Phylogenetic analyses of Reticulitermes (Blattodea: Rhinotermitidae) from California and other western states: multiple genes confirm undescribed species identified by cuticular hydrocarbons.

Subterranean termites in the genus Reticulitermes Holmgren 1913 are among the most economically important wood-destroying pests in the western United States. Yet, there remains uncertainty regarding the taxonomy and biology of the species in this genus. The 2 species described as having distributions in this region are the western subterranean termite, Reticulitermes hesperus Banks, and the arid land subterranean termite, Reticulitermes tibialis Banks. Taxonomic studies utilizing cuticular hydrocarbon (CHC) profiles, agonistic behavior, flight phenology, and mitochondrial DNA (mtDNA) suggested that R. hesperus is a species complex comprised of 2 or more sympatric, yet reproductively isolated species. To further delineate these taxa, we examined multiple genes from samples of Reticulitermes collected in the western United States. Alates collected after recent spring and fall mating flights, as well as previously collected workers, were subjected to CHC phenotyping and DNA sequence analyses that targeted mitochondrial cytochrome oxidase subunit II (COII), mitochondrial 16S rRNA, and nuclear Internal Transcribed Spacer 1 and 2 (ITS1 and 2). Phylogenetic analyses conducted also included published sequences of other putative western Reticulitermes species. Results suggest that at least 5 species of Reticulitermes may be present in California and that Reticulitermes in Arizona consistently group into multiple clades, including samples previously identified as R. tibialis in a sister clade. These analyses further support the species status of qualitatively different CHC phenotypes and that alates swarming in spring vs. fall are reproductively isolated species.

"Scent of a fruit fly": Cuticular chemoprofiles after mating in differently fed Drosophila melanogaster (Diptera: Drosophilidae) strains.

In the world of complex smells in natural environment, feeding and mating represent two important olfactory-guided behaviors in Drosophila melanogaster (Diptera: Drosophilidae). Diet affects the chemoprofile composition of the individuals, which, indirectly, may significantly affect their mating success. In this study, chemoprofiles of recently mated flies belonging to four D. melanogaster strains, which were fed for many generations on different substrates (standard cornmeal-S strain; banana-B strain; carrot-C strain; tomato-T strain) were identified and quantified. In total, 67 chemical compounds were identified: 48 compounds were extracted from males maintained on banana and carrot, and 47 compounds from males maintained on cornmeal and tomato substrates, while total of 60 compounds were identified in females from all strains. The strains and the sexes significantly differed in qualitative nature of their chemoprofiles after mating. Significant differences in the relative amount of three major male pheromones (cis-vaccenyl acetate-cVA, (Z)-7-pentacosene, and (Z)-7-tricosene) and in female pheromone (Z,Z)-7,11-nonacosadiene among strains were also recorded. Furthermore, multivariate analysis of variance (MANOVA) pointed to significant differences between virgin and mated individuals of all strains and within both sexes. Differences in some of the well known sex pheromones were also identified when comparing their relative amount before and after mating. The presence of typical male pheromones in females, and vice versa may indicate their bidirectional transfer during copulation. Our results confirm significant effect of mating status on cuticular hydrocarbon (CHC) phenotypes in differently fed D. melanogaster flies.

A shift to shorter cuticular hydrocarbons accompanies sexual isolation among Drosophila americana group populations.

Because sensory signals often evolve rapidly, they could be instrumental in the emergence of reproductive isolation between species. However, pinpointing their specific contribution to isolating barriers, and the mechanisms underlying their divergence, remains challenging. Here, we demonstrate sexual isolation due to divergence in chemical signals between two populations of Drosophila americana (SC and NE) and one population of D. novamexicana, and dissect its underlying phenotypic and genetic mechanisms. Mating trials revealed strong sexual isolation between Drosophila novamexicana males and SC Drosophila americana females, as well as more moderate bi‐directional isolation between D. americana populations. Mating behavior data indicate SC D. americana males have the highest courtship efficiency and, unlike males of the other populations, are accepted by females of all species. Quantification of cuticular hydrocarbon (CHC) profiles—chemosensory signals that are used for species recognition and mate finding in Drosophila—shows that the SC D. americana population differs from the other populations primarily on the basis of compound carbon chain‐length. Moreover, manipulation of male CHC composition via heterospecific perfuming—specifically perfuming D. novamexicana males with SC D. americana males—abolishes their sexual isolation from these D. americana females. Of a set of candidates, a single gene—elongase CG17821—had patterns of gene expression consistent with a role in CHC differences between species. Sequence comparisons indicate D. novamexicana and our Nebraska (NE) D. americana population share a derived CG17821 truncation mutation that could also contribute to their shared “short” CHC phenotype. Together, these data suggest an evolutionary model for the origin and spread of this allele and its consequences for CHC divergence and sexual isolation in this group.

Chemical and molecular identification of the invasive termite Zootermopsis nevadensis (Isoptera: Archotermopsidae) in Japan

Numerous termite species have been introduced outside their native ranges by human transport, and some have become invasive. The dampwood termite Zootermopsis nevadensis (Hagen), which is native to western North America, has been introduced to and become established in Kawanishi City, Hyogo Prefecture, Japan. Zootermopsis nevadensis is subdivided into two subspecies based on cuticular hydrocarbon (CHC) phenotypes: Z. nevadensis nevadensis and Z. nevadensis nuttingi (Haverty and Thorne). Here, we identified Z. nevadensis in Japan as hybrids between the two subspecies. Chemical analysis showed the presence of 7,15-dimethylhenicosane and 5,17-dimethylhenicosane in the CHCs of Z. nevadensis in Japan, corresponding to the CHC phenotype of Z. n. nevadensis. Conversely, all mitochondrial cytochrome c oxidase subunit I sequences of Z. nevadensis in Japan were identical to sequences from Z. n. nuttingi and hybrids between the two subspecies from a native hybrid zone in California, USA. In addition, phylogenetic analysis showed that Z. nevadensis in Japan formed a clade with Z. n. nuttingi and hybrids between the two subspecies. Our results show discordance between the chemical and genetic features of Z. nevadensis in Japan, indicating that individuals of Z. nevadensis in Japan are hybrids between the two subspecies.

The effect of dietary fatty acids on the cuticular hydrocarbon phenotype of an herbivorous insect and consequences for mate recognition.

The cuticular hydrocarbon (CHC) profile of the mustard leaf beetle Phaedon cochleariae is known to mediate mate recognition and is dependent on food plant species; beetles previously were shown to prefer mates that fed on the same plant species and which have a similar CHC pattern. In order to elucidate whether the pattern of ingested fatty acids affects the CHC pattern of P. cochleariae adults, we fed beetles: (a) with two different host plant species differing in fatty acid profile; and (b) artificial diets differing mainly in their composition of mono-, di-, and triunsaturated fatty acids. Analyses of the beetles' CHCs revealed that ingestion of different fatty acid blends results in quantitative effects on the beetle's straight-chain and methyl-branched CHCs. Interestingly, CHC patterns of males and females were affected differently by ingestion of fatty acids. In contrast to the effect on mating caused by feeding on different host plant species, beetles that were fed with different artificial diets, leading to different beetle CHC profiles, did not exhibit mating preference. We suggest that the occurrence of CHC-dependent assortative mating in P. cochleariae does not depend on the dietary fatty acids offered to the beetles in this study, but on other food constituents that affect CHC biosynthesis.

Looking for a similar partner: host plants shape mating preferences of herbivorous insects by altering their contact pheromones

The role of phenotypical plasticity in ecological speciation and the evolution of sexual isolation remains largely unknown. We investigated whether or not divergent host plant use in an herbivorous insect causes assortative mating by phenotypically altering traits involved in mate recognition. We found that males of the mustard leaf beetle Phaedon cochleariae preferred to mate with females that were reared on the same plant species to females provided with a different plant species, based on divergent cuticular hydrocarbon profiles that serve as contact pheromones. The cuticular hydrocarbon phenotypes of the beetles were host plant specific and changed within 2 weeks after a shift to a novel host plant species. We suggest that plant-induced phenotypic divergence in mate recognition cues may act as an early barrier to gene flow between herbivorous insect populations on alternative host species, preceding genetic divergence and thus, promoting ecological speciation.

Cuticular Hydrocarbon Phenotypes Do not Indicate Cryptic Species in Fungus-Growing Termites (Isoptera: Macrotermitinae)

In several termite species, distinct differences in the composition of cuticular hydrocarbons among colonies correspond to high genetic divergence of mitochondrial DNA sequences. These observations suggest that hydrocarbon phenotypes represent cryptic species. Different cuticular hydrocarbon phenotypes also are found among colonies of fungus-growing termites of the genus Macrotermes. To determine if these hydrocarbon differences in Macrotermes also indicate cryptic species, we sequenced the mitochondrial CO I gene from species in West and East Africa. Among individuals of a supposed species but belonging to different cuticular hydrocarbon phenotypes, the genetic distances are much smaller than distances between species. Unlike what has been observed in other termites, Macrotermes hydrocarbon phenotypes do not represent cryptic species. Our findings suggest fundamental differences in the evolution and/or function of cuticular hydrocarbons among different termite lineages.

Aphid species identification using cuticular hydrocarbons and cytochrome b gene sequences

Abstract: In Tunisia, four major aphid species have been identified based on adult female's morphological characters: Aphis gossypii Glover, Aphis craccivora Koch, Myzus persicae Sluzer and Macrosiphum euphorbiae Thomas. Species identification at individual collection sites is often difficult because adults are much fewer in number than larvae which are not so easy to distinguish morphologically. We therefore set up an experiment to determine if cuticular hydrocarbon phenotypes and mitochondrial DNA haplotypes could be used to distinguish such sympatric species. Results showed that each species had an unique cuticular hydrocarbon phenotype and mitochondrial cytochrome b sequence. Cytochrome b restriction fragment‐length polymorphism markers, especially DdeI, identified in this sudy constitute a relatively simple and useful approach to distinguish the four species even at the nymphal stage.

Flight Phenology of Sympatric Populations of <I>Reticulitermes</I> (Isoptera: Rhinotermitidae) in Northern California: Disparate Flight Intervals Indicate Reproductive Isolation Among Cuticular Hydrocarbon Phenotypes

The diurnal flights of Reticulitermes spp. have been reported to occur in the spring and early fall in northern California. From the spring of 1994 until the fall of 2001, we collected hundreds of alates of Reticulitermes spp. cuticular hydrocarbon phenotypes CA-A/A′ and CA-D during flights from multiple locations in the San Francisco Bay Area. As predicted, there were two flight seasons: one in the spring from February to May and one in the fall from October to December. The flights in the spring were made only by phenotypes CA-A/A′; those in the fall exclusively included phenotype CA-D. These disparate flight times indicate that reproductive isolation is complete and provide further biological evidence that phenotypes CA-A and CA-A′ are variants of one taxon and are distinct from CA-D.

The nucleotide changes governing cuticular hydrocarbon variation and their evolution in Drosophila melanogaster.

The cuticular hydrocarbon (CH) pheromones in Drosophila melanogaster exhibit strong geographic variation. African and Caribbean populations have a high ratio of 5,9 heptacosadiene/7,11 heptacosadiene (the "High" CH type), whereas populations from all other areas have a low ratio ("Low" CH type). Based on previous genetic mapping, DNA markers were developed that localized the genetic basis of this CH polymorphism to within a 13-kb region. We then carried out a hierarchical search for diagnostic nucleotide sites starting with four lines, and increasing to 24 and 43 lines from a worldwide collection. Within the 13-kb region, only one variable site shows a complete concordance with the CH phenotype. This is a 16-bp deletion in the 5' region of a desaturase gene (desat2) that was recently suggested to be responsible for the CH polymorphism on the basis of its expression [Dallerac, R., Labeur, C., Jallon, J.-M., Knipple, D. C., Roelofs, W. L. & Wicker-Thomas, C. (2000) Proc. Natl. Acad. Sci. 97, 9449--9454]. The cosmopolitan Low type is derived from the ancestral High type, and DNA sequence variations suggest that the former spread worldwide with the aid of positive selection. Whether this CH variation could be a component of the sexual isolation between Zimbabwe and other cosmopolitan populations remains an interesting and unresolved question.

Correspondence of soldier defense secretion mixtures with cuticular hydrocarbon phenotypes for chemotaxonomy of the termite genus Reticulitermes in North America.

Soldier defense secretions from samples of Reticulitermes collected in California, Nevada, Arizona, New Mexico, and Georgia were characterized and correlated with cuticular hydrocarbon phenotypes. Twenty-seven cuticular hydrocarbon phenotypes have been defined, and soldier defense secretion (SDS) phenotypes have been described for 25 of these. Forty-five terpenoid compounds were found, including monoterpenes, sesquiterpenes, and a few diterpenes. The monoterpenes include (-)-alpha-pinene, (-)-beta-pinene, (-)-camphene, myrcene, (Z)- and (E)-ocimene, and (-)-limonene. The major sesquiterpenes produced are (+)-gamma-cadinene, (+)-gamma-cadinene aldehyde, (-)-germacrene A, germacrene B, gamma-himachalene, and beta-bisabolene. Some SDS phenotypes pair with more than one cuticular hydrocarbon phenotype; however, with two exceptions, each hydrocarbon phenotype is associated with only one SDS phenotype. These chemical characterizations lend support to the conclusion that there are numerous undescribed species of Reticulitermes in North America.

Size and Dispersion of Colonies of <I>Reticulitermes</I> spp. (Isoptera: Rhinotermitidae) in a Wildland and a Residential Location in Northern California

Mark-release-recapture studies were conducted to estimate foraging populations, maximum foraging distances between foraging sites, and minimum total foraging distance for three colonies of two cuticular hydrocarbon phenotypes of Reticulitermes at a wildland site near Placerville, CA, in the Sierra Nevada foothills and for six colonies of three phenotypes at two residential sites in Marin County. At Placerville, the hydrocarbon phenotype B colony had the fewest termites, with an estimated foraging population of 4,476-13,602, and occupied only one monitoring station. The two phenotype A colonies had foraging populations estimated to range from 40,809 to 128,597; one inhabited one monitoring station, whereas the other occupied three stations with a maximum distance between monitoring stations of 6.3 m. At the Marin County sites, two phenotype D colonies were estimated to have foraging populations ranging from 9,191 to 194,692; each foraged at a single monitoring station. Estimated foraging populations for the three phenotype A' colonies ranged from 71,483 to 491,901 with the maximum distance between monitoring stations ranging from 11.7 to 25.3 m. The phenotype A colony was estimated to have 8,747-25,190 foragers, with a maximum distance between monitoring stations of 1.8 m.

Correlation of mitochondrial haplotypes with cuticular hydrocarbon phenotypes of sympatric Reticulitermes species from the southeastern United States.

Three sympatric Reticulitermes species have been identified in Georgia, USA, based on morphological characters from alates and soldiers: R. flavipes, R. virginicus, and R. hageni, but species identification at individual collection sites is often difficult because alate production is seasonal and soldiers comprise 1–3% of the colony. We therefore set up an experiment to determine if chemical phenotypes and mtDNA haplotypes can be used together to separate species of subterranean termites. Subterranean termites of the order Reticulitermes (Isoptera: Rhinotermitidae) were collected from 20 inspection ports across four soil provinces in Georgia. Each collection was identified to species using dichotomous keys. Two collections, HH11 and BH25, however, could not be unequivocally keyed to species and were classified as unknown. The mitochondrial cytochrome oxidase II (COII) gene was sequenced from individual members of each collection and the variation in cuticular hydrocarbon phenotypes from these same collections was characterized. The cuticular hydrocarbon and mtDNA phylogenetic analyses show agreement with both unknown collections falling out in a separate clade. Specimens from HH11 nad BH25 are different morphologically, chemically, and genetically from the three known sympatric species in Georgia. Our results suggest that these two collections may represent at least one new taxon in Reticulitermes. Furthermore, the association of cuticular hydrocarbon phenotypes and mtDNA haplotypes demonstrates that, when combined with morphological characters, they are useful in separating known species, determining new species, and understanding termite evolution.

Agonistic Behavior and Cuticular Hydrocarbon Phenotypes of Colonies of Reticulitermes (Isoptera: Rhinotermitidae) from Northern California

Reticulitermes in northern California is supposed to be represented by R. hesperus Banks and R tibialis Banks, yet at least 5 distinct cuticular hydrocarbon phenotypes have been characterized. Three hydrocarbon phenotypes from the Institute of Forest Genetics near Placerville. CA, and 3 cuticular hydrocarbon phenotypes from 2 separate sites in Marin County were used to characterize interactions of foraging groups or colonies at each site. Pairings from the same foraging group or different foraging groups of the same colony rarely resulted in immediate aggression and never resulted in high mortality. Pairings of workers from different foraging groups of the same cuticular hydrocarbon phenotype from either the Placerville or Marin sites resulted in few bouts (8 and 15%, respectively ) with immediate aggression, but after 24 h, mortality was high in 56 and 81% of the bouts, respectively. Pairings from different cuticular hydrocarbon phenotypes resulted in immediate aggression 48.8 and 61.5% of the time, respectively, nearly all of these (>99%) resulted in high mortality after 24 h. These results suggest that these Reticulitermes recognize hydrocarbon phenotypes, and can differentiate colony mates and alien workers within a cuticular hydrocarbon phenotype. Because kin discriminal ion suggests genetic relatedness among individuals, this bioassay will be useful for determining the association of foraging groups in ecological studies of Reticulitermes colonies in northern California and indirectly may indicate relatedness among colonies of the same hydrocarbon phenotype.

Cuticular Hydrocarbons of Reticulitermes (Isoptera: Rhinotermitidae) from Northern California Indicate Undescribed Species

Cuticular hydrocarbon mixtures of Reticulitermes samples from disparate locations in northern California were characterized and correlated with species determinations. Literature records indicate that there are two extant species of Reticulitermes in northern California: Reticulitermes hesperus Banks and R. tibialis Banks. We identified five distinct hydrocarbon phenotypes in Reticulitermes from northern California. Termites were diagnosed morphologically as R. tibialis for all five cuticular hydrocarbon phenotypes based on available keys to soldiers. Commonly accepted distributions of Reticulitermes in northern California indicate that our primary collection sites should only produce R. hesperus. On the basis of cuticular hydrocarbons alone we suggest that there are two or more undescribed taxa of Reticulitermes in northern California, or that described species of Reticulitermes occur in northern California, but have never been noticed before. Therefore, we conclude that the taxonomy of Reticulitermes in North America is in need of revision.

Allozymic Relationships among Cuticular Hydrocarbon Phenotypes of Zootermopsis Species (Isoptera: Termopsidae)

Genetic relationships among and within three species of Zootermopsis were examined using protein electrophoresis. Results were compared with four cuticular hydrocarbon phenotypes: Z. angusticollis (Hagen), Z. laticeps (Banks), Z. nevadensis nevadensis (Hagen), and Z. nevadensis nuttingi Haverty and Thorne. Allozyme data supported the position of Z. angusticollis, Z. laticeps, and Z. nevadensis as separate species and were consistent with cuticular hydrocarbon groupings. However, the two subspecies or cuticular hydrocarbon groupings within Z. nevadensis were not genetically distinct. If these subspecies are not interbreeding, divergence has been recent because genetic differentiation has not occurred. Further studies are required to determine the basis for the inconsistency between cuticular hydrocarbon patterns and allozymes within the Z. nevadensis complex.

Cuticular hydrocarbons and defensive compounds ofReticulitermes flavipes (Kollar) andR. santonensis (feytaud): Polymorphism and chemotaxonomy

Colonies ofReticulitermes flavipes andR. santonensis were collected from the southeastern United States (Georgia) and the southwest of France (Charente-maritime). Defensive compounds and cuticular hydrocarbons were identified by gas chromatography-mass spectrometry and quantified by gas chromatography using an internal standard for each caste and all colonies. These analyses show that although the cuticular hydrocarbons ofR. santonensis in Europe andR. flavipes in Georgia are identical, their relative proportions are different. However, the defensive compounds synthesized by their soldiers are different. A strong chemical polymorphism between sympatric colonies ofR. flavipes in the SW United States was detected in terms of both the hydrocarbons of the workers and soldiers and in the defensive secretions of the soldiers. The six defensive secretion phenotypes are based on the presence or absence of terpenes whereas the cuticular hydrocarbon phenotypes are based on significant differences in the proportions of the various components. A multivariate analysis (analysis of principal components) clearly permitted discrimination of four phenotypes (three inR. flavipes and one inR. santonensis) without intermediates. The hydrocarbons responsible for these variations were identified, and it was shown that the variations are neither seasonal nor geographic. The phenotypes of the cuticular hydrocarbons (workers and soldiers) and defensive compounds are linked in each colony, forming in three groups inR. flavipes Georgia, one subdivided into four subgroups according to the defensive secretion phenotypes. The role of these polymorphisms is discussed and ethological tests indicate that the chemical polymorphism do not determine aggressive behavior. The taxonomic significance of these results is considered and two hypothesis are formulated: (1) We only detected a strong genetic polymorphism in one unique species, and we believe thatR. santonensis was introduced into Europe in the last century from oneR. flavipes colony. (2) Chemical variability characterizes the sibling species that can be grouped into the same subspeciesR. flavipes. Unknown mechanisms of reproductive isolation separate them.

Cuticular hydrocarbons of dampwood termites,Zootermopsis: Intra- and intercolony variation and potential as taxonomic characters

Colonies ofZootermopsis were collected from the central Sierra Nevada and the Monterey Penninsula in California, and from southern Arizona. Cuticular hydrocarbons were identified by gas chromatography-mass spectrometry (GC-MS) and quantified by gas-liquid chromatography (GLC) for each caste of all colonies. Four consistent and distinct cuticular hydrocarbon patterns, or chemical phenotypes, were identified. Unique and abundant monomethyl- and dimethylalkanes, and ann-alkene provided easy separation of the various phenotypes. Significant differences in the proportions of the various components were found among castes within a colony and colonies within phenotypes from California. Differences in the hydrocarbon proportions for castes were not consistent between colonies. The current taxonomy of the genusZootermopsis recognizes three species. Our identification of four consistent, unique cuticular hydrocarbon phenotypes from the three described species should alert systematists and others to a major concern. If there are truly only three extant species, then the hypothesis that cuticular hydrocarbon profiles in this genus are species specific is not acceptable. Conversely, if cuticular hydrocarbon profiles are truly species specific, then there is at least one new, undescribed species ofZootermopsis.
