Antibacterial property of dual-network hydrogel filled with amino functionalized graphene quantum dots

Bacterial infections seriously impair wound healing, so the development of wound dressings with high antimicrobial activity is of critical importance. In this study, amino functionalized graphene quantum dots (His-GQDs) were prepared via thermal pyrolysis of citric acid and histidine, and doped into a dual-network gelatin/sodium alginate (His-GQDs/G/SA) hydrogel to improve the antibacterial activity. The His-GQDs/G/SA hydrogel exhibited excellent antibacterial activity after 6 h of contact with bacteria. Simultaneously, the composite hydrogel could effectively alleviate the cytotoxicity of His-GQDs. The results of antimicrobial and biocompatibility tests demonstrate that the His-GQDs/G/SA dual-network hydrogel is a promising wound dressing to promote healing of infected wounds.

Gas foamed scaffolds as smart 3D structures in skin tissue engineering

Globally, wound care is a major problem that poses big issues for national healthcare systems. Consequently, the development of wound dressings, systems applied over a wound to prevent complications and speed up recovery, is receiving increasing attention. Electrospun nanofibrous scaffolds gained great interest in the medical field due to their many advantages and beneficial characteristics. However, they are formed by of densely packed fiber layers, which render the available space between adjacent fibers minimal. In this study, nanometric fibers from pullulan polymeric blends were developed using cricket powder as novel component, due to its high chitin and chitosan content. Hydroxyapatite was also added due to its biocompatibility and capability to interact with the host tissues. The 2D mats based on electropsun fibers in membrane were converted to a 3D structure by gas foaming, involving NaBH4. The obtained 3D systems were characterized for their morphology and composition, and mechanical properties. The scaffolds biocompatibility was tested on normal human dermal fibroblasts and mesenchymal stem cells, and cell adhesion was assessed. TNF-α secretion was thereafter characterized to evaluate the anti-inflammatory effects of the scaffolds on macrophages, and their antioxidant activity was also investigated. Finally, an evaluation of the scaffolds’ safety and efficacy in vivo on a murine incisional and burn model was performed. The results obtained not only revealed greater structural and mechanical integrity of the gas-foamed scaffolds containing cricket powder, but also higher cytocompatibility towards different cell lines together with anti-inflammatory and antioxidant properties, thereby making them innovative tools to improve the process of wound healing.

Electrospun zein nanofibers loaded with curcumin as a wound dressing: enhancing properties with PSS and PDADMAC layers

Development of wound dressings with enhanced therapeutic properties is of great interest in the modern healthcare. In this study, a zein-based nanofibrous wound dressing containing curcumin as a therapeutic agent was fabricated through electrospinning technique. In order to achieve desirable properties, such as antibacterial characteristics, reduced contact angle, and enhanced mechanical properties, the layer-by-layer technique was used for coating the surfaces of drug-loaded nanofibers by sequentially incorporating poly (sodium 4-styrene sulfonate) as a polyanion and poly (diallyldimethylammonium chloride) (PDADMAC) as a polycation. Various analyses, including scanning electron microscopy, Fourier transform infrared spectroscopy, drug release assessment., and mechanical tests were employed to assess the characteristics of the prepared wound dressings. Based on the results, coating with polyelectrolytes enhanced the Young’s modulus and tensile strength of the electrospun mat from 1.34 MPa and 4.21 MPa to 1.88 MPa and 8.83 MPa, respectively. The coating also improved the controlled release of curcumin and antioxidant activity, while the outer layer, PDADMAC, exhibited antibacterial properties. The cell viability tests proved the appropriate biocompatibility of the prepared wound dressings. Moreover, our findings show that incorporation of the coating layers enhances cell migration and provides a favorable surface for cell attachment. According to the findings of this study, the fabricated nanofibrous wound dressing can be considered a promising and effective therapeutic intervention for wound management, facilitating the healing process.

Development of collagenous scaffolds for wound healing: characterization and in vivo analysis.

The development of wound dressings from biomaterials has been the subject of research due to their unique structural and functional characteristics. Proteins from animal origin, such as collagen and chitosan, act as promising materials for applications in injuries and chronic wounds, functioning as a repairing agent. This study aims to evaluate in vitro effects of scaffolds with different formulations containing bioactive compounds such as collagen, chitosan, N-acetylcysteine (NAC) and ε-poly-lysine (ε-PL). We manufactured a scaffold made of a collagen hydrogel bioconjugated with chitosan by crosslinking and addition of NAC and ε-PL. Cell viability was verified by resazurin and live/dead assays and the ultrastructure of biomaterials was evaluated by SEM. Antimicrobial sensitivity was assessed by antibiogram. The healing potential of the biomaterial was evaluated in vivo, in a model of healing of excisional wounds in mice. On the 7th day after the injury, the wounds and surrounding skin were processed for evaluation of biochemical and histological parameters associated with the inflammatory process. The results showed great cell viability and increase in porosity after crosslinking while antimicrobial action was observed in scaffolds containing NAC and ε-PL. Chitosan scaffolds bioconjugated with NAC/ε-PL showed improvement in tissue healing, with reduced lesion size and reduced inflammation. It is concluded that scaffolds crosslinked with chitosan-NAC-ε-PL have the desirable characteristics for tissue repair at low cost and could be considered promising biomaterials in the practice of regenerative medicine.

In situ light‐activated materials for skin wound healing and repair: A narrative review

AbstractDermal wounds are a major global health burden made worse by common comorbidities such as diabetes and infection. Appropriate wound closure relies on a highly coordinated series of cellular events, ultimately bridging tissue gaps and regenerating normal physiological structures. Wound dressings are an important component of wound care management, providing a barrier against external insults while preserving the active reparative processes underway within the wound bed. The development of wound dressings with biomaterial constituents has become an attractive design strategy due to the varied functions intrinsic in biological polymers, such as cell instructiveness, growth factor binding, antimicrobial properties, and tissue integration. Using photosensitive agents to generate crosslinked or photopolymerized dressings in situ provides an opportunity to develop dressings rapidly within the wound bed, facilitating robust adhesion to the wound bed for greater barrier protection and adaptation to irregular wound shapes. Despite the popularity of this fabrication approach, relatively few experimental wound dressings have undergone preclinical translation into animal models, limiting the overall integrity of assessing their potential as effective wound dressings. Here, we provide an up‐to‐date narrative review of reported photoinitiator‐ and wavelength‐guided design strategies for in situ light activation of biomaterial dressings that have been evaluated in preclinical wound healing models.

Chitosan-based films filled with nanoencapsulated essential oil: Physical-chemical characterization and enhanced wound healing activity

The economic burden of chronic wounds, the complexity of the process of tissue repair and the possibility of resistant bacterial infections, have triggered a significant research interest in the application of natural alternative therapies for wound healing. Biomolecules are intrinsically multi-active, as they affect multiple mechanisms involved in tissue repair phenomenon, including immunomodulatory, anti-inflammatory, cell proliferation, extra cellular matrix remodeling and angiogenesis. Chitosan features a unique combination of attributes, including intrinsic hemostatic, antimicrobial, and immunomodulatory properties, that make it an exceptional candidate for wound management, in the development of wound dressings and scaffolds. In this study, we produced nanoemulsions (NE) loaded with SFO, characterized them, and evaluated their tissue repairing properties. Dynamic light scattering (DLS) analysis confirmed the formation of a nanoemulsion with a droplet size of 21.12 ± 2.31 nm and a polydispersity index (PdI) of 0.159, indicating good stability for up to 90 days. To investigate the potential wound healing effects, SFO-loaded NE were applied on male C57BL/6 mice for seven consecutive days, producing a significantly higher wound closure efficiency (p < 0.05) for the group treated with SFO-loaded NE compared to the control group treated with the saline solution. This finding indicates that the SFO-loaded NE exhibits therapeutic properties that effectively promote wound healing in this experimental model. Then, SFO-loaded NE were incorporated into chitosan:polyvinyl alcohol (PVA)-based films. The inclusion of NE into the polymer matrix resulted in increased lipophilicity reflected by the contact angle results, while decreasing moisture absorption, water solubility, and crystallinity. Moreover, FTIR analysis confirmed the formation of new bonds between SFO-NE and the film matrix, which also impacted on porosity properties. Thermal analysis indicated a decrease in the glass transition temperature of the films due to the presence of SFO-NE, suggesting a plasticizing role of NE, confirmed by XRD results, that showed a decrease in the crystallinity of the blend films upon the addition of SFO-NE. AFM images showed no evidence of NE droplet aggregation in the Chitosan:PVA film matrix. Moisture absorption and water content decreased upon incorporation of SFO-loaded NE. Although the inclusion of NE increased hydrophobicity and water contact angle, the values remained within an acceptable range for wound healing applications. Overall, our results emphasize the significant tissue repairing properties of SFO-loaded NE and the potential of Chitosan:PVA films containing nanoencapsulated SFO as effective formulations for wound healing with notable tissue repairing properties.

Hemostatic and antibacterial calcium-copper zeolite gauze for infected wound healing.

The design and development of wound dressings with excellent procoagulant and antibacterial activity to achieve high wound healing effectiveness are highly desirable in clinical applications. In this work, we develop a calcium-copper zeolite gauze (CaCu-ZG) by a two-step process involving calcium and copper ion exchange in a zeolite gauze. The CaCu-ZG exhibits remarkable procoagulant and antibacterial abilities, as well as good biocompatibility. Compared with the medical gauze, the blood clotting time of CaCu-ZG significantly decreases and the antibacterial activity increases in both in vivo and in vitro experiments. The remarkable ability of wound healing has been verified using a mouse dorsal skin-infected wound model, demonstrating its great potential for wound treatment in clinical applications.

Recent Advances in Biodegradable and Biocompatible Synthetic Polymers Used in Skin Wound Healing.

The treatment of skin wounds caused by trauma and pathophysiological disorders has been a growing healthcare challenge, posing a great economic burden worldwide. The use of appropriate wound dressings can help to facilitate the repair and healing rate of defective skin. Natural polymer biomaterials such as collagen and hyaluronic acid with excellent biocompatibility have been shown to promote wound healing and the restoration of skin. However, the low mechanical properties and fast degradation rate have limited their applications. Skin wound dressings based on biodegradable and biocompatible synthetic polymers can not only overcome the shortcomings of natural polymer biomaterials but also possess favorable properties for applications in the treatment of skin wounds. Herein, we listed several biodegradable and biocompatible synthetic polymers used as wound dressing materials, such as PVA, PCL, PLA, PLGA, PU, and PEO/PEG, focusing on their composition, fabrication techniques, and functions promoting wound healing. Additionally, the future development prospects of synthetic biodegradable polymer-based wound dressings are put forward. Our review aims to provide new insights for the further development of wound dressings using synthetic biodegradable polymers.

Alginate/Gauze Incorporated with Hibiscus sabdariffa Linn. Extract as a Bioactive Agent for Wound Dressing Application

In recent years, the development of wound dressings has changed from a passive to an active form by adding antiseptic agents such as polyvinylpyrrolidone (PVPI) and nanoparticles such as silver nanoparticles to facilitate the wound healing process. However, PVPI is toxic to human skin fibroblasts, and silver nanoparticles may result in metal accumulation in the tissues due to their nanosize. Therefore, a new approach to using natural sources from plants such as Hibiscus sabdariffa Linn. (HSL) is promising due to its phytochemical constituents that can serve as a potential therapeutic agent in wound dressings. In this study, freeze-dried alginate/gauze (FDA/gauze) without and with HS extract and calcium alginate/gauze (CA /gauze) with and without HS extract were developed by freeze-drying and gelation, respectively. This study aimed to characterize the properties of alginate/gauze-dressings as wound dressings. The morphology of the designed dressing; FDA/gauze, looks evenly distributed on the surface of the gauze compared to CA/gauze. It was found that FDA/gauze dressings have higher water absorbency than CA /gauze dressings, ranging from 690.43 to 1340.86% from day 1 until day 14. The percentage of water uptake ability increases as the concentration of alginate increases. In addition, the seeded human skin fibroblasts' growth on the alginate/gauze dressings infused with HSL extract growth in clustered, bright, and colourless proves that the cells were viable. Thus, the addition of HSL extract shows to support the process of cell viability.

An injectable, natural peptide hydrogel with potent antimicrobial activity and excellent wound healing-promoting effects

Wounds, especially refractory wounds infected by drug-resistant bacteria, threaten patients’ health seriously. The development of wound dressings is urgently needed to address this problem. Herein, we report a peptide hydrogel formed by natural antimicrobial peptide Jelleine-1 and its potential use for MRSA-infected wounds. We studied the formation of Jelleine-1 hydrogel, its antimicrobial activity, biodegradability, biocompatibility, and wound healing effects. Our results showed that Jelleine-1 hydrogel was formed by the physical crosslinking of self-assembled Jelleine-1 nanofibers in PBS without any other chemical crosslinkers. It exhibited broad spectrum of antimicrobial activity and excellent biocompatibility. Notably, the hydrogel showed great potential in promoting wound healing in a mouse model with MRSA-infected burn wound. Our study would provide an inherent antimicrobial hydrogel dressing without loading antibiotics for wounds infected by drug-resistant bacteria, broaden the application of antimicrobial peptides, and pave an alternative strategy for the design of antimicrobial hydrogel.

3D Matrices for Enhanced Encapsulation and Controlled Release of Anti-Inflammatory Bioactive Compounds in Wound Healing

Current trends in the development of wound dressings are oriented towards the use of biopolymer-based materials, due to their unique properties such as non-toxicity, hydrophilicity, biocompatibility and biodegradability, properties that have advantageous therapeutic characteristics. In this regard, the present study aims to develop hydrogels based on cellulose and dextran (CD) and to reveal their anti-inflammatory performance. This purpose is achieved by incorporating plant bioactive polyphenols (PFs) in CD hydrogels. The assessments include establishing the structural characteristics using attenuated total reflection Fourier transformed infrared (ATR-FTIR) spectroscopy, the morphology by scanning electron microscopy (SEM), the swelling degree of hydrogels, the PFs incorporation/release kinetics and the hydrogels' cytotoxicity, together with evaluation of the anti-inflammatory properties of PFs-loaded hydrogels. The results show that the presence of dextran has a positive impact on the hydrogel's structure by decreasing the pore size at the same time as increasing the uniformity and interconnectivity of the pores. In addition, there is an increased degree of swelling and of the encapsulation capacity of PFs, with the increase of the dextran content in hydrogels. The kinetics of PFs released by hydrogels was studied according to the Korsmeyer-Peppas model, and it was observed that the transport mechanisms depend on hydrogels' composition and morphology. Furthermore, CD hydrogels have been shown to promote cell proliferation without cytotoxicity, by successfully culturing fibroblasts and endothelial cells on CD hydrogels (over 80% viability). The anti-inflammatory tests performed in the presence of lipopolysaccharides demonstrate the anti-inflammatory properties of the PFs-loaded hydrogels. All these results provide conclusive evidence on the acceleration of wound healing by inhibiting the inflammation process and support the use of these hydrogels encapsulated with PFs in wound healing applications.

Enrichment of Cellulose Acetate Nanofibrous Scaffolds with Retinyl Palmitate and Clove Essential Oil for Wound Healing Applications.

The use of biocompatible materials and fabrication methods is of particular importance in the development of wound dressings. Cellulose acetate (CA) has excellent properties for wound dressing applications, but it is insufficient for the wound healing process due to its lack of bioactive and antibacterial properties. In this study, CA was electrospun with retinyl palmitate (RP) and clove essential oil (CLV) to fabricate a novel antibacterial and antioxidant biomaterial. The effects of RP and CLV incorporation on the surface morphology, fiber diameter, antioxidant activity, antibacterial activity, cell viability, and release behavior of the fabricated CA mats were investigated. In light of these studies, it was determined that the nanofiber mat, fabricated with a 15% w/v CA polymer concentration, a 1% w/w RP ratio, and a 5% w/w CLV ratio, was biocompatible with L929 fibroblast cells with antibacterial and antioxidant properties. Overall, results showed that this nanofiber offers promise for use as a wound dressing.

Multi-functional carboxymethyl chitosan/sericin protein/halloysite composite sponge with efficient antibacterial and hemostatic properties for accelerating wound healing

The development of wound dressings with hemostatic and antibacterial properties has attracted great attention. In this study, we prepared a multi-functional natural substance sponge (CMC/Ser-Ag/HNT) composed of carboxymethyl chitosan (CMC), sericin-silver nanoparticle (Ser-Ag), and halloysite (HNT). CMC/Ser-Ag/HNT sponge was demonstrated to bear desired hygroscopicity, porosity, compressive strength and compressive stability, cytocompatibility, and hemocompatibility. The mechanical properties (compressive strength of 100 kPa) and hemostatic capacity (hemostasis time of 15 ± 3 s in the liver injury model and 12 ± 3 s in the caudal injury model) were enhanced by introducing HNT into the CMC sponge. Ser-Ag was synthesized in situ via the redox nature of tyrosine residues in sericin in a “one-step, green” way to enhance the antibacterial activity of the hybrid sponge against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). In addition, the rat full-thickness skin defect model experiments demonstrated that the CMC/Ser-Ag/HNT4 sponge significantly promoted epithelialization and collagen formation. Immunofluorescence staining assays revealed that the composite sponge reduced inflammation by downregulating the expression of IL-6 and enhanced angiogenesis by upregulating VEGF expression. All the findings demonstrated the great potential of CMC/Ser-Ag/HNT sponge as versatile clinical wound dressing, especially for hemorrhagic and infected wounds.

Plasma-Functionalised Dressings for Enhanced Wound Healing.

Fundamental knowledge about cell-surface interactions can be applied in the development of wound dressings and scaffolds to encourage wounds to heal. As surfaces produced with acid-functionalised monomers encourage keratinocyte adhesion, proliferation and migration, whilst amine functionalisation enhances fibroblast proliferation and migration in vitro, standard care wound dressings were plasma-coated with either acrylic acid or allylamine and applied to 6 mm excisional wounds on the backs of mice to test their effectiveness in vivo. At day 3, the rate of wound healing was increased in mice treated with dressings that were plasma-coated with allylamine compared to uncoated dressings, with a significantly reduced wound area. However, healing may be impaired following prolonged treatment with allylamine-functionalised dressings, with delayed re-epithelialisation and increased cellularisation of the wound site at later timepoints. Acrylic acid functionalisation, however, offered no early improvement in wound healing, but wounds treated with these dressings displayed increased collagen deposition at day 7 post wounding. These results suggest that plasma polymerisation may allow for the development of new dressings which can enhance wound closure by directing cell behaviour, but that the application of these dressings may require a timed approach to enhance specific phases of the wound healing response.

Rotary Jet Spinning (RJS): A Key Process to Produce Biopolymeric Wound Dressings.

Wounds result from different causes (e.g., trauma, surgeries, and diabetic ulcers), requiring even extended periods of intensive care for healing, according to the patient's organism and treatment. Currently, wound dressings generated by polymeric fibers at micro and nanometric scales are promising for healing the injured area. They offer great surface area and porosity, mimicking the fibrous extracellular matrix structure, facilitating cell adhesion, migration, and proliferation, and accelerating the wound healing process. Such properties resulted in countless applications of these materials in biomedical and tissue engineering, also as drug delivery systems for bioactive molecules to help tissue regeneration. The techniques used to engineer these fibers include spinning methods (electro-, rotary jet-), airbrushing, and 3D printing. These techniques have important advantages, such as easy-handle procedure and process parameters variability (type of polymer), but encounter some scalability problems. RJS is described as a simple and low-cost technique resulting in high efficiency and yield for fiber production, also capable of bioactive agents' incorporation to improve the healing potential of RJS wound dressings. This review addresses the use of RJS to produce polymeric fibers, describing the concept, type of configuration, comparison to other spinning techniques, most commonly used polymers, and the relevant parameters that influence the manufacture of the fibers, for the ultimate use in the development of wound dressings.

Poly(aspartic acid)-based self-healing hydrogel with precise antibacterial ability for rapid infected-wound repairing

The development of wound dressings with antibacterial activities and simultaneous pro-healing functions are always urgent in treating bacterial wound infection. Herein, a novel multifunctional self-healing hydrogel was designed and prepared by crosslinking quaternary ammonium/boronic acid modified poly(aspartic acid) and poly (vinyl alcohol) polymers with targeted peptide MP196- conjugated polydopamine. The formation of this hydrogel not only improves the biocompatibility of quaternary poly(aspartic acid), but also enhances antibacterial efficacy by pH-triggering dissociation under the low pH bacterial microenvironment. Moreover, precise photothermal treatment can be achieved. In vitro study suggested high synergistic antibacterial efficiency(∼100 %) under near-infrared light, significantly higher than a single antibacterial strategy (66.0–82.6 %). In vivo study suggested infected wounds treated with the hydrogel showed an optimal healing rate(92.0 %) after 7 days. The survival rate of the bacteria in the epidermal tissues was reduced to 2.3 %. Besides, the suitable self-healing property of this hydrogel facilitated its application in the diversity of wound shapes. Thus, the novel poly(aspartic acid) hydrogel might be a promising candidate for precise therapy of bacteria-infected wounds.

Engineered reversible adhesive biofoams for accelerated dermal wound healing: Intriguing multi-covalent phenylboronic acid/cis-diol interaction

Development of wound dressings that not only have multiple advantages including good biocompatibility, barrier properties, adhesive properties, but also exhibit anti-bacterial, anti-infection, and promote cell adhesion, accelerated wound healing process, remains challenging in the skin tissue engineering. In this work, a noninvasive adhesive biofoam with interconnectivity structure and antibacterial property was first-time designed through solid surfactant stabilized high internal phase emulsion (HIPE) template, where bacteriostatic Zn-CuO@GO nanosheets dynamic assemble with 4-vinylphenylboronic acid at the oil/water interface as cooperative emulsifier. Taking advantage of the cooperative assembly of the PBA/Zn-CuO@GO, the biofoams display microfibrousity and interconnectivity to facilitate material-cellular interaction as well nutrient exchange, and can effectively inhibit the growth of E. coli and S. aureus. Meanwhile, phenylboronic acid functional groups not only endow biofoam reversible adhesive property though dynamic phenylboronic acid/cis-diol interaction, but also facilitate the rearrangement of Zn-CuO@GO nanosheets in more uniform way at emulsion interface. Taken together, the biofoams can strongly adhere to the wound bed reducing the risk of infection and accelerate earlier tissue regeneration, which exhibit great potential in bioengineering application.

Near-infrared light-responsive and antibacterial injectable hydrogels with antioxidant activity based on a Dopamine-functionalized Gellan Gum for wound healing

The development of wound dressings with combined antioxidant, antibacterial and tissue adhesion functions has been a difficult medical task for the treatment of wound infections. We synthetized a dopamine and PEG functionalized Gellan Gum (GG) to produce an injectable hydrogel with radical scavenging activity having both specific and aspecific antibiotic/antimicrobial properties. Using starting GG with different molecular weights, we obtained two derivatives that have been used to prepare the gel precursor dispersion, that undergoes gelation in the presence of colistin and dried microparticles (MPs) functionalized on the surface with polydopamine (pDA). Both were used to dope the hydrogel, increase the radical scavenger activity and impart near-infrared light (NIR) responsiveness. Indeed, with an irradiation of 810 nm, the incorporated microparticles exhibit photothermal transformation properties and improve the release of antibiotics on demand. The combination of photothermal and antibiotic therapy with synergistic antibacterial action acts on Pseudomonas aeruginosa and leads to a bactericidal effect in a few hours, while on Staphylococcus aureus there is an effect of inhibition of growth over time due only to the hyperthermic effect. We believe this study provides a promising method for fabricating a multifunctional injectable hydrogel for the potential treatment of infected skin wounds.

Recent Progress in Electrospun Polyacrylonitrile Nanofiber-Based Wound Dressing.

Bleeding control plays a very important role in worldwide healthcare, which also promotes research and development of wound dressings. The wound healing process involves four stages of hemostasis, inflammation, proliferation and remodeling, which is a complex process, and wound dressings play a huge role in it. Electrospinning technology is simple to operate. Electrospun nanofibers have a high specific surface area, high porosity, high oxygen permeability, and excellent mechanical properties, which show great utilization value in the manufacture of wound dressings. As one of the most popular reactive and functional synthetic polymers, polyacrylonitrile (PAN) is frequently explored to create nanofibers for a wide variety of applications. In recent years, researchers have invested in the application of PAN nanofibers in wound dressings. Research on spun nanofibers is reviewed, and future development directions and prospects of electrospun PAN nanofibers for wound dressings are proposed.
