Phosphatidylcholine Surface Hydration-Dependent Adsorption to Mucin Enhances Intestinal Mucus Barrier Function.

The crucial role of zwitterionic phosphatidylcholines (PC) within mucus gel is essential for maintaining intestinal homeostasis, while the underlying mechanism remains incompletely understood. Herein, we compared the dynamic interfacial adsorption behavior of saturated dipalmitoylphosphatidylcholine (DPPC) and unsaturated dioleoylphosphatidylcholine (DOPC) to intestinal mucin and their impact on the intestinal mucus barrier function. Results of quartz crystal microbalance with dissipation showed that the highly surface-hydrated DPPC vesicles exhibited significantly faster and more extensive adsorption to purified intestinal mucin than the slightly surface-hydrated DOPC vesicles. Utilizing an intestinal Caco-2/HT29-MTX coculture model, we observed that DPPC vesicles adsorbed much more to the mucus gel compared to DOPC vesicles. Additionally, DPPC vesicle adsorption displayed increased wetting, and converse for DOPC vesicles. Interestingly, both of them exhibited nearly the same protective effects against cell injury induced by peptic-tryptic digests of gliadin (PTG). The partial mechanism involved the binding of PTG to DPPC and DOPC within the mucus gel, thereby restricting PTG contact with the underlying epithelial cells. These findings shed light on the intricate interfacial dynamics of PC adsorption to mucin and their implications for maintaining the integrity of the intestinal mucus barrier.

Peptide Permeation across a Phosphocholine Membrane: An Atomically Detailed Mechanism Determined through Simulations and Supported by Experimentation.

Cell-penetrating peptides (CPPs) facilitate translocation across biological membranes and are of significant biological and medical interest. Several CPPs can permeate into specific cells and organelles. We examine the incorporation and translocation of a novel anticancer CPP in a dioleoylphosphatidylcholine (DOPC) lipid bilayer membrane. The peptide, NAF-144-67, is a short fragment of a transmembrane protein, consisting of hydrophobic N-terminal and charged C-terminal segments. Experiments using fluorescently labeled NAF-144-67 in ∼100 nm DOPC vesicles and atomically detailed simulations conducted with Milestoning support a model in which a significant barrier for peptide-membrane entry is found at the interface between the aqueous solution and membrane. The initial step is the insertion of the N-terminal segment and the hydrophobic helix into the membrane, passing the hydrophilic head groups. Both experiments and simulations suggest that the free energy difference in the first step of the permeation mechanism in which the hydrophobic helix crosses the phospholipid head groups is -0.4 kcal mol-1 slightly favoring motion into the membrane. Milestoning calculations of the mean first passage time and the committor function underscore the existence of an early polar barrier followed by a diffusive barrierless motion in the lipid tail region. Permeation events are coupled to membrane fluctuations that are examined in detail. Our study opens the way to investigate in atomistic resolution the molecular mechanism, kinetics, and thermodynamics of CPP permeation to diverse membranes.

Supported Planar Single and Multiple Bilayer Formation by DOPC Vesicle Rupture on Mica Substrate: A Mechanism as Revealed by Atomic Force Microscopy Study

A planar lipid bilayer on a solid support serves as model system that explains fundamental aspects of membrane biology and enables us to characterize wide-range surface-sensitive techniques, including molecular engineering. The present study aims at understanding the process of single and multiple bilayer formation after the exposure of small unilamellar vesicles (SUVs) of dioleoyl phosphatidylcholine (DOPC) to mica substrate. Isolated single bilayer formation and co-existence of double and triple lipid bilayers in the aqueous medium have been quantitatively measured by atomic force microscopy and discussed the physicochemical mechanism. It has been observed that due to the strong adhesion of DOPC SUV to mica surface, vesicles of diluted solution rupture spontaneously and form isolated bilayer patches when they come in contact with the mica surface. No further lateral growth or movement of the bilayer patches has been observed upon increase of incubation time. However, the increase of vesicle number on the same surface area by successive deposition of DOPC solution of same concentration and increasing incubation time shows merging of the nearby patches as well as development of stacked second and third bilayers due to edge-guided rupture of adsorbed vesicles on first or second bilayer patches. Mechanisms of single and multi-bilayer formation and a theoretical interpretation of the process have been elucidated.

Cardiolipin Preferentially Partitions to the Inner Leaflet of Mixed Lipid Large Unilamellar Vesicles.

Cardiolipin (CL), an anionic phospholipid constituting 20% of the inner mitochondrial membrane (IMM) of eukaryotes, stabilizes electron transport chain (ETC) complexes and is a signaling agent in the early stages of apoptosis. For apoptosis, CL moves from the inner to the outer leaflet of the IMM via a poorly understood mechanism. Relative to cylindrically shaped lipids like dioleoylphosphatidylcholine (DOPC) and dioleoylphosphatidylglycerol (DOPG), cone-shaped CL should prefer the concave surfaces of lipid bilayers. Using the fluorophore, 1,1,2,2-tetrakis[4-(2-trimethylammonioethoxy)phenyl]ethene, we have measured CL versus DOPG partitioning to the inner versus the outer leaflet of liposomes in mixed lipid systems with DOPC. DOPG shows no leaflet preference. However, CL has a 4:1 preference for the concave surface of the inner leaflet of liposomes. To further test the inner leaflet preference of CL, we show that cytochrome c binding to the outer convex surface of 20% CL/80% DOPC vesicles is strongly attenuated. Because the outer leaflet of intracristal regions of the IMM has a concave curvature, the preference of CL for concave surfaces may facilitate the transport of CL from the inner to the outer leaflet of the IMM needed for CL remodeling, the optimal functioning of the ETC, and signaling in the early stages of apoptosis.

Accumulation of phosphatidylcholine on gut mucosal surface is not dominated by electrostatic interactions

The accumulation of phosphatidylcholine (PC) in the intestinal mucus layer is crucial for the protection of colon epithelia from the bacterial attack. It has been reported that the depletion of PC is a distinct feature of ulcerative colitis. Here we addressed the question how PC interacts with its binding proteins, the mucins, which may establish the hydrophobic barrier against colonic microbiota. In the first step, the interactions of dioleoylphosphatidylcholine (DOPC) with two mucin preparations from porcine stomach, have been studied using dynamic light scattering, zeta potential measurement, and Langmuir isotherms, suggesting that mucin binds to the surface of DOPC vesicles. The enthalpy of mucin-PC interaction could be determined by isothermal titration calorimetry. The high affinity to PC found for both mucin types seems reasonable, as they mainly consist of mucin 2, a major constituent of the flowing mucus. Moreover, by the systematic variation of net charges, we concluded that the zwitterionic DOPC has the strongest binding affinity that cannot be explained within the electrostatic interactions between charged molecules.

Substituents modulate biphenyl penetration into lipid membranes

Electrochemical impedance techniques and fluorescence spectroscopic methods have been applied to the study of the interaction of ortho (o)-, meta (m)- and para (p)-Cl-, o-, m- and p-HO-, p-H3CO-, p-H3C-, p-NC- and p-O3−S- substituted biphenyls (BPs) with Hg supported dioleoyl phosphatidylcholine (DOPC) monolayers and DOPC vesicles. Non-planar o-substituted BPs exhibit the weakest interactions whereas planar p-substituted BPs interact to the greatest extent with the DOPC layers. The substituted BP/DOPC monolayer and bilayer interaction depends on the effect of the substituent on the aromatic electron density, which is related to the substituents' mesomeric Hammetts constants. Substituted BPs with increased ring electron density do not increase the DOPC monolayer thickness on Hg and penetrate the DOPC vesicle membranes to the greatest extent. Substituted BPs with lower ring electron density can cause an increase in the monolayer's thickness on Hg depending on their location and they remain in the interfacial and superficial layer of the free standing DOPC membranes. Quantum mechanical calculations correlate the binding energy between the substituted BP rings and methyl acetate, as a model for the –CH2-(CO)O-CH2- fragment of a DOPC molecule, with the location of BPs within the DOPC monolayer.

Effects of Wild Type and Mutant HA Fusion Peptides on Kinetics and Activation Thermodynamics of Stalk and Pore Formation: Mechanistic Implications

Viral fusion peptides are 20-25 residue N-terminal regions of type 1 viral fusion proteins that are known to be critical to viral infection and to promote fusion between model membrane vesicles brought into close contact by poly(ethylene glycol) (PRG). However, almost nothing is known about how they function. We report the effects of wild-type (WT) hemagglutinin fusion peptide and its G1S, G1V and W14A mutants on the kinetics of PEG-mediated fusion of 23 nm vesicles of lipid composition dioleoylphosphatidylcholine, dioleoylphosphatidylethanolamine, sphingomyelin and cholesterol in a molar ratio of 35:30:15:20. Time courses of lipid mixing, content mixing, and content leakage were obtained using fluorescence assays at multiple temperatures, and analyzed according to a 2-step or 3-step (Biophys. J., 2007, 92; 4012) sequential model, in order to obtain activation thermodynamics for each step of the fusion process (Biophys J., 2012, 103: 2751). We also used fluorescence probes to monitor the influence of peptides on bilayer interfacial and acyl chain order, bilayer free volume and water penetration. These data were all considered in terms of recently published mechanistic models for the transition states for each of the steps of fusion (Biophys J., 2012, 103: 2751). WT peptide is proposed to catalyze step 1 due to its ability to occupy membrane free volume and stabilize TS1; it eliminates step 2 by destabilizing the second intermediate; and catalyzes step 3 by favoring correlated fluctuations of lipids from cis and trans leaflets of an unstable second intermediate. Polydopamine-Supported Lipid Bilayers

We report the formation of lipid membranes supported by a soft polymeric cushion of polydopamine. First, 20 nm thick polydopamine films were formed on mica substrates. Atomic force microscopy imaging indicated that these films were also soft with a surface roughness of 2 nm under hydrated conditions. A zwitterionic phospholipid bilayer was then deposited on the polydopamine cushion by fusion of dimyristoylphosphatidylcholine (DMPC) and dioleoylphosphatidylcholine (DOPC) vesicles. Polydopamine films preserved the lateral mobility of the phospholipids as shown by fluorescence microscopy recovery after photobleaching (FRAP) experiments. Diffusion coefficients of ~5.9 and 7.2 µm2 s−1 were respectively determined for DMPC and DOPC at room temperature, values which are characteristic of lipids in a free standing bilayer system.

Chloroform-Enhanced Incorporation of Hydrophobic Gold Nanocrystals into Dioleoylphosphatidylcholine (DOPC) Vesicle Membranes

Vesicles of dioleoylphosphatidylcholine (DOPC) formed by extrusion (liposomes) with hydrophobic alkanethiol-capped Au nanocrystals were studied. Dodecanethiol-capped 1.8-nm-diameter Au nanocrystals accumulate in the lipid bilayer, but only when dried lipid-nanocrystal films were annealed with chloroform prior to hydration. Without chloroform annealing, the Au nanocrystals phase separate from DOPC and do not load into the liposomes. Au nanocrystals with slightly longer capping ligands of hexadecanethiol or with a larger diameter of 4.1 nm disrupted vesicle formation and created lipid assemblies with many internal lamellar attachments.

Reduced Graphene Oxide as the Support for Lipid Bilayer Membrane

Supported lipid membranes of dioleoylphosphatidylcholine (DOPC) were formed on reduced graphene oxide (r-GO) by the vesicle fusion method. Graphene oxide prepared by chemical exfoliation was deposited on a thermally oxidized SiO2/Si substrate, and was reduced by hydrazine vapour. Flat lipid membranes were formed on the r-GO surface after the r-GO/SiO2/Si substrate was incubated in the DOPC vesicle suspension. We propose a structural model in which a lipid bilayer and monolayer stacked on r-GO based on the results of fluorescence microscope and atomic force microscope observation.

Red‐Emitting Dyes with Photophysical and Photochemical Properties Controlled by pH

New unsymmetrical zinc azaphthalocyanines, bearing one substituted aniline as a peripheral substituent, were prepared by using a statistical condensation approach. Both fluorescence and singlet oxygen quantum yields were extremely low in DMF (Φ(F)<0.01, Φ(Δ)<0.02, respectively), but increased after the addition of sulfuric acid, reaching values comparable to controls without aniline substituents (Φ(F)=0.22-0.29, Φ(Δ)=0.40-0.59, respectively). This behavior was attributed to the deactivation of excited states by intramolecular charge transfer from a donor site (aniline), which was blocked after protonation in acidic media. In the protonated form, all of the compounds efficiently emitted light with λ(em) in the region of 662-675 nm. The investigated compounds were anchored to dioleoylphosphatidylcholine (DOPC) unilamellar vesicles and showed response to buffer pH. They were highly fluorescent at low pH values and almost nonfluorescent in neutral solutions. The pK(a) values were determined in DOPC vesicles and ranged between 2.2 and 4.2.

Characterization of a Listeria monocytogenes Ca2+ Pump: A SERCA-TYPE ATPase WITH ONLY ONE Ca2+-BINDING SITE

We have characterized a putative Ca(2+)-ATPase from the pathogenic bacterium Listeria monocytogenes with the locus tag lmo0841. The purified and detergent-solubilized protein, which we have named Listeria monocytogenes Ca(2+)-ATPase 1 (LMCA1), performs a Ca(2+)-dependent ATP hydrolysis and actively transports Ca(2+) after reconstitution in dioleoylphosphatidyl-choline vesicles. Despite a high sequence similarity to the sarcoplasmic reticulum Ca(2+)-ATPase (SERCA1a) and plasma membrane Ca(2+)-ATPase (PMCA), LMCA1 exhibits important biochemical differences such as a low Ca(2+) affinity (K(0.5) ∼80 μm) and a high pH optimum (pH ∼9). Mutational studies indicate that the unusually high pH optimum can be partially ascribed to the presence of an arginine residue (Arg-795), corresponding in sequence alignments to the Glu-908 position at Ca(2+) binding site I of rabbit SERCA1a, but probably with an exposed position in LMCA1. The arginine is characteristic of a large group of putative bacterial Ca(2+)-ATPases. Moreover, we demonstrate that H(+) is countertransported with a transport stoichiometry of 1 Ca(2+) out and 1 H(+) in per ATP hydrolyzed. The ATPase may serve an important function by removing Ca(2+) from the microorganism in environmental conditions when e.g. stressed by high Ca(2+) and alkaline pH.

Spontaneous insertion of lipopolysaccharide into lipid membranes from aqueous solution

Lipopolysaccharide (LPS), one of the main components of outer membranes of Gram-negative bacteria, consists of a hydrophobic lipid (lipid A) with six hydrocarbon chains and a large hydrophilic polysaccharide chain. LPS plays endotoxic roles and can stimulate macrophages and B cells. To elucidate the mechanism of the interaction of LPS with various cell membranes, it is important to investigate the interaction of wild type LPS in a buffer with lipid membranes. In this report we investigated the interaction of low concentrations of LPS in a buffer with giant unilamellar vesicles (GUVs) of dioleoylphosphatidylcholine (DOPC) membrane in the liquid-crystalline (Lα) phase and sphingomyelin (SM)/cholesterol(chol) (molar ration; 6/4) membrane in the liquid-ordered (lo) phase. We found that low concentrations (less than critical micelle concentration) of LPS in aqueous solution induced the shape changes such as the transformation from a prolate to a two-spheres-connected by a very narrow neck in the DOPC-GUVs and also in the SM/chol (6/4)-GUVs above their threshold concentrations. The analysis of the shape changes of the GUVs indicates that the monomers of LPS can insert spontaneously into the external monolayer of the lipid membranes of these GUVs from the aqueous solution. Moreover, higher concentrations of LPS induced the vesicle fission of SM/chol(6/4)-GUVs above its higher threshold concentration. The vesicle fission of GUVs is similar to those induced by single long chain amphiphiles such as lysophosphatidylcholine. On the basis of these results, we discuss the interaction of wild type LPS with lipid membranes and cell membranes. These results suggest that LPS molecules can insert spontaneously into the external monolayer of the plasma membranes composed of the Lα phase-membrane and the microdomain in the lo phase.

Interaction of n-Octyl β,d-Glucopyranoside with Giant Magnetic-Fluid-Loaded Phosphatidylcholine Vesicles: Direct Visualization of Membrane Curvature Fluctuations As a Function of Surfactant Partitioning between Water and Lipid Bilayer

The present study deals with the morphological modifications of giant dioleoyl phosphatidylcholine vesicles (DOPC GUVs) induced by the nonionic surfactant n-octyl β,D-glucopyranoside at sublytic levels, i.e., in the first steps of the vesicle-to-micelle transition process, when surfactant inserts into the vesicle bilayer without disruption. Experimental conditions were perfected to exactly control the surfactant bilayer composition of the vesicles, in line with former work focused on the mechanical properties of the membrane of magnetic-fluid-loaded DOPC GUVs submitted to a magnetic field. The purpose here was to systematically examine, in the absence of any external mechanical constraint, the dynamics of giant vesicle shape and membrane deformations as a function of surfactant partitioning between the aqueous phase and the lipid membrane, beforehand established by turbidity measurements from small unilamellar vesicles.

Self-Reproduction of Fatty Acid Vesicles: A Combined Experimental and Simulation Study

Dilution of a fatty acid micellar solution at basic pH toward neutrality results in spontaneous formation of vesicles with a broad size distribution. However, when vesicles of a defined size are present before dilution, the size distribution of the newly formed vesicles is strongly biased toward that of the seed vesicles. This so-called matrix effect is believed to be a key feature of early life. Here we reproduced this effect for oleate micelles and seed vesicles of either oleate or dioleoylphosphatidylcholine. Fluorescence measurements showed that the vesicle contents do not leak out during the replication process. We hypothesized that the matrix effect results from vesicle fission induced by an imbalance of material across both leaflets of the vesicle upon initial insertion of fatty acids into the outer leaflet of the seed vesicle. This was supported by experiments that showed a significant increase in vesicle size when the equilibration of oleate over both leaflets was enhanced by either slowing down the rate of fatty acid addition or increasing the rate of fatty acid transbilayer movement. Coarse-grained molecular-dynamics simulations showed excellent agreement with the experimental results and provided further mechanistic details of the replication process.

Adsorption of DOPC vesicles on hydrophobic substrates in the presence of electrolytes: A QCM and reflectometry study

The adsorption of lipid Dioleoylphosphatidylcholine (DOPC) vesicles on a hydrophobic substrate has been investigated in aqueous buffer solution by means of the quartz crystal microbalance (QCM) and reflectometry. DOPC vesicles were prepared by the injection method on a hydrophobic substrate using 1-octadecanethiol (ODT) self-assembled on a gold-coated AT-cut quartz. The reflection spectrometry measurements of the adsorbed vesicles showed nearly monolayer formation in few cases, while in most other experiments, the frequency changes measured suggested multilayer formation assuming the usual Sauerbrey equation to hold in the present system as well. Presence of NaCl, KCl, Na2SO4 and ethanol in the aqueous phase during the formation of vesicles suggest that the multilayer formation can be hastened in some cases. Atomic force microscopic study corroborate the thicknesses that range between 8 and 20 nm for high concentration of electrolytes or ethanol suggesting coalescence of vesicles leading to several bilayers possibly stacked over each other.

Interaction between Lamellar (Vesicles) and Nonlamellar Lipid Liquid-Crystalline Nanoparticles as Studied by Time-Resolved Small-Angle X-ray Diffraction

The kinetics of structure change when dispersions of two different types of lipid-based liquid-crystalline phases, one lamellar and one reversed, are mixed has been investigated using synchrotron small-angle X-ray diffraction and ellipsometry. The systems studied were (i) cubic-phase nanoparticles (CPNPs) based on glycerol monooleate (GMO) stabilized with a nonionic block copolymer, Pluronic F-127; (ii) CPNPs based on phytantriol (PtOH) stabilized with D-alpha-Tocopheryl polyethylene glycol 1000 succinate (Vitamin E TPGS); and (iii) hexagonal-phase nanoparticles (HPNPs) based on a lipid mixture of diglycerol monooleate/glycerol dioleate, stabilized by Pluronic F-127. Time-resolved small-angle X-ray diffraction was used to track structural changes within nonlamellar nanoparticles when they interact with uni- and multilamellar vesicles of dioleoylphosphatidylcholine and dipalmitoylphatidylcholine. The results are very dependent on the type of nanoparticles under investigation. For GMO-based CPNPs, a strong interaction is observed on mixing with vesicular dispersions that leads to large changes in unit size dimensions as well as a later transition from cubic to lamellar structure. These results are in good agreement with previous studies on the interaction of GMO-based CPNPs with planar bilayers using neutron reflectivity, where the diffraction peak shifted with time upon mixing. The structural changes are much less prominent for the PtOH-based CPNPs and the HPNPs upon mixing with phospholipid vesicles. These results are correlated with those from measurement studying interactions between the liquid-crystalline nanoparticles and supported phospholipid bilayers by ellipsometry. Also, here the GMO-based CPNPs show more pronounced and rapid adsorption and interaction with the supported bilayer surface than do the other types of nonlamellar nanoparticles. The interaction also depends on the bilayer properties, where significantly slower lipid mixing is observed for a bilayer in the gel state compared to a bilayer in the liquid-crystalline phase. This study is not only relevant for drug-delivery applications but also shows the potential of synchrotron small-angle X-ray diffraction in studying time-dependent structural changes as a consequence of the interaction between different lipid self-assembled aggregates in complex systems.

An Electrochemical Study into the Interaction between Complement-Derived Peptides and DOPC Mono- and Bilayers

Electrochemical methods employing the hanging mercury drop electrode were used to study the interaction between variants of the complement-derived antimicrobial peptide CNY21 (CNYITELRRQH ARASHLGLAR) and dioleoyl phosphatidylcholine (DOPC) monolayers. Capacitance potential and impedance measurements showed that the CNY21 analogues investigated interact with DOPC monolayers coating the mercury drop. Increasing the peptide hydrophobicity by substituting the two histidine residues with leucine resulted in a deeper peptide penetration into the hydrophobic region of the DOPC monolayer, indicated by an increase in the dielectric constant of the lipid monolayer (Deltaepsilon = 2.0 after 15 min interaction). Increasing the peptide net charge from +3 to +5 by replacing the histidines by lysines, on the other hand, arrests the peptide in the lipid head group region. Reduction of electroactive ions (Tl+, Pb2+, Cd2+, and Eu3+) at the monolayer-coated electrode was employed to further characterize the types of defects induced by the peptides. All peptides studied permeabilize the monolayer to Tl+ to an appreciable extent, but this effect is more pronounced for the more hydrophobic peptide (CNY21L), which also allows penetration of larger ions and ions of higher valency. The results for the various ions indicate that charge repulsion rather than ion size is the determining factor for cation penetration through peptide-induced defects in the DOPC monolayer. The effects obtained for monolayers were compared to results obtained with bilayers from liposome leakage and circular dichroism studies for unilamellar DOPC vesicles, and in situ ellipsometry for supported DOPC bilayers. Trends in peptide-induced liposome leakage were similar to peptide effects on electrochemical impedance and permeability of electroactive ions for the monolayer system, demonstrating that formation of transmembrane pores alone does not constitute the mechanism of action for the peptides investigated. Instead, our results point to the importance of local packing defects in the lipid membrane in close proximity to the adsorbed peptide molecules.

Synthesis and bandgap variation of molecular-size CdSe clusters via electroporation ofvesicles

Molecular-size uncapped CdSe clusters are synthesized via the electroporationof synthetic dioleoylphosphatidylcholine vesicles of diameter 178 nm.Cd2+ ions, originally entrapped in the vesicles, are ejected through the transientpores of the vesicle membrane into the bulk where they can react withSeSO32− ions in basic condition to form CdSe clusters at room temperature. Growth andself-aggregation of CdSe clusters is slowed down to timescales of days by their adsorption atthe exterior surface of the vesicle, which permits spectral monitoring of the continuousgrowth process over a long period of time. Growth in the molecular size regimeis found to entail novel blue-shifts of the characteristic absorption band. Theblue-shift of the transition energy is in agreement with an analogous oscillation ofthe HOMO–LUMO gap calculated by the use of density functional theory for(CdSe)n clusters. The reason for the blue-shift absorption spectra of molecular-size CdSe clusters isinterpreted as the higher stability and bigger gaps of magic number. On the basis of thesimilarity between the experimental and theoretical trends, the characteristic absorptionpeaks of 256 nm, 215 nm can be assigned to the monomer or dimer, trimer clusters ofCdSe, respectively. Any further growth beyond trimers is associated with thecustomary monotonic red-shift of the absorption band, due to quantum size effects.

Entropic Stabilization and Equilibrium Size of Lipid Vesicles

We have studied the phase behavior of zwitterionic phospholipid dioleoylphosphatidylcholine (DOPC) vesicles (membranes) and interpreted our results using scaling arguments in combination with molecular realistic self-consistent field (SCF) calculations. DOPC membranes acquire a partial negative charge per lipid molecule at intermediate NaBr concentrations. As a result of this, dilute DOPC solutions form stable unilamellar vesicles. Both at low and high salt concentrations phase separation into a lamellar and a vesicular phase is observed. The vesicle radius decreases as a power law with decreasing lipid concentration. This power-law concentration dependence indicates that the vesicle phase is entropically stabilized; the size of the DOPC vesicles result from a competition between the bending energy and translation and undulation entropy. This scaling behavior breaks down for very small vesicles. This appears to be consistent with SCF predictions that point to the fact that in this regime the mean bending modulus kc increases with curvature. The SCF theory predicts that, at low ionic strength, the membrane stability improves when there is more charge on the lipids. Upon a decrease of the ionic strength, lipids with a full negative charge form vesicles that grow exponentially in size because the mean bending modulus increases with decreasing ionic strength. At the same time the Gaussian bending modulus becomes increasingly negative such that the overall bending energy tends to zero. This indicates that small micelles become the dominant species. The SCF theory thus predicts a catastrophic break down of giant vesicles in favor of small micelles at sufficiently low ionic strength and high charge density on the lipids.
