The Impact of Ethinyl Estradiol on Metformin Action on Prolactin Levels in Women with Hyperprolactinemia.

Metformin reduced prolactin levels only in women with hyperprolactinemia. The purpose of this case-control study was to compare metformin action on lactoctrope function between women receiving oral contraceptive pills and women not using hormonal contraception. The study included two groups of matched women with elevated prolactin levels and new-onset prediabetes or diabetes. The first group consisted of 20 women using oral contraceptive pills for at least 12 months before entering the study, while the second group included 20 patients not using any hormonal contraception. Over the whole study period, all women were treated with metformin (1.7-3 g daily). Circulating levels of glucose, insulin, prolactin, thyrotropin, free thyroid hormones, adrenocorticotropic hormone, gonadotropins and insulin-like growth factor-1 were measured at the beginning and at the end of the study (16 weeks later). Thirty-eight patients completed the study. Metformin reduced plasma glucose levels and improved insulin sensitivity but the latter effect was stronger in women receiving oral contraceptive pills than in women not using any contraception. Although metformin treatment decreased plasma prolactin levels in both study groups, this effect was stronger in women taking oral contraceptive pills. Only in this group of women, metformin increased plasma luteinizing hormone levels. The changes in plasma prolactin correlated with their baseline insulin sensitivity and the effect of metformin on insulin sensitivity. Metformin did not affect plasma levels of thyrotropin, free thyroxine, free triiodothyronine, follicle-stimulating hormone, adrenocorticotropic hormone and insulin-like growth factor-1. The obtained results suggest that the effect of metformin on overactive lactotropes depends on estrogen levels.

Decreased prolactin levels reduce parental commitment, egg temperatures, and breeding success of incubating male Adélie penguins

Hormones regulate many aspects of an individual's phenotype, including various physiological and behavioral traits. Two hormones have been described as important players in the regulation of parental investment in birds: the glucocorticoid hormone corticosterone and prolactin, a pituitary hormone, widely involved in mediating parental behavior. In comparison with corticosterone, the role of prolactin on parental investment remains poorly documented, and most studies so far have been correlative. In this study, the effects of an experimental decrease of prolactin levels on the incubation behavior of a long-lived seabird species were assessed. Male Adélie penguins were treated with self-degradable bromocriptine pellets, inhibiting prolactin secretion. Filming and subsequent video analysis allowed the determination of a behavioral time budget for birds and their position on the nest, while dummy eggs recorded incubation parameters. Incubation duration and breeding success at hatching were also monitored. As expected, bromocriptine-treatment significantly decreased plasma prolactin levels, but did not affect corticosterone levels. The behavioral time budget of penguins was not affected by the treatment. However, treated birds spent significantly more time in an upright position on the nest. These birds also incubated their eggs at lower temperatures and turned their eggs more frequently than controls, resulting in a lengthened incubation period. Despite this, the treatment was insufficient to trigger nest desertion and eggs of treated birds still hatched, indicating that several endocrine signals are required for the induction of nest abandonment. We suggest that the decreased prolactin levels in treated birds offset their timeline of breeding, so that birds displayed behavior typical of early incubation.

Neuroendocrine regulation of rearing behavior in the native Thai hen

Vasoactive intestinal peptide (VIP) is the avian prolactin releasing factor and changes in the concentrations of plasma prolactin (PRL) are found during the avian reproductive cycle. This study investigated the changes in the VIP/PRL system of native Thai hens rearing their young as compared to hens deprived of rearing their chicks. The number of VIP-immunoreactive (VIP-ir) neurons in the Nucleus inferioris hypothalami (IH) and Nucleus infundibuli hypothalami (IN) of hens rearing chicks (R) were compared with those of non-rearing chicks (NR). Plasma PRL levels were determined by enzyme-linked immunosorbent assay. The localization and number of VIP-ir neurons were determined by immunohistochemistry. The numbers of VIP-ir neurons in the IH–IN areas were high in the R hens, whereas the number of VIP-ir neurons decreased in the NR hens as compared to their respective R hens. During the rearing period, changes in the VIP-ir neurons within the IH–IN were correlated with plasma PRL levels. The results of the present study indicate for the first time that the VIP/PRL system plays a role in neuroendocrine reorganization to establish maternal behavior in native Thai chickens. The VIP/PRL system functions not only as a well established key regulator of incubation behavior, but is also involved in the regulation of rearing behavior. It is possible that VIP and the decline in the number of VIP-ir neurons and in turn VIPergic activity and the decrease in PRL levels are related to their contribution to rearing behavior of this non-seasonal breeding, equatorial precocial species.

Role of Ammonia and Nitric Oxide in the Decrease in Plasma Prolactin Levels in Prehepatic Portal Hypertensive Male Rats

Objectives: Since very little is known about neuroendocrine changes that occur in portal-systemic hepatic encephalopathy, we studied plasma prolactin (PRL) levels and the involvement of hyperammonemia, nitric oxide (NO) and dopaminergic and adrenergic systems in the control of this hormone secretion in a male rat model of prehepatic portal hypertension (PH). Methods: We conducted in vivo studies to determine plasma ammonia and PRL levels. Dopamine (DA), dihydroxyphenylacetic acid (DOPAC), epinephrine and norepinephrine content in medial basal hypothalamus (MBH) and anterior pituitary (AP) were measured. In addition, NO synthase (NOS) activity and protein expression were evaluated in APs. In in vitro studies, the APs from intact rats were incubated with different doses of ammonia and PRL secretion was determined. In ex vivo studies, the APs from normal and PH rats were incubated in the presence of ammonia and/or a NOS inhibitor, N^G-nitro-L-arginine-methyl ester (L-NAME) and PRL secretion was determined. Results: PH rats had a significant increase in plasma ammonia levels (p < 0.001) and a decrease in plasma PRL levels (p < 0.05). Neither DA nor DOPAC content or DOPAC/DA ratios were modified in both MBH and APs; however, we observed a significant increase in norepinephrine content in both MBH and AP (p < 0.001 and p < 0.05, respectively) and a significant increase in epinephrine in APs (p < 0.001). Moreover, PH produced an increase in NOS activity (p < 0.01) and NOS protein expression (p < 0.0001) in APs. The ammonia (100 µM) significantly reduced PRL secretion from APs in vitro (p < 0.05). The presence of L-NAME, an inhibitor of NOS, abrogated the inhibitory effect of ammonia on PRL secretion from APs from control and PH rats. Conclusions: We found that plasma PRL levels were decreased in PH rats probably due to the high ammonia levels. The central noradrenergic system could also mediate this decrease. Also, the increase in NOS activity and/or content in AP induced NO production that directly inhibited PRL secretion from the AP, without the participation of the dopaminergic system.

Experimental allergic encephalomyelitis in pituitary-grafted Lewis rats.

Treatment of susceptible rats with dopaminergic agonists that reduce prolactin release decreases both severity and duration of clinical signs of experimental allergic encephalomyelitis (EAE). To assess to what extent the presence of an ectopic pituitary (that produces an increase in plasma prolactin levels mainly derived from the ectopic gland) affects EAE, 39 male Lewis rats were submitted to pituitary grafting from littermate donors. Another group of 38 rats was sham-operated by implanting a muscle fragment similar in size to the pituitary graft. All rats received subcutaneous (s.c.) injections of complete Freund's adjuvant (CFA) plus spinal cord homogenate (SCH) and were monitored daily for clinical signs of EAE. Animals were killed by decapitation on days 1, 4, 7, 11 or 15 after immunization and plasma was collected for prolactin RIA. In a second experiment, 48 rats were immunized by s.c. injection of a mixture of SCH and CFA, and then received daily s.c. injections of bromocriptine (1 mg/kg) or saline. Groups of 8 animals were killed on days 8, 11 or 15 after immunization and plasma prolactin was measured. Only sham-operated rats exhibited clinical signs of the disease when assessed on day 15 after immunization. A progressive decrease in plasma prolactin levels was observed in pituitary-grafted rats, attaining a minimum 15 days after immunization, whereas plasma prolactin levels were increased during the course of the disease in sham-operated rats. Plasma prolactin levels were higher in pituitary-grafted rats than in sham-operated rats 1 day after immunization, but lower on days 7, 11 and 15 after immunogen injection. Further supporting a correlation of suppressed prolactin levels with absence of clinical signs of EAE, rats that were administered the dopaminergic agonist bromocriptine showed very low plasma prolactin levels and did not exhibit any clinical sign of EAE. These results indicate that low circulating prolactin levels coincide with absence of clinical signs of EAE in Lewis rats.

Sumanirole, a Highly Dopamine D2-Selective Receptor Agonist: In Vitro and in Vivo Pharmacological Characterization and Efficacy in Animal Models of Parkinson's Disease

The purpose of this study is to demonstrate that sumanirole is a novel dopamine receptor agonist with high in vitro and in vivo selectivity for the D(2) receptor subtype. Sumanirole, (R)-5,6-dihydro-5-(methylamino)-4H-imidazo[4,5,1-ij]quinolin-2(1H)-one (Z)-2-butenedioate (1:1), is unique; it has greater than 200-fold selectivity for the D(2) receptor subtype versus the other dopamine receptor subtypes in radioligand binding assays. In cell-based assays, sumanirole is a fully efficacious agonist, with EC(50) values between 17 and 75 nM. In animals, sumanirole elicits many physiological responses attributed to D(2)-like receptor function. In rats, sumanirole is a full agonist for elevation of striatal acetylcholine levels (ED(50) = 12.1 micromol/kg i.p.). Sumanirole s.c. dose dependently decreased plasma prolactin levels and depressed dopamine neuron firing rates in the substantia nigra pars compacta with an ED(50) of 2.3 micromol/kg i.v. This high selectivity for D(2) receptors translates into excellent locomotor stimulant activity in animal models of Parkinson's disease. In reserpinized, alpha-methyl-para-tyrosine-treated rats, sumanirole caused a significant and sustained increase in horizontal activity at doses > or =12.5 micromol/kg s.c. In unilateral 6-hydroxydopamine-lesioned rats, sumanirole caused profound, sustained rotational behavior and was substantially more efficacious than any other agonist tested. Sumanirole-stimulated rotational behavior was blocked by the dopamine receptor antagonist haloperidol. Sumanirole dose dependently improved disability scores and locomotor activities of two of three 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-lesioned monkeys. In summary, sumanirole is the first published selective D(2) receptor agonist. The compound has activity in animal models of dopamine hypofunction and has a high level of efficacy in animal models of Parkinson's disease.

Modulation of prolactin but not corticosterone responses to stress in relation to parental effort in a long-lived bird

We tested the hypothesis that parental effort modulates the magnitude of corticosterone and prolactin responses to stress in a long-lived bird, the Black-legged kittiwake ( Rissa tridactyla). To do so, we compared corticosterone and prolactin responses to capture/restraint stress between chick-rearing birds and failed breeders (no parental effort). We predicted that (1) the increase in plasma corticosterone levels in response to stress should be lower in chick-rearing birds, (2) the decrease in plasma prolactin levels in response to stress should be lower in chick-rearing birds, and (3) as both sexes care for the chick, there should be no sex difference in the hormonal response to stress. Baseline plasma corticosterone and prolactin levels were higher in chick-rearing birds and were not influenced by body condition. Failed breeders were in better condition than chick-rearing individuals. Corticosterone response to stress was unaffected by parental effort as both chick-rearing and failed birds exhibited a robust corticosterone increase. Prolactin response to stress was however clearly influenced by parental effort: chick-rearing birds showed a modest 9% prolactin decrease whereas in failed birds prolactin concentrations fell by 41%. Body condition did not influence hormonal responses to stress. When facing stressful condition, breeding kittiwakes attenuate their prolactin response to stress while enhancing their secretion of corticosterone. Increasing corticosterone secretion triggers foraging efforts and diminishes nest attendance whereas an attenuation of prolactin response to stress maintains parental behavior. We suggest that this hormonal mechanism facilitates a flexible time-budget that has been interpreted as a buffer against environmental variability.

Cabergoline treatment of risperidone-induced hyperprolactinemia: a pilot study.

D(2) blockers, including the atypical antipsychotic risperidone, induce hyper-prolactinemia in a significant number of patients treated. The endocrine and sexual side effects related to hyperprolactinemia significantly impair tolerability and compliance in patients, including those with a good response to risperidone. This pilot study aimed to evaluate the efficacy and tolerability of a low dose of cabergoline, a D(2) agonist, in the treatment of risperidone-induced hyperprolactinemia. Nineteen male and female DSM-IV-defined schizophrenic patients who were clinical responders to risperidone but were suffering from symptomatic hyperprolactinemia were treated with cabergoline, 0.125 to 0.250 mg/week for 8 weeks. Plasma prolactin level was assessed at baseline and at the end of the study. Data were collected from January 2002 to April 2003. After cabergoline treatment, the mean decrease in plasma prolactin levels was statistically significant (p <.05) for the total sample, and 11 patients showed remission of clinical signs with prolactin values within the normal range. No side effect was observed or reported, and the patients' psychopathology was unchanged. Results suggest that low-dose cabergoline treatment of risperidone-induced hyperprolactinemia may be safe and clinically effective in a relevant number of patients.

Changes in substance P content at the hypothalamic-pituitary axis during the Wallerian degeneration of peripheral sympathetic neurons after superior cervical ganglionectomy in male rats: effect of hyperprolactinemia.

The effects of Wallerian degeneration of the peripheral sympathetic neurons projecting to the hypothalamus on the mechanism of interaction between prolactin and substance P (SP) were examined. The effects of superior cervical ganglionectomy (SCGx) on SP content in various hypothalamic regions and in the hypophysis were evaluated in control and hyperprolactinemic rats. Male rats that received pituitary transplants at the age of 5 days and age-matched sham-operated controls were used. Pituitary grafting significantly increased circulating values of prolactin, as did SCGx. In hyperprolactinemic rats, SCGx partially decreased plasma prolactin levels. Neonatal hyperprolactinemia decreased SP content in the anterior (AH) and posterior (PH) hypothalamus and in the median eminence (ME), but increased it in the mediobasal hypothalamus (MBH). Acute SCGx significantly increased SP in the MBH, PH, and ME. SCGx in hyperprolactinemic animals further increased SP content in MBH. In the ME and Ah, SCGx in pituitary grafted rats decreased SP content as compared with the controls. In the pituitary gland (PG), SCGx only decreased SP content in hyperprolactinemic, but not in control rats. An interaction between peripheral nor-adrenergic neurons and prolactin to regulate SP within the hypothalamus was positive in the MBH, AH, ME, and PG, but not in the PH. These data indicate the existence of interactive mechanisms between prolactin and the peripheral sympathetic neurons to regulate SP content at the hypothalamic-pituitary axis. Interrelationships between prolactin and SP were also observed.

Extrapyramidal and neuroendocrine effects of AM404, an inhibitor of the carrier-mediated transport of anandamide

A selective inhibitor of the carrier-mediated transport of endogenous cannabinoids, N-(4-hydroxyphenyl)-arachidonylethanolamide (AM404), has been recently synthesized and proposed as a useful tool for studying the physiological effects of endogenous cannabinoids and as a potential therapeutic agent in a variety of diseases. In the present study, we have examined the effects of this compound in two important brain processes in which a role for anandamide and other endogenous cannabinoids has been claimed: neuroendocrine regulation and extrapyramidal motor activity. A single and well-characterized dose of AM404, which presumably resulted in a significant elevation of the levels of endogenous cannabinoids, produced a marked decrease in plasma prolactin (PRL) levels, with no changes in luteinizing hormone (LH) levels. This decrease in PRL levels was accompanied by an increase in the activity of tyrosine hydroxylase (TH) in the medial basal hypothalamus. Both decreased PRL secretion and increased hypothalamic TH activity have been reported to occur after the administration of anandamide. Administration of AM404 also produced a marked motor inhibition in the open-field test, as also reported for anandamide, with a decrease in ambulatory and exploratory activities and an increase in the time spent in inactivity. This was accompanied by a decrease in the activity of TH in the substantia nigra, an effect also previously observed for anandamide.

Stereotactic Radiotherapy for Cushing's Disease and Prolactinoma

Stereotactic radiotherapy (SRT) for secretory pituitary adenoma offers precise localization of dose with potential sparing of optic and hypothalamic functions. We collated the indications, treatment parameters, and both the clinical and biochemical results of Stereotactic radiotherapy for ACTH- 1 and prolactin-secreting pituitary adenoma. Between May 1990 and February 1996, nine patients (4 males and 5 females) with a mean age of 31.9 years had fractionated SRT for ACTH- or prolactin-secreting pituitary adenomas (Cushing's disease: n = 5; prolactinoma: n = 3; Nelson's syndrome: n = 1). These 9 patients had subtotal transphenoidal resections before SRT. An additional patient with Cushing's disease had aborted transphenoidal surgery and single-fraction Stereotactic radiosurgery (SRS). Prior to SRT, 4 patients received planned conventional fractionated external beam radiotherapy (XRT) (median dose 33.3 Gy). The median SRT dose was 34.5 Gy (range 14.4–50.4 Gy). The median total radiation dose (XRT plus SRT) for patients receiving fractionated treatment was 47.7 Gy. The median follow-up was 62 months (range 22–91 months). One patient with Nelson's syndrome was lost to follow-up. Of the 9 evaluable patients, none required reoperation for growth of pituitary tumor and all had radiographic control as shown by serial MRI scans. In 4 of the 6 patients with Cushing's disease, treatment resulted in normalization of hormone levels and resolution of symptoms. All 4 patients were able to discontinue medications. In 2 patients, normalization occurred within 4 months of treatment. Two patients with Cushing's disease failed, requiring adrenalectomy. For patients with prolactinoma, treatments resulted in a significant decrease of plasma prolactin levels. All patients treated for prolactinoma became asymptomatic; 2 were able to discontinue bromocriptine and 1 patient had a successful pregnancy. After radiation therapy, 1 patient developed new hypopituitarism. None of the patients developed worsened visual acuity, visual fields, or new neurological deficits. SRT with or without XRT may yield meaningful reductions in hormone levels with concomitant clinical improvement in selected patients with prolactinomas or Cushing's disease. In certain situations, combining conventional XRT with SRT is appropriate. Modifications of the Stereotactic radiation delivery may maximally reduce the hypothalamic dose, thereby possibly reducing the risk of clinical hypopituitarism. This treatment is safe and is without visual or other neurological toxicity.

Time course of the effects of different cannabimimetics on prolactin and gonadotrophin secretion: Evidence for the presence of CB 1 receptors in hypothalamic structures and their involvement in the effects of cannabimimetics

Several reports have demonstrated that (−)- Δ 9-tetrahydrocannabinol ( Δ 9-THC) and arachidonylethanolamide [anandamide (AEA)] were able to inhibit prolactin (PRL) secretion from the anterior pituitary gland in male rodents, whereas ovarian phase-dependent effects were seen in females. However, in most of these studies, the analysis of PRL levels was performed at times longer than 30 min after cannabinoid administration. In the present study, we examined the time course of the effects of three different Cannabimimetics, Δ 9-THC, AEA, and AM356 ( R-methanandamide), a more stable analog of AEA, on PRL and gonadotrophin secretion in male Wistar rats. In addition, we characterized the presence of cannabinoid receptors in hypothalamic structures related to neuroendocrine control and studied their potential involvement in the effects of Cannabimimetics. We found that the three compounds decreased plasma luteinizing hormone (LH) levels, although only the effects of Δ 9-THC were statistically significant. The inhibitory effect was already apparent at 40 min after administration, but only in the case of Δ 9-THC did it persist up to 180 min after administration. No significant changes were seen in plasma follicle-stimulating hormone (FSH) levels after the administration of any of the three different Cannabimimetics at any of the four times analyzed. Both AEA and AM356 produced a significant decrease in plasma PRL levels, which appeared at 20 min after administration and persisted up to 60 min, waning after this time. Interestingly, the time course of the effect of Δ 9-THC resembled that of AEA and AM356 only during the later part of the response, because Δ 9-THC produced a marked increase in plasma PRL levels at 20 min, no changes at 40 min and a decrease from 60 min up to 180 min. In additional experiments, we tried to elucidate which of these two phases observed after Δ 9-THC administration was mediated by the activation of cannabinoid receptors. These receptors are present in hypothalamic structures related to neuroendocrine control, with the highest densities in the arcuate nucleus (dorsal area) and the medial preoptic area, and the lowest in the lateral hypothalamic area, although none of these regions exhibited high densities for this receptor as compared with classical regions containing cannabinoid receptors, such as the basal ganglia. The activation of these receptors by Δ 9-THC seems to be involved in the inhibitory phase of the effect of this cannabinoid on PRL release, but not in the early stimulation; when these receptors were blocked with a specific antagonist, SR141716, the stimulation by Δ 9-THC was still observed, but the late inhibition was abolished. In summary, AEA and AM356 markedly decreased PRL release and slightly decreased LH secretion, with no changes on FSH release. Δ 9-THC also produced a marked inhibition of LH secretion, but its effects on PRL were biphasic with an early stimulation not mediated by the activation of cannabinoid receptors, followed by a late and cannabinoid receptor-mediated inhibition. Their site of action may well be the hypothalamic structures related to neuroendocrine control, which contain a small, but probably very active, population of cannabinoid receptors.

Hyperprolactinaemia and cyclosporine treatment on secretion of adenohypophyseal hormones

The possibility of interactions between the effects of cyclosporine (CyA) and prolactin to regulate various physiological functions has been suggested. To further analyze these interactions, the effect of chronic CyA administration on the secretion of adenohypophyseal hormones in hyperprolactinemic male rats, was studied. Male rats were grafted one pituitary gland under the kidney capsule or were sham-operated at 30 days of age. Both pituitary-grafted and sham-operated rats were given s.c. injections of vehicle or CyA (5mg/kg/day) for 30 days beginning on the day of surgery. Pituitary-grafting increased the plasma leveles of prolactin and decreased the plasma levels of luteinizing hormone (LH), whereas growth hormone (GH), thyroid stimulating hormone (TSH) and follicle stimulating hormone (FSH) did not change. CyA treatment to sham-operated rats did not modify plasma prolactin levels and decreased plasma levels of GH and LH. Administration of the immunosuppressor to pituitary-grafted rats decreased plasma prolactin levels and augmented plasma levels of LH and FSH. Plasma TSH levels did not change after CyA treatment either in sham-operated or in pituitary-grafted rats. These data suggest that CyA may modify the effects induced by prolactin on pituitary hormone secretion, although direct effects of the drug on the hypothalamic-pituitary axis cannot be excluded, according to the data obtained in sham-operated rats.

Behavioral and endocrine effects of growth hormone administration in aged female rats.

Growth hormone (GH) secretion declines during normal aging along with reproductive activity in mammalian species. Various behavioral changes also occur in aged animals. In these experiments we have studied the effects of GH administration on behavioral and endocrine alterations exhibited by aged (18 months old) female rats of the Sprague-Dawley strain. Animals were selected showing at least 2 weeks of cornified vaginal smears (constant estrous) and treated with GH (0.1 mg/kg SC) daily for 8 weeks. Vaginal smears performed during the drug treatment revealed a recovery of estrous cycle in 60% of animals. GH treatment was also followed by an increased acquisition of shuttle-box active avoidance behavior and a facilitated retention of passive avoidance response. Compared to saline-injected controls, female rats treated with GH also exhibited a decrease of novelty-induced excessive grooming. The endocrine pattern of GH-treated aged female rats revealed a decrease in plasma prolactin levels and an increase in luteinizing hormone and 17 beta-estradiol levels as compared to those of control animals. These results support the concept that behavioral and endocrine alterations occurring in aging are not irreversible and that GH may interfere with these changes probably by means of its trophic action on different target organs.

Bromocryptine is effective in reducing milk production in ewes during lactation, but has no additional effect during drying off

AbstractThe effect of bromocryptine on milk production was investigated in 32 crossbred ewes (Flemish Milksheep х Suffolk) with one sucking lamb, during lactation and during drying off. After weaning the lambs, all ewes were hand milked twice daily for 3 days. From day 4 onwards, the ewes were divided into four groups with equal mean daily milk production. Ewes of groups A and B were hand milked twice daily during the following 9 days. Ewes of groups C and D were dried off by reducing the milking frequency. They were milked on days 5, 8 and 12. In groups B and D, bromocryptin (1 ml of 1 g/l ethanol 50%) was injected intramuscularly twice daily while control ewes (groups A and C) received 1 ml ethanol 50%. Blood samples were taken by venipuncture prior to each injection and were assayed for prolactin, growth hormone, thyrotropin and cortisol.Bromocryptine decreased plasma prolactin levels but did not alter growth hormone or cortisol concentrations during lactation or drying off. Thyrotropin levels were elevated by bromocryptine during drying off but were unaffected during galactopoiesis.During lactation the daily milk production remained unchanged in control ewes while in bromocryptine-treated ewes a proportional decrease of 0·5 was observed. In all ewes the decrease in milk production occurring at drying off could not be accelerated or stimulated by bromocryptine.It is concluded that prolactin plays an important role in galactopoiesis in ewes but its role is comparatively unimportant compared with other factors during mammary involution.

Blood luteinizing hormone and prolactin concentrations in response to naltrexone challenge: Studies on rats with diabetes induced by different doses of streptozotocin

Opiate system involvement in diabetes induced by three different doses of streptozotocin (STZ; 40, 50, and 60 mg/kg body weight [BW]) was studied by monitoring luteinizing hormone (LH) and prolactin (PRL) levels as a response to naltrexone (Nalt) administration. After four weeks of diabetes a marked decrease in BW, as well as severe hyperglycemia and increased blood urea nitrogen (BUN) levels were found. The rats, whose diabetes was induced by 50 mg/kg of STZ, exhibited the highest amount of blood glucose ( P < 0.05, compared with the 40 mg/kg induced group) and BUN levels ( P < 0.004 compared with the order two groups) and BW loss. The normal response to Nalt, which is expressed by elevation of plasma LH and decreased plasma PRL levels was observed only in the low-dose STZ (40 mg/kg BW) diabetes-induced group, while in the other two diabetic groups (50 and 60 mg/kg BW) there was no significant change in plasma LH and PRL as a result of the Nalt challenge. Presensitization of the endogenous opioid receptors by morphine in normoglycemic (control) and “low-dose diabetic” rats (40 mg/kg BW of STZ), presented a clear difference between the two. Morphine pretreatment inhibited LH response to Nalt in the low-dose, STZ-induced diabetic rats, while no effect of morphine pretreatment on LH response to Nalt could be recorded in the normoglycemic group. Thus it can be concluded that in STZ-induced diabetes, plasma glucose and BUN levels do not reflect the neuroendocrine injury observed when monitored by LH and PRL secretion in response to Nalt challenge. Supersensitization of the opioid receptors before the Nalt challenge may increase the ability to reveal neuroendocrine system impairment.

Reduction of plasma prolactin by acute administration of CB-154 in ovariectomized F344 and Holtzman rats treated with diethylstilbestrol: A comparison of RIA and Nb 2 lymphoma bioassay

Fischer 344 (F344) and Holtzman Sprague-Dawley female rats were ovariectomized and implanted with a s.c. Silastic capsule of diethylstilbestrol (DES). At 1,4 and 8 weeks of DES exposure, blood samples were obtained by infraorbital sinus puncture under light ether anesthesia before and 2 hours after s.c. administration of CB-154 (2.5 mg/rat). The plasma obtained was assayed for prolactin by radioimmunoassay (RIA) and Nb 2 lymphoma cell bioassay (BA). At 1 week of DES treatment plasma prolactin in F344 rats measured by RIA was decreased by CB-154 (≈ 60%) whereas the level measured by BA was not changed and the BA:RIA ratio was increased from 2.4 +/− 0.2 to 4.8 +/− 0.9. CB-154 decreased plasma prolactin levels at 4 weeks but the effect seen by RIA (∼80%) was greater than that seen by BA (∼60%) and the BA:RIA ratio was increased (2.2 +/− 0.2 vs 3.4 +/− 0.5). By 8 weeks of DES exposure, CB-154 was as effective in reducing the levels measured by BA (∼89%) as those measured by RIA (∼85%) and the BA:RRIA was not affected by the dopamine agonist. In Holtzman rats CB-154 decreased prolactin levels measured by RIA and BA to the same extent at both 1 and 4 weeks resulting in no change in the BA:RIA ratio, but at 8 weeks the BA:RIA was decreased by CB-154 (2.1 +/− 0.1 to 1.7 +/−0.1). As was observed in F344 rats, the reduction in plasma prolactin induced by CB-154 increased as the duration of DES treatment increased (1 week ∼45%; 4 weeks ∼55–60%; 8 weeks ∼80–85% inhibition) regardless of assay method. It is concluded that DES increases the BA:RIA ratio of plasma prolactin and that acute CB-154 treatment increases, decreases or does not change the BA:RIA ratio depending on the strain of rat used and the duration of DES treatment.

Prolactin Levels in the Western Spotted Skunk: Changes during Pre- and Periimplantation and Effects of Melatonin and Lesions to the Anterior Hypothalamus1

Prolactin (PRL) is the primary pituitary hormone responsible for initiating increased function of the corpus luteum and blastocyst implantation in the western spotted skunk. Therefore, we have designed experiments to validate a PRL RIA, characterize the preimplantation profile in PRL secretion, and determine the effects of exogenous melatonin and lesions to the anterior hypothalamic area (AHA) on PRL secretion in the skunk. These objectives were investigated with a heterologous RIA using canine PRL standards and antiserum. Displacement curves of skunk pituitary extract and serum were parallel to the canine PRL standard curve. Growth hormone-releasing hormone injection did not cause a significant change in plasma PRL levels as detected by the assay (p = 0.74). Injection of pimozide increased and bromocriptine decreased plasma PRL levels (p less than 0.05). A seasonal trend in plasma PRL levels was observed during the preimplantation period, with mean concentrations ranging from 5 ng/ml during the period of short day length in January to 17.1 ng/ml during the long day photoperiod in early May. The average date of blastocyst implantation in this study was 2 May (n = 16). Silastic capsules containing melatonin (n = 5) significantly delayed both the seasonal rise in plasma PRL levels and the time of implantation (p less than 0.05) compared to controls with empty capsules (n = 4). Lesions to the AHA (AHAx, n = 5) eliminated these effects of melatonin on both the rise in PRL and time of implantation. Effects of Heterologous Growth Hormones on Hypothalamic and Pituitary Function in Transgenic Mice

The expression of the mouse metallothionein-I (mMT) promoter/human growth hormone (hGH) fusion gene in transgenic mice leads to female sterility and major alterations in the function of the hypothalamic-adenohypophyseal system. These alterations include increases in median-eminence norepinephrine turnover and plasma LH levels, and a decrease in plasma prolactin levels in intact males, and an increase in median-eminence dopamine turnover combined with the suppression of plasma prolactin levels in ovariectomized females. To further characterize these changes and to determine whether they are due to the lactogenic or somatotropic activity of hGH, we have studied hypothalamic and pituitary function in transgenic mice expressing mMT/hGH, mMT/hGH.B 'variant', or mMT/bGH fusion genes. In males, the expression of the hGH.B gene was associated with a reduction in pituitary prolactin release in vitro and an increase in LH response to LHRH stimulation, while the bGH transgene did not affect any of the examined parameters of LH and prolactin release. Median-eminence norepinephrine turnover was increased in each of the three lines of transgenic males, while median-eminence dopamine turnover was reduced only in animals expressing the hGH.B gene. In ovariectomized females, plasma LH was suppressed by hGH variant expression, while median-eminence norepinephrine turnover was suppressed in both hGH.B and bGH animals. The turnover of dopamine was increased in the median eminence of females expressing either of the human genes (hGH or gHG.B) and reduced in the median eminence of ovariectomized bGH females. We conclude that the hGH.B gene is weakly lactogenic in mice, and that the chronic stimulation of either GH receptors (by bGH) or both GH and prolactin receptors (by hGH or hGH.B) can lead to profound alterations in the metabolism of hypothalamic neurotransmitters and pituitary hormone release.

Possible Involvement of Hypothalamic Monoamines in Mediating the Action of Alpha-2u-Globulin on the Pituitary-Testicular Axis in Rats

A major androgen-dependent urinary protein of male rodents, alpha 2u-globulin, has been shown to influence adenohypophyseal hormone release. In an attempt to elucidate the mechanisms of its action, we have examined several parameters of hypothalamic and pituitary function in adult male rats treated for 2 weeks with two injections daily of 0.75 mg alpha 2u-globulin and sacrificed 16 h after the last injection. This treatment led to an increase in plasma luteinizing hormone levels, a decrease in plasma prolactin levels, an increase in testosterone concentrations in both plasma and testicular tissue, and increases in testicular weights. The norepinephrine turnover in median eminence and anterior hypothalamus was increased in alpha 2u-globulin-injected animals, while the norepinephrine turnover in the remaining medial basal hypothalamus was reduced. alpha 2u-Globulin-treated animals had a significantly decreased dopamine turnover in the anterior hypothalamus, while in the medial basal hypothalamus the dopamine metabolism was increased. These data suggest that alpha 2u-globulin-induced changes in gonadotropin and prolactin secretion are mediated by changes in catecholamine metabolism in several hypothalamic regions. Increased testosterone secretion appears to be due to increased secretion of gonadotropins from the pituitary.
