Ektacytometry: characterization of the erythrocytes of rats subjected to hypothermia and receiving natural zeolites

The article presents data as to the effect of a diet containing natural zeolite, taken from the Mysovsky mine (Khanty-Mansi Autonomous Okrug - Ugra), and applied in powdered form on the ability of erythrocytes of white outbred rats to change their shape in shear flow under normal and hypothermic conditions. To assess the deformability of erythrocytes, a laser diffraction technique combined with a redesigned version of the ektacytomer has been used in this work. For comparison, zeolite from the Kholinsky mine (Zabaykalsky Krai), which is well-studied and widely used for medicines and nutraceuticals has been provided. It has been shown that hypothermia causes a significant increase in the parameter such as erythrocyte deformability in rats in a cold group and is associated with enhanced lipid peroxidation when compared to animals in a control group. These alterations were a result of macrocytosis, revealed by flow cytometry analysis. Natural zeolite powder used in animal feeds had antioxidant and membrane-protective effects on animals in both groups. These included a significant increase in the parameter such as erythrocyte deformability with a decrease in lipid peroxidation processes and expansion of the Price-Jones curve to the left, unlike those observed in animals subjected to hypothermia but not receiving zeolites. The results obtained demonstrated that the biological effects of natural zeolite-containing preparation which contains a clinoptilolite component are similar and that zeolites from the Mysovsky mine can be used as a potent raw material for drugs.

Effect of Hydrogen Sulfide on Deformability of Rat Erythrocytes.

The influence of a hydrogen sulfide donor NaHS (2×10-5-10-3 M) on the rat erythrocyte deformability was analyzed by laser diffractometry. NaHS (6×10-5 M) increased, while NaHS (10-3 M) reduced erythrocyte deformability. The effect of NaHS (6×10-5 M) was similar to that of NO donor sodium nitroprusside (SNP, 10-7 M). However, simultaneous use of NaHS (6×10-5 M) and SNP induced less pronounced changes in erythrocyte deformability than their individual application. It is likely H2S, similar to NO, is involved in the regulation of erythrocyte deformability in the microvascular bed.

Effects of intravenous ibuprofen and lornoxicam on erythrocyte deformability in rats undergoing hind limb ischemia reperfusion injury.

Acute hind limb ischemia reperfusion (I/R) injury is a common consequence of abdominal aorta cross‑clamping during aortic surgery. Erythrocyte deformability is affected by I/R process and may lead to increased tissue and organ injury. Lornoxicam and intravenous ibuprofen are becoming commonly used as non-steroidal anti-inflammatory drugs (NSAID) for postoperative analgesia. In this study, we aimed to investigate the effects of lornoxicam (2 mg/kg iv) and intravenous ibuprofen (30 mg/kg iv) on erythrocyte deformability in I/R model in rats. Four study groups, each containing 6 Wistar rats were created. Laparotomy was performed in all groups under general anesthesia with ketamine and xylazine. In all groups except sham group, ischemia and reperfusion were achieved by clamping and declamping the infrarenal abdominal aorta for 120 minutes. Rats in Group IR+L received intravenous infusion of lornoxicam (2 mg/kg) while rats in Group IR+I received intravenous infusion of ibubrofen (30 mg/kg) following 2 hours of ischemic period. At the end of reperfusion period, erythrocyte packs were prepared from heparinized blood samples. Erythrocyte suspensions with hematocrit at a concentration of 5% in a phosphate‑buffered saline (PBS) were used in order to perform deformability measurements. The value of p<0.05 was considered statistically significant. Relative resistance has increased in ischemia reperfusion group when compared to control group (p < 0.0001). Lornoxicam or ibuprofen intravenous treatments did not change the erythrocyte deformability during ischemia reperfusion period in rats (p=0.851, p=0.690). Intravenous ibuprofen or lornoxicam administrations during ischemia reperfusion period in rats have no negative effect on erythrocyte deformability. The findings of the study should be supported with more detailed and extensive clinical/experimental studies in the future (Fig. 1, Ref. 18).

Obstructive nephropathy is a common cause of renal insufficiency in adults. Unilateral ureteral obstruction (UUO) is a well-established model known to imitate the process of obstructive nephropathy in a simple, accelerated and species-independent manner.

Investigation of the effects of propofol and vitamin C administration on erythrocyte deformability in rats with streptozotocin-induced diabetes mellitis.

In the current study we aim to investigate the effects of vitamin C and profol on red blood cell deformability in diabetic rats. Twenty- eight Wistar Albino rats were included in the study after streptozocin (60 mg/kg) treatment for 4 weeks of observation for diabetes presence. Twenty-eight rats were allocated to 4 groups. In group DP (n = 7) 150 mg.kg-1 of propofol was injected intraperitoneally. In group DP-vit C (n = 7) rats 100 mg/kg of vitamin C (Ascorbic acid, Redoxon® 1000 mg/5 mL - Roche) were applied one hour before administrating 150 mg.kg-1 of propofol, while rats in control group (n = 7), and diabetic control group (n = 7) received intraperitoneally physiological saline. Deformability measurements were achieved by using erythrocyte suspensions with hematocrit level of 5 % in PBS buffer. Erythrocyte deformability was significantly higher in diabetic control group than in control and vitamin C plus propofol groups (p = 0.00, p = 0.025, respectively). Erythrocyte deformability indexes were found similar in control group and vitamin C plus propofol group (p = 0.949). Relative resistance was increased in diabetic rat model. Erythrocyte deformability was damaged in rats with diabetes. This injury might lead to further problems in microcirculation. Application of propofol did not alter red cell deformability in diabetic rats. Vitamin C supplementation seems to reverse those negative effects and variations in erythrocyte deformability (Fig. 2, Ref. 57).

Simvastatin and GGTI-2133, a geranylgeranyl transferase inhibitor, increase erythrocyte deformability but reduce low O2tension-induced ATP release

Statin drugs inhibit 3-hydroxy-3-methylglutaryl CoA reductase, which reduces the synthesis of both cholesterol and isoprenoids (geranylgeranyl pyrophosphate and farnesyl pyrophosphate), with the latter being lipid molecules responsible for the posttranslational modification of small GTP-binding proteins such as Rho. Effects of statins, independent of lowering blood cholesterol levels, are thought to occur by inhibition of Rho/Rho kinase. The Rho kinase inhibitor Y-27632 has been reported to increase both erythrocyte deformability and low O2 tension-induced ATP release. Here, we tested the hypothesis that by inhibiting Rho/Rho kinase, simvastatin would increase both erythrocyte deformability and low O2 tension-induced ATP release. Male Sprague-Dawley rats were divided into two groups, control or simvastatin treated [simvastatin-supplemented chow (0.02%)], for 4 wk. Simvastatin treatment increased rat erythrocyte deformability compared with controls (n = 6, P < 0.05). However, erythrocytes of simvastatin-treated rats (n = 9, P < 0.05) exhibited impaired low O2 tension-induced ATP release. Similarly, the geranylgeranyl transferase inhibitor GGTI-2133 (10 μM) also increased deformability and impaired low O2 tension-induced ATP release in healthy human erythrocytes (P < 0.05). Interestingly, ATP release in response to mastoparan 7 (n = 7, P < 0.05), which directly activates Gi, and isoproterenol (n = 5, P < 0.05), which signals through Gs, was not altered by incubation with GGTI-2133. These results suggest that although statins increase erythrocyte deformability, likely by inhibiting geranylgeranylation, the finding that both statins and a geranylgeranyl transferase inhibitor attenuated low O2 tension-induced ATP release demonstrates that factors in addition to erythrocyte deformability are critical for ATP release in response to this physiological stimulus.

Mechanism of endurance training-induced erythrocyte deformability in rats involves erythropoiesis

Higher erythrocyte deformability may reduce the risk of circulatory diseases by enhancing oxygen delivery and reducing the load on the cardiovascular system. The effect of endurance training on erythrocyte deformability is not clear. This study explored the impact of endurance training on erythrocyte deformation and shape and investigated the underlying mechanisms of hemorheological alterations. Forty male Wistar rats were randomly divided into two groups: sedentary (S; n = 21) and exercised (E; n = 19). Hematological indices and erythrocyte shape were measured at the end of the 11th week. The gene expression of erythropoietin (Epo) and the Epo receptor (EpoR) was quantified using reverse transcription-PCR (RT-PCR), and Epo protein expression was analyzed using Western blotting. Endurance training significantly decreased the abnormality ratio of erythrocyte shape (P < 0.01). The deformability indicator (DI) of red blood cells was lower in the E group than in the S group (P < 0.01). Eleven weeks of endurance training increased Epo mRNA and protein expression in the kidney (P < 0.01), EpoR mRNA expression in the bone marrow (P < 0.05), and relative circulating Epo (P < 0.01) compared to the sedentary group. The decrease in the erythrocyte morphological index and the maximum of deformability indicator were associated with an increase in relative circulating Epo. In conclusion, 11 weeks of endurance training increased erythrocyte deformability. Epo and EpoR may contribute to the decreased morphological index and deformability indicator in erythrocytes during endurance training in rats.

The effects of low-high doses of dexmedetomidine on erythrocyte deformability in rats

Dexmedetomidine is an anesthetic agent frequently used for sedation, intensive care units, and general anesthesia. The purpose of our study was to investigate the effect of two different doses of dexmedetomidine on erythrocyte deformability in rats. The study was performed on 21 male rats, with 7 rats in each study group and the control group. The rats in the study groups were administered dexmedetomidine (low dose 5 µg.kg-, high dose 10 µg.kg-) intraperitoneally, and the rats in the control group were administered physiological saline. Erythrocyte packs were prepared using heparinized total blood samples. Deformability measurements were done by erythrocyte suspensions in phosphate buffered saline (PBS) buffer. A constant flow filtrometer system was used to measure erythrocyte deformability, and the relative resistance was calculated. Use of a high dose dexmedetomidine resulted in an increase in relative resistance, which is an indicator for erythrocyte deformability in control rats (p=0.014). High dose dexmedetomidine via negative change in erythrocyte deformability may cause a functional deterioration in blood flow and tissue perfusion. Our results showed that low dose dexmedetomidine protects erythrocyte deformability better than the high dose (Fig. 1, Ref. 23).

The evaluation of effects of lornoxicam on blood flow and erythrocyte deformability in comparison with iv paracetamol in rats

Lornoxicam and iv paracetamol are commonly preferred to be used for postoperative analgesia. Although Aspirin is a well known non-steroid anti-inflammatory drug that decreases the erythrocyte deformability, there is no study comparing lornoxicam and iv paracetamol regarding their effects on erythrocyte deformability recorded in literature. The aim of this study was to compare the effects of lornoxicam and IV paracetamol on blood perfusion and erythrocyte deformability on rats. Twenty male Wistar Albino rats were randomly divided into three groups as Lornoxicam group (Group L), IV paracetamol group (Group P), and control group (Group C). Intraperitoneal administrations were done in all groups except Group C. Liver and renal blood flows were conducted by laser Doppler and euthanasia was performed via intraabdominal blood uptake. Erythrocyte deformability was measured using a constant flow filtrometer system. Lornoxicam increased the relative resistance which shows the erythrocyte deformability in rats (p<0.05). The comparison of Groups C and P revealed no statistically different results (p=0.731) while Group L revealed statistically higher results than Group C (p=0.022). No statistically significant differences were found between groups L and P (p=0.073). Liver and renal blood flow values in Group L were just numerically decreased not statistically whilst no statistically significant difference was found between the three groups. Lornoxicam have lead to functional disorders related to tissue perfusion as a result of both decreased blood flow and erythrocyte deformability (Fig. 3, Ref. 21). Full Text in PDF www.elis.sk.

Does vitamin C prevent the effects of high dose dexmedetomidine on rat erythrocyte deformability?

Dexmedetomidine is an anesthetic agent frequently used for sedation at the intensive care units and during general anesthesia. The purpose of our study was to investigate whether vitamin C prevents the effect of high dose dexmedetomidine on erythrocyte deformability in rats. The study was performed on 21 male rats, with 7 rats in each study groups and the control group. The rats in the study groups were treated with intraperitoneal dexmedetomidine (10 µg/kg) and intraperitoneal dexmedetomidine plus Vitamin C (ascorbic acid) (100 mg/kg ascorbic acid administered 1 hour before administration of 10 µg/kg dexmedetomidine), respectively. Intraperitoneal physiological saline was administered in the control group. Erythrocyte packs were prepared using heparinized total blood samples. Deformability measurements were done by erythrocyte suspensions in phosphate buffered saline (PBS) buffer. A constant flow filtrometer system was used to measure erythrocyte deformability and the relative resistance was calculated. Erythrocyte deformability was significantly higher in dexmedetomidine group than in control and vitamin C plus dexmedetomidine groups (p=0.003, p=0.013, respectively). Erythrocyte deformability indexes were found similar in the control group and in the vitamin C plus dexmedetomidine group (p=0.383). High dose dexmedetomidine may cause functional deterioration in blood flow and tissue perfusion with negative effects in erythrocyte deformability. Vitamin C supplementation seems to reverse those negative effects and variations in erythrocyte deformability. However, our preliminary results should be confirmed in wider serious of experimental and clinical trials (Fig. 1, Ref. 27).

Effects of Cold Water Swimming on Blood Rheological Properties and Composition of Fatty Acids in Erythrocyte Membranes of Untrained Older Rats

This is the first report on the effects of a single bout of swimming to exhaustion in cold water on rat erythrocyte deformability, aggregation and fatty acid composition in erythrocyte membranes. The results indicate that there was a significant decrease in body temperature of experimental rats swimming in water at 4 degrees C and 25 degrees C when compared to the control. Erythrocyte aggregation indices did not change after swimming in water at 4 degrees C whereas erythrocyte deformability increased at shear stress 1,13 [Pa] and 15,96 [Pa]. Physical effort performed in water at 4 degrees C when compared to the control group resulted in an increase in monounsaturated and polyunsaturated n-3 fatty acid content in erythrocyte membranes that influenced the increase in their fluidity and permeability even though that of polyunsaturated n-6 fatty acids decreased. Physical effort performed in 25 degrees C water resulted in an increase in saturated fatty acid content and a decrease in all polyunsaturated fatty acids and polyunsaturated n-6 fatty acids when compared to the control group. Swimming of untrained old rats in cold water affected rheological properties oferythrocytes in a negligible way while changes in the fatty acid composition of erythrocyte membranes were more pronounced.

Reduction of erythrocyte deformability in rats with cerebral ischemia.

Erythrocyte deformability was studied by laser interferometry in rats with cerebral ischemia. Ninety minutes after ligation of both common carotid arteries the erythrocyte deformability coefficient decreased by 11+/-2% in experimental group in comparison with the baseline level and by 12% in comparison with sham-operated animals and remained 16% decreased after 4 days.

Pharmacological studies of Reiousan, a drug containing bezoar and ginseng--effects on the blood rheology

Effects of Reiousan, a crude drug preparation consisting of bezoar and ginseng, on blood rheology were studied. Reiousan improved the deformability of rat erythrocytes exposed to hyperosmorality and treated with phenylhydrazine. The ATP depletion in erythrocytes, the polybrene-induced erythrocyte aggregation and the oxidation of low density lipoprotein were suppressed by Reiousan. Oral administration of Reiousan also improved the erythrocyte deformability in phenylhydrazine-treated rats and delayed the thromboembolic death induced by arachidonic acid in mice. These results suggest that Reiousan has an ameliorative effect on blood rheology related to "Oketsu" syndrome in Kampo diagnostics.

Hemolysis and deformability of erythrocytes exposed to butoxyacetic acid, a metabolite of 2-butoxyethanol: I. Sensitivity in rats and resistance in normal humans.

The effects of butoxyacetic acid (BAA), a metabolite of the important solvent 2-butoxyethanol, on rat and human red blood cells (RBCs) were investigated. Rat RBCs demonstrated decreased deformability as assessed by a sensitive polycarbonate sieve filtration technique and an increased mean cellular volume after incubation with 0.2 and 2.0 mM BAA for 1-4 h. Evaluation of erythrocyte morphology showed that rat RBCs exposed to BAA became spherocytic and lysed to form erythrocyte membrane ghosts. Hemolysis of rat erythrocytes was rapid in 2.0 mM BAA. Changes in the deformability of rat erythrocytes appear to precede hemolysis. Treated rat erythrocytes also demonstrated a tendency to agglutinate and to release hemoglobin, which formed visible precipitates. None of the adverse effects seen in the RBCs of rats were observed in human blood similarly incubated with 2.0 mM BAA.

Effects of dietary tea catechins on alpha-tocopherol levels, lipid peroxidation, and erythrocyte deformability in rats fed on high palm oil and perilla oil diets.

The effects of dietary tea catechins on the levels of alpha-tocopherol and lipid peroxidation in both plasma and erythrocytes, as well as their effects on erythrocyte deformability, were examined in rats fed on high palm and perilla oil diets. The decrease in alpha-tocopherol concentration and the increase in lipid peroxidation level were much more pronounced in the perilla oil group than in the palm oil group. The addition of tea catechins to these diets significantly prevented the alpha-tocopherol concentration from decreasing. These results suggest that tea catechins may counteract a decrease in alpha-tocopherol by acting as an antioxidant in vivo. Furthermore, the lipid peroxidation in the plasma of rats fed perilla oil was slightly but significantly reduced by the supplemented tea catechins. However, no measurable differences were observed in the deformability of the erythrocytes in any of the groups. It is therefore likely that the erythrocytes are not severely enough affected by the lipid peroxidation to influence their deformability.

The effect of macrocytosis on rat erythrocyte deformability during recovery from phenylhydrazine-induced anemia

To investigate the role of red blood cell (RBC) geometry in determining the filterability of stress-induced macrocytes, measurements of RBC volume, diameter, and passage time through 3 micron and 5 micron filters were made on RBCs from control rats and from phenylhydrazine-treated rats during a 28-day recovery period following the peak hemolytic response to the anemia induced by phenylhydrazine (PHZ). Mean cell volumes (MCV) were calculated from volume distribution curves; RBC diameters were obtained from peripheral blood smears; surface area (SA) and mean cylindrical diameter (MCD) were calculated using a biconcave erythrocyte model. At the time of the peak macroreticulocyte response to PHZ, MCD and MCV were significantly increased compared to controls (3.07 vs 2.57 microns, and 102.1 vs 59.7 cubic microns, respectively), the ratio SA/MCV was significantly reduced (1.405 vs 1.670), and RBC filterability through both 3 and 5 micron cylindrical pores was significantly reduced. During the 28-day recovery period, MCV, MCD, SA/MCV, and filterability all returned toward the control values, MCD was inversely correlated with filterability through both 3 micron (r = -0.861, p less than .01) and 5 micron (r = -0.767, p less than .01) pores, and MCD fell below 3.6 microns in 97.5% of the animals despite the persistence of large macrocytic subpopulations. These results emphasize the correlation between RBC geometry and filterability, and demonstrate that macrocytes with favorable geometry can persist in the circulation for extended periods without remodeling or reduction in size.

Effects of Diltiazem on the Physicochemical Properties of Rat Erythrocyte and Liposome Membrane: Comparison with Pentoxifylline and Propranolol

In vitro effects of the coronary vasodilator diltiazem on rat erythrocytes and liposomes were studied in comparison with propranolol and pentoxifylline. Diltiazem improved the deformability of rat erythrocytes, reduced the viscosity of rat erythrocyte suspensions, and protected the erythrocyte against hypotonic hemolysis at concentrations above 10−4 M, 10−5 M and 5×10−7 M, respectively. Diltiazem at 5×10−4 M also improved the impaired deformability of ATP-depleted erythrocytes, whereas it affected neither the adenine nucleotide level nor the phosphorylation of spectrin in the erythrocytes. Diltiazem enhanced the interaction of 1-anilino-naphthalene-8-sulfonate, a fluorescent probe, with erythrocyte ghosts at concentrations above 5×10−6 M and, above 5×10−5 M, inhibited the (Na+ + K+)-ATPase activity of the erythrocyte ghosts. Diltiazem reduced the microviscosity of both erythrocyte ghosts and liposomes prepared from rat erythrocyte lipids. Diltiazem induced aggregation of the liposomes prepared from rat erythrocyte lipids, phos-phatidylserine or phosphatidylinositol, but it did not affect the liposomes prepared from a mixture of phosphatidylethanolamine and phosphatidylcholine (1:1). /-Diltiazem, a stereo-isomer of diltiazem, exhibited equipotent effects, compared with diltiazem, on these parameters. Propranolol showed similiar properties, but pentoxifylline shared none of the above properties, except that it improved the deformability of erythrocytes. From these results, it is suggested that diltiazem may affect the erythrocyte membrane by interacting with acidic phospholipids and thus reduce the microviscosity of the membrane, improve erythrocyte deformability and protects the erythrocyte against hypotonic hemolysis.
