Dietary supplementation of phytoncide and soybean oil increases milk conjugated linoleic acid and depresses methane emissions in Holstein dairy cows

The objective of this study was to determine whether adding phytoncide oil (PO) and soybean oil (SBO) to the dairy cow diet could increase milk conjugated linoleic acid (CLA) and depress methane (CH4) emissions in Holstein dairy cows. Rumen fermentation was conducted at four levels of SBO (0, 1, 2, and 4%, on DM basis) and two levels of PO (0 and 0.1%, on DM basis) with in vitro experiment. To evaluate blood parameters, fecal microbe population, milk yield and fatty acid compositions, and CH4 production, in vivo experiment was conducted using 38 Holstein dairy cows divided into two groups of control (fed TMR) and treatment (fed TMR with 0.1% PO and 2% SBO as DM basis). In the in vitro study (Experiment 1), PO or SBO did not affect rumen pH. However, SBO tended to decrease ruminal ammonia-N (p = 0.099). Additionally, PO or SBO significantly decreased total gas production (p = 0.041 and p = 0.034, respectively). Both PO and SBO significantly decreased CH4 production (p < 0.05). In addition, PO significantly increased both CLA isomers (c9, t11 and t10, c12 CLA) (p < 0.001). Collectively, 0.1% PO and 2% SBO were selected resulting in most effectively improved CLA and decreased CH4 production. In the in vivo study (Experiment 2), 0.1% PO with 2% SBO (PSO) did not affect complete blood count. However, it decreased blood urea nitrogen and magnesium levels in blood (p = 0.021 and p = 0.01, respectively). PSO treatment decreased pathogenic microbes (p < 0.05). It increased milk yield (p = 0.017) but decreased percentage of milk fat (p = 0.013) and MUN level (p < 0.01). In addition, PSO treatment increased both the concentration of CLA and PUFA in milk fat (p < 0.01). Finally, it decreased CH4 emissions from dairy cows. These results provide compelling evidence that a diet supplemented with PSO can simultaneously increase CLA concentration and decrease CH4 production with no influence on the amount of milk fat (kg/day) in Holstein dairy cows.

Progress of Conjugated Linoleic Acid on Milk Fat Metabolism in Ruminants and Humans.

As a valuable nutrient in milk, fat accounts for a significant proportion of the energy requirements of ruminants and is largely responsible for determining milk quality. Fatty acids (FAs) are a pivotal component of milk fat. Conjugated linoleic acid (CLA) is one of the naturally occurring FAs prevalent in ruminant dairy products and meat. Increasing attention has been given to CLA because of its anti-cancer, anti-inflammatory, immune regulation, and lipid metabolism regulation properties, and these benefits potentially contribute to the growth and health of infants. In breast milk, CLA is present in trace amounts, mainly in the form of cis-9, trans-11 CLA. Notably, cis-9, trans-11 CLA improves the milk fat rate while trans-10, cis-12 CLA inhibits it. Apart from having multiple physiological functions, CLA is also a pivotal factor in determining the milk quality of ruminants, especially milk fat rate. In response to growing interest in green and healthy functional foods, more and more researchers are exploring the potential of CLA to improve the production performance of animals and the nutritional value of livestock products. Taken together, it is novel and worthwhile to investigate how CLA regulates milk fat synthesis. It is the purpose of this review to clarify the necessity for studying CLA in ruminant milk fat and breast milk fat.

Effect of genotype, lactation and climatic factors on fatty acid profile of bovine milk

Milk fat from bovine milk contains fatty acids that may have favourable properties for human health, for example, conjugated linoleic acid (CLA) has nutraceutical activity. This research aimed to know the effects of genotype, days of lactation and climatic factors on the fatty acids (FA) profile of milk and particularly the content of CLA in milk fat. Seventeen first-calving milking cows in early lactation were used for the assessment of milk; 12 were Gyr and five were F1 (Holstein/Gyr) crosses. Sampling was carried out every 15 days, from the beginning to the end of lactation (300 days). Fatty acids were analyzed employing gas chromatography. The genotype did not influence the content of the fatty acid groups: saturated fatty acids (SFA), monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA) and CLA. The highest MUFA and PUFA contents were recorded at 100 days of lactation (32.334 and 3.553 g/100 g of FA, respectively), while SFA and CLA had their highest production at 200 days of lactation (63.238 and 1.378 g/100 g of FA, respectively). Regarding the climate, the highest temperature caused a decrease in the CLA content, because temperatures above 30 °C caused a decrease in the grazing time.

Comparative effect of Volvariella volvacea-treated rice straw and purple corn stover fed at different levels on predicted methane production and milk fatty acid profiles in tropical dairy cows

The objective of this study was to determine the effect of different concentrations of rice straw (RS) and purple corn stover (PCS) treated with Volvariella volvacea fed in total mixed rations (TMR) on ruminal fermentation, blood metabolites, calculated methane (CH4) production, and milk fatty acid profile in mid-lactation crossbred Holstein Friesian cows. The experimental design was a 2 × 2 factorial arrangement of treatments in a 4 × 4 Latin square design. Factor A included roughage type (V. volvacea-treated RS and V. volvacea-treated PCS), and Factor B included roughage (R)-to-concentrate (C) ratio (R:C) (50:50 and 60:40). The four dietary treatment combinations were 1) RS with an R:C ratio of 50:50 (RS50); 2) RS with an R:C ratio of 60:40 (RS60); 3) PCS with an R:C ratio of 50:50 (PCS50); and 4) PCS with an R:C ratio of 60:40 (PCS60). Different roughage sources did not alter total tract digestibility of DM, OM, and NDF (P > 0.05), but PCS increased NDF digestibility by 4.6 percentage units and CP digestibility by 15.6 percentage units. Roughage types influenced rumen propionic acid (C3) concentration 4 h after feeding with PCA increasing higher C3 concentration compared to RS at both R:C ratios. There was no interaction between roughage types and the R:C ratio for CH4 emission calculated based on volatile fatty acid profiles. However, roughage type impacted CH4 emission. Calculated CH4 concentration was lower in PCS than RS. Changing the R:C ratio did not alter milk production, milk composition, or somatic cell count (SCC). Moreover, roughage type did not influence milk production and milk composition, except for SCC which was lower in PCS. Roughage type and the R:C ratio independently influenced milk fat arachidonic acid (C20:4n-6) concentration. Furthermore, the PCS group had higher conjugated linoleic acid (CLA) than the RS groups. The proportion of total C18:0 and cis-9 C18:1 in milk fat increased 10% with the feeding of PCS50 and PCS60. However, milk cis-9, trans-11 C18:2 increased by 61% in the PCS group (P < 0.01). In conclusion, PCS increased rumen C3 and milk fat CLA and arachidonic acid concentration and decreased CH4 production compared to the RS. The PCS treated with V. volvacea could be used for up to 60% of TMR for dairy cows.

Anticancer activity of milk fat rich in conjugated linoleic acid against Ehrlich ascites carcinoma cells in female Swiss albino mice.

Background and Aim:The major conjugated linoleic acid (CLA) isomers have anticancer effect, especially breast cancer cells, inhibits cell growth and induces cell death. Also, CLA has several health benefits in vivo, including antiatherogenesis, antiobesity, and modulation of immune function. The present study aimed to assess the safety and anticancer effects of milk fat CLA against in vivo Ehrlich ascites carcinoma (EAC) in female Swiss albino mice. This was based on acute toxicity study, detection of the tumor growth, life span of EAC bearing hosts, and simultaneous alterations in the hematological, biochemical, and histopathological profiles.Materials and Methods:One hundred and fifty adult female mice were equally divided into five groups. Groups (1-2) were normal controls, and Groups (3-5) were tumor transplanted mice (TTM) inoculated intraperitoneally with EAC cells (2×106/0.2 mL). Group (3) was (TTM positive control). Group (4) TTM fed orally on balanced diet supplemented with milk fat CLA (40 mg CLA/kg body weight). Group (5) TTM fed orally on balanced diet supplemented with the same level of CLA 28 days before tumor cells inoculation. Blood samples and specimens from liver and kidney were collected from each group. The effect of milk fat CLA on the growth of tumor, life span of TTM, and simultaneous alterations in the hematological, biochemical, and histopathological profiles were examined.Results:For CLA treated TTM, significant decrease in tumor weight, ascetic volume, viable Ehrlich cells accompanied with increase in life span were observed. Hematological and biochemical profiles reverted to more or less normal levels and histopathology showed minimal effects.Conclusion:The present study proved the safety and anticancer efficiency of milk fat CLA and provides a scientific basis for its medicinal use as anticancer attributable to the additive or synergistic effects of its isomers.

Evaluation of the Impact of Ruminant Trans Fatty Acids on Human Health: Important Aspects to Consider

The definition and evaluation of trans fatty acids (TFA) with regard to foodstuffs and health hazard are not consistent. Based on the current situation, the term should be restricted only to TFA with isolated double bonds in trans-configuration. Conjugated linoleic acids (CLA) should be separately assessed. Ideally, the origin of the consumed fat should be declared, i.e., ruminant TFA (R-TFA) and industrial TFA (non-ruminant; I-TFA). In ruminant fat, more than 50% of R-TFA consists of vaccenic acid (C18:1 t11). In addition, natural CLA, i.e., c9,t11 CLA is also present. Both are elevated in products from organic farming. In contrast to elaidic acid (t9) and t10, which occur mainly in partially hydrogenated industrial fat, t11 is partially metabolized into c9,t11 CLA via Δ9-desaturation. This is the major metabolic criterion used to differentiate between t11 and other trans C18:1. t11 indicates health beneficial effects in several studies. Moreover, CLA in milk fat is associated with the prevention of allergy and asthma. An analysis of the few studies relating to R-TFA alone makes clear that no convincing adverse physiological effect can be attributed to R-TFA. Only extremely high R-TFA intakes cause negative change in blood lipids. In conclusion, in most European countries, the intake of R-TFA is assessed as being low to moderate. Restriction of R-TFA would unjustifiably represent a disadvantage for organic farming of milk.

Effects of conjugated linoleic acids and dietary concentrate proportion on performance, milk composition, milk yield and metabolic parameters of periparturient dairy cows

The study aimed to examine effects of supplemented conjugated linoleic acids (CLA) to periparturient cows receiving different concentrate proportions ante partum (a.p.) to investigate CLA effects on lipid mobilisation and metabolism. Compared to adapted feeding, a high-concentrate diet a.p. should induce a ketogenic metabolic situation post partum (p.p.) to better understand how CLA works. Sixty-four pregnant German Holstein cows had ad libitum access to partial mixed rations 3 weeks prior to calving until day 60 p.p. Ante partum, cows received control fat (CON) or a CLA supplement at 100 g/d, either in a low-concentrate (CON-20, CLA-20) or high-concentrate diet (CON-60, CLA-60). Post partum, concentrate proportion was adjusted, while fat supplementation continued. After day 32 p.p., half of the animals of CLA-groups changed to CON supplementation (CLA-20-CON, CLA-60-CON). A ketogenic metabolic situation p.p. was not achieved and therefore impacts of CLA could not be examined. Live weight, milk yield and composition, blood parameters remained unaffected by the treatments. Only a slightly reduced milk fat yield (not significant) was recorded for Group CLA-20. The proportion of trans-10,cis-12 (t10,c12) CLA in milk fat was significantly increased in CLA-groups compared to CON-groups. With the exception of a reversible CLA effect on milk fat in Group CLA-20, no post-treatment effects occurred. Dry matter intake (DMI) of Group CLA-60 was highest before calving, resulting in a significantly improved estimated energy balance after calving. Ante partum, net energy intakes were significantly increased in high-concentrate groups. Overall, supplemented CLA preparation did not relieve metabolism and lipid mobilisation of early lactating cows. But feeding CLA in a high-concentrate diet a.p. seems to increase DMI and thereby improve the energy balance of cows immediately after calving.

An unprotected conjugated linoleic acid supplement decreases milk production and secretion of milk components in grazing dairy ewes

Feeding conjugated linoleic acid (CLA) in a rumen-inert form to dairy ewes has been shown to increase milk production, alter milk composition, and increase the milk fat CLA content. However, few studies have tested ruminally unprotected CLA sources. The objective of this study was to evaluate the effects of an unprotected CLA supplement (29.8% of cis-9,trans-11 and 29.9% of trans-10,cis-12 isomers as methyl esters) on milk yield and composition of dairy ewes. Twenty-four lactating Lacaune ewes were used in a crossover design and received 2 dietary treatments: (1) control: basal diet containing no supplemental lipid and (2) basal diet plus CLA (30g/d). The CLA supplement was mixed into the concentrate and fed in 2 equal meals after morning and afternoon milkings. Each experimental period consisted of 21 d: 7 d for adaptation and 14 d for data collection. The CLA supplement decreased milk fat content and yield by 31.3 and 38.0%, respectively. Milk yield and secretion of milk lactose and protein were decreased by 8.0, 9.8, and 5.6%, respectively. On the other hand, milk protein content and linear SCC score were 1.8 and 17.7% higher in ewes fed the CLA supplement. The concentration of milk fatty acids originating from de novo synthesis (<C16) was decreased by 25%, whereas the concentration of milk fatty acids taken up preformed from the plasma (>C16) was increased by 22.6% in ewes fed the CLA supplement. The CLA supplement decreased C14:1/C14:0, C16:1/C16:0, and C18:1/C18:0 desaturase indexes by 25, 18.7, and 0.1%, respectively, but increased the cis-9,trans-11 CLA/trans-11 C18:1 ratio by 8.6%. The concentrations of trans-10,cis-12 CLA and cis-9,trans-11 CLA in milk fat was 309 and 33.4% higher in ewes fed CLA. Pronounced milk fat depression coupled with the deleterious effects on milk yield, milk SCC, and secretion of all milk solids observed in ewes fed an unprotected CLA supplement is likely to be associated with high doses of trans-10,cis-12 CLA reaching the mammary gland, corroborating previous results obtained with dairy cows.

Effects of long-term supplementation of dairy cow diets with rumen-protected conjugated linoleic acids (CLA) on performance, metabolic parameters and fatty acid profile in milk fat

The supplementation of conjugated linoleic acids (CLA) to the rations of dairy cows represents an opportunity to reduce the content of milk fat. Therefore, CLA have the potential beneficial effect of reducing energy requirements of the early lactating cow. The present study aimed at the examination of long-term and post-treatment effects of dietary CLA intake on performance, variables of energy metabolism-like plasma levels of non esterified fatty acids (NEFA) and ß-hydroxybutyrate (BHB), and fatty acid profile in milk fat. Forty-six pregnant German Holstein cows were assigned to one of three dietary treatments: (1) 100 g/d of control fat supplement (CON), (2) 50 g/d of control fat supplement and 50 g/d of CLA supplement (CLA-1) and (3) 100 g/d of CLA supplement (CLA-2). The lipid-encapsulated CLA supplement consisted of approximately 10% of trans-10, cis-12 CLA and cis-9, trans-11 CLA each. The experiment started 1 d after calving and continued for about 38 weeks, divided into a supplementation (26 weeks) and a depletion period (12 weeks). Over the first 7 weeks of treatment, 11 and 16% reductions in dry matter intake compared to control were observed for the cows fed CLA-1 and CLA-2 supplements respectively. Consequently, the calculated energy balance for these two CLA groups was lower compared to the control. Plasma levels of NEFA and BHB remained unaffected. Later in lactation the highest CLA supplementation resulted in a reduction of milk fat content of 0.7%. However, no reduction in milk fat yield, and accordingly no milk fat depression (MFD), could be shown. The trans-10, cis-12 CLA in milk fat increased with increasing dietary CLA supplementation in a dose-dependent manner. The proportion of C16 in milk fat was decreased by the highest CLA supplementation. With the exception of an increase in plasma glucose level in the CLA-2 group, no post-treatment effects were observed. Overall, under the conditions of the present study no improvement in the calculated energy balance by CLA supplementation could be shown for the entire evaluation period.

Comparison of Conjugated Linoleic Acid and Other Fatty Acid Content of Milk Fat of Mafriwal and Jersey Cows

Special attention has been given to the milk Fatty Acids (FA) that have a beneficial effect for human health such as mono and poly unsaturated fatty acids in particularly the Conjugated Linoleic Acids (CLA). This study was undertaken to investigate the milk fat contents of CLA variables (CLA and CLA-desaturase index) and other FA composition of Mafriwal and Jersey cows under same feeding system. In addition, the relationship between these two CLA variables with milk production and milk fat percent was determined. All the cows were grazed on pasture and given 5.5 kg of concentrate per head daily. Milk FA composition was determined using gas chromatography after extraction of milk fat using modified Folch's method. The results showed a significant variation (p<0.05) in the FA contents of the two breeds. The cis-9, trans-II CLA and CLA-desaturase index in milk fat of Mafriwal were significantly higher (p<0.05) than that of Jersey cows. Mafriwal cows produced significantly (p<0.05) higher concentrations of C18:0, C18:1cis-9, C18:3 and C20:1 than that of Jersey, while Jersey cows produced significantly (p<0.05) higher concentrations of C12:0 and C14:0 than Mafriwal cows. Additionally, significant positive correlations were observed between CLA variables and milk production. This study indicates that the breed of cows has an effect on CLA and other FA composition of milk fat and Mafriwal cows produced significantly higher percentages of CLA than Jersey cows which would provide better benefits for human health. Furthermore, the milk fat content of CLA and CLA-desaturase index were positively related to the milk production.

Milk Conjugated Linoleic Acid (CLA) Profile and Metabolic Responses of Dairy Cows Fed with High-temperature-micro-time (HTMT) Treated Diets Containing High Quantity Extruded Soybean (ESB)

A feeding trial was conducted to examine the effect of high-temperature-micro-time (HTMT) processing of diets containing extruded soybean (ESB) in high quantity on milk fat production, metabolic responses, and the formation of conjugated linoleic acid (CLA) and trans-vaccenic acid (TVA). Twenty-one multiparous Holstein cows in mid-lactation were blocked according to milk yield in the previous lactation. Cows within each block were randomly assigned to either normal concentrate or HTMT treated diets containing ESB (7.5% HTMT-ESB and 15% HTMT-ESB). It was hypothesized that the HTMT-ESB would affect the undegradable fatty acids in the rumen and, thus, would modify the fatty acid profile of milk fat. Both 7.5% and 15% HTMT-ESB did not affect milk yield, fat, protein, lactose and solid-not-fat (SNF), but the proportion of cis-9, trans-11 CLA in milk fat was significantly increased by these treatments. Content of TVA in milk fat was not affected by HTMT-ESB. The HTMT-ESB influenced the fatty acid profile in milk fat, but there was little difference between 7.5% and 15% of supplementation. HTMT-ESB feeding significantly decreased the concentration of plasma insulin and glucose, while plasma growth hormone (GH), triglyceride (TG), non-esterified fatty acid (NEFA) and HDL-cholesterol were increased by 7.5% and 15% ESB-HTMT supplementation in comparison to the control group (p 0.05). The present results showed that cis-9, trans-11 CLA production was increased by HTMT treatment of dietary ESB without reduction of milk fat, and the unchanged milk fat and yield was assumed to be associated with the constant level of thyroid hormones, leptin, and IGF-1.

Milk production and fatty acid profile of dairy cows supplemented with flaxseed oil, soybean oil, or extruded soybeans

The objective of the study was to investigate the effects of oil sources in the diet on milk yield, milk composition, and fatty acid (FA) profiles in mid-lactating dairy cows. Forty-eight Chinese Holstein dairy cows averaging 150 days in milk (DIM) at the start of the experiment (body weight = 596±19 kg; milk yield = 29.7±3.00 kg/d) were used in a completely randomized block design. The animals were assigned into four dietary treatments according to DIM and milk yield, and supplemented with no oil (control), 2% flaxseed oil (FSO), 2% soybean oil (SBO), and 2% oil from extruded soybeans (ESB). The experiment lasted nine weeks including the first week for adaptation. Milk yields, milk compositions (fat, protein, and lactose), and milk FA profiles were measured. Daily milk yield from cows fed with FSO, SBO, and ESB were higher than milk yield of the control cows (27.0, 27.0, and 26.5 vs. 25.4 kg/d). Milk fat percentage of the control cows was greater than those cows fed with oil-supplemented diets. However, increasing dietary fat content resulted with no change in fat-corrected milk yield. The FA profile of milk was changed by fat supplementation. Feeding oil reduced the proportion of both short-chain (C8:0 to C12:0) and medium-chain (C14:0 to C16:1) FAs, and increased the proportion of long-chain (≥C18:0) FAs in milk fat. Cis-9, trans-11 conjugated linoleic acid (CLA) in milk fat was increased from 0.38% for the control to 0.79, 1.51, and 1.56% of fat for the cows supplemented with FSO, SBO, and ESB, respectively. Feeding oils rich in linoleic acid (SBO and ESB) was more effective in enhancing cis-9, trans-11 CLA in milk fat than oils containing linolenic acid (FSO). There was a linear relationship between transvaccenic acid and cis-9, trans-11 CLA content in milk. Overall, feeding the FSO, SBO, and ESB diets increased monounsaturated and polyunsaturated fatty acids and decreased the saturated fatty acid in milk fat.

Effects of Protected Conjugated Linoleic Acid Supplementation on Milk Fatty Acid in Dairy Cows

The objective was to determine the effects of supplementation of protected conjugated linoleic acid (CLA), CLA-20 comprising 10% each of cis-9, trans-11 and trans-10, cis-12, on milk production and fatty acid profiles in plasma and milk in lactating dairy cows. Five mid-lactation, multiparous crossbred Holstein Friesian cows with average 402±20 kg BW were used in a 5x5 Latin square design for 21-d periods. Cows were given a total mixed ration (TMR) and supplemented with CLA-20 at 0, 20, 40, 80 and 160 g/d. The results showed that dry matter intake depression occurred in cows supplemented with CLA-20 at 160 g/d. Milk production slightly increased when CLA-20 supplementation was at 20, 40 and 80 g/d. However, 3.5% fat-corrected milk (FCM) was not affected by CLA-20 supplementation. Increased levels of CLA-20 supplementation resulted in a significantly decreased percentage of milk fat. Plasma concentrations of fatty acid were not altered by the amounts of CLA-20 supplementation except for the concentration of trans10, cis-12 CLA. For all dietary treatments, percentages of fatty acids (C4:0, C6:0, C8:0, C13:0, C14:0 C14:1 C15:0 C15:1 C16:0, C16:1, C18:1n9t, C18:2n6t, C18:2n6c, C20:0, C18:3n6, C18:3n3, C20:1 and C20:3n6) in milk fat were similar. Concentrations of C10:0, C11:0, C12:0 and C18:1n9c were decreased cubically and C18:0 was elevated linearly (p<0.01) according to the increased amounts of CLA-20 supplemented. The linear increase was observed for cis-9, trans-ll CLA (0.62, 1.17, 1.94, 1.87 and 1.82% of total fatty acid), trans-10, cis-12 CLA (0.01, 0.63, 0.67, 0.93 and 0.95% of total fatty acid) and total CLA (0.80, 2.25, 3.16, 3.97 and 3.94% of total fatty acid) in milk fat from 0 to 160 g/d of CLA-20 supplement. In conclusion, concentration of cis-9, trans-ll CLA in milk fat was concomitantly elevated at an increasing rate with the increased amounts of CLA-20. Based on the results in this study, supplementation of CLA-20 at 80 g/d optimally enhanced total CLA in milk fat.

Designing milk fat to improve healthfulness and functional properties of dairy products: from feeding strategies to a genetic approach

The present review, in the first part, deals with the most effective feeding strategies applied to dairy ruminants in order to enhance the healthfulness of milk fat. The largest changes in milk fatty acid (FA) composition have been obtained either by changing the amounts and the nature of forages in the diets of ruminants, particularly pasture, or by adding plant or marine oils to the diet. Alpine and legume based pastures are associated with high levels of conjugated linoleic acid (CLA), omega-3 FA and with low levels of saturated FA, but further studies are needed with the aim to better explain the effect of some plant species on milk FA composition. Linseed, soybeans safflower and sunflower are the most effective sources of unsaturated plant lipids used to enhance CLA and unsaturated FA content in milk fat. Among animal sources, marine oil is more effective than plant oils for enhancing CLA, vaccenic acid (VA) and omega-3 FA in milk fat, especially when fish oil is fed in combination with oil supplements rich in linoleic acid. In the second part of the review the potential contribute of genetic improvement to modifying milk FA composition is discussed. Recent studies have suggested that the genetic improvement of the nutritional quality of milk based on FA profile may be possible. At this aim, genetic parameters of milk FA composition have been estimated in Dutch, US and Italian Holstein populations and in a Belgian multiple breed population of dairy cows. In dairy sheep and goat there is still a lack of knowledge about genetic parameters and correlations of milk FA. The develop of specific selection indexes aimed to improve the nutritional properties of milk could be supported by the new insight about potential candidate genes able to affect a significant quote of the milk FA variability. Increasing evidences indicate the Stearoyl-CoA Desaturase (SCD) and Diacylglycerol acyltransferase (DGAT-1) genes as possible sources of FA variation in milk. Evidences from quantitative and molecular studies suggest a potential future animal selection, but some question about the kind of selection mechanism and the economic incentive for the improvement should be preliminarily answered.

The effect of dietary inclusion of olive tree leaves and grape marc on the content of conjugated linoleic acid and vaccenic acid in the milk of dairy sheep and goats

Olive tree leaves (OTL) and grape marc (GM) are by-products with high linolenic (LNA) and linoleic (LA) acid content, respectively, which can be used as dietary ingredients to increase the cis-9 trans-11 conjugated linoleic acid (CLA) content of milk fat in sheep and goats. An experiment was conducted with 16 Friesian ewes and 16 Alpine goats to study the effect of OTL and GM inclusion in sheep and goat diets on their milk fatty acid profile, with emphasis on cis-9, trans-11 CLA and vaccenic acid (VA). Ewes and the goats were fed the control (C) diets from parturition to the 90 days in milk (DIM) and then both groups were divided into two sub-groups (treatments). The control groups of both species continued to be fed the C diets, whereas the treated groups were gradually switched over a 2-week period (DIM=91-105) from the C diets to that of treatment 1, which contained air-dried OTL. These OTL diets were fed ad libitum for 1 month (DIM=106-135). After that period, the same treated groups, after 2 weeks of gradual adaptation (DIM=136-150), were switched to treatment 2, which contained air-dried GM. The GM diets were fed ad libitum for 1 month (151-180 DIM). Concentrations of polyunsaturated fatty acids (PUFA) and mono-unsaturated fatty acids increased significantly in milk fat of sheep fed OTL v. C. For goats, only the PUFA in milk fat was increased by feeding OTL compared with C. Relative to C, GM increased significantly the concentration of PUFA only in milk fat of sheep. OTL and GM diets increased the cis-9, trans-11 CLA and VA content in milk fat, compared with C, only in sheep. GM caused a sharp increase in 18:0 only in sheep milk fat, while the OTL diet increased significantly the 18:0 in milk fat of goats. GM and OTL diets also had opposite effects on the 18:1/18:0 ratio of sheep milk fat. In conclusion, OTL and GM, when included in sheep diets altered the milk fatty acid profile with a pronounced increase in cis-9, trans-11 CLA and VA contents. The results show that the response of sheep and goats to OTL and GM diets was different, suggesting a species difference that needs further investigation.

Effect of dietary sources of roasted oilseeds on blood parameters and milk fatty acid composition

The aim of this experiment was to investigate the effect of supplementing the basal diet with oilseeds on blood parameters and composition of milk fatty acids, especially conjugated linoleic acid (CLA). Forty-eight lactating Holstein cows in early lactation were used in a randomized block design. The cows in each group were fed the control basal diet (the control diet) or diets containing roasted soybean (RSB), roasted linseed (RLS), roasted sunflower seed (RSS), hulled roasted peanut (HRP) and roasted cottonseed (RCS), respectively. Milk yield and dry matter intake (DMI) were not significantly different. Milk fat percentage and yield decreased (&lt;I&gt;P&lt;/I&gt; &lt; 0.05) in RLS, RSS and RCS diets compared with the control. Feeding various oilseeds had no effect on plasma parameters, but it tended to increase concentrations of &lt;I&gt;trans&lt;/I&gt; C18:1 and C18:2 in plasma. In milk fat, the concentrations of short and medium fatty acids decreased while C&lt;sub&gt;18&lt;/sub&gt; unsaturated fatty acids increased when the cows were fed oilseed diets. &lt;I&gt;Ci&lt;/I&gt;s-9, &lt;I&gt;trans &lt;/I&gt; -11 CLA content increased (&lt;I&gt;P&lt;/I&gt; &lt; 0.01) in the milk fat of cows fed oilseeds. RSB treatment produced the highest (&lt;I&gt;P&lt;/I&gt; &lt; 0.01) content of&lt;I&gt; cis&lt;/I&gt;-9, &lt;I&gt;trans &lt;/I&gt; -11 CLA, which was a 60% increase compared with the control. The results indicate that the diets supplemented with oilseeds improve the content of C&lt;sub&gt;18&lt;/sub&gt; unsaturated fatty acids and CLA in milk fat, and soybeans seem to be the optimal source to improve the nutritive value of milk compared with other oilseeds.

Temporal changes in milk conjugated linoleic acidfor cows fed a pasture- or conserved-forages-based diet and supplemented with fish oil and sunflower oil

The objective of this experiment was to evaluate the effect of forage source on the temporal changes in milk conjugated linoleic acid (CLA). Fourteen Holstein cows were divided into two groups. One group (LOT) was fed a corn silage-alfalfa hay mix ad libitum, while the other group (PAS) grazed on alfalfa-grass pasture. Additionally, both groups were fed 8.2 kg d-1 grain supplement containing 650 g of oil composed of fish oil and sunflower oil (1:4). The concentration of cis-9, trans-11 CLA in milk fat of both diets reached maximum levels on day 3 of oil supplementation, declined during day 6, and then remained constant during the reminder of the experiment. Milk vaccenic acid (VA) and trans-10 C18:1 reached their maximum concentrations on days 3 and 6, respectively, of oil supplementation with both diets. In conclusion, the decline in milk cis-9, trans-11 CLA over time was not affected by forage source and may have resulted from an increase in formation of trans-10 C18:1 and C18:0 at the expenses of VA. Performance Comparison of UV and FT-Raman Spectroscopy in the Determination of Conjugated Linoleic Acids in Cow Milk Fat

The determination of conjugated linoleic acids (CLA) in cow milk fat was studied by using UV (210-250 nm) and Fourier transform (FT)-Raman (900-3400 cm (-1)) spectroscopy in order to determine the best spectrophotometric technique for routine analysis of milk fat. A collection of 57 milk fat samples was randomly divided into two sets, a calibration set and a validation set, representing two-thirds and one-third of the samples, respectively. All calculations were performed on the calibration set and then applied to the validation set. The CLA content ranged from 0.56 to 4.70%. A comparison of various spectral pretreatments and different multivariate calibration techniques, such as partial least-squares (PLS) and multiple linear regression (MLR), was done. This paper shows that UV spectroscopy is as reliable as FT-Raman spectroscopy to monitor CLA in cow milk fat. The best calibration for FT-Raman was given by a PLS model of seven factors with a standard error of prediction (SEP) of 0.246. For UV spectroscopy, PLS models were also better than MLR models. The most robust PLS model was constructed with only one factor and with SEP=0.288.

Long-Term Effects of Feeding Monensin on Milk Fatty Acid Composition in Lactating Dairy Cows

The objective of this study was to determine the long-term effects of feeding monensin on milk fatty acid (FA) profile in lactating dairy cows. Twenty-four lactating Holstein dairy cows (1.46±0.17 parity; 620±5.9kg of live weight; 92.5±2.62 d in milk) housed in a tie-stall facility were used in the study. The study was conducted as paired comparisons in a completely randomized block design with repeated measurements in a color-coded, double blind experiment. The cows were paired by parity and days in milk and allocated to 1 of 2 treatments: 1) the regular milking cow total mixed ration (TMR) with a forage-to-concentrate ratio of 60:40 (control TMR; placebo premix) vs. a medicated TMR [monensin TMR; regular TMR + 24mg of Rumensin Premix per kg of dry matter (DM)] fed ad libitum. The animals were fed and milked twice daily (feeding at 0830 and 1300h; milking at 0500 and 1500h). Milk samples were collected before the introduction of treatments and monthly thereafter for 6 mo and analyzed for FA composition. Monensin reduced the percentage of the short-and medium-chain saturated FA 7:0, 9:0, 15:0, and 16:0 in milk fat by 26, 35, 19, and 6%, respectively, compared with the control group. Monensin increased the percentage of the long-chain saturated FA in milk fat by 9%, total monounsaturated FA by 5%, total n-6 polyunsaturated FA (PUFA) by 19%, total n-3 PUFA by 16%, total cis-18:1 by 7%, and total conjugated linoleic acid (CLA) by 43% compared with the control group. Monensin increased the percentage of docosahexaenoic acid (22:6n-3), docosapentaenoic acid (22:5n-3), and cis-9, trans-11 CLA in milk fat by 19, 13, and 43%, respectively, compared with the control. These results suggest that monensin was at least partly effective in inhibiting the biohydrogenation of unsaturated FA in the rumen and consequently increased the percentage of n-6 and n-3 PUFA and CLA in milk, thus enhancing the nutritional properties of milk with regard to human health.

Influence of Supplementing Dairy Cows Grazing on Pasture with Feeds Rich in Linoleic Acid on Milk Fat Conjugated Linoleic Acid (CLA) Content

Three experiments were conducted to investigate the hypothesis that cows grazing on pasture produce the highest proportion of c-9 t-11 CLA in milk fat and no further increase can be achieved through supplementation of diets rich in linoleic acid, such as full-fat extruded soybeans or soybean oil. In experiment 1, 18 lactating Holstein cows were used in a randomized complete block design with measurements made from wk 4 to 6 of the experiment. In experiment 2, three cannulated lactating Holstein cows were used in a 3x3 Latin square design. Each period was 4 wk with measurements made in the final wk of each period. Cows in both experiments were assigned at random to treatments: a, conventional total mixed ration (TMR); b, pasture (PS); or c, PS supplemented with 2.5 kg/cow per day of full-fat extruded soybeans (PES). In both experiments, feed intake, milk yield, milk composition, and fatty acid profile of milk and blood serum were measured, along with fatty acid composition of bacteria harvested from rumen digesta in experiment 2. In experiment 3, 10 cows which had continuously grazed a pasture for six weeks were assigned to two groups, with one group (n = 5) on pasture diet alone (PS) and the other group (n = 5) supplemented with 452 g of soy oil/cow per day for 7 d (OIL). In experiment 1, cows in PS treatment produced 350% more c-9, t-11 CLA compared with cows in TMR treatment (1.70 vs. 0.5% of fat), with no further increase for cows in PES treatment (1.50% of fat). Serum c-9, t-11 CLA increased by 233% in PS treatment compared with TMR treatment (0.21 vs. 0.09% of fat) with no further increase for cows in PES treatment (0.18% of fat). In experiment 2, cows in PS treatment produced 300% more c-9 t-11 CLA in their milk fat compared with cows in TMR treatment (1.77 vs. 0.59% of fat), but no further increase for cows in PES treatment (1.84% of fat) was observed. Serum c-9, t-11 CLA increased by 250% for cows in PS treatment compared with cows in TMR treatment (0.27 vs. 0.11% of fat), with no further increase for cows in PES treatment (0.31% of fat). The c-9, t-11 CLA content of ruminal bacteria for cows in PS treatment was 200% or more of TMR treatment, but no further increase in bacterial c-9, (-11 CLA for cows in PES treatment was observed. Supplementation of soy oil in experiment 3 also did not increase the c-9 t-11 CLA content of milk fat compared with cows fed a full pasture diet (1.60 vs. 1.54% of fat). Based on these findings, it was concluded that supplementing with feeds rich in linoleic acid, such as full-fat extruded soybeans or an equivalent amount of soy oil, to cows grazing perennial ryegrass pasture may not increase milk fat c-9 t-11 CLA contents.
