Development of a double antibody sandwich ELISA method for the quantitative detection of serum C-reactive protein based on nanobody

In this study, we successfully developed a nanobody-based double antibody sandwich ELISA kit for the detection of clinical serum C-reactive protein (CRP) by using two novel CRP specific nanobodies.The developed method exhibited a linear detection range of approximately 6–200 ng/mL, with a detection limit of 1 ng/mL. Furthermore, the method demonstrated excellent specificity, as there was no cross-reactivity with interfering substances such as total bilirubin and hemoglobin and so on. To assess reproducibility, independent measurements of the samples were conducted under experimental conditions, resulting in intra- and inter-batch coefficients of variation below 10% and a recovery rate of 93%–102%. These results indicate robust reproducibility of the method.To evaluate the performance of the developed kit, we collected 90 clinical samples for correlation analysis with commercial kits. The results showed a high correlation coefficient value (R2) of 0.98, indicating accurate concordance between the developed and commercial kits.In conclusion, our study successfully developed a nanobody-based double antibody sandwich ELISA kit to detect clinical serum CRP. The utilization of nanobodies represents a significant advancement in the field of CRP immunoassay development. The developed kit demonstrates excellent performance characteristics and holds promise for clinical applications.

Fabrication of a Disposable Electrochemical Immunosensor Based on Nanochannel Array Modified Electrodes and Gated Electrochemical Signals for Sensitive Determination of C-Reactive Protein.

Sensitive determination of C-reactive protein (CRP) is of great significance because it is an early indicator of inflammation in cardiovascular disease and acute myocardial infarction. A disposable electrode with an integrated three-electrode system (working, reference, and counter electrodes) has great potential in the detection of biomarkers. In this work, an electrochemical immunosensing platform was fabricated on disposable and integrated screen-printed carbon electrode (SPCE) by introducing nanochannel arrays and gated electrochemical signals, which can achieve the sensitive detection of CRP in serum. To introduce active reactive groups for the fabrication of immuno-recognitive interface, vertically-ordered mesoporous silica-nanochannel film (VMSF) with rich amino groups (NH2-VMSF) was rapidly grown by electrochemical assisted self-assembly (EASA). The electrochemically reduced graphene oxide (ErGO) synthesized in situ during the growth of NH2-VMSF was used as a conductive adhesive glue to achieve stable bonding of the nanochannel array (NH2-VMSF/ErGO/SPCE). After the amino group on the outer surface of NH2-VMSF reacted with bifunctional glutaraldehyde (GA/NH2-VMSF/ErGO/SPCE), the converted aldehyde surface was applied for covalent immobilization of the recognitive antibody (Ab) followed with the blocking of the non-specific sites. The fabricated immunosensor, Ab/GA/NH2-VMSF/ErGO/SPCE, enables sensitive detection of CRP in the range from 10 pg/mL to 100 ng/mL with low limit of detection (LOD, 8 pg/mL, S/N = 3). The immunosensor possessed high selectivity and can realize reliable determination of CRP in human serum.

Long period grating in double cladding fiber coated with graphene oxide as high-performance optical platform for biosensing

In this work, the development and testing of a novel fiber-optic based label-free biosensor is presented, whose performance were verified through the detection of C-reactive protein (CRP) in serum. The device is based on a long period grating fabricated in a double cladding fiber with a W-shaped refractive index (RI) profile. As a result, the working point of the device was tuned to the mode transition region by chemical etching of the outer fiber cladding, obtaining a significant enhancement of the RI sensitivity and an excellent visibility of the grating resonances due to the mode transition in an all-silica structure. The fiber transducer was coated with a nanometric thin layer of graphene oxide in order to provide functional groups for the covalent immobilization of the biological recognition element. A very low limit of detection of about 0.15 ng/mL was obtained during the detection of CRP in serum, and a large working range (1 ng/mL – 100 μg/mL) of clinical relevance has been also achieved.

This paper describes a simple and sensitive immuno-based biosensor for interference-reduced detection of C-reactive protein (CRP) in serum. The detection was performed by using a non-competitive sandwich immunoassay in combination with surface plasmon field-enhanced fluorescence spectroscopy (SPFS). CRP is an important marker for the diagnosis of inflammatory processes and cardiovascular diseases (CVD). It is nowadays detected by high-sensitivity enzyme-linked immunosorbent assays (ELISA) in blood serum. CRP was used as a model analyte in this work because it is well-characterized. However, interfering effects of matrix components affect the limit of detection (LOD) and quantification (LOQ) in general. Therefore, the availability of fast, sensitive and robust analytical methods is of major interest. A number of biosensor approaches have been described already, but only a few have demonstrated their usefulness in authentic samples such as serum. Thus our aim was to develop a simple and sensitive immunoassay-based biosensor for an interference-reduced detection of CRP in serum with surface plasmon enhanced fluorescence spectroscopy (SPFS). LODs and LOQs were experimentally determined both for CRP spiked buffer and serum. SPFS in combination with our biosensor allows sensitive analysis of CRP, achieving in buffer a LOD of 0.016 μg mL(-1) and a LOQ of 0.049 μg mL(-1). In serum the accomplished LOD was 0.026 μg mL(-1) and the LOQ was found to be 0.08 μg mL(-1). These low LODs and LOQs demonstrate the applicability of the designed biosensor for qualitative and semi-quantitative analysis of trace amounts of substances in very small sample volumes of body fluids.

The prognostic value of detection of serum C-reactive protein in the patients with stage I lung cancer

背景与目的急性炎症反应包括白细胞及急性期炎症蛋白的变化被较多研究证实是各种肿瘤包括非小细胞肺癌(non-small cell lung cancer), NSCLC的预后因素之一。本研究旨在观察Ⅰ期NSCLC患者术前C反应蛋白(C-reactive protein, CRP)水平与临床特征之间的关系及其预后意义。方法应用胶乳免疫透射比浊法计算血清CRP含量回顾性收集2000年1月-2004年1月间上海市长宁区中心医院及上海市胸科医院共96例完整手术切除的Ⅰ期NSCLC患者资料, 采用ANOVA检验分析不同临床特征的患者中CRP水平是否存在差异。采用Kaplan-Meier曲线及Logrank检验进行生存分析和比较, 并采用COX多因素回归分析与生存相关的因素。结果96例患者中CRP≤5 mg/L者66例。鳞癌(P < 0.001)、肿瘤直径 > 3 cm(P < 0.001)的患者CRP水平较高。多元线性回归分析提示肿瘤最大径与血清CRP水平相关(β=0.322, P=0.039)。CRP > 5 mg/L组患者的5年生存率低于CRP≤5 mg/L组(54.1% vs 78.2%, P=0.021)。COX分析表明CRP水平是影响Ⅰ期NSCLC患者总生存期的独立预后因素(P=0.023)。结论Ⅰ期NSCLC患者术前CRP水平与肿瘤最大径呈正相关CRP > 5 mg/L组患者的总生存期低于CRP≤5 mg/L组可能是影响患者预后的不良因素。

Evaluation of 2-methacryloyloxyethyl phosphorylcholine polymeric nanoparticle for immunoassay of C-reactive protein detection.

To prepare novel 2-methacryloyloxyethyl phosphorylcholine (MPC)-polymeric nanoparticle (MPC-PNP), water-soluble amphiphilic phospholipid polymer, poly [MPC-co-n-butyl methacrylate (BMA)-co-p-nitrophenyloxycarbonyl poly(ethylene glycol) methacrylate (MEONP) (PMBN)], which has active ester groups for bioconjugation on the side chains, was synthesized. MPC-PNP was prepared by a solvent evaporation technique where the poly(l-lactic acid) was used as core and PMBN was applied as an emulsifier and a surface modifier under systematical design of well-arranged phospholipids polar groups in its surface. Characteristics for MPC-PNP were thoroughly investigated with dynamic light scattering, electrophoresis light scattering, X-ray photoelectron spectroscopy, and field emission scanning electron microscopy measurements. Through a protein adsorption test, the phosphorylcholine group on the surface of MPC-PNPs, which had their active ester groups substituted by glycine, were shown to suppress the nonspecific adsorption of bovine serum albumin. These particles were used for C-reactive protein (CRP) detection, where anti-CRP monoclonal antibodies were immobilized on the MPC-PNP using the active ester group, while the remaining active ester groups were thoroughly reacted with glycine. The detection limit about serum-free CRP in the calibration curve was shown to extend from 0.01 to 10 mg/dL when anti-CRP antibody immobilized MPC-PNP was used for serum-free CRP detection. This compares favorably with measurement using polystyrene nanoparticles that were shown to detect from 0.1 to 10 mg/dL by an immunoagglutination technique. Also, for the detection of CRP in serum, MPC-PNP was shown to give the same calibration curve explained by the efficient suppression of nonspecific binding. Furthermore, denaturation of immobilizing anti-CRP antibody on the MPC-PNP hardly occurred despite increasing the temperature. It is concluded that MPC-PNP is unique due to the design of its interfacial properties, also it will perform well in a diagnostic immunoassay because of its optimized material properties.
