ANALYSIS OF TOTAL FLAVONOID CONTENT AND ANTIOXIDANT ACTIVITY ASSAY OF GUAVA VARIETY CRYSTAL (Psidium guajava L.) LEAVES EXTRACT

The study investigates the antioxidant activity and total flavonoid compounds in leaves crystal guava, a plant rich in vitamin C and high in antioxidant activity. The research involved maceration extraction of leaves cystal guava using methanol solvent, then fractionation with ethyl acetate and n-hexane solvents in stages to obtain extracts methanol, ethyl acetat and n-hexane; thereafter extracts were subjected to phytochemical screening, total flavonoid content determination, and antioxidant activity using the DPPH method. The study found secondary metabolite compounds which is found in leaves crystal guava such as flavonoids, alkaloids, steroids, terpenoids, saponins and tannins; that ethyl acetate extract had the highest flavonoid content of 171.91 mg/L and highest antioxidant activity with IC50 value of 9.59 ppm, followed by n-hexane and methanol extracts.

HPLC-ESI/MS-MS metabolic profiling of white pitaya fruit and cytotoxic potential against cervical cancer: Comparative studies, synergistic effects, and molecular mechanistic approaches

Natural approach became a high demand for the prevention and treatment of such diseases for their proven safety and efficacy. This study is aimed to perform comparative phytochemical analysis of white pitaya (Hylocereus undatus) peel, pulp and seed extracts via determination of total flavonoid content, phenolic content, and antioxidant capacity, coupled with HPLC-ESI/MS-MS analysis. Further, we evaluated the synergistic cytotoxic potential with Cisplatin against cervical cancer cells with investigation of underlying mechanism. The highest content of phenolics and antioxidants were found in both seed and peel extracts. The HPLC-ESI/MS-MS revealed identification of flavonoids, phenolic acids, anthocyanin glycosides, lignans, stilbenes, and coumarins. The cytotoxicity effects were evaluated by MTT assay against prostate, breast and cervical (HeLa) and Vero cell lines. The seed and peel extracts showed remarkable cytotoxic effect against all tested cell lines. Moreover, the selectivity index confirmed high selectivity of pitaya extracts to cancer cells and safety on normal cells. The combined therapy with Cisplatin effectively enhanced its efficacy and optimized the treatment outcomes, through the apoptotic ability of pitaya extracts in HeLa cells, as evaluated by flow cytometry. Besides, RT-PCR and western blotting analysis showed downregulation of Bcl-2 and overexpression of P53, BAX among HeLa cells treated with pitaya extracts, which eventually activated apoptosis process. Thus, pitaya extract could be used as adjuvant therapy with cisplatin for treatment of cervical cancer. Furthermore, in-vivo extensive studies on the seed and peel extracts, and their compounds are recommended to gain more clarification about the required dose, and side effects.

ANALISIS KADAR FENOLIK TOTAL DAN FLAVONOID TOTAL EKSTRAK ETANOL KULIT PISANG KEPOK (Musa pradisiaca L)

Banana peel is a very large amount of waste material (banana fruit waste). In general, banana peels have not been used optimally. Based on the research results, kepok banana peel contains secondary metabolites in the form of flavonoids, alkaloids, tannins and saponins. This study aims to determine the total phenolic content and total flavonoid content of the ethanol extract of kepok banana skin (Musa pradisiaca L). Extraction was carried out by maceration method with 96% ethanol solvent. Determination of total phenol content used the Folin-Ciocalteu method with gallic acid as comparator compound and the determination of total flavonoid content used the colorimetric method with 10% AlCl3 and 5% acetic acid as reagents. The results showed that kepok banana peel extract had an average total phenolic content of 13.4495 mg GAE/g ± 1.5473206 and an average total flavonoid content of 8.5468 mg QE/g ± 0.947422695.

Analysis of Total Flavonoid and Phenol Content from the Combination of Red Spinach (Amaranthus tricolor L.) Ethanolic Extract and Chrysanthemum Flower (Chrysanthemum morifolium) Ethanolic Extract as a Potential Anti-anemic

Red spinach and chrysanthemum are two plants that contain various active compounds, including flavonoids and phenols which have good antioxidant activity. One of the benefits of antioxidants is as an anti-anemia. The combination of red spinach and chrysanthemum extracts is expected to be a potential alternative treatment for anemia by combining high Fe and antioxidants found in both herbal ingredients. Research related to test the levels of flavonoids and phenols in red spinach and chrysanthemum extracts (in combination) has not been carried out. This study aimed to measure the total flavonoid and phenol levels of the combination of red spinach ethanol extract (EEBM) and chrysanthemum flower ethanol extract (EEBK). The design of this research is descriptif quantitative. The method used for the extraction is maceration. The solvent used is 70% ethanol. Then the two extracts were combined by mixing the two extracts with a ratio of 5.6 ml EEBM and 1.4 ml EEBK. The process of mixing the extracts was carried out by the centrifugation method. The suspension of the combination of EEBM and EEBK was then measured for the total flavonoid and phenol content. Determination of total flavonoid content (TFC) was carried out using UV-Vis spectrophotometry method with AlCl3 reagent at 425 nm. TFC results are expressed in quercetin equivalent (QE). The TFC result from the combination of EEBM and EEBK was 85.33 mg QE/g. Determination of total phenol content (TPC) was carried out using UV-Vis spectrophotometry method with Folin-Ciocalteu LP reagent at 735 nm. TPC results are expressed in gallic acid equivalent (GAE). The TPC result from the combination of EEBM and EEBK is 25.22 GAE/g. Flavonoids and phenols contained in the combination of EEBM + EEBK extracts can be an alternative anti-anemia because they have the effect of helping the absorption of iron in the intestine

Evaluation of the Total Content of Phenols and Flavonoids in Two Different Extracts of Orbea wissmannii O. Schwart and their Toxic Effect on WI38 and HepG2 Cell Lines.

This study aimed at estimating the total phenolic content, total flavonoid content and evaluation of toxic effect of chloroform and aqueous methanol extracts of Orbea wissmannii against WI38 and HepG2 cell lines. Orbea wissmannii was collected from Qaren region near Amran governorate, Yemen. Chloroform and methanol extracts of whole parts of Orbea wissmannii were prepared by maceration and decantation. The total polyphenol content was estimated using Folin–Ciocalteau method. Aluminum chloride colorimetric method was applied for the determination of total flavonoid content. The cytotoxic activity was measured using the MTT assay. The total phenolic content of chloroform and methanol extracts was 70.21 mg GAE/g, 122 mg GAE/g respectively (milligrams of gallic acid equivalents per gram), while the flavonoid content was 21.80 mg QE/g and 54.24 mg QE/g respectively (milligrams of quercetin dihydrate equivalents per gram). Dependently, the two extracts significantly and concentratedly reduced the viability of lung tissue-derived fibroblasts (wi38) and human hepatoma cells (HepG2). The two extracts showed effective effect on HepG2 at 250, 500 and 1000 μg/ml, while the same extracts showed their effect on the WI38 at 500 and 1000 μg/ml. The toxicological properties shown by the extracts of this plant require further study to isolate and purify the active substances responsible for this high anti-cancer efficacy. Phytochemical investigation on chloroform/methanol extracts and their structures is recommended.

PENETAPAN KADAR TOTAL FENOLIK-FLAVONOID EKSTRAK ETANOL 70% KULIT BATANG TANDUI (Mangifera rufocostata Kosterm.)

Tandui (Mangifera rufocostata Kosterm.) is a species of the genus Mangifera which is known to contain phenolic and flavonoid compounds and is one of the plants used as an alternative medicine by the people of South Kalimantan. The purpose of this study was to determine the total phenolic and flavonoid content of 70% ethanol extract of tandui bark. Qualitative analysis using phenolic and flavonoid phytochemical screening with 3% FeCl3, Mg-HCl reagents and Thin Layer Chromatography (TLC) with spot appearance 10% FeCl3 for phenolic and 5% AlCl3 for flavonoid compounds. Quantitative analysis using UV-Vis spectrophotometry, determination of total phenolic content using Folin-Ciocalteu reagent with gallic acid standard solution, while determination of total flavonoid content using AlCl3 reagent with quercetin standard solution. The results of identification of phenolic and flavonoid compounds by phytochemical screening and TLC showed that 70% ethanol extract of tandui bark contained phenolic and flavonoid. The results of the determination of the total phenolic content using UV-Vis spectrophotometry were 471.3126 mgGAE/g or 47.1312%, while the total flavonoid content was 872.075 mgQE/g extract or 87.2075%. Based on these results, it can be concluded that 70% ethanol extract of tandui bark contains secondary metabolites in the form of phenolics and flavonoids which have potential as medicine.

Tectona Grandis Leaves: Determination of Total Flavonoid Content, Phenolic Content, Characterization of the Leaves, and Compound Identification in GC-MS

Nowadays, traditional plant study has grown in importance as their use has been increased. Tectona grandis (teak) is one of several plants that have been studied for its phytochemical and pharmacological properties. This plant includes a number of secondary metabolites, which may explain its diverse pharmacological properties. Although teak leaf compounds have been examined in the past, there is still little information on the diversity of teak leaf compounds. As a result, we are employing several methodologies to determine the total flavonoids content, phenolic content, water extractable matter, ethanol extractable matter, total ash content, and other component analysis of teak leaves. We found that the average of total flavonoid content in the teak leaves is about 3.93 ± 0.008%w/w, while the average total phenolic concentration in teak leaves is about 4.3±0.15%w/w. The Gas Chromatography- Mass Spectrometry GC-MS study of the methanol extract of T. grandis leaves also found twenty-three active chemical compounds (phytochemical components)

Antioxidant Activity of Flavonoids from Cassava Leaves (Manihot esculenta Crantz)

The community uses cassava leaves for cooking because of their high protein content, β-carotene, Fe, Mg, Zn, S, Ca, Zn, Ni, and K. Cassava leaves contain various secondary metabolites, including flavonoids, saponins, tannins, phenolics, steroids, anthocyanins, and anthraquinones. The antioxidant activity of flavonoid compounds in cassava leaves (Manihot esculenta Crantz) has been successfully investigated. This study consisted of six stages: the preparation of ethanol extract of cassava leaves, phytochemical screening, determination of total flavonoid content in the extract, isolation of flavonoids, and purity testing of flavonoid isolates using the chromatographic method, identification of the structure of pure flavonoid isolates using a UV-Vis spectrophotometer with the addition of a shear reagent, FTIR, and LC-MS/MS. As well as test the antioxidant activity using the DPPH method. The yield of ethanol extract from cassava leaf in this study was 14.67%. The results of the phytochemical screening showed that the cassava leaves and the ethanol extract of cassava leaves contained alkaloids, flavonoids, saponins, tannins, quinones, steroids, phenolics, and triterpenoids. The total content of flavonoids in the ethanol extract was 35.71 mg EQ/g extract. UV-Vis, FTIR, and LC-MS/MS analysis revealed that the flavonoid isolate was quercetin. The results of the antioxidant activity of the ethanol extract and BC isolate showed IC50 of 81.76 ± 0.505 mg/L and 77.85 ± 4.708 mg/L.

Antioxidant Assay and Total Flavonoid Determination of Ethanolic Extract of Walnut (Canarium indicum L.) Leaves and Its Fractions

Antioxidants are compounds that inhibit free radicals. Many plants have antioxidant activity because they contain antioxidant compounds such as flavonoids. This study aimed to evaluate the antioxidant activity, phytochemical contents, total flavonoid content and to establish the relationship of total flavonoid content and antioxidant activity of extract and fractions of walnut (Canarium indicum L.) leaves. Antioxidant activity of ethanolic extract, n-hexane fraction, ethyl acetate fraction, and ethanolic-water fraction of walnut leaves was examined by using the DPPH method and determination of total flavonoid content was determined spectrophotometrically. The results showed that antioxidant activity of ethanolic extract, n-hexane fraction, ethyl acetate fraction, and ethanolic-water fraction based were 25.294 ± 0.055; 175.245 ± 0.4999; 20.135 ± 0.009; and 28.806 ± 0.0424 μg/ml, respectively. The phytochemical content of ethanol extract of walnut leaves are saponins, flavonoids, tannins, and polyphenols. The total flavonoid content of ethanolic extract, n-hexane fraction, ethyl acetate fraction and ethanolic-water fraction were 2.624 ± 0.012; 0.499 ± 0.023; 3.846 ± 0.006; and 1.596 ± 0.006 gram quercetin equivalent/gram extract, respectively. Correlation between antioxidant activity and total flavonoid content revealed that 63.2 % of antioxidant activity was influenced by the presence of flavonoid compounds.

Determination of Total Flavonoid Content from Ethanol Extract and Various Paliasa (Kleinhovia Hospita L.) Leaf Fractions on its Potential as an Antioxidant

Paliasa (Kleinhoivia hospita L.) is a plant of the Sterculiaceae family which is known to have potential as an antioxidant. This study aims to determine the total flavonoid content of ethanol extract and various fractions of paliasa leaves on their potential as antioxidants. Samples were macerated with 70% ethanol for 24 hours at room temperature. The ethanol extract obtained was then evaporated at 50 o C to produce a viscous extract which was then fractionated using n -hexane, dichloromethane and water as solvents. The content of chemical compounds from the ethanol extract of paliasa leaves are alkaloids, flavonoids, saponins, phenols and tannins. The content of chemical compounds from the n -hexane fraction is tannins. The content of chemical compounds from the dichloromethane fraction are flavonoids, phenols and tannins. The content of chemical compounds from the water fraction are alkaloids, saponins, phenols and tannins. Tests for total flavonoid compounds found in the ethanol extract and dichloromethane fraction using rutin as a comparison at λ max 419 nm were 3.10% and 3.73%. Antioxidant activity testing using the 1,1-Diphenyl-2-Picrilhydrazil (DPPH ) method showed that the ethanol extract and fraction ( n -hexane, dichloromethane and water) had an IC of 50 166.55 µg/mL, 351.19 µg/mL, 85 .87 µg/mL, and 226.52 µg/mL, while gallic acid comparator has a very strong antioxidant activity with an IC50 value of 10.75 µg/mL

Determination of Total Flavonoid Content of Limpasu Pericarp Extract (Baccaurea lanceolata) by Spectrophotometric Uv-Vis

Limpasu (Baccaurea lanceolata) is a native plant of Borneo which is used empirically to protect the skin from sun exposure. Limpasu contains flavonoid compounds which generally act as antioxidants. This study aims to determine the total flavonoid content of limpasu pericarp extract by using UV-Vis spectrophotometry method. Limpasu pericarp was extracted by using the stratification maceration method. This extraction was carried out by immersing the simplicia into three types of solvents with different polarities, respectively, namely n-hexane (nonpolar), ethyl acetate (semi-polar), and methanol (polar). The concentration of n-hexane, ethyl acetate, and methanol extracts was determined using a UV-Vis spectrophotometer at a maximum wavelength of 510 nm with quercetin as standard. Based on the test, it was found that the total flavonoid content of pericarp extracts, hexane, ethyl acetate, and methanol, respectively, was 44.1552 g/ml; 155.8506 g/ml; and 25.8506 g/ml. The highest total flavonoid content was found in the ethyl acetate extract.

Mustard is a vegetable that has many benefits for the body.The stages of this research include sample collection and plant processing, plant identification, manufacture of ethanol extract of bitter mustard leaf, determination of water content, phytochemical screening and determination of total flavonoid content by UV-Visible spectrophotometry.This study aims to determine the total flavonoid content of bitter mustard greens and to determine the characteristics, class of chemical compounds, and total flavonoid value of bitter mustard (Brassica juncea (L.) Czern) extract by UV-VIS spectrophotometry.

Physical Stability Test and Determination of Total Flavonoid Content of Watermelon (Citrullus lanatus) Fruit Mesocarp Extract Gel Preparation

The content of flavonoids in the white skin or mesocarp of watermelon has antioxidant power so that the active substance can be used in the manufacture of cosmetics, one of which is a gel dosage form. This type of research is observational with a descriptive research design. The purpose of this study was to obtain data on the stability of the watermelon (Citrullus lanatus) mesocarp extract gel preparation using the cycling test method. Physical stability evaluations were carried out, including organoleptic, homogeneity, pH, adhesion, spreadability, and viscosity. The results showed that the organoleptic observation of the preparation was in the form of a semi-solid, had a characteristic aromatic odor of the extract, and had a brownish-yellow color. As for the test results for the entire homogeneous cycle, the average pH range is 4.75 - 5, adhesion is 2.45 - 3.96 seconds, dispersion is 5.59 - 6.67 cm, and viscosity is 5426 - 6830 cps, and total flavonoid content of the gel preparation was 5.27%. This study concluded that the gel preparation was physically stable before and after 12 days of storage and contained 5.27% flavonoids.

DETERMINATION OF TOTAL FLAVONOID LEVELS OF ETHANOL EXTRACT MORINGA LEAVES (Moringa oleifera, LamK) BY SPECTROPHOTOMETRY UV-Vis

Moringa (Moringa oleifera L.) is one of the plants that can be used both as food and medicine. The parts of this plant which often used as medicine are the seeds, leaves, and barks. The purpose of this study was to determine the total flavonoid of the ethanol extract of Moringa leaves by UV-Vis spectrophotometry. Extraction method for Moringa leaves using maceration method with 96% ethanol solvent.Furthermore, phytochemical screening was carried out on the ethanolic extract of Moringa leaves using the Thin Layer Chromatography (TLC) method and determination of total flavonoid content by UV-Vis spectrophotometry. The results of the research are Moringa contains alkaloids, flavonoids, tannins, free steroids/terpenoids, and steroids/terpenoid glycosides. Each gram of Moringa (Moringa oleifera, LamK) contain a total flavonoid compound content of 32.261 gQE/g extract with a percentage yield of 5,11%. The conclusion that Moringa leaves contain flavonoids with total flavonoid content of 32.261 μgQE / g extract.

Determination of Total Flavonoid Content of Saputangan Leaves (Maniltoa grandiflora (A. Gray) Scheff) and Its Ability as Antioxidant

Determination of phenolic content using uv-visible and measured IC50 as antioxidant of saputangan leaves. Methods: Screening was used FeCl3, NH3, Alkali and Pb(CH3COO)2. The maceration process was carried out using ethanol as a solvent and soaked for more than 24 hours. The measurement of total flavonoid levels began with determining the maximum wavelength of quercetin, which was 442 nm. The linear regression equation for the resulting quercetin calibration curve was the equation y = 0.0155x + 0.0387 with a correlation value (R2) of 0.9943. The linear regression of antioxidant of saputangan leaves on percentage of inhibition is y = 0.6899x + 1.5616 with a correlation (R2) of 0.9895. The Maniltoa grandiflora (A. Gray) Scheff leaves contained flavonoid compounds which are proven based on the results of screening. Meanwhile, the weight of the extract after partitioning using ethyl acetate and n-hexane was 50 gr. Total flavonoid levels in Maniltoa grandiflora (A. Gray) Scheff leaves were 33.87 mg QE/g extract or 33.87%. IC50 value of 70.2108 ppm which was categorized as an antioxidant at the medium (medium) level. Conclusions: Total flavonoid content was 3.87 mg QE/g extract or 33.87% and categorized as antioxidant on medium level.

Investigation of Changes of Antioxidant Properties of Coffee through Fermentation by Using Saccharomyces Cerevisiae and Bacillus Subtilis

The study was carried out to investigate the changes of antioxidant properties and quality of green coffee beans fermented by using Bacillus subtilis and Saccharomyces cerevisiae as starter cultures in sugarcane and banana juice at different times fermentation. The fermentations were conducted with different juice concentrations for different fermentation times 24, 48, and 72 hours at room temperature. Antioxidant properties were determined by Folin-Ciocalteu reagent for the determination of total phenolic content, DPPH assay for the determination of antioxidant capacity, and colorimetric method used for determination of total flavonoid content. As a result, coffee fermentation at 10oBrix, 107 cells/mL for each type of microbe, and the time (48 hours) had a positive effect on antioxidant properties. However, there was not a significant difference in terms of antioxidant properties between fermented and original coffee beans at optimal conditions. Therefore, antioxidant properties were dramatically reduced during the early stages of fermentation, but it was shown to be able to overcome it through this study. This study is a premise to applying microbial products through fermentation to create a coffee with high antioxidant activity compared to conventional coffee products.

PENETAPAN KADAR FLAVONOID DAN AKTIVITAS ANTIOKSIDAN PADA EKSTRAK BATANG BROTOWALI (Tinospora crispa L.) DENGAN METODE CUPRAC

Oxidative stress is damage caused by reactive oxygen species Compounds that can prevent oxidative stress are antioxidants. The brotowali plant (Tinospora crispa) has been used for the treatment of diabetes and hypertension. The brotowali plant contains secondary metabolites such as flavonoids. Flavonoids are known to be a class of compounds that have antioxidant activity. so it can be kind of solvent which is the most excellent in the test plant extract brotowali. Extraction was conducted by reflux-rise that use nhexane, ethyl acetate and ethanol 96%. Determination of antioxidant activity using them CUPRAC method. Determination of total flavonoid content using colorimetric aluminum chloride method with quercetin as standard. The results showed that the ethyl acetate extract sample had strong antioxidant activity compared to the extract sample with 96% ethanol and n-hexane. The EC50 value of the ethyl acetate extract was 53.637 g/mL. Extract with n-hexane solvent had the highest flavonoid content, namely 9,937 ± 0.009 gQE/100 g extract.

Combined Metabolome and Transcriptome Analyses Reveal the Flavonoids Changes and Biosynthesis Mechanisms in Different Organs of Hibiseu manihot L.

Hibiseu manihot L. (Jinhuakui, JHK), also known as a garden landscape plant, is widely cultivated as a landscape plant having pharmacological effects due to its high flavonoids content. Although flavonoids were the main active pharmaceutical ingredients in JHK, little information was obtained about the content, composition, and accumulation pattern of flavonoids in different tissues. Most studies only identified a few kinds of flavonoids in JHK limited by separation and identification problems. Therefore, combined metabolome and transcriptome analysis was performed to explore the accumulation patterns and biosynthesis mechanisms of flavonoids in JHK. In this study, we identified 160 flavonoids in 15 samples of JHK (flower, leaf, root, stem, and seeds) by using LC-MS/MS. Consistent with the total flavonoid content determination, these flavonoids were significantly accumulated in flowers, followed by leaves, stems, roots, and seeds. Among them, certain flavonoids, with high content, were also identified for the first time in JHK, such as tricetin, catechin, hesperidin, ncyanidin-3-O-sambubioside, astragalin, procyanidin B2/B3/C1, apigenin-5-O-glucoside, etc. Different tissues underwent significantly reprogramming of their transcriptomes and metabolites changes in JHK, particularly in the flavonoid, flavone, and flavonol biosynthesis pathways. We conducted a correlation analysis between RNA-seq and LC-MS/MS to identify the key genes and related flavonoids compounds, rebuild the gene-metabolites regulatory subnetworks, and then identified 15 key genes highly related to flavonoids accumulation in JHK. These key genes might play a fine regulatory role in flavonoids biosynthesis by affecting the gene expression level in different organs of JHK. Our results could be helpful for the improvement of the market/industrial utilization value of different parts of JHK, to pave the way for the regulatory mechanism research of flavonoids biosynthesis, and provide insight for studying the production quality improvement of JHK.

Antioxidant activity, total flavonoid, and total tannin content of ethanol extract of avocado peel (Persea americana Mill.)

Background: Avocado is a plant that can grow in tropical climates. In general, people only consume the fruit while the skin and seeds are not used. Avocado fruit, skin and seeds contain secondary metabolites such as flavonoids and tannins which have antioxidant activity. Objective: To determine antioxidant activity, total flavonoid, and total tannin content of the ethanol extract of avocado peel. Methods: P. americana Mill. was collected from Lubuk Linggau, South Sumatra. The ethanol extract of P. americana Mill. peel obtained by maceration with ethanol. Qualitative analysis of flavonoids using Mg and HCl while tannins using FeCl3. Determination of total flavonoid content was carried out with AlCl3 reagent and Folin-Ciocalteu reagent to determine the total tannin content and was measured by a UV-Vis Spectrophotometer. Determination of antioxidant activity using the DPPH method. Result: The results of the qualitative analysis showed that ethanol extract of avocado peel contained flavonoids and tannins. It also had antioxidant activity with IC50 of 41,93 µg/mL. The total flavonoid and tannin content of it were 125,01 mgQE/g extract and 53,82 mgTAE/g extract respectively. Conclusion: Ethanol extract of avocado peel had antioxidant activity, while the total flavonoid compounds was higher than the total tannins. Keywords: antioxidant, DPPH, total flavonoid, total tannins
