Apoptosis induction in human breast cancer (MCF-7) cells by a novel venom l-amino acid oxidase (Rusvinoxidase) is independent of its enzymatic activity and is accompanied by caspase-7 activation and reactive oxygen species production

We report the elucidation of a mechanism of apoptosis induction in breast cancer (MCF-7) cells by an L-amino acid oxidase (LAAO), Rusvinoxidase, purified from the venom of Daboia russelii russelii. Peptide mass fingerprinting analysis of Rusvinoxidase, an acidic monomeric glycoprotein with a mass of ~57kDa, confirmed its identity as snake venom LAAO. The enzymatic activity of Rusvinoxidase was completely abolished after two cycles of freezing and thawing; however, its cytotoxicity toward MCF-7 cells remained unaffected. Dose- and time-dependent induction of apoptosis by Rusvinoxidase on MCF-7 cells was evident from changes in cell morphology, cell membrane integrity, shrinkage of cells and apoptotic body formation accompanied by DNA fragmentation. Rusvinoxidase induced apoptosis in MCF-7 cells by both the extrinsic (death-receptor) and intrinsic (mitochondrial) signaling pathways. The former pathway of apoptosis operated through activation of caspase-8 that subsequently activated caspase-7 but not caspase-3. Rusvinoxidase-induced intrinsic pathway of apoptosis was accompanied by a time-dependent depolarization of the mitochondrial membrane through the generation of reactive oxygen species, followed by a decrease in cellular glutathione content and catalase activity, and down-regulation of expression of anti-apoptotic proteins Bcl-XL and heat-shock proteins (HSP-90 and HSP-70). Rusvinoxidase treatment resulted in increase of the pro-apoptotic protein Bax, subsequently leading to the release of cytochrome c from mitochondria to the cytosol and activating caspase-9, which in turn stimulated effector caspase-7. Rusvinoxidase at a dose of 4mg/kg was non-toxic in mice, indicating that it may be useful as a model for the development of peptide-based anticancer drugs.

Se-methylselenocysteine modulates antioxidant response of rat spleen to ionizing radiation

Whole body irradiation with a single 10-Gy dose caused increase in lipid peroxidation and a transient decrease in cellular glutathione content in rat spleen, demonstrating elevated oxidative stress. The irradiation also caused increases in activities of glutathione peroxidase (GPx), glutathione reductase (GR), and glucose 6-phosphate dehydrogenase (G6PD), the enzymes carrying out glutathione redox cycling; but not glutamate cysteine ligase (GCL), the enzyme in glutathione synthesis process. Increases in catalase (CAT) activity and heme oxygenase-1 (HO-1) and glutathione S-transferase pi (GSTpi) protein levels were also exhibited after irradiation. Administration of Se-methylselenocysteine (MSC) (0.75 mg/rat/day, for 1 week) resulted in increases in GPx, G6PD, and CAT activities and GSTpi protein level in non-irradiated spleen, without affecting glutathione and lipid peroxidation levels. The MSC pretreatment prior to irradiation abrogated the irradiation-induced increase in lipid peroxidation, and it induced increases in glutathione content, GCL, GPx and CAT activities, and HO-1, GSTpi, and peroxiredoxin 2 protein levels upon irradiation. Our results suggest a role for MSC pretreatment in prevention of irradiation-induced oxidative damage in spleen by reinforcing antioxidant capacity, particularly the glutathione system.

Cytoprotective Effects of Organosulfur Compounds against Methimazole Induced Toxicity in Isolated Rat Hepatocytes.

Methimazole is a drug widely used in hyperthyroidism. However, life threatening hepatotoxicity has been associated with its clinical use. No protective agent has been found to be effective against methimazole induced hepatotoxicity yet. Hence, the capacity of organosulfur compounds to protect rat hepatocytes against cytotoxic effects of methimazole and its proposed toxic metabolite, N-methylthiourea was evaluated. Hepatocytes were prepared by the method of collagenase enzyme perfusion via portal vein. Cells were treated with different concentrations of methimazole, N methylthiourea, and organosulfur chemicals. Cell death, protein carbonylation, reactive oxygen species formation, lipid peroxidation, and mitochondrial depolarization were assessed as toxicity markers and the role of organosulfurs administration on them was investigated. Methimazole caused a decrease in cellular glutathione content, mitochondrial membrane potential (ΔΨm) collapse, and protein carbonylation. In addition, an increase in reactive oxygen species (ROS) formation and lipid peroxidation was observed. Treating hepatocytes with N methylthiourea caused a reduction in hepatocytes glutathione reservoirs and an elevation in carbonylated proteins, but no significant ROS formation, lipid peroxidation, or mitochondrial depolarization was observed. N-acetyl cysteine, allylmercaptan, and diallyldisulfide attenuated cell death and prevented ROS formation and lipid peroxidation caused by methimazole. Furthermore, organosulfur compounds diminished methimazole induced mitochondrial damage and reduced the carbonylated proteins. In addition, these chemicals showed protective effects against cell death and protein carbonylation induced by methimazole metabolite. Organosulfur chemicals extend their protective effects against methimazole-induced toxicity by attenuating oxidative stress caused by this drug and preventing the adverse effects of methimazole and/or its metabolite (s) on subcellular components such as mitochondria.

The preventive effect of Se-methylselenocysteine on γ-radiation-induced oxidative stress in rat lungs

We investigated the preventive effect of Se-methylselenocysteine (MSC) administration on γ-radiation (whole body irradiation, single 10-Gy dose)-induced oxidative damage in rat lungs. Rats were pretreated with MSC (0.75mg/rat/day) for 1 week before γ-irradiation. The MSC pretreatment prevented the irradiation-induced increase in lipid peroxidation and the concomitant decrease in cellular glutathione content. The prevention of irradiation-induced oxidative damage in MSC-pretreated rat lungs appeared to be associated with increased antioxidant capacity, particularly in the glutathione system. The 1-week MSC treatment resulted in an increase in glutathione peroxidase, glutathione reductase, and glucose 6-phosphate dehydrogenase activities, which are involved in glutathione redox cycling. An increase in catalase activity was also observed in the rat lungs. Additionally, a significantly increased level of nuclear factor erythroid 2-related factor 2 (Nrf2) was exhibited in the MSC-treated rat lungs. Heme oxygenase 1, glutathione S-transferase pi, and peroxiredoxin 1, which are known target proteins of Nrf2, were also increased in MSC-treated lungs. These results implicate Nrf2 signaling in the MSC-induced activation of the antioxidant system.

The effect of vitamin C on amiodarone-induced toxicity in rat thymocytes

Abstract Although, the antiarrhythmic effect of amiodarone (AMD) is well characterized, the mechanism of its toxicity on extracardiac tissues is still poorly understood. Several antioxidants have been shown to prevent AMD-induced toxicity by antioxidant and/or non-antioxidant mechanisms. In the current study, we evaluated the possible protective effect, in vitro, of vitamin C on AMD-induced toxicity in rat thymocytes. Rat thymocytes were cultured with increasing AMD concentrations (1–20 μM) with or without vitamin C (1000 μg/ml), for 24 hours. Cells treatment with AMD resulted in a concentration-dependent increase of hypodiploid cells and a significant decrease in cellular glutathione content. Vitamin C combined with AMD significantly decreased the proportion of hypodiploid cells and markedly increased the cellular glutathione content, compared with AMD treatment alone. These results suggest that treatment with vitamin C may prevent AMD-induced toxicity in rat thymocytes by restoring cellular glutathione content.

(R)- and (S)-Verapamil Differentially Modulate the Multidrug-resistant Protein MRP1

The multidrug-resistant protein MRP1 (involved in the cancer cell multidrug resistance phenotype) has been found to be modulated by racemic verapamil (through stimulation of glutathione transport), inducing apoptosis of human MRP1 cDNA-transfected baby hamster kidney 21 (BHK-21) cells and not of control BHK-21 cells. In this study, we show that the two enantiomers of verapamil have different effects on MRP1 activity. Only the S-isomer (not the R-isomer) potently induced the death of MRP1-transfected BHK-21 cells. The decrease in cellular glutathione content induced by the S-isomer, which was not observed with the R-isomer, was stronger than that induced by the racemic mixture, indicating that the R-isomer antagonized the S-isomer effect. Both enantiomers altered leukotriene C(4) and calcein transport by MRP1. Thus, the R-isomer behaved as an inhibitor, which was confirmed by its ability to revert the multidrug resistance phenotype toward vincristine. Molecular studies on purified MRP1 using fluorescence spectroscopy showed that both enantiomers bound to MRP1 with high affinity, with the binding being prevented by glutathione. Furthermore, conformational changes induced by the two enantiomers (monitored by sodium iodide accessibility of MRP1 tryptophan residues) were quite different, correlating with their distinct effects. (S)-Verapamil induces the death of potentially resistant tumor cells, whereas (R)-verapamil sensitizes MRP1-overexpressing cells to chemotherapeutics. These results might be of great potential interest in the design of new compounds able to modulate MRP1 in chemotherapy.

Apoptosis initiated by carbon tetrachloride in mitochondria of rat primary cultured hepatocytes

To investigate the mitochondria-initiated apoptosis pathway involved in Carbon tetrachloride (CCl4) hepatotoxicity in vitro. Several cytotoxicity endpoints, including WST-8 metabolism, lactate dehydrogenase leakage and morphological changes, were examined. The 5,5'-dithio-bis(2-nitrobenzoic acid) reaction was used to measure reduced glutathione level, and the malondialdehyde level was determined using the thiobarbituric acid assay. The release of cytochrome c and Bcl-X(L) was detected by Western blot. Caspase-3 activity was measured using the fluorogenic substrate Ac-DEVD-AMC. DNA fragmentation was used to evaluate cell apoptosis. A time- and dose-dependent decrease in cellular glutathione content was observed, along with a concomitant increase in malondialdehyde levels following the application of CCl4. Caspase 3 activity was stimulated at all doses of CCl4, with the most significant activation at 3 mmol/L. Cytochrome c was released obviously after CCl4 treatment. A time-dependent decrease in Bcl-X(L) expression was observed. DNA fragmentation results revealed apoptosis and necrosis following CCl4 treatment. Oxidative damage is one of the essential mechanisms of CCl4 hepatotoxicity, which triggers apoptosis via the mitochondria-initiated pathway.

L-2-Oxothiazolidine-4-Carboxylate: An Agent that Modulates Lipopolysaccharide-Induced Peritonitis in Rats

L-2-Oxothiazolidine-4-carboxylate (OTZ), a cysteine precursor, is a substrate for intracellular glutathione synthesis. As shown previously, OTZ prevents free-radical induced cellular damage during in vitro simulation of peritoneal dialysis. In the present study, we examined the effect of adding OTZ to peritoneal dialysis solution on peritoneal function and structure during lipopolysaccharide (LPS)-induced peritonitis in rats. In addition, we studied the effects of pretreatment with OTZ (given orally) on the effects of LPS-induced peritonitis in rats. Peritonitis was induced in rats by adding LPS (5 microg/mL) to the dialysis fluid. For acute experiments, rats were exposed to a single infusion of dialysis solution containing LPS or to LPS plus 5 mmol/L OTZ; peritoneal cell counts and permeability were determined after 4 hours. Alternatively, rats were pretreated with OTZ added to the drinking water (0.1%) for 10 days prior to infusion of LPS. For chronic experiments, peritoneal dialysis was performed over a 3-week period in rats with implanted peritoneal catheters. On days 8, 9, and 10 of the experiment, the rats were infused intraperitoneally with solution containing LPS (5 micro/mL), or LPS plus 5 mmol/L OTZ, to induce acute peritonitis. At the end of dialysis (10 days after the episodes of peritonitis), peritoneal function was assessed using a peritoneal equilibration test (PET), and peritoneal biopsies were taken to assess effects on peritoneal morphology. In the acute experiments, exposure to LPS led to increased peritoneal cell counts (+61% vs control, p < 0.05) and enhanced permeability of the peritoneum, leading to a loss in ultrafiltration (-63%, p < 0.0005). The glutathione concentration in peritoneal leukocytes also decreased during acute peritonitis (-31%, p < 0.05). During LPS-induced peritonitis, OTZ prevented the increase in dialysate cell count and the decrease in cellular glutathione content. Simultaneous administration of OTZ did not prevent the increased peritoneal permeability induced by LPS. However, in rats pretreated with OTZ, LPS-induced permeability to protein was significantly lower than in the nontreated animals (0.049 +/- 0.011 vs 0.087 +/- 0.034, p < 0.05). In the chronic experiments, LPS-induced peritonitis did not lead to any functional differences in peritoneal transport at the end of 3 weeks of dialysis. However, LPS-induced peritonitis led to increased thickness of the peritoneum and neovascularization within peritoneal interstitium compared to peritonitis-free animals. In contrast to the LPS-treated animals, the peritoneum of the rats exposed to LPS in the presence of OTZ was of a thickness similar to that in the control rats. Supplementation of dialysis fluid with OTZ prevented changes in cellular glutathione levels and dialysate cell counts during acute peritonitis in rats. During chronic dialysis in rats exposed to intermittent peritonitis episodes, OTZ prevented increased thickening and neovascularization of the peritoneum. Our results suggest this may help to protect the peritoneal membrane during episodes of peritonitis.

Role of glutathione metabolism in the glutamate-induced programmed cell death of neuronal-like PC12 cells

In addition to its well-known interaction with ionotropic and metabotropic receptors, glutamate may, at high concentrations, interfere with a cystine-glutamate antiport designated as X c − and lead to a significant decrease in cystine uptake and intracellular glutathione level. These effects, in turn, may induce death in various cellular bodies including astrocytes, rat glioma cells and cortical neurons in culture. In the present paper we demonstrate that the toxicity evoked by glutamate in a neuronal-like model is indeed related to the metabolism of glutathione since glutamate toxicity is preceded by a significant depletion of intracellular glutathione and is abolished in the presence of precursors of glutathione synthesis such as cystine and N-acetylcysteine. It also appears that prolonged incubation in cystine-free medium leads to cell detachment and death, a phenomenon which is progressively abolished in the presence of increasing concentrations of cystine. In addition, buthionine sulfoximine, a known inhibitor of glutathione synthesis, also induces cell lysis with a time-course very similar to that of glutamate. However, depletion of glutathione is probably not sufficient to trigger the death signal since cycloheximide, which inhibits the toxic effect of both glutamate and buthionine sulfoximine, does not block the decrease in cellular glutathione content induced by these drugs. Our results therefore confirm that oxidative stress and intracellular glutathione depletion are able to trigger programmed cell death in neuronal-like cells, although the exact nature of the death mechanisms remains largely unknown.

Potentiation of cocaine hepatotoxicity in human hepatocytes by ethanol

The cytotoxic effect of cocaine on human hepatocytes was evaluated after 24 hr of exposure to cocaine by measuring the leakage of intracellular lactate dehydrogenase and the reduction of MTT. According to these endpoint parameters, the half-maximal cytotoxic concentration (IC 50) of cocaine was 6.8 and 7.8 m m, respectively. Lower concentrations of cocaine, however, impaired basic metabolic functions of human hepatocytes. Exposure of cells to 2 m m-cocaine resulted in a 50% decrease in hepatic glycogen, a 40% decrease in cellular glutathione content and a 40% decrease in urea synthesis with respect to control values. Ethanol greatly potentiated cocaine-induced hepatotoxicity. After a 48-hr pretreatment of human hepatocytes with 50 m m-ethanol, concentrations of cocaine (0.25 m m) that had no effects on hepatocyte metabolism in the absence of ethanol, caused a 20% inhibition of the urea synthesis rate, a 40% depletion of glycogen stores and a 30% reduction in glutathione content. The results of our work show that ethanol significantly increases the toxic effects of cocaine on human hepatocytes.

Potentiation of cocaine hepatotoxicity by ethanol in human hepatocytes

The hepatotoxic effects of cocaine on the human liver and the effect of ethanol on cocaine-induced hepatotoxicity have been examined in adult human hepatocytes cultured in chemically defined conditions. Cultures were exposed to concentrations of cocaine ranging from 10 −2 to 10 −5 m. Cytotoxicity was evaluated after 24 hr of continuous exposure to cocaine by measuring the leakage of intracellular LDH and the ability of cells to reduce MTT. According to these end-point parameters, half-maximal cytotoxic concentrations of cocaine for human hepatocytes (IC50) were 6.8 and 7.8 m m, respectively. Lower concentrations of cocaine, however, impaired basic metabolic functions of human hepatocytes. Exposure of cells to 2 m m cocaine for 24 hr resulted in a 50% decrease in hepatic glycogen, a 40% decrease in cellular glutathione content, and a 40% decrease in urea synthesis with respect to control values. For most of the metabolic parameters assayed, significant alterations were observed at 0.5 m m cocaine. Glycogen reloading of hepatocytes began to be inhibited in the presence of 0.60 m m cocaine (IC10). Ethanol greatly potentiated cocaine-induced hepatotoxicity. After a 48 hr pretreatment of human hepatocytes with 50 m m ethanol, low concentrations of cocaine (0.25 m m) that had no effects on hepatocyte metabolism in the absence of ethanol caused a 20% inhibition of the urea synthesis rate, a 40% degradation of glycogen stores, and a 30% reduction in glutathione content. The results of our work show that ethanol increases the effect of cocaine on human hepatocytes by a factor of 10.

Alteration of Glutathione Level in Human Melanoma Cells: Effect of N‐Acetyl‐L‐Cysteine and its Analogues

The effects of N-acetyl-L-cysteine (L-NAC), N,N-diacetyl-L-cystine (oxidized form of L-NAC) and N-acetyl-D-cysteine on the intracellular glutathione (GSH) level and their toxicity were investigated in the human melanoma cell culture IGR1. L-NAC applied in 3 mM concentration for 24 hr decreased; when applied for 48 hr it did not alter the intracellular GSH level. Treatment with 1 mM L-NAC for 24 hr had no effect on cellular glutathione, whereas the same concentration applied for 48 hr resulted in an increase in the level of GSH. Both concentrations also induced cell injury as determined by protein assay and trypan blue staining. N,N-diacetyl-L-cystine (0.5 and 1.5 mM, 24 hr) induced a decrease in cellular glutathione content without any apparent cell toxicity. D-NAC (1 and 3 mM, 24 hr) did not influence the GSH level of the melanoma cells; however, it had toxic effects resulting in cell loss.
