The Effects of Long-Term Precipitation Exclusion on Leaf Photosynthetic Traits, Stomatal Conductance, and Water Use Efficiency in Phyllostachys edulis

Ongoing climate change is projected to intensify drought stress globally. Understanding the response mechanisms of Phyllostachys edulis (Carrière) J. Houz. (moso bamboo) to long-term drought is crucial, given its significance as a carbon sequestration resource. In this study, precipitation exclusion was implemented to simulate drought stress and we investigated the effects of long-term drought on the photosynthetic parameters, stomatal conductance, and water use efficiency of moso bamboo. The results showed that throughout all growth seasons, the maximum net photosynthetic rates (Pmax) of bamboo at all ages under long-term drought conditions (after 8 years of precipitation exclusion treatment) were significantly lower than those of the control (p < 0.05). It can be concluded that long-term drought reduced the maximum photosynthetic capacity of the bamboo at all ages. Under long-term drought conditions, there were many seasons where the light saturation point (LSP) of first-degree (1–2 years old) bamboo and third-degree (5–6 years old) bamboo under drought was significantly lower than those of the control, while the LSP value of second-degree (3–4 years old) bamboo under drought was significantly higher than that of the control. This suggests that long-term drought reduced the ability of first-degree and third-degree bamboo to utilize strong light, while improving the ability of second-degree bamboo to utilize strong light in summer, autumn, and winter. Under long-term drought conditions, the light compensation point (LCP) and the apparent quantum efficiency (AQY) of the bamboo decreased. It can be concluded that long-term drought reduced the ability of first-degree bamboo to utilize weak light in all seasons, as well as the ability of second-degree bamboo to utilize weak light in spring and autumn; meanwhile, it improved the ability of second-degree bamboo to utilize weak light in summer and winter, and the ability of third-degree bamboo to utilize weak light in spring, summer, and autumn. In the high light range (PARi > 1000 µmol · m−2 · s−1), there were significant differences in stomatal conductance (gs) among different the different treatments of bamboo, which were influenced by both the growing season and the forest age. Compared to the control, under drought conditions, the stomatal conductance of third-degree bamboo increased in spring and that of the second-degree bamboo increased in autumn. The correlation analysis showed that the relationship between the stomatal conductance and vapor pressure deficit (VPDL) of bamboo under long-term drought conditions showed a significant polynomial relationship in both high and low light ranges. The correlation between the instantaneous water use efficiency (iWUE) and VPDL for the drought and control treatments of bamboo also showed a significant polynomial relationship in high light ranges. It was found that long-term drought changed the photosynthetic parameters of the bamboo, reflecting its ability to tolerate and adapt to drought in different seasons. Age-related differences in photosynthetic parameters should be fully considered in forest age structure adjustments and forest thinning procedures to strengthen the light intensity and maintain the opening of the stoma. These results provide a theoretical basis for the efficient and sustainable cultivation of bamboo under global climate change.

Early testing for improving growth under water shortage in Eucalyptus globulus Labill.

Aim of study: We aimed at identifying differences in the response to water shortage between Eucalyptus globulus clones. Area of study: Field trials were established in SW Spain. Material and methods: Potted plants from six clones were grown in a greenhouse for 35 days under two watering regimes. Two clones were F0 genotypes and the other four were F1 (hybrid) genotypes, including one inbred clone. Differences in stomatal conductance, hydraulic traits, growth and specific leaf area (SLA) were analyzed. Main results: Water shortage decreased SLA, growth in height and leaf area and leaf-specific hydraulic conductivity (KLmax). We measured the highest growth in F1 genotypes and the lowest in the clone in which SLA was lowest. The inbred clone showed the highest growth reduction under water shortage. There was substantial hysteresis between leaf water potential (Y) and native embolism, most probably a result of combined cavitation and refilling. High losses of hydraulic conductance were compatible with high stomatal conductances. Maximum values of stomatal conductance decreased with the soil water content estimated from predawn Y and were lowest in the inbred clone, showing less plasticity and a diminished ability to cope with high temperatures, which could explain its poor development under field conditions. Research highlights: Soil water content and predawn Y appeared as critical factors controlling stomata closure, while stomatal conductance and SLA could be useful to predict differences in growth and survival from early trials.

Endophyte mediated Populus trichocarpa water use efficiency is dependent on time of day and plant water status

Endophytes are potential partners for improving the resource use efficiency of bioenergy feedstock systems such as short rotation coppiced Populus. Endophytes isolated from members of the Salicaceae family have broad host compatibility and can improve water use efficiency (WUE) through decreases in stomatal conductance. However, the literature is inconsistent with regards to the environmental conditions and temporal patterns of these benefits. This study investigates how endophyte mediated changes in Populus trichocarpa (Torr. and Gray) ‘Nisqually-1’ stomatal conductance and WUE shift with time and scale in response to water-deficit stress. Leaf gas exchange and above-ground productivity were used to evaluate the carbon and water balances of glasshouse grown plants in response to endophyte inoculation and water-deficit. Differences in stomatal conductance between control and inoculated plants were more pronounced (39.7 % decrease, Welch’s two-sample t(14.34 dfadj) = -2.358, p-value = 0.033) under water-deficit conditions in the late morning during a period of higher light intensity. The decrease in stomatal conductance accompanied a substantial increase in intrinsic WUE (iWUE) for water-deficit inoculated plants. However, increases in iWUE did not result in improvements in aboveground productivity or shoot biomass WUE (WUEsb) for water-deficit inoculated plants. This decoupling between iWUE and aboveground productivity may be an indicator of assimilate allocation to microbial metabolism as an additional carbon sink or a shift in carbon allocation towards belowground biomass. Future work should take a whole-plant approach that accounts for diurnal patterns in incident irradiance to evaluate the impact of endophyte inoculation on host WUE and stress tolerance.

Leaf and tree water-use efficiencies of Populus deltoides × P. nigra in mixed forest and agroforestry plantations.

In a global context where water will become a scarce resource under temperate latitudes, managing tree plantations with species associations, i.e., forest mixture or agroforestry, could play a major role in optimizing the sustainable use of this resource. Conceptual frameworks in community ecology suggest that, in mixed plantations, environmental resources such as water may be more efficiently used for carbon acquisition and tree growth thanks to niche complementarity among species. To test the hypotheses behind these conceptual frameworks, we estimated water-use efficiency (WUE) for poplar trees grown in a monoculture, in association with alder trees (forest mixture) and in association with clover leys (agroforestry) in an experimental plantation located in northeastern France. Water-use efficiency was estimated (i) at leaf level through gas exchange measurements and analysis of carbon isotope composition, (ii) at wood level through carbon isotope composition and (iii) at tree level with sap flow sensors and growth increment data. We hypothesized that species interactions would increase WUE of poplars in mixtures due to a reduction in competition and/or facilitation effects due to the presence of the N2-fixing species in mixtures. Poplar trees in both mixture types showed higher WUE than those in the monoculture. The differences we found in WUE between the monoculture and the agroforestry treatment were associated to differences in stomatal conductance and light-saturated net CO2 assimilation rate (at the leaf level) and transpiration (at the tree level), while the differences between the monoculture and the forest mixture were more likely due to differences in stomatal conductance at the leaf level and both transpiration and biomass accumulation at the tree level. Moreover, the more WUE was integrated in time (instantaneous gas exchanges < leaf life span < seasonal wood core < whole tree), the more the differences among treatments were marked.

Competition between Rice and Weedy Rice under Different Water Regimes

The rainfed lowland (RFL) rice ecosystem is prone to drought stress and high weed infestation, including weedy rice (WR), which often negatively affects the yield. This study examined the competitive ability of rice and WR under mixed culture and drought stress. Two rice genotypes (drought-susceptible IR64 and drought-tolerant INL73) and a WR were grown in root boxes with soil under mono (rice or WR only) or mixed (rice/WR) cultures and subjected to either continuously waterlogged (CWL) or progressive drought (PDR) conditions during the early vegetative stage. Under PDR, SDW in all planting cultures was reduced relative to their CWL counterparts. The WR in mixed culture had higher SDW than its monoculture counterpart, regardless of water treatments. SDW in mixed cultures under PDR was not affected in IR64, reduced in INL73, and increased in WR relative to their monocultures. Under PDR, water use (WU) was higher in INL73/ WR than in IR64/WR mixed culture. Between two mixed cultures, WR mixed with INL73 had higher SDW than when mixed with IR64. The differences in stomatal conductance between INL73 and WR under mixed culture were attributed to the greater increase in total lateral root length of the latter than the former genotype under PDR. Overall, the results showed that WR had greater competitive ability than rice under mixed cultures, especially with a drought-tolerant rice variety. This calls for careful planning of cultivar adoption in rainfed systems where WR is a problem.

Incorporating cultivar-specific stomatal traits into stomatal conductance models improves the estimation of evapotranspiration enhancing greenhouse climate management

The effect of considering cultivar differences in stomatal conductance (g s ) on relative air humidity (RH)-related energy demand was addressed. We conducted six experiments in order to study the variation in evapotranspiration (ET c ) of six pot rose cultivars, investigate the underlying processes and parameterise a g s -based ET c model. Several levels of crop ET c were realised by adjusting the growth environment. The commonly applied Ball–Woodrow–Berry g s -sub-model (BWB-model) in ET c models was validated under greenhouse conditions, and showed a close agreement between simulated and measured ET c . The validated model was incorporated into a greenhouse simulator. A scenario simulation study showed that selecting low-g s cultivars reduces energy demand (≤5.75%), depending on the RH set point. However, the BWB-model showed poor prediction quality at RH lower than 60% and a good fit at higher RH. Therefore, an attempt was made to improve model prediction: the in situ -obtained data were employed to adapt and extend either the BWB-model, or the Liu-extension with substrate water potential (Ψ; BWB-Liu-model). Both models were extended with stomatal density (D s ) or pore area. Although the modified BWB-Liu-model (considering D s ) allowed higher accuracy (R 2 = 0.59), as compared to the basic version (R 2 = 0.31), the typical lack of Ψ prediction in greenhouse models may be problematic for implementation into real-time climate control. The current study lays the basis for the development of cultivar specific cultivation strategies as well as improving the g s sub-model for dynamic climate conditions under low RH using model-based control systems. • Relative air humidity control is a key but costly aspect of greenhouse climate. • Accurate estimation of evapotranspiration (ET c ) is essential for relative air humidity regulation. • Six pot rose cultivars and several cultivation regimes were used. • ET c estimation was improved using cultivar-specific stomatal conductance parameters. • The use of low-stomatal conductance cultivars secures energy saving (2.5–5.75%).

Seedlings from marginal and core populations of European beech (Fagus sylvatica L.) respond differently to imposed drought and shade

Key messageLocal adaptation largely segregates between traits affecting light responses and water relations, but population-level differences in stomatal conductance when growth is unconstrained converge among populations under water stress.Warming temperatures and altered precipitation patterns threaten plant populations worldwide. European beech (Fagus sylvatica) is a species that expresses both high phenotypic plasticity and local adaptation among populations. Beech seedlings’ susceptibility to prolonged drought may be dictated by their immediate light environment. We tested whether seedlings of four beech provenances, from contrasting edaphoclimatic environments, expressed differences in trait responses to imposed water stress under sun and shade treatments. Populations from the southern range margin were expected to display greater water-stress tolerance and core populations’ faster growth rates in the absence of abiotic limitations. Both high light and water stress induced differences in trait responses among provenances, but traits that failed to respond to our experimental treatments likewise did not segregate at the provenance level. Hence, those traits responding to light, e.g., increasing leaf flavonol index and leaf mass area, also tended to differ among provenances. Similarly, there was evidence of local adaptation among provenances in traits, like midday leaf water potential, responding to water stress. Exceptionally, there was a three way interaction water- × -light- × -provenance for stomatal conductance which converged among provenances under water stress. Leaf chlorophyll content also varied both with light and water in a provenance-specific manner. We found core provenances’ growth traits to outperform others under favourable conditions, whereas southern and high-elevation populations displayed traits adapted to tolerate high irradiance. Only stomatal conductance produced a complementary interactive response between light- × -water across provenances, whereas other traits responded less to combined water stress and high irradiance than to either treatment alone.

Long-term transcriptional memory in rice plants submitted to salt shock.

A first salt shock event alters transcriptional and physiological responses to a second event, being possible to identify 26 genes associated with long-term memory. Soil salinity significantly affects rice cultivation, resulting in large losses in growth and productivity. Studies report that a disturbing event can prepare the plant for a subsequent event through memory acquisition, involving physiological and molecular processes. Therefore, genes that provide altered responses in subsequent events define a category known as "memory genes". In this work, the RNA-sequencing (RNA-Seq) technique was used to analyse the transcriptional profile of rice plants subjected to different salt shock events and to characterise genes associated with long-term memory. Plants subjected to recurrent salt shock showed differences in stomatal conductance, chlorophyll index, electrolyte leakage, and the number of differentially expressed genes (DEGs), and they had lower Na+/K+ ratios than plants that experienced only one stress event. Additionally, the mammalian target of rapamycin (mTOR) pathways, and carbohydrate and amino acid-associated pathways were altered under all conditions. Memory genes can be classified according to their responses during the first event (+ or -) and the second shock event (+ or -), being possible to observe a larger number of transcripts for groups [+ /-] and [-/ +], genes characterised as "revised response." This is the first long-term transcriptional memory study in rice plants under salt shock, providing new insights into the process of plant memory acquisition.

Element content and expression of genes of interest in guard cells are connected to spatiotemporal variations in stomatal conductance.

Element content and expression of genes of interest on single cell types, such as stomata, provide valuable insights into their specific physiology, improving our understanding of leaf gas exchange regulation. We investigated how far differences in stomatal conductance (gs ) can be ascribed to changes in guard cells functioning in amphistomateous leaves. gs was measured during the day on both leaf sides, on well-watered and drought-stressed trees (two Populus euramericana Moench and two Populus nigra L. genotypes). In parallel, guard cells were dissected for element content and gene expressions analyses. Both were strongly arranged according to genotype, and drought had the lowest impact overall. Normalizing the data by genotype highlighted a structure on the basis of leaf sides and time of day both for element content and gene expression. Guard cells magnesium, phosphorus, and chlorine were the most abundant on the abaxial side in the morning, where gs was at the highest. In contrast, genes encoding H+ -ATPase and aquaporins were usually more abundant in the afternoon, whereas genes encoding Ca2+ -vacuolar antiporters, K+ channels, and ABA-related genes were in general more abundant on the adaxial side. Our work highlights the unique physiology of each leaf side and their analogous rhythmicity through the day.

The quantitative impact of different leaf temperature determination on computed values of stomatal conductance and internal CO2 concentrations

The LI-COR 6400 portable photosynthesis system is the most widely used and cited instrument for measuring leaf gas exchange in plant physiology. The focus of this paper is to evaluate three ways of computing/measuring leaf temperature: the LI-COR energy budget computation, the LI-COR leaf thermocouple (T/C), and an infra-red (IR) sensor chip (MLX90615) incorporated into the LI-6400 leaf chamber. The IR-sensor was calibrated against known targets, and then used to cross check the other two methods. The results showed that the IR-sensor was superior to the LI-COR thermocouple and agreed closer with the energy budget values of leaf temperature. The magnitudes of the errors were quantified in terms of °C difference in leaf temperature measured by T/C (as control) and IR sensor versus percentage difference in stomatal conductance (gs) and intercellular CO2 concentration (Ci) computed from leaf temperature. The temperature difference (IR versus other methods) in the range of -1.89 to 1.58 °C resulted in relative errors in gs from 14.7% to -18.0% and Ci from 16.5% to -7.8% of the values reported by the standard LI-6400 calculations depending on species and temperature. Reasons are given for trusting the IR-sensor over the other methods and it is suggested that the next generation LI-COR photosynthesis system include an IR-sensor.

Diverse Stomatal Behaviors Mediating Photosynthetic Acclimation to Low Temperatures in Hordeum vulgare.

Photosynthetic acclimation to cold conditions is an important factor influencing freezing tolerance of plants. Photosynthetic enzyme activities increase as part of a photochemical mechanism underlying photosynthetic acclimation to low temperatures. Additionally, a non-photochemical mechanism may be activated to minimize photooxidative damage. The aim of this study was to test the hypothesis that differences in stomatal conductance in Hordeum vulgare plants with contrasting freezing tolerances induce various strategies for photosynthetic acclimation to cold stress. Different stomatal behaviors during the prehardening step resulted in diverse plant reactions to low-temperature stress. Plants with a relatively low freezing tolerance exhibited decreased stomatal conductance, resulting in decreased photochemical activity, faster induction of the non-photochemical mechanism, and downregulated expression of two Rubisco activase (RcaA) splicing variants. In contrast, plants with a relatively high freezing tolerance that underwent a prehardening step maintained the stomatal conductance at control level and exhibited delayed photochemical activity and RcaA expression decrease, and increased Rubisco activity, which increased net photosynthetic rate. Thus, in barley, the induction of photoinhibition avoidance (i.e., non-photochemical photoacclimation mechanism) is insufficient for an effective cold acclimation. An increase in cold-induced net photosynthetic rate due to open stomata is also necessary.

Genotypic variation in whole-plant transpiration efficiency in sorghum only partly aligns with variation in stomatal conductance.

Water scarcity can limit sorghum (Sorghum bicolor (L.) Moench) production in dryland agriculture, but increased whole-plant transpiration efficiency (TEwp, biomass production per unit of water transpired) can enhance grain yield in such conditions. The objectives of this study were to quantify variation in TEwp for 27 sorghum genotypes and explore the linkages of this variation to responses of the underpinning leaf-level processes to environmental conditions. Individual plants were grown in large lysimeters in two well-watered experiments. Whole-plant transpiration per unit of green leaf area (TGLA) was monitored continuously and stomatal conductance and maximum photosynthetic capacity were measured during sunny conditions on recently expanded leaves. Leaf chlorophyll measurements of the upper five leaves of the main shoot were conducted during early grain filling. TEwp was determined at harvest. The results showed that diurnal patterns in TGLA were determined by vapour pressure deficit (VPD) and by the response of whole-plant conductance to radiation and VPD. Significant genotypic variation in the response of TGLA to VPD occurred and was related to genotypic differences in stomatal conductance. However, variation in TGLA explained only part of the variation in TEwp, with some of the residual variation explained by leaf chlorophyll readings, which were a reflection of photosynthetic capacity. Genotypes with different genetic background often differed in TEwp, TGLA and leaf chlorophyll, indicating potential differences in photosynthetic capacity among these groups. Observed differences in TEwp and its component traits can affect adaptation to drought stress.

Salinity alleviates zinc toxicity in the saltmarsh zinc-accumulator Juncus acutus

The potential importance of Juncus acutus for remediation of Zn-contaminated lands has been recognized, because of its Zn tolerance and capacity to accumulate Zn. Since it is also a halophyte, the extent to which salinity influences its Zn tolerance requires investigation. A factorial greenhouse experiment was designed to assess the effect of NaCl supply (0 and 85 mM NaCl) on the growth, photosynthetic physiology and tissue ions concentrations of plants exposed to 0, 30 and 100 mM Zn. Our results indicated that NaCl supplementation alleviated the effects of Zn toxicity on growth, as Zn at 100 mM reduced relative growth rate (RGR) by 60% in the absence of NaCl but by only 34% in plants treated also with NaCl. This effect was linked to a reduction in Zn tissue concentrations, as well as to overall protective effects on various stages in the photosynthetic pathway. Thus, at 85 mM NaCl plants were able to maintain higher net photosynthesis (AN) than in the absence of added NaCl, although there were no differences in stomatal conductance (gs). This contributed to preserving the trade-off between CO2 acquisition and water loss, as indicated by higher intrinsic water use efficiency (iWUE). Hence, AN differences were ascribed to limitation in the RuBisCO carboxylation, manifested as higher intercellular CO2 concentration (Ci), together with dysfunction of PSII photochemistry (in term of light harvest and energy excess dissipation), as indicated by higher chronic photoinhibition percentages and variations in the photosynthetic pigment profiles in presence of Zn under non-saline conditions.

Effect of root infection by Macrophomina phaseolina on stomatal conductance, canopy temperature and yield of soybean

Charcoal rot, caused by Macrophomina phaseolina, is a widespread drought-associated, soilborne disease of soybean. Generally, disease severity is assessed indirectly as host colonization at the end of the growing season. Few studies have measured the effects of disease during the season. Two moderately charcoal rot resistant cultivars (‘DT 97-4290‘ and ‘Delta Pineland 4546‘), a drought tolerant cultivar (‘R01-1581F’) and a susceptible cultivar (‘LS 980358‘) were planted in microplots filled with uninoculated steam pasteurized soil or pasteurized soil infested with M. phaseolina. Half of the plots were irrigated and half were drought stressed. Stomatal conductance and infrared canopy temperatures were measured 47, 70, 77 and 98 days after planting (DAP) in 2008 and 52, 63, 72 and 84 DAP in 2009. Yields were taken at the end of the season. There were significant differences in stomatal conductance. In 2008, stomatal conductance was significantly lower 47 DAP in the non-irrigated/inoculated plots than the other treatments. At 98 DAP, stomatal conductance was highest in the irrigated/non-inoculated plots. In 2009, stomatal conductance was significantly higher in the non-infested plots than the infested plots of ‘LS 890358‘ and ‘R01-1581F’ across sample times, but not in plots of the resistant cultivars ‘DT 97-4290‘ or ‘DPL 4546‘. These differences were reflected in significantly greater canopy temperatures and lower yields in the infested compared with the non-infested plots of the susceptible, but not the resistant cultivars. These results show that root infection by M. phaseolina can reduce water movement in soybean and reduce yields in susceptible cultivars.

Comparisons of the Performance of δ13C and δ18O of Fagus sylvatica, Pinus sylvestris, and Quercus petraea in the Record of Past Climate Variations

AbstractClimate reconstructions in temperate Europe have been widely based on oak species. However, other co‐occurring species, largely distributed in Europe, may be used for recording climate variability. In this paper, we documented the intertrees and interspecies variations over 1960–2007 of oxygen and carbon isotopic compositions in ring cellulose of Fagus sylvatica, Pinus sylvestris, and Quercus petraea co‐occurring in the Fontainebleau forest (France). Our results indicated that large levels of series replication (11 trees on average) were required to generate isotopic mean series representative of the populations. We calculated mean isotopic ratios higher in pine than in the deciduous species and hypothesized that these contrasts resulted from differences in stomatal conductance, phenology, and canopy structure and, for oxygen, also in water uptake depth and isotopic exchange rate. We found that δ18O and δ13C chronologies were significantly correlated to one another in the three species and responded primarily to air moisture and Tmax, which indicated that stomatal conductance was an important driver of changes in both types of records. We determined that the correlations were strong with the May to July climate variables in F. sylvatica and with the July and August ones in Q. petraea and P. sylvestris. We showed that the oxygen records were systematically more coherent than those of carbon. This study demonstrated that δ18O, and to a lesser extent δ13C, from the three different species are reliable proxies for reconstructing past hydroclimatic variations in Europe.

A novel QTL associated with rice canopy temperature difference affects stomatal conductance and leaf photosynthesis.

Canopy temperature can be a good indicator of stomatal conductance. To understand the genetic basis of phenotypic differences in stomatal conductance between average and high-yielding rice (Oryza sativa L.) cultivars, we conducted a quantitative trait locus (QTL) analysis of canopy temperature. We developed reciprocal series of backcross inbred lines (BC1F6) derived from a cross between the average-yielding japonica cultivar ‘Koshihikari’ and the high-yielding indica cultivar ‘Takanari’. A stable QTL, qCTd11 (QTL for canopy temperature difference on chromosome 11) on the short arm of chromosome 11, accounted for 10.4 and 19.8% of the total phenotypic variance in the two lines; the ‘Takanari’ allele decreased the canopy temperature difference value. A chromosome segment substitution line carrying the Takanari qCTd11 showed a greater reduction in canopy temperature than ‘Koshihikari’, and had higher stomatal conductance and photosynthetic rate. These results suggest that qCTd11 is not only involved in canopy temperature, but is also involved in both stomatal conductance and photosynthetic rate.

Indirect selection for potential yield in early-generation, spaced plantings of wheat and other small-grain cereals: a review

Early-generation (e.g. F2–F4) selection for grain yield itself is frustrated in particular by the small amounts of seed available. However, there has long been an interest in traits related to yield and reasonably faithfully expressed in spaced planting arrangements using little seed; these are potentially useful as indirect selection criteria for yield, with the view to increasing genetic progress per unit cost. This subject is revisited in this review, targeting potential yield (yield in the absence of abiotic and biotic stresses) of small-grain cereals. A brief assessment of current breeding systems for self-pollinated crops such as wheat reveals that all have some stage during which selection among visually acceptable spaced plants has to, or could, be practiced. The relative performance of different genotypes in such spaced plantings is then explored, highlighting interactions arising from intergenotypic competition as well as from the extra space itself. The theory of indirect selection is presented, along with some practical examples. After a brief survey of possible selection traits and developments in high-throughput measurement, harvest index, fruiting efficiency and stomatal conductance (and its surrogates) are chosen for in-depth review. All three traits show promise, especially in the light of possible new ways of reducing the cost of their measurement in early generations. Remote sensing of foliage temperature for the detection of genotypic differences in stomatal conductance makes this clearly the most promising trait for thorough testing in commercial breeding populations. Such traits could be used directly or they could complement genomic selection in early generations.

Screening for Natural Variation in Water Use Efficiency Traits in a Diversity Set of Brassica napus L. Identifies Candidate Variants in Photosynthetic Assimilation.

Seed yield and quality of crop species are significantly reduced by water deficit. Stable isotope screening (δ13C) of a diversity set of 147 accessions of Brassica napus grown in the field identified several accessions with extremes in water use efficiency (WUE). We next conducted an investigation of the physiological characteristics of selected natural variants with high and low WUE to understand how these characteristics translate to differences in WUE. We identified an interesting Spring accession, G302 (Mozart), that exhibited the highest WUE in the field and high CO2 assimilation rates coupled with an increased electron transport capacity (Jmax) under the imposed conditions. Differences in stomatal density and stomatal index did not translate to differences in stomatal conductance in the investigated accessions. Stomatal conductance response to exogenous ABA was analyzed in selected high and low WUE accessions. Spring lines showed little variation in response to exogenous ABA, while one Semi-Winter line (SW047) showed a significantly more rapid response to exogenous ABA, that corresponded to the high WUE indicated by δ13C measurements. This research illustrates the importance of examining natural variation at a physiological level for investigation of the underlying mechanisms influencing the diversity of carbon isotope discrimination values in the field and identifies natural variants in B. napus with improved WUE and potential relevant traits.

Evidence for Active Uptake and Deposition of Si-based Defenses in Tall Fescue.

Silicon (Si) is taken up from the soil as monosilicic acid by plant roots, transported to leaves and deposited as phytoliths, amorphous silica (SiO2) bodies, which are a key component of anti-herbivore defense in grasses. Silicon transporters have been identified in many plant species, but the mechanisms underpinning Si transport remain poorly understood. Specifically, the extent to which Si uptake is a passive process, driven primarily by transpiration, or has both passive and active components remains disputed. Increases in foliar Si concentration following herbivory suggest plants may exercise some control over Si uptake and distribution. In order to investigate passive and active controls on Si accumulation, we examined both genetic and environmental influences on Si accumulation in the forage grass Festuca arundinacea. We studied three F. arundinacea varieties that differ in the levels of Si they accumulate. Varieties not only differed in Si concentration, but also in increases in Si accumulation in response to leaf damage. The varietal differences in Si concentration generally reflected differences in stomatal density and stomatal conductance, suggesting passive, transpiration-mediated mechanisms underpin these differences. Bagging plants after damage was employed to minimize differences in stomatal conductance between varieties and in response to damage. This treatment eliminated constitutive differences in leaf Si levels, but did not impair the damage-induced increases in Si uptake: damaged, bagged plants still had more leaf Si than undamaged, bagged plants in all three varieties. Preliminary differential gene expression analysis revealed that the active Si transporter Lsi2 was highly expressed in damaged unbagged plants compared with undamaged unbagged plants, suggesting damage-induced Si defenses are regulated at gene level. Our findings suggest that although differences in transpiration may be partially responsible for varietal differences in Si uptake, they cannot explain damage-induced increases in Si uptake and deposition, suggesting that wounding causes changes in Si uptake, distribution and deposition that likely involve active processes and changes in gene expression.

Evidence for adaptation and acclimation in a widespread eucalypt of semi-arid Australia

As climates change, restoration programmes need to maximize the capacity of regenerating ecosystems to adapt to new environments, particularly when planting long-lived trees. By using a combined phenotypic and genomic approach, we assessed evidence of adaptation capacity in Eucalyptus loxophleba ssp. lissophloia, a mallee eucalypt important for restoration and oil production in Western Australia. Assessment of leaf traits in nine wild populations across a rainfall gradient showed that two traits were correlated with long-term moisture availability. Populations in more arid environments had lower specific leaf area and lower stomatal conductance, consistent with a stable plastic or adaptive response. Other leaf size and shape traits were correlated with short-term climate variables, suggesting a dynamic plastic response. Genome-wide scans with 4851 DArTseq markers and outlier analysis detected 50 markers showing signals of disruptive selection, consistent with local adaptation. Sixteen markers showed allele frequencies correlated with aridity; three were also associated with differences in stomatal conductance. Multivariate analysis and marker-environment associations signalled secondary directions of adaptation associated with maximum temperatures and soil phosphorus. We suggest that adaptation and acclimation are both likely determinants of functional phenotype in E. loxophleba, and argue that aridity is a critical driver of adaptation in this widespread species.
