Metabolic activity of Planktothrix rubescens and its consequences on oxygen dynamics in laboratory experiment: A stable isotope study.

Fluctuations in dissolved oxygen (DO) contents in natural waters can become intense during cyanobacteria blooms. In a reconnaissance study, we investigated DO concentrations and stable isotope dynamics during a laboratory experiment with the cyanobacterium Planktothrix rubescens in order to obtain insights into primary production under specific conditions. This observation was extended to sub-daily timescales with alternating light and dark phases. Dissolved oxygen concentrations and its isotopes (δ18ODO) ranged from 0.02 to 0.06 mmol · L-1 and from +9.6‰ to +23.4‰. The δ18ODO proved to be more sensitive than concentration measurements in response to metabolic variation and registered earlier shifts to dominance by respiration. Oxygen (O2) contents in the headspace and its isotopes (δ18OO2) ranged from 2.62 to 3.20 mmol · L-1 and from +9.8‰ to +21.9‰. Headspace samples showed less fluctuations in concentration and isotope trends because aquatic processes were hardly able to alter signals once the gas had reached the headspace. Headspace δ18OO2 values were corrected for gas-water equilibration and were determinedto be higher than the mean δ18OH2O of -8.7‰. This finding suggests that counteracting respiration was important even during the highest photosynthetic activity. Additionally, headspace analyses led to the definition of a fractionation factor for respiration (αR) of this cyanobacterium with a value of 0.980. This value confirms the one commonly used for cyanobacteria. Our findings may become important for the management of water bodies where decreases in DO are caused by cyanobacteria.

Fate of Planktothrix-derived toxins in aquatic food webs: A case study in Lake Mindelsee (Germany)

Blooms of the red, filamentous cyanobacterium Planktothrix rubescens occur frequently in pre-alpine lakes in Europe, often with concomitant toxic microcystin (MC) production. Trophic transfer of MCs has been observed in bivalves, fish, and zooplankton species, while uptake of MCs into Diptera species could facilitate distribution of MCs into terrestrial food webs and habitats. In this study, we characterized a Planktothrix bloom in summer 2019 in Lake Mindelsee and tracked possible trophic transfer and/or bioaccumulation of MCs via analysis of phytoplankton, zooplankton (Daphnia) and emergent aquatic insects (Chaoborus, Chironomidae and Trichoptera). Using 16 S rRNA gene amplicon sequencing, we found that five sequence variants of Planktothrix spp. were responsible for bloom formation in September and October of 2019, and these MC-producing variants, provisionally identified as P. isothrix and/or P. serta, occurred exclusively in Lake Mindelsee (Germany), while other variants were also detected in nearby Lake Constance. The remaining cyanobacterial community was dominated by Cyanobiaceae species with high species overlap with Lake Constance, suggesting a well-established exchange of cyanobacteria species between the adjacent lakes. With targeted LC–HRMS/MS we identified two MC-congeners, MC-LR and [Asp3]MC-RR with maximum concentrations of 45 ng [Asp3]MC-RR/L in lake water in September. Both MC congeners displayed different predominance patterns, suggesting that two different MC-producing species occurred in a time-dependent manner, whereby [Asp3]MC-RR was clearly associated with the Planktothrix spp. bloom. We demonstrate an exclusive transfer of MC-LR, but not [Asp3]MC-RR, from phytoplankton into zooplankton reaching a 10-fold bioconcentration, yet complete absence of these MC congeners or their conjugates in aquatic insects. The latter demonstrated a limited trophic transfer of MCs from zooplankton to zooplanktivorous insect larvae (e.g., Chaoborus), or direct transfer into other aquatic insects (e.g. Chironomidae and Trichoptera), whether due to avoidance or limited uptake and/or rapid excretion of MCs by higher trophic emergent aquatic insects.

Cascading climate effects in deep reservoirs: Full assessment of physical and biogeochemical dynamics under ensemble climate projections and ways towards adaptation

AbstractWe coupled twenty-first century climate projections with a well-established water quality model to depict future ecological changes of Rappbode Reservoir, Germany. Our results document a chain of climate-driven effects propagating through the aquatic ecosystem and interfering with drinking water supply: intense climate warming (RCP8.5 scenario) will firstly trigger a strong increase in water temperatures, in turn leading to metalimnetic hypoxia, accelerating sediment nutrient release and finally boosting blooms of the cyanobacterium Planktothrix rubescens. Such adverse water quality developments will be suppressed under RCP2.6 and 6.0 indicating that mitigation of climate change is improving water security. Our results also suggested surface withdrawal can be an effective adaptation strategy to make the reservoir ecosystem more resilient to climate warming. The identified consequences from climate warming and adaptation strategies are relevant to many deep waters in the temperate zone, and the conclusion should provide important guidances for stakeholders to confront potential climate changes.

Complex effects of chytrid parasites on the growth of the cyanobacterium Planktothrix rubescens across interacting temperature and light gradients

Chytrids are important drivers of aquatic ecosystems as phytoplankton parasites. The interaction between these parasites and their hosts are shaped by abiotic factors such as temperature and light. Here, we performed a full-factorial experiment to study how temperature and light interact to affect the dynamics of the bloom-forming toxic cyanobacterium Planktothrix rubescens and its chytrid parasite. We used a dynamic host-parasite model to explore how temperature and light affect long term dynamics. At low temperatures, chytrids do not survive. Higher light and temperature levels stimulated both phytoplankton and chytrid growth, with complex effects on their dynamics. Model exploration indicates that increasing temperature and light shifts equilibrium outcomes from P. rubescens persisting alone to stable coexistence and then to limit cycles. This provides an alternative biological explanation for why P. rubescens is mainly found in the relatively cold and dark lake metalimnion – it may enable avoidance of its parasite. Our study emphasizes the importance of investigating how abiotic factors interact with biotic interactions to drive complex outcomes.

The Red Harmful Plague in Times of Climate Change: Blooms of the Cyanobacterium Planktothrix rubescens Triggered by Stratification Dynamics and Irradiance.

Planktothrix rubescens is a harmful planktonic cyanobacterium, forming concentrated metalimnetic populations in deep oligo- and mesotrophic lakes, even after successful restoration. In Lake Zurich (Switzerland), P. rubescens emerged as a keystone species with annual mass developments since the 1970s. Its success was partly attributed to effects of lake warming, such as changes in thermal stratification and seasonal deep mixing. However, recent observations based on a biweekly monitoring campaign (2009–2020) revealed two massive breakdowns and striking seasonal oscillations of the population. Here, we disentangle positive from negative consequences of secular lake warming and annual variations in weather conditions on P. rubescens dynamics: (i) despite the high survival rates of overwintering populations (up to 25%) during three consecutive winters (2014–2016) of incomplete deep convective mixing, cyanobacterial regrowth during the following stratified season was moderate and not overshooting a distinct standing stock threshold. Moreover, we recorded a negative trend for annual population maxima and total population size, pointing to a potential nutrient limitation after a series of incomplete winter mixing. Thus, the predication of steadily increasing blooms of P. rubescens could not be confirmed for the last decade. (ii) The seasonal reestablishment of P. rubescens was strongly coupled with a timely formation of a stable metalimnion structure, where the first positive net growth in the following productive summer season was observed. The trigger for the vertical positioning of filaments within the metalimnion was irradiance and not maximal water column stability. Repetitive disruptions of the vernal metalimnion owing to unstable weather conditions, as in spring 2019, went in parallel with a massive breakdown of the standing stock and marginal regrowth during thermal stratification. (iii) Driven by light intensity, P. rubescens was entrained into the turbulent epilimnion in autumn, followed by a second peak in population growth. Thus, the typical bimodal growth pattern was still intact during the last decade. Our long-term study highlights the finely tuned interplay between climate-induced changes and variability of thermal stratification dynamics and physiological traits of P. rubescens, determining its survival in a mesotrophic temperate lake.

Lake Ecosystem Robustness and Resilience Inferred from a Climate-Stressed Protistan Plankton Network.

Network analyses of biological communities allow for identifying potential consequences of climate change on the resilience of ecosystems and their robustness to resist stressors. Using DNA metabarcoding datasets from a three-year-sampling (73 samples), we constructed the protistan plankton co-occurrence network of Lake Zurich, a model lake ecosystem subjected to climate change. Despite several documentations of dramatic lake warming in Lake Zurich, our study provides an unprecedented perspective by linking changes in biotic association patterns to climate stress. Water temperature belonged to the strongest environmental parameters splitting the data into two distinct seasonal networks (October–April; May–September). The expected ecological niche of phytoplankton, weakened through nutrient depletion because of permanent thermal stratification and through parasitic fungi, was occupied by the cyanobacterium Planktothrix rubescens and mixotrophic nanoflagellates. Instead of phytoplankton, bacteria and nanoflagellates were the main prey organisms associated with key predators (ciliates), which contrasts traditional views of biological associations in lake plankton. In a species extinction scenario, the warm season network emerged as more vulnerable than the cold season network, indicating a time-lagged effect of warmer winter temperatures on the communities. We conclude that climate stressors compromise lake ecosystem robustness and resilience through species replacement, richness differences, and succession as indicated by key network properties.

The formation of a metalimnetic oxygen minimum exemplifies how ecosystem dynamics shape biogeochemical processes: A modelling study

Metalimnetic oxygen minima are observed in many lakes and reservoirs, but the mechanisms behind this phenomena are not well understood. Thus, we simulated the metalimnetic oxygen minimum (MOM) in the Rappbode Reservoir with a well-established two-dimensional water quality model (CE-QUAL-W2) to systematically quantify the chain of events leading to its formation. We used high-resolution measured data to calibrate the model, which accurately reproduced the physical (e.g. water level and water temperature), biogeochemical (e.g. nutrient and oxygen dynamics) and ecological (e.g. algal community dynamics) features of the reservoir, particularly the spatial and temporal extent of the MOM. The results indicated that around 60% of the total oxygen consumption rate in the MOM layer originated from benthic processes whereas the remainder originated from pelagic processes. The occurrence of the cyanobacterium Planktothrix rubescens in the metalimnion delayed and slightly weakened the MOM through photosynthesis, although its decaying biomass ultimately induced the MOM. Our research also confirmed the decisive role of water temperature in the formation of the MOM since the water temperatures, and thus benthic and pelagic oxygen consumption rates, were higher in the metalimnion than in the hypolimnion. Our model is not only providing novel conclusions about the drivers of MOM development and their quantitative contributions, it is also a new tool for understanding and predicting ecological and biogeochemical water quality dynamics.

Metalimnetic oxygen minimum and the presence of Planktothrix rubescens in a low-nutrient drinking water reservoir

Dissolved oxygen is a key player in water quality. Stratified water bodies show distinct vertical patterns of oxygen concentration, which can originate from physical, chemical or biological processes. We observed a pronounced metalimnetic oxygen minimum in the low-nutrient Rappbode Reservoir, Germany. Contrary to the situation in the hypolimnion, measurements of lateral gradients excluded the sediment contact zone from the major sources of oxygen depletion for the metalimnetic oxygen minimum. Instead, the minimum was the result of locally enhanced oxygen consumption in the open water body. A follow-up monitoring included multiple chlorophyll a fluorescence sensors with high temporal and vertical resolution to detect and document the evolution of phytoplankton. While chlorophyll fluorescence sensors with multiple channels detected a mass development of the phycoerythrin-rich cyanobacterium Planktothrix rubescens in the metalimnion, this species was overlooked by the commonly used single-channel chlorophyll sensor. The survey indicated that the waning P. rubescens fluorescence was responsible for the oxygen minimum in the metalimnion. We hypothesize that pelagic processes, i.e., either oxygen use through decomposition of dead organic material originating from P. rubescens or P. rubescens extending its respiration beyond its photosynthetic activity, induced the metalimnetic oxygen minimum. The deeper understanding of the oxygen dynamics is mandatory for optimizing reservoir management.

Rapid detection and quantification of the potentially toxic cyanobacterium Planktothrix rubescens by in-vivo fluorometry and flow cytometry

We combined profiling of the bloom-forming and potentially toxigenic cyanobacterium Planktothrix rubescens using a multiparameter probe equipped with a phycoerythrin sensor (in vivo fluorometry, IVL) in Lake Mondsee, Austria, with flow cytometric live analyses of discrete samples taken from several depths in the upper 20 m of the water column. Results obtained by IVL and acoustic flow cytometry (AFC) were compared to microscopic analyses of integrated (0–21 m) water samples using fixed material. This comparison was made because the integrated samples are used for the Austrian monitoring programme according to the EU Water Framework Directive. We demonstrate that AFC provides quantitative analyses of the filaments of P. rubescens that are significantly correlated to IVF and microscopic analyses, allowing rapid (within hours) and more precise calculation of P. rubescens biomass than estimates derived from IVL. Our analysis shows that vertically integrated water samples provide unreliable information on the concentration of P. rubescens in the upper surface waters and on the peak concentration of P. rubescens within the water column. We conclude that the protocol that we developed is superior to the current monitoring practice.

Do current European lake monitoring programmes reliably estimate phytoplankton community changes?

Many European lakes are monitored according to the EU Water Framework Directive (WFD), with focus on phytoplankton biomass and species composition. However, the low-frequency WFD monitoring may miss short-term phytoplankton changes. This is an important issue because short-term extreme meteorological events (heat waves and heavy rain) are predicted to increase in frequency and intensity with climate change. We used records from Lake Mondsee (Austria) from 2009 to 2015 to test if a reduction from monthly to seasonal sampling affected the average annual phytoplankton biovolume. Furthermore, we combined inverted light microscopy, FlowCAM and flow cytometry to estimate the effect of sampling during extreme events on average phytoplankton biovolume. Relative to monthly sampling, seasonal sampling significantly overestimated phytoplankton biomass. A heat wave in 2015 and two episodes of heavy rain in 2015 and 2016 caused species-specific changes; biovolumes of chlorophytes and the filamentous cyanobacterium Planktothrix rubescens (De Candolle ex Gomont) Anagnostidis & Komárek increased significantly during the heat wave. Using live material with FlowCAM and flow cytometry, we detected small and fragile cells and colonies that were either ignored or underrepresented by analysing fixed samples with light microscopy. We suggest a modified sampling and analysis strategy to capture short-term changes within the phytoplankton community.

Modelling the plankton groups of the deep, peri-alpine Lake Bourget

Predicting phytoplankton succession and variability in natural systems remains to be a grand challenge in aquatic ecosystems research. In this study, we identified six major plankton groups in Lake Bourget (France), based on cell size, taxonomic properties, food-web interactions and occurrence patterns: cyanobacterium Planktothrix rubescens, small and large phytoplankton, mixotrophs, herbivorous and carnivorous zooplankton. We then developed a deterministic dynamic model that describes the dynamics of these groups in terms of carbon and phosphorus fluxes, as well as of particulate organic phosphorus and dissolved inorganic phosphorus. The modular and generic model scheme, implemented as a set of modules under Framework for Aquatic Biogeochemical Models (FABM) enables run-time coupling of the plankton module an arbitrary number of times, each time with a prescribed position across the autotrophy/heterotrophy continuum. Parameters of the plankton groups were mainly determined conjointly by the taxonomic and allometric relationships, based on the species composition and average cellular volume of each group. The biogeochemical model was coupled to the one-dimensional General Ocean Turbulence Model (GOTM) and forced with local meteorological conditions. The coupled model system shows very high skill in predicting the spatiotemporal distributions of water temperature and dissolved inorganic phosphorus for five simulated years within the period 2004 to 2010, and intermediate skill in predicting the plankton succession. We performed a scenario analysis to gain insight into the factors driving the sudden disappearance of P. rubescens in 2010. Our results provide evidence for the hypothesis that the abundance of this species before the onset of stratification is critical for its success later in the growing season, pointing thereby to a priority effect.

Grazing of Nuclearia thermophila and Nuclearia delicatula (Nucleariidae, Opisthokonta) on the toxic cyanobacterium Planktothrix rubescens

During the last decades, the planktonic cyanobacterium Planktothrix rubescens became a dominant primary producer in many deep pre-alpine lakes. While altered physiochemical conditions due to lake warming seem to favour this cyanobacterial species, its dominance is partly attributed to factors conferring grazing resistance. The rigid structure of the cyanobacterial filaments and toxic secondary metabolites (e.g. microcystins) protect against diverse grazers. Nonetheless, species of the protistan genus Nuclearia (Nucleariidae, Opisthokonta) are able to overcome this grazing protection. Time lapse video documentation served as tool to record slow feeding processes of N. thermophila and N. delicatula. Three different feeding strategies could be distinguished: (i) Phagocytosis of small fragments, (ii) serial break-ups of cyanobacterial cells and (iii) bending and breaking of filaments. While observations revealed mechanical manipulation to be important for the efficient breakdown of P. rubescens filaments, the toxin microcystin had no pronounced negative effects on nucleariid cells. Growth experiments with N. thermophila/N. delicatula and different accompanying bacterial assemblages pointed to a pivotal role of distinct prokaryotic species for toxin degradation and for the growth success of the protists. Thus, the synergistic effect of nucleariids and specific bacteria favours an efficient degradation of P. rubescens along with its toxin.

Changes in N:P Supply Ratios Affect the Ecological Stoichiometry of a Toxic Cyanobacterium and Its Fungal Parasite.

Human activities have dramatically altered nutrient fluxes from the landscape into receiving waters. As a result, not only the concentration of nutrients in surface waters has increased, but also their elemental ratios have changed. Such shifts in resource supply ratios will alter autotroph stoichiometry, which may in turn have consequences for higher trophic levels, including parasites. Here, we hypothesize that parasite elemental composition will follow changes in the stoichiometry of its host, and that its reproductive success will decrease with host nutrient limitation. We tested this hypothesis by following the response of a host–parasite system to changes in nitrogen (N) and phosphorus (P) supply in a controlled laboratory experiment. To this end, we exposed a fungal parasite (the chytrid Rhizophydium megarrhizum) to its host (the freshwater cyanobacterium Planktothrix rubescens) under control, low N:P and high N:P conditions. Host N:P followed treatment conditions, with a decreased N:P ratio under low N:P supply, and an increased N:P ratio under high N:P supply, as compared to the control. Shifts in host N:P stoichiometry were reflected in the parasite stoichiometry. Furthermore, at low N:P supply, host intracellular microcystin concentration was lowered as compared to high N:P supply. In contrast to our hypothesis, zoospore production decreased at low N:P and increased at high N:P ratio as compared to the control. These findings suggest that fungal parasites have a relatively high N, but low P requirement. Furthermore, zoospore elemental content, and thereby presumably their size, decreased at high N:P ratios. From these results we hypothesize that fungal parasites may exhibit a trade-off between zoospore size and production. Since zooplankton can graze on chytrid zoospores, changes in parasite production, stoichiometry and cell size may have implications for aquatic food web dynamics.

Restoring lakes through external phosphorus load reduction: the case of Lake Pusiano (Southern Alps)

We estimated the external phosphorus load (EPL) between 1960 and 2015 in Lake Pusiano, a mid-sized subalpine lake that reached its maximum trophic state in the mid-1980s. Using historical data we also estimated the internal phosphorus load (IPL) between 1972 and 2015. EPL reached its maximum in the mid-1980s with phosphorus (P) values ~21 t yr−1 compared to the current value close to 6 t yr−1. IPL was one order of magnitude less and ranged between 0.25 t yr−1 (2015) and 3.6 (1985) t yr−1. The strong reduction of the P load determined a marked decrease of both P (from 200 to 23 μg L−1) and chlorophyll a (from 18 to 8 μg L−1) in-lake concentrations. The process of eutrophication and subsequent recovery, however, showed hysteresis between P load and in-lake P concentrations. In recent years, in particular, the P concentrations at winter overturn seem to be independent from the P load, related to modifications in the hydrological management of the lake that favored P flush-out in October–November, when the EPL is maximal. This process led to a marked decrease of the total P concentrations at winter overturn (23 μg L−1), which recently resulted in lower-than-target concentrations (30 μg L−1) established by the Lombardy Region. The lake, nevertheless, suffers from the presence of the toxic cyanobacterium Planktothrix rubescens, and eradicating this species and further improving water quality will likely require an additional abatement of the EPL.

Compound-specific carbon and nitrogen isotopic compositions of chlorophyll a and its derivatives reveal the eutrophication history of Lake Zurich (Switzerland)

To reconstruct the impact of eutrophication on phototrophic communities and the biogeochemical cycling of carbon and nitrogen in the surface water, we investigated the distributions and carbon and nitrogen isotopic compositions (δ13C and δ15N values) of chlorins in the sediments of Lake Zurich. The chlorin distributions were dominated by chlorophyll a (Chl a) and its derivatives, which reflect rapid degradation to the pheopigments in the water column and sediments of the lake. The δ13C values of these sedimentary chlorins followed the historical trends of eutrophication and reoligotrophication, except in the surface sediments, which were characterised by higher relative contributions of aged, redeposited organic matter (OM). The δ13C values of the sedimentary chlorins together with bulk sediment δ13C values and C/N ratios indicate that the phototrophic communities in the lake used a 13C-depleted carbon source, which is mainly of aquatic origin. The δ15N values of chlorins reflect the predominance of nitrate assimilating phototrophs, especially the non-N2-fixing cyanobacterium Planktothrix rubescens prevalent during sediment deposition. Shifts in δ15N values of Chl a followed mostly the trends in eutrophication and reoligotrophication, but were also affected by community assemblage shifts to diatoms and/or other cyanobacteria at the end of the 19th century and during the eutrophication maximum in the 1970s. The lower δ15NChl-a values in the surface sediments coincide with increasing nitrogen to phosphorus ratios and reduced water column mixing that characterise the recent reoligotrophication period and may explain the predominance of P. rubescens in Lake Zurich. In contrast, the higher contributions of laterally transported OM explains the large offset of δ15N values of the pheopigments relative to Chl a, which is supported by the high radiocarbon age of the surface sediments.

Prolongation, deepening and warming of the metalimnion change habitat conditions of the harmful filamentous cyanobacterium Planktothrix rubescens in a prealpine lake

The most prominent responses of Lake Zurich to climate warming include the increase of surface water temperatures, a reduced depth of spring mixing, and the persistent thriving of the harmful cyanobacterium Planktothrix rubescens, a low-light adapted species concentrating in the metalimnion during summer. To study changes of its habitat, we assessed the spatio-temporal metalimnetic boundaries by applying low-pass filtering and binary thresholding to temperature profiles of long-term data (1978–2013, weekly measurements). Due to increasing temperatures over the last 3.5 decades, the onset and duration of metalimnion formation changed significantly (stratification increased by 33 days). Moreover, the upper metalimnetic boundary has undergone a significant drawdown of 2.3 m, accounting for an overall decrease in metalimnion thickness. Being the most abundant species in this zone, P. rubescens was not negatively affected by altered temporal or spatial stratification patterns during its phase of net growth (July–September), as the zone of its optimal light-dependent buoyancy was still located within the metalimnion. The biomass of P. rubescens in August was even significantly related to increasing temperatures. Nevertheless, a further depression of the metalimnetic top boundary may eventually restrict P. rubescens by forcing it into layers of unfavourable light conditions or into the turbulent epilimnetic zone.

Multiannual trend of microcystin production in the toxic cyanobacterium Planktothrix rubescens in Lake Garda (Italy)

ABSTRACTMassive developments (blooms) of cyanobacteria represent a major concern in many natural and artificial waterbodies, because of the high levels of toxins that can be released into water. Microcystins, potent hepatotoxins, represent the most frequent toxins produced by cyanobacteria. The determination of the microcystins’ profile in waterbodies is important for determining the real toxic potential. We have investigated the evolution of the microcystins’ profile of Lake Garda (Italy) from both a qualitative and a quantitative point of view over a period of five years. We have also analysed possible correlations between toxin profile and selected environmental parameters. We found that a demethylated microcystin variant (RRdm) was always dominant but showing a cyclic trend, alternating periods in which it represented the only variant, and periods in which other variants were present (LRdm, HtyrRdm, RR, and LR). These changes are probably due to changes in chemotypes composition inside the resident Planktothrix rubescens population and do not seem to be exclusively linked to the considered environmental variables. Total microcystin concentrations were always low (in the ng L−1 scale), and showed typical peaks in the metalimnetic layer during summer–autumn time of each year.

Cyanobacteria biennal dynamic in a volcanic mesotrophic lake in central Italy: Strategies to prevent dangerous human exposures to cyanotoxins

Vico Lake, a volcanic meso-eutrophic lake in Central Italy, whose water is used for drinking and recreational activities, experienced the presence of the microcystins (MC) producing cyanobacterium Planktothrix rubescens. In order to assess the human health risks and to provide the local health authorities with a scientific basis for planning tailored monitoring activities, we studied P. rubescens ecology and toxicity for two years. P. rubescens generally dominated the phytoplankton community, alternating with Limnothrix redekei, potentially toxic. P. rubescens was distributed throughout the water column during winter; in summer it produced intense blooms where drinking water is collected (−20 m); here MC were detected all year round (0.5–5 μg/L), with implications for drinking water quality. In surface waters, MC posed no risk for recreational activities in summer, while in winter surface blooms and foams (containing up to 56 μg MC/L) can represent a risk for people and children practicing water sports and for animals consuming raw water.Total phosphorus, phosphate and inorganic nitrogen were not relevant to predict densities nor toxicity; however, a strong correlation between P. rubescens density and aminopeptidase ectoenzymatic activity, an enzyme involved in protein degradation, suggested a role of organic nitrogen for this species. The fraction of potentially toxic population, determined both as mcyB+/16SrDNA (10–100%) and as the MC/mcyB+ cells (0.03–0.79 pg MC/cell), was much more variable than usually observed for P. rubescens. Differently from other Italian and European lakes, the correlation between cell density or the mcyB+ cells and MC explained only ∼50 and 30% of MC variability, respectively: for Vico Lake, monitoring only cell or the mcyB+ cell density is not sufficient to predict MC concentrations, and consequently to protect population health. Finally, during a winter bloom one site has been sampled weekly, showing that monthly sampling during such a phase could greatly underestimate the ‘hazard’. Our results highlight the need to adopt a stepwise monitoring activity, considering the lake and the cyanobacteria specific features. This activity should be complemented with communication to the public and involvement of stakeholders.

Emergence of nontoxic mutants as revealed by single filament analysis in bloom-forming cyanobacteria of the genus Planktothrix.

BackgroundBloom-forming cyanobacteria cause toxic algae outbreaks in lakes and reservoirs. We aimed to explore and quantify mutation events occurring within the large mcy gene cluster (55 kbp) encoding microcystin (MC) biosynthesis that inactivate MC net production. For this purpose we developed a workflow to detect mutations in situ occurring anywhere within the large mcy gene cluster as amplified from one single filament of the red-pigmented cyanobacterium Planktothrix rubescens. From five lakes of the Alps eight hundred Planktothrix filaments were isolated and each individual filament was analyzed for mutations affecting the mcy genes.ResultsMutations inactivating MC synthesis were either through an insertion element ISPlr1 or the partial deletion of mcy genes. Neutral mutations not affecting MC biosynthesis occurred within two intergenic spacer regions, either through the insertion of a Holliday-junction resolvase RusA or ISPlr1. Altogether, the insertions affected a few mcy genes only and their location was correlated with regions similar to repetitive extragenic palindromic DNA sequences (REPs). Taking all of the filaments together, the mutations leading to the inactivation of MC synthesis were more rare (0.5–6.9 %), when compared with the neutral mutations (7.5–20.6 %). On a spatial-temporal scale the ratio of MC synthesis-inactivating vs. neutral mutations was variable, e.g., the filament abundance carrying partial deletion of mcyD (5.2–19.4 %) and/or mcyHA (0–7.3 %) exceeded the abundance of neutral mutations.ConclusionsIt is concluded that insertion events occurring within the Planktothrix mcy gene cluster are predictable due to their correlation with REPs. The frequency of occurrence of the REPs within the mcy gene cluster of Planktothrix relates to the rather common mutation of mcy genes in Planktothrix. Spatial-temporal variable conditions may favor the emergence of partial mcy deletion mutants in Planktothrix, in particular a higher proportion of genotypes resulting in inactivation of MC synthesis might be caused by increased ISPlr1 activity.Electronic supplementary materialThe online version of this article (doi:10.1186/s12866-016-0639-1) contains supplementary material, which is available to authorized users.
