Determination of phthalate esters contents in different types of cheese produced and consumed in Saudi Arabia

Background: Phthalates’ esters are commonly incorporated into plastic materials to enhance their elasticity, thereby facilitating their migration into food items. Aims: To evaluate the levels of phthalate esters in six distinct varieties of locally produced cheese and sold in Saudi Arabia, at the time of purchase (t=0) and after storage for one and two months. Methods: Six types of local cheeses (Cheddar, Mozzarella, Feta, Cheeseburger slices, Haloumi and Kashkaval) were purchased and analyzed at three different intervals: at the time of purchase (t=0), after one month, and after two months of storage. The Phthalate ester concentrations were determined using indirect gas chromatography with results expressed in mg/kg of cheese. Results: Among the cheese samples analyzed, di(2-ethylhexyl) phthalate (DEHP) was found to be the most prevalent. Halloumi cheese exhibited the highest average concentration of DEHP (4.29 ± 0.51 mg/kg) followed by Mozzarella cheese (3.65 ± 0.46 mg/kg). After two months of storage, more than two-thirds (n=13) of the samples exceeded the permissible limits content of DEHP content while one-third (n=6) and more than half (n=9) exceeded the limits for dibutyl phthalate (DBP) and di-n-octyl phthalate (DNOP), respectively. The levels of phthalates in all cheese types increased with prolonged storage duration with Cheddar cheese exhibiting the highest rate of increase, exceeding fivefold (from 0.34 to 2.29 mg/kg). Conclusion: This study confirms the presence of phthalate in cheese samples, with concentrations varying depending on the cheese type, specific phthalate compound, and storage duration. Heightened consumer awareness regarding cheese storage practices is imperative to mitigate potential health risks associated with phthalate consumption. Accumulation and Transport of Phthalic Acid Esters in the Soil-Plant System of Agricultural Fields with Different Years of Film Mulching

In this study, we investigated the accumulation and transport patterns of phthalate ester (PAEs) compounds in the soil-plant system of maize and cabbage fields under different film mulching years. The total content of five PAEs in the maize fields soil ranged from 156.19 to 566.1 μg kg−1 with film mulching for 0–20 years. The total content of five PAEs in the cabbage field soils ranged from 252.48 to 559.07 μg kg−1 with film mulching for 0–30 years. The PAEs content in cabbage and maize field soils was lower than the environmental quality standards for soil (GB 15618-2008). With the increase in film mulching years, both maize and cabbage soil PAEs contents increased significantly (p < 0.05), while among the five PAEs, the contents of di-n-butyl phthalate (DnBP) and di(2-ethylhexyl) phthalate (DEHP) increased the most significantly, and the content of DEHP in the soils was the highest. The DEHP content in soils planted with maize ranges from 134.03 to 406.79 μg kg−1 with the film mulching for 20 years; while in soil planted with cabbage, it ranges from 229.35 to 405.2 μg kg−1 with the film mulching for 30 years. The DEHP content in all of the soils has not exceed the limit value established by the USEPA in U.S. Five PAEs were detected in maize seeds and cabbage leaves, among which the DEHP content exceeded the maximum residue limit set by the National Food Safety Standard (GB 9685-2016) for DEHP in food after more than 10 years of mulching. The bioaccumulation and translocation factors in cabbage showed a significant positive correlation with the film mulching years. However, the bioaccumulation factor and translocation factor in maize have weak correlations with the film mulching years, possibly due to the lower PAEs content in the deep soil layers where maize roots are distributed. Our research revealed a significant association between the presence and distribution of PAEs in crops and mulching years. These novel discoveries provide fundamental information for controlling PAEs pollution in agricultural environments.

Effect of particle size and environmental conditions on the release of di(2-ethylhexyl) phthalate from microplastics

The extensive use and improper handling of plastics have caused extensive microplastic (MP) pollution in terrestrial environments. Di(2-ethylhexyl) phthalate (DEHP), the main additive used in plastics, is toxic to organisms and may pose risks to human and animal reproductive functions. However, research on the release behavior of DEHP from MPs is scarce. In this study, the effects of particle size and environmental conditions (temperature, pH, ionic strength, and cation type) on DEHP release from polylactide (PLA), polystyrene (PS), and polyvinyl chloride (PVC) MPs were determined by performing leaching experiments. The results showed that when particle size decreased, the content of DEHP in the MPs and the amount of released DEHP increased though increasing specific surface area. An increase in temperature also promoted DEHP release; when the temperature increased from 15 °C to 45 °C, the amount of DEHP released from PLA, PS, and PVC increased by 38.4%, 71.0%, and 109%, respectively. The lower the crystallinity, the greater the increase in the amount of DEHP released. Ionic strength inhibited the release of DEHP from MPs. When Na+ concentration increased from 0 to 200 mM, the amount of DEHP released from PLA, PS, and PVC decreased by 27.4%, 41.6%, and 35.3%, respectively. The effect of Ca2+ on DEHP release from MPs was greater than that of Na+. In addition, the process of DEHP release from MPs fit well with a pseudo-first-order kinetic model. The results of this study provide a theoretical basis for managing and controlling the risks associated with plastic wastes.

Distribution of Micro-Nano PS, DEHP, and/or MEHP in Mice and Nerve Cell Models In Vitro after Exposure to Micro-Nano PS and DEHP.

Polystyrene (PS) and di-(2-ethylhexyl) phthalate (DEHP) exist widely in the environment. However, their distribution in organisms remains unclear. We used three sizes (50 nm, 500 nm, and 5 μm) of PS and DEHP to study the distribution and accumulation of PS, DEHP, and mono(2-ethylhexyl) phthalate (MEHP) in mice and nerve cell models (HT22 and BV2 cells) and their potential toxicity. Results showed that PS entered the blood of mice, and the distribution of different particle sizes in different tissues was different. After the combined exposure to PS and DEHP, PS carried DEHP, which significantly increased the DEHP content and MEHP content and the highest content of MEHP was in the brain. With the decrease in PS particle size, the contents of PS, DEHP, and MEHP in the body increased. The levels of inflammatory factors were increased in the serum of the PS or/and DEHP group. In addition, 50 nm polystyrene can carry MEHP into nerve cells. These results suggest for the first time that PS and DEHP combined exposure can induce systemic inflammation, and the brain is an important target organ of PS and DEHP combined exposure. This study may serve as a reference for further evaluation of the neurotoxicity induced by combined exposure to PS and DEHP.

Determination and Traceability Analysis of Phthalic Acid Esters in Garlic (Allium stivum L.) from Jiangsu Province, China

Phthalic acid esters (PAEs) are ubiquitous environmental pollutants and are recognized as a threat to the environment and agricultural product safety across the world. In order to investigate the level of PAEs in garlic, soils, and agricultural films from Pizhou City, Jiangsu province, China, 11 garlic samples, 106 soil samples, and 4 agricultural film samples were collected and analyzed using GC-MS. In addition, the uptake and transport characteristics of six PAEs compounds classified as priority pollutants by the United States Environmental Protection Agency (EPA) in the garlic cultivar Daqingke were investigated under hydroponic conditions. The results indicated that dibutyl phthalate (DBP) and di-(2-ethylhexyl) phthalate (DEHP) were the dominant PAEs species in garlic cloves of the different garlic varieties from Pizhou City. The average contents of DBP and DEHP in garlic cloves were 0.611 mg·kg-1 and 0.167 mg·kg-1, respectively, which were significantly higher than those of the commercial varieties of garlic. The concentrations of DBP and DEHP differed in three tissues of garlic bulbs, ordered as the skin of garlic bulb>skin of garlic clove>garlic clove. Dimethyl phthalate (DMP), diethyl phthalate (DEP), diisobutyl phthalate (DIBP), DBP, and DEHP were the main PAEs species and were detected in all the surface soils collected from Pizhou City. Compared with the soil allowable concentrations of the six PAEs in the United States, the DMP and DBP concentrations in approximately 100% and 63.2% of soil samples exceeded the recommended allowable concentrations set by the EPA. However, the levels of DEP, DIBP, and DEHP in the soils were below the maximum allowable concentrations set by the EPA. Nevertheless, the average content of DEHP in soils was 486 μg·kg-1 and was found to be much higher than that in the other four PAEs. Six PAEs, including DMP, DEP, DIBP, DBP, butyl benzyl phthalate (BBP), and DEHP, were detected in all the agricultural film samples. Among them, the contents of DBP and DEHP in the agricultural films were the highest, accounting for 53.7%-63.2% of the total PAEs. The amount of PAEs present in the residual film was significantly lower than that in the new film, and all six PAEs were detected in garlic or soil samples, suggesting that agricultural film can be an important source of PAEs in garlic farming soils and garlic. Furthermore, the garlic plants absorbed DMP and DEP efficiently from the substrate and showed higher translocation factors (TFs) for DMP and DEP than those for DBP, BBP, DEHP, and di-n-octyl phthalate (DnOP), resulting in a higher accumulation of DMP and DEP in the over-ground parts of garlic. In contrast, DBP and BBP in roots of garlic displayed higher bioconcentration factors (57.4 and 81.5, respectively) compared to those of the other four PAEs, whereas the TFs of DBP and BBP were lower; this may have contributed to the high accumulation of DBP in garlic bulbs. The BCFs and TFs of DEHP and DnOP in garlic were relatively lower, but the DEHP had been detected in all garlic cloves, which may be a result of the higher DEHP contents in soils.

EFFECT OF SOUS VIDE HEAT TREATMENT ON PHTHALIC ACID ESTERS CONTENT IN MEAT

The chemical composition and content of DBP (di-n-butyl phthalate) and DEHP (di-2-ethylhexyl phthalate) in pork shoulder before and after heat treatment in the package by the sous vide method was analyzed. The meat was analyzed raw and after heat treatment at 50 °C and 60 °C. The heat treatment time in the sous vide water bath was 4 hours, 4 hours + 1 hour after 24 hours, 8 hours and 8 hours and 1 hour after 24 hours. The fat content in meat treated at 50 °C was 6.04 in raw meat and after heat treatment after 4 hours. 7.51 and after 8 h. 6.81 g.100g-1, in the shoulder after heat treatment at 60 °C was after 4 hours 6.24 and after 8 h. 6.76 g.100g-1. The content of fatty acids during the sous vide treatment did not significantly change with exception of vaccenic acid, the content of which was statistically significantly reduced at temperatures of 50 °C also at 60 °C. The DBP content in raw shoulder at 50 °C during sous vide heat treatment it increased to 1.91 μg.g-1. The DEHP content during the heat treatment it increased to 23.95 μg.g–1 in the treatment of 4+1 hours. The DBP content in raw shoulder after heat treatment at 60 °C increased to 1.84 μg.g–1 during treatment for 4+1 hours. The DEHP content decreased to 8.72 in the treatment of 4+1 hours and to 4.021 μg.g–1. Based on our results, we can conclude that at both monitored temperatures of sous vide method, the DBP content increased and the DEHP content decreased. The DBP content in raw shoulder at 50°C heat treatment increased to 1.91 μg.g-1 at 4+1 hour heat treatment and to 3.02 at 8+1 hour. The DEHP content increased to 23.95 μg.g-1 in the treatment of 4+1 hours. The content of DBP and DEHP in the packaging material before use was 29.08 μg.g-1, it gradually decreased with the length of the heat treatment, to 15.09 μg.g-1 in the treatment of 8+1 hours. The DEHP content in the unused package decreased to1.27 μg.g-1 at heat treatment of 8+1 hours. At the heat treatment at of 60°C in the packaging material gradually decreased to 3.18 μg.g-1. The DEHP content decreased to 2.54 μg.g-1.

Rice-associated with Bacillus sp. DRL1 enhanced remediation of DEHP-contaminated soil and reduced the risk of secondary pollution through promotion of plant growth, degradation of DEHP in soil and modulation of rhizosphere bacterial community

Di (2-ethylhexyl) phthalate (DEHP) attracted much attention since its excessive accumulation in soil and potential endocrine disrupting effects on human. In order to find an efficient approach for remedying DEHP-contaminated soil while reducing crop safety risk, a plant growth-promoting rhizobacteria (PGPR) with DEHP-degrading ability was isolated and identified as Bacillus sp. DRL1. The phytoremediation system that integrated the use of rice and DRL1 into remediation of DEHP-contaminated soil was evaluated. The results showed that DRL1-assisted rice effectively remove 83.9 % DEHP from soil within 30 d. Interestingly, the DEHP content in rice was significantly reduced by 7.7 % compared to non-inoculation treatment, which meant that due to its degradation ability, DRL1 could directly participate in the DEHP degradation in soil. Moreover, the inoculation of DRL1 could alleviate the negative effects of DEHP on rice and then improved its shoot height (31.4 %), root length (31.9 %) and dry weight (24.1 %), which might indirectly improve the phytoremediation efficiency of DEHP-contaminated soil. Above all, this study provided new perspective for the improvement of DEHP removal from soil by a phytoremediation approach while decreasing the secondary pollution risk. The important roles of soil enzyme and rhizosphere bacterial community in this phytoremediation process were also investigated and discussed.

Diethylhexyl phthalate as a current problem of hygienic safety of packaging and packaged drinking water

Introduction. The chemical safety of packaged drinking water is influenced by many factors, including the quality of packaging materials. The chemical components included in the packaging can migrate into drinking water and have a negative impact on human health. The most common types of packaging for drinking water are polyethylene terephthalate (PET, PET) and polycarbonate (PC) bottles. In the production of polymers, to soften and increase the flexibility of the finished product, special substances are added to the material - plasticisers, including dialkyl or alkyl aryl esters of orthophthalic acid (phthalates), among which di (2-Ethylhexyl) phthalate (DEHP), which has toxic properties. The purpose of this study is to analyse various samples of plastic containers in contact with drinking water for the presence of DEHP in polymers and to study the emission of DEHP from polymer packaging into a model environment. Materials and methods. Research has been carried out on 11 samples of containers for drinking water of domestic production, of which nine samples were represented by PET bottles of various sizes and two samples of containers from PC with a volume of 19.0 litres. There was determined the content of DEHP in the container material and the level of migration of DEHP into the aqueous model environment. Sanitary and hygienic studies were carried out based on the Chemical Analytical Center “Arbitrage” of the DI. Mendeleev All-Russian Scientific Research Institute using gas chromatography with mass spectrometric detection (GC-MS). GSO 11366-2019 for the composition of a solution of orthophthalic acid esters in methanol was used as a standard sample for the calibration characteristics; a deuterated analogue of the analyte, DEHP-D4, was chosen as an internal standard. Results. In all studied samples of packaging materials, the residual content of DEHP was in the range of 1.7-4.2 mg/kg of polymer; the levels of DEHP migration into aquatic model media were determined at T = 20 °C and T = 40 °C in the range of (8.6-71.0) μg/dm3. Conclusion. One of the sources of contamination of packaged drinking water with DEHP is PET and PC containers. The results obtained indicate the need to include DEHP in the list of monitored sanitary and hygienic safety indicators in the technical regulations of the EAEU TR 044/2017 and TR CU 005/2011 and to consider the issue of restrictions on its use in the production of food packaging. Limitations of the study. As part of this work, samples of bottles from six domestic container manufacturers were analysed. To study the migration of the target component, a model medium was used - distilled water, which does not imitate the mineral composition of drinking water and has certain pH and electrical conductivity values. Studies of the level of DEHP migration were carried out at an ambient temperature of 20 °C and 40 °C and an exposure time of 30 days. The measurement range of DEHP in polymers and the model medium in this study was (0.1-5) µg/sample. The expanded uncertainty of the results of measurements of the DEHP content in the entire measurement range did not exceed 20%.

Storage lesions in red blood cell-saline adenine glucose mannitol: In-vitro and in-vivo analysis over 42 days and its implications in routine transfusion practice

Background and Objectives: Indian studies on evaluation of storage lesions in red blood cells (RBCs) are either limited to 21 or 28 days or have evaluated limited parameters for 42 days. Moreover, issue of transfusion of “fresh” versus “old” RBC is far from settled. The study serially assesses, up to 42 days, in vitro and in vivo RBC storage lesion parameters, including di-(2-ethylhexyl) phthalate (DEHP) leaching and its comparison with published literature. Methods: The study serially assessed in vitro RBC storage lesion parameters including potassium, lactate, glucose, pH, supernatant hemoglobin, percentage-hemolysis, and DEHP leaching of RBC unit during storage till 42 days. The study also evaluated in vivo recovery of potassium after transfusion of “older” RBC. Results: Serial monitoring of in vitro biochemical parameters showed increase in potassium, lactate, supernatant Hb, and hemolysis% and reduction in glucose and pH. DEHP content of the RBC bag was within no-observed adverse effect limit on days 42. Measurement of serum potassium after transfusion of “older” RBC unit revealed that levels of potassium were within normal limit in all four patients. Sterility testing done on days 42 was negative for all 24 bags. Conclusion: Development of storage lesions is inevitable. Appropriate storage limits the RBC lesions to within normal limits. The increase in potassium, lactate, or hemolysis consequent to aging of blood has little clinical significance in routine transfusion practice.

Does heated erythrocyte suspension transfusion with medical devices containing phthalates increase DEHP and MEHP levels?

It is commonly known that stored blood and blood products are heated before transfusion to prevent hypothermia, which leads to increased di-(2-ethylhexyl) phthalate (DEHP) content leaching into the blood and blood products and thereby causes greater conversion of DEHP to mono (2-ethylhexyl) phthalate (MEHP). However, there has been no study in the literature reporting on the amount of toxic phthalates in blood following the erythrocyte suspension (ES) transfused via warming. In this study, we aimed to investigate the DEHP and MEHP content in blood following the heated ES transfusions administered by DEHP-containing and DEHP-free infusion sets. The study included 30 patients that were randomly divided into two groups with 15 patients each: group I underwent ES transfusion via DEHP-containing infusion sets warmed with blood-fluid warmers, and group II underwent ES transfusion via DEHP-free infusion sets warmed with blood-fluid warmers. DEHP and MEHP levels were measured both before and after transfusion. DEHP-free infusion sets led to no increase in the phthalate content, whereas DEHP-containing infusion sets significantly increased the DEHP and MEHP, where the DEHP level increased almost four times (P=.001). DEHP-containing products lead to toxicity. Therefore, using DEHP-free medical devices may prevent toxicity in patients undergoing ES transfusion.

Phthalate Exposures in the Neonatal Intensive Care Unit.

Background: Di-2-ethylhexyl phthalate (DEHP), a phthalate compound found in medical devices, may cause toxic effects in premature infants. In this study, the objective is to quantify DEHP exposures from various intravenous and respiratory therapy devices, and to use these values to predict typical exposure for an infant in a neonatal unit. Methods: Common IV products used on infants are directed through various types of IV tubing (IVT) and analyzed for DEHP content. DEHP exposure for infants receiving respiratory therapy was determined indirectly through analysis of urine DEHP metabolites. By deriving these values for DEHP we calculated the daily exposure to DEHP from common IV fluids (IVF) and respiratory devices during hospitalization in a neonatal unit. Results: IVF labeled DEHP-positive showed very high concentrations of DEHP, but when passed through IVT, substantial amounts were adsorbed. DEHP was undetectable with all DEHP-negative IVF tests, except when passed through DEHP-positive IVT. The DEHP leached from most respiratory devices was relatively modest, except that detected from bubble CPAP. In 14 very low birthweight infants, the mean DEHP exposure was 182,369 mcg/kg over 81.2 days of the initial hospitalization. Ninety-eight percent of the exposure was from respiratory devices, with bubble CPAP accounting for 95% of the total DEHP exposure in these infants. Conclusions: The DEHP exposure in our neonatal unit can be reduced markedly by avoiding or modifying bubble CPAP equipment and avoiding IV tubing containing DEHP.

Phthalates levels in olive oils and olive pomace oils marketed in Turkey

ABSTRACT Phthalates are used as additives and plasticisers in packaging for personal care and food products. Several investigations reported the harmful impact of phthalates on human health. In this study, different types of olive oils (12 olive oil; 20 extra virgin oil; 4 refined pomace oil) in different packaging materials [polyethylene terephthalate (PET), glass and metal] obtained from local markets in Turkey in 2019, were analysed using GC-MS for the presence of benzyl butyl phthalate (BBP), di(2-ethylhexyl) phthalate (DEHP), diisononyl phthalate (DINP), dibutyl phthalate (DBP), and diisodecyl phthalate (DIDP). The average recoveries of the 5 phthalates in olive oils were 87%-100%, with limits of quantification (LOQs) of 0.09–2.28 mg/kg. DEHP was the abundant phthalate in all olive oil samples ranging from below the LOQ (0.23 mg/kg) to 602 mg/kg. In all analysed samples, the levels of DINP and DIDP were less than their LOQ, thus these phthalates were not detected. The highest DEHP content was found in an olive oil sample containing 602 mg/kg, whilst 5 samples did not contain detectable phthalate esters.

Photosynthetic and antioxidant response of wheat to di(2-ethylhexyl) phthalate (DEHP) contamination in the soil

Di(2-ethylhexyl) phthalate (DEHP) is a commonly used, artificially-synthesized, industrial chemical that can be released into the soil. However, to date, there is no comprehensive study on the effects of DEHP on photosynthesis, induction of reactive oxygen species, and response of the antioxidant defense system in wheat plants growing in DEHP contaminated soil. This study was conducted to address this gap in knowledge. Our results showed that after application of 10, 20, and 40 mg/kg DEHP, photosynthetic parameters, fluorescence parameters, and chlorophyll content of wheat leaves at seedling, jointing, and booting stages decreased, while the intercellular carbon dioxide concentration increased. This indicates that the observed decrease in net photosynthetic rate in wheat leaves was due to a non-stomatal limitation, wherein DEHP seems to have hindered the photoelectron transfer process. Both superoxide anion (O2-) and hydrogen peroxide (H2O2) content increased in the roots, stems, and leaves in plant under DEHP treatment compared with those in the control plants. Antioxidant enzyme activity increased with increasing DEHP stress, except under the 40 mg/kg treatment at the seedling stage. The antioxidant system had a certain protective effect on wheat, but DEHP still caused peroxidation of cell membrane lipids. The extent of DEHP damage to the roots, stems, and leaves was concentration dependent. Furthermore, enzymatic activity tolerance increased with metabolism, and long-term effects of DEHP gradually decreased with plant growth. Finally, the toxic effects of DEHP on root tissues were more serious at the seedling and jointing stages.

Phthalates in meat products in dependence on the fat content

The content of dibutylphthalate (DBP) and di-(2-ethylhexyl) phthalate (DEHP) in samples of packages intended for thermally processed meat products and release of phthalates from packages into meat products in dependence on the fat content were observed. 80 samples of packages were analyzed, 5 of them wereselected due to exceeding the specific migration limit. The raw meat was prepared, one type with the fat content of 10% and second one with the fat content of 50%. The both types of raw meat were analyzed for the content of DBP and DEHP and packed into chosen packages.The samples of meat products were thermally processed (70 ℃, 10 min in the core), stored until the expiration date at 4 °C and gradually analyzed after 1st, 7th, 14th, 21st and 28th day of storage. Determination of phthalates was carried out by high performance liquid chromatography (HPLC) in the Zorbax Eclipse C8column and by UV detection at a wavelength of 224 mm. The phthalate content in the raw meat was under the limit of detection. According to the EU Commission Regulation no. 10/2011 the specific migration limit of products intended for the contact with food for DEHP (max. 1.5 mg.kg-1of food stimulant and DBP max. 0.3 mg.kg-1 of food stimulant), wasexceeded already after first day of storage, in case of DBP in two samples with 10% of fat and after 7th day of storage in one sample. In the samples with 50% of fat, SML was exceeded after first day of storage in four samples and in one sample after 14th day of storage. Regarding DEHP in the samples with 10% of fat SML was exceeded after 1st day of storage in one sample and after 7th day of storage also in one sample and after 21st day of storage similarly in one sample. Four samples with 50% of fat had SML exceeded in case of DEHP already after 1st day of storage. By comparison of PAE migration depending on the fat content we concluded that leaching of PAE from a package into food was 2 - 21 times higher in samples with 50% of fat than in samples with 10% of fat.

There are growing concerns on long-term health consequences, notably on fertility rates, of plasticizers such as phthalates. While di(2-ethylhexyl)phthalate (DEHP) is currently used in several medical devices, newborns in the neonatal intensive care unit are both more exposed and more vulnerable to DEHP. The objectives of this study were to identify, count, and describe possible sources of DEHP in a neonatal care unit. Our method consisted in the listing and the inspection of the information on packaging, complemented by contact with manufacturers when necessary. According to the results, 6% of all products and 10% of plastic products contained some DEHP; 71% of these involved respiratory support devices. A vast majority of the items showed no information on the content of DEHP. Further research is needed, particularly to determine the effects of such an early exposure and to study and develop safer alternatives.

SLEDOVÁNÍ FTALÁTŮ VE TKÁNÍCH KUŘAT V ZÁVISLOSTI NA JEJICH OBSAHU V KRMIVU

For the monitoring of distribution and accumulation of phthalic acid esters (PAE) in animal tissues, samples of muscle, mesenteric fat (fat), skin and liver from broiler chicks ROSS 308 were used. The chicks were divided into 4 groups (50 chicks each). All the chicks were given commercial diets (complete feed, KKS) for broiler chicks (starter – BR1; grower – BR2 and finisher – BR3). The experimental diets were supplemented with vegetable oil (RO) with low (group N) or high (group V) phthalate contents, or animal fat with a high phthalate content (group Z). Neither the control diets (K), nor the grower (BR1) diets contained vegetable oils or animal fat. The N chicks were given the grower (BR2) and finisher (BR3) diets supplemented with 5% and 3% vegetable oil, respectively. The V chicks were given BR2 and BR3 diets with 5% and 3% vegetable oil, respectively. The Z chicks were given BR2 and BR3 diets with 5% and 3% animal fat, respectively. Di-n-butyl phthalate (DBP) and di-(2-ethylhexyl) phthalate (DEHP) were found in the tissues of chicks in all the experimental groups. The DBP content in the muscle ranged from 0.03 to 0.55 mg . kg−1, in the adipose tissue from < 0.20 to 2.56 mg . kg−1, in the skin from < 0.20 to 1.49 mg . kg−1, and in the liver from 0.03 to 0.13 mg . kg−1. The content of DEHP in the muscle ranged from 0.03 to 1.15 mg . kg−1, in the adipose tissue from 0.25 to 9.85 mg . kg−1, in the skin from < 0.20 to 4.68 mg . kg−1, and in the liver from 0.16 to 0.24 mg . kg−1. The highest concentrations of DBP of 1.28 ± 1.00 mg . kg−1 of fresh sample (an average value from 8 chicks) was determined in the adipose tissue of V chicks. The highest concentration of DEHP of 3.27 ± 2.87 mg . kg−1 of fresh sample (mean of 8 chicks) was also determined in the V group. The accumulation of DEHP was 3.2; 2.6 and 2.9 times higher than that of DBP in the muscle, adipose tissue and skin, respectively.

Exposure risk assessment of plasticizer in dietary food in Xiamen

To understand the dietary consumption of residents in Xiamen and the content of phthalic acid esters (PAEs) in food, and to assess the plasticizer exposure risk of diet in Xiamen. The survey was conducted by stratified cluster random sampling method in Xiamen from September to October in 2010. According to the Xiamen administrative division, six neighborhood communities were selected as sampling units, then 25 families were randomly chosen from each sampling units.From the above 150 families, the permanent residents over the age of six were permitted to our study. The survey included 495 residents totally. These participants' information, such as basic personal information, physical activity levels, meal frequency and the average consumption of 33 kinds of food in 13 categories were collected using questionnaires. Thirteen categories included cereal and tubers, beans, vegetables, fungi and algae, fruits, dairy products, meat, seafood, eggs, snacks, beverages, cooking oil and spices. The height and weight of residents were measured and the average daily dietary intake was calculated. Thirty-three kinds of food in 13 categories were collected in supermarkets in Xiamen. According to the annual sales ranking, the top three-five brands of each kinds of food were selected and numbered, then two or three brands were chosen by random number table method from them; three completely individual packed samples in the same batch of each brand were detected; 243 samples were included in our study.100-500 g solid samples or 100-500 ml liquid samples were collected. The content of diethyl phthalate (DEP), dibutyl phthalate (DBP), di (2-ethylhexyl) phthalate (DEHP) in food were detected by liquid chromatography mass spectrometry, which expressed by median (minimum-maximum). The exposure dose, contribution rate and risk index of PAEs were calculated by point estimation method. According to the average daily dietary intake of residents in Xiamen, the top three ones in 13 categories of food were cereal and tubers (337.16 g/d, 18.21%), vegetables (309.12 g/d, 16.69%) and fruits (213.20 g/d, 11.51%). The content of DEP, DBP or DEHP among different categories of food was significantly different (χ² values were 58.05, 50.19 and 102.10, P < 0.01). Among 13 categories of food, seafood contained the most DEP (0.090 (0.000-0.324)mg/kg); cooking oil had the most DBP (0.700(0.000-2.980) mg/kg) and DEHP (5.115(0.000-24.160) mg/kg). DEP, DBP and DEHP exposure(0.19, 4.20, 18.10 µg × kg⁻¹ ×d ⁻¹)in dietary food in Xiamen were less than the reference dose(RfD) (800, 100, 20 µg × kg⁻¹ × d⁻¹) proposed by the United States Environmental Protection Agency (EPA), and the risk indexes were 0.02%, 4.20% and 90.50%, respectively. Among 13 categories of foods, seafood was the main source of DEP dietary exposure. The exposure dose and contribution rate of DEP in seafood were 0.18 µg × kg⁻¹ × d⁻¹ and 94.74%, respectively.Vegetables were the main source of DBP and DEHP dietary exposure. The exposure dose and contribution rate of DBP and DEHP were 1.48 µg × kg⁻¹ × d⁻¹, 35.24% and 6.07 µg × kg⁻¹ × d⁻¹, 33.54%, respectively. The food consumed by residents in Xiamen was overall in a safe state, but to some extent, there still exists DEHP exposure risk in foods.

Plasticizer migration from PVC cable insulation – The challenges of extrapolation methods

A single strand PVC-P insulation including an internal metal conductor removed from the jacketed assemblies of a signal cable showed brittleness after 30 years service at 25 ± 3 °C in air. The PVC compound contained diisodecyl phthalate (DIDP), di(2-ethylhexyl) phthalate (DEHP) and a sizeable fraction of filler. Single strand insulation samples with internal metal conductor were aged in air at elevated temperatures for different periods of time after which the strain at break, the Young's modulus and the plasticizer content were assessed by tensile testing and liquid chromatography. Isothermal evaporation rates from pristine DIDP and DEHP and solutions of the two plasticizers were obtained by thermogravimetry. Data for Young's modulus, strain at break and plasticizer contents were extrapolated to service temperature using two different extrapolation methods, Arrhenius extrapolation (constant activation energy) and a method based on models by Langmuir, Clausius–Clapeyron and Kirchhoff. These methods assume that evaporation of plasticizers to the surrounding gas phase is the dominant deterioration mechanism. Both methods predicted only a minor decrease in plasticizer content after 30 years of ageing at 28 °C and thus a material with adequate mechanical properties. Liquid chromatography showed that the single strand cable samples contained a very low DIDP content (4 wt.%) and an anomalously high DEHP content; a finding that cannot be explained by the expected evaporative loss mechanism. It is suggested that DIDP was efficiently extracted by contact with a DEHP-rich interface at the insulation surface, a process which is active during plant operation, but could not be simulated by controlled laboratory accelerated ageing studies.

Plasticizer Contamination in Edible Vegetable Oil in a U.S. Retail Market

With the wide application of plastics, the contamination of plasticizers migrating from plastic materials in the environment is becoming ubiquitous. The presence of phthalates, the major group of plasticizers, in edible items has gained increasingly more concern due to their endocrine disrupting property. In this study, 15 plasticizers in 21 edible vegetable oils purchased from a U.S. retail market were analyzed using gas chromatograph-mass spectrometry. Di(2-ethylhexyl) phthalate (DEHP) and diisobutyl phthalate (DiBP) were detected in all oil samples. Benzylbutyl phthalate (BzBP), dibutyl phthalate (DBP), and diethyl phthalate (DEP) were detected at a rate of 95.2, 90.5, and 90.5%, respectively. The detection rates for all other plasticizers ranged from 0 to 57.1%. The content of total plasticizers in oil samples was determined to be 210-7558 μg/kg, which was comparable to the content range in oil marketed in Italy. Although no significant difference (p = 0.05) in the total content of plasticizer was observed among oil species (soybean, canola, corn, and olive), the wider range and higher average of total content of plasticizers in olive oil than other oil species indicated the inconsistence of plasticizer contamination in olive oil and a possible priority for quality monitoring. No significant difference (p = 0.05) in the total content of plasticizers was found among glass-bottle (n = 4), plastic-bottle (n = 14), and metal-can (n = 3) packaging, implying that oil packaging is not the major cause of plasticizer contamination. The daily intake amount of plasticizers contained in edible oil on this U.S. retail market constituted only a minimum percentage of reference dose established by US EPA, thus no obvious toxicological effect might be caused. However, the fact that DEHP content in two olive oils exceeded relevant special migration limits (SMLs) of Europe and China might need attention.

Uptake of Di(2-ethylhexyl) Phthalate (DEHP) by the Plant Benincasa hispida and Its Use for Lowering DEHP Content of Intercropped Vegetables

Uptake of di(2-ethylhexyl) phthalate (DEHP) by the plant Benincasa hispida and its use for topical phytoremediation were investigated by cultivation of plants in DEHP-contaminated environments. The results showed that major plant organs of B. hispida , including leaves, stems, and fruits, readily absorbed DEHP from the air. The amount of DEHP that accumulated in leaves, stems, and fruits was mainly dependent upon exposure time, and most DEHP accumulated in their inner tissues. A single plant of B. hispida with a gourd was able to absorb more than 700 mg of DEHP when it was exposed to DEHP-contaminated air for 6 week. B. hispida reduced air DEHP concentration by 65-76% as the air DEHP concentration ranged from 2351 to 3955 μg/m³ (high DEHP level) and 85-92% as the air DEHP concentration ranged from 35.1 to 65.3 μg/m³ (low DEHP level) in greenhouse experiments. When intercropping of B. hispida and Brassica chinensis or Brassica campestris , B. hispida reduced more than 87% of DEHP accumulation in the latter, which indicates that B. hispida has excellent use potential for lowering the DEHP content of intercropped vegetables.
