Effect of partial O-methylation in dehydrodieugenol on its antitrypanosomal activity - correlation with the toxicity using cell membrane models

Biseugenol (1), a neolignan with antiprotozoal activity against Trypanosoma cruzi, was partially methylated, and the compound obtained – methyl biseugenol (2) – had its activity evaluated against the extracellular (trypomastigotes) and intracellular (amastigotes) forms of T. cruzi. It was observed that both compounds 1 and 2 exhibited similar effects against trypomastigotes (IC50 of 11.7 and 16.2 μM, respectively), whereas compound 2 displayed higher activity against amastigotes (IC50 = 8.2 μM) in comparison with biseugenol (IC50 = 15.4 μM). Additionally, reduced toxicity against NCTC cells for compound 2 was observed (CC50 > 200 μM), differently from compound 1 with CC50 = 58.0 μM. Aiming to understand better the molecular mechanism of the biological action of compound 2, the prodrug was incorporated into cellular membrane models constituted of Langmuir monolayers of the lipids dipalmitoylphosphatidylcholine (DPPC), dipalmitoylphosphatidylethanolamine (DPPE), dipalmitoylphosphatidylserine (DPPS), and dipalmitoylphosphatidylglycerol (DPPG). The lipid-drug interaction was inferred through tensiometry, surface potential, infrared spectroscopy (PM-IRRAS), and Brewster angle microscopy (BAM). The prodrug expanded DPPC and DPPG monolayers and condensed DPPE ones, as well as presented characteristic behaviors regarding the chemical structure of the lipid considering expansion-compression curves, surface potential-area isotherms, and stability of previously compressed monolayers to relevant-biological surface pressures. PM-IRRAS indicated a molecular disorder for DPPC and DPPS alkyl chains in the presence of the drug. BAM revealed the presence of domains in the DPPG and DPPE monolayers, which was probably induced by the prodrug. These data suggest, in general, that the lipid composition modulates the interaction of compound 2, whose results are expected to correlate to its trypanocidal activity, which involves the plasma membrane of T. cruzi as the primary target, i.e., the first barrier that the compound should encounter to interact with the microorganism.

How do antimicrobial peptides disrupt the lipopolysaccharide membrane leaflet of Gram-negative bacteria?

HypothesisIt is widely regarded that antimicrobial peptides (AMPs) kill bacteria by physically disrupting microbial membranes and causing cytoplasmic leakage, but it remains unclear how AMPs disrupt the outer membrane (OM) of Gram-negative bacteria (GNB) and then compromise the inner membrane. We hypothesise that different AMPs impose different structural disruptions, with direct implications to their antimicrobial efficacies. ExperimentsThe antimicrobial activities of three typical AMPs, including the designed short AMP, G3, and two natural AMPs, melittin and LL37, against E. coli and their haemolytic activities were studied. Lipopolysaccharide (LPS) and anionic di-palmitoyl phosphatidyl glycerol (DPPG) monolayer models were constructed to mimic the outer membrane and inner membrane leaflets of Gram-negative bacteria. The binding and penetration of AMPs to the model lipid monolayers were systematically studied by neutron reflection via multiple H/D contrast variations. FindingG3 has relatively high antimicrobial activity, low cytotoxicity, and high proteolytic stability, whilst melittin has significant haemolysis and LL37 has weaker antimicrobial activity. G3 could rapidly lyse LPS and DPPG monolayers within 10–20 min. In contrast, melittin was highly active against the LPS membrane, but the dynamic process lasted up to 80 min, with excessive stacking in the OM. LL37 caused rather weak destruction to LPS and DPPG monolayers, leading to massive adsorption on the membrane surface without penetrating the lipid tail region. These findings demonstrate that the rationally designed AMP G3 was well optimised to impose most effective destruction to bacterial membranes, consistent with its highest bactericidal activity. These different interfacial structural features associated with AMP binding shed light on the future development of active and biocompatible AMPs for infection and wound treatments.

Interactions of l-arginine with Langmuir monolayers of common phospholipids at the air-water interface

The interactions of l-arginine (l-arg) with Langmuir monolayers of three most common phospholipids, which are sodium salt of dipalmitoylphosphatidylglycerol (DPPG), dipalmitoylphosphatidylcholine (DPPC) and dipalmitoylphosphatidylethanolamine (DPPE), have been investigated at the air-water interface. The surface pressure-area (π-A) isotherms of these monolayers have been measured with a film balance and monolayer morphology has been observed by a Brewster angle microscopy (BAM). The DPPG monolayers on pure water do not show any phase transition but show irregular shaped condensed phases formed just after evaporation of the solvent at 20 °C. However, this monolayer on l-arg solution subphase indicates a first-order phase transition from liquid expanded to liquid condensed (LE-LC) phases and forms LC domains at the same temperature. With an increase in the l-arg concentration in the subphase up to 5.0 × 10−4 M, the π-A shows an overall increasingly greater expansion in the molecular area. All of the π-A isotherms recorded on ≥5.0 × 10−4 M l-arg solution subphases almost coincide with each other. These changes in the phase behavior have been explained by the fact that l-arg having guanidinium cationic group undergoes strong hydrogen bonding interaction with the anionic phosphatidylglycerol (PG-) head group. The bonding between two molecules is further strengthened by electrostatic attraction between cationic l-arg and anionic PG- ions. The BAM observation of the monolayer morphology supports this explanation. On the other hand, a very negligible interaction has been observed between l-arg and DPPC or DPPE monolayers. The π-A isotherms in the presence of l-arg for both the amphiphiles show a very little expansion only in the LE phase region, but coincide in the so called solid phase region. The monolayer morphology of both the monolayers also supports these results. This little effect of expansion in the LE region may be explained by the ion-pair formation between cationic l-arg and anionic head groups in the monolayers at lower pressures. However, due to compression at high pressure, the l-arg molecules are squeezed out from the amphiphile head groups.

Evaluation of the effects in cellular membrane models of antitrypanosomal poly-thymolformaldehyde (PTF) using Langmuir monolayers

The polymerization of bioactive compounds may be interesting because the supramolecular structures formed can boost biological action on microorganism membranes. In the present work, poly-thymolformaldehyde (PTF) activity, prepared by condensation of thymol and formaldehyde, was evaluated against trypomastigote forms of Trypanosoma cruzi and related with the physicochemical changes provided by the incorporation of the compound in protozoan cell membrane models. PTF exhibited an EC50 value of 23.4 μg/mL and no toxicity against mammalian cells (CC50 > 200 μg/mL). To understand the molecular action of PTF as an antiprotozoal candidate, this compound was incorporated in Langmuir monolayers of dipalmitoylphosphatidylglycerol (DPPG) as a model for parasite cell membranes. PTF shifted DPPG surface pressure-area isotherms to higher areas, indicating its incorporation in the lipid films. Additionally, it changed the thermodynamic, compressional, structural, and morphological properties of the floating monolayers, decreasing the collapse pressure, reducing the surface elasticity, and segregating molecules at the interface, forming domains with different reflectivities. Infrared spectroscopy showed that the lipid films with PTF presented an increased rate of gauche/all-trans conformers for the methylene groups from the acyl chains, indicating molecular disorder. Therefore, these results show that PTF alters the physicochemical properties of DPPG monolayers as a model for protozoa cell membranes, which can enhance the comprehension of the parasitic action of PTF against T. cruzi.

Interaction of Short Pentavalent Cationic Peptides with Negatively Charged DPPG Monolayers and Bilayers: Influence of Peptide Modifications on Binding.

The binding of oligopeptides with the structure (RX)4R and (KXX)4K, with X being the amino acid G or A, to lipid monolayers and bilayers of dipalmitoyl-phosphatidylglycerol (DPPG) was studied and compared to the binding effects of peptides with the structure (KX)4K. The monolayer adsorption experiments again showed the superposition of condensation effects due to charge compensation and insertion of amino acid side chains leading to expansion of the monolayer. The latter effect was enhanced when glycine was replaced by alanine. The thermotropic phase behavior of dipalmitoyl-phosphatidylglycerol (DPPG) bilayer membranes and their mixtures with short cationic model peptides was investigated by differential scanning calorimetry and infrared spectroscopy. Increasing the charge distance of the lysine residues in the series (K)5, (KG)4K, and (KGG)4K results in an upshift of the main phase transition of DPPG up to 5 K, as predicted for pure electrostatic binding. All peptides exhibit only unordered structures in bulk solution as well as when bound to DPPG bilayers. (KGG)4K additionally shows a high propensity of turn structures due to its flexibility. The exchange of glycine by alanine in (KAA)4K leads only to a marginal increase in Tm, in contrast to the binding of (KA)4K where the formation of intervesicular antiparallel β-sheets occurs, leading to a much more pronounced stabilization of the gel phase. This shows that the sequence and flexibility of the oligopeptides has an important influence on the formation of secondary structures bound to the bilayers. Binding of (RX)4R peptides to DPPG bilayers has almost no influence on the lipid phase transition in bilayers. Here, condensation and insertion effects almost compensate, as the results of monolayer experiments show. This is due to the higher propensity of arginine side chains to insert into the lipid headgroup region.

Differences in the Aspect Ratio of Gold Nanorods that Induce Defects in Cell Membrane Models.

Understanding the interactions between biomolecules and nanomaterials is of great importance for many areas of nanomedicine and bioapplications. Although studies in this area have been performed, the interactions between cell membranes and nanoparticles are not fully understood. Here, we investigate the interactions that occur between the Langmuir monolayers of dipalmitoylphosphatidyl glycerol (DPPG) and dipalmitoylphosphatidyl choline (DPPC) with gold nanorods (NR)-with three aspect ratios-and gold nanoparticles. Our results showed that the aspect ratio of the NRs influenced the interactions with both monolayers, which suggest that the physical morphology and electrostatic forces govern the interactions in the DPPG-NR system, whereas the van der Waals interactions are predominant in the DPPC-NR systems. Size influences the expansion isotherms in both systems, but the lipid tails remain conformationally ordered upon expansion, which suggests phase separation between the lipids and nanomaterials at the interface. The coexistence of lipid and NP regions affects the elasticity of the monolayer. When there is coexistence between two phases, the elasticity does not reflect the lipid packaging state but depends on the elasticity of the NP islands. Therefore, the results corroborate that nanomaterials influence the packing and the phase behavior of the mimetic cell membranes. For this reason, developing a methodology to understand the membrane-nanomaterial interactions is of great importance.

Experimental evidence for the mode of action based on electrostatic and hydrophobic forces to explain interaction between chitosans and phospholipid Langmuir monolayers

The interaction between chitosans and Langmuir monolayers mimicking cell membranes has been explained with an empirical scheme based on electrostatic and hydrophobic forces, but so far this has been tested only for dimyristoyl phosphatidic acid (DMPA). In this paper, we show that the mode of action in such a scheme is also valid for dipalmitoyl phosphatidyl choline (DPPC) and dipalmitoyl phosphatidyl glycerol (DPPG), whose monolayers were expanded and their compressibility modulus decreased by interacting with chitosans. In general, the effects were stronger for the negatively charged DPPG in comparison to DPPC, and for the low molecular weight chitosan (LMWChi) which was better able to penetrate into the hydrophobic chains than the high molecular weight chitosan (Chi). Penetration into the hydrophobic chains was confirmed with polarization-modulated infrared reflection absorption spectroscopy (PM-IRRAS) and sum frequency generation (SFG) spectroscopy. A slight reduction in conformational order of the lipid chains induced by the chitosans was quantitatively estimated by measuring the ratio between the intensities of the methyl (r+) and methylene (d+) peaks in the SFG spectra for DPPG. The ratio decreased from 35.6 for the closely packed DPPG monolayer to 7.0 and 6.6 for monolayers containing Chi and LMWChi, respectively. Since in both cases there was a significant phospholipid monolayer expansion, the incorporation of chitosans led to chitosan-rich and lipid-rich condensed domains, which mantained conformational order for their hydrophobic tails. The stronger effects from LMWChi are ascribed to an easier access to the hydrophobic tails, as corroborated by measuring aggregation in solution with dynamic light scattering, where the hydrodynamic radius for LMWChi was close to half of that for Chi. Taken together, the results presented here confirm that the same mode of action applies to different phospholipids that are important constituents of mammalian (DPPC) and bacterial (DPPG) cell membranes.

Understanding the biocide action of poly(hexamethylene biguanide) using Langmuir monolayers of dipalmitoyl phosphatidylglycerol

The disinfectant activity of poly(hexamethylene biguanide) (PHMB) has been explored in industrial applications, in agriculture and in food manipulation, but this biocide action is not completely understood. It is believed to arise from electrostatic interactions between the polyhexanide group and phosphatidylglycerol, which is the main phospholipid on the bacterial membrane. In this study, we investigated the molecular-level interactions between PHMB and dipalmitoyl phosphatidylglycerol (DPPG) in Langmuir monolayers that served as cell membrane models. PHMB at a concentration of 2×10−4gL−1 in a Theorell–Stenhagen at pH 3.0 and in a phosphate at pH 7.4 was used as a subphase to prepare the DPPG monolayers. Surface pressure–area isotherms showed that PHMB adsorbs and penetrates into the DPPG monolayers, expanding them and increasing their elasticity under both conditions examined. Results from polarization-modulated infrared reflection absorption spectroscopy (PM-IRRAS) indicated that PHMB induces disorder in the DPPG chains and dehydrates their CO groups, especially for the physiological medium. Overall, these findings point to hydrophobic interactions and dehydration being as relevant as electrostatic interactions to explain changes in membrane fluidity and permeability, believed to be responsible for the biocide action of PHMB.

Chitosan as a subphase disturbant of membrane lipid monolayers. The effect of temperature at varying pH: I. DPPG

Temperature-mediated effects of chitosan dissolved in acetate buffer at different pHs, on the structural and thermodynamic characteristics of dipalmitoylphosphatidylglycerol (DPPG) monolayers, employed here as a model of the bacterial cell membrane, were investigated. The investigation was done to improve the understanding if in its antibacterial activity chitosan disturbs the bacterial cell membrane. The Langmuir film technique was employed, where the compression surface pressure–area (π–A) isotherms of DPPG monolayer formed at the air-buffer interface were measured at five temperatures in the range 15–37°C for three pHs in the range 3.5–6.0, with the chitosan concentration in the subphase varied between 0.002 and 1mg/mL. Illustrating the extent of DPPG–chitosan interactions, the characteristics of the monolayers on the chitosan-containing solutions were assessed by monitoring differences in the course of the π–A isotherms relative to pure DPPG films. The characteristics revealed that chitosan interacted with the lipid film not only superficially but also inserted to a certain degree into the film. Increasing temperature enhanced these effects, the more strongly, the lower was the pH. Furthermore, the transition of the monolayer from the liquid-expanded to liquid-condensed phase was found to be an endothermic process accompanied by an increase in disorder. The effects were most pronounced at pH 3.5, and were markedly enhanced by chitosan. Most interestingly, the analysis of the critical temperatures provided evidence that the interaction of chitosan with DPPG monolayers is not only due to electrostatic but also to non-electrostatic contributions. The most effective disturbing effects of chitosan on DPPG monolayer were observed at the highest temperature 37°C applied at pH 3.5.

Probing the interaction of oppositely charged gold nanoparticles with DPPG and DPPC Langmuir monolayers as cell membrane models

The growing use of nanoparticles in a variety of applications calls for detailed studies of their toxicology, which in turn require understanding the interactions between nanoparticles and living cells. Since simulating the interaction with real cell membranes is rather complex, Langmuir monolayers (LMs) have been used to mimic the first barrier encountered by a nanoparticle as it approaches a biological membrane to assess molecular-level interactions. In this study, we show how oppositely charged gold nanoparticles (Au-NPs) interact with monolayers of the zwitterionic dipalmitoylphosphatidyl choline (DPPC) and negatively charged dipalmitoylphosphatidyl glycerol (DPPG). The monolayers were spread on subphases containing two concentrations of either negatively charged Au-NPs coated with citrate anions or positively charged Au-NPs functionalized with the cationic polyelectrolyte poly(allylamine hydrochloride) (PAH). For DPPG, electrostatic effects dominated which depended strongly on the NPs capping agent, being obviously larger for the positive nanoparticles. The in-plane elasticity for DPPG monolayers within the surface pressure range corresponding to real cell membranes increased with adsorption of positively charged NPs, but decreased with the negative ones. For the zwitterionic DPPC, on the other hand, significant effects only occurred for negatively charged NPs, including a decrease in elasticity. Therefore, it is concluded that the nature, namely the charge of the capping agents, is crucial for the interaction of charged NPs with the cell membrane.

Interaction of toremifene with dipalmitoyl-phosphatidyl-glycerol in monolayers at the air–water interface followed by fluorescence microscopy in Langmuir–Blodgett films

Langmuir monolayers of dipalmitoyl-phosphatidyl-glycerol (DPPG) containing toremifene (TOR), an antiestrogen drug derivative of tamoxifen, were prepared in order to study the interaction of the drug with the cell membrane. TOR is not surface active but it remains at the interface in DPPG rich monolayers anchored by electrostatic interaction with the anionic DPPG up to the equimolar composition. The fluidity of mixed monolayers increases up to the TOR mole fraction XTOR=0.3, remaining practically invariant for higher compositions. Brewster angle microscopy shows that the TOR disturbs the DPPG organization and the liquid condensed (LC) domains of DPPG become undetectable for XTOR≥0.4. Laser scanning confocal fluorescence microscopy images of the LB films doped with rhodamine B-piperazine amide dye confirm the progressive reduction in size of LC domains, from which TOR and rhodamine are excluded. The incorporation of TOR in DPPG monolayers up to the equimolar composition supports the formation of a TOR:DPPG complex (1:1) due to electrostatic interactions between the negatively charged polar groups of DPPG and protonated TOR.

Effect of thiol-functionalisation on chitosan antibacterial activity: Interaction with a bacterial membrane model

The antibacterial activity of chitosan modified with the thiol-containing 2-iminothiolane HCl (TC-IMI) and N-acetyl-l-cysteine (TC-NAC) was studied by Langmuir film balance technique using a dipalmitoylphosphatidylglycerol (DPPG) monolayer bacterial membrane model. The interactions of the biopolymer with the membrane model were assessed by monitoring the differences in the shape of the compression isotherms recorded in the absence and presence of chitosan and thiolated conjugates in the subphase. A low molecular weight chitosan (15kDa) shifted the compression isotherms of DPPG monolayers towards larger areas (A0,CS=145Å2), confirming its membrane disturbance capacity. Further thiolation induced higher yield of expansion, more pronounced in the case of TC-IMI. The expansion of the monolayer increased significantly (A0,TC-NAC=150Å2vs A0,TC-IMI=175Å2) and the elasticity at a surface pressure of 30mN/m, typical for bio-membranes decreased to a greater extent Cs,30TC-NAC-1=120mN/mvs Cs,30TC-IMI-1=87mN/m) in presence of TC-IMI. Antibacterial tests against a Gram-negative Escherichia coli and a Gram-positive Staphylococcus aureus were in good agreement with these findings, suggesting that chitosan thiolated with 2-iminothiolane HCl acts as a bactericide disrupting the integrity of the bacterial cell membrane.

Adsorption behaviors and structural transitions of organic cations on an anionic lipid monolayer at the air–water interface

We studied the adsorption behaviors of organic cations, malachite green (MG), on an anionic dipalmitoylphosphatidylglycerol (DPPG) monolayer at the air–water interface for different thermodynamic phases of the lipid monolayer. Simultaneous monitoring of both the surface pressure and the optical second harmonic generation intensity, combined with the fluorescence microscopy and X-ray reflectivity measurement, gave detailed structural information on the adsorbed MG, showing three distinct types of adsorption behaviors depending on the initial phase of the monolayer. We provided plausible molecular models for the vertical position and orientation of MG in each phase: penetration into the DPPG monolayer with a vertical one direction at a fluid phase; vertically but oppositely paired orientations at a more condensed phase; parallel adsorption at the interface near the lipid headgroup region at a highly packed condensed phase. This result can be usefully expanded for the understanding of the penetration mechanism and conformational change of biological adsorbates at the cellular membrane.

The role of the C-terminal region of pulchellin A-chain in the interaction with membrane model systems

Pulchellin is a Ribosome Inactivating Protein containing an A-chain (PAC), whose toxic activity requires crossing the endoplasmic reticulum (ER) membrane. In this paper, we investigate the interaction between recombinant PAC (rPAC) and Langmuir monolayers of dipalmitoyl phosphatidyl glycerol (DPPG), which served as membrane model. Three catalytically active, truncated PACs with increasing deletion of the C-terminal region, possessing 244, 239 and 236 residues (rPAC244, rPAC239 and rPAC236), were studied. rPAC had the strongest interaction with the DPPG monolayer, inducing a large expansion in its surface pressure–area isotherm. The affinity to DPPG decreased with increased deletion of the C-terminal region. When the C-terminal region was deleted completely (rPAC236), the interaction was recovered, probably because other hydrophobic regions were exposed to the membrane. Using Polarization Modulated-Infrared Reflection Absorption Spectroscopy (PM-IRRAS) we observed that at a bare air/water interface rPAC comprised mainly α-helix structures, the C-terminal region had unordered structures when interacting with DPPG. For rPAC236 the α-helices were preserved even in the presence of DPPG. These results confirm the importance of the C-terminal region for PAC-ER membrane interaction. The partial unfolding only with preserved C-terminal appears a key step for the protein to reach the cytosol and develop its toxic activity.

Probing the Modes of Antibacterial Activity of Chitosan. Effects of pH and Molecular Weight on Chitosan Interactions with Membrane Lipids in Langmuir Films

Chitosan, a cationic biopolymer derived from chitin, has been described as having antibacterial activity. The modes of this activity, however, have not been established. One mode proposed is that chitosan perturbs bacterial cell membranes. To validate this proposal, in this study we investigated chitosan interactions with lipids in Langmuir monolayers as model membranes. The interactions were assessed by monitoring differences in the shape of the compression isotherms measured in the absence and presence of chitosan in the subphase (acetate buffer). To appraise the contribution of electrostatic interactions versus hydrogen bonding and hydrophobic interactions, three membrane lipids differing in charge were studied-anionic dipalmitoylphosphatidylglycerol (DPPG), zwitterionic dipalmitoylphosphatidylcholine (DPPC), and neutral cholesterol-and the pH of the subphase was varied between 3.5 and 6.0. In addition, the impact of the molecular weight of chitosan on the interactions was assessed at pH 3.5. It was found that while chitosan had a negligible effect on DPPC monolayers over the pH range studied, it distinctly affected DPPG and cholesterol monolayers. The effect on DPPG was found to decrease with increasing pH, that at pH 3.5 being ascribed to the charge-mediating action of chitosan on the local ionic environment and that at higher pHs to the intercalation of chitosan to the monolayers. Practically independent of pH, the effect of chitosan on cholesterol was accounted for by the formation of cholesterol-chitosan hydrogen bonds. Chitosan of lower molecular weight facilitated the interactions with all the three lipids studied. The results obtained may be helpful in identifying the mode of antibacterial activity of chitosan versus other modes that involve the disturbance of cell life cycles.

Influence of the lipid composition of biomimetic monolayers on the structure and orientation of the gp41 tryptophan-rich peptide from HIV-1

The tryptophan-rich peptide of gp41 (so-called gp41W), one of the two envelope glycoproteins of HIV-1, is known to play a crucial role in the fusion between this virus and the host cell membranes. The influence of lipids on this role was investigated using different lipid monolayers at the air–water interface. Gp41W affinity for the lipid monolayer was measured by following the peptide-induced variation in the lateral surface pressure and we demonstrated that gp41W binds to monolayers containing the saturated zwitterionic dipalmitoylphosphatidylcholine (DPPC) as well as to the anionic dipalmitoylphosphatidylglycerol (DPPG) and to mixed monolayers containing DPPC and cholesterol (Chol). The secondary structure of gp41W in the presence of these lipid monolayers was determined by polarization modulation infrared reflection absorption spectroscopy (PM-IRRAS). The data showed that gp41W was an oriented α-helix in the presence of DPPG. However this spectroscopic method was unable to detect the gp41W structure in the presence of DPPC and DPPC/Chol monolayer. The peptide-induced modifications of the DPPC/Chol, DPPC and DPPG monolayer morphology were analyzed by Brewster angle microscopy (BAM). The peptide-induced changes in the DPPG monolayer morphology suggest that gp41W disturbed the lipid intermolecular interactions. Furthermore the peptide delayed the condensed state of DPPC and DPPC/Chol, indicating that, although gp41W was not detected by PM-IRRAS, it was present in these lipid monolayers.

Fluid and Condensed ApoA-I/Phospholipid Monolayers Provide Insights into ApoA-I Membrane Insertion

Apolipoprotein A-I (ApoA-I) is a protein implicated in the solubilization of lipids and cholesterol from cellular membranes. The study of ApoA-I in phospholipid (PL) monolayers brings relevant information about ApoA-I/PL interactions. We investigated the influence of PL charge and acyl chain organization on the interaction with ApoA-I using dipalmitoyl-phosphatidylcholine, dioleoyl-phosphatidylcholine and dipalmitoyl-phosphatidylglycerol monolayers coupled to ellipsometric, surface pressure, atomic force microscopy and infrared (polarization modulation infrared reflection–absorption spectroscopy) measurements. We show that monolayer compressibility is the major factor controlling protein insertion into PL monolayers and show evidence of the requirement of a minimal distance between lipid headgroups for insertion to occur, Moreover, we demonstrate that ApoA-I inserts deepest at the highest compressibility of the protein monolayer and that the presence of an anionic headgroup increases the amount of protein inserted in the PL monolayer and prevents the steric constrains imposed by the spacing of the headgroup. We also defined the geometry of protein clusters into the lipid monolayer by atomic force microscopy and show evidence of the geometry dependence upon the lipid charge and the distance between headgroups. Finally, we show that ApoA-I helices have a specific orientation when associated to form clusters and that this is influenced by the character of PL charges. Taken together, our results suggest that the interaction of ApoA-I with the cellular membrane may be driven by a mechanism that resembles that of antimicrobial peptide/lipid interaction.

Interaction of a partially fluorinated alcohol (F8H11OH) with biomembrane constituents in two-component monolayers

The interaction of (perfluorooctyl)undecanol (F8H11OH) with dipalmitoylphosphatidylcholine (DPPC), dipalmitoylphosphatidylglycerol (DPPG), and cholesterol (Ch) has been investigated in two-component monolayers at the air–water interface. The thermodynamic properties and phase behavior of the monolayers have been systematically elucidated employing isotherm measurements and several methods at the mesoscopic and microscopic scales. Addition of F8H11OH induces strong condensation in DPPC and DPPG monolayers. The magnitude for the Gibbs free energy and the interaction parameter between the two components for the DPPC/F8H11OH and DPPG/F8H11OH systems are twice as large as those for the Ch/F8H11OH system. Judging from the evidence that disordered/ordered transition pressures and collapse pressures for the binary monolayers change with molar fraction of F8H11OH, F8H11OH was (partially) miscible with all the compounds investigated. The miscibility of the two components in the monolayer is also supported by the in situ (Brewster angle and fluorescence microscopies) and ex situ (atomic force microscopy after transfer on a mica plate) morphological aspects. The present study reports firstly on the interaction between Ch and the fluorinated amphiphile by simply employing the monolayer technique. The molecular function of Ch in pulmonary surfactants and lipid rafts is now extensively investigated. To our knowledge, Ch does not interact strongly with lipid components in biomembranes. However, this study indicates that the partially fluorinated amphiphile has some affinity for Ch. This study provides a fundamental insight into the molecular function of biomembrane components and support for biomedical use of fluorinated materials.

Microphase Separation in Mixed Monolayers of DPPG with a Double Hydrophilic Block Copolymer at the Air−Water Interface: A BAM, LSCFM, and AFM Study

Phase separation and interactions in mixed monolayers of dipalmitoylphosphatidylglycerol (DPPG) with the rhodamine B end-labeled double-hydrophilic block copolymer (DHBC), poly(N,N-dimethylacrylamide)-block-poly(N,N-diethylacrylamide) (RhB-PDMA(207)-b-PDEA(177)), was studied at the air-water interface. Surface pressure versus area isotherms indicate that both components behave almost independently. Brewster angle microscopy (BAM) images show a random distribution of liquid condensed (LC) domains of DPPG in an apparent homogeneous matrix of DHBC, excluding the macroscopic phase separation. The laser scanning confocal fluorescence microscopy (LSCFM) of the rhodamine dye at the end of the PDMA chain showed how the DHBC is distributed in Langmuir-Blodgett (LB) mixed monolayers. The high spatial resolution of atomic force microscopy (AFM) combined with the LCSFM images indicates that DHBC incorporates in the expanded phase of DPPG to form mixed domains, being excluded from the condensed regions. Upon compression, nanosized LC domains of DPPG nucleate inside the mixed domains corralled in the nanopatterning of pure DHBC. The negatively charged polar group of DPPG inhibits rhodamine aggregation, while the long polymer chains promote the formation of corralled nanodomains of DPPG in two dimensions.

Enzyme Activity of Catalase Immobilized in Langmuir−Blodgett Films of Phospholipids

A major challenge for producing low cost biosensors based on nanostructured films with control of molecular architectures is to preserve the catalytic activity of the immobilized biomolecules. In this study, we show that catalase (HRP) keeps its activity if immobilized in Langmuir-Blodgett (LB) films of dipalmitoyl phosphatidylglycerol (DPPG). The incorporation of catalase into a DPPG monolayer at the air-water interface was demonstrated with surface pressure and surface potential isotherms, in addition to polarization-modulated infrared reflection absorption spectroscopy (PM-IRRAS). According to the PM-IRRAS data, catalase was not denatured upon adsorption on a preformed DPPG monolayer and could be transferred onto a solid substrate. The catalytic activity of catalase in a mixed LB film with DPPG was ca. 13% higher than in solution. The control of molecular architecture and choice of a suitable phospholipid matrix allows catalase-containing LB films to be used in sensing hydrogen peroxide.
