Mercury monitoring in farmed Pacific bluefin tuna (Thunnus orientalis) using liquid asymmetric-electrode plasma optical emission spectroscopy: Ventricle tissue as a potential indicator

Tuna muscle is known to contain high levels of mercury, necessitating the monitoring of mercury concentrations in farmed fish at production sites. There is currently no straightforward in on-site method for testing whether the mercury levels in farmed tuna exceed regulatory limits. This study investigated the potential application of liquid asymmetric-electrode plasma optical emission spectroscopy (LAEP-OES) to estimate mercury concentrations in the tissues of cultured Pacific bluefin tuna at aquaculture farms. No significant differences in mercury concentrations were observed between the ventricle tissue and biopsied tissue from tuna bleeding site (TTBS), which can be harvested without reducing the market value of farmed tuna (Hg concentration of the ventricle tissue and the TTBS, median: 0.41 and 0.40 mg/kg, respectively). In addition, the relationship between mercury concentrations in the ventricle tissue and in the edible portions of three tuna fish was examined. The findings showed that total mercury level in the edible tissues varied, with the ratio of the mercury level in edible tissues to that in the ventricle ranging from 0.39 to 1.08. A positive correlation was observed between the average total mercury levels in edible tissues and ventricle tissue. These results suggest that determining the total mercury level in the ventricle, which is typically discarded at processing sites, may serve as a convenient proxy for quality control and monitoring purposes using the LAEP-OES method. However, larger sample sizes are needed to validate the proposed approach more robustly.

Total mercury in fur, whiskers and whole blood of Caspian seals (Pusa capsica) from north-east of Caspian Sea (Kazakhstan)

Total Mercury (THg) content was determined in the fur of 64 Caspian seals, in the whiskers of 59 individuals and whole blood of 29 individuals. The THg content in Caspian seal fur varied from 258 to 8511 μg/kg, in whiskers from 954 to 12,957 μg/kg, and in whole blood from 88 to 350 μg/l. There were no statistically significant differences in mercury concentration in biomaterial between males and females (Kruskal-Wallis test, p < 0.05). The 1–2-year-old seals contained less mercury compared to older seals. The THg content in Caspian seal samples was comparable to seals from different regions of North Eurasia. Four individuals had mercury concentrations in their fur above the threshold values that can lead to nervous system disorders (>5400 μg/kg).

Seasonal and age-dependent differences in mercury concentrations in Apodemus sp. in the north-western region of Slovakia

Pollution of ecosystems by heavy metals such as mercury is currently a great concern. Mercury (Hg) can be released into the environment anthropogenically, but it is also naturally present in small quantities in all environmental compartments. Many different factors contribute to different rates of Hg deposition in animal bodies. The aim of this work is to describe how Hg concentrations in the bodies of small rodents change throughout the season at a site where massive anthropogenic pollution is not expected. Mice of the genus Apodemus were sampled during the whole year. Samples of blood, hair, liver, kidney, and brain were analyzed. Total Hg concentrations were measured by DMA-80. The mean Hg concentrations in examined organs were in the order hairs > kidney > liver > blood > brain, and their values decreased from 0.0500 to 0.0046 mg kg−1 dry weight. Males and females did not differ in contamination levels, but age-dependent differences in Hg concentrations were found. It was also identified how Hg concentrations in different organs correlate with each other. Different levels of seasonal variability were detected in Hg concentrations in blood, hair, and kidney.

Evaluation of Mercury Pollution and Risk Assessment of Human Health due to Consumption of Common Carp (Cyprinus Carpio) from the Caspian Sea

Background: In the present study, the main goal was to measure the concentration of mercury in the muscle tissue of common carp and evaluate the health risk to consumers using different indices, and evaluate the possible difference between the values of the studied element in the sampling stations. Methods: Samples of the studied species were collected from four different stations, Bandar torkaman, Faridunknar, Chalus, and Bandar Anzali, from the southern shores of the Caspian sea. Kruskal-wallis and Mann-whitney u tests were used to determine the difference in mercury concentration in fish muscle tissue at the stations.

Mercury distribution in fish organs sampled along the Mauritanian Atlantic coast and their potential human health risks

This paper aimed at assessing total mercury concentration in seven common fish species (Auxis rochei, Caranx rhonchus, Sardina pilchardus, Sardinella aurita, Sardinella maderensis, Scomber colias and Trachurus trecae) and a relationship between Hg organotropism and food regimes along the Mauritanian Atlantic coast. Results show that total mercury concentration in fish collected along five sites ranged from 0.027 to 0.533 mg/kg dry weight. Significant differences were observed among species depending on feeding behavior. Muscle tissues of carnivorous fish presented significantly higher levels of total mercury than that of omnivorous fish, except for Scomber colias, suggesting mercury biomagnification through the food chain. Significant differences in mercury concentrations were observed between muscle tissues and liver, for Auxis rochei, Trachurus trecae, and Caranx rhonchus.The mean concentrations in the different species are however low and none of the concentration values exceed the World Health Organization's threshold for human consumption.

Mercury Concentration in Saliva and the Impact of Chewing: An Inductively Coupled Plasma Mass Spectrometry Study.

Mercury, which is found in dental amalgams, is considered to be the most toxic non-radioactive element. However, the health policies of different countries have not reached a consensus on the use and safety of amalgam. This study aims to investigate the effect of amalgam restorations on mercury concentration in saliva, as well as the effect of restoration number, surface number, and chewing on this concentration. A total of 86 participants were included in this study (an equal number for the study and control groups). The number of amalgam restorations and their surfaces were recorded. While both unstimulated and stimulated saliva were collected from the study group, only unstimulated saliva was collected from the control group. The effect of chewing on mercury concentration was examined in the study group with unstimulated and stimulated saliva specimens using inductively coupled plasma mass spectrometry device. Mercury concentration in the unstimulated saliva was found to be higher in the study group compared to the control group (p= 0.000). As the number of amalgam restorations and the number of amalgam restoration surfaces increased, the mercury concentration in the saliva increased (p= 0.015 and p= 0.021, respectively). There was no statistically significant difference between mercury levels in the unstimulated saliva and the stimulated saliva (p=0.316). Chewing presented an insignificant difference in mercury concentration. Given this surprising result, the effect of chewing on mercury concentrations should be explored more extensively in future research.

Spatial and temporal variability of mercury in Upper and Lower Red Lake Walleye.

Mercury is a global pollutant that is released into our environment by natural and anthropogenic processes resulting in extensive studies of mercury cycling in aquatic ecosystems, and the issuance of human-health-based fish-consumption advisories. We examined total mercury concentrations in Walleye Sander vitreus from Upper and Lower Red Lakes, located in north central Minnesota, between 2019 and 2020. Sampled Walleye (n = 265) ranged from 158 to 610 mm in total length from an age range of young-of-the year to 16 years. Mercury concentrations within the Walleye ranged from 0.030 mg/kg to 0.564 mg/kg (x̄ = 0.179 ± 0.105 mg/kg; x̄ = mean ± sd, all fish-mercury concentrations expressed on wet-weight basis). The best supported model for predicting mercury concentrations in Red Lake Walleye included the independent variables: length, age, sex, and lake basin. This model indicated that there was a significant difference in mercury concentrations between Upper and Lower Red Lake (x̄ = 0.215 ± 0.117 and 0.144 ± 0.077 mg/kg, respectively), and also suggests that individuals who rely on fish for subsistence should target Walleye that are ≤ 400 mm from Lower Red Lake. Observed differences in mercury concentrations could be linked to wetland area influences, fish growth rates, and physicochemical parameters between the two basins. Given that our results illustrated a significant difference in fish-mercury concentrations between basins, future pollutant monitoring efforts should treat Upper and Lower Red Lake as separate lakes and not assume that data from one basin can apply to the other.

Significance of Intestinal Helminth Infection and Animal Sex for Mercury Concentrations in Two Rodent Species.

We compared the effects of animal gender, species, and intestinal helminth burden on mercury concentrations in rodents. Total mercury concentrations were determined in the liver and kidney tissues of 80 small rodents (44 yellow-necked mice, Apodemus flavicollis, and 36 bank voles, Myodes glareolus) captured in the Ore Mountains (northwest Bohemia, Czech Republic). Overall, 25/80 (32%) of animals were infected by intestinal helminths. The differences in mercury concentration between rodents infected and not infected with intestinal helminths were not statistically significant. Statistically significant differences in mercury concentrations were found only between voles and mice (that were not infected with intestinal helminths). This suggests the differences may be associated with host genetics. Apodemus flavicollis body tissues had significantly lower (P=0.01) mean Hg concentrations (0.032 mg/kg) than Myodes glareolus (0.279 mg/kg), provided that animals were not infected by intestinal helminths; if the animals were infected by intestinal helminths, the difference between both groups was insignificant. The effect of gender in this study was significant only for voles (without helminth infection); for mice (either with or without helminth infection) the differences between genders were not significant. Myodes glareolus males had significantly lower (P=0.03) Hg concentrations in liver and kidney tissues (0.050 mg/kg) than Myodes glareolus females (0.122 mg/kg). These results reveal the importance of considering species and gender when evaluating mercury concentrations.

Differences in Mercury Concentrations in Water and Hydrobionts of the Crimean Saline Lakes: Does Only Salinity Matter?

Of significant scientific and public concern is the high toxicity, significant bioaccumulation, and magnified concentration within the food web of mercury (Hg). Hg content both dissolved and in suspended forms in water as well as in biomass of different hydrobiont taxa was studied in 18 saline lakes in Crimea from 2012 to 2021. The impact of different factors (salinity, seasonality, anthropogenic activities, geological background, etc.) was analyzed. The generalization of data for all lakes showed that the average concentration of Hg in dissolved form was 129 ng L−1, varying over a wide range. The content of Hg in total suspended substrates was an average of 151 ng L−1, and the total content of Hg in lake water averaged 291 ng L−1. Geological background and anthropogenic activities can determine the total Hg content in lakes. In most lakes, a significant positive correlation was noted between the concentration of one, two, or all three indicators of Hg content and the month of the year, with indicators increasing from winter through to autumn. When analyzing the entire data array, a significant positive correlation was found between the concentration of the suspended form of Hg and salinity; such correlation between the concentration of the dissolved form of Hg and salinity was absent. The highest average Hg concentrations in biomasses were noted in Artemia and were the lowest in plants. Geological background and human activity contribute to high Hg content in lakes. Hydrobionts can significantly influence the distribution and behavior of Hg, being an important factor of its cycle in the lakes.

Fish consumption habits of pregnant women in Itaituba, Tapajós River basin, Brazil: risks of mercury contamination as assessed by measuring total mercury in highly consumed piscivore fish species and in hair of pregnant women.

The Tapajós River basin in the Amazon region, Brazil is one of the most active gold mining areas in the world. In this study, we evaluated fish consumption habits and mercury exposure in 110 pregnant women in the city of Itaituba by measuring their total hair mercury concentrations. In addition, we investigated seasonal differences in mercury concentrations in two highly consumed piscivorous fish species, tucunaré (Cichla spp.) and pescada (Plagioscion squamosissimus). Total fish mercury concentrations (THg) during the dry season were 0.62±0.07 mg/kg for Cichla spp. and 0.73±0.08 mg/kg for P. squamosissimus. During the rainy season they were 0.39±0.04 and 0.84±0.08 mg/ kg, respectively. Of our participants 44 % declared that they ate Cichla spp. and 67 % P. squamosissimus. Mean mercury concentration in their hair was 1.6±0.2 mg/kg and was above the US Environmental Protection Agency (US EPA) reference dose of 1 mg/kg in 48 % of them. Mean fish THg concentrations were also above the joint Food and Drug Administration and US EPA safety limit of 0.5 mg/kg for P. squamosissimus during both seasons and for Cichla spp. during the dry season only. These results show that pregnant women should avoid consumption of these piscivorous fish species during pregnancy and call for a regular programme to monitor Hg levels in that area.

Variations in mercury concentrations in the muscles of fish in biotopes within the water body and in different water bodies of Russia

Abstract. In recent decades, researchers from Papanin Institute for Biology of Inland Waters Russian Academy of Sciences have been studying mercury concentrations in the muscles of fish from water bodies situated in different natural and climatic zones. Overall, more than 5000 fish samples from 102 lakes and 35 rivers in Russia were analysed. In the absence of local sources of mercury entering the water body, its concentrations in the fish muscles varied in wide ranges, exceeding two orders of magnitude. Minimum concentration (&lt;0.03 mg/kg wet weight) was recorded in the muscles of omul, rotan (Chinese sleeper), minnow, and bleak (Lake Baikal, Transbaikalia and European Russia). Maximum mercury concentration (2-3 mg/kg wet weight) was recorded in the muscles of perches from lakes in the northwest of Russia (Vologda and Novgorod regions, Karelia). Most of the measurement results ranged from 0.05 to 0.30 mg/kg wet weight. Differences in mercury concentrations in the muscles of fish of the same species, similar in size and from one water body, as a rule, did not exceed ranges of one order of magnitude. In the absence of local mercury sources, mercury concentrations in fish muscles from closely spaced water bodies could have more than tenfold differences.

Utilization of black-tailed gull (Larus crassirostris) eggs for monitoring of mercury levels in coastal areas of South Korea: Preliminary study

Studies on the monitoring of mercury accumulation using high trophic-level predators of the marine ecosystem have been scarce in South Korea. In this study, we compared the mercury concentrations of the eggs of the black-tailed gulls, a higher-order predator, breeding in two coastal areas. Breeding sites with varying mercury concentrations in land-origin freshwater fish and freshwater and marine sediments were selected in the southeastern (Hongdo Island) and western (Baengnyeongdo Island) seas. The 5-year mean total mercury concentration in eggs collected during the breeding seasons from 2012 to 2016 was higher in those collected from Hongdo than in those collected from Baengnyeongdo. This difference in mercury concentration in eggs was observed for each year. In addition, the total mercury concentration in eggs was consistently higher on Hongdo, which also had higher mercury pollution, than on Baengnyeongdo Island. These results support the suitability of black-tailed gull eggs for monitoring of mercury pollution.

Mercury contamination of the snow voles (Chionomys nivalis) in the West Carpathians.

The subject of this research is mercury pollution in the alpine zone of the High Tatra Mountains (the West Carpathians). The snow vole (Chionomys nivalis) was chosen as a bioindicator. Mercury concentrations in the tail tips from captured individuals were examined. The samples were collected over the span of 2 years, from May 2016 to November 2017. The smaller (1-year-old) individuals had higher mercury concentrations in their tails than the bigger (2-year-old) individuals, suggesting recent aerial contamination of alpine habitats. The amount of mercury found in the tails did not differ between sexes. The differences in mercury concentration between animals caught in the spring and autumn were also insignificant. The possible relationship between mercury in the tissues and the seasonal variability of the water cycle in the mountains (snow melting, precipitation) is discussed.

Total Mercury Levels in Commercial Fish in Market of the Republic of Srpska, Bosnia and Herzegovina.

The mercury concentration was quantified in frozen and canned fish found in the market of the Republic of Srpska, Bosnia and Herzegovina. Using the method of amalgamation by atomic absorption spectrophotometry with a direct mercury analyzer (DMA-80), significant differences in mercury concentrations among the analyzed fish were found. Frozen pangasius and canned tuna had the highest mercury concentration (59.1μg/kg and 54.1μg/kg, respectively). Also, fish from Vietnam had the highest level of total mercury in comparison to fish from other countries. Determined levels of mercury in our samples were below the limit set by the European and Bosnian and Herzegovinian legislation (500μg/kg wet weight). We have concluded that consumption of commercial fish in the market of the Republic of Srpska does not present any health risk for the average consumer. Nevertheless, a surveillance system of mercury content in fishery products, especially in certain species that concentrate mercury, is crucial for public health protection.

Ecotoxicoparasitology of mercury and trace elements in semi-aquatic mammals and their endoparasite communities

Many contaminants persist in the environment for decades or more, influencing ecosystem health. Environmental contamination with mercury (Hg) is a particular concern due to its ability to biomagnify in food webs and its lethal and sub-lethal effects in exposed organisms. Despite the known impacts of anthropogenic contamination, there remains a need for data on wildlife exposure to Hg and other contaminants, and the effects of exposure on wildlife health. The objectives of this study were to: 1) quantify differences in concentrations of mercury and other trace elements among three sympatric semiaquatic mammals of different assumed trophic position: North American river otter (Lontra canadensis), raccoon (Procyon lotor), and North American beaver (Castor canadensis), 2) compare trace element concentrations between animals captured on the Savannah River Site (SRS) in South Carolina, USA, where known inputs of Hg and other trace elements have occurred, and reference sites in South Carolina (SC) and Georgia (GA), USA, and 3) investigate the relationship between host trace element concentrations and endoparasite communities. River otters, beavers, and raccoons were sampled from the SRS, SC, and GA to quantify trace element concentrations in liver tissue and quantify endoparasite communities. Both species and sampling location were important factors determining hepatic trace element concentration, however, there was no consistent trend of elevated trace element concentrations among animals sampled on the SRS. Only Hg demonstrated biomagnification based on assumed trophic position, with river otters having the highest Hg concentrations among the sampled species. Additionally, the results suggest a possible relationship between host hepatic mercury concentration and endoparasite abundance, while hepatic selenium concentration may be related to endoparasite diversity. These findings further demonstrate how wildlife can accumulate anthropogenic contamination, although future research is needed to determine the mechanisms contributing to patterns observed between endoparasite communities and the contaminant concentrations of their mammalian hosts.

Effect of Metallothionein-III on Mercury-Induced Chemokine Gene Expression.

Mercury compounds are known to cause central nervous system disorders; however the detailed molecular mechanisms of their actions remain unclear. Methylmercury increases the expression of several chemokine genes, specifically in the brain, while metallothionein-III (MT-III) has a protective role against various brain diseases. In this study, we investigated the involvement of MT-III in chemokine gene expression changes in response to methylmercury and mercury vapor in the cerebrum and cerebellum of wild-type mice and MT-III null mice. No difference in mercury concentration was observed between the wild-type mice and MT-III null mice in any brain tissue examined. The expression of Ccl3 in the cerebrum and of Cxcl10 in the cerebellum was increased by methylmercury in the MT-III null but not the wild-type mice. The expression of Ccl7 in the cerebellum was increased by mercury vapor in the MT-III null mice but not the wild-type mice. However, the expression of Ccl12 and Cxcl12 was increased in the cerebrum by methylmercury only in the wild-type mice and the expression of Ccl3 in the cerebellum was increased by mercury vapor only in the wild-type mice. These results indicate that MT-III does not affect mercury accumulation in the brain, but that it affects the expression of some chemokine genes in response to mercury compounds.

Spatial and Ontogenetic Variation in Mercury in Lake Superior Basin Sea Lamprey (Petromyzon marinus).

Mercury concentrations were measured in eggs, larvae, and adult spawning-phase sea lampreys (Petromyzon marinus) collected in tributaries of Lake Superior to investigate spatial and ontogenetic variation. There were significant differences in mercury concentrations between all three life stages, with levels highest in adults (mean = 3.01µg/g), followed by eggs (mean = 0.942µg/g), and lowest in larvae (mean = 0.455µg/g). There were no significant differences in mercury concentrations by location for any life stage or by sex in adults. Mercury was not correlated with adult or larval lamprey length or mass. Mercury levels in adult lampreys exceeded U.S. and Canadian federal guidelines for human consumption. Mercury concentrations in all life stages exceeded criteria for the protection of piscivorous wildlife, posing a threat to local fish, birds, and mammals. High mercury levels in adult lampreys combined with their semelparous life history make them a potential source of lake-derived mercury to spawning streams.

Direct and indirect causes of sex differences in mercury concentrations and parasitic infections in a marine bird

In many animal species, males and females differ in their levels of contaminants and/or parasitic infections. Most contaminants and gastro-intestinal parasites are obtained through prey ingestion, and thus the causes of sex differences in the distribution of these factors might follow similar pathways. We studied the northern common eider duck (Somateria molissima borealis) as an avian model, and used directed separation path analysis to explore the causes of sex differences in mercury (Hg) and gastro-intestinal helminths. Two trophically transmitted helminths were examined: a cestode (Lateriporus sp.) and an acanthocephalan (Polymorphus sp). We found that the number of Lateriporus sp. varied positively with stable isotope signature (as indicated by δ15N in eider breast muscle tissue), and negatively with crustaceans being present in the short term diet. We also found that Polymorphus sp. varied positively with eider tissue stable isotope signature. However, Polymorphus sp. varied negatively with sex indirectly through condition and liver mass. Similarly, Hg concentrations also varied negatively with sex indirectly through condition and liver mass, with both Polymorphus sp. intensity and Hg concentrations significantly higher in males. We found that model fit increased when a negative relationship between the two helminth species was included, suggesting a yet unknown causal mechanism linking these parasites. Our findings suggest that although Hg and gastro-intestinal parasites are both trophically transmitted through the eider's prey items, the factors that contribute towards bioaccumulation of these two burdens differ in source, likely caused by several different factors and may potentially influence each other.

Scrobicularia plana (Mollusca, Bivalvia) as a biomonitor for mercury contamination in Portuguese estuaries

The main goal of this research was to test the effectiveness of Scrobicularia plana to reflect environmental mercury concentrations, with the aim to propose it as a suitable, sentinel mercury biomonitor.Significant differences in mercury concentrations were observed between small and larger sized individuals in the most contaminated sampling sites, suggesting progressive mercury accumulation with size. Larger size classes did not significantly differ, however, indicating that organisms attain an equilibrium state between the mercury assimilation and excretion rates. Therefore, the average concentration of individuals larger than 2cm shell length is indicative of the local contaminant bioavailability.The diet emerges as the source of contaminant incorporation in this species, which reflects sediment and suspended particulate matter (SPM) contamination regardless of the degree of environmental contamination.Such consistent bioaccumulation behavior, its widespread distribution, abundance, ease of collection, taxonomic identification and resistance to contamination, emphasize the usefulness of S. plana as a cosmopolitan biomonitor of mercury contamination in temperate estuarine systems.

Distribution of Mercury in a Gadoid Fish Species, Tusk (Brosme brosme), and Its Implication for Food Safety

The tusk (Brosme brosme) is a popular gadoid fish for human consumption, which also is susceptible to mercury accumulation. However, few data of mercury levels in tusk exist, and it is difficult to compare tusk fished from assumed contaminated areas with background levels. The aim of this study was to increase the information of mercury levels observed in tusk from the Northeast Atlantic Ocean. Samples of tusk were collected from seven different localities in open ocean areas, nine along the Norwegian coastal current (NCC), and from two fjord stations. Determinations of mercury in muscle and liver were carried out using inductive coupled plasma mass spectrometry (ICPMS) after microwave-assisted wet digestion. The highest concentrations of mercury in muscle were found in tusk captured from the fjords Hardangerfjord and Sorfjord, and had mean concentrations of mercury at 1.5 and 1.7 mg kg -1 wet weight (ww), respectively, where all of the tusk exceeded EU's upper limit of 0.5 mg kg -1 ww. The lowest concentrations of mercury were observed in tusk captured from the open ocean areas, and the concentrations ranged from 0.08 to 0.30 mg kg -1 ww. Mercury concentrations of tusk captured along the NCC ranged from 0.21 to 0.49 mg kg -1 ww. Mercury concentrations increased with the weight of the tusk, and variance analysis (ANOVA) of weight normalized mercury concentrations showed that there were significant differences in mercury concentrations between tusk collected from the open ocean, NCC and fjords, which indicates that differences in mercury concentration between different areas arecaused by other factors.
