Strategies for Bovine Respiratory Disease (BRD) Diagnosis and Prognosis: A Comprehensive Overview.

Despite significant advances in vaccination strategies and antibiotic therapy, bovine respiratory disease (BRD) continues to be the leading disease affecting the global cattle industry. The etiology of BRD is complex, often involving multiple microbial agents, which lead to intricate interactions between the host immune system and pathogens during various beef production stages. These interactions present environmental, social, and geographical challenges. Accurate diagnosis is essential for effective disease management. Nevertheless, correct identification of BRD cases remains a daunting challenge for animal health technicians in feedlots. In response to current regulations, there is a growing interest in refining clinical diagnoses of BRD to curb the overuse of antimicrobials. This shift marks a pivotal first step toward establishing a structured diagnostic framework for this disease. This review article provides an update on recent developments and future perspectives in clinical diagnostics and prognostic techniques for BRD, assessing their benefits and limitations. The methods discussed include the evaluation of clinical signs and animal behavior, biomarker analysis, molecular diagnostics, ultrasound imaging, and prognostic modeling. While some techniques show promise as standalone diagnostics, it is likely that a multifaceted approach-leveraging a combination of these methods-will yield the most accurate diagnosis of BRD.

Comparative diagnoses of respiratory disease in preweaned dairy calves using sequential thoracic ultrasonography and clinical respiratory scoring.

Bovine respiratory disease (BRD) has serious impacts on dairy production and animal welfare. It is most commonly diagnosed based on clinical respiratory signs (CRS), but in recent years, thoracic ultrasonography (TUS) has emerged as a diagnostic tool with improved sensitivity and specificity. This study aimed to assess the alignment of BRD diagnoses based on a Clinical Respiratory Scoring Chart (CRSC) and weekly TUS findings throughout the progression of BRD of variable severity in preweaned Holstein dairy heifers. A total of 60 calves on two farms were followed from the 2nd week of life through the 11th week of life and assessed on a weekly basis for CRS and lung consolidation via TUS. The alignment of BRD diagnoses based on CRSC scores and TUS findings was evaluated across disease progression (pre-consolidation, onset, chronic, or recovered) and severity (lobular or lobar lung consolidation) using receiver operator curves and area under the curves combined with Cohen's kappa (κ), sensitivity, and specificity. The diagnosis of BRD using CRSC scores ≥5 aligned best with the onset of lobar lung consolidation (>1 cm in width and full thickness). This equated to an acceptable level of discrimination (AUC = 0.76), fair agreement (κ = 0.37), and a sensitivity of 29% and specificity of 99%. Similarly, there was acceptable discrimination (AUC = 0.70) and fair agreement (κ = 0.33) between CRSC ≥5 and the onset of a less severe threshold of disease based on lobular (1-3 cm2 but not full thickness) or lobar consolidation. Discrimination remained acceptable (AUC = 0.71) with fair agreement (κ = 0.28) between CRSC scores ≥2 for nasal discharge and/or cough (spontaneous or induced) and the onset of lobar consolidation. However, sensitivity was <40% across comparisons and outside of the onset of disease there tended to be poor discrimination, slight agreement, and lowered sensitivity between CRS and TUS diagnoses of lobular or lobar consolidation (pre-consolidation, chronic, or recovered). Conversely, specificity was relatively high (≥92%) across comparisons suggesting that CRSC diagnoses indicative of BRD and associated lung consolidation tend to result in few false positive diagnoses and accurate identification of healthy animals. Although we found the specificity of clinical signs for diagnosing lung consolidation to be ≥92% across all methods of TUS evaluations, the low levels of sensitivity dictate that clinical assessments lead to many false negative diagnoses. Consequently, depending on clinical signs alone to diagnose BRD within populations of dairy calves will likely result in overlooking a substantial proportion of subclinically affected animals that could inform the success of treatment and prevention protocols and guide management decisions.

Short communication: Circadian variations and day-to-day variability of clinical signs used for the early diagnosis of pneumonia within and between calves

For rational antimicrobial use, a timely and correct diagnosis of bovine respiratory disease, especially pneumonia, in calves is required. Current approaches often rely on clinical signs observed at a single time point, and do not take potential diurnal patterns in the manifestation of these clinical signs into account. Therefore, the aim of this pilot study was to investigate how clinical signs utilized for the (early) detection of pneumonia vary both within and between calves, throughout the day and across days. A longitudinal study was conducted in which 36 pre-weaned Holstein-Friesian calves were clinically examined eight times over the course of 48 h. The following parameters were considered: respiratory rate, type of respiration, dyspnea, stridor, induced cough (trachea reflex), spontaneous cough, eye and nasal discharge, ear positions, head tilt, rectal temperature, diarrhea, milk residue, body posture, Wisconsin and Davis BRD scorecard. The advent of thoracic ultrasonography (TUS) enables detection of (sub)clinical pneumonia in a more reliable way, compared to the diagnosis based solely on clinical signs. In this study, 14% (5/36) of the calves had an ultrasound confirmed pneumonia (consolidation ≥1 cm in depth). No variations were observed in the prevalence of clinical signs at the various time points of the day. However, we did observe a difference in the manifestation of clinical signs in individual calves (intra) and between (inter) them. Due to the significant intra-calf variability, diagnosing pneumonia based solely on a single observation of clinical signs, is likely to be insufficient. Hence, misdiagnosis might lead to incorrect use of antimicrobials.

Differential gene expression in peripheral leukocytes of pre-weaned dairy calves with respiratory disease

Bovine respiratory disease (BRD) is a leading cause of calf morbidity and mortality in both beef and dairy sectors of the United States cattle industry resulting in financial losses and compromised welfare. Differential gene expression (DGE) al­lows detailed insight into individual immune responses and can identify enriched pathways and biomarkers that contrib­ute to disease susceptibility and outcome. Differential expres­sion of genes related to innate immune function has been well documented in both feedlot and post-weaned dairy cattle with experimentally induced or naturally occurring BRD. However, little is known regarding DGE in pre-weaned Holstein calves. Therefore, the aims of this study were to evaluate gene expres­sion in peripheral leukocytes relative to BRD diagnosis, dis­ease progression and age progression in Holstein heifer calves throughout the preweaning period.

Treatment with 2 commercial antibiotics reduced clinical and systemic signs of pneumonia and the abundance of pathogenic bacteria in the upper respiratory tract of preweaning dairy calves

The aim of this study was to evaluate the effect of therapeutically administered tildipirosin or florfenicol + flunixin meglumine for the treatment of bovine respiratory disease (BRD) accompanied by fever in calves before weaning compared with diseased and untreated animals. As specific objectives, we evaluated the composition of the bacterial microbiota of the upper respiratory tract (URT) and blood and health parameters of the animals. Preweaning Holstein female calves diagnosed with naturally acquired pneumonia were randomly assigned to one of the following experimental groups on the day of diagnosis (d 0): (1) TLD (n = 36): single subcutaneous injection with 4 mg/kg tildipirosin; (2) FLF (n = 33): single subcutaneous injection with an antimicrobial (40 mg/kg florfenicol) combined with a nonsteroidal anti-inflammatory drug (2.2 mg/kg flunixin meglumine); and (3) NEG (n = 35): no treatment within the first 5 d following enrollment. The NEG treatment group was closely monitored for 5 d, and calves were removed from the study following a standardized late treatment protocol, when necessary, to minimize health concerns. Healthy untreated calves (CTR; n = 31) were also selected for the study and used as controls. Blood samples used for biochemical analysis and nasopharyngeal swabs used for evaluation of URT microbiota were collected daily from d 0 until d 5 and then weekly until weaning. Next-generation sequencing of the 16S rRNA gene was used to assess the URT microbiota at the phylum and genus levels. Clinical signs associated with pneumonia and otitis media were assessed daily, as was the need for antibiotic interventions. Calves in the TLD and FLF groups had faster recovery from fever within the first 5 d after enrollment. In addition, antibiotic-treated calves reached the same serum haptoglobin levels as healthy calves on d 2 after diagnosis, whereas calves in the NEG group had higher haptoglobin levels than the CTR group until at least d 5 after BRD diagnosis. Calves in the TLD and FLF groups had a lower risk of treatment for pneumonia (FLF = 22.8%; TLD = 27.7%) from d 5 to weaning than calves in the NEG group (54.7%). Furthermore, FLF treatment had a significantly lower risk of nasal discharge, otitis media, and treatment failure compared with the NEG group, but did not differ from the TLD group. Differences in the composition of the URT microbiota were found between groups, and the genus Mycoplasma was the most abundant in samples collected from the URT of calves with and without pneumonia. Both drugs were effective in reducing the mean relative abundance (MRA) of important genera associated with pneumonia (Mannheimia and Pasteurella), although an increase in Mycoplasma MRA was observed for tildipirosin-treated calves. In conclusion, both drugs were effective in reducing the inflammatory signs of pneumonia and the need for antimicrobial treatment after enrollment compared with no treatment. In addition, both TLD and FLF were effective in reducing the MRA of important bacterial genera associated with pneumonia; however, TLD treatment was associated with increased Mycoplasma MRA compared with healthy and untreated calves.

Cross-Section Observational Study to Assess Antimicrobial Resistance Prevalence among Bovine Respiratory Disease Bacterial Isolates from Commercial US Feedlots.

Antimicrobial resistance (AMR) is a global public health threat that jeopardizes efficacy of antibiotics in veterinary and human medicine. Antibiotics are commonly administered to target the bacterial component of bovine respiratory disease (BRD). The objectives of this study were to obtain a better understanding of antibiotic resistance in BRD-associated bacteria (Mannheimia haemolytica, Pasteurella multocida, and Histophilus somni), investigate the clinical significance of AMR by monitoring clinical outcomes, and determine if regional differences exist in AMR trends. Deep pharyngeal swabs were used to sample beef cattle at initial BRD diagnosis (n = 453) from US feedlots representing three geographic regions. Organisms were identified by bacterial culture and subjected to broth microdilution antimicrobial susceptibility testing. Bacterium prevalence include P. multocida (36.0%), M. haemolytica (32.7%), and H. somni (28.5%). Of the Histophilus isolates, 39.5% were resistant to at least one antimicrobial, compared to 11.7% and 8.8% Pasteurella and Mannheimia, respectively. Non-susceptibility across all organisms was 5.7 X more likely in animals that received metaphylaxis, than those that did not (p < 0.0001; OR 5.7; CI 2.6-12.5). During days on feed 21-40, non-susceptibility of Histophilus was 8.7 X more likely than Mannheimia (p = 0.0002; OR 8.7; CI 2.8 to 27.4) and 6 X more likely than Pasteurella (p = 0.0016; OR 6.0; CI 2.0-18.0).

Technological Tools for the Early Detection of Bovine Respiratory Disease in Farms

Simple SummaryThe inclusion of remote automatic systems that use continuous learning technology are of great interest in precision livestock cattle farming, since the average size of farms is increasing while time for individual observation is decreasing. Bovine respiratory disease is a main concern in both fattening and heifer rearing farms due to its impact on antibiotic use, loss of performance, mortality, and animal welfare. Much scientific literature has been published regarding technologies for continuous learning and monitoring of cattle’s behavior and accurate correlation with health status, including early detection of bovine respiratory disease. This review summarizes the up-to-date technologies for early diagnosis of bovine respiratory disease and discusses their advantages and disadvantages under practical conditions.Classically, the diagnosis of respiratory disease in cattle has been based on observation of clinical signs and the behavior of the animals, but this technique can be subjective, time-consuming and labor intensive. It also requires proper training of staff and lacks sensitivity (Se) and specificity (Sp). Furthermore, respiratory disease is diagnosed too late, when the animal already has severe lesions. A total of 104 papers were included in this review. The use of new advanced technologies that allow early diagnosis of diseases using real-time data analysis may be the future of cattle farms. These technologies allow continuous, remote, and objective assessment of animal behavior and diagnosis of bovine respiratory disease with improved Se and Sp. The most commonly used behavioral variables are eating behavior and physical activity. Diagnosis of bovine respiratory disease may experience a significant change with the help of big data combined with machine learning, and may even integrate metabolomics as disease markers. Advanced technologies should not be a substitute for practitioners, farmers or technicians, but could help achieve a much more accurate and earlier diagnosis of respiratory disease and, therefore, reduce the use of antibiotics, increase animal welfare and sustainability of livestock farms. This review aims to familiarize practitioners and farmers with the advantages and disadvantages of the advanced technological diagnostic tools for bovine respiratory disease and introduce recent clinical applications.

Ultrasonographic diagnosis of clinical and subclinical bovine respiratory disease in Holstein calves.

Background and Aim:Bovine respiratory disease (BRD) is the main cause of death in calves, and early BRD diagnosis saves lives. This study aimed to diagnose clinical and subclinical BRD in calves by assessing some biochemical alterations and ultrasonography (USG).Materials and Methods:Fifty-four Holstein dairy calves in Al-Sharqiyah Province, Egypt, were used in the study. They were divided into three groups. The first control group consisted of 10 clinically healthy calves. The second group consisted of 34 calves suffering from clinical lower respiratory tract disorders. The third group consisted of 10 subclinical BRD-affected calves. Ultrasonographic examinations of chest and thoracic ultrasound scoring were performed once per 2 weeks for each calf. Blood samples were collected for serum separation to measure albumin (ALB), total protein (TP), ALB, globulin, and haptoglobin (HP).Results:The USG revealed small consolidation areas within an aerated lung lobe, a hypoechoic parenchyma of the entire distal lung lobe, and a hypoechoic-circumscribed structure surrounded by an echogenic wall appeared within the lung tissue in calves that suffered from lobular pneumonia, lobar pneumonia, and lung abscess, respectively. However, subclinical cases showed a small consolidation area in the cranial aspects of the right cranial lung lobe. The ultrasound lung score (ULS) was greater in clinical than in subclinical cases. The BRD-affected calves recorded significant increases in serum TP, globulin, and HP. Meanwhile, serum ALB decreased significantly.Conclusion:Thoracic ultrasound had a reliable tool in the BRD diagnosis, especially in the early prediction of subclinical cases in newborn calves. In addition, the ULS appeared to be a better classifier than the clinical respiratory score (CRS) for BRD diagnosis. On the other side, it was found that regression models were very useful in assessing the prediction of biochemical blood parameters based on the ULS and CRS in diseased cases.

Risk factors for mid- and late-feeding-stage bovine respiratory morbidity and mortality based on individual animal treatments of beef feedlot cattle

Previously published research on bovine respiratory disease (BRD) has emphasized the initial portion of the feeding phase; yet, little work has investigated risk factors associated with BRD incidence later in the feeding period. Our study objective was to identify risk factors associated with BRD morbidity and mortality timing within the feeding phase. Individual animal records for first BRD treatment (n = 188,437) or BRD mortality (n = 13,991) were classified as early stage, middle stage, or late stage based on the percentage of their cohorts’ feeding phase that was complete at day of event (event days on feed/cohort total days on feed). There were 141,097 early stage, 33,871 middle stage, and 13,469 late stage at first treatments for BRD and 7,821 early stage, 3,625 middle stage, and 2,545 late stage at death. Two ordinal logistic regression models (morbidity, mortality) were used to evaluate the association of cattle demographic factors (arrival weight, arrival lot size, sex, metaphylaxis, quarter of arrival) and the probability of an individual animal first treatment for BRD or mortality from BRD being early, middle, or late stage. All 2-way interactions of cattle demographics for morbidity were significant (P < 0.05), whereas for mortality, main effects of arrival weight and metaphylaxis were significant (P < 0.05) and only the 2-way interactions of sex by quarter of arrival and sex by lot size were significant (P < 0.05). In general, heifers, heavier animals at arrival, and cattle that arrived at the yard in the first and second quarter were the most likely demographic categories to have an initial diagnosis of BRD in the mid or late stage of the feeding phase.

Feeding behavior and activity levels are associated with recovery status in dairy calves treated with antimicrobials for Bovine Respiratory Disease

Calves with Bovine Respiratory Disease (BRD) have different feeding behavior and activity levels prior to BRD diagnosis when compared to healthy calves, but it is unknown if calves who relapse from their initial BRD diagnosis are behaviorally different from calves who recover. Using precision technologies, we aimed to identify associations of feeding behavior and activity with recovery status in dairy calves (recovered or relapsed) over the 10 days after first antimicrobial treatment for BRD. Dairy calves were health scored daily for a BRD bout (using a standard respiratory scoring system and lung ultrasonography) and received antimicrobial therapy (enrofloxacin) on day 0 of initial BRD diagnosis; 10–14 days later, recovery status was scored as either recovered or relapsed (n = 19 each). Feeding behaviors and activity were monitored using automated feeders and pedometers. Over the 10 days post-treatment, recovered calves showed improvements in starter intake and were generally more active, while relapsed calves showed sickness behaviors, including depressed feed intake, and longer lying times. These results suggest there is a new potential for precision technology devices on farms in evaluating recovery status of dairy calves that are recently treated for BRD; there is opportunity to automatically identify relapsing calves before re-emergence of clinical disease.

Daily behavioral measures recorded by precision technology devices may indicate bovine respiratory disease status in preweaned dairy calves

Precision technology devices can measure and detect relative changes in an animal's behavior to possibly create alerts to intervene and to administer treatments. However, the association of relative changes in daily feeding and activity behaviors in calves with bovine respiratory disease (BRD) status is still largely unexplored. The objective of this case-control study was to determine if daily behavioral patterns of preweaning dairy calves (measured by precision technologies) change before BRD diagnosis. This case-control study enrolled 33 pairs of calves (33 BRD calves matched by age, sex, and birthdate to 33 controls) health scored daily for BRD for the preweaning period (until 50 d on the automated feeder). A pedometer (IceQube, Ice Robotics) was attached to the left rear leg to track activity (lying time, lying bouts, total steps, and acceleration activity index). At 3.0 ± 2.0 d of age, calves were trained to use the automated feeder, which recorded milk and calf starter intake, drinking speed, and feeder visits. Calves were allotted a maximum 10 L/d of milk replacer (Cow's Match, Land O'Lakes Animal Milk Products Co.; 140 g/L) and starter (Special Calf Starter and Grower, Baghdad Feeds), ad libitum. Calves were scored daily for signs of BRD using the Wisconsin scoring system, and their lungs were scored with ultrasonography twice weekly. Outward signs of BRD as defined by the Wisconsin scoring system and an area of consolidated lung ≥3.0 cm2 was identified as BRD (d 0). Relative changes in daily behaviors were calculated using d -5 before BRD diagnosis as the baseline for each calf for each behavior. Linear mixed models were used to investigate the association of BRD status with feeding behaviors, activity, and relative changes in calf behavior over a 5-d period before diagnosis, as well as the BRD status × day interaction. Calves with a BRD bout were diagnosed at an average age of 33.0 ± 9.0 d (mean ± standard deviation) and weighed 56.1 ± 9.7 kg. Over the period, BRD calves had reduced milk and starter intake, greater lying times, and fewer lying bouts, step counts, and activity indices when compared with healthy calves. Furthermore, there was a BRD status × day interaction for relative changes in unrewarded visits, and relative changes in calf starter intake. Specifically, BRD calves had a decline in relative changes in their unrewarded visits on d -4, -2, -1, and 0 compared with healthy calves. This study suggests that there is the potential to use feeding and activity behaviors to identify BRD development in preweaning calves. However, utilization of an animals' daily behavioral patterns in real time is fundamental for developing disease detection algorithms, thus we suggest relative changes in unrewarded visits may be useful for algorithm development when d -5 is used as a baseline. Future research should investigate the potential of feeding behavior and activity levels collectively to indicate BRD status in calves using machine learning techniques.

Galyean Appreciation Club Review: revisiting nutrition and health of newly received cattle-what have we learned in the last 15 years?

Our objective was to review the literature related to the health and management of newly received cattle published since a previous review by Duff and Galyean (2007). Bovine respiratory disease (BRD) continues to be a major challenge for the beef industry. Depending on disease severity, animals treated for BRD have decreased performance and lowered carcass value. Diagnosis of BRD is less effective than desired, and progress on developing real-time, chute-side methods to diagnose BRD has been limited. Systems that combine lung auscultation with temperature and body weight data show promise. Assessment of blood metabolites and behavior monitoring offer potential for early identification of morbid animals. Vaccination and metaphylaxis continue to be important tools for the prevention and control of BRD, but antimicrobial resistance is a concern with antibiotic use. Dietary energy concentration and roughage source and level continue to be important topics. Mineral supplementation has received considerable attention, particularly the use of organic vs. inorganic sources and injectable minerals or drenches given on arrival. The use of probiotics and prebiotics for newly received cattle has shown variable results, but further research is warranted. The health and nutrition of newly received cattle will continue to be an important research area in the years to come.

Use of nCounter mRNA profiling to identify at-arrival gene expression patterns for predicting bovine respiratory disease in beef cattle

BackgroundTranscriptomics has identified at-arrival differentially expressed genes associated with bovine respiratory disease (BRD) development; however, their use as prediction molecules necessitates further evaluation. Therefore, we aimed to selectively analyze and corroborate at-arrival mRNA expression from multiple independent populations of beef cattle. In a nested case-control study, we evaluated the expression of 56 mRNA molecules from at-arrival blood samples of 234 cattle across seven populations via NanoString nCounter gene expression profiling. Analysis of mRNA was performed with nSolver Advanced Analysis software (p < 0.05), comparing cattle groups based on the diagnosis of clinical BRD within 28 days of facility arrival (n = 115 Healthy; n = 119 BRD); BRD was further stratified for severity based on frequency of treatment and/or mortality (Treated_1, n = 89; Treated_2+, n = 30). Gene expression homogeneity of variance, receiver operator characteristic (ROC) curve, and decision tree analyses were performed between severity cohorts.ResultsIncreased expression of mRNAs involved in specialized pro-resolving mediator synthesis (ALOX15, HPGD), leukocyte differentiation (LOC100297044, GCSAML, KLF17), and antimicrobial peptide production (CATHL3, GZMB, LTF) were identified in Healthy cattle. BRD cattle possessed increased expression of CFB, and mRNA related to granulocytic processes (DSG1, LRG1, MCF2L) and type-I interferon activity (HERC6, IFI6, ISG15, MX1). Healthy and Treated_1 cattle were similar in terms of gene expression, while Treated_2+ cattle were the most distinct. ROC cutoffs were used to generate an at-arrival treatment decision tree, which classified 90% of Treated_2+ individuals.ConclusionsIncreased expression of complement factor B, pro-inflammatory, and type I interferon-associated mRNA hallmark the at-arrival expression patterns of cattle that develop severe clinical BRD. Here, we corroborate at-arrival mRNA markers identified in previous transcriptome studies and generate a prediction model to be evaluated in future studies. Further research is necessary to evaluate these expression patterns in a prospective manner.

Pulmonary Ultrasound in the Diagnosis of Respiratory Disease in Dairy Calves - Systematic Review / Ultrassonografia Pulmonar no Diagnóstico de Doença Respiratória em Bezerras Leiteiras- Revisão Sistemática

Bovine respiratory disease (BRD) is a common health challenge for dairy calves during the first weeks of life. It can be caused by several environmental factors and pathogens, such as bacteria and viruses, presenting alterations called pulmonary consolidations. BRD shows nonspecific clinical signs, a high rate of subclinical cases and triggers several health consequences, such as impaired growth, reduced weight gain, interference in production and reproduction during the first lactation, and in some cases, animals can also die. In addition economic losses such as: increase in the rate of involuntary culling and reduction of the source of replacement of productive cows in the herd can also occur. There are some forms of diagnosis evaluating clinical signs such as fever and discharges. However, these methods are not sufficient to confirm the clinical diagnosis and do not allow the measurement of the degree of pulmonary lesions. However, the disease can also be diagnosed by pulmonary ultrasound, presenting a high rate of accuracy. This study reviews systematically the main studies that performed pulmonary ultrasound evaluation in the diagnosis of BRD and reported results that showed the efficacy of the use of this method in the clinical diagnosis of the disease. The technique is considered noninvasive, fast, capacity of detecting animals with subclinical BRD and uses the same ultrasound device used in the reproductive management of cows. In addition, ultrasonography has other benefits such as cost reduction, reduced use of antimicrobials, evaluation of the extent of lung lesions, positive influence on animal health and support of profitability of a dairy operation.

Treatment of bovine respiratory disease with a single administration of tulathromycin and ketoprofen.

The therapeutic strategy of bovine respiratory disease (BRD) often involves a combination of an antibiotic with an anti-inflammatory agent. Aim of this study was to evaluate the clinical effect of a new combination product containing tulathromycin and ketoprofen for the treatment of naturally occurring BRD. Two hundred and eighty animals were randomized upon diagnosis of BRD. One hundred forty animals each were treated once subcutaneously with tulathromycin-ketoprofen or tulathromycin. Rectal temperature of each animal was measured at 1, 2, 4, 6, 8, 10, 12 and 24 h post-treatment. Individual respiration and depression scores were determined at 6 h post-treatment. Daily rectal temperature, respiration and depression scores were recorded from day 2 to 14 and on day 21. The tulathromycin-ketoprofen and tulathromycin treatment group demonstrated a treatment success rate of 94.2% and 95.0%, respectively and a relapse rate of 3.8% and 4.0%, respectively. Tulathromycin-ketoprofen demonstrated superior pyrexia control compared to tulathromycin within the first 24 h following treatment. Tulathromycin-ketoprofen-treated animals demonstrated faster improvement of their clinical symptoms (respiration and depression score). Efficacy of tulathromycin-ketoprofen for the treatment of BRD was non-inferior to tulathromycin. The combination product clearly exhibited more pronounced fever control than tulathromycin which is considered beneficial for animal welfare.

Infectious Bovine Respiratory Diseases in Adult Cattle: An Extensive Necropsic and Etiological Study.

Simple SummaryAnimal necropsy can accurately determine the cause of its death. However, studies based on large-scale necropsies of cattle are rare because they require special skills and equipment. This study places particular emphasis on fatal respiratory diseases in adult cattle. The objectives of this study were to assess the importance of respiratory diseases as a cause of death in adult cattle and to determine associated lesions as well as associated pathogens of infectious causes of respiratory diseases in adult cattle. This study showed that respiratory diseases are the second leading cause of death in adult cattle after digestive diseases. Among respiratory diseases, we noticed a strong predominance of infectious pulmonary lesions, mainly characterized by fibrinous, hemorrhagic and/or necrotic bronchopneumonia. These bronchopneumonia are mainly associated with the detection of a bacteria, Mannheimia haemolytica. This study suggests that Mannheimia haemolytica should be included in the differential diagnosis of BRD in adult cattle.In young cattle, bovine respiratory disease (BRD) is a major cause of death and Mannheimia haemolytica is a frequent pathogen. Knowledge of fatal BRD in adult cattle is more limited. We assessed the importance of infectious BRD as a cause of death in adult cattle and determined the associated pathogens. We analyzed data from 737 adult cattle necropsies at the Pathology Unit for Large Animals at Oniris, Nantes, France over a 6 year period (2013–2019). Each carcass was subjected to a complete necropsy. Lungs showing macroscopic lesions were classified into three categories: infectious primary pulmonary (IPP) lesions, thromboembolic pneumonia (TEP) and others (aspiration pneumonia, verminous pneumonia, and local extension of an extra-pulmonary inflammatory process). Half of the lungs with IPP macroscopic lesions were sampled for histology and submitted for polymerase chain reaction. BRD was the second leading cause of death (15.7%) after digestive diseases (32.2%). A strong predominance of IPP lesions (42.3%) and TEP lesions (39.6%) was also demonstrated. In IPP macroscopic lesions, fibrinous, hemorrhagic and/or hecrotic (FHN) bronchopneumonia accounted for 77.6% of macroscopic lesions. Mannheimia haemolytica was significantly associated with FHN bronchopneumonia macroscopic lesions. This study suggests that Mannheimia haemolytica should be included in the differential diagnosis of BRD in adult cattle.

Clinical and Ultrasonographic Studies in Bovine Respiratory Disease (BRD) Affected Calves

Bovine Respiratory Diseases (BRD) are one of the most common and important disease problem affecting newborn calves. The aim of this study was to use clinical respiratory score and thoracic ultrasonography as a screening method for the diagnosis of BRD in newborn calves. This study was carried out on 59 newborn calves in the Al-salhia farm for investment in Alsharqia Governorate, Egypt. These calves were divided into two groups. The first group (G1) including 16 healthy control calves and the second group (G2) including 43 newborn calves suffered from respiratory disorders. The most prominent clinical finding in BRD-affected calves was fever, nasal and ocular discharge, coughing with abnormal tracheal and lung sounds. According to the ultrasonographic examination of the lung, the lower respiratory tract disorders classified lesion into lobar pneumonia 12 calves (35.3%), lobular pneumonia 16 calves (47.1%), mixed pneumonia 3 calves (8.82%) in addition to lung abscess 3 calves (8.82%). Clinical respiratory score and thoracic ultrasound score showed a highly significant increase in BRD affected calves, meanwhile, CRS showed a non-significant change in calves suffered from upper and lower respiratory tract disorders. TUS revealed a non-significant change in calves with lobar and mixed pneumonia and in lung abscess, in addition to a significant increase in calves with lobar pneumonia when compared with lobular pneumonia. BRD affected calves had CRS ≥ five meanwhile calves with TUS ≥ two were considered sick.

Messenger RNA biomarkers of Bovine Respiratory Syncytial Virus infection in the whole blood of dairy calves

Bovine Respiratory Syncytial Virus (BRSV) is a primary viral cause of Bovine Respiratory Disease (BRD) in young calves, which is responsible for substantial morbidity and mortality. Infection with BRSV induces global gene expression changes in respiratory tissues. If these changes are observed in tissues which are more accessible in live animals, such as whole blood, they may be used as biomarkers for diagnosis of the disease. Therefore, the objective of the current study was to elucidate the whole blood transcriptomic response of dairy calves to an experimental challenge with BRSV. Calves (Holstein–Friesian) were either administered BRSV inoculate (103.5 TCID50/ml × 15 ml) (n = 12) or sterile phosphate buffered saline (n = 6). Clinical signs were scored daily and whole blood was collected in Tempus RNA tubes immediately prior to euthanasia, at day 7 post-challenge. RNA was extracted from blood and sequenced (150 bp paired-end). The sequence reads were aligned to the bovine reference genome (UMD3.1) and EdgeR was subsequently employed for differential gene expression analysis. Multidimensional scaling showed that samples from BRSV challenged and control calves segregated based on whole blood gene expression changes, despite the BRSV challenged calves only displaying mild clinical symptoms of the disease. There were 281 differentially expressed (DE) genes (p < 0.05, FDR < 0.1, fold change > 2) between the BRSV challenged and control calves. The top enriched KEGG pathways and gene ontology terms were associated with viral infection and included “Influenza A”, “defense response to virus”, “regulation of viral life cycle” and “innate immune response”. Highly DE genes involved in these pathways may be beneficial for the diagnosis of subclinical BRD from blood samples.

Bovine respiratory disease in Brasil: a short review

The bovine respiratory disease (BRD) complex is a multifactorial and multietiological disease entity described in all geographic regions of Brazil. This brief review discusses aspects related to epidemiology, etiologic agents, clinical and pathological manifestations, and challenges in the diagnosis of BRD in Brazil. The main infectious disease agents associated with respiratory outbreaks in cattle from Brazil are bovine alphaherpesvirus type 1, bovine viral diarrhea virus, bovine respiratory syncytial virus, and Mycoplasma bovis. Ovine gammaherpesvirus-2 and HoBi-like pestivirus have been associated with the development of pneumonia in adult cattle and calves, respectively in Brazil, and should be considered as possible causes of BRD. Additionally, studies using epidemiological data, histopathological and molecular associations with morbidity and mortality should be carried out in Brazil, to demonstrate the real impacts of BRD on livestock.
