Coated oregano essential oil and cinnamaldehyde compounds supplementation improves growth performance, enhances immune responses and inhibits cecal Escherichia coli proliferation of broilers.

Plant essential oils are unstable due to high volatility and easy oxidation, while microencapsulation provides a potentially effective strategy for increasing the stability of natural essential oils and preserving their function. This study examined the effects of feeding coated oregano essential oil and cinnamaldehyde (COEC) compounds on growth, immune organ development, intestinal morphology, mucosal immune function, and the cecal microbiota populations of broilers. Three hundred one-day-old male Arbor Acres broiler chicks were organized into five groups: (1) negative control fed basal diet alone (NC), (2) positive control receiving basal diet plus 50mg/kg of chlortetracycline (CTC), (3) basal diet plus 150mg/kg COEC (COEC150), (4) plus 300mg/kg COEC (COEC300), and (5) plus 450mg/kg COEC (COEC450). The supplement trial was continued for 42 days. The results showed that CTC, COEC300 and COEC450 treatments decreased the feed conversion ratio of broilers both in the starter and whole experiment phases, increased the height of jejunal villi at 21 d and the number of goblet cells and IgA-producing cells at 21 or 42 d compared with NC group (P < 0.05). Members of the COEC300 treatment group had a higher thymus weight index and jejunum length index than birds of NC or CTC groups at 21 d (P < 0.05). CTC and all COEC treatments decreased malondialdehyde content in jejunal mucosa at 42 d (P < 0.05). The population of Escherichia coli in the cecal digesta at 21 d was lower in the CTC, COEC300 and COEC450 treatment groups compared with the NC group (P < 0.05). In contrast to the CTC group, COEC supplementation dose-dependently accelerated body weight gain, improved jejunal morphology, decreased malondialdehyde content in jejunal mucosa, increased numbers of jejunal goblet cells and IgA-producing cells, and decreased the Escherichia coli population in cecal digesta at 21 or 42 d (P < 0.05). Thus, we concluded that feeding broiler chickens with 300 or 450mg/kg in antibiotic-free diets can improve growth performance, enhance immune responses and inhibit the proliferation of cecal pathogenic bacteria.

Mitigation strategy of saline stress in Fragaria vesca using natural and synthetic brassinosteroids as biostimulants.

Bassinosteroids (BRs) can induce plant defence responses and promote plant growth. In this work, we evaluated the effect of a natural (EP24) and a synthetic (BB16) brassinosteroid on strawberry (Fragaria vesca ) plants exposed to saline stress. Treated plants showed higher shoot dry weight and root growth compared to untreated control plants. In BR-treated plants, crown diameters increased 66% and 40%, leaf area 148% and 112%, relative water content in leaves 84% and 61%, and SPAD values 24% and 26%, in response to BB16 and EP24, respectively. A marked stomatal closure, increased leaflet lignification, and a decrease in cortex thickness, root diameter and stele radius were also observed in treated plants. Treatments also reduces stress-induced damage, as plants showed a 34% decrease in malondialdehyde content and a lower proline content compared to control plants. A 22% and 15% increase in ascorbate peroxidase and total phenolic compound activities was observed in response to BB16, and a 24% increase in total flavonoid compound in response to both BRs, under stress conditions. These results allow us to propose the use of BRs as an environmentally safe crop management strategy to overcome salinity situations that severely affect crop yield.

Novel Insights into Ethanol-Soluble Oyster Peptide-Zinc-Chelating Agents: Structural Characterization, Chelation Mechanism, and Potential Protection on MEHP-Induced Leydig Cells.

Numerous studies have reported that mono-(2-ethylhexyl) phthalate (MEHP) (bioactive metabolite of Di(2-ethylhexyl) phthalate) has inhibitory effects on Leydig cells. This study aims to prepare an oyster peptide-zinc complex (PEP-Zn) to alleviate MEHP-induced damage in Leydig cells. Zinc-binding peptides were obtained through the following processes: zinc-immobilized affinity chromatography (IMAC-Zn2+), liquid chromatography-mass spectrometry technology (LC-MS/MS) analysis, molecular docking, molecular dynamic simulation, and structural characterization. Then, the Zn-binding peptide (PEP) named Glu-His-Ala-Pro-Asn-His-Asp-Asn-Pro-Gly-Asp-Leu (EHAPNHDNPGDL) was identified. EHAPNHDNPGDL showed the highest zinc-chelating ability of 49.74 ± 1.44%, which was higher than that of the ethanol-soluble oyster peptides (27.50 ± 0.41%). In the EHAPNHDNPGDL-Zn complex, Asn-5, Asp-7, Asn-8, His-2, and Asp-11 played an important role in binding to the zinc ion. Additionally, EHAPNHDNPGDL-Zn was found to increase the cell viability, significantly increase the relative activity of antioxidant enzymes and testosterone content, and decrease malondialdehyde (MDA) content in MEHP-induced TM3 cells. The results also indicated that EHAPNHDNPGDL-Zn could alleviate MEHP-induced apoptosis by reducing the protein level of p53, p21, and Bax, and increasing the protein level of Bcl-2. These results indicate that the zinc-chelating peptides derived from oyster peptides could be used as a potential dietary zinc supplement.

Combination of Exogenous Spermidine and Phosphocreatine Efficiently Improved the Quality and Antioxidant Capacity of Cryopreserved Boar Sperm and Reduced Apoptosis-Like Changes.

The low resistance of boar sperm to cryopreservation dictates that addition antioxidants and energetic substances to the diluent to improve sperm quality is necessary. This study evaluated the effect of spermidine and phosphocreatine in combination on the quality, antioxidant capacity, and antiapoptotic-like changes capacity of cryopreserved boar sperm based on previous reports. The results showed that the combined application of spermidine and phosphocreatine significantly enhanced the motility, average path velocity, straight-line velocity, curvilinear velocity, beat cross frequency, acrosome integrity, plasma membrane integrity, mitochondrial activity, and DNA integrity compared with the control group (p < 0.05). In addition, the combined application of spermidine and phosphocreatine significantly enhanced the total antioxidant capacity, superoxide dismutase activity, glutathione peroxidase activity, and catalase activity while significantly decreasing malondialdehyde content and hydrogen peroxide content (p < 0.05). Western Blot analysis further showed that spermidine and phosphocreatine significantly decreased the expression of CASP3 and BAX and significantly enhanced the expression of BCL2 (p < 0.05); therefore, the combination of spermidine and phosphocreatine has potentially positive implications for improving the quality of cryopreserved boar sperm.

Schisandrol A Alleviates Allergic Asthma in Mice via Regulating the NF-κB/IκBα and Nrf2/HO-1 Signaling Pathways.

Schisandra chinensis (Turcz) Baill (S. chinensis) is the key traditional Chinese medicine for the treatment of asthma used by ancient and modern medical practitioners. However, the material basis and the main mechanism of its antiasthmatic effect remain unclear. Our preliminary results showed that schisandrol A (SCA), a representative monomer of Schisandra lignans, had the best relaxation effect on tracheal rings in isolated rats. In this research, a mouse asthma model was prepared by combining ovalbumin (OVA) with Al (OH)3 for exploring the antiasthmatic action and the underlying mechanism of SCA. The study results demonstrated that SCA improved the behavior of mice with asthma and pathological changes in their lung tissues and airways, decreased serum immunoglobulin E (IgE) and OVA-IgE levels, interleukin-4 (IL-4), IL-5, IL-13, and eotaxin contents, and leukocytes number in bronchoalveolar lavage fluid. SCA downregulated the gene expressions of keratinocyte-derived protein chemokines and ILs and reduced the expressions of phosphorylated IκB kinase α (p-IKKα) and p-nuclear factor kappa-B (NF-κB) proteins in lung tissues. In addition, it was found that SCA could significantly increase T-superoxide dismutase and catalase activities, decrease malondialdehyde content, and elevate p-IκBα, NF-E2-related-factor 2 (Nrf2), and heme oxygenase-1 (HO-1) protein expressions. In summary, SCA treatment resulted in a significant improvement in the allergic bronchial asthma in mice, and its mechanisms may involve the regulation of the NF-κB/IκBα pathway to reduce inflammatory response and the Nrf2/HO-1 pathway to improve the body's antioxidant capacity. These results suggest that SCA is a key component of S. chinensis in exerting antiasthmatic effects.

Retinal ischemia-reperfusion injury and pretreatment with Lycium barbarum glycopeptide.

To investigate the antioxidant protective effect of Lycium barbarum glycopeptide (LbGP) pretreatment on retinal ischemia-reperfusion (I/R) injury (RIRI) in rats. RIRI was induced in Sprague Dawley rats through anterior chamber perfusion, and pretreatment involved administering LbGP via gavage for 7d. After 24h of reperfusion, serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and creatinine (CREA) levels, retinal structure, expression of Caspase-3 and Caspase-8, superoxide dismutase (SOD) activity, and malondialdehyde (MDA) in the retina were measured. The pretreatment with LbGP effectively protected the retina and retinal tissue from edema and inflammation in the ganglion cell layer (GCL) and nerve fiber layer (NFL) of rats subjected to RIRI, as shown by light microscopy and optical coherence tomography (OCT). Serum AST was higher in the model group than in the blank group (P=0.042), but no difference was found in ALT, AST, and CREA across the LbGP groups and model group. Caspase-3 expression was higher in the model group than in the blank group (P=0.006), but no difference was found among LbGP groups and the model group. Caspase-8 expression was higher in the model group than in the blank group (P=0.000), and lower in the 400 mg/kg LbGP group than in the model group (P=0.016). SOD activity was lower in the model group than in the blank group (P=0.001), and the decrease was slower in the 400 mg/kg LbGP group than in the model group (P=0.003). MDA content was higher in the model group than in the blank group (P=0.001), and lower in the 400 mg/kg LbGP group than in the model group (P=0.016). The pretreatment with LbGP did not result in any observed liver or renal toxicity in the model. LbGP pretreatment exhibits dose-dependent anti-inflammatory, and antioxidative effects by reducing Caspase-8 expression, preventing declines of SOD activity, and decreasing MDA content in the RIRI rat model.

Low voltage electrostatic field combined with ice-temperature to improve the quality of litchi during storage

Litchi is popular among consumers due to its delicious taste, however, the extremely short shelf life limits its commercial value. Few studies have been conducted to develop new technologies to extend the shelf life of litchi. Therefore, this study applied a novel technique (low voltage electrostatic field, LVEF) combined with ice-temperature (0 °C) treatment on litchi and evaluated its quality characteristics during storage. The results demonstrated that low voltage electrostatic field with ice-temperature storage (LVEF-IT) decreased malondialdehyde content, relative electrolyte leakage, and polyphenol oxidase and peroxidase activities, successfully delayed the start of peel browning. Additionally, it preserved the fruit pulp’s excellent quality, which included high concentrations of soluble solids, ascorbic acid, and soluble protein. These findings imply that LVEF-IT treatment may be a novel technique for extending the storage time of litchi.

Para-coumaric acid and cinnamic acid enhance resistance of Agaricus bisporus mushrooms to Brown blotch disease caused by Pseudomonas tolaasii

The effects of postharvest pre-treatments with para-coumaric acid (PCA) and cinnamic acid (CA) against brown blotch disease in Agaricus bisporus mushrooms caused by Pseudomonas tolaasii were investigated. PCA and CA at the concentration of 300 mg L−1 provided the highest inhibition ratio of 28.9% and 20.9%, respectively, on the in vitro growth of P. tolaasii. In addition, PCA and CA pre-treatments dramatically reduced the P. tolaasii-caused brown blotch disease in postharvest A. bisporus. Biochemical analysis revealed that PCA and CA inhibited P. tolaasii-induced activation of polyphenol oxidase and melanin production, increased activities of β-N-acetylglucosaminidase, β-1,3-glucanase and chitinase, promoted phenylpropanoid biosynthetic enzyme activities and metabolite production, enhanced antioxidant enzyme activities, and decreased malondialdehyde content in A. bisporus mushrooms. Collectively, these results demonstrated that PCA and CA can induce resistance to postharvest brown blotch disease in A. bisporus mushrooms, and have good prospects for disease control in edible mushrooms.

Effects of Bacillus subtilis on cotton physiology and growth under water and salt stress

The scarcity of fresh water resources has severely limited agricultural production in arid areas. Although brackish water irrigation or fresh water deficit irrigation can alleviate the water resources crisis, both may cause water and salt stress to crop. Therefore, this study is based on the functional advantages of Bacillus subtilis in soil improvement and crop growth promotion to alleviate water and salt stress and build safe and efficient water-saving irrigation patterns. In this study, cotton (No. 50 Chuangmian) was selected as the research crop, and five application rates of Bacillus subtilis (0, 22.5, 45, 67.5 and 90 kg·ha−1) were combined with three irrigation patterns (brackish water, fresh water and fresh water deficit irrigations) to study the effects of Bacillus subtilis on soil moisture and salinity, soil microbial community, cotton physiology and growth under water and salt stress. The results showed that Bacillus subtilis could enhance soil water retention capacity, promote soil desalination, improve cotton growth indices (plant height, stem diameter, leaf area index, dry matter accumulation), and then increase yield and water use efficiency (WUE). Compared with the control treatment, the yield and WUE of Bacillus subtilis application treatments increased by 3.32–54.67 % and 1.68–41.07 %, respectively. In the cotton physiology characteristics, Bacillus subtilis increased proline content and the activity of superoxide dismutase, peroxidase and catalase while decreased malondialdehyde content in cotton leaves. Bacillus subtilis could enhance the relative abundance of bacteria with the functions of nitrogen fixation, stress resistance and biocontrol. A structural equation model proved that Bacillus subtilis could improve yield and WUE indirectly by directly improving soil microbial diversity, alleviating water and salt stress, and then improving cotton physiology and growth. According to a comprehensive evaluation of cotton physiology and growth, it was determined that the optimal improvement effect was achieved when the application rate of Bacillus subtilis was 45 kg ha−1; the synergistic effect of brackish water irrigation and Bacillus subtilis (45 kg·ha−1) was superior to that of fresh water deficit irrigation combining with Bacillus subtilis (45 kg·ha−1), which could be considered a priority strategy for alleviating the fresh water crisis in arid areas and promoting the efficient increase in cotton yield.

Safflower Yellow Injection Alleviates Myocardial Ischemia/Reperfusion Injury by Reducing Oxidative and Endoplasmic Reticulum Stress.

Safflower yellow is an extract of the famous Chinese medicine Carthamus tinctorious L, and safflower yellow injection (SYI) is widely used clinically to treat angina pectoris. However, there are few studies on the anti-myocardial ischemia/reperfusion (I/R) injury effect of SYI, and its mechanisms are unclear. In the present study, we aimed to investigate the protective effect of SYI on myocardial I/R injury and explore its underlying mechanisms. Male Sprague Dawley rats were randomly divided into a control group, sham group, model group, and SYI group (20 mg/kg, femoral vein injection 1 h before modeling). The left anterior descending coronary artery was ligated to establish a myocardial I/R model. H9c2 cells were exposed to oxygen-glucose deprivation/reoxygenation (OGD/R) after incubation with 80 μg/mL SYI for 24 h. In vivo, TsTC, HE, and TUNEL staining were performed to evaluate myocardial injury and apoptosis. A kit was used to detect superoxide dismutase (SOD) and malondialdehyde (MDA) to assess oxidative stress. In vitro, flow cytometry was used to detect the reactive oxygen species (ROS) content and apoptosis rate. Protein levels were determined via Western blotting. Pretreatment with SYI significantly reduced infarct size and pathological damage in rat hearts and suppressed cardiomyocyte apoptosis in vivo and in vitro. In addition, SYI inhibited oxidative stress by increasing SOD activity and decreasing MDA content and ROS production. Myocardial I/R and OGD/R activate endoplasmic reticulum (ER) stress, as evidenced by increased expression of activating transcription factor 6 (ATF6), glucose-regulated protein 78 (GRP78), cysteinyl aspartate-specific proteinase caspase-12, and C/EBP-homologous protein (CHOP), which were all inhibited by SYI. SYI ameliorated myocardial I/R injury by attenuating apoptosis, oxidative damage, and ER stress, which revealed new mechanistic insights into its application.

Effects of Low-Temperature Stress on Physiological Characteristics and Microstructure of Stems and Leaves of Pinus massoniana L.

Pinus massoniana L. is one of the most important conifer species in southern China and is the mainstay of the forest ecosystem and timber production, yet low temperatures limit its growth and geographical distribution. This study used 30-day-old seedlings from families of varying cold-tolerance to examine the morphological traits of needles and stems, chlorophyll fluorescence characteristics, protective enzymes, and changes in starch and lignin under different low-temperature stresses in an artificial climate chamber. The results showed that the seedlings of Pinus massoniana exhibited changes in phenotypic morphology and tissue structure under low-temperature stress. Physiological and biochemical indexes such as protective enzymes, osmoregulatory substances, starch, and lignin responded to low-temperature stress. The cold-tolerant family increased soluble sugars, starch grain, and lignin content as well as peroxidase activity, and decreased malondialdehyde content by increasing the levels of actual photochemical efficiency (ΦPSII), electron transport rate (ETR), and photochemical quenching (qP) to improve the cold tolerance ability. This study provides a reference for the selection and breeding of cold-tolerant genetic resources of Pinus massoniana and the mechanism of cold-tolerance, as well as the analysis of the mechanism of adaptation of Pinus massoniana in different climatic regions of China.

Functional analysis of maize GRAS transcription factor gene ZmGRAS72 in response to drought and salt stresses

Abiotic stresses, such as drought and salt, are major factors affecting plant growth, development, and productivity. The GRAS gene family is a class of transcriptional regulators in plants that influence plant responses to various biotic and abiotic stresses. In this study, we cloned the maize (Zea mays L.) GRAS gene ZmGRAS72 and preliminarily analyzed its biological function. ZmGRAS72 was highly expressed in maize stems and young leaves, and was induced by abiotic stress and phytohormone treatments. Transient expression assays of maize protoplasts showed that ZmGRAS72 was localized to the nucleus. Heterologous expression of ZmGRAS72 in A. thaliana significantly improved plant tolerance to drought and salt stresses, increased chlorophyll content, decreased malondialdehyde content, and enhanced peroxidase activity. In addition, heterologous expression of ZmGRAS72 in A. thaliana upregulated or downregulated the expression levels of abscisic acid biosynthesis genes (NCED3), signaling genes (ABI1, ABI2, ABI4, and ABI5), and stress-related genes (RD22, RD29A, and KIN1) under abiotic stress. These results indicate that ZmGRAS72 may be responsive to abiotic stress, which forms a basis for further research on the mechanisms underlying the action of ZmGRAS72 in maize.

Untargeted metabolomics reveals the mechanism of ultrasound combined with L-cysteine treatment inhibiting the browning of fresh-cut potatoes

Freshly cut potatoes tend to brown during processing and preservation, which is an important issue worldwide. Ultrasound (US) combined with L-cysteine (L-Cys) treatment (US-L-Cys) has been reported to inhibit the browning of fresh-cut potatoes in our laboratory. However, the underlying mechanism remains to be elucidated. Untargeted metabolomics, a popular high-throughput technique, has been used to investigate the biochemical changes that are closely associated with the browning of fresh-cut fruits and vegetables. The present study investigated the effect of US-L-Cys treatment on the browning index (BI), related enzyme activities, total phenolic content, antioxidant capacity, malondialdehyde (MDA) content, and relative electrical conductivity of fresh-cut potatoes during 6-d storage. Metabolomics was used to compare the metabolites of US-L-Cys-treated and control samples on days 0 and 6. The results showed that US-L-Cys treatment reduced BI; decreased polyphenol oxidase, peroxidase, and phenylalanine ammonia-lyase activities; maintained high total phenolic content; increased antioxidant capacity; and decreased MDA content and relative electrical conductivity in fresh-cut potatoes during storage. Metabolomic analysis revealed that tyrosine and amino acid metabolisms were enriched. Higher levels of alanine and lower levels of dopamine were observed in US-L-Cys-treated samples compared with control samples. Therefore, depressed tyrosine and activated alanine metabolisms may be involved in the mechanism through which US-L-Cys inhibits the enzymatic browning of fresh-cut potatoes during storage.

Genome-wide identification and expression analysis of the BZR gene family in Zanthoxylum armatum DC and functional analysis of ZaBZR1 in drought tolerance.

In this study, six ZaBZRs were identified in Zanthoxylum armatum DC, and all the ZaBZRs were upregulated by abscisic acid (ABA) and drought. Overexpression of ZaBZR1 enhanced the drought tolerance of transgenic Nicotiana benthamian. Brassinosteroids (BRs) are a pivotal class of sterol hormones in plants that play a crucial role in plant growth and development. BZR (brassinazole resistant) is a crucial transcription factor in the signal transduction pathway of BRs. However, the BZR gene family members have not yet been identified in Zanthoxylum armatum DC. In this study, six members of the ZaBZR family were identified by bioinformatic methods. All six ZaBZRs exhibited multiple phosphorylation sites. Phylogenetic and collinearity analyses revealed a closest relationship between ZaBZRs and ZbBZRs located on the B subgenomes. Expression analysis revealed tissue-specific expression patterns of ZaBZRs in Z. armatum, and their promoter regions contained cis-acting elements associated with hormone response and stress induction. Additionally, all six ZaBZRs showed upregulation upon treatment after abscisic acid (ABA) and polyethylene glycol (PEG), indicating their participation in drought response. Subsequently, we conducted an extensive investigation of ZaBZR1. ZaBZR1 showed the highest expression in the root, followed by the stem and terminal bud. Subcellular localization analysis revealed that ZaBZR1 is present in the cytoplasm and nucleus. Overexpression of ZaBZR1 in transgenic Nicotiana benthamiana improved seed germination rate and root growth under drought conditions, reducing water loss rates compared to wild-type plants. Furthermore, ZaBZR1 increased proline content (PRO) and decreased malondialdehyde content (MDA), indicating improved tolerance to drought-induced oxidative stress. The transgenic plants also showed a reduced accumulation of reactive oxygen species. Importantly, ZaBZR1 up-regulated the expression of drought-related genes such as NbP5CS1, NbDREB2A, and NbWRKY44. These findings highlight the potential of ZaBZR1 as a candidate gene for enhancing drought resistance in transgenic N. benthamiana and provide insight into the function of ZaBZRs in Z. armatum.

Antihypertensive effects of whey protein hydrolysate involve reshaping the gut microbiome in spontaneously hypertension rats

Novel angiotensin-converting enzyme (ACE) inhibitory peptides were identified from whey protein hydrolysates (WPH) in vitro in our previous study and the antihypertensive abilities of WPH in vivo were further investigated in the current study. Results indicated that WPH significantly inhibited the development of high blood pressure and tissue injuries caused by hypertension. WPH inhibited ACE activity (20.81 %, P < 0.01), and reduced renin concentration (P < 0.05), thereby reducing systolic blood pressure (SBP) (12.63 %, P < 0.05) in spontaneously hypertensive rats. The increased Akkermansia, Bacteroides, and Lactobacillus abundance promoted high short chain fatty acid content in feces after WPH intervention. These changes jointly contributed to low blood pressure. The heart weight and cardiomyocyte injuries (hypertrophy and degeneration) were alleviated by WPH. The proteomic results revealed that 19 protein expressions in the heart mainly associated with the wingless/integrated (Wnt) signaling pathway and Apelin signaling pathway were altered after WPH supplementation. Notably, WPH alleviated serum oxidative stress, indicated by the decreased malondialdehyde content (P < 0.01), enhanced total antioxidant capacity (P < 0.01) and superoxide dismutase activity (P < 0.01). The current study suggests that WPH exhibit promising antihypertensive abilities in vivo and could be a potential alternative for antihypertensive dietary supplements.

The physiological and biochemical role of silicon in enhancing the resistance of maize to root‐lesion nematode

AbstractSilicon (Si) plays an important role in enhancing the tolerance of plants to biotic and abiotic stress in soil ecosystems. Root‐lesion nematodes (Pratylenchus scribneri; RLNs) cause root damage and diseases that result in quality deterioration and economic loss. This study investigated the effects of Si application on maize plants and its interaction with RLN infection. We set up different treatments to evaluate the role of silicon application in maize root growth and RLN resistance. This study conducted analysis by combining measurements of the metabolism and root activity of maize under different conditions. The results suggested that Si application (0.5 g/kg) significantly promoted fresh shoot weight, plant height, SPAD value (chlorophyll content), and root activity of maize, regardless of RLN inoculation. The highest SPAD value was observed in the Si treatment, which was significantly higher than in the control (CK) and RLN (N) treatments. Analysis of enzyme activity revealed that nematode inoculation reduced catalase (CAT) activity and increased malondialdehyde (MDA) concentration, while Si application increased CAT activity and decreased MDA concentration. In the SiN treatment, there was increased CAT activity at 0, 12, 48, 72 and 96 h compared with the N treatment. In parallel, nematode inoculation increased phenylalanine ammonia‐lyase (PAL), and polyphenol oxidase (PPO) activities, while SiN treatment further enhanced their activities. These findings indicate that Si application enhances maize resistance to nematode infection and improves plant growth and antioxidant defence mechanisms.

Salicylic acid-mediated alleviation of salt stress: Insights from physiological and transcriptomic analysis in Asarum sieboldii Miq

As global agriculture faces the pressing threat of salt stress, innovative solutions are imperative for sustainable agriculture. The remarkable potential of salicylic acid (SA) in enhancing plant resilience against environmental stressors has recently gained attention. However, the specific molecular mechanisms by which SA mitigates salt stress in Asarum sieboldii Miq., a valuable medicinal plant, remain poorly understood. Here, we evaluated the physiological and transcriptomic regulatory responses of A. sieboldii under salt stress (100 mM NaCl), both in the presence (1 mM SA) and absence of exogenous SA. The results highlighted that SA significantly alleviates salt stress, primarily through enhancing antioxidant activities as evidenced by increased superoxide dismutase, and peroxidase activities. Additionally, we observed an increment in chlorophyll (a and b), proline, total soluble sugar, and plant fresh weight, along with a decrease in malondialdehyde contents. Transcriptome analysis suggested consistency in the regulation of many differentially expressed genes and transcription factors (TFs); however, genes targets (GSTs, TIR1, and NPR1), and TFs (MYB, WRKY, TCP, and bHLH) possessed expressional uniqueness, and majority had significantly up-regulated trends in SA-coupled salt stress treatments. Further, bioinformatics and KEGG enrichment analysis indicated several SA-induced significantly enriched biological pathways. Specifically, plant hormone signal transduction was identified as being populated with key genes distinctive to auxin, cytokinin, ethylene, and salicylic acid signaling, suggesting their important role in salt stress alleviation. Inclusively, this report presents a comprehensive analysis encompassing gene targets, TFs, and biological pathways, and these insights may offer a valuable contribution to our knowledge of SA-mediated regulation and its crucial role in enhancing plant defense against diverse abiotic stressors.

Postprandial Antioxidative Response to Ingestion of Formulated Date- and Fruit-Based Nutritional Bars by Healthy Individuals.

Nutritional bars (NBs) are gaining popularity among healthy and athletic individuals, but postprandial antioxidative response has not been investigated. Therefore, the current study examined the postprandial alterations in total phenolic content (TPC), total antioxidant capacity (T-AOC), malondialdehyde (MDA), and Superoxide dismutase (SOD) in the plasma of healthy individuals after the ingestion of 140 g (510 Kcal) from formulated date-based bars (DBBs) or fruit-based bars (FBBs). Firstly, the free and bound phenolic contents (PCs) were determined to be 10.15 and 12.98 and 6.19 and 3.57 mg GAE g-1, respectively. FBBs were significantly higher in free PC than DBBs, while DBBs were considerably higher in bound PC than FBBs. Secondly, twenty participants with age, height, weight, body mass index (BMI), fat mass, and fat-free mass averages of 21.4 years, 170.0 cm, 66.3 kg, 22.9 kg m2, 14.5, and 29.2 kg, respectively, were subjected to metabolic experiments (ISRCTN19386758). Ingestion of 140 g of FBB or DBB resulted in 288.50 or 302.14 µg TPC mL-1 blood, respectively. Postprandial TPC content increased with time progression and peaked after 120 min. T-AOC contents averaged 22.63 and 23.61 U mL-1 before ingestion of FBBs or DBBs, respectively. The T-AOC content increased significantly 120 and 180 min after ingestion of DBBs, while no significant change was noted after consuming FBBs. A significant decrease in MDA content was observed 180 min after consuming DBBs, while no significant change was noted after consuming FBBs. SOD concentrations ranged from 193.99 to 201.07 U L-1 in FBBs and DBBs, respectively. No considerable response was noted up to 3 h after ingestion of FBBs. On the contrary, a significant response was found 120 min after consuming DBBs. Pearson's correlation coefficient indicated a highly significant positive correlation coefficient (p < 0.01) between T-AOC and either MDA or SOD, as well as between MDA and SOD. The principal component analysis demonstrated a strong and positive relationship between SOD and TPC at 60 and 120 min after DBB ingestion. In conclusion, the relative changes in postprandial responses in T-AOC and MDA did not significantly (p > 0.05) differ between DBBs and FBBs, except for TPC (p = 0.04, paired t-test) and SOD (p = 0.003, paired t-test). Further studies with an extended experimental time are needed to confirm the current findings.

Pharmacokinetic study and neuropharmacological effects of atractylenolide Ⅲ to improve cognitive impairment via PI3K/AKT/GSK3β pathway in intracerebroventricular-streptozotocin rats

Ethnopharmacological relevanceThe traditional Chinese herbal remedy Atractylodes macrocephala Koidz is renowned for its purported gastrointestinal regulatory properties and immune-enhancing capabilities. Atractylenolide III (ATL III), a prominent bioactive compound in Atractylodes macrocephala Koidz, has demonstrated significant pharmacological activities. However, its impact on neuroinflammation, oxidative stress, and therapeutic potential concerning Alzheimer's disease (AD) remain inadequately investigated.Aim of the study: This study aims to assess the plasma pharmacokinetics of ATL III in Sprague-Dawley (SD) rats and elucidate its neuropharmacological effects on AD via the PI3K/AKT/GSK3β pathway. Through this research, we endeavor to furnish experimental substantiation for the advancement of novel therapeutics centered on ATL III. Materials and methodsThe pharmacokinetic profile of ATL III in SD rat plasma was analyzed using liquid chromatography-tandem mass spectrometry (LC-MS/MS). AD models were induced in SD rats through bilateral intracerebroventricular (ICV) administration of streptozotocin (STZ). ATL III was administered at doses of 0.6 mg/kg, 1.2 mg/kg, and 2.4 mg/kg, while donepezil (1 mg/kg) served as control. Cognitive function assessments were conducted employing behavioral tests including the Morris Water Maze and Novel Object Recognition. Neuronal pathology and histological changes were evaluated through Nissl staining and Hematoxylin-Eosin (HE) staining, respectively. Oxidative stress levels were determined by quantifying malondialdehyde (MDA) content and total superoxide dismutase (T-SOD) activity. Molecular docking analysis was employed to explore the direct binding between ATL III and its relevant targets, followed by validation using Western blot (WB) experiments to assess the expression of p-Tau, PI3K, AKT, GSK3β, and their phosphorylated forms. ResultsWithin the concentration range of 5–500 ng/mL, ATL III demonstrated exceptional linearity (R2 = 0.9991), with a quantification limit of 5 ng/mL. In male SD rats, ATL III exhibited a Tmax of 45 min, a t1/2 of 172.1 min, a Cmax of 1211 ng/L, and an AUC(0-t) of 156031 ng/L*min. Treatment with ATL III significantly attenuated Tau hyperphosphorylation in intracerebroventricular-streptozotocin (ICV-STZ) rats. Furthermore, ATL III administration mitigated neuroinflammation and oxidative stress, as evidenced by reduced Nissl body loss, alleviated histological alterations, decreased MDA content, and enhanced T-SOD activity. Molecular docking analyses revealed strong binding affinity between ATL III and the target genes PI3K, AKT, and GSK3β. Experimental validation corroborated that ATL III stimulated the phosphorylation of PI3K and AKT while reducing the phosphorylation of GSK3β. ConclusionsOur results indicate that ATL III can mitigate Tau protein phosphorylation through modulation of the PI3K/AKT/GSK3β pathway. This attenuation consequently ameliorates neuroinflammation and oxidative stress, leading to enhanced learning and memory abilities in ICV-STZ rats.

Desloratadine via its anti-inflammatory and antioxidative properties ameliorates TNBS-induced experimental colitis in rats

Background Intestinal mucosal immune cells, notably mast cells, are pivotal in ulcerative colitis (UC) pathophysiology. Its activation elevates tissue concentrations of histamine. Inhibiting colonic histamine release could be an effective therapeutic strategy for treating UC. Experimental model like 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced colitis in rats mimic human IBD, aiding treatment investigations. Drug repurposing is a promising strategy to explore new indications for established drugs. Desloratadine (DES) is second-generation antihistamine utilized for managing allergies by blocking histamine action in the body. It also has reported anti-inflammatory and antioxidant actions. Objective DES was investigated for its repurposing potential in UC by preclinical screening in TNBS-induced colitis in Wistar rats. Methods Therapeutic efficacy of DES was evaluated both individually and in combination with standard drug 5-aminosalicylicacid (5-ASA). Rats were orally administered DES (10 mg/kg), 5-ASA (25 mg/kg), and DES + 5-ASA (5 mg + 12.15 mg) following the induction of colitis. Parameters including disease activity score rate (DASR), colon/body weight ratio (CBWR), colon length, diameter, pH, histological injury, and scoring were evaluated. Inflammatory biomarkers such as IL-1β, TNF-α, along with reduced glutathione (GSH), and malondialdehyde (MDA) were assessed. Results Significant protective effects of DES, especially in combination with 5-ASA, against TNBS-induced inflammation were observed as evidenced by reduced DASR, CBWR, and improved colon morphology. Drugs significantly lowered plasma and colon histamine and, cytokines levels. GSH restoration, and decreased MDA content were also observed. Conclusion DES and DES + 5-ASA demonstrated potential in alleviating colonic inflammation associated with TNBS-induced colitis in rats. The effect can be attributed to its antihistamine, anticytokine, and antioxidative properties.
