Novel, acidic, and cold-adapted glycoside hydrolase family 8 endo-β-1,4-glucanase from an Antarctic lichen-associated bacterium, Lichenicola cladoniae PAMC 26568.

Endo-β-1,4-glucanase is a crucial glycoside hydrolase (GH) involved in the decomposition of cellulosic materials. In this study, to discover a novel cold-adapted β-1,4-D-glucan-degrading enzyme, the gene coding for an extracellular endo-β-1,4-glucanase (GluL) from Lichenicola cladoniae PAMC 26568, an Antarctic lichen (Cladonia borealis)-associated bacterium, was identified and recombinantly expressed in Escherichia coli BL21. The GluL gene (1044-bp) encoded a non-modular polypeptide consisting of a single catalytic GH8 domain, which shared the highest sequence identity of 55% with that of an uncharacterized protein from Gluconacetobacter takamatsuzukensis (WP_182950054). The recombinant endo-β-1,4-glucanase (rGluL: 38.0 kDa) most efficiently degraded sodium carboxymethylcellulose (CMC) at pH 4.0 and 45°C, and showed approximately 23% of its maximum degradation activity even at 3°C. The biocatalytic activity of rGluL was noticeably enhanced by >1.3-fold in the presence of 1 mM Mn2+ or NaCl at concentrations between 0.1 and 0.5 M, whereas the enzyme was considerably downregulated by 1 mM Hg2+ and Fe2+ together with 5 mM N-bromosuccinimide and 0.5% sodium dodecyl sulfate. rGluL is a true endo-β-1,4-glucanase, which could preferentially decompose D-cellooligosaccharides consisting of 3 to 6 D-glucose, CMC, and barley β-glucan, without other additional glycoside hydrolase activities. The specific activity (15.1 U mg–1) and kcat/Km value (6.35 mg–1 s–1mL) of rGluL toward barley β-glucan were approximately 1.8- and 2.2-fold higher, respectively, compared to its specific activity (8.3 U mg–1) and kcat/Km value (2.83 mg–1 s–1mL) toward CMC. The enzymatic hydrolysis of CMC, D-cellotetraose, and D-cellohexaose yielded primarily D-cellobiose, accompanied by D-glucose, D-cellotriose, and D-cellotetraose. However, the cleavage of D-cellopentaose by rGluL resulted in the production of only D-cellobiose and D-cellotriose. The findings of the present study imply that rGluL is a novel, acidic, and cold-adapted GH8 endo-β-1,4-glucanase with high specific activity, which can be exploited as a promising candidate in low-temperature processes including textile and food processes.

Cellulosic Materials Characterisation of Kenaf Core from Retting Process Using Thermal Analysis

The waste from the kenaf water retting; kenaf core can be used to create by-product. The characteristics of the kenaf are important to create new by-product. Cellulosic materials can be obtained from fibrous plants which known as cellulose, hemicelluloses, lignin, waxes, and several water-soluble compounds. Kenaf was categorized as one of the fibrous family plants. Hence, its cellulosic materials need to be determined before extraction process. This research was conducted to determine the cellulosic materials concentration in kenaf core using Thermal Gravimetry Analyzer (TGA) and Differential Scanning Calorimetry (DSC). As result, the percentage of degradation and decomposition of cellulosic materials were recorded and analyzed as reported in this paper.

Microbial community structures in an integrated two-phase anaerobic bioreactor fed by fruit vegetable wastes and wheat straw

The microbial community structures in an integrated two-phase anaerobic reactor (ITPAR) were investigated by 16S rDNA clone library technology. The 75L reactor was designed with a 25L rotating acidogenic unit at the top and a 50L conventional upflow methanogenic unit at the bottom, with a recirculation connected to the two units. The reactor had been operated for 21 stages to co-digest fruit/vegetable wastes and wheat straw, which showed a very good biogas production and decomposition of cellulosic materials. The results showed that many kinds of cellulose and glycan decomposition bacteria related with Bacteroidales, Clostridiales and Syntrophobacterales were dominated in the reactor, with more bacteria community diversities in the acidogenic unit. The methanogens were mostly related with Methanosaeta, Methanosarcina, Methanoculleus, Methanospirillum and Methanobacterium; the predominating genus Methanosaeta, accounting for 40.5%, 54.2%, 73.6% and 78.7% in four samples from top to bottom, indicated a major methanogenesis pathway by acetoclastic methanogenesis in the methanogenic unit. The beta diversity indexes illustrated a more similar distribution of bacterial communities than that of methanogens between acidogenic unit and methanogenic unit. The differentiation of methanogenic community composition in two phases, as well as pH values and volatile fatty acid (VFA) concentrations confirmed the phase separation of the ITPAR. Overall, the results of this study demonstrated that the special designing of ITPAR maintained a sufficient number of methanogens, more diverse communities and stronger syntrophic associations among microorganisms, which made two phase anaerobic digestion of cellulosic materials more efficient.

Investigation of thermochemistry associated with the carbon-carbon coupling reactions of furan and furfural using ab initio methods.

Upgrading furan and small oxygenates obtained from the decomposition of cellulosic materials via formation of carbon-carbon bonds is critical to effective conversion of biomass to liquid transportation fuels. Simulation-driven molecular level understanding of carbon-carbon bond formation is required to design efficient catalysts and processes. Accurate quantum chemical methods are utilized here to predict the reaction energetics for conversion of furan (C4H4O) to C5-C8 ethers and the transformation of furfural (C5H6O2) to C13-C26 alkanes. Furan can be coupled with various C1 to C4 low molecular weight carbohydrates obtained from the pyrolysis via Diels-Alder type reactions in the gas phase to produce C5-C8 cyclic ethers. The computed reaction barriers for these reactions (∼25 kcal/mol) are lower than the cellulose activation or decomposition reactions (∼50 kcal/mol). Cycloaddition of C5-C8 cyclo ethers with furans can also occur in the gas phase, and the computed activation energy is similar to that of the first Diels-Alder reaction. Furfural, obtained from biomass, can be coupled with aldehydes or ketones with α-hydrogen atoms to form longer chain aldol products, and these aldol products can undergo vapor phase hydrocycloaddition (activation barrier of ∼20 kcal/mol) to form the precursors of C26 cyclic hydrocarbons. These thermochemical studies provide the basis for further vapor phase catalytic studies required for upgrading of furans/furfurals to longer chain hydrocarbons.

Simulation Model for Composting Cellulosic (Bagasse) Substrates

A dynamic model of aerobic composting with solid substrates was developed as a first step to predict the rate of decomposition of cellulosic materials. The model was based on heat and mass transfer principles and reaction kinetics of biological processes. The relations describing the rates of reaction and the degradability potential of the substrates were determined based on laboratory-scale experiments. The model was developed using a graphical software package, Stella II® (which was able to solve feedback loops encountered in actual composting processes.The model was validated with temperature, oxygen distribution and mass degradation accompanying the composting of bagasse. Experimental setups consisted of 30 gallon drum reactors, continuously aerated. Temperature and oxygen in exhaust gases were monitored on an hourly basis and measurements of moisture content, pH and volatile solids were done on a weekly basis.The data fit with the model was excellent for mass degraded and temperatures for the first 10 days. From day 10 to 14, the deviations between model predictions (as solved with Stella II®) and actual measurements did not exceed 2.6 percent. The concentrations of oxygen in outlet gases showed fair agreement between predicted and experimental values. Simulating the model with varying airflow rates showed that the maximum mass breakdown rates occurred for airflow rates of 0.12 kg dry air/(kg dry compost.hr) while maintaining the temperatures below 50°C, through a temperature feedback control strategy.

Effects of glyphosate on cellulose decomposition in two soils

Glyphosate with an equivalent concentration of either 0, 2.16 or 8.64 kg/hm2 was sprayed on to cellulosic materials before burying in two soil types; peat (soil I) and sandy clay loam (soil II). Alternatively the soils were sprayed with 0, 20 or 150 ppm of the herbicide before burying the cellulosic material either immediately or after preincubation for 4 weeks. In soil I, the increase in glyphosate concentrations substantially reduced the decomposition of cellulosic material regardless of the method of application employed. Glyphosate at 8.64 kg/hm2 reduced the mass loss of the treated substrate by 83%. However, cellulose decomposition in soil preincubated for 4 weeks before burying was affected almost to the same extent as the untreated control. Glyphosate stimulated cellulose decomposition when substrates were buried in soil II. Mass loss in soil treated with 150 ppm increased by about 100% while when glyphosate was sprayed directly to the substrate (at 8.64 kg/hm2), the loss was about 25%.

ESR g-shift measures pyrolytic deoxygenation and the effect of flame retardants

The theory of g-factors in aromatic radicals proposed by McConnell and Robertson and the statistical theory of Boltzmann were used to show that a small decrease in the g-factor of the ESR spectrum of the pyrolysis residue of cellulosic materials is proportional to a small decrease in the ratio of the bound oxygen atoms to that of the carbon atoms in the system. An average g-factor decrement was defined and used to evaluate the effect of a flame retardant on the rate of deoxygenation in the thermal decomposition of cellulosic materials at various temperatures.
