPA Administration in a Knock‐in Mouse Model of DOPA‐Responsive Dystonia

Abnormal dopamine (DA) neurotransmission is associated with many dystonic disorders; however, the precise nature of the defects in DA transmission that result in dystonia are not known. To address this question, we used a knock‐in mouse model of L‐DOPA‐responsive dystonia (DRD). DRD mice display the core features of the human disorder, including reduced striatal DA levels and abnormal movement that improves in response to L‐DOPA, the precursor to DA. In the striatum, DA promotes movement by activating D1 receptors (D1Rs) expressed on direct pathway spiny projection neurons (SPNs) and inhibits movement by activating D2 receptors (D2Rs) expressed on indirect pathway SPNs. D1Rs stimulate adenylate cyclase (AC) activity and D2Rs inhibit AC; however, in DRD mice, D1Rs display an exaggerated AC response to D1 agonist stimulation and D2Rs exhibit a change in valence whereby D2R agonists increase AC. Our objective was to determine signaling alterations in both SPN subtypes downstream of AC, specifically phosphorylated ERK (p‐ERK), that contribute to the alleviation of dystonia by L‐DOPA to identify downstream targets that may serve as novel therapeutic targets. DRD and control mice were subcutaneously injected with L‐DOPA (10 mg/kg), the D1R agonist SKF 81297 (0.2 mg/kg), the D2R agonist quinpirole (0.1 mg/kg), or saline. Mice were perfused 45 min after drug treatment and striatal sections were immunostained for p‐ERK. DRD mice showed a significant and robust increase in p‐ERK levels in the dorsal striatum in response to L‐DOPA treatment compared to controls and saline‐treated mice. In contrast, SKF 81297 or quinpirole treatment evoked only modest changes in p‐ERK. The L‐DOPA specific increase in p‐ERK suggests that D1R and D2R co‐activation may regulate p‐ERK and/or that p‐ERK may serve as a coincidence detector for convergent DA and glutamatergic signaling that occurs when endogenous DA signaling is restored in the striatum. Kainate Receptors Inhibit Glutamate Release Via Mobilization of Endocannabinoids in Striatal Direct Pathway Spiny Projection Neurons.

Kainate receptors are members of the glutamate receptor family that function by both generating ionotropic currents through an integral ion channel pore and coupling to downstream metabotropic signaling pathways. They are highly expressed in the striatum, yet their roles in regulating striatal synapses are not known. Using mice of both sexes, we demonstrate that GluK2-containing kainate receptors expressed in direct pathway spiny projection neurons (dSPNs) inhibit glutamate release at corticostriatal synapses in the dorsolateral striatum. This inhibition requires postsynaptic kainate-receptor-mediated mobilization of a retrograde endocannabinoid (eCB) signal and activation of presynaptic CB1 receptors. This pathway can be activated during repetitive 25 Hz trains of synaptic stimulation, causing short-term depression of corticostriatal synapses. This is the first study to demonstrate a role for kainate receptors in regulating eCB-mediated plasticity at the corticostriatal synapse and demonstrates an important role for these receptors in regulating basal ganglia circuits.SIGNIFICANCE STATEMENT The GRIK2 gene, encoding the GluK2 subunit of the kainate receptor, has been linked to several neuropsychiatric and neurodevelopmental disorders including obsessive compulsive disorder (OCD). Perseverative behaviors associated with OCD are known to result from pathophysiological changes in the striatum and kainate receptor knock-out mice have striatal-dependent phenotypes. However, the role of kainate receptors in striatal synapses is not known. We demonstrate that GluK2-containing kainate receptors regulate corticostriatal synapses by mobilizing endocannabinoids from direct pathway spiny projection neurons. Synaptic activation of GluK2 receptors during trains of synaptic input causes short-term synaptic depression, demonstrating a novel role for these receptors in regulating striatal circuits.

M4 Muscarinic Receptor Signaling Ameliorates Striatal Plasticity Deficits in Models of L-DOPA-Induced Dyskinesia

A balanced interaction between dopaminergic and cholinergic signaling in the striatum is critical to goal-directed behavior. But how this interaction modulates corticostriatal synaptic plasticity underlying learned actions remains unclear--particularly in direct-pathway spiny projection neurons (dSPNs). Our studies show that in dSPNs, endogenous cholinergic signaling through M4 muscarinic receptors (M4Rs) promoted long-term depression of corticostriatal glutamatergic synapses, by suppressing regulator of G protein signaling type 4 (RGS4) activity, and blocked D1 dopamine receptor dependent long-term potentiation (LTP). Furthermore, in a mouse model of L-3,4-dihydroxyphenylalanine (L-DOPA)-induced dyskinesia (LID) in Parkinson's disease (PD), boosting M4R signaling with positive allosteric modulator (PAM) blocked aberrant LTP in dSPNs, enabled LTP reversal, and attenuated dyskinetic behaviors. An M4R PAM also was effective in a primate LID model. Taken together, these studies identify an important signaling pathway controlling striatal synaptic plasticity and point to a novel pharmacological strategy for alleviating LID in PD patients.

Cell type-specific plasticity of striatal projection neurons in parkinsonism and L-DOPA-induced dyskinesia.

SummaryThe striatum is widely viewed as the fulcrum of pathophysiology in Parkinson's disease (PD) and L-DOPA-induced dyskinesia (LID). In these disease states, the balance in activity of striatal direct pathway spiny projection neurons (dSPNs) and indirect pathway spiny projection neurons (iSPNs) is disrupted, leading to aberrant action selection. However, it is unclear whether countervailing mechanisms are engaged in these states. Here we report that iSPN intrinsic excitability and excitatory corticostriatal synaptic connectivity were lower in PD models than normal; L-DOPA treatment restored these properties. Conversely, dSPN intrinsic excitability was elevated in tissue from PD models and suppressed in LID models. Although the synaptic connectivity of dSPNs did not change in PD models, it fell with L-DOPA treatment. In neither case, however, was the strength of corticostriatal connections globally scaled. Thus, SPNs manifested homeostatic adaptations in intrinsic excitability and in the number but not strength of excitatory corticostriatal synapses.

In vivo Photometry during Movement Changes Our Understanding of the Direct and Indirect Pathways

The role of the direct and indirect pathways in movement and their dysfunction in the pathophysiology of most movement disorders is difficult to grasp for clinicians. The current model whereby the direct pathway has a role akin to the accelerator when driving and the indirect to the brake makes sense at a basic level for many disorders of movement. However, some clinical phenomenology such as hemiballismus are difficult to reconcile with the current model. Cui et al. (1), using an elegant experimental paradigm employing photometric methods in vivo in a mouse model have observed the moving brain at work and have attempted to disentangle the complex network of spiny projection neurons involved in the direct and indirect pathways. The authors reflect on the prevailing model whereby two distinct projection circuits composed of spiny projection neurons in the striatum act in an opposing fashion to either promote movement (direct pathway) or inhibit movement (indirect pathway). The anatomical location of these pathways is the striatum. Surmeier in an accompanying editorial further identifies the striatum as the seat of “past experiences” and when “asked” by cortex what way to proceed, the current model which he identifies as intrinsically binary sends “go” or “no-go” instructions to cortex, via the thalamus (2). Cui et al. reflect on the fact that despite increased understanding of basal ganglia circuitry, we still do not understand the physiological relationship and interactions between these pathways. To do so we need to observe the activity of the direct and indirect pathway spiny projection neurons in “behaving” animal models (1). However, it is difficult to distinguish direct and indirect spiny projection neuron firing using electrophysiological recordings and antidromic stimulation is unreliable and difficult due to “cross talk” between axons of both pathways (1). The authors highlight that optogenetic technology in vivo facilitates the identification of specific cell types and the monitoring of activity of specific cell types (1). Limitations of this technology include the requirement for head restraint, which limits the study of structures involved in movement and the technology does not penetrate to subcortical structures therein limiting the ability to study the basal ganglia (1). Cui et al. developed an experimental paradigm utilizing in vivo photometry to optically record neural activity within the striatum during free movement in a mouse model (1). They implanted a fiber-optic probe to deliver laser pulses and record emitted photons in the dorsal striatum over time. The optical indicator was a genetically encoded calcium indicator (GECI), selectively expressed in the direct pathway spiny projection neurons by injection with a recombinant viral vector to produce D1-Cre mice (2). A similar technique was used to selectively express the indicator in the indirect pathway spiny projection neurons of A2A-Cre mice (1). Focusing specifically on their examination of the relationship between activity in the direct and indirect pathway spiny projection neurons during movement, the authors measured fluorescence whilst mice performed a lever-pressing task. They demonstrated time-locked increases in fluorescence during the initiation of the task with a similar temporal profile in both the direct and indirect pathways in all of the mice (1). They identified transient increases in fluorescence throughout the completion of different tasks and in addition during the monitoring sessions, mice were exposed to a series of active states and periods of inactivity (1). Using the classical direct and indirect pathway model it would be logical to hypothesize that during the active periods there should be relatively more transients identified in the direct pathway spiny projection neurons compared to the indirect and during periods of inactivity there should be more transients identified in the indirect spiny projection neurons compared to the direct (1). Cui et al. identified increased activity in both direct and indirect pathways during movement and silence of both pathways during periods of inactivity. These results question the accelerator-brake metaphor applied to our current understanding of the interaction between the direct and indirect pathways. Further analysis of self-paced movement initiation toward the contralateral side demonstrated co-activation of both the direct and indirect pathways and furthermore activity in both pathways preceded movement initiation. Activity in both pathways predicted the occurrence of specific movements within 500 ms of transient initiation. In the accompanying editorial it is suggested that rather than giving sequential “go” or “no-go” recommendations, the striatum is simultaneously making recommendations about “what to do and what not to do” (2). Through the interrogation of the direct and indirect pathways in vivo and during movement, this study builds upon and changes our current understanding of the function of the basal ganglia. Whilst the results challenge the current concepts used to explain movement, further work is required including lesional studies to translate this theory into the clinical environment in order to explain the clinical phenomenology we see at the bedside.
