An Effective Method to Prepare Curcumin-Loaded Soy Protein Isolate Nanoparticles Co-Stabilized by Carrageenan and Fucoidan.

In this study, a novel and simple strategy is proposed based on 3D network formed by easily blending polysaccharide carrageenan (Car) and fucoidan (Fuc) without a crosslinker. The Fuc/Car dual coating effectively assists the self-assembly of soy protein-isolated (SPI)/curcumin (Cur, C) composite microcapsules (SPI/C) and achieves an excellent curcumin encapsulation efficiency (EE) up to 95.28% with a 4.16% loading capacity (LC) under optimal conditions. The resulting nanocomposites achieved a satisfying redispersibility in aqueous solution and enhanced the water solubility with a lower size dispersity index (PDI) of 0.12 and a larger zeta potential of -29.67 mV. The Fuc/Car double-layer network not only dramatically improved its thermal stability and photostability, but also provided controlled release and enhanced antioxidant activity in in vitro conditions. The underlying mechanism of the self-assembly of the curcumin-loaded nanoparticles was also addressed. The results proved the feasibility of the encapsulation of unstable hydrophobic bioactive substances (curcumin) with the dual anionic polysaccharide Fuc/Car co-stabilized SPI nanoparticles. This study paves the way for an alternative way of developing novel curcumin delivery systems and will have broad prospects in the pharmaceutical industries.

The application of phenylboronic acid pinacol ester functionalized ROS-responsive multifunctional nanoparticles in the treatment of Periodontitis

Periodontitis is an inflammatory disease induced by the complex interactions between the host immune system and the microbiota of dental plaque. Oxidative stress and the inflammatory microenvironment resulting from periodontitis are among the primary factors contributing to the progression of the disease. Additionally, the presence of dental plaque microbiota plays a significant role in affecting the condition. Consequently, treatment strategies for periodontitis should be multi-faceted. In this study, a reactive oxygen species (ROS)-responsive drug delivery system was developed by structurally modifying hyaluronic acid (HA) with phenylboronic acid pinacol ester (PBAP). Curcumin (CUR) was encapsulated in this drug delivery system to form curcumin-loaded nanoparticles (HA@CUR NPs). The release results indicate that CUR can be rapidly released in a ROS environment to reach the concentration required for treatment. In terms of uptake, HA can effectively enhance cellular uptake of NPs because it specifically recognizes CD44 expressed by normal cells. Moreover, HA@CUR NPs not only retained the antimicrobial efficacy of CUR, but also exhibited more pronounced anti-inflammatory and anti-oxidative stress functions both in vivo and in vitro. This provides a good potential drug delivery system for the treatment of periodontitis, and could offer valuable insights for dental therapeutics targeting periodontal diseases.

Curcumin-loaded mesoporous polydopamine nanoparticles modified by quaternized chitosan against bacterial infection through synergistic effect

Clinically, open wounds caused by accidental trauma and surgical lesion resection are easily infected by external bacteria, hindering wound healing. Antibacterial photodynamic therapy has become a promising treatment strategy for wound infection. In this study, a novel antibacterial nanocomposite material (QMC NPs) was synthesized by curcumin, quaternized chitosan and mesoporous polydopamine nanoparticles. The results showed that 150 μg/mL QMC NPs had good biocompatibility and exerted excellent antibacterial activity against Staphylococcus aureus and Escherichia coli after blue laser irradiation (450 nm, 1 W/cm2). In vivo, QMC NPs effectively treated bacterial infection and accelerated the healing of infected wounds in mice.

Eficacia de nuevos viscosuplementos activos en el comportamiento de un modelo experimental de osteoartritis

ObjectiveTo evaluate with an animal model of osteoarthritis (New Zealand rabbits) the effectiveness of treatment with active viscosupplements (hyaluronic acid loaded with nanoparticles (NPs) that encapsulate anti-inflammatory compounds or drugs. Material and methodsExperimental study composed of 5 groups of rabbits in which section of the anterior cruciate ligament and resection of the internal meniscus were performed to trigger degenerative changes and use it as a model of osteoarthritis. The groups were divided into osteoarthrosis without treatment (I), treatment with commercial hyaluronic acid (HA) (II), treatment with HA with empty nanoparticles (III), treatment with HA with nanoparticles encapsulating dexamethasone (IV) and treatment with HA with nanoparticles that encapsulate curcumin (V). In groups II to V, the infiltration of the corresponding compound was carried out spaced one week apart.Macroscopic histological analysis was performed using a scale based on the Outerbridge classification for osteoarthritis. ResultsWe observed that this osteoarthritis model is reproducible and degenerative changes similar to those found in humans are observed.The groups that were infiltrated with hyaluronic acid with curcumin-loaded nanoparticles (V), followed by the dexamethasone group (IV) presented macroscopically less fibrillation, exposure of subchondral bone and sclerosis (better score on the scale) than the control groups (I) (osteoarthritis without treatment), group (II) treated with commercial hyaluronic acid and hyaluronic acid with nanoparticles without drug (III). ConclusionsThe use of active viscosupplements could have an additional effect to conventional hyaluronic acid treatment due to its antioxidant and anti-inflammatory effect. The most promising group was hyaluronic acid with nanoparticles that encapsulate curcumin and the second group was the one that encapsulates dexamethasone.

Enhanced Drug Carriage Efficiency of Curcumin-Loaded PLGA Nanoparticles in Combating Diabetic Nephropathy via Mitigation of Renal Apoptosis.

The Curcuma-derived diferuloylmethane compound CUR, loaded on Poly (lactide-co-glycolic) acid (PLGA) nanoparticles was utilized to combat DN-induced renal apoptosis by selectively targeting and modulating Bcl2. Upon in silico molecular docking and screening study CUR was selected as the core phytocompound for nanoparticle formulation. PLGA-nano-encapsulated-curcumin (NCUR) were synthesized following standard solvent displacement method. The NCUR were characterized for shape, size and other physico-chemical properties by Atomic Force Microscopy (AFM), Dynamic Light Scattering (DLS) and Fourier-Transform Infrared (FTIR) Spectroscopy studies. For in vivo validation of nephro-protective effects, Mus musculus were pre-treated with CUR at a dose of 50 mg/kg b.w. and NCUR at a dose of 25 mg/kg b.w. (dose 1), 12.5 mg/kg b.w (dose 2) followed by alloxan administration (100 mg/kg b.w) and serum glucose levels, histopathology and immunofluorescence study were conducted. The in silico study revealed a strong affinity of CUR towards Bcl2 (dock score -10.94 Kcal/mol). The synthesized NCUR were of even shape, devoid of cracks and holes with mean size of ~80 nm having -7.53 mV zeta potential. Dose 1 efficiently improved serum glucose levels, tissue-specific expression of Bcl2 and reduced glomerular space and glomerular sclerosis in comparison to hyperglycaemic group. This study essentially validates the potential of NCUR to inhibit DN by reducing blood glucose level and mitigating glomerular apoptosis by selectively promoting Bcl2 protein expression in kidney tissue.

DESIGN OF JACKFRUIT GUM-BASED GENIPIN CROSSLINKED NANOPARTICLES FOR SUSTAINED RELEASE OF CURCUMIN: OPTIMIZATION AND IN VITRO CHARACTERIZATION

The present work aims to design jackfruit gum-based curcumin-loaded nanoparticles (CUR-NPs) for improved drug entrapment and modified release of CUR using ionotropic gelation. Briefly, the optimization of CUR-NPs was confirmed using a 32 response surface methodology. The diffractogram and thermogram of CUR-NPs confirmed reduction of crystallinity of CUR (optimized batch: F5) due to jackfruit gum and genipin cross-linked polymeric network. The particle size and zeta potential analysis confirmed formation of nanosized and stable CUR-NPs, respectively. Also, the nanoparticles demonstrated 83.99 ± 1.23% entrapment efficiency, whereas they showed 98.36 ± 0.96% of CUR release within 12 h at pH 7.4. The CUR-NPs exhibited good mucoadhesive properties due to the presence of jackfruit gum. Finally, the MTT assay showed a decrease in colorectal cancer cell viability due to tailored CUR release from CUR-NPs. In conclusion, jackfruit gum-genipin-based CUR-NPs offered high entrapment efficiency, tailored releases of CUR, good mucoadhesive property and improved anticancer activity.

Preparation, characterization, and ex-vivo evaluation of curcumin-loaded niosomal nanoparticles on the equine sperm quality during cooled storage

Oxidative damage to sperm during cooled storage is a significant issue, and curcumin, with its antioxidant properties, could be a solution. However, its low bioavailability presents a challenge that this study aims to address. The primary objective of this study was to investigate the potential of curcumin-loaded niosomal nanoparticles (Cur-LNN) to enhance the antioxidant properties of curcumin and its effect on sperm quality during 72 h cooled storage. The thin-film hydration procedure was applied to prepare Cur-LNN. The fabricated noisomal nanocarriers were characterized using dynamic light scattering (DLS), transmission electron microscopy (TEM), field emission scanning electron microscopy (FESEM), and Fourier-transform infrared (FT-IR) spectroscopy. Moreover, the encapsulation and loading efficiency, in vitro release study, and ex-vivo antioxidant functionality of Cur-LNN on the stallion sperm preserved under cooled storage conditions were assessed. The fabricated Cur-LNN was spherical in shape and had an average particle size of 163.1 ± 1.8 nm, a zeta potential of −34.1 ± 1.9 mV, a poly-dispersity index of 0.339 ± 0.045, an encapsulation efficiency of 92.34 ± 0.18 %, and a loading efficiency of 35.57 ± 1.36 %. Ex-vivo evaluation revealed that supplementation of the semen extender with Cur-LNN has the potential to enhance sperm quality by improving total and progressive motility, plasma membrane functionality, and lipid peroxidation. These results demonstrate that Cur-LNN exhibited stronger antioxidant and protective effects than curcumin. Although further in vivo investigations are warranted, our ex-vivo results suggest that Cur-LNN has the potential to attenuate oxidative damage and can be used to fortify the antioxidant capacity of equine semen under cooled storage conditions.

Fenpropathrin provoked kidney damage via controlling the NLRP3/Caspase-1/GSDMD–mediated pyroptosis: The palliative role of curcumin-loaded chitosan nanoparticles

This study assessed the ability of formulated curcumin-loaded chitosan nanoparticles (CU-CS-NPs) to reduce the kidney damage resulting from fenpropathrin (FPN) in rats compared to curcumin (CU) in rats. Sixty male Sprague Dawley rats were separated into six groups and orally administered 1 mL/kg b.wt corn oil, 50 mg CU/kg b.wt, 50 mg CU-CS-NPs /kg b.wt., 15 mg FPN /kg b.wt, CU+ FPN or CU-CS-NPs + FPN for 60 days. Then, serum renal damage products were assessed. Total antioxidant capacity, reactive oxygen species, interleukin 1β (IL-1β), malondialdehyde, NF-κB P65, cleaved-Caspase-1, and Caspase-8 were estimated in kidney homogenates. The cleaved Caspase-3 and TNF-α immunoexpression and pyroptosis-related genes were determined in renal tissues. The results showed that CU-CS-NPS significantly repressed the FPN-induced increment in kidney damage products (urea, uric acid, and creatinine). Moreover, the FPN-associated hypo-proteinemia, renal oxidative stress and apoptotic reactions, and impaired renal histology were considerably repaired by CU and CU-CS-NPs. Additionally, compared to FPN-exposed rats, CU, and CU-CS-NPs-treated rats had considerably lower immunoexpression of cleaved Caspase-3 and TNF-α in renal tissue. The pyroptosis-related genes NLRP3, GSDMD, IL-18, Caspase-3, Caspase-1, IL-1β, Caspase-8, TNF-α, and NF-κB dramatically upregulated by FPN exposure in the renal tissues. Yet, in CU and CU-CS-NPs-treated rats, the gene above expression deviations were corrected. Notably, CU-CS-NPs were superior to CU in preventing oxidative damage and inflammation and regulating pyroptosis in the renal tissues of the FPN-exposed group. The results of the present study conclusively showed the superior favorable effect of CU-CS-NPs in counteracting renal impairment linked to environmental pollutants.

A new green self-assembly strategy for preparing curcumin-loaded starch nanoparticles based on natural deep eutectic solvent: Development, characterization and stability

At present, the primary solvents in nanocarrier systems for curcumin encounter challenges related to rapid degradation, and biosafety of organic reagents, surfactants and lipids, which are not suitable for food applications. Therefore, this research proposed an eco-friendly method for the first time, using choline chloride-lactic acid deep eutectic solvent as a solvent for curcumin and a self-assembly medium for short-chain amylose to prepare curcumin-loaded starch nanoparticles. This study investigated the interactions between curcumin and short-chain amylose as well as improved stability. Curcumin was successfully loaded into starch nanoparticles as demonstrated by scanning electron microscopy and confocal laser scanning microscopy analyses. The loading capacity of curcumin-loaded starch nanoparticles on curcumin reached up to 7.99 mg/g, with an encapsulation efficiency of 56.18%. 13C nuclear magnetic resonance spectroscopy peaks at 93–107 ppm and Fourier transform-infrared spectroscopy signals at 3386 cm−1 indicated the formation of a non-inclusion complex through hydrogen bond interactions between curcumin and short-chain amylose. While A-type crystalline pattern and weakening ordered crystallization structure were also probed. Additionally, the formation of non-inclusion complex improved the pH stability and photostability of curcumin through physical barrier of starch nanoparticles and the hydrogen bond interaction between curcumin and starch. Overall, this study established a novel and eco-friendly technique for loading and stabilizing curcumin with nanoscale dimensions, offering potential applications in edible packaging and functional foods.

Curcumin-conjugated Nanoparticles: An Approach to Target Mitochondria

Background: Unconjugated nanoparticles used in the treatment of cancer and various metabolic and neurodegenerative disorders exhibit extended blood circulation time, inhibition of enzymatic degradation, and increased chemical stability of the encapsulated therapeutic molecules. However, the unconjugated nanoparticles often show off-target effects and lack of specificity, which limit their applications in the healthcare system. Objective: Curcumin, a polyphenolic compound known for its proficiency to scavenge reactive oxygen species attributed to antioxidant activity and anticancer effects associated with mitochondrial dysfunction, is one of the most effective therapeutic agents. Therefore, with respect to chemotherapeutic strategies, the development of mitochondrial targeting curcumin nanoparticles for target-specific drug delivery has been extensively studied. Conclusion: The present review has focused on diverse curcumin-loaded nanoparticles targeting mitochondria along with their improved delivery potential, enhanced localization and biodistribution profile.

Injectable, Antioxidative, and Tissue-Adhesive Nanocomposite Hydrogel as a Potential Treatment for Inner Retina Injuries.

Reactive oxygen species (ROS) have been recognized as prevalent contributors to the development of inner retinal injuries including optic neuropathies such as glaucoma, non-arteritic anterior ischemic optic neuropathy, traumatic optic neuropathy, and Leber hereditary optic neuropathy, among others. This underscores the pivotal significance of oxidative stress in the damage inflicted upon retinal tissue. To combat ROS-related challenges, this study focuses on creating an injectable and tissue-adhesive hydrogel with tailored antioxidant properties for retinal applications. GelCA, a gelatin-modified hydrogel with photo-crosslinkable and injectable properties, is developed. To enhance its antioxidant capabilities, curcumin-loaded polydopamine nanoparticles (Cur@PDA NPs) are incorporated into the GelCA matrix, resulting in a multifunctional nanocomposite hydrogel referred to as Cur@PDA@GelCA. This hydrogel exhibits excellent biocompatibility in both in vitro and in vivo assessments, along with enhanced tissue adhesion facilitated by NPs in an in vivo model. Importantly, Cur@PDA@GelCA demonstrates the potential to mitigate oxidative stress when administered via intravitreal injection in retinal injury models such as the optic nerve crush model. These findings underscore its promise in advancing retinal tissue engineering and providing an innovative strategy for acute neuroprotection in the context of inner retinal injuries.

Curcumin-loaded pH-sensitive carboxymethyl chitosan nanoparticles for the treatment of liver cancer

In this study, the pH-responsive API-CMCS-SA (ACS) polymeric nanoparticles (NPs) based on 1-(3-amino-propyl) imidazole (API), stearic acid (SA), and carboxymethyl chitosan (CMCS) were fabricated for the effective transport of curcumin (CUR) in liver cancer. CUR-ACS-NPs with various degrees of substitution (DS) were employed to prepare through ultrasonic dispersion method. The effect of different DS on NPs formation was discussed. The obtained CUR-ACS-NPs (DSSA=12.4%) had high encapsulation rate (more than 85%) and uniform particle size (186.2 ± 1.42 nm). The CUR-ACS-NPs showed better stability than the other groups. Drug release from the CUR-ACS-NPs was pH-dependent, and more than 90% or 65% of CUR was released in 48 h in weakly acid medium (pH 5.0 or 6.0, respectively). Additionally, the CUR-ACS-NPs increased the intracellular accumulation of CUR and demonstrated high anticancer effect on HepG2 cells compared with the other groups. CUR-ACS-NPs prolonged the retention time of the drug, and the area under the curve (AUC) increased significantly in vivo. The in vivo antitumor study further revealed that the CUR-ACS-NPs exhibited the capability of inhibiting tumor growth and lower systemic toxicity. Meanwhile, CUR, CUR-CS-NPs, and CUR-ACS-NPs could be detected in the evaluated organs, including tumor, liver, spleen, lung, heart, and kidney in distribution studies. Among them, CUR-ACS-NPs reached the maximum concentration at the tumor site, indicating the tumor-targeting properties. In short, the results suggested that CUR-ACS-NPs could act a prospective drug transport system for effective delivery of CUR in cancer treatment.

Photostabilities and anti-tumor effects of curcumin and curcumin-loaded polydopamine nanoparticles.

Currently, the photostability of photosensitizer curcumin is the main bottleneck limiting their application, reducing the bioavailability of curcumin. Studying the effect of different light sources on the photostabilities of curcumin and loading it onto polydopamine nanocarriers with better biocompatibility will help improve its light utilization efficiency. In this study, we investigated the photostabilities of curcumin and a polydopamine-based nanoparticle (polydopamine-curcumin composite nanoparticles, PDA-Cur NPs) loaded with curcumin through in vitro and in vivo experiments to achieve better antitumor effects. The results demonstrated that curcumin has good photostability in dark, but with significant photodegradation rates in both red and blue light. Blue light has a faster effect on the photodegradation of curcumin, with a degradation rate of 42.1% after 10 minutes, which is about 1.7 times that of the red light. Our study successfully synthesized PDA-Cur NPs, demonstrating its ability to stably load and release curcumin, with a loading percentage of 65.7% after 2 hours and 41.9% release in 8 hours (pH 6.0). Compared with single curcumin treatments, the photodegradation rates of PDA-Cur NPs in red and blue light treatments were reduced by 46% and 50%, respectively. Meanwhile, PDA-Cur NPs exhibited remarkable antitumor efficacy due to PDT and promote apoptosis in cancer cells, which both better than that of single curcumin treatments. Moreover, in MCF-7 tumor-bearing mice, the PDA-Cur NPs led to significant tumor growth inhibition effects, without causing evident systemic damage in vivo. The findings highlight the potential of PDA-Cur NPs as anticancer photosensitizer with greatly increased utilization of curcumin in PDT.

Fabrication and characterization of curcumin-loaded composite nanoparticles based on high-hydrostatic-pressure-treated zein and pectin: Interaction mechanism, stability, and bioaccessibility

We successfully designed curcumin (Cur)-loaded composite nanoparticles consisting of high-hydrostatic-pressure-treated (HHP-treated) zein and pectin with a pressure of 150 MPa (zein-150 MPa-P-Cur), showing nano-spherical structure with high zeta-potential (−36.72 ± 1.14 mV) and encapsulation efficiency (95.64 ± 1.23 %). We investigated the interaction mechanism of the components in zein-150 MPa-P-Cur using fluorescence spectroscopy, molecular dynamics simulation, Fourier-transform infrared spectrometry and scanning electron microscopy techniques. Compared with zein-P-Cur, the binding sites and binding energy (−53.68 kcal/mol vs. − 44.22 kcal/mol) of HHP-treated zein and Cur were increased. Meanwhile, the interaction force among HHP-treated zein, pectin, and Cur was significantly enhanced, which formed a tighter and more stable particle structure to further improve package performance. Additionally, Cur showed the best chemical stability in zein-150 MPa-P-Cur. And the bioavailability of Cur was increased to 65.53 ± 1.70 %. Collectively, composite nanoparticles based on HHP-treated zein and pectin could be used as a promising Cur delivery system.

Toxicity Evaluation and Controlled-Release of Curcumin-Loaded Amphiphilic Poly-N-vinylpyrrolidone Nanoparticles: In Vitro and In Vivo Models.

Curcumin attracts huge attention because of its biological properties: it is antiproliferative, antioxidant, anti-inflammatory, immunomodulatory and so on. However, its usage has been limited by poor water solubility and low bioavailability. Herein, to solve these problems, we developed curcumin-loaded nanoparticles based on end-capped amphiphilic poly(N-vinylpyrrolidone). Nanoparticles were obtained using the solvent evaporation method and were characterized by dynamic and electrophoretic light scattering, transmission electron (TEM) and atomic force (AFM) microscopy. The average particle size was 200 nm, and the ζ-potential was -4 mV. Curcumin-release studies showed that nanoparticles are stable in aqueous solutions. An in vitro release study showed prolonged action in gastric, intestinal and colonic fluids, consistently, and in PBS. In vitro studies on epidermoid carcinoma and human embryonic kidney cells showed that the cells absorbed more curcumin in nanoparticles compared to free curcumin. Nanoparticles are safe for healthy cells and show high cytotoxicity for glioblastoma cells in cytotoxicity studies in vitro. The median lethal dose was determined in an acute toxicity assay on zebrafish and was 23 μM. Overall, the curcumin-loaded nanoparticles seem promising for cancer treatment.

Sport Medicine Principles Augment Healing Response in Spinal Cord Injury in a Rat Model Treated with a Curcumin-Loaded Nanocomposite Hydrogel

In this research, the potential healing function of regular exercise on spinal cord injury healing response was investigated in a rat model. The animals were treated with curcumin-loaded chitosan nanoparticles dispersed in a calcium alginate hydrogel containing endometrial stem cells. Study showed the animals that received both regular exercise and hydrogel showed significantly better histopathological signs and functional recovery than the other groups. Histopathological studies showed that edema, vacuolation, and fibrosis were significantly lower in this group. Gene expression studies showed that the hybrid treatment upregulated the expression levels of VEGF, b-FGF, and TGF-β genes.

Folic acid-conjugated cancer drug curcumin-loaded albumin nanoparticles: Investigation of curcumin release kinetics

The aim of this study is to design a drug delivery system in which both the carrier material and the flavonoid anticancer, chemopreventive and chemotherapeutic active substance to be loaded into this material are biologically sourced soft material. In this study, bovine serum albumin nanoparticles (BSANPs) were synthesized using “Desolvation Method”. Characterization studies were carried out by performing SEM, TEM, FT-IR, XRD, TGA, DSC and Zetasizer analyses. The most suitable BSA concentration was determined to be 25 mg/ml, and the size of BSANPs was measured at 156.9 nm with a PDI value of 0.078 in a medium with pH adjusted to 10 after 24 h of cross-linking. The optimum ratio of distilled water (solvent) to ethanol (non-solvent) for desolvation was found to be 1:2. The maximum CUR encapsulation efficiency and capacity of BSANPs were obtained as 90.2 % and 44.22 %, respectively, at a cross-linking time of 24 h using 15 mg of CUR and 110 μl of glutaraldehyde. The cumulative CUR release percentage from CUR-BSANPs containing 0.5, 1.0 and 1.5 mg CUR was determined to be 51.6 %, 67.3 % and 56.2 %, respectively, after 72–168 h in pH 5.6 PBS buffer. The conjugation of folic acid to CUR-BSANPs slightly reduced the release rate and the cumulative percentage release of CUR, but still provided a sustained and controlled release of CUR. In order to analyze the CUR release kinetic mechanism, the fit of CUR release profiles to zero order, first order, Higuchi, Hixson-Crowell and Korsmeyer-Peppas models was examined. The values of kinetic constants for each model were calculated.

Curcumin-loaded chitosan-protamine nanoparticles: A promising approach to ameliorate nicotine-induced reproductive disorders in male rats

Nicotine (NIC) exposure has been shown to impair male fertility, affecting sperm quality and motility. Curcumin (CUR) has been shown to have anti-inflammatory and antioxidant properties and has demonstrated potential in treating a range of health conditions, including infertility. The current study aimed to investigate the effects of curcumin encapsulated in chitosan-protamine nanoparticles (CUR-CPNPs) against nicotine-induced reproductive dysfunction in male rats. CUR-CPNPs were chemically prepared and characterized by TEM imaging, Zetasizer, and FT-IR spectroscopy. eight groups (6 rats per each) of male rats were used: first & second groups acted as control, third group received CUR-CPNPs (250 mg/kg), fourth group received CUR (250 mg/kg), fifth group received CPNPs (250 mg/kg), sixth group received 0.6 mg/kg nicotine (NIC). Finally, seventh and eighth groups are received NIC + CUR, NIC + CUR-CPNPs, respectively. After the end of the treatment period, the rats were anesthetized then the relative weight of the reproductive organs, semen analysis, testosterone levels and oxidative stress biomarkers were assessed, and finally a histopathological analysis of the testicular tissue was performed.The impacts of NIC administration resulted in a significant reduction in relative reproductive organs weights, exacerbated oxidative stress, decreased serum testosterone level, and negatively impacted semen count, motility, and viability. Testicular histopathology revealed extensive degenerative alterations. On the other hand, the ameliorative role of CUR-CPNPs clearly reversed the devastating effect caused by nicotine.

Fabrication and characterization of curcumin-loaded nanoparticles using licorice protein isolate from Radix Glycyrrhizae

Licorice was widely used in food and herbal medicine. In its extract industry, a substantial amount of licorice protein was produced and discarded as waste. Herein, we extracted Licorice Protein Isolate (LPI) and explored its potential as a curcumin nanocarrier. Using a pH-driven method, we fabricated LPI-curcumin nanoparticles with diameters ranging from 129.30 ± 3.21 nm to 75.03 ± 1.19 nm, depending on the LPI/curcumin molar ratio. The formation of LPI-curcumin nanoparticles was primarily driven by hydrophobic interactions, with curcumin entrapped in LPI being in an amorphous form. These nanoparticles significantly enhanced curcumin properties in terms of solubility, photochemical stability, and stability under varying pH, storage, and physiological conditions. Moreover, the loaded curcumin exhibited a 2.58-fold increase in cellular antioxidant activity on RAW 264.7 cells and a 1.86-fold increase in antitumor activity against HepG2 cells compared to its free form. These findings suggested that LPI could potentially serve as a promising novel delivery material.

Screening the statistical impact of some independent variables on the particle size and entrapment efficiency (EE) for the design and development of curcumin-loaded PLGA-Tf nanoparticles

The present study provides the insight of statistical aspects of various independent variables like Tf-PLGA Curcumin ratio, stirring speed and emulsifying concentration over dependent variables like particle size and EE of the drug. Design Expert Version 13.1.5 was used to analyse the impact of independent variables over dependent variables. To study the effect of intendent variables over particle size, the valve Predicted R² was 0.9701 is in reasonable agreement with the Adjusted R² of 0.9907; i.e., the difference is less than 0.2. indicating that stirring speed have a great impact of particle size. While in another fact also shown the Adeq Precision measures the signal to noise ratio. A ratio greater than 4 is desirable and in respect of that the ratio of 9.630 indicates an adequate signal, i.e. it greatly gives impact on size of nanoparticles. In the case of entrapment efficacy (EE) the Predicted R² of 0.9892 is in reasonable agreement with the Adjusted R² of 0.9910; i.e. the difference is less than 0.2 which support the significance of the study and impact of Tf- PLGA curcumin ratio over EE. Adeq Precision measures the signal to noise ratio. A ratio greater than 4 is desirable but for this study the ratio was calculated as14.989 indicates an adequate signal. Therefore, the two independent variables like, Tf-PLGA Curcumin ratio and stirring speed have significant impact on EE and particle size of nanoparticles.
