Ovary structure and symbiotic associates of a ground mealybug, Rhizoecus albidus (Hemiptera, Coccomorpha: Rhizoecidae) and their phylogenetic implications.

The ovary structure and the organization of its symbiotic system of the ground mealybug, Rhizoecus albidus (Rhizoecidae), were examined by means of microscopic and molecular methods. Each of the paired elongated ovaries of R. albidus is composed of circa one hundred short telotrophic-meroistic ovarioles, which are radially arranged along the distal part of the lateral oviduct. Analysis of serial sections revealed that each ovariole contains four germ cells: three trophocytes (nurse cells) occupying the tropharium and a single oocyte in the vitellarium. The ovaries are accompanied by giant cells termed bacteriocytes which are tightly packed with large pleomorphic bacteria. Their identity as Brownia rhizoecola (Bacteroidetes) was confirmed by means of amplicon sequencing and fluorescence in situ hybridization techniques. Moreover, to our knowledge, this is the first report on the morphology and ultrastructure of the Brownia rhizoecola bacterium. In the bacteriocyte cytoplasm bacteria Brownia co-reside with sporadic rod-shaped smaller bacteria, namely Wolbachia (Proteobacteria: Alphaproteobacteria). Both symbionts are transmitted to the next generation vertically (maternally), that is, via female germline cells. We documented that, at the time when ovarioles contain oocytes at the vitellogenic stage, these symbionts leave the bacteriocytes and move toward the neck region of ovarioles (i.e. the region between tropharium and vitellarium). Next, the bacteria enter the cytoplasm of follicular cells surrounding the basal part of the tropharium, leave them and enter the space between the follicular epithelium and surface of the nutritive cord connecting the tropharium and vitellarium. Finally, they gather in the deep depression of the oolemma at the anterior pole of the oocyte in the form of a 'symbiont ball'. Our results provide further arguments strongly supporting the validity of the recent changes in the classification of mealybugs, which involved excluding ground mealybugs from the Pseudococcidae family and raising them to the rank of their own family Rhizoecidae.

The substance P and neurokinin-1 receptor system in human thyroid cancer: an immunohistochemical study.

To develop a new therapeutic strategy against thyroid cancer (TC), the expression of both substance P (SP) and neurokinin-1 receptor (NK-1R) must be demonstrated in TC cells. This study aims to examine by immunohistochemistry the localization of SP and the NK-1R in human TC samples (papillary, follicular, medullary, anaplastic) in metastasis and in healthy thyroid samples. SP and the NK-1R were expressed in all normal and TC samples. In healthy glands, SP was located in follicular cells (nucleus) and colloid, and NK-1R in follicular cells (cytoplasm) and stroma. In TC samples, SP was visualized in follicular cells (nucleus and cytoplasm), stroma and colloid, and NK-1R in follicular cells (cytoplasm), stroma and colloid. A semiquantitative scoring system (Allred Unit Scoring System) was applied. The expression (Allred total score) of SP and NK-1R was weaker in normal thyroid glands than in TC. In comparison with TC samples, a lower intensity/proportion of SP (nucleus and cytoplasm of follicular cells; stroma) was observed in normal samples. By contrast, in the colloid of TC samples the presence of SP was lower than in normal samples. In comparison with TC samples, the presence of the NK-1R in the cytoplasm of follicular cells and colloid was lower in normal thyroid samples, whereas the expression of this receptor in the stroma was higher. The results reported in this study suggest that the NK-1R could be a new target for the treatment of TC and use of the NK-1R antagonists could serve as a new anti-TC therapeutic strategy.

Expression and regulation of Smad2 by gonadotropins in the protogynous hermaphroditic ricefield eel (Monopterus albus).

Smad2, a receptor-activated Smad, plays a critical role in regulating gametogenesis. In this study, a smad2 homologue was identified and sequenced from ricefield eel ovary cDNA, and its mRNA and protein expression levels were analysed during oocyte development. The cDNA sequence of ricefield eel smad2 consisted of 1863bp encoding a 467-amino acid protein that had high sequence homology with Smad proteins in other teleosts, especially in Poeciliopsis prolifica. The results of real-time quantitative PCR (RT-qPCR) analysis revealed that smad2 is expressed in the ovary during gonad development, increased continuously until the early vitellogenic stage in the ovaries, and then decreased with ovary maturation. Smad2 protein immunoreactivity was localized in the cytoplasm of follicular cells, oogonia, and primary growth stage oocytes. In vitro experiments revealed that follicle-stimulating hormone (FSH) and human chorionic gonadotropin (hCG) promoted smad2 expression in ovary tissue in a time- and dose-dependent manner, respectively. In summary, Smad2 plays a potentially vital role in ricefield eel ovary development.

Evidence for a transcellular route for vitellogenin transport in the telotrophic ovary of Podisus nigrispinus (Hemiptera: Pentatomidae)

Vitellogenin is the main yolk precursor protein in insect oocytes. It is synthesized in the fat body and released into the hemolymph. To reach the oocyte surface, vitellogenin must cross a single layer of follicular epithelium cells. The transport of vitellogenin across the follicular epithelium has been suggested to occur through the enlarged intercellular spaces (patency) by a paracellular route or by endocytosis by follicular cells and release onto oocyte surface in a transcelluar route. In this study, we investigated whether vitellogenin transport in the meroistic telotrophic ovary of Podisus nigrispinus (Hemiptera) occurs via a paracellular or transcellular route. Light and transmission electron microscopies showed that short cell–cell contacts with well-developed occluding septate junctions were present in follicular cells with patency. Immunofluorescence microscopy revealed the presence of vitellogenin receptors in the plasma membrane and of vitellogenin in the cytoplasm of follicular cells. Data suggest that cell–cell contacts serve as a barrier to large vitellogenin molecules and that this protein is transported via a transcellular route of receptor-mediated endocytosis.

Vitellogenin transcytosis in follicular cells of the honeybee Apis mellifera and the wasp Polistes simillimus.

Vitellogenin receptor (VgR) is a low-density lipoprotein receptor responsible for the mediated endocytosis of vitellogenin (Vg) during egg formation in insects. The maturing oocyte is enveloped by a follicular epithelium, which has large intercellular spaces during Vg accumulation (patency). However, Vg has been reported in the cytoplasm of follicular cells, indicating that there may be a transcellular route for its transport. This study verified the presence of VgR in the follicular cells of the ovaries of the honeybee Apis mellifera and the wasp Polistes simillimus in order to evaluate if Vg is transported via transcytosis in these insects. Antibodies specific for vitellogenin receptor (anti-VgR), vitellogenin (anti-Vg), and clathrin (anti-Clt) were used for immunolocalization. The results showed the presence of VgR on the apical and basal plasma membranes of follicular cells of the vitellogenic follicles in both species, indicating that VgR may have been transported from the basal to the apical cell domain, followed by its release into the perivitelline space, evidenced by the presence of apical plasma membrane projections containing VgR. Co-localization proved that Vg bind to VgR and that the transport of this protein is mediated by clathrin. These data suggest that, in these social insects, Vg is transported via clathrin-mediated VgR transcytosis in follicular cells.

Effect of follicle cell autophagy on gonadal development of triploid female rainbow trout (Oncorhynchus mykiss).

Autophagy is a cellular process which occurs in eukaryotic cells. To study the mechanism regulating polyploid fish growth and development is of significance in genetic, because of its growth advantages and economic values. This study focused on triploid female rainbow trout (RBT) which discusses the effects of autophagy on gonadal development of polyploid fish. Autophagy-related genes of RBT lc3b, atg12, atg4b, gabarap1, and bcl2 were cloned, and autophagy gene expressions in gonads were analyzed at different developmental period. Gonadal ultrastructures were observed under transmission electron microscopy. To detect autophagy protein expression and localization, antibodies of RBT-LC3B and RBT-ATG12 were produced. Results showed clear evidence that autophagy-related genes were highly expressed during 200-300 days post fertilization (dpf), in which autophagosome structures were identified. In this stage, the conversion of LC3B-I to LC3B-II was greater than those in other stages. Immunolabeling-manifested autophagy occurred intensively in the cytoplasm of follicular cells. The morphology of follicular cells was gradually changed, leading to gonadal fibrosis and regression. This autophagic research is a new study area on gonadal development of polyploid fish.

Immunohistochemical, morphological and histometrical analyses of follicular development in Astyanax bimaculatus (Teleostei: Characidae) exposed to an organochlorine insecticide

Thiodan® is an organochlorine insecticide used in agriculture that can reach aquatic ecosystems where it can affect fish reproduction. This research aimed to evaluate follicular development and the expression of integrin β1, collagen type IV and caspase 3 by morphological, histometrical and immunohistochemical analyses of Astyanax bimaculatus exposed to Thiodan®. Treatments included three sublethal concentrations of Thiodan® (1.15, 2.30, and 5.60μgL−1) for 96h and a control without the insecticide. Mature females with ovaries in advanced follicular development were chosen for study (average weight: 11.52 ± 2.0g; average total length: 9.12 ± 0.64cm). The follicles of A. bimaculatus exhibited normal morphology, while the diameters of secondary follicles showed an increase (P< 0.05) in all concentrations when compared to the control group; a characteristic of follicles undergoing the initial stages of intoxication. Immunohistochemical analysis revealed secondary follicles with greater expression of integrin β1 and collagen type IV in cytoplasm of follicular cells than in the primary follicles in all treatments and in the control. Immunolocalization of caspase 3 was detected in follicular cells during secondary development in all tested concentrations of Thiodan® and the control. These analyses demonstrate positive immunolocalization throughout the course of follicular development, even in fish exposed to varying concentrations of Thiodan® for 96h, demonstrating that follicular cells retain their physiological integrity.

Sulcia symbiont of the leafhopper Macrosteles laevis (Ribaut, 1927) (Insecta, Hemiptera, Cicadellidae: Deltocephalinae) harbors Arsenophonus bacteria.

The leafhopper Macrosteles laevis, like other plant sap-feeding hemipterans, lives in obligate symbiotic association with microorganisms. The symbionts are harbored in the cytoplasm of large cells termed bacteriocytes, which are integrated into huge organs termed bacteriomes. Morphological and molecular investigations have revealed that in the bacteriomes of M. laevis, two types of bacteriocytes are present which are as follows: bacteriocytes with bacterium Sulcia and bacteriocytes with Nasuia symbiont. We observed that in bacteriocytes with Sulcia, some cells of this bacterium contain numerous cells of the bacterium Arsenophonus. All types of symbionts are transmitted transovarially between generations. In the mature female, the bacteria Nasuia, bacteria Sulcia, and Sulcia with Arsenophonus inside are released from the bacteriocytes and start to assemble around the terminal oocytes. Next, the bacteria enter the cytoplasm of follicular cells surrounding the posterior pole of the oocyte. After passing through the follicular cells, the symbionts enter the space between the oocyte and follicular epithelium, forming a characteristic “symbiont ball.”

Bacterial symbionts of the leafhopper Evacanthus interruptus (Linnaeus, 1758) (Insecta, Hemiptera, Cicadellidae: Evacanthinae).

Plant sap-feeding hemipterans harbor obligate symbiotic microorganisms which are responsible for the synthesis of amino acids missing in their diet. In this study, we characterized the obligate symbionts hosted in the body of the xylem-feeding leafhopper Evacanthus interruptus (Cicadellidae: Evacanthinae: Evacanthini) by means of histological, ultrastructural and molecular methods. We observed that E. interruptus is associated with two types of symbiotic microorganisms: bacterium ‘Candidatus Sulcia muelleri’ (Bacteroidetes) and betaproteobacterium that is closely related to symbionts which reside in two other Cicadellidae representatives: Pagaronia tredecimpunctata (Evacanthinae: Pagaronini) and Hylaius oregonensis (Bathysmatophorinae: Bathysmatophorini). Both symbionts are harbored in their own bacteriocytes which are localized between the body wall and ovaries. In E. interruptus, both Sulcia and betaproteobacterial symbionts are transovarially transmitted from one generation to the next. In the mature female, symbionts leave the bacteriocytes and gather around the posterior pole of the terminal oocytes. Then, they gradually pass through the cytoplasm of follicular cells surrounding the posterior pole of the oocyte and enter the space between them and the oocyte. The bacteria accumulate in the deep depression of the oolemma and form a characteristic ‘symbiont ball’. In the light of the results obtained, the phylogenetic relationships within modern Cicadomorpha and some Cicadellidae subfamilies are discussed.

Chaetal loss and replacement inPseudopotamilla reniformis(Sabellida, Annelida)

Chaetae are continually being replaced during the life of a polychaete, but the process of chaetal degeneration during replacement is still poorly known. Chaetal loss occurs either one at a time through special degenerative sites, or all chaetae in a chaetal sac are lost simultaneously in the absence of degenerative sites. Old chaetae can be either resorbed inside the coelom or shed. This study describes chaetal degradation, loss, and subsequent replacement during experimentally induced regeneration in the sabellid polychaete Pseudopotamilla reniformis. During post-traumatic regeneration in P. reniformis, abdominal-type chaetae fall from the chaetal sac simultaneously without degenerative sites, and are replaced by thoracic-type chaetae. Chaetal sac degradation occurs in three main stages: (1) formation of numerous lysosomes and small electron-transparent vesicles in follicle cell cytoplasm; (2) disintegration of follicular cell membranes; and (3) disintegration of follicular cell cytoplasm. Description of changes during parapodial transformation allows identification of signs of degradation that can serve as histological indicators of chaetal degeneration in a wide range of polychaetes. Our study suggests that chaetal degeneration and loss in polychaetes can occur either with degenerative sites or without them, depending on the type of chaetal replacement.

The natural insect peptide Neb-colloostatin induces ovarian atresia and apoptosis in the mealworm Tenebrio molitor

BackgroundThe injection of Neb-colloostatin into T. molitor females causes gonadoinhibitory effects on ovarian development. This peptide inhibits intercellular space formation (patency) in follicular epithelium and results in slowed vitellogenesis, delayed ovulation, reduced number of eggs laid and presumably cell death in the terminal follicles. However, as does the form of cell death in the terminal follicle, the mode of action of Neb-colloostatin remains unknown.ResultsWe tested Neb-colloostatin for a sterilizing effect on females of Tenebrio molitor. We report that injection of nanomolar doses of Neb-colloostatin induce ovarian follicle atresia in 4-day old females during their first gonadotropic cycle. Light microscope observations revealed morphological changes in the ovary: after Neb-colloostatin injection the terminal oocytes are significantly smaller and elicit massive follicle resorption, but the control terminal follicles possess translucent ooplasm in oocytes at different stages of vitellogenesis. A patency is visible in follicular epithelium of the control vitellogenic oocytes, whereas peptide injection inhibits intercellular space formation and, in consequence, inhibits vitellogenesis. Confocal and electron microscope examination showed that peptide injection causes changes in the morphology indicating death of follicular cells. We observed F-actin cytoskeleton disorganization, induction of caspase activity, changes in chromatin organization and autophagic vacuole formation. Moreover, the apical cytoplasm of follicular cells is filled with numerous free ribosomes, probably indicating a higher demand for protein biosynthesis, especially in preparation for autophagic vacuole formation. On the other hand, the process of polyribosomes formation is inhibited, indicating the contributing effect of this hormone.ConclusionNeb-colloostatin induces atresia in the mealworm ovary. Degeneration of T. molitor follicles includes changes in morphology and viability of follicular cells, and oosorption as a consequence of these changes.

Expression profiles and localization of vitellogenin mRNA and protein during ovarian development of the giant tiger shrimp Penaeus monodon

Ovarian maturation of the giant tiger shrimp (Penaeus monodon) results from rapid synthesis and accumulation of a major yolk protein, vitellin which is derived from its precursor, vitellogenin. The expression profiles and localization of P. monodon vitellogenin 1 (PmVtg1) were examined. PmVtg1 mRNA was expressed only in ovaries and hepatopancreas but not in other tissues of female and testes of male broodstock. In wild intact broodstock, PmVtg1 mRNA was expressed at a low level in stage I ovaries and up-regulated in stages II and III being significantly reduced in stage IV ovaries (P<0.05). In eyestalk-ablated broodstock, the expression level of PmVtg1 mRNA was increased to nearly a peak level in vitellogenic (stage II) ovaries and peaked in cortical rod (stage III) ovaries (P<0.05). The level of PmVtg1 mRNA in hepatopancreas of intact shrimp with stages II and III ovaries was significantly greater than those with stages IV and V (post-spawning) ovaries. Interestingly, PmVtg1 mRNA in hepatopancreas was 25–40 times higher than that in ovaries of intact shrimp with stages I–III of ovarian development. In situ hybridization revealed that PmVtg1 mRNA was clearly localized in the cytoplasm of follicular cells surrounding late previtellogenic, vitellogenic and cortical rod oocytes. Quantitative real-time PCR against different periods of spawned eggs suggested that PmVtg1 mRNA was also synthesized in oocytes. Western blot analysis revealed a similar profile of ovarian P. monodon vitellin and PmVtg1 mRNA during ovarian development and also suggested that the Vtg protein in hepatopancreas and Pm-vitellin in ovaries should have encoded from different Vtg genes. Immunohistochemistry confirmed that Pm-vitellin was localized in developing, vitellogenic and mature oocytes but not in the follicular cells.

Molecular cloning and expression of progestin membrane receptor component 1 ( Pgmrc1) of the giant tiger shrimp Penaeus monodon

Knowledge on molecular mechanisms of steroid hormonal induction on oocyte development may lead to the possible ways to effectively induce ovarian maturation in shrimp. In this study, progestin membrane receptor component 1 ( Pgmrc1) of the giant tiger shrimp ( Penaeus monodon) initially identified by EST analysis was further characterized. The full-length cDNA of Pgmrc1 was 2015 bp in length containing an ORF of 573 bp corresponding to a polypeptide of 190 amino acids. Northern blot analysis revealed a single form of Pgmrc1 in ovaries of P. monodon. Quantitative real-time PCR indicated that the expression level of Pgmrc1 mRNA in ovaries of both intact and eyestalk-ablated broodstock was greater than that of juveniles ( P < 0.05). Pgmrc1 was up-regulated in mature (stage IV) ovaries of intact broodstock ( P < 0.05). Unilateral eyestalk ablation resulted in an earlier up-regulation of Pgmrc1 since the vitellogenic (II) ovarian stage. Moreover, the expression level of Pgmrc1 in vitellogenic, early cortical rod and mature (II–IV) ovaries of eyestalk-ablated broodstock was greater than that of the same ovarian stages in intact broodstock ( P < 0.05). Pgmrc1 mRNA was clearly localized in the cytoplasm of follicular cells, previtellogenic and early vitellogenic oocytes. Immunohistochemistry revealed the positive signals of the Pgmrc1 protein in the follicular layers and cell membrane of follicular cells and various stages of oocytes. Taken the information together, Pgmrc1 gene products seem to play the important role on ovarian development and may be used as the bioindicator for monitoring progression of oocyte maturation of P. monodon.

Ultrastructural deformity of ovarian follicles induced by different cryopreservation protocols

Ultrastructural deformities were monitored after cryopreservation of F1 (B6CBAF1) mouse ovaries by either slow freezing or vitrification. Vacuole formation in the ooplasm, zona pellucida, and the cytoplasm of follicular cells, and mitochondrial deformity were detected. These types of cryodamage demonstrated protocol specificity.

Immunohistochemical localization of NPY, VIP and 5-HT in the thyroid gland of the lizard, Podarcis sicula.

The thyroid gland of the lizard Podarcis sicula was immunohistochemically studied in adult male specimens using specific antibodies against NPY, VIP and 5-HT and the avidin-biotin peroxidase complex (ABC) procedure to localize the three peptides. Fine beaded VIP-immunoreactive nerve fibers ran between the follicles, and VIP-immunoreactivity was evenly distributed in the apical cytoplasm of follicular cells. NPY-immunoreactive fibers were found around the follicles, and, in the cells, immunoreactivity was localizated only in the cellular apices. Immunoreactivity to 5-HT was observed in the colloid, with a concentration in the follicular lumen exceeding that in the follicular cells. In fact, most follicles showed immunoreactivity in the cytoplasmic bridges formed between the apical portion of the follicular cells and the colloid.

Identifying genes related to choriogenesis in insect panoistic ovaries by Suppression Subtractive Hybridization

BackgroundInsect ovarioles are classified into two categories: panoistic and meroistic, the later having apparently evolved from an ancestral panoistic type. Molecular data on oogenesis is practically restricted to meroistic ovaries. If we aim at studying the evolutionary transition from panoistic to meroistic, data on panoistic ovaries should be gathered. To this end, we planned the construction of a Suppression Subtractive Hybridization (SSH) library to identify genes involved in panoistic choriogenesis, using the cockroach Blattella germanica as model.ResultsWe constructed a post-vitellogenic ovary library by SSH to isolate genes involved in choriogenesis in B. germanica. The tester library was prepared with an ovary pool from 6- to 7-day-old females, whereas the driver library was prepared with an ovary pool from 3- to 4-day-old females. From the SSH library, we obtained 258 high quality sequences which clustered into 34 unique sequences grouped in 19 contigs and 15 singlets. The sequences were compared against non-redundant NCBI databases using BLAST. We found that 44% of the unique sequences had homologous sequences in known genes of other organisms, whereas 56% had no significant similarity to any of the databases entries. A Gene Ontology analysis was carried out, classifying the 34 sequences into different functional categories. Seven of these gene sequences, representative of different categories and processes, were chosen to perform expression studies during the first gonadotrophic cycle by real-time PCR. Results showed that they were mainly expressed during post-vitellogenesis, which validates the SSH technique. In two of them corresponding to novel genes, we demonstrated that they are specifically expressed in the cytoplasm of follicular cells in basal oocytes at the time of choriogenesis.ConclusionThe SSH approach has proven to be useful in identifying ovarian genes expressed after vitellogenesis in B. germanica. For most of the genes, functions related to choriogenesis are postulated. The relatively high percentage of novel genes obtained and the practical absence of chorion genes typical of meroistic ovaries suggest that mechanisms regulating chorion formation in panoistic ovaries are significantly different from those of meroistic ones.

Substance P: Immunohistochemical localization and possible function in thyroid gland of the lizard podarcis sicula (reptilia, lacertidae)

Tachykinins, such as substance P (SP), are a family of regulatory peptides involved in the modulation of the morphophysiology of the thyroid gland. The paper investigated by light microscopy techniques and radioimmunoassay the effects of SP on Podarcis sicula thyroid gland morphology and release of thyroid hormones (T3 and T4) and thyroid‐stimulating hormone (TSH). Substance P significantly increased the plasma T3 and T4 levels and reduced TSH release. In agreement with these results, the morphology of the thyroid gland also appeared influenced after treatment with SP. Besides, SP was immunolocalized in the thyroid gland of P. sicula with the ABC complex. Substance P‐immunoreactive fibres ran between the follicles and SP‐immunoreactivity was evenly distributed on the apical cytoplasm of follicular cells. The results obtained indicate that this peptide may participate in the paracrine control of the P. sicula thyroid gland.

The orphan receptor BmHNF-4 of the silkmoth Bombyx mori: ovarian and zygotic expression of two mRNA isoforms encoding polypeptides with different activating domains

Two silkmoth nuclear receptor isoforms, BmHNF-4a and BmHNF-4b, that are related to the mammalian orphan receptor HNF-4, were characterized. Their characterization revealed that they differ from each other only in their 5′ UTR and N-terminus of the predicted polypeptides. In ovarian tissue, the two receptors are expressed as a delayed response to 20-hydroxy-ecdysone and their expression increases during vitellogenesis. BmHNF-4 mRNA is localized in the cytoplasm of follicular cells and a binding activity that recognizes a mammalian HNF-4 response element is present in follicular cell nuclear extracts. BmHNF-4 mRNA is also present in the oocyte, the unfertilized egg and the early embryo, thus displaying a behavior reminiscent of maternal mRNA. Both mRNA isoforms are found in the embryo following fertilization and their abundance is modulated during ensuing embryogenesis. In contrast to the rather limited distribution of HNF-4 in mammalian tissues, BmHNF-4 is expressed in most larval and pharate adult tissues of the silkmoth.

A method for a selective local application of radioactive precursors of nucleic acids to the cow ovary.

Local application of /5-3H/uridine or /6-3H/thymidine to the cow ovary was obtained by the instillation of the radioactive solution into a rubber balloon ('bursa') tightly enclosing the organ exposed by laparotomy. After 30-90 min the exteriorized ovary was replaced in the abdominal cavity. Ovaries were recovered 1-8 days later and processed for autoradiography. After /5-3H/uridine application the peripheral layer of the ovary, containing the follicles in different stages of development was most intensely labelled. The labelling by /5-3H/uridine could be attributed prevailingly to the cytoplasm of different cells constituting the ovary but in some cases the labelling was higher in nuclei. The highest level of labelling was seen in the cytoplasm of follicular cells of follicles composed of one or two layers of cuboidal cells. Labelling was also detected in the ooplasm of oocytes with the maximal number of grains being seen in the first phase of their growth. On the other hand, no labelling was detected in the ooplasm of oocytes isolated from antral follicles the growth of which had been induced previously by hormonal treatment. It is believed that the labelling obtained by the described procedure represents to a significant degree RNA. A similar local application of /6-3H/thymidine allowed us to obtain labelling restricted specifically to nuclei of different cells composing the ovarian tissue. In the presented results, the entry of flattened follicular cells of primordial follicles in vivo in the S-phase of the cell cycle was demonstrated as well as an evident different proliferation rate in the successive stages of follicle development. The method proposed here may probably be the way for testing the effects of different substances, available only in small quantities, on the cow ovary.

Development Regulation of a Silkworm Gene Encoding Multiple GATA-type Transcription Factors by Alternative Splicing

Gene BmGATAβ of the silkworm Bombyx moriwas previously shown to encode factor BCFI, which regulates the expression of a class of chorion genes expressed during the late stages of choriogenesis. We now show that the expression of the BmGATAβ gene is spatially and temporally regulated by alternative splicing that generates two major (BmGATAβ1 and BmGATAβ2) and one minor (BmGATAβ3) mRNA isoforms of non-identical tissue distribution. The three isoforms differ in the organization of the DNA-binding domains of the corresponding polypeptides. While all three isoforms are expressed in ovarian follicular cells and in testes, only one of them, BmGATAβ1, is gonad-specific. BmGATAβ2 is expressed in a variety of other larval and pupal tissues, while BmGATAβ3 is detected in some pupal but none of the larval tissues. Analysis of RNA isolated from follicular cells of developing ovarian follicles has shown that the onset of ovarian transcription for all three mRNA isoforms occurs during late vitellogenesis, and that the level of accumulated mRNA declines significantly at the onset of choriogensis. Coincident with the onset of late chorion gene expression, we have observed a significant change in the preference of splice site selection in favour of the one that results in the generation of BmGATAβ1 mRNA. The transcriptional activation of the BmGATAβ gene in follicular cells during late vitellogenesis correlates with the previously demonstrated initial accumulation of factor BCFI in the cytoplasm of follicular cells and its appearance in follicular cell nuclei only during the late stages of choriogenesis. The relationship between factor BCFI and the different polypeptides encoded by the three BmGATAβ mRNA isoforms is discussed.
