Optimized Feeding Strategies for Biosurfactant Production from Acetate by Alcanivorax borkumensis SK2

Biosurfactants are much-discussed alternatives to petro- and oleochemical surfactants. Alcanivorax borkumensis, a marine, Gram-negative γ-proteobacterium, produces a glycine-glucolipid biosurfactant from hydrocarbons, pyruvate, and acetate as carbon sources. Sustainable acetate production from lignocellulose or syngas adds to its relevance for the bioeconomy. This study investigated nitrogen sources and carbon-to-nitrogen ratios (C/N) to optimize fed-batch fermentation for biosurfactant production using A. borkumensis with acetate as the carbon source. Urea enabled high biosurfactant production, which was confirmed in DO-based fed-batch fermentation. Varying C/N ratios led to increased glycine-glucolipid production and decreased biomass production, with improvement plateauing at a C/N ratio of 26.7 Cmol Nmol−1. pH-stat fed-batch fermentation using glacial acetic acid as the carbon source and a pH-adjusting agent doubled the biosurfactant production. Finally, bubble-free membrane aeration was used to prevent extensive foam formation observed during conventional bubble aeration. The efficient production made it possible to investigate the bioactivity of glycine-glucolipid in combination with antibiotics against various microorganisms. Our findings allow for the leverage of glycine-glucolipid biosurfactant production using acetate as a carbon source.

Acetylene Tunes Microbial Growth During Aerobic Cometabolism of Trichloroethene.

Microbial aerobic cometabolism is a possible treatment approach for large, dilute trichloroethene (TCE) plumes at groundwater contaminated sites. Rapid microbial growth and bioclogging pose a persistent problem in bioremediation schemes. Bioclogging reduces soil porosity and permeability, which negatively affects substrate distribution and contaminant treatment efficacy while also increasing the operation and maintenance costs of bioremediation. In this study, we evaluated the ability of acetylene, an oxygenase enzyme-specific inhibitor, to decrease biomass production while maintaining aerobic TCE cometabolism capacity upon removal of acetylene. We first exposed propane-metabolizing cultures (pure and mixed) to 5% acetylene (v v-1) for 1, 2, 4, and 8 d and we then verified TCE aerobic cometabolic activity. Exposure to acetylene overall decreased biomass production and TCE degradation rates while retaining the TCE degradation capacity. In the mixed culture, exposure to acetylene for 1-8 d showed minimal effects on the composition and relative abundance of TCE cometabolizing bacterial taxa. TCE aerobic cometabolism and incubation conditions exerted more notable effects on microbial ecology than did acetylene. Acetylene appears to be a viable approach to control biomass production that may lessen the likelihood of bioclogging during TCE cometabolism. The findings from this study may lead to advancements in aerobic cometabolism remediation technologies for dilute plumes.

Differential responses of two fenugreek (Trigonella foenum-graecum L.) landraces pretreated with melatonin to prolonged drought stress and subsequent recovery

BackgroundDrought impairs growth, disturbs photosynthesis, and induces senescence in plants, which results in crop productivity reduction and ultimately jeopardizes human food security. The objective of this study was to determine major parameters associated with drought tolerance and recovery ability of fenugreek (Trigonella foenum-graecum L.), by examining differential biochemical and phenological responses and underlying enzyme activities as well as melatonin roles during drought stress and re-watering for two contrasting landraces. Moreover, the relative expression of three key genes involved in the biosynthesis pathway of diosgenin, including SQS, CAS, and BG, was investigated.ResultsDepending on the conditions, drought stress enhanced the activity of antioxidant enzymes and the osmoregulating compounds, non-enzymatic antioxidants, hydrogen peroxide content, and lipid peroxidation levels in most cases. Severe drought stress accelerated flowering time in Shushtar landrace (SHR) but had no significant effects on Varamin (VR). Pretreatment with melatonin delayed flowering time in SHR and caused high drought resistance in this landrace. Furthermore, melatonin significantly enhanced drought adaptability in VR by improving plant recovery ability.DiscussionBased on our results plants’ responses to drought stress and melatonin pretreatment were completely landrace-specific. Drought stress caused an increase in the relative expression of CAS gene and ultimately the accumulation of steroidal saponins in SHR. Melatonin compensated for the decrease in biomass production due to drought stress and finally increased steroidal saponins performance in SHR. Our study showed that melatonin can improve drought stress and recovery in fenugreek, but different factors such as genotype, melatonin concentration, and plant age should be considered.

Physiological and agronomical traits effects of titanium dioxide nanoparticles in seedlings of Solanum lycopersicum L

BackgroundTitanium dioxide nanoparticles (TiO2 NPs) have been reported to have contrasting effects on plant physiology, while their effects on sugar, protein, and amino acid metabolism are poorly understood. In this work, we evaluated the effects of TiO2 NPs on physiological and agronomical traits of tomato (Solanum lycopersicum L.) seedlings. Tomato seeds were treated with TiO2 NPs (1000 and 2000 mg L− 1), TiO2 microparticles (µPs, 2000 mg L− 1) as the size control, and ultrapure water as negative control.ResultsThe dry matter of stems (DMs), leaves (DMl) and total dry matter (DMt) decreased as particle concentration increased. This trend was also observed in the maximum quantum yield of light-adapted photosystem II (PSII) (Fv´/Fm´), the effective quantum yield of PSII (ΦPSII), and net photosynthesis (Pn). The concentrations of sugars, total soluble proteins, and total free amino acids were unaffected, but there were differences in the daily dynamics of these compounds among the treatments.ConclusionOur results suggest that treating tomato seeds with TiO2 might affect PSII performance, net photosynthesis and decrease biomass production, associated with a concentration- and size-related effect of TiO2 particles.

Recycling of nutrient medium to improve productivity in large-scale microalgal culture using a hybrid electrochemical water treatment system

Recycling and reusing of nutrient media in microalgal cultivation are important strategies to reduce water consumption and nutrient costs. However, these approaches have limitations, e.g., a decrease in biomass production, (because as reused media can inhibit biomass growth). To address these limitations, we applied a novel membrane filtration‒electrolysis‒ultraviolet hybrid water treatment method capable of laboratory-to-large-scale operation to increase biomass productivity and enable nutrient medium disinfection and recycling. In laboratory-scale experiments, electrolysis effectively remove the biological contaminants from the spent nutrient medium, resulting in a high on-site removal efficiency of dissolved organic carbon (DOC; 80.3 ± 5 %) and disinfection (99.5 ± 0.2 %). Compared to the results for the recycling of nutrient medium without water treatment, electrolysis resulted in a 1.5-fold increase in biomass production, which was attributable to the removal of biological inhibitors from electrochemically produced oxidants (mainly OCl−). In scaled-up applications, the hybrid system improved the quality of the recycled nutrient medium, with 85 ± 2 % turbidity removal, 75 ± 3 % DOC removal, and 99.5 ± 2 % disinfection efficiency, which was beneficial for biomass growth by removing biological inhibitors. After applying the hybrid water treatment method, we achieved a Spirulina biomass production of 0.47 ± 0.03 g L−1, similar to that obtained using a fresh medium (0.53 ± 0.02 g L−1). The on-site disinfection process described herein is practical and offers a cost-saving and environmental friendly alternative for nutrient medium recycling and reusing water in mass and sustainable cultivation of microalgae.

Comparison of Salt Stress Tolerance among Two Leaf and Six Grain Cultivars of Amaranthus cruentus L.

Amaranths (Amaranthus L.) are multi-use crop species renowned for their nutritional quality and their tolerance to biotic and abiotic stresses. Since the soil salinity of croplands is a growing problem worldwide, we tested the salinity tolerance of six grain and two leaf cultivars of Amaranthus cruentus L. The plants were grown for 53 days under hydroponic conditions at 0, 50 and 100 mM NaCl. We investigated the growth rate, photosynthetic activity, mineral content, pigments and biochemical compounds involved in oxidative stress. Although 100 mM NaCl always decreased biomass production, we highlighted Don Leon and K91 as tolerant cultivars under moderate salt stress (50 mM NaCl). Under salinity, sodium accumulated more in the shoots than in the roots, particularly in the stems. Sodium accumulation in the plants decreased the net photosynthetic rate, transpiration rate and stomatal conductance but increased water use efficiency, and it decreased chlorophyll, betalain and polyphenol content in the leaves. It also decreased the foliar content of calcium, magnesium and potassium but not the iron and zinc content. The physiological parameters responded differently to sodium accumulation depending on the cultivar, suggesting a different relative importance of ionic and osmotic phases of salt stress among cultivars. Our results allowed us to identify the morpho-physiological traits of the cultivars with different salt tolerance levels.

PHYTOREMEDIATION AND EXTRACTION OF COLLOIDAL CONTAMINANTS FROM MINE EFFLUENTS THROUGH LEGUME PLANTS

The bioaccumulation and environmental toxicity of colloidal contaminants have always been the focus of extensive research. Here we report efficient extraction of iron oxide colloidal contaminants from mine drainage effluents by phytoremediation with legume plants. The smallest fine particles with size less than 10 nm exhibit two wide absorption peaks at 350 and 495 nm, respectively. They have good dispersibility in aqueous environment and are highly resistant to removing or purification by any gravitational settings. The ultra-small nanosized particles (with average diameter 2-3 nm) can be bioaccumulated into the roots of soybean sprouts and might cause physiological stress and inhibition of the germination process. All experimental data found delayed growth, as well as decreased biomass production, which was observed in both the stem and root system of treated sprouts. Nevertheless, the immersed in mine effluent plants showed some resistance to the colloid toxicity. The light microscopic analysis also revealed the occurrence of more intensive symbiosis between the root tubers and the rhizosphere microbial communities in the case of phytorenmediation. The absorbance spectroscopic measurement demonstrated about 90 % efficient colloidal removement from the suspension within 10 days treatment. The atomic absorption spectroscopy analysis of mineralised sprouts cultured in synthetic mine effluents found that the highest concentration of accumulated iron in the legume plants was 258,02 ± 23,48 mg/100 g, which was achieved in suspension of 25 mM FeSO4. The presented report enables potential practical application of legume plants in the circumneutral passive treatment systems for remediation of mine effluents.

Modeling‐based performance assessment of an indigenous macro‐catchment water harvesting technique (Marab) in the Jordanian Badia

AbstractWater resources management is fundamental for rural communities in drylands, where water harvesting technologies (WHT) can be used for intercepting surface runoff and storing water in soils. The so‐called “Marab” WHT was initially developed by Middle Eastern agro‐pastoralists that reside or commute in semi‐arid and arid rangelands. The Marab WHT is a macro‐catchment measure consisting of earth dams and stone spillways along the contours of a lowland depression or floodplain. Dependent on the local context (i.e., climate, soil, management, etc.), the established Marabs show highly variable effectiveness and little scientific evidence is supporting the scaling out of the technology. This study aims at filling the knowledge gap on the Marab performance in different environments by simulating its hydro‐agrological effects for different soils and climatic conditions using the AquaCrop model. A case study performed for a Jordanian Marab over three seasons (2019–2022) confirms its huge improvement potential for barley production. Through Marab‐based farming, barley production reached 8.37 t ha−1on average, versus highly variable 0.34 t ha−1without the WHT. The simulation‐based assessment of soil textures identified that silty soils have the largest potential for producing up to 9.25 t ha−1barley, compared to 6.60 t ha−1produced in clay soils. Assessing different climate scenarios, a slight increase in daily average temperatures (+0.5°C) led to a considerable production decline of 4%–8%, while a significant reduction of precipitation (−20%) decreased biomass production by a similar rate (4%–10%). This underlines the robustness of the “Marab” WHT to rainfall amount variation. However, simulations also highlight the sensitivity of timing and frequency of flood events: removing the last and the first flood event reduced biomass production by approximately 50% and 80%, respectively, while the barley fails to develop if both events were suppressed.

Impact of the combined effect of seawater exposure with wastewater and Fe2O3 nanoparticles on Chlorella vulgaris microalgae growth, lipid content, biochar, and bio-oil production

Microalgae is one the promising source of energy for the production of biofuel and other value-added products to replace the existing conventional fossil fuels. However, low lipid content and poor cell harvesting are the key challenges. Based on the growth conditions the lipid productivity will be affected. The current study examines the mixtures of both wastewater and NaCl on the microalgae growth was studied. The microalgae used for conducting the tests were Chlorella vulgaris microalgae. Mixtures of the wastewater was prepared under the different concentrations of the seawater, classified as S0%, S20%, and S40%. The growth of microalgae was studied in the presence of these mixtures, and the addition of Fe2O3 nanoparticles was included to stimulate the growth. The results showed that increasing the salinity in the wastewater resulted in decreased biomass production, but significantly increased lipid content compared to S0%. The highest lipid content was recorded at S40%N with 21.2%. The Highest lipid productivity was also witnessed for S40% with 45.6 mg/Ld. The cell diameter was also found to increase with increasing salinity content in the wastewater. The addition of Fe2O3 nanoparticles in the seawater was found to enhance the productivity of the microalgae extensively, resulting in 9.2% and 6.15% increased lipid content and lipid productivity respectively compared to conventional cases. However, the inclusion of the nanoparticles slightly increased the zeta potential of microalgal colloids, with no noticeable effects on the cell diameter or bio-oil yields. Based on these findings, Chlorella vulgaris was identified as a suitable candidate for treating wastewater with high salinity exposure.

Fungicide Effect of a Novelty Antimicrobial Biosurfactant Extract Alone or Combined with Copper Oxychloride on Botrytis cinerea Cells

Microbial contaminations represent an important issue for crop production, resulting in tons of losses worldwide every year. One of the highest-risk pathogens involved in these infections is Botrytis cinerea, which is responsible for the grey mold disease. In this study, a biosurfactant extracted from corn steep liquor (CSL), an agro-industrial residue from the corn-milling industry, was used in combination with copper oxychloride (Cu-Oxy), a copper-based pesticide, to evaluate their fungitoxic activity on B. cinerea cells. The results showed that the mixture of 2 g/L of Cu-Oxy with 16 g/L of the biosurfactant extract from CSL, named biosurfactant pesticide formulation (BS-P), helped in the reduction of colony growth, in the decrease of biomass production, and in the improvement of copper bioaccumulation on fungal mycelium in comparison with aqueous solutions of commercial and pure-copper-based fungicides. A factorial design was conducted to obtain the best operational conditions for enhancing the bioaccumulation of copper by B. cinerea cells in the presence of BS-P, resulting in a maximum copper uptake of 2122 µg Cu/g dry weight when using 1 g/L of Cu-Oxy combined with 8 g/L of CSL biosurfactant extract at an incubation temperature of 15 °C. It was also proved that, due to the absence of the biosurfactant extract, the amount of copper bio-adsorbed by fungal mycelium was considerably decreased and, therefore, so was its fungitoxic activity. These results suggest that the biosurfactant extract studied could be involved in eco-friendlier pesticide formulations, reducing the environmental impact of copper-based active principles.

Response of Sagebrush Steppe Grass Species to AMF Inoculum Sources and Biochar.

The sagebrush steppe has presented increasing levels of degradation. The addition of arbuscular mycorrhizal fungi (AMF) and biochar have been suggested to restore ecosystems. However, little is known about their effects on sagebrush steppe plants. We tested three sources of AMF inoculum: soil from a disturbed site (Inoculum A), soil from an undisturbed site (Inoculum B), and commercial inoculum (Inoculum C), all with and without biochar, to test if they could mediate growth of Pseudoroegneria spicata (native perennial), Taeniatherum caput-medusae (early seral; exotic annual) and Ventenata dubia (early seral; exotic annual) under greenhouse conditions. We measured AMF colonization and biomass. We hypothesized that the plant species would be differently affected by the inoculum types. The colonization of T. caput-medusae and V. dubia was greatest when inoculated with Inoculum A (38.8% and 19.6%). In contrast, the colonization of P. spicata was greatest with Inoculum B and Inoculum C (32.1% and 32.2). Biochar decreased biomass production but increased colonization with Inoculum A for P. spicata and V. dubia and with Inoculum C for T. caput-medusae. This study reveals the response of early and late seral sagebrush steppe grass species to contrasting sources of AMF and suggests that late seral plant species respond better to late seral inocula.

Two-stage lipid induction in the microalga Tetraselmis striata CTP4 upon exposure to different abiotic stresses

Tetraselmis striata CTP4 is a euryhaline, robust, fast-growing microalga suitable for wastewater treatment and industrial production. Lipid production was induced through a two-stage cultivation strategy: a 1st stage under standard growth-promoting conditions (100 μmol photons m−2 s−1, salinity 36 ppt and 20 °C) to achieve high biomass concentration and a 2nd stage of 6 days for lipid induction by the application of abiotic stresses such as nutrient depletion, high light intensity (200 and 400 μmol photons m−2 s−1), high salinity (75 and 100 ppt), and extreme temperatures (5 and 35 °C). Although nutrient depletion always resulted in a decrease in biomass productivity, it had also the highest impact on lipid induction. The highest lipid content (43.2%) and lipid productivity (29.2 mg L−1 d−1) were obtained using a combination of nutrient depletion and high light intensity (400 μmol m−2 s−1). The fatty acid profile was mainly composed of C16:0 (palmitic), C18:1 (oleic) and C18:2 (linoleic) acids. The low content of unsaturated fatty acids and absence of C18:3 (linolenic) acid render the oil of this microalga suitable for biodiesel production, a renewable source of energy.

Microplastics reduce microalgal biomass by decreasing single-cell weight: The barrier towards implementation at scale

Microplastics (MPs) are a widespread environmental threat, especially to aquatic and urban systems. Water quality is vital for biomass production in microalgal-based industries. Here, industrially relevant microalgae Tetraselmis suecica, Scenedesmus armatus, and Nannochloropsis gaditana were exposed to PS- and PE-MPs (polystyrene and polyethylene, respectively – 10-20 μm) contaminated waters (5 and 10 mg/L). Following industrial empirical and ecotoxicological procedures, the production period was established as four days (exponential growth phase). 27-long day experiments were conducted to determine the chronic effects of MPs contamination in microalgal biomass yields. MPs induced different responses in cell density: T. suecica decreased (up to 11 %); S. armatus showed no changes; and N. gaditana increased (up to 6 %). However, all three microalgae exhibited significant decreases in biomass production (up to 24, 48, and 52 %, respectively). S. armatus exposed to PS-MPs and N. gaditana exposed to PE-MPs were the most impacted regarding biomass production. The decrease in biomass yield was due to the reduction in single-cell weight (up to 14, 47, and 43 %), and/or the production of smaller-sized cells (T. suecica). In response to chronic exposure, microalgae showed signs of cell density adaptation. Despite cell density normalizing, biomass production was still reduced compared to biomass production in clean water. Computational modelling highlighted that MPs exposure had a concentration-dependent negative impact on microalgae biomass. The models allow the evaluation of the systematic risks that MPs impose in microalgal-based industries and stimulate actions towards implementing systems to contain/eliminate MPs contamination in the waters used in microalgae production.

Nanoarchitectonics of nitric oxide releasing supramolecular structures for enhanced antibacterial efficacy under visible light irradiation

Light‐controlled therapies offer a promising strategy to prevent and suppress infections caused by numerous bacterial pathogens. Excitation of exogenously supplied photosensitizers (PS) at specific wavelengths elicits levels of reactive oxygen intermediates toxic to bacteria. Porphyrin-based supramolecular nanostructure frameworks (SNF) are effective PS with unique physicochemical properties that have led to their widespread use in photomedicine. Herein, we developed a nitric oxide (NO) releasing, biocompatible, and stable porphyrin-based SNF (SNF-NO), which was achieved through a confined noncovalent self-assembly process based on π–π stacking. Characterization of the SNFs via scanning electron microscopy (SEM) and transmission electron microscopy (TEM) analysis showed the formation of three-dimensional, well-defined octahedral structures. These SNF-NO were shown to exhibit a red shift due to the noncovalent self-assembly of porphyrins, which also show extended light absorption to broadly cover the entire visible light spectrum to enhance photodynamic therapy (PDT). Under visible light irradiation (46 J cm−2), the SNF generates high yields of singlet oxygen (1O2) radicals, hydroxyl radicals (HO), superoxide radicals (O2), and peroxynitrite (ONOO−) radicals that have shown potential to enhance antimicrobial photodynamic therapy (APDT) against Gram-positive methicillin-resistant Staphylococcus aureus (MRSA) and Gram-negative Escherichia coli (E. coli). The resulting SNFs also exhibit significant biofilm dispersion and a decrease in biomass production. The combination of robust photosensitizer SNFs with nitric oxide-releasing capabilities is dynamic in its ability to target pathogenic infections while remaining nontoxic to mammalian cells. The engineered SNFs have enormous potential for treating and managing microbial infections.

Determination of Chlorophyll Levels of Water Kale Plants (Ipomoea aquatica Forkss) Experiencing Nutrient Deficiencies

The effect of nutrient deficiency on plants can cause a decrease in plant growth and productivity. The decrease in biomass production is related to the decrease in photosynthesis rate caused by the inhibition of chlorophyll synthesis process. This study aims to determine the chlorophyll content of kale (Ipomoea aquatica) that experienced nutrient deficiencies. The type of research used is an experiment using a completely randomized design (CRD) with three replications. The treatment given was the provision of nutrient solutions: complete, -Ca, -S, -Mg, -K, -N, -P, Fe, -Micronutrients, distilled water and water. The solvent used for maceration was DMSO. The parameters observed were chlorophyll a, chlorophyll b and total chlorophyll content of kale leaves. The results showed that the complete solution had the highest total chlorophyll content of 6.387 mg/L. While the distilled water solution has the lowest total chlorophyll content of 2.982 mg/L. The complete solution has all the nutrients needed by plants so that the growth and development process can run optimally, characterized by high chlorophyll content.

The Interaction of Fungicide and Nitrogen for Aboveground Biomass from Flag Leaf Emergence and Grain Yield Generation under Tan Spot Infection in Wheat

Pyrenophora tritici-repentis (Died.) Drechs., the causal agent of tan spot, is one of the most serious biotic diseases affecting wheat worldwide (Triticum aestivum L.). Studying the interaction between different fungicide mixtures and nitrogen (N) rates under tan spot outbreaks is of key importance for reducing aboveground biomass and grain yield losses. Taking this into account, our study took a mechanistic approach to estimating the combined effect of different fungicides and N fertilization schemes on the severity of tan spot, green leaf area index, SPAD index, aboveground biomass dynamics, and yield in a wheat crop affected at the reproductive stage. Our results indicated that reductions in green leaf area, healthy area duration (HAD), and the chlorophyll concentration (SPAD index) due to increases in the percentage of damage led to decreases in biomass production (-19.2%) and grain yield (-48.1%). Fungicides containing triazole + strobilurin + carboxamides (TSC) or triazole + strobilurin (TS) combined with high N doses showed the most efficient disease control. The positive physiological effects of TSC fungicides, such as extending the green leaf area, are probably responsible for the greater production of aboveground biomass (+29.3%), as well as the positive effects on grain yield (+15.8%) with respect to TS. Both fungicide treatments increased grains per spike, kernel weight, spikes m-2, grains m-2, and grain yield. The increase in biomass in the TSC tended to cause slighter non-significant increases in grains per spike, 1000-kernel weight and grain yield compared with TS. The linear regression revealed positive associations among the extension of HAD and biomass (+5.88 g.m-2.HAD-1.day-1), grain yield (+38 kg.ha.HAD-1.day-1), and grain number (100.7 grains m2.HAD-1.day-1), explained by the interactions of high N doses and fungicides. Our study is the first report of the positive effect of TSC fungicides with high N doses on grain yield related-traits under tan spot infections in wheat.

Utilization of Various Levels of Shading and Organic Fertilizer on Morphology, Production, and Nutrient Composition of Panicum maximum cv. Mombasa

Panicum maximum cv. Mombasa is a type of grass that is known resistant to shade and responsive to fertilization. This study aimed to determine the morphology and production of Panicum maximum cv. Mombasa on various levels of shading and organic fertilizer. The soil used in this study was brown latosol. This study used a completely randomized design (CRD) with a factorial pattern consisting of three levels of shade (0%, 36%, and 72%) and two dose factors of organic fertilizer (0 g/pot and 500 g/pot). The parameters in this study consisted of morphology (plant height, number of leaves, number of tillers), plant production (biomass production, stem production, leaf production, and leaf/stem ratio), and nutrient composition (crude protein, extract ether, gross energy, and crude fibre). All data were observed in five replicates. The results showed that the level of shade up to 72% did not affect plant morphology. An increasing level of shading showed a significant increase (p<0.05) in the number of leaves of plant. Shading level decrease (p<0.05) biomass production, leaf, stem, LSR, and crude fibre content. The fertilizer application improved (p<0.05) plant morphology and production. The best interaction between growth and production was obtained in the treatment control (without shading) and fertilization of 500 g/pot. Based on the finding of this study, Panicum maximum cv. Physiological and morphological responses of Chlorella pyrenoidosa to different exposure methods of graphene oxide quantum dots

Graphene oxide quantum dots (GOQDs) can convert the ultraviolet (200– 380 nm) into available wavelength (400– 700 nm) for microalgae cultivation. However, it has not been applied in large-scale microalgae culture due to its high cost and difficulties in recovery. This study proposed a new strategy for the sustainable use of GOQDs, namely, GOQDs solution was added to the outer sandwich of the reactor. Herein, the effects of direct and indirect exposure of different GOQDs concentrations (0, 100, and 1000 mg/L) on the microalgae culture were compared. When microalgae were directly exposed to the GOQDs, 100 mg/L of GOQDs increased the biomass production of microalgae by 24.0 %, while 1000 mg/L of GOQDs decreased biomass production by 31 %. High concentration of GOQDs (direct exposure) could cause extra oxidative stress in the microalgae cells and result in a significant reduction of pigment content. When microalgae were indirectly exposed to the GOQDs, the increased concentration of GOQDs enhanced the growth of microalgae. Compared to the blank group, 1000 mg/L of GOQDs increased the microalgae biomass production and bioenergy by 14.1 % and 40.17 %, respectively. The indirect exposure of GOQDs can effectively avoid photo-oxidation and organelle damage to the microalgae cells. Overall, the indirect exposure of GOQDs is a sustainable way for effectively promoting microalgae growth and reducing the application cost.

Pilot-scale annual production of Scenedesmus almeriensis using diluted pig slurry as the nutrient source: Reduction of water losses in thin-layer cascade reactors

The microalga Scenedesmus almeriensis was produced using diluted pig slurry as the sole nutrient source during one year using a 30 m2 thin-layer cascade photobioreactor located outdoors. Evaporation contributes significantly to the water requirements of microalgae production; approximately 1000 L of water are required to produce 1 kg of microalgal biomass. The aim of this study was to assess the potential of plastic covers to avoid or minimise water evaporation in open photobioreactors. The N–NH4+ concentrations at the inlets ranged from 40 to 100 mg L−1, depending on the experimental run. These values could not be increased because of the dark colour and turbidity of the pig slurry. The biomass productivity of the system was 17.6, 18.8, 23.4, and 17.3 g m−2·day−1 in winter, spring, summer, and autumn, respectively. Covering the reactors led to a decrease in biomass productivity that ranged from 8.5 to 16.9% depending on the season. However, water evaporation was reduced by 24.9–42.0% depending on the season. In summer, covering the reactors reduced the amount of water evaporated from 6.3 to 3.6 L m−2·day−1. In a theoretical 1 ha production facility, this would represent 2700 L of water saved per day. Overall, the processing of pig slurry using microalgae is challenging because of their variable nutrient concentration, colour and turbidity. Covering the reactors with inexpensive commercial plastics could significantly reduce the water requirements of the system.

Modulated stress to balance Nannochloropsis oculata growth and eicosapentaenoic acid production.

Two environmental parameters, temperature and light intensity, were independently used as stress modulators to enhance eicosapentaenoic acid (EPA) production by the microalga Nannochloropsis oculata, without hindering biomass production. A sinusoidal approach was used, as environmental conditions were alternated between optimum and stress status in multi-day cycles. Low temperatures (5 and 10°C) and light intensities (30 and 50μmol photons/m2/s) were tested. Results revealed that the modulated stress approach used was able to avoid decreases in biomass production. Temperature stress (10°C) presented the highest impact, increasing EPA content to 12.8 mgEPA/L, 158% more than the amount obtained in optimum (non-modulated) growth conditions at that point in time, while the lower light intensity stress was able to increase to 126% more. It is important to point out that in both cases increases in EPA amounts resulted from increased content in each individual cell and not just from increased biomass contents.
