Exploring Refinement Strategies for Single Housing of Male C57BL/6JRj Mice: Effect of Cage Divider on Stress-Related Behavior and Hypothalamic-Pituitary-Adrenal-Axis Activity.

Introduction: Single housing of laboratory mice is a common practice to meet experimental needs, or to avoid intermale aggression. However, single housing is considered to negatively affect animal welfare and may compromise the scientific validity of experiments. The aim of this study was to investigate whether the use of a cage with a cage divider, which avoids physical contact between mice while maintaining sensory contact, may be a potential refinement strategy for experiments in which group housing of mice is not possible.Methods: Eight-week-old male C57BL/6JRj mice were single housed, pair housed or pair housed with a cage divider for four (experiment 1) or ten (experiment 2) weeks, after which we performed an open field test, Y-maze spontaneous alternation test, elevated plus maze test, an auditory fear conditioning task, and assessed responsiveness of the hypothalamic-pituitary-adrenal (HPA) axis.Results: Housing conditions did not affect body weight, exploratory activity, anxiety, working memory, fear memory processing or markers for HPA-axis functioning in either experiment 1 or experiment 2. There was an increased distance traveled in mice housed with a cage divider compared to pair housed mice after 4 weeks, and after 10 weeks mice housed with a cage divider made significantly more arm entries in the Y-maze spontaneous alternation test.Conclusion: Taken together, our study did not provide evidence for robust differences in exploratory activity, anxiety, working memory and fear memory processing in male C57BL/6JRj mice that were single housed, pair housed or pair housed with a cage divider.

Effect of Neuropeptide S Administration on Ultrasonic Vocalizations and Behaviour in Rats with Low vs. High Exploratory Activity.

Neuropeptide S (NPS) is a peptide neurotransmitter that in animal studies promotes wakefulness and arousal with simultaneous anxiety reduction, in some inconsistency with results in humans. We examined the effect of NPS on rat ultrasonic vocalizations (USV) as an index of affective state and on behaviour in novel environments in rats with persistent inter-individual differences in exploratory activity. Adult male Wistar rats were categorised as of high (HE) or low (LE) exploratory activity and NPS was administered intracerebroventricularly (i.c.v.) at a dose of 1.0 nmol/5 µL, after which USVs were recorded in the home-cage and a novel standard housing cage, and behaviour evaluated in exploration/anxiety tests. NPS induced a massive production of long and short 22 kHz USVs in the home cage that continued later in the novel environment; no effect on 50 kHz USVs were found. In LE-rats, the long 22 kHz calls were emitted at lower frequencies and were louder. The effects of NPS on behaviour appeared novelty- and test-dependent. NPS had an anxiolytic-like effect in LE-rats only in the elevated zero-maze, whereas in HE-rats, locomotor activity in the zero-maze and in a novel standard cage was increased. Thus NPS appears as a psychostimulant peptide but with a complex effect on dimensions of affect.

Does consistent individual variability in pelagic fish larval behaviour affect recruitment in nursery habitats?

Individual animals across all taxa differ consistently in behaviour, i.e. they show personality traits. This inter-individual variability has significant ecological and evolutionary consequences, since it affects a range of population-level processes. Here, we focus on the selection and recruitment of nursery habitats in temperate fish larvae. The “Sense Acuity and Behavioural Hypothesis” has proposed that fish larvae could detect and follow environmental cues to actively choose suitable nursery habitats. We empirically tested this hypothesis questioning if this non-random active process occurs and if it could be linked to consistency in individual behaviours. Individual larvae of the white seabream Diplodus sargus (Linnaeus, 1758) were tested repeatedly at different ages in a two-channel choice-chamber apparatus exposing them to a flow with different stimuli, as nursery habitats (lagoon, coastal), different temperatures or salinities and recording exploratory activity and preference in the different conditions. Most larvae changed behaviour during ontogeny, but they were also significantly consistent in their behaviour, revealing strong individuality; yet, no significant preference for the presented stimuli emerged, nor it was related to individuality. Exploratory activity was higher when larvae showed unresponsive or inconclusive behaviours, meaning that the larvae tried to find a different stimulus from the one that we were offering or had random habitat selection. Individual behavioural consistency could influence the process of searching for suitable nursery habitats and, consequently, dispersion and connectivity of white seabream population. Characterizing the behaviour of temperate pelagic marine fish larvae may shed light on fish recruitment variability, help refining larval dispersion models and possibly help understanding effects of climate change on population distribution and connectivity. A Chinese idiom says that “It is easier to change mountains and rivers than to alter one’s character.” What about fish? Well, fish can exhibit individuality traits that control autoecological and demecological processes. For example, shy fish have lower fitness while the rate of invasion progress is faster in populations with bolder individuals. Individuality studies rarely focused on fish larvae, except for coral fish. So, we tested if temperate fish larvae display consistent behaviour throughout ontogeny. This goal delves into the Sense Acuity And Behavioural Hypothesis which incorporated behaviour into the hypotheses deeming to explain fish recruitment variability. We found that temperate fish larvae display consistent individual behavioural differences in exploratory activity since early in ontogeny. This confirms the deterministic role of pelagic fish larvae behaviour on population connectivity processes, namely to control their dispersion and choose a nursery habitat.

Individual differences in exploratory activity relate to cognitive judgement bias in carpenter ants

Emotional state may influence cognitive processes such as attention and decision-making. A cognitive judgement bias is the propensity to anticipate either positive or negative consequences in response to ambiguous information. Recent work, mainly on vertebrates, showed that the response to ambiguous stimuli might change depending on an individual’s affective state, which is influenced by e.g. the social and physical environment. However, the response to ambiguous stimuli could also be affected by the individual’s behavioural type (personality), a question that has been under-investigated. We studied the link between individual differences in exploratory activity and the response to an ambiguous stimulus in the ant Camponotus aethiops. Exploratory behaviour, quantified with an open-field test, was variable among individuals but consistent over time within individuals. Individual ants learned to associate a spatial position to a reinforcement and another spatial position to a punishment. Once the ants had acquired this discrimination, cognitive judgement bias was tested with the stimulus in an intermediate position. Fast explorers in the open-field took significantly more time to approach the ambiguous stimulus compared to slow explorers, suggesting a negative judgement bias for fast explorers and a positive bias for slow explorers. This previously unknown link between individual difference in exploratory activity and cognitive bias in a social insect may help understanding the evolution and organization of social life.

Motivational disturbances and effects of L-dopa administration in neurofibromatosis-1 model mice.

Children with neurofibromatosis type 1 (NF1) frequently have cognitive and behavioral deficits. Some of these deficits have been successfully modeled in Nf1 genetically-engineered mice that develop optic gliomas (Nf1 OPG mice). In the current study, we show that abnormal motivational influences affect the behavior of Nf1 OPG mice, particularly with regard to their response to novel environmental stimuli. For example, Nf1 OPG mice made fewer spontaneous alternations in a Y-maze and fewer arm entries relative to WT controls. However, analysis of normalized alternation data demonstrated that these differences were not due to a spatial working memory deficit. Other reported behavioral results (e.g., open-field test, below) suggest that differential responses to novelty and/or other motivational influences may be more important determinants of these kinds of behavior than simple differences in locomotor activity/spontaneous movements. Importantly, normal long-term depression was observed in hippocampal slices from Nf1 OPG mice. Results from elevated plus maze testing showed that differences in exploratory activity between Nf1 OPG and WT control mice may be dependent on the environmental context (e.g., threatening or non-threatening) under which exploration is being measured. Nf1 OPG mice also exhibited decreased exploratory hole poking in a novel holeboard and showed abnormal olfactory preferences, although L-dopa (50 mg/kg) administration resolved the abnormal olfactory preference behaviors. Nf1 OPG mice displayed an attenuated response to a novel open field in terms of decreased ambulatory activity and rearing but only during the first 10 min of the session. Importantly, Nf1 OPG mice demonstrated investigative rearing deficits with regard to a novel hanging object suspended on one side of the field which were not rescued by L-dopa administration. Collectively, our results provide new data important for evaluating therapeutic treatments aimed at ameliorating NF1-associated cognitive/behavioral deficits.

D-Cycloserine enhances social exploration in the Balb/c mouse

Inbred Balb/c mice show deficits of sociability. The endogenous tone of NMDA receptor-mediated neurotransmission is altered in Balb/c mice, which may explain the beneficial effect of d-cycloserine on impaired sociability. In the current study, Balb/c mice spent more time than the Swiss Webster comparator strain in the open arms of an elevated plus maze (EPM), suggesting that they are not more anxious or fearful in the absence of a social stimulus mouse. Moreover, Balb/c and Swiss Webster mice did not differ in the amount of time they spent exploring an inanimate object in an open field. Differences in exploratory activity between strains emerged only when a salient social stimulus mouse was enclosed in the open field. d-Cycloserine increased the amount of time Balb/c mice spent exploring the enclosed stimulus mouse to levels observed in vehicle-treated Swiss Webster mice. Finally, irrespective of strain, d-cycloserine increased exploratory activity as measured in open arm entries in the EPM, when no enclosed stimulus mouse was present. The data show that mouse strain influences d-cycloserine's effect on exploration in the presence of a salient social stimulus mouse. In the absence of an enclosed stimulus mouse, d-cycloserine increased open arm entries significantly in both the sociability-impaired Balb/c and comparator Swiss Webster strains. Thus, d-cycloserine positively affects exploratory activity in general, but strain differences emerge when the stimulus eliciting exploration is a salient social stimulus mouse versus an inanimate object. Further, the sociability deficit of the Balb/c mouse is not an epiphenomenon of increased generalized anxiety.

Impaired behavior of female tg-ArcSwe APP mice in the IntelliCage: A longitudinal study

Transgenic animals expressing mutant human amyloid precursor protein (APP) are used as models for Alzheimer disease (AD). Ideally, behavioral tests improve the predictive validity of studies on animals by mirroring the functional impact of AD-like neuropathology. Learning and memory studies in APP transgenic models have been difficult to replicate. Standardization of procedures, automatization or improved protocol design can improve reproducibility. Here the IntelliCage, an automated system, was used for behavioral testing of APP female transgenic mice with both the Arctic and Swedish mutations, the tg-ArcSwe model. Protocols covering exploration, operant learning, place learning and extinction of place preference as well as passive avoidance tests were used for longitudinal characterization of behavior. Differences in exploratory activity were significant at four months of age, when plaque-free tg-ArcSwe mice visited less frequently the IntelliCage corners and initially performed fewer visits with licks compared to non-tg animals, inside the new environment. Fourteen months old tg-ArcSwe mice required a longer time to re-habituate to the IntelliCages than non-tg mice. At both ages tg-ArcSwe mice perseverated in place preference extinction test. Fourteen months old tg-ArcSwe mice were impaired in hippocampus-dependent spatial passive avoidance learning. This deficit was found to inversely correlate to calbindin-D28k immunoreactivity in the polymorphic layer of the dentate gyrus. Reduced water intake and body weight were observed in 4 months old tg-ArcSwe animals. The body weight difference increased with age. Thus behavioral and metabolic changes in the tg-ArcSwe APP model were detected using the IntelliCage, a system which provides the opportunity for standardized automated longitudinal behavioral phenotyping.

Differential gene expression in a rat model of depression based on persistent differences in exploratory activity

Affective disorders are often accompanied by changes in motivation and anxiety. We investigated the genome-wide gene expression patterns in an animal model of depression that separates Wistar rats belonging into clusters of persistently high anxiety/low motivation to explore and low anxiety/high motivation to explore (low explorers and high explorers, LE and HE, respectively), in three brain regions previously implicated in mood disorders (raphe, hippocampus and the frontal cortex). Several serotonin-, GABA-, and glutamatergic genes were differentially expressed in LE- and HE-rats. The analysis of Gene Ontology biological process terms associated with the differentially regulated genes identified a significant overrepresentation of genes involved in the neuron development, morphogenesis, and differentiation; the most enriched pathways from the Kyoto Encyclopedia of Genes and Genomes were the Wnt signalling, MAPK signalling, long-term potentiation, and long-term depression pathways. These findings corroborate some expression data from other models of depression, and suggest additional targets.

Effects of caloric restriction on cognition and behavior in developing mice

The effects of caloric restriction (CR) on cognition and behavior in developing mice were investigated in this study. Male and female Kunmin mice were fed a standard rodent diet ad libitum (Control); 80% of control (20% CR) or 65% of control (35% CR) for 6 months. Body weight gain was significant reduced in CR mice relative to control. Learning and memory retention test in a Y maze demonstrated that CR increased learning but not retention in male mice, whereas CR did not affect learning or retention in females. Open field test revealed no difference in exploratory activity between CR and control mice. These findings suggest that CR produce sex-dependent effect on cognition, but not exploratory activity, in developing animals.

Effects of dietary restriction on growth, neurobehavior, and reproduction in developing Kunmin mice.

The effects of dietary restriction (DR) on growth, neurobehavior, and reproduction in developing Kunmin mice were investigated in this study. Male and female mice were fed a standard rodent diet ad libitum (control), 80% of control (20% DR), or 65% of control (35% DR) for 3 months. Body weight of DR mice was reduced relative to control except that of females in the 20% DR group (no difference as compared with control group). Learning and memory retention test in a Y maze demonstrated that DR increased learning, but not retention, in male mice, whereas neither learning nor retention was affected in females. The open-field test revealed no difference in exploratory activity in all groups. Reproductive assessment showed that 35% DR retarded the maturation of reproductive function and reduced fertility compared with other groups. Furthermore, both 20% and 35% DR led to a lower level of sperm motility and a higher level of abnormal sperm relative to control mice. These findings indicate that DR does not cause damaging effects on growth and neurobehavior, but imposes a risk to reproductive development events.

Behavioral and pharmacological effects of centrally administered aminooxyacetic acid in rats

In the present experiment unilateral intrastriatal injections of aminooxyacetic acid (1, 2.5, 5 μmol) to freely moving animals and pentobarbital-anesthetized rats produced contralateral jerks and dose-dependent mortality, but no barrel rotation. At 10–12 days there were no significant differences in exploratory activity, passive avoidance behavior, and elevated plus-maze test in aminooxyacetic acid-treated animals as compared with controls. However freely moving animals microinjcctcd with aminooxyacetic acid (but not the pentobarbital-pretreated group) had impaired learning activity in an active avoidance conditioning test, and showed reduced striatal concentrations of substance P and GABA. Intrastriatal injections of aminooxyacetic acid therefore result in both acute and chronic behavioral changes which are attenuated by pentobarbital anesthesia.

Passive avoidance in rats and gerbils as a function of species-specific exploratory tendencies

Rats were found superior to gerbils in acquisition of step-down passive avoidance in each of three experiments. In Experiment 1 size of the safe platform was varied and rats showed comparable passive avoidance on both large and small platforms while gerbils were reliably superior in passive avoidance on the large platform relative to the small. Experiment 2 revealed that gerbils took significantly longer than rats to reach a 180-sec passive avoidance criterion, although the two species did not differ in the number of shocks received prior to reaching criterion. Finally, Experiment 3 showed that platform confinement prior to training retarded passive avoidance acquisition in gerbils while having no effect on rat performance. The results of all three experiments were interpreted in terms of species differences in exploratory activity in aversive situations. Specifically, observed species differences in passive avoidance were attributed to a stronger exploratory tendency in gerbils relative to rats.

Sex differences in extinction of food-rewarded approach responses

Female rats exhibited slower extinction of a food-rewarded running response than did males, but here was no sex difference in extinction of a barpress response that was rewarded by food. It is suggested that sex differences in exploratory activity underlie the differences in extinction.

A note strain differences in exploratory activity (rearing)

Contrary to the findings of other workers, the results of this study (based on relatively large population sample) clearly demonstrate a strain difference in the frequency of rearing responses of the Maudsley strains of rats which have been selectively brad for differences in emotional elimination. An explanation of these findings is offered which makes it unnecessary to posit differences in innate CNS excitability between the strains.

Conditioned-hunger drive as indicated by avoidance of a situation previously associated with hunger

This investigation concerned evidence for conditioned hunger in terms of the tendency for animals to leave an environment previously associated with hunger. The Ss were placed in a distinctive white box for 30 minutes a day for 45 days; one group always was 21½-hours hungry, a control group was relatively satiated (one-hour hungry). The Ss were then tested by being allowed a 10-minute period in the white box during which they could move freely into (or out of) a black box attached to one side of the white box. The amount of time out of the 10-minute period that Ss spent in the black box was recorded along with the number of crossings between the white and black box. The Ss were tested on two separate days. They were satiated the first test day, 22-hours hungry the second test day. Experimental Ss spent significantly more time in the black box than the controls and made significantly more crossings. Various interpretations of the differences in exploratory behavior were discussed. In view of the greater number of crossings the results were regarded as reflecting differences in exploratory activity rather than avoidance of the white box.
