A bioprocess engineering approach for the production of hydrocarbons and fatty acids from green microalga under high cobalt concentration as the feedstock of high-grade biofuels

Botryococcus braunii, a colonial green microalga which is well-known for its capacity to synthesize hydrocarbons, has significant promise as a long-term source of feedstock for the generation of biofuels. However, cultivating and scaling up B. braunii using conventional aqua-suspended cultivation systems remains a challenge. In this study, we optimized medium components and light intensity to enhance lipid and hydrocarbon production in a multi-cultivator airlift photobioreactor. BBM 3N medium with 200 μmol/m2/s light intensity and a 16 h light–8 h dark regimen yielded the highest biomass productivity (110.00 ± 2.88 mg/L/day), as well as the highest lipid and hydrocarbon content. Cultivation in a flat-panel bioreactor resulted in significantly higher biomass productivity (129.11 ± 2.74 mg/L/day), lipid productivity (32.21 ± 1.31 mg/L/day), and hydrocarbon productivity (28.98 ± 2.08 mg/L/day) compared to cultivation in Erlenmeyer flasks and open 20-L raceway pond. It also exhibited 20.15 ± 1.03% of protein content including elevated levels of chlorophyll a, chlorophyll b, and carotenoids. This work is noteworthy since it is the first to describe fatty acid and hydrocarbon profiles of B. braunii during cobalt treatment. The study demonstrated that high cobalt concentrations (up to 5 mg/L of cobalt nitrate) during Botryococcus culture affected hydrocarbon synthesis, resulting in high amounts of n-alkadienes and trienes as well as lipids with elevated monounsaturated fatty acids concentration. Furthermore, pyrolysis experiments on microalgal green biomass and de-oiled biomass revealed the lipid and hydrocarbon compounds generated by the thermal degradation of B. braunii that facilitate extra economical value to this system.

A Mediterranean diet improves glycation markers in healthy people and in those with chronic diseases: a systematic review of clinical trials.

Consumption of the Mediterranean diet (MedDiet) has beneficial effects on cardiometabolic health and aging. This systematic review aimed to critically investigate the effect of the MedDiet on glycation markers in healthy or overweight individuals with type 2 diabetes or cardiovascular disease. MEDLINE, EMBASE, Web of Science, and the Cochrane Library were searched, using the terms "Mediterranean diet" AND "glycation end products, advanced". Three randomized and 3 nonrandomized clinical trials, containing data on 2935 adult and elderly individuals with normal weight or overweight, were included. All extracted data were compiled, compared, and critically analyzed. The authors of most of the studies demonstrated a reduction in serum concentrations of advanced glycation end products (AGEs), such as εN-carboxymethyllysine and methylglyoxal, and in skin autofluorescence levels after at least 4 weeks of adherence to the MedDiet. The MedDiet also led to positive effects on gene expression of receptors for AGEs, as RAGE and AGER1, and an enzyme involved in detoxification (glyoxalase I). There is no evidence that short-term adherence affects glycation markers. Glycation markers improved in response to the MedDiet. The possible mechanisms involved may be related to the low AGE and refined sugars content of the diet, as well as its high monounsaturated fatty acid, phenolic compound, and dietary fiber contents. Normal fat intake with high MUFA content and an appropriate SFA/MUFA/PUFA ratio improved the health of rats

AbstractThe quantity and quality of dietary fats have a significant impact on human health. This study aimed to investigate the effects of different cooking oils (olive oil (OO), soybean oil (SO), and blended oil (BO)) on growth, glucolipid metabolism, inflammation, and oxidative stress of rats fed normal and high‐fat diets. The results showed that the OO‐fed group had lower concentrations of serum triglyceride (TG), low‐density lipoprotein cholesterol (LDL‐C), malondialdehyde (MDA), oxidized LDL‐C (ox‐LDL‐C), and serum total cholesterol (TC) than the normal‐fat SO‐fed group, and rats in the BO‐fed group had lower values of serum interleukin‐6 (IL‐6), high‐sensitivity C‐reactive protein (hs‐CRP), tumor necrosis factor (TNF‐α), and nonesterified fatty acids (NEFA) than the normal‐fat SO‐fed groups. However, higher serum glutathione peroxidase (GSH‐Px) activity was observed in the OO‐fed group, while those fed with BO had higher serum superoxide dismutase (SOD) and GSH‐Px activities. Generally, most health‐beneficial fatty acid profiles have a high content of monounsaturated fatty acids (MUFAs), an appropriate saturated fatty acid (SFA)/MUFA/poly unsaturated fatty acid (PUFA) ratio, and a low n‐6/n‐3 PUFA ratio, while diets contained a high content of fat is detrimental to health.

Carcass and meat characteristics of Nellore young bulls fed diet using cottonseed cake as a replacer of the forage fiber source

The aim of this study was to assess the effects of substituting traditional forage fiber sources with cottonseed cake in the diet on both the quantitative and qualitative characteristics of carcass and meat in Nelore young bulls. Twenty-four Nelore steers starting with an average weight of 377.8 ± 43.5 kg, were individually housed in stalls and provided with individualized feeding over a 112-day confinement period. The study followed a completely randomized design with two treatments and 12 replications. The diets incorporated either whole plant corn silage (WPCS) and, cottonseed cake (CSC) as fiber sources, at a rate of 300 g/kg of dry matter. The CSC diet promoted higher carcass weight. Aging animal meat for seven days significantly decreased the shear force from 83.4 to 71.6 N. Although diets did not influence meat composition, WPCS diet provided higher concentrations of C16:1, C18:1n9c, C18:3n3, and C22:2 acid, and CSC diet higher concentrations of C15:0, C18:1n9t, C18:2n6c, and 20:3n3. The WPCS diet provided higher concentrations of monounsaturated fatty acids and ω9, and the CSC diet had higher concentrations of ω6 and ω6:ω3 ratio in meat. Cottonseed cake used as a fiber source increases the concentration of polyunsaturated fatty acids and ω6 fatty acids in the meat of young bulls finished in feedlot.

Effects of high moisture ear corn on production performance, milk fatty acid composition, serum antioxidant status, and immunity in primiparous dairy cows.

This study evaluated the effects of high moisture ear corn (HMEC) on production performance, milk fatty acid composition, serum antioxidant status, and immunity in primiparous dairy cows. A total of 45 healthy primiparous Holstein cows (36.50±4.30 kg of milk/day, 201±9.00 lactating days in milk) were sorted into 3 groups: control group (CG, n = 15), 50% HMEC (replacing 50% steam-flaked corn with HMEC, n = 15), and 100% HMEC (replacing steam-flaked corn with HMEC, n = 15) on an equal dry matter (DM) basis. The study consisted of adaptation period of 14 days, followed by a formal period of 60 days. Feed intake and milk yield were recorded daily. Milk and blood samples were collected on 1, 30, and 60 d of the experimental period. The 50% HMEC group and 100% HMEC group significantly increased (p<0.05) milk yield and dry matter intake (DMI) in dairy cows compared to the control group (CG). The 100% HMEC group showed an increase (p<0.05) in 4% fat-corrected milk (4% FCM). Both the 50% HMEC group and 100% HMEC group exhibited significant decreases (p<0.05) in the content of C10:0, C12:0, and C14:0 fatty acids, along with a significant increase (p<0.05) in cis-9C18:1 content. The saturated fatty acid (SFA) content was significantly lower (p<0.05) in the 50% HMEC and 100% HMEC groups than that of CG. Conversely, the monounsaturated fatty acid (MUFA) content was higher (p<0.05) in the 50% HMEC and 100% HMEC groups than that in CG. Notably, the 100% HMEC group significantly increased (p<0.05) the serum superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) content, while also decreasing the serum malondialdehyde (MDA) content (p<0.05). Moreover, the 100% HMEC group significantly increased (p<0.05) the content of immunoglobulin G (IgG) and immunoglobulin M (IgM). High moisture ear corn could improve production performance and milk fatty acid levels and enhance immunity and antioxidant capacity in dairy cows. These results lay the foundation for the wider application of HMEC in ruminant animal diets.

Cocoa by-product inclusion in dairy sheep diet: effect on sensory, volatile and antioxidant properties of cheese

The possibility of inclusion of agro-industrial by-products in the diet of small ruminants represents both an economical and an environmental strategy for reducing waste management by industries and the cost of feeding as well as the impact of livestock farming. Large amounts of wastes from the cocoa industry are annually produced with a considerable part represented by cocoa bean shells, considered a suitable ingredient to be included in the diet of ruminants within the limits established by European legislation. The aim of this study was to assess the effect of including cocoa bean shells in the diet of dairy sheep on the sensory, volatile, and antioxidant properties of cheese. To this purpose, 20 Comisana lactating ewes were randomly assigned to 2 experimental groups: control (CTRL) and cocoa bean shells (CBS), and received alfalfa hay ad libitum and 800g of conventional (CTRL) or experimental (CBS) concentrate containing 11.7% CBS to partially replace corn and barley of the CTRL concentrate. Bulk milk collected from each group was used to produce a total of 15 cheeses per group, obtained in 5 different days of cheese-making (3 cheeses a day per group). After 60 d of aging, each cheese of each experimental group was sampled for the analyses. The results on chemical composition revealed a greater monounsaturated fatty acids content and an increase in the nutritional indices suggesting a favorable role of cocoa bean shells dietary inclusion on the nutritive value of the cheese. The cheese sensory profile was affected by the cocoa bean shells inclusion, with more pronounced appearance, odor, aroma, and taste attributes in the product. The volatile profile showed only a few significant differences, mainly related to the cheese ripening process, and no differences were found in α-tocopherol contents in cheese fat between the 2 groups. Therefore, the coca bean shells inclusion in the diet of dairy sheep allowed to obtain a good quality cheese, without altering the characteristics associated with the typical profiles of sheep cheese. Furthermore, the use of this by-product could contribute to decrease feed costs and waste management, representing a good practice for increasing the sustainability of dairy products.

High Dietary Folic Acid Supplementation Reduced the Composition of Fatty Acids and Amino Acids in Fortified Eggs.

The study aimed to evaluate the effects of dietary folic acid (FA) on the production performance of laying hens, egg quality, and the nutritional differences between eggs fortified with FA and ordinary eggs. A total of 288 26-week-old Hy-Line Brown laying hens (initial body weights 1.65 ± 0.10 kg) with a similar weight and genetic background were used. A completely randomized design divided the birds into a control group and three treatment groups. Each group consisted of six replicates, with twelve chickens per replicate. Initially, all birds were fed a basal diet for 1 week. Subsequently, they were fed a basal diet supplemented with 0, 5, 10, or 15 mg/kg FA in a premix for a duration of 6 weeks. Supplementation of FA could significantly (p < 0.05) enhance the FA content in egg yolks, particularly when 10 mg/kg was used, as it had the most effective enrichment effect. Compared to the control group, the Glu content in the 10 and 15 mg/kg FA groups showed a significant (p < 0.05) decrease. Additionally, the contents of Asp, Ile, Tyr, Phe, Cys, and Met in the 15 mg/kg FA group were significantly (p < 0.05) lower compared to the other groups. Adding FA did not have significant effects on the levels of vitamin A and vitamin E in egg yolk, but the vitamin D content in the 5 and 10 mg/kg FA groups showed a significant (p < 0.05) increase. Furthermore, the addition of FA did not have a significant effect on the levels of Cu, Fe, Mn, Se, and Zn in egg yolk. The dietary FA did not have a significant effect on the total saturated fatty acids (SFA) and polyunsaturated fatty acid (PUFA) content in egg yolk. However, the total monounsaturated fatty acid (MUFA) content in the 5 and 10 mg/kg groups significantly (p < 0.05) increased. These changes in nutritional content might be attributed to the increased very low-density lipoprotein (VLDL) protein content. The significant decrease in solute carrier family 1 Member 1 (SLC1A1), solute carrier family 1 Member 2 (SLC1A2), and solute carrier family 1 Member 3 (SLC1A3) gene expression compared to the control group appeared to be the reason for the decrease in amino acid content in egg yolk within the dietary FA group. The findings suggest that the appropriate addition of FA can enhance the levels of MUFA and vitamin D in egg yolks, thereby improving their nutritional value. Excessive intake of FA can decrease the effectiveness of enriching FA in egg yolk and impact the enrichment of certain amino acids. The yolk of eggs produced by adding 10 mg/kg of FA to the feed contains the optimal amount of nutrients. This study informs consumers purchasing FA-fortified eggs.

Artificial neural networks optimize the establishment of a Brazilian germplasm core collection of winter squash (Cucurbita moschata D.)

With widespread cultivation, Cucurbita moschata stands out for the carotenoid content of its fruits such as β and α-carotene, components with pronounced provitamin A function and antioxidant activity. C. moschata seed oil has a high monounsaturated fatty acid content and vitamin E, constituting a lipid source of high chemical–nutritional quality. The present study evaluates the agronomic and chemical–nutritional aspects of 91 accessions of C. moschata kept at the BGH-UFV and propose the establishment of a core collection based on multivariate approaches and on the implementation of Artificial Neural Networks (ANNs). ANNs was more efficient in identifying similarity patterns and in organizing the distance between the genotypes in the groups. The averages and variances of traits in the CC formed using a 15% sampling of accessions, were closer to those of the complete collection, particularly for accumulated degree days for flowering, the mass of seeds per fruit, and seed and oil productivity. Establishing the 15% CC, based on the broad characterization of this germplasm, will be crucial to optimize the evaluation and use of promising accessions from this collection in C. moschata breeding programs, especially for traits of high chemical–nutritional importance such as the carotenoid content and the fatty acid profile.

Relationships of quality indices with wooden breast myopathy severity in chicken breast meat under refrigerated storage

ABSTRACT 1. This study investigated the relationships of quality indices with the severity of wooden breast (WB) myopathy in chicken breast meat under refrigerated storage. The physicochemical properties, water-holding capacity (WHC), microbial quality and fatty acid profiles of normal chicken breast meat samples (NOR samples, n = 63), moderate WB (MWB, n = 63) myopathy and severe WB (SWB, n = 63) myopathy (MWB and SWB samples, respectively) were evaluated immediately after sampling and after 4 and 8 d of refrigerated storage at 4°C. 2. Total collagen, fat, saturated and monounsaturated fatty acid contents, redness and pH of the SWB and MWB samples were higher than the NOR samples. The SWB samples that were stored for 8 d had poor WHC, total viable counts (TVC) of higher than 7.0log colony-forming units, total volatile basic nitrogen (TVB-N) content of greater than 15 mg/100 g and a thiobarbituric acid – reactive substance level of higher than 1 mg/kg malondialdehyde. 3. No significant difference was observed in the TVB-N content and TVC of the MWB and NOR samples during storage. Polyunsaturated fatty acid content was lower in the SWB and MWB samples than in the NOR samples. The SWB samples were tougher than the MWB and NOR samples after 8 d of refrigeration. 4. In conclusion, the quality of chicken breast meat with SWB myopathy degraded considerably over time; thus, such meat should not be subjected to extended refrigeration for storage.

Effect of Olive Oil Hydrogel as a Fat Replacer in Beef Meatballs.

Meat and meat products are essential sources of dietary saturated fatty acids. However, excessive consumption of meat and meat products may be harmful to human health. The study evaluates the effect of fat replacement with hydrogels (olive oil in water emulsions gelled by gelatine) in meatballs. The effect of replacing fat with different ratios of hydrogel (control, 25 (F25), 50 (F50), 75 (F75) and 100 % (F100)) on the chemical (fatty acids and thiobarbituric acid reactive substances (TBARS)) and physical (cooking loss, diameter reduction, fat retention, water retention, colour and texture analysis) characteristics of the meatballs were analyzed. The fat content of raw meatball samples was reduced from (31.2±2.2) to (10.5±0.4) % in the sample with the highest fat substitution (F100). The energy levels of the F100 samples were almost 56 % lower than of the control group. Monounsaturated fatty acids (MUFAs) represented the dominant group in all substitution rates of the meatballs, followed by saturated fatty acids (SFAs) and finally polyunsaturated fatty acids (PUFAs). Among the raw meatball samples, the highest oxidation occurred in the F50 and F100 groups. However, it was determined that the difference between F25 and F75 and the difference between control and F75 were not statistically significant (p˃0.05). When the cooked samples were compared, the highest thiobarbituric acid (TBA) value was found in the F50 sample, followed by the F100 and F75 samples. The difference between the mean values of springiness and cohesiveness of the samples was not significant (p˃0.05). The hardness value of samples decreased significantly (p˂0.001) with >75 % fat replacement. It can be concluded that the oil replacement rate that may satisfy consumer demand without impairing the product technological and chemical quality should be <75 %. As the fat replacement ratio increases, the SFA content of cooked meatballs decreases, while the MUFA and PUFA contents increase. Considering the positive effects of reducing the intake of SFAs and increasing the intake of unsaturated fatty acids on non-communicable diseases such as cardiovascular diseases, fat replacement in meatballs is important for future developments.

Dynamic changes of rumen microbiota and serum metabolome revealed increases in meat quality and growth performances of sheep fed bio-fermented rice straw

BackgroundProviding high-quality roughage is crucial for improvement of ruminant production because it is an essential component of their feed. Our previous study showed that feeding bio-fermented rice straw (BF) improved the feed intake and weight gain of sheep. However, it remains unclear why feeding BF to sheep increased their feed intake and weight gain. Therefore, the purposes of this research were to investigate how the rumen microbiota and serum metabolome are dynamically changing after feeding BF, as well as how their changes influence the feed intake, digestibility, nutrient transport, meat quality and growth performances of sheep. Twelve growing Hu sheep were allocated into 3 groups: alfalfa hay fed group (AH: positive control), rice straw fed group (RS: negative control) and BF fed group (BF: treatment). Samples of rumen content, blood, rumen epithelium, muscle, feed offered and refusals were collected for the subsequent analysis.ResultsFeeding BF changed the microbial community and rumen fermentation, particularly increasing (P < 0.05) relative abundance of Prevotella and propionate production, and decreasing (P < 0.05) enteric methane yield. The histomorphology (height, width, area and thickness) of rumen papillae and gene expression for carbohydrate transport (MCT1), tight junction (claudin-1, claudin-4), and cell proliferation (CDK4, Cyclin A2, Cyclin E1) were improved (P < 0.05) in sheep fed BF. Additionally, serum metabolome was also dynamically changed, which led to up-regulating (P < 0.05) the primary bile acid biosynthesis and biosynthesis of unsaturated fatty acid in sheep fed BF. As a result, the higher (P < 0.05) feed intake, digestibility, growth rate, feed efficiency, meat quality and mono-unsaturated fatty acid concentration in muscle, and the lower (P < 0.05) feed cost per kg of live weight were achieved by feeding BF.ConclusionsFeeding BF improved the growth performances and meat quality of sheep and reduced their feed cost. Therefore, bio-fermentation of rice straw could be an innovative way for improving ruminant production with minimizing production costs.Graphical

Disordered endogenous synthesis as well as changes in the quantity and qualitative composition of fatty acids in the diet can become pathogenetic factors in a number of metabolic diseases. The purpose of this article was a comparative analysis of the content of monounsaturated fatty acids (palmitoleic, elaidic, oleic, gondoic, erucic and nervonic) in the peripheral blood of residents of different climatic regions. Materials and methods. The research was conducted between 2008 and 2018 and involved 697 subjects aged 22 to 60 years. Depending on the region of permanent residence, all the participants were divided into three groups: 1) residents of the Arctic region (AR); 2) people living in the circumpolar region (CR); 3) residents of the South Caucasus (SC). The level of monounsaturated fatty acids (MUFAs) in the blood serum was evaluated using gas-liquid chromatography. Results. The analysis of the data obtained demonstrates that the total concentration of MUFAs in the peripheral blood of AR and CR residents is statistically significantly lower compared to the people of SC. In particular, northerners showed lower concentrations of palmitoleic, oleic and gondoic acids. Moreover, in the AR group, the level of elaidic acid was higher than in the SC group, while the concentration of erucic acid was lower than in the CR and SC groups. In terms of nervonic acid, a statistically significantly lower content was detected in SC residents compared to the CR group, while the probability of differences between SC and AR residents was at the trend level. Thus, extreme factors of the North cause adaptive rearrangements in fat metabolism at the level of major and minor fatty acid components. At the same time, the changes are closely linked with the structure of nutrition and can be attributed to an unbalanced diet.

Analysis of contents of different kinds of fatty acids in carotid atherosclerotic plaques

Objective: To analyze the contents of different kinds of fatty acids in carotid atherosclerotic plaques. Methods: A total of 24 patients who underwent carotid endarterectomy at the Second Affiliated Hospital of Naval Medical University from October 2021 to September 2022 due to moderate and severe carotid artery stenosis were retrospectively enrolled, including 20 males and 4 females, with a median age[M(Q1, Q3)] of 68.5 (63.5, 72.3) years. According to the symptoms of cerebral ischemia, the patients were divided into a symptomatic group (12 cases) and an asymptomatic group (12 cases). Regarding the pathological characteristics, the patients were divided into a stable group (14 cases) and a vulnerable group (10 cases) according to carotid plaque pathology scores. The expression differences of different types of fatty acids in carotid plaques were analyzed by targeted fatty acid metabolomics technology based on ultra-performance liquid chromatography-mass spectrometry (UPLC-ESI-MS/MS) analysis. Results: In the 24 samples, the median amount of fatty acids [M (Q1, Q3)] was 1 113 (330, 5 897) ng/g. A total of 13 medium and long-chain fatty acids were detected, including saturated fatty acids, monounsaturated fatty acids and polyunsaturated fatty acids. The content of saturated fatty acids was 584 (290, 9 888) ng/g, accounting for the highest proportion of 51.8%. The content of polyunsaturated fatty acids was 1 444 (393, 4 264) ng/g, accounting for 44.4%. The content of monounsaturated fatty acids was 2 793 (1 558, 3 247) ng/g, accounting for 3.8%. The contents of linoleic acid, α-linolenic acid and oleic acid in carotid plaques in the symptomatic group were 1 760 (581, 3 006), 682 (527, 886) and 2 081 (1 358, 2 907) ng/g, respectively, which were lower than those in the asymptomatic group 3 149 (2 226, 4 683), 1 423 (964, 2 270) and 3 178 (2 352, 3 993) ng/g (all P<0.05). The contents of linoleic acid, α-linolenic acid and oleic acid in carotid plaques in the vulnerable group were 1 537 (588, 2 921), 649 (477, 850) and 2 081 (1 129, 2 831) ng/g, respectively, which were lower than those in the stable group 3 149 (2 047, 4 416), 1 423 (940, 2 184) and 3 091 (2 201, 3 973) ng/g (all P<0.05). There were no significant differences in the contents of 11, 14-eicosadienoic acid, γ-linolenic acid, eicosapentaenoic acid, arachidonic acid, erucic acid, margaric acid, pentadecanoic acid, stearic acid, dodecanoic acid and palmitic acid (all P>0.05). Conclusions: Saturated fatty acids are the main type in carotid plaques. The contents of oleic acid, α-linolenic acid and linoleic acid decrease in vulnerable plaques.

Effects of estradiol on fatty acid composition and tissue structure of hepatopancreas in Procambarus clarkii

The hepatopancreas of Procambarus clarkii provides lipids and other nutrients for ovarian development and contains hormones such as estradiol related to ovarian development. During the development of the P. clarkii ovary, the hepatopancreas’ tissue structure and fatty acid content undergo corresponding changes. A controlled indoor experiment was conducted to investigate the effects of exogenous estradiol injection on the tissue structure and fatty acid content in the hepatopancreas of P. clarkii. The experiment consists of three experimental groups and one control group. The P. clarkii in the experimental group were injected with 5, 0.5, and 0.05 μg/(g body weight) of estradiol once every 5 days for 5 times and with normal saline injection as the control. After 25 days, the content of fatty acids and changes in tissue structure in the hepatopancreas were measured. The results showed that the main fatty acids in P. clarkii hepatopancreas were C18:4n, C20:3n, and EPA, and polyunsaturated fatty acid (PUFA) content was 57.17%±4.76%. Injecting estradiol mainly affected the content of C16:0, C16:1n, C18:0, C18:1n9, C18:2n, C18:3n6, C18:3n6, C18:4n, C20:1n, C20:2n, C20:3n, C20:4n3, C20:5n3, C22:n (n=1-4), C22:5n, and C22:6n in the hepatopancreas. Injecting estradiol significantly reduced the total content of PUFA in hepatopancreas with the reduction of C18:3n6, C18:4n, C20:3n, C20:4n3, C20:5n3 C22: n (n=1-4) (P&lt;0.05); Estradiol injection significantly increased the total content of monounsaturated fatty acid (MUFA) in hepatopancreas with the increase of C18:1n9, and C20:1n (P&lt;0.05); Injection of estradiol (0.05 μg/g) significantly increased the total content of saturated fatty acids (SFA) in the hepatopancreas (P&lt;0.05) with the increase of C16:0 and C18:0 (P&lt;0.05). Injecting estradiol can increase the volume and quantity of hepatopancreatic B cells, and the 5 μg/g group had the most obvious changes.

Conjugated Linoleic Acid Reduces Lipid Accumulation via Down-regulation Expression of Lipogenic Genes and Up-regulation of Apoptotic Genes in Grass Carp (Ctenopharyngodon idella) Adipocyte In Vitro.

The relationship between conjugated linoleic acid (CLA) and lipogenesis has been extensively studied in mammals and some cell lines, but it is relatively rare in fish, and the potential mechanism of action of CLA reducing fat mass remains unclear. The established primary culture model for studying lipogenesis in grass carp (Ctenopharyngodon idella) preadipocytes was used in the present study, and the objective was to explore the effects of CLA on intracellular lipid and TG content, fatty acid composition, and mRNA levels of adipogenesis transcription factors, lipase, and apoptosis genes in grass carp adipocytes in vitro. The results showed that CLA reduced the size of adipocyte and lipid droplet and decreased the content of intracellular lipid and TG, which was accompanied by a significant down-regulation of mRNA abundance in transcriptional regulators including peroxisome proliferator-activated receptor (PPAR) γ, CCAAT/enhancer-binding protein (C/EBP) α, sterol regulatory element-binding protein (SREBP) 1c, lipase genes including fatty acid synthase (FAS), acetyl-CoA carboxylase (ACC), lipoprotein lipase (LPL). Meanwhile, it decreased the content of saturated fatty acids (SFAs) and n - 6 polyunsaturated fatty acid (n-6 PUFA) and increased the content of monounsaturated fatty acid (MUFA) and n - 3 polyunsaturated fatty acid (n-3 PUFA) in primary grass carp adipocyte. In addition, CLA induced adipocyte apoptosis through downregulated anti-apoptotic gene B-cell CLL/lymphoma 2 (Bcl-2) mRNA level and up-regulated pro-apoptotic genes tumor necrosis factor-α (TNF-α), Bcl-2-associated X protein (Bax), Caspase-3, and Caspase-9 mRNA level in a dose-dependent manner. These findings suggest that CLA can act on grass carp adipocytes through various pathways, including decreasing adipocyte size, altering fatty acid composition, inhibiting adipocyte differentiation, promoting adipocyte apoptosis, and ultimately decreasing lipid accumulation.

STK11 (LKB1) mutation suppresses ferroptosis in lung adenocarcinoma by facilitating monounsaturated fatty acid synthesis.

Serine/threonine kinase 11 (STK11), a tumor suppressor gene, exhibits frequent mutations in lung adenocarcinoma (LUAD). However, the specific molecular mechanisms by which STK11 mutations exert an influence on the biosynthesis of monounsaturated fatty acids (MUFAs) and subsequently affect ferroptosis in LUAD remain indistinct. In this study, bioinformatic analysis was employed to probe into the linkage between STK11 and key inhibitory genes of ferroptosis, namely SLC7A11 and SCD1, in LUAD tissues. Quantitative reverse transcription polymerase chain reaction was employed to assess the expression of STK11 in both wild-type and mutant STK11 LUAD cells, cell counting kit-8 to assess cell viability, and flow cytometry to detect apoptosis. A transmission electron microscope was utilized to observe mitochondrial morphology, and Western blot to ascertain the protein expression of STK11, ferroptosis-related proteins, and the enzyme SCD1 involved in MUFA synthesis. Oil red O staining was employed to test the distribution of lipid droplets in cancer cells, and a lipid quantification method to measure the content of MUFAs. Commercial kits were employed to assess the levels of lipid reactive oxygen species, malondialdehyde, glutathione, and Fe2+ in cells. The result revealed a negative correlation between STK11 and SLC7A11 as well as SCD1, with STK11 expression downregulated in mutant STK11 LUAD cells. Furthermore, STK11 mutations were found to suppress ferroptosis in LUAD cells by affecting MUFA synthesis. Subsequent rescue assays demonstrated that STK11 mutations hindered ferroptosis by impacting the synthesis of MUFAs in LUAD cells. This study provided evidence that STK11 mutations suppressed ferroptosis in LUAD cells by promoting MUFA synthesis, thus offering a novel research direction in the management of LUAD.

Minerals and fatty acid profile of small indigenous fish species from homestead ponds within a Sub-tropical coastal region

Malnutrition has emerged as a noticeable obstruction to the socio-economic advancement of rural areas along the coastal regions of Bangladesh. Small indigenous fish species (SIS) have the potential to alleviate the malnutrition issue because of having higher nutritional compositions. However, prior research has overlooked the detailed nutritional value of SIS originating from coastal regions. Consequently, the current investigation sought to analyze the mineral and fatty acid composition of twelve SIS obtained from coastal homestead ponds. The findings indicated that the mineral composition in SIS exhibited the following descending order: calcium (Ca) > phosphorus (P) > potassium (K) > magnesium (Mg) > iron (Fe) > zinc (Zn). Furthermore, when considering the overall mineral content in SIS, it ranked in the following decreasing order: A. testudineus, M. tengara, C. punctatus, N. nandus, P. sophore, C. fasciatus, A. mola, C. batrachus, H. fossilis, P. sarana, M. aculeatus, and O. pabda. The analysis of the fatty acid profile further revealed that SIS is a rich source of palmitic acid, linoleic acid, oleic acid, stearic acid, myristic acid, palmitoleic acid, and linolenic acid. The saturated fatty acid content of the SIS varied between 42.66 % and 63.37 %, and the highest content was found in A. mola whereas the lowest was in A. testudineus. On the other hand, the total monounsaturated fatty acid content of the SIS ranged from 26.49 % (A. mola) to 46.12 % (P. sarana), and the total PUFAs contents among the fish species ranged from 5.7 % (A. mola) to 16.54 % (H. fossilis). Therefore, SIS could be a key source of minerals and essential fatty acids for human well-being. This can help fulfill nutrient requirements and reduce malnutrition among coastal populations. It can be said that, if these fishes are introduced in the culture systems, it will be used for consumption as well as support the livelihood of coastal people.

HEMPSEED CAKE IN RABBIT NUTRITION: LIVESTOCK PERFORMANCES, QUALITY OF MEAT, DIGESTIBILITY OF NUTRIENTS AND ANIMAL HEALTH STATUS

The objective of this study was to evaluate the effect of dried hempseed cake (by-product of oil production) supplementation (5 % − EG1 and 10 % − EG2) to rabbit feed mixture on livestock performances, quality of meat, digestibility of nutrients and animal health status. The chemical composition of the Longissimus thoracis and lumborum muscle including the content of fatty acids and amino acids, and the biochemical parameters in caecum and blood of growing rabbits were investigated. No significant differences among the experimental groups in feed intake, body weight and carcass parameters were found. The digestibility trial was performed using the balance method. Feed mixtures differed in digestible energy content i.e., crude protein, crude fibre and fat. The resulting digestibility coefficients for protein fell within the range of 66.72 − 74.18 %, fat digestibility was in the interval from 88.73 to 89.34 % and crude fibre digestibility was in the range from 27.56 to 34.59 %. Fatty acid profile in intramuscular lipids represents the highest content of monounsaturated fatty acids (MUFA; 47.72 − 48.51 %), followed by the content of saturated acids (SFA; 33.75 − 33.87 %) and a low content of polyunsaturated fatty acids (PUFA; 11.23 − 12.08 %). Hempseed oil cake could be included in rabbits' diet with beneficial effect on carcass quality and can enhance the nutritional quality of rabbit meat with the focus on essential amino acids. The tested blood serum parameters were within the range of the physiological reference values with no statistical differences between experimental groups, except for glucose and cholesterol content. The data on volatile fatty acids (VFA) show that most intensive process was in the caecum of rabbits in the experimental groups EG1 and EG2. All obtained data let us to recommend the inclusion of hempseed cake up to 10 % in rabbit diet without any negative effect on animal welfare, livestock performance and quality of meat.

Cold pressed cooking oils: An enigma of choice based on Omega-6 to Omega-3 ratio

A range of cold pressed edible oils which are commonly used in households of India were decoded with gas chromatography for their fatty acid composition and n-6/n-3 ratios. Among the nine cold pressed oils tested coconut oil contained highest amount of saturated fat. Safflower and Mustard had about similar amount of unsaturated fatty acids, however the content of monounsaturated fatty acids was high in case of mustard oil (59.16%) and poly unsaturated fatty acids were high in safflower oil (73.61 %). Every oil examined, with the exception of mustard oil, has much more omega 6 than omega 3 fatty acids. The ratio of n-6 to n-3 fatty acids was significantly higher in other oils and more favourable in case of mustard oil.

Exploitation of inland salt lake water by dilution and nutrient enrichment to cultivate Vischeria sp. WL1 (Eustigmatophyceae) for biomass and oil production

Salt lakes are significant components of global inland waters. Salt lake (SL) water can provide precious mineral resource for microbial growth. The prospect of utilizing diluted SL water for cultivation of a terrestrial oil-producing microalga Vischeria sp. WL1 was evaluated under laboratory conditions. Based on the detected mineral element composition, the water from Gouchi Salt Lake was diluted 2, 4, 6 and 8 folds and used with supplementation of additional nitrogen, phosphorus and iron (SL+ water). It was found that 4 folds diluted SL+ water was most favorable for biomass and oil production. When cultivated in this condition, Vischeria sp. WL1 gained a biomass yield of 0.82 g L−1 and an oil yield of 0.56 g L−1 after 24 days of cultivation, which is comparable to the optimum productivity we previously established. In addition, total monounsaturated fatty acid contents (64.4∼68.1 %) of the oils resulted from cultures in diluted SL+waters were higher than that in the control (55.5 %). It was also noteworthy that in all these cultures the oil contents (652.0∼681.0 mg g−1) accounted for the most of the biomass, which are far more than the protein and starch contents. This study demonstrates the feasibility of using SL water as a cost-effective mineral resource to cultivate microalgae for biomass and oil production.
