Uptake of hexavalent chromium by Lactuca sativa and Triticum aestivum plants and mediated effects on their performance, linked with associated public health risks

Plants exposed to toxic Cr(VI) concentrations show reduced biomass production and yield. The present study examined (1) the impacts of increasing Cr(VI) exposure (0.05, 0.5, 1, 5 and 10 mg L−1 K2Cr2O7 in irrigation water) on the growth and development, yield, and stress physiology of two important crop species, such as wheat (Triticum aestivum L.) and lettuce (Lactuca sativa L.), and (2) the associated human health risks due to the consumption of these commodities. Plants in all Cr(VI) treatments preserved similar growth rates and phenotypes with control, untreated plants, with no remarkable modulations in biomass yield (fresh/dry weight, plant height, straw/grain weight). Grains harvested from plants exposed to all Cr(VI) treatments had increased size and 1000 grain weight, partially suggesting hormetic effect. Elevated K2Cr2O7 concentrations (5 and 10 mg L−1) resulted in increased photosynthetic pigments’ levels in lettuce leaves. Lipid peroxidation and H2O2 content also revealed the absence of oxidative stress in lettuce plants. Non-carcinogenic (target hazard quotient) and carcinogenic risks from the consumption of these commodities grown under the conditions described in this study were found de minimis. However, more studies are needed in order to obtain more solid information on the safety of Cr(VI)-contaminated water irrigation.

Physiological and antioxidant responses of Medicago sativa-rhizobia symbiosis to cyanobacterial toxins (Microcystins) exposure

Toxic cyanobacteria in freshwaters can induce potent harmful effects on growth and development of plants irrigated with contaminated water. In this study, the effect of cyanobacteria extract containing Microcystins (MC) on Medicago sativa-rhizobia symbiosis was investigated in order to explore plants response through biomass production, photosynthetic pigment and antioxidant enzymes analysis: Peroxidase (POD), Polyphenoloxidase (PPO) and Catalase (CAT). Alfalfa plants were inoculated with two endosymbiotic rhizobial strains: RhOL1 (MC less sensitive strain) and RhOL3 (MC more sensitive strain), to evaluate the rhizobial contribution on the plant response cultured under cyanobacterial toxins stress. The two rhizobia strains were identified as Ensifer meliloti by sequence analysis of their rrs and atpD genes. The chronic exposure to MC extract showed shoot, root and nodules dry weight decrease, in both symbiosis cultures. The rate of decline in plants inoculated with RhOL3 was higher than that in symbiosis with RhOL1 mainly at 20 μg L−1 of MC. Cyanotoxins also reduced photosynthetic pigment content and generated an oxidative stress observed at cellular level. POD, PPO and CAT activities were significantly increased in leaves, roots and nodules of alfalfa plants exposed to MC. These enzyme activities were higher in plants inoculated with RhOL3 especially when alfalfa plants were exposed to 20 μg L−1 of MC. The present paper reports new scientific finding related to the behavior of rhizobia-M. sativa associations to MC (Microcystins) for later recommendation concerning the possible use of these symbiosis face to crops exposure to MC contaminated water irrigation.

Persistence of Escherichia coli on injured iceberg lettuce in the field, overhead irrigated with contaminated water.

Fresh produce is increasingly implicated in food-related illnesses. Escherichia coli can survive in soil and water and can be transferred onto plant surfaces through farm management practices such as irrigation. A trial was conducted to evaluate the impact of field conditions on E. coli persistence on iceberg lettuce irrigated with contaminated water, and the impact of plant injury on the persistence of E. coli. Lettuce heads were injured at 14, 7, 3, 2, 1, and 0 days before inoculation, with uninjured heads used as a control. All lettuce heads (including controls) were overhead irrigated with a mixture of nonpathogenic E. coli strains (10(7) CFU/ml). E. coli counts were measured on the day of inoculation and 5 days after, and E. coli was detected on all lettuce head samples. Injury immediately prior to inoculation and harvest significantly (P = 0.00067) increased persistence of E. coli on lettuce plants. Harsh environmental conditions (warm temperatures, limited rainfall) over 5 days resulted in a 2.2-log reduction in E. coli counts on uninjured lettuce plants, and lettuce plants injured more than 2 days prior to inoculation had similar results. Plants with more recent injuries (up to 2 days prior to inoculation) had significantly (P = 7.6 x 10(-6)) greater E. coli persistence. Therefore, growers should postpone contaminated water irrigation of lettuce crops with suspected injuries for a minimum of 2 days, or if unavoidable, use the highest microbiological quality of water available, to minimize food safety risks.

Lysimeter Studies on Nitrogen Transport and Transformation as Affected by Contaminated Water Irrigation

Contaminated river water irrigation was conducted in two couples of lysimeters with 1 and 3.5m soil depths, respectively, to identify factors affecting transport and transformation of organic matters and nitrogen compounds. During 2months of experimentation, 372kg∕ha of total nitrogen mineralized into inorganic nitrogen. The amounts of nitrification and denitrification were as high as 387.8 and 321.8kg∕ha, respectively. 40.5kg∕ha of NO3−–N leached into groundwater, which amounted to 78% of the initial NO3−–N. Therefore, denitrification and the leaching are the main processes for the loss of nitrogen in soil. Soil moisture was a primary factor affecting the transport and transformation of nitrogen (N) compounds. Lysimeter studies demonstrate that chemical oxygen demand (COD) and NH4+–N concentrations changed with spatial and temporal variations of the water content and the concentration of NH4+–N is highly positively correlated with COD. However, there is no significant correlation between soil moisture and NO3−–N, and the relationship between NH4+–N and NO3−–N is poor.
