Fenpropathrin provoked kidney damage via controlling the NLRP3/Caspase-1/GSDMD–mediated pyroptosis: The palliative role of curcumin-loaded chitosan nanoparticles

This study assessed the ability of formulated curcumin-loaded chitosan nanoparticles (CU-CS-NPs) to reduce the kidney damage resulting from fenpropathrin (FPN) in rats compared to curcumin (CU) in rats. Sixty male Sprague Dawley rats were separated into six groups and orally administered 1 mL/kg b.wt corn oil, 50 mg CU/kg b.wt, 50 mg CU-CS-NPs /kg b.wt., 15 mg FPN /kg b.wt, CU+ FPN or CU-CS-NPs + FPN for 60 days. Then, serum renal damage products were assessed. Total antioxidant capacity, reactive oxygen species, interleukin 1β (IL-1β), malondialdehyde, NF-κB P65, cleaved-Caspase-1, and Caspase-8 were estimated in kidney homogenates. The cleaved Caspase-3 and TNF-α immunoexpression and pyroptosis-related genes were determined in renal tissues. The results showed that CU-CS-NPS significantly repressed the FPN-induced increment in kidney damage products (urea, uric acid, and creatinine). Moreover, the FPN-associated hypo-proteinemia, renal oxidative stress and apoptotic reactions, and impaired renal histology were considerably repaired by CU and CU-CS-NPs. Additionally, compared to FPN-exposed rats, CU, and CU-CS-NPs-treated rats had considerably lower immunoexpression of cleaved Caspase-3 and TNF-α in renal tissue. The pyroptosis-related genes NLRP3, GSDMD, IL-18, Caspase-3, Caspase-1, IL-1β, Caspase-8, TNF-α, and NF-κB dramatically upregulated by FPN exposure in the renal tissues. Yet, in CU and CU-CS-NPs-treated rats, the gene above expression deviations were corrected. Notably, CU-CS-NPs were superior to CU in preventing oxidative damage and inflammation and regulating pyroptosis in the renal tissues of the FPN-exposed group. The results of the present study conclusively showed the superior favorable effect of CU-CS-NPs in counteracting renal impairment linked to environmental pollutants.

Sport Medicine Principles Augment Healing Response in Spinal Cord Injury in a Rat Model Treated with a Curcumin-Loaded Nanocomposite Hydrogel

In this research, the potential healing function of regular exercise on spinal cord injury healing response was investigated in a rat model. The animals were treated with curcumin-loaded chitosan nanoparticles dispersed in a calcium alginate hydrogel containing endometrial stem cells. Study showed the animals that received both regular exercise and hydrogel showed significantly better histopathological signs and functional recovery than the other groups. Histopathological studies showed that edema, vacuolation, and fibrosis were significantly lower in this group. Gene expression studies showed that the hybrid treatment upregulated the expression levels of VEGF, b-FGF, and TGF-β genes.

Preparation and characterization of 3D bioprinted gelatin methacrylate hydrogel incorporated with curcumin loaded chitosan nanoparticles for in vivo wound healing application

This study developed a biomimetic composite bioink consisting of gelatin methacrylate (GelMA) /chitosan nanoparticles (CSNPs) for extrusion-based 3D bioprinting. Additionally, curcumin(Cur)-loaded nanoparticles were incorporated which increased the proliferation and antibacterial activity of biomimetic skin constructs. The hydrogel, curcumin-loaded NPs, and the biocomposite was characterized chemically and physically. The results indicated proper modified gelatin with tunable physical characteristics, e.g., swelling ratio and biodegradability up to 1200 % and 25 days, respectively. In addition, the characterized CSNPs showed good distribution with a size of 370 nm and a zeta potential of 41.1 mV. We investigated the mechanical and cytocompatibility properties of chitosan nanoparticles encapsulated in hydrogel for emulating an extracellular matrix suitable for skin tissue engineering. CSNPs entrapped in GelMA (15 % w/v) exhibited controlled drug release during 5 days, which was fitted into various kinetic models to study the mass transfer mechanism behavior. Also, the composite hydrogels were effective as a barrier against both gram-positive and gram-negative bacteria at a concentration of 50 μg/ml nanoparticles in GelMA 15 %. Furthermore, the biocomposite was applied on Wistar rats for wound healing. As a result, this study provides a GelMA-NP50-Cur3 scaffold that promotes cell proliferation and decreases microbial infections in wounds.

Curcumin-loaded chitosan nanoparticles alleviate fenpropathrin-induced hepatotoxicity by regulating lipogenesis and pyroptosis in rats.

In this study, the probable alleviative role of curcumin (CMN) (50mg/kg b.wt) or curcumin-loaded chitosan nanoparticle (CLC-NP) (50mg/kg b.wt) was assessed against the hepatotoxic effect of a widely used pyrethroid insecticide, fenpropathrin (FEN) (15mg/kg b.wt) in rats in a 60-day experiment. The results revealed that CMN and CLC-NP significantly suppressed the FEN-induced increment in serum hepatic enzyme activities (ALT, AST, and ALP) and hyperbilirubinemia. Moreover, FEN-associated dyslipidemia, hepatic oxidative stress, and altered hepatic histology were significantly rescued by CMN and CLC-NP. Furthermore, the increased TNF-α and Caspase-3 immunoexpression in hepatic tissues of FEN-exposed rats was significantly reduced in CMN and CLC-NP-treated ones. FEN exposure significantly upregulated the pyroptosis-related genes, including GSDMD, Casp-1, Casp-3, Casp-8, IL-18, TNF-α, IL-1β, and NF-κB and altered the expression of lipogenesis-related genes including SREBP-1c, PPAR-α, MCP1, and FAS in the hepatic tissues. Nevertheless, the earlier disturbances in gene expression were corrected in CMN and CLC-NP-treated groups. Of note, compared to CMN, CLC-NP was more effective at inhibiting oxidative damage and controlling lipogenesis and pyroptosis in the hepatic tissues of FEN-exposed rats. Conclusively, the current study findings proved the superior and useful role of CLC-NP in combating pollutants associated with hepatic dysfunction.

Evaluation of curcumin-loaded chitosan nanoparticles for wound healing activity.

Wound healing is a biological process that can be difficult to manage clinically. In skin wound healing, the interaction of many cells, growth factors, and cytokines reveals an outstanding biological function mechanism. Wound healing that occurs naturally restores tissue integrity, however, it is usually restricted to wound repair. Curcumin synthesised in a chitosan matrix can be used to heal skin sores. The ionotropic gelation procedure required crosslinking chitosan with a tripolyphosphate (TPP) crosslinker to generate curcumin nanoparticles encapsulated in chitosan. The nanoparticles were between 200 and 400 nm in size, with a strong positive surface charge and good entrapment efficacy, according to SEM and TEM investigations. Curcumin and chitosan compatibility was investigated using FTIR spectroscopy. All batches showed consistent drug release, with the F5 batch having the highest curcumin release, at 75% after 16 hours. On L929 cells, scratch assays were utilised to assess wound healing. Wound closure with widths of 59 and 65 mm with curcumin and 45 and 78 mm with curcumin-loaded chitosan nanoparticles was seen after 24 and 48 hours of examination. According to the findings, prepared curcumin chitosan nanoparticles are beneficial in healing skin damage.

Synthesis, characterization and evaluation of anti-arthritic and anti-inflammatory potential of curcumin loaded chitosan nanoparticles

Curcumin is a phytochemical isolated from the dried rhizome of Curcuma longa L. family Zingiberaceae which possesses versatile biological activities and has hydrophobic properties. The current study was conducted to fabricate, and optimize curcumin loaded chitosan and Sodium tripolyphosphate (STPP) nanoparticles (NPs) to improve the bioavailability of curcumin. NPs were fabricated employing the Ionic gelation method. Four formulations were developed based on the selected variables like STPP and chitosan concentration, rotations per minute (rpm), temperature, and pH of chitosan solution. NPs were characterized for morphology, drug-polymer compatibility, yield, particle size, encapsulation efficiency, release behavior, anti-inflammatory and anti-arthritic activity compared to curcumin and standard drug. Fourier transform infrared spectroscopy (FTIR) shows nanoparticle compatibility. The maximum yield was 60%. Entrapment efficiency ranged from 45 to 65%. Curcumin NPs and standard drug 600 µg/ml shows 59% and 70% anti-inflammatory activity by HRB membrane stabilization method respectively which are greater than curcumin alone whereas anti-arthritic activity by protein denaturation method which is also comparable to standard drug and greater than curcumin was 66 and 70% respectively. Statistical analysis shows the mean significant difference at p ≤ 0.05. The study concluded that curcumin-loaded chitosan and STPP NPs formulated successfully by the Ionic gelation method, which increased curcumin absorption leading to a reduced dosing rate and improved patient compliance.

In vitro anticancer activity of curcumin loaded chitosan nanoparticles (CLCNPs) against Vero cells

The global burden of cancer is rising due to increasing pollution, change in lifestyles and dietary habits rendering it as one of the leading cause of death worldwide. Poor availability of anticancer drugs in affected tissues necessitates higher doses and longer treatment regimens to treat most cancers. In the present investigation curcumin, an anticancer drug was loaded in chitosan nanoparticles by ionic gelation method and their anticancer effect was evaluated against the Vero cell line. The characteristics of nanoparticles were studied by UV–Vis spectroscopy, Fourier Transform Infrared spectroscopy (FTIR), Thermogravimetric analysis (TGA), Differential thermal analysis (DTA), Scanning electron microscopy (SEM) and Transmission electron microscopy (TEM). UV–Vis spectroscopy confirmed the synthesis of CLCNPs, whereas FTIR indicated different functional groups associated with the formation of these nanoparticles. TGA and DTA confirmed the stability of nanoparticles at high temperature. SEM revealed spherical shape of CLCNPs whereas TEM confirmed loading of curcumin in chitosan nanoparticles. The anticancer activity of CLCNPs was evaluated against Vero cell line in different concentrations (10, 20, and 50 µg) and it was observed that 50 µg concentration was most effective. CLCNPs can be used to treat cancer cells as curcumin is poorly soluble in aqueous solutions.

The dual gastro- and neuroprotective effects of curcumin loaded chitosan nanoparticles against cold restraint stress in rats

Stress is a condition affecting different body systems. Curcumin (CUR) is a natural compound that has various pharmacological benefits. However, its poor oral bioavailability limits its therapeutic value. This study aimed to formulating curcumin loaded chitosan nanoparticles (CS.CUR.NPs) and investigate its gastroprotective and neuroprotective effects in rats subjected to cold restraint stress (CRS), in reference to conventional oral CUR preparation, and explore its underlying mechanism. Treated groups received either CUR or CS.CUR.NPs (100 mg∕kg) orally for 14 days before exposure to CRS. CRS elicited marked behavioral changes and gastric ulcer accompanied by histopathological abnormalities of the brain and stomach along with elevation of pain score. CUR and CS.CUR.NPs improved stress-induced gastric ulcer, cognitive performance, and pain sensation. Mechanistically, CRS disrupts oxidative and inflammatory status of the brain as manifested by high malondialdehyde and IL-6 and low total antioxidant capacity and IL-10, along with high C-reactive protein level. CRS decreased nuclear factor erythroid 2-related factor2 (Nrf2) and increased nuclear factor-kappa B (NF-κB) expressions. Furthermore, brain levels of unphosphorylated signal transducer and activator of transcription3 (U-STAT3) and glial fibrillary acidic protein (GFAP) were upregulated with stress. CUR and CS.CUR.NPs provided beneficial effects against harmful consequences resulting from stress with superior beneficial effects reported with CS.CUR.NPs. In conclusion, these findings shed light on the neuroprotective effect of CUR and CS.CUR.NPs against stress-induced neurobehavioral and neurochemical deficits and protection against stress-associated gastric ulcer. Moreover, we explored a potential crosslink between neuroinflammation, U-STAT3, NF-κB, and GFAP in brain dysfunction resulted from CRS.

Dietary supplementation of curcumin-loaded chitosan nanoparticles stimulates immune response in the white leg shrimp Litopenaeus vannamei challenged with Vibrio harveyi

Pathogen infections in shrimps trigger the release of reactive oxygen species (ROS) as a part of immune response. The excessive accumulation of ROS causes the production of oxidative stress, which leads to oxidative damage of the biomolecules in the host cells. The inclusion of dietary antioxidants is known to mitigate oxidative stress and stimulate immunity. Curcumin, a potential antioxidant was encapsulated in chitosan nanoparticles to surge its bioavailability and was administered orally to Vibrio harveyi challenged and non-challenged Litopenaeus vannamei. The non-challenged shrimps fed with curcumin-loaded chitosan nanoparticles (Cur-CSNPs) showed a significant increase (p ≤ 0.05) in the specific growth rate, daily growth coefficient and survival rate. A significant increase (p ≤ 0.05) in the phenoloxidase activity, total hemocyte count and superoxide dismutase activity was observed in both the challenged and non-challenged shrimps fed with Cur-CSNPs. Additionally, a significant increase (p ≤ 0.05) in the relative mRNA expression of lysozyme, cMnSOD and lectin was observed in the Cur-CSNPs fed shrimps. The findings of this research suggest that Cur-CSNPs reinforce the immune system of L. vannamei against V. harveyi infection. Moreover, the non-challenged shrimps showed improvement in the growth parameters in addition to immunostimulation. Thereby a routine inclusion of dietary Cur-CSNPs could mitigate the oxidative damage caused by the incidence of environmental or pathogen-mediated oxidative stress.

Therapeutic effects of EGF-modified curcumin/chitosan nano-spray on wound healing.

Dermal injury, including trauma, surgical incisions, and burns, remain the most prevalent socio-economical health care issue in the clinic. Nanomedicine represents a reliable administration strategy that can promote the healing of skin lesions, but the lack of effective drug delivery methods can limit its effectiveness. In this study, we developed a novel nano-drug delivery system to treat skin defects through spraying. We prepared curcumin-loaded chitosan nanoparticles modified with epidermal growth factor (EGF) to develop an aqueous EGF-modified spray (EGF@CCN) for the treatment of dermal wounds. In vitro assays showed that the EGF@CCN displayed low cytotoxicity, and that curcumin was continuously and slowly released from the EGF@CCN. In vivo efficacy on wound healing was then evaluated using full-thickness dermal defect models in Wistar rats, showing that the EGF@CCN had significant advantages in promoting wound healing. On day 12 post-operation, skin defects in the rats of the EGF@CCN group were almost completely restored. These effects were related to the activity of curcumin and EGF on skin healing, and the high compatibility of the nano formulation. We therefore conclude that the prepared nano-scaled EGF@CCN spray represents a promising strategy for the treatment of dermal wounds.

Novel curcumin-loaded chitosan-polyelectrolyte complexed nanoparticles and their characteristics

Curcumin was incorporated into oil/water (o/w) emulsion by dissolving it in soybean oil to cover the surface of the oil droplets with a pH-sensitive polyelectrolyte complex (PEC) composed of chitosan and fucoidan molecules. This curcumin-loading efficiency of the suspension system was investigated. The size and zeta potential distributions of novel curcumin-loaded chitosan nanoparticles self-assembled with fucoidan (CFNPLC) were assessed at various pH and fucoidan-to-chitosan mass ratio (FCMR). The release behavior of curcumin from CFNPLC was confirmed quantitatively by the superimposition of relevant release behavior. The release of curcumin from CFNPLC prepared with a chitosan solution of pH 6 for one to two days was slower than that from CFNPLC prepared with a chitosan solution of pH 3.7 for five to 10 hrs. This was attributed to the higher affinity of a chitosan molecule to curcumin molecules at a higher pH. The centrifugation of release medium accelerated the release of curcumin droplet from the surface of CFNPLC into the release medium much faster than a conventional curcumin release. Not to mention a high stability of curcumin blended with soybean oil, encapsulated in CFNPLC, the advantage of the CFNPLC is addressed in such a way that curcumin releasing period of a conventional curcumin delivery using CFNPLC, is supposed to be extended for much longer time than one to two days of the curcumin release period in this study. Thus, CFNPLC can bring about an enhanced effect of curcumin bioavailability resulting from the high curcumin stability and its extended release because it was dissolved in soybean oil and CFNPLC sustained slow curcumin-release for more than days in the oral drug (curcumin) delivery system. Therefore, CFNPLC can be treated mainly as a food-functional additive or healthy dietary product for tumor patients.

Wound healing performance of PCL/chitosan based electrospun nanofiber electrosprayed with curcumin loaded chitosan nanoparticles

In this study, the electrospun poly(ε-caprolactone) (PCL)/Chitosan (CS)/curcumin (CUR) nanofiber was fabricated successfully with curcumin loaded chitosan nano-encapsulated particles (CURCSNPs). The morphology of the produced CURCSNPs, PCL, PCL/CS, PCL/CS/CUR, and PCL/CS/CUR electrosprayed with CURCSNPs were analyzed by scanning electron microscopy (SEM). The physicochemical properties and biological characteristics of fabricated nanofibers such as antibacterial, antioxidant, cell viability, and in vivo wound healing efficiency and histological assay were tested. The electrospraying of CURCSNPs on surface PCL/CS/CUR nanofiber resulted in the enhanced antibacterial, antioxidant, cell proliferation efficiencies and higher swelling and water vapor transition rates. In vivo examination and Histological analysis showed PCL/CS/CUR electrosprayed with CURCSNPs led to significant improvement of complete well-organized wound healing process in MRSA infected wounds. These results suggest that the application of PCL/CS/CUR electrosprayed with CURCSNPs as a wound dressing significantly facilitates wound healing with notable antibacterial, antioxidant, and cell proliferation properties.

Curcumin loaded chitosan nanoparticles fortify shrimp feed pellets with enhanced antioxidant activity

Chitosan nanoparticles (CSNPs) have been recently explored as a potential drug carrier to enhance the bioavailability and aqueous solubility of drugs. Curcumin, an antioxidant with a remarkable antiradical scavenging activity was encapsulated in CSNPs to revamp its bioavailability. While changes in the optimal farming condition can induce oxidative stress in the animals, curcumin loaded chitosan nanoparticles (Cur-CSNPs) were amalgamated into shrimp feed pellets to ameliorate its antioxidant content in an attempt to bolster the organisms against oxidative stress. Cur-CSNPs were synthesized in two different concentrations of curcumin as Cur-CSNPs A and B. Characterization of the synthesized Cur-CSNPs revealed asymmetrical nanoparticles with semispherical geometry and a zeta potential ˃50 mV. HPLC studies substantiated encapsulation efficiencies of 77.53% and 80.35% for Cur-CSNPs A and B respectively. DPPH, ABTS and FRAP assays manifested a significant enhancement in the antioxidant property of the Cur-CSNPs fortified feed pellets. This is the first study to investigate and demonstrate the ability of Cur-CSNPs to enhance the antioxidant property of aquaculture feed pellets. These findings substantiate that Cur-CSNPs fortified feed may be applied to reinforce aquaculture animals against oxidative stress.

Cold atmospheric plasma modification of curcumin loaded in tri‐phosphate chitosan nanoparticles enhanced breast cancer cells apoptosis

The anti‐cancer mechanisms of curcumin have been reported to include suppressions of angiogenesis and tumor proliferation. The main goal of this research is to increase the solubility of curcumin by cold atmospheric plasma (CAP) and assess the effects of modified curcumin by charging with tri‐polyphosphate chitosan nanoparticles for MCF‐7, MDA‐MB‐231 breast cancer cells, and human fibroblast cells. Curcumin modification was done by CAP and its solubility was evaluated by spectrophotometry. After loading modified curcumin into nano‐chitosan‐TPP, nanocurcumin was characterized by scanning electron microscopy. Cellular viability and apoptosis of treated cells were assessed by MTT and Annexin V. The changes of messenger RNA expression of TP5353 and VEGF genes were analyzed by real‐time PCR. CAP was able to transform the curcumin to possess hydrophilic characteristics after 90 seconds. The mean diameter of Curcumin loaded chitosannanoparticles (NPs) were determined as 48 nm. MTT results showed that the IC50 of nano Cur‐chitosan‐TPP was effectively decreased compared to free curcumin in MCF‐7 (15 μg/mL at 72 hours vs 20 μg/mL at 48 hours). Additionally, nano Cur‐chitosan‐TPP had no significant effect on normal cells (Human dermal fibroblas: HDF), whereas it also decreased the viability of triple negative breast cancer cell line (MDA‐MB‐231). Real‐time PCR results showed that expression level of TP53 gene was upregulated (P = .000), whereas VEGF gene downregulated (P = .000) in treated MCF‐7 cells. Curcumin loaded chitosan nanoparticles have led to an induction of apoptosis (79.93%) and cell cycle arrest (at S and G2M). Modified‐curcumin‐tri‐polyphosphate chitosan nanoparticles using CAP can be considered as a proper candidate for breast cancer treatment.

Preparation and antibiofilm studies of curcumin loaded chitosan nanoparticles against polymicrobial biofilms of Candida albicans and Staphylococcus aureus

Polymicrobial biofilms related infections are an important clinical problem with classical antibiotics being not sufficient in therapy. Here, curcumin (Cur) was loaded on positively charged chitosan nanoparticles (CSNP). The antibiofilm activities against mono- and polymicrobial biofilms of Candida albicans and Staphylococcus aureus were evaluated. The average diameter of CSNP-Cur was 134.37 ± 1.99 nm and its surface charge was +18.10 ± 0.82 mV. Cur released from NPs was slower at pH 7.4 than at pH 5.4. Scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM) were used to observe biofilm architecture and live/dead organisms within biofilm on medical silicone surface. CSNP-Cur exhibited excellent antibiofilm activity against planktonic bacteria or fungi, mono- and polymicrobial biofilm formations and preformed biofilms. SEM and CLSM showed that CSNP-Cur was able to reduce biofilm thickness as well as kill microbial cells embedded in biofilm on silicone surfaces.

Formulation and comparative characterization of nanoparticles of curcumin using natural, synthetic and semi-synthetic polymers for wound healing

AimThe present research work aimed to prepare and characterize nanoparticles of curcumin using polymers from different sources like natural, synthetic and semi-synthetic, and compare their activity for wound healing. Curcumin, BCS class II drug, is a polyphenol with proven wound healing activity. Material and methodsThe curcumin-loaded chitosan and carboxymethylcellulose (CMC) nanoparticles were prepared by ionotropic gelation method. In contrast, poly-lactic co-glycolic acid (PLGA) nanoparticles were prepared by a double emulsion solvent evapouration method. The different sources of polymers include natural (chitosan), synthetic (PLGA) and semi-synthetic CMC were used for the preparation of nanoparticles. Key findingsThe percentage entrapment efficiency of curcumin-loaded nanoparticles was found to be in the order of polymers PLGA>chitosan>CMC. The in-vitro release study of carboxymethyl cellulose -curcumin nanoparticles was found to be 74.96% for 24 h. The presence of a specific peak of curcumin in all the polymeric nanoparticles specifies drug incorporation in the polymeric matrix. The in-vivo study revealed that curcumin-loaded chitosan nanoparticles fasten the healing process of the wound due to the synergistic effect produced by using a combination of curcumin and chitosan. SignificanceCurcumin-loaded nanoparticles showed significant enhancement in wound healing action by lowering the dose of curcumin and effecting synergistically due to the use of chitosan.

Nephroprotective effects of curcumin loaded chitosan nanoparticles in cypermethrin induced renal toxicity in rabbits.

Cypermethrin, a pyrethroid insecticide, may cause several adverse effects including nephrotoxicity. Curcumin is a nutraceutical with many pharmacological effects including nephroprotective effects. But its effective clinical use is limited due to poor bioavailability, physicochemical instability, low bioactive absorption, quick metabolization, less penetration, and targeting efficacy. To resolve these issues, curcumin is incorporated in chitosan nanoparticles. The focus of the study was to prepare and characterize curcumin loaded chitosan nanoparticles and evaluate their nephroprotective activity in a cypermethrin induced renal toxicity. The curcumin loaded chitosan nanoparticles were prepared by using solvent displacement method and characterized by particle size, zeta potential, polydispersity index, entrapment efficiency, and FTIR. The prepared formulation was stable and lies within nanometer range (264.8nm), and possessed high drug loading capacity (84.64%). Cypermethrin (24mg/kg body weight) and Curcumin loaded chitosan nanoparticles (15mg/kg and 30mg/kg body weight) were orally administered to 20 rabbits (4 groups) for 28days.
