MiRNA 24-3p-rich exosomes functionalized DEGMA-modified hyaluronic acid hydrogels for corneal epithelial healing.

The damage of corneal epithelium may lead to the formation of irreversible corneal opacities and even blindness. The migration rate of corneal epithelial cells directly affects corneal repair. Here, we explored ocu-microRNA 24-3p (miRNA 24-3p) that can promote rabbit corneal epithelial cells migration and cornea repair. Exosomes, an excellent transport carrier, were exacted from adipose derived mesenchymal stem cells for loading with miRNA 24-3p to prepare miRNA 24-3p-rich exosomes (Exos-miRNA 24-3p). It can accelerate corneal epithelial migration in vitro and in vivo. For application in cornea alkali burns, we further modified hyaluronic acid with di(ethylene glycol) monomethyl ether methacrylate (DEGMA) to obtain a thermosensitive hydrogel, also reported a thermosensitive DEGMA-modified hyaluronic acid hydrogel (THH) for the controlled release of Exos-miRNA 24-3p. It formed a highly uniform and clear thin layer on the ocular surface to resist clearance from blinking and extended the drug-ocular-epithelium contact time. The use of THH-3/Exos-miRNA 24-3p for 28 days after alkali burn injury accelerated corneal epithelial defect healing and epithelial maturation. It also reduced corneal stromal fibrosis and macrophage activation. MiRNA 24-3p-rich exosomes functionalized DEGMA-modified hyaluronic acid hydrogel as a multilevel delivery strategy has a potential use for cell-free therapy of corneal epithelial regeneration.

A Temperature/pH Double-Responsive and Physical Double-Crosslinked Hydrogel Based on PLA and Histidine.

Hydrogel is a good drug carrier, widely used in the sustained-release aspect of tumor drugs, which can achieve the continuous release of drugs to the tumor sites. In this study, diethylene glycol monomethyl ether methacrylate (MEO2MA) and poly (ethylene glycol) methyl ether methacrylate (OEGMA) are temperature-sensitive monomers. N-Methacryloyl-L-Histidine (Mist) is pH sensitive monomer and ligand for metal coordination bond. The temperature-sensitive monomers and pH sensitive monomer with stereocomplex of modified polylactic acid (HEMA-PLLA30/PDLA30) were mixed, under 2,2’-azobis (2-methylpropionitrile) (AIBN) as radical initiator, polymer was formed by free-radical polymerization. The polymer was then immersed in ZnSO4 solution, the imidazole group of Mist monomer forms a tridentate metal coordination bond with Zn2+, temperature/pH double-responsive and physical double-crosslinked hydrogel was finally obtained. Comparing the hydrogen bond hydrogel, hydrogen bond and metal coordination bond double crosslinking hydrogel, metal coordination bond hydrogel, testing thermal stability, viscoelasticity, swelling, and morphology of three hydrogels. In addition, using UV-Visible spectroscopy (UV-Vis) to test the sustained release of the hydrophobic drug doxorubicin hydrochloride (DOX-HCl) in the human tumor environment (37 °C, pH = 5). We found that the temperature/pH double-responsive and physical double-crosslinked hydrogel had the most potential for the sustained drug release.

Effect of Thermal Stimulus on Kinetic Rehydration of Thermoresponsive Poly(diethylene glycol monomethyl ether methacrylate)-block-poly(poly(ethylene glycol) methyl ether methacrylate) Thin Films Probed by In Situ Neutron Reflectivity.

The kinetic rehydration of thin di-block copolymer poly(diethylene glycol monomethyl ether methacrylate)-block-poly(poly(ethylene glycol) methyl ether methacrylate) (PO2-b-PO300) films containing two thermoresponsive components is probed by in situ neutron reflectivity (NR) with different thermal stimuli in the D2O vapor atmosphere. The transition temperatures (TTs) of PO2 and PO300 blocks are 25 and 60 °C, respectively. After the one-step stimulus (rapid decrease in temperature from 60 to 20 °C), the film directly switches from a collapsed to a fully swollen state. The rehydration process is divided into four steps: (a) D2O condensation, (b) D2O absorption, (c) D2O evaporation, and (d) film reswelling. However, the film presents a different rehydration behavior when the thermal stimulus is separated into two smaller steps (first decrease from 60 to 40 °C and then to 20 °C). The film first switches from a collapsed to a semiswollen state caused by the rehydrated PO300 blocks after the first step of thermal stimulus (60 to 40 °C) and then to a swollen state induced by the rehydrated PO2 blocks after the second step (40 to 20 °C). Thus, the kinetic responses are distinct from that after the one-step thermal stimulus. Both the time and extent of condensation as well as evaporation processes are significantly reduced in these two smaller steps. However, the final states of the rehydrated PO2-b-PO300 films are basically identical irrespective of the applied thermal stimulus. Thus, the final state of thermoresponsive di-block copolymer films is not affected by the external thermal stimuli, which is beneficial for the design and preparation of sensors or switches based on thermoresponsive polymer films.

Ultrathin Yet Robust Single Lithium-Ion Conducting Quasi-Solid-State Polymer-Brush Electrolytes Enable Ultralong-Life and Dendrite-Free Lithium-Metal Batteries.

Quasi-solid-state polymer electrolytes are one of the most promising candidates for long-life lithium-metal batteries. However, introduction of plasticizers for high ion conductivity at room temperature inevitably gives rise to poor mechanical strength and requires a very thick electrolyte membrane, which is detrimental to safety and energy density of the batteries. Herein, inspired by tube brushes coupling hardness with softness, a novel superstructured polymer bottlebrush BC-g-PLiSTFSI-b-PEGM (BC = bacterial cellulose; PLiSTFSI = poly(lithium 4-styrenesulfonyl-(trifluoromethylsulfonyl) imide); PEGM = poly(diethylene glycol monomethyl ether methacrylate)) with a hard nanofibril backbone and soft functional polymer side-chains is reported as an effective strategy to well balance the mechanical strength and ion conductivity of quasi-solid-state polymer electrolytes. The resulting single lithium-ion conducting quasi-solid-state polymer-brush electrolytes (SLIC-QSPBEs) integrate the features of the ultrathin membrane thickness (10µm), the nanofibril backbone-strengthened porous nanonetwork (Young's modulus = 1.9GPa), and the high-rate single lithium-ion conducting diblock copolymer brushes. As a result, the ultrathin yet robust SLIC-QSPBEs enable ultralong-term (over 3300 h) reversible and stable lithium plating/stripping in Li/Li symmetrical cell at a current density of 1mA cm-2 for lithium anode. This work affords a promising strategy to develop advanced electrolytes for solid-state lithium-metal batteries.

Hydration and Thermal Response Kinetics of a Cross-Linked Thermoresponsive Copolymer Film on a Hydrophobic PAN Substrate Coating Probed by In Situ Neutron Reflectivity.

The hydration and thermal response kinetics of the cross-linked thermoresponsive copolymer poly((diethylene glycol monomethyl ether methacrylate)-co-poly(ethylene glycol) methyl ether methacrylate), abbreviated as P(MEO2MA-co-OEGMA300), thin film on a hydrophobic polyacrylonitrile (PAN) substrate coating, which resembles a synthetic fabric, is probed by in situ neutron reflectivity (NR). The PAN and monomer (MEO2MA and OEGMA300) solutions are sequentially spin-coated onto a silicon (Si) substrate. Afterward, plasma treatment is applied to realize the cross-linking of PAN and monomers. The as-prepared cross-linked P(MEO2MA-co-OEGMA300) film on the hydrophobic PAN substrate coating presents a two-layer structure: a substrate-near layer, which is a mixture of PAN and P(MEO2MA-co-OEGMA300), and a main layer, which is composed of pure hydrophilic P(MEO2MA-co-OEGMA300). During hydration in D2O vapor atmosphere, the hydrophobic PAN component prevents the formation of D2O enrichment in the substrate-near layer. However, an additional vapor-near layer is observed on top of the main layer, which is enriched with D2O. The hydration process is constrained by the cross-linking points in the film, inducing the relaxation time to be longer than that in a spin-coated P(MEO2MA-co-OEGMA300) film. Because the as-prepared cross-linked film presents a transition temperature (TT) at 38 °C, the hydrated film switches to the collapsed state when the temperature is increased from 23 to 50 °C. The response to a thermal stimulus is also slower due to the existence of the internal cross-linking points as compared to the spin-coated film. Interestingly, no reswelling is observed at the end of the thermal stimulus, which can be also attributed to the presence of internal cross-linking points.

Thermo-responsive cotton fabric prepared by enzyme-initiated “graft from” polymerization for moisture/thermal management

Thermo-responsive textiles with moisture/thermal management characteristics have attracted extensive attention for the purpose of improving human comfort or special job requirements. In this study, a thermo-responsive cotton (TR-cotton) fabric for moisture/thermal management was prepared by horseradish peroxidase/acetylacetone (ACAC)/hydrogen peroxide (H2O2)-initiated “graft from” polymerization on the fiber surface. Enzymatic graft polymerization is a new environmentally friendly method to prepare thermo-responsive textiles. Diethylene glycol monomethyl ether methacrylate (MEO2MA) and poly(ethylene glycol) methyl ether methacrylate (OEGMA500) were selected as thermosensitive monomers for graft copolymerization. The lower critical solution temperature (LCST) of P(MEO2MA-co-OEGMA500) was 29 °C. TR-cotton was hydrophilic at ambient temperature below the LCST and hydrophobic at a temperature above the LCST. The air/moisture permeability of TR-cotton at a high temperature (> LCST) was better than that at a low temperature (< LCST). In a cold environment, the fabric exhibited a heat preservation effect (~ 1.5 °C warmer than that observed for unmodified cotton fabric). This study could provide an enzyme catalysis method for the development of “smart textiles”.

Impact of Thermal History on the Kinetic Response of Thermoresponsive Poly(diethylene glycol monomethyl ether methacrylate)-block-poly(poly(ethylene glycol)methyl ether methacrylate) Thin Films Investigated by In Situ Neutron Reflectivity.

The impact of thermal history on the kinetic response of thin thermoresponsive diblock copolymer poly(diethylene glycol monomethyl ether methacrylate)-block-poly(poly(ethylene glycol) methyl ether methacrylate), abbreviated as PMEO2MA-b-POEGMA300, films is investigated by in situ neutron reflectivity. The PMEO2MA and POEGMA300 blocks are both thermoresponsive polymers with a lower critical solution temperature. Their transition temperatures (TTs) are around 25 °C (TT1, PMEO2MA) and 60 °C (TT2, POEGMA300). Thus, by applying different temperature protocols (20 to 60 or 20 to 40 to 60 °C), the PMEO2MA-b-POEGMA300 thin films experience different thermal histories: the first protocol directly switches from a swollen to a collapsed state, whereas the second one switches first from a swollen to a semicollapsed and finally to a collapsed state. Although the applied thermal histories differ, the response and final state of the collapsed films are very close to each other. After the thermal stimulus, both films present a complicated response composed of an initial shrinkage, followed by a rearrangement. Interestingly, a subsequent reswelling of the collapsed film is only observed in the case of having applied a thermal stimulus of 20 to 40 °C. The normalized film thickness and the D2O amount of each layer in the PMEO2MA-b-POEGMA300 films are consistent at the end of the two different thermal stimuli. Hence, it can be concluded that the thermal history does not influence the final state of the PMEO2MA-b-POEGMA300 films upon heating. Based on this property, these thin films are especially suitable for the temperature switches on the nanoscale, which may experience different thermal histories.

The Importance of Cooperativity in Polymer Blending: Toward Controlling the Thermoresponsive Behavior of Blended Block Copolymer Micelles.

Understanding, predicting, and controlling the self-assembly behavior of stimuli-responsive block copolymers remains a pertinent challenge. As such, the copolymer blending protocol provides an accessible methodology for obtaining a range of intermediate polymeric nanostructures simply by blending two or more block copolymers in the desired molar ratio to target specific stimuli-responsiveness. Herein, thermoresponsive diblock copolymers are blended in various combinations to investigate whether the resultant cloud point temperature can be modulated by simple manipulation of the molar ratio. Thermoresponsive amphiphilic diblock copolymers composed of statistical poly(n-butyl acrylate-co-N,N-dimethylacrylamide) core-forming blocks and four different thermoresponsive corona-forming blocks, namely poly(diethylene glycol monomethyl ether methacrylate) (p(DEGMA)), poly(N-isopropylacrylamide), poly(N,N-diethylacrylamide), and poly(oligo(ethylene glycol) monomethyl ether methacrylate) (p(OEGMA)) are selected for evaluation. Using variable temperature turbidimetry, the thermoresponsive behavior of blended diblock copolymer self-assemblies is assessed and compared to the thermoresponsive behavior of the constituent pure diblock copolymer micelles to determine whether comicellization is achieved and more significantly, whether the two blended corona-forming thermoresponsive blocks exhibit cooperative behavior. Interestingly, blended diblock copolymer micelles composed of p(DEGMA)/p(OEGMA) mixed coronae display cooperative behavior, highlighting the potential of copolymer blending for the preparation of stimuli-responsive nanomaterials in applications such as oil recovery, drug delivery, biosensing, and catalysis.

Structural Colored Balloon Composed of Temperature-Responsive Polymers Showing LCST Behavior.

Structural colored balloons (SCBs) consisting of polymer thin film developed structural color by thin-layer interference on the shell. Thermoresponsive SCBs were prepared with poly(diethylene glycol monomethyl ether methacrylate)- co-poly( N-phenylacrylamide), which shows lower critical solution temperature (LCST) behavior. When cooling gelatin aqueous solution in which osmotic pressure was not operated, only hydration of the copolymer progressed due to LCST transition. The optical path length of the SCB increased due to swelling by water and subsequently decreased due to dissolution. The structural color changed according to the change in optical path length. In cold pure water, in addition to the hydration, osmotic pressure was operated to induce an influx of the outer solvent and the resulting diameter change also affected the shell thickness. The structural color change was analyzed to reveal that the dissolution of the polymer had significant effect on the developed structural color.

Folate-Conjugated and Dual Stimuli-Responsive Mixed Micelles Loading Indocyanine Green for Photothermal and Photodynamic Therapy.

A folic acid targeted mixed micelle system based on co-assembly of poly(ε-caprolactone)-b-poly(methoxytri(ethylene glycol) methacrylate-co-N-(2-methacrylamido)ethyl folatic amide) and poly(ε-caprolactone)-b-poly(diethylene glycol monomethyl ether methacrylate) is developed to encapsulate indocyanine green (ICG) for photothermal therapy and photodynamic therapy. In this study, the use of folic acid is not only for specific cancer cell recognition, but also in virtue of the carboxylic acid on folic acid to regulate the pH-dependent thermal phase transition of polymeric micelles for controlled drug release. The prepared ICG-loaded mixed micelles possess several superior properties such as a preferable thermoresponsive behavior, excellent storage stability, and good local hyperthermia and reactive oxygen species generation under near-infrared (NIR) irradiation. The photototoxicity induced by the ICG-loaded micelles has efficiently suppressed the growth of HeLa cells (folate receptor positive cells) under NIR irradiation compared to that of HT-29, which has low folate receptor expression. Hence, this new type of mixed micelles with excellent features could be a promising delivery system for controlled drug release, effective cancer cell targeting, and photoactivated therapy.

Synthesis and self-assembly of double-hydrophilic pentablock copolymer with pH and temperature responses via sequential atom transfer radical polymerization

ABCBA pentablock double-hydrophilic copolymer of poly(diethylene glycol monomethyl ether methacrylate)- block-poly(2-( N,N-dimethylamino)ethyl methacrylate)- block-poly(ethylene glycol)- block-poly(2-( N,N-dimethylamino)ethyl methacrylate)- block-poly(diethylene glycol monomethyl ether methacrylate) (PMEO 2MA- b-PDMAEMA- b-PEG- b-PDMAEMA- b-PMEO 2MA) was synthesized by sequential atom transfer radical polymerization (ATRP). The copolymer is hydrophilic at low temperature and pH values (at temperatures lower than 26 °C and acidic pH values), and became amphiphilic when pH and (or) temperature changed. Investigation shows that the copolymer can self-assembly into micelles with various structures under the conditions of temperature higher than 26 °C or basic solutions. The self-assembly behavior and multi-responsive properties of the copolymer in water were investigated by DLS and TEM. The self-assembly behavior of the copolymer can be adjusted by the alteration of pH values or temperature. Therefore, the copolymer will have the potential applications in biomedical field.

Fluorescent water-soluble responsive polymers site-specifically labeled with FRET dyes possessing pH- and thermo-modulated multicolor fluorescence emissions as dual ratiometric probes

We reported on the synthesis of well-defined thermoresponsive polymers labeled with fluorescence resonance energy transfer (FRET) pairs at chain middle and terminals, which can act as single chain-based dual ratiometric fluorescent probes for pH and temperature under extremely dilute conditions. Starting from difunctional initiator containing a 7-nitro-2,1,3-benzoxadiazole (NBD) moiety, the atom transfer radical polymerization (ATRP) of oligo(ethylene glycol) monomethyl ether methacrylate (OEGMA) and di(ethylene glycol) monomethyl ether methacrylate (DEGMA), and the subsequent terminal group functionalization with Rhodamine B (RhB)-ethylenediamine derivative afforded thermoresponsive NBD-P(OEGMA-co-DEGMA)-RhB2, which were labeled with FRET donor (NBD) and acceptor moieties (RhB) at the chain middle and terminals. The fluorescence emission of terminal RhB functionalities is highly pH-dependent, i.e, non-fluorescent in neutral or alkaline media (spirolactam form) and highly fluorescent in acidic media (ring-opened acyclic form), thus the off/on switching of FRET process can be facilely modulated by solution pH. Moreover, at acidic pH and highly dilute conditions, the thermo-induced chain collapse and extension of NBD-P(OEGMA-co-DEGMA)-RhB2 can effectively modulate the spatial distance between FRET donor and acceptor moieties, leading to prominent changes in FRET efficiencies. The site-specific incorporation of one FRET donor and two pH-switchable acceptors at the chain middle and terminals of thermoresponsive polymers allows for the effective off/on switching and the modulation of efficiency of FRET processes by dually playing with solution pH and temperatures. This work represents the first report of single thermoresponsive polymer chains acting as dual ratiometric fluorescent probes under highly dilute conditions.

Synthesis of Hetero-Telechelic α,ω Bio-Functionalized Polymers

Reversible addition-fragmentation chain transfer (RAFT) polymerization was used to synthesize poly[diethylene glycol monomethylether methacrylate] (PDEGMA) (M(n) = 6250 g/mol, PDI = 1.14) with a pentafluorophenyl (PFP) activated ester and a dithioester end group. The hormone thyroxin (T4) was quantitatively attached to the PFP activated ester alpha end group via its amino group. The omega-terminal dithioester was not harmed by this reaction and was subsequently aminolyzed in the presence of N-biotinylaminoethyl methanethiosulfonate, yielding a polymer with a thyroxin and a biotin end group with very high heterotelechelic functionality. The polymer was characterized by (1)H, (13)C, and (19)F NMR, UV-vis, and IR spectroscopy and gel permeation chromatography. The thyroxin transport protein prealbumin with two thyroxin binding sites and streptavidin, which has four biotin binding sites, was conjugated using the biotarget labeled polymer, resulting in the formation of a protein-polymer network, confirming the heterotelechelic nature of the polymer. Polymer-protein microgel formation was observed with dynamic light scattering. To realize a directed protein assembly, prealbumin was immobilized onto a surface, exposing one of its two thyroxin binding groups and thus allowing the conjugation with the thyroxin alpha end group of the heterotelechelic polymer. The biotin omega end group of the attached polymer layer enabled the subsequent immobilization of streptavidin, yielding a defined multilayer system of two proteins connected with the synthetic polymer (efficiency of streptavidin immobilization 81% based on prealbumin). Without the polymer, no streptavidin immobilization occurred. The layer depositions were monitored by surface plasmon resonance. The synthetic approach of combining PFP activated esters with functional MTS reagents presents a powerful method for obtaining well-defined heterotelechelic (bio-) functionalized polymers.

Synthesis of Reactive Telechelic Polymers Based on Pentafluorophenyl Esters

A diazo initiator and a chain transfer agent (CTA), both containing a pentafluorophenyl (PFP) activated ester, were synthesized. In a RAFT polymerization using the functionalized chain transfer agent (PFP-CTA), methyl methacrylate (MMA), diethylene glycol monomethyl ether methacrylate (DEGMA), poly(ethylene glycol) monomethyl ether methacrylate (PEGMA), and lauryl methacrylate (LMA) could successfully be polymerized into homopolymers and diblock copolymers with good control over molecular weight, very high conversions, and narrow molecular weight distributions. Polymers derived from the PFP−CTA possessed an activated ester at the α-end of the polymer chain, which could be reacted with amines with high conversions. The terminal ω-dithioester group of each polymer chain could quantitatively be removed by treating the polymer with an excess of AIBN, leaving the α-PFP ester functionality intact. Accordingly, the pentafluorophenyl ester diazo compound could successfully be employed to functionalize RAFT polymers with a PFP ester at their ω-end. As a consequence, functionalization of both end groups was possible and led to telechelic polymers, exhibiting an active ester at both ends of the polymer chain. As an example, a high molecular weight PMMA was prepared by polycondensation with ethylenediamine.

Preparation of Cholesteric (Hydroxypropyl)cellulose/Polymer Networks and Ion-Mediated Control of Their Optical Properties

(Hydroxypropyl)cellulose (HPC)/vinyl polymer networks were synthesized in film form from liquid-crystalline solutions of HPC in a mixed solvent of methacrylate monomer/methanol/water (2:1:2 in weight) containing cross-linking agents, via photopolymerization of the methacrylate monomer. Di(ethylene glycol) monomethyl ether methacrylate (DEGMEM) or 2-hydroxypropyl methacrylate (HPMA) was used as the polymerizing monomer, and tetra(ethylene glycol) diacrylate and glutaraldehyde were the cross-linkers for the monomers and HPC, respectively. The polymer composite films, HPC/PDEGMEM and HPC/PHPMA, prepared at ca. 60-70 wt % concentrations of HPC in the starting solutions, were iridescently colored due to the selective light reflection, originating from the cholesteric helical arrangement carried over successively into the network system. When the cholesteric films were immersed and swollen in water containing an inorganic neutral salt, their coloration and optical turbidity varied according to a strength of 'chaotropicity' of the impregnant ions. This ionic effect may be interpreted as essentially identical with that found formerly in the coexistent salt-sort dependence of the cholesteric pitch and lower critical solution temperature for HPC aqueous solutions. It is also demonstrated that visual appearance of the swollen networks can be changed by application of an electric potential of practical magnitude between both edges of the samples of rectangular shape.
