Production of Conjugated Linoleic Acid from Safflower Oil as Precursor by Probiotic Cultures

Four potentially probiotic strains of lactobacillus and Bifidobacterium were evaluated for their ability to produce conjugated linoleic acid (CLA) from safflower oil in vitro. All the four strains were found to be capable of converting linoleic acid to CLA when using lipolysed safflower oil as a precursor for free linoleic acid. Production of CLA by four probiotics bacterial strains increases in presence 0.6% lipolysed safflower oil as maximum level for 48 h at 37°C and Lb. plantartum has higher CLA content in MRS broth media than Lb. acidophilus, Lb. casei and B. lactis. Also, Supplementation with amounts higher than 0.6% lipolysed safflower oil reduced the CLA content. The research on the ability of converting CLA of probiotics cultures could be basis for the future research and development of fermented dairy products.

Dietary supplementation of phytoncide and soybean oil increases milk conjugated linoleic acid and depresses methane emissions in Holstein dairy cows

The objective of this study was to determine whether adding phytoncide oil (PO) and soybean oil (SBO) to the dairy cow diet could increase milk conjugated linoleic acid (CLA) and depress methane (CH4) emissions in Holstein dairy cows. Rumen fermentation was conducted at four levels of SBO (0, 1, 2, and 4%, on DM basis) and two levels of PO (0 and 0.1%, on DM basis) with in vitro experiment. To evaluate blood parameters, fecal microbe population, milk yield and fatty acid compositions, and CH4 production, in vivo experiment was conducted using 38 Holstein dairy cows divided into two groups of control (fed TMR) and treatment (fed TMR with 0.1% PO and 2% SBO as DM basis). In the in vitro study (Experiment 1), PO or SBO did not affect rumen pH. However, SBO tended to decrease ruminal ammonia-N (p = 0.099). Additionally, PO or SBO significantly decreased total gas production (p = 0.041 and p = 0.034, respectively). Both PO and SBO significantly decreased CH4 production (p < 0.05). In addition, PO significantly increased both CLA isomers (c9, t11 and t10, c12 CLA) (p < 0.001). Collectively, 0.1% PO and 2% SBO were selected resulting in most effectively improved CLA and decreased CH4 production. In the in vivo study (Experiment 2), 0.1% PO with 2% SBO (PSO) did not affect complete blood count. However, it decreased blood urea nitrogen and magnesium levels in blood (p = 0.021 and p = 0.01, respectively). PSO treatment decreased pathogenic microbes (p < 0.05). It increased milk yield (p = 0.017) but decreased percentage of milk fat (p = 0.013) and MUN level (p < 0.01). In addition, PSO treatment increased both the concentration of CLA and PUFA in milk fat (p < 0.01). Finally, it decreased CH4 emissions from dairy cows. These results provide compelling evidence that a diet supplemented with PSO can simultaneously increase CLA concentration and decrease CH4 production with no influence on the amount of milk fat (kg/day) in Holstein dairy cows.

Inducing Conjugated Linoleic Acid Concentration in Tallaga-Like Cheese by Using Probiotics, Inulin and Lipolysed Milk Containing Safflower Oil as Precursor on Its Quality Characteristics

Probiotic functional dairy products are known of their positive impact on overall health. Conjugated linoleic acid (CLA) has a healthy magical effect on human health. So, a new Tallaga-like cheese was prepared by using some probiotic strains and lipolyzed milk with Safflower oil as a source of free linolenic acid as well as with or without inulin as prebiotic and followed its impact on cheese quality characteristics of Tallaga-like cheese stored at 5°C for 28 day. Mixture of equal parts of fresh buffaloe`s and cow`s milk were standardized to 3.60 % fat, pasteurized, cooled to 37°C, salted, mixed well with 0.6% (v/v) lipolyzed milk with Safflower oil, inoculated with probiotic bacteria (Lb. plantarum, Lb. acidophilus, Lb. casei and B. bifidum) and fortified with/without 1.5% inulin to induce CLA formation Results showed that using inulin with 0.6% safflower oil and probiotic bacteria increased significantly the CLA concentration as compared to using probiotics with lipolysed milk only. Addition of different probiotics with inulin increased significantly the titratable acidity, total lactic acid bacteria with higher rheological characteristics and gained higher total acceptability scores. It can be recommended that functional Tallaga-like cheese with more than double CLA, presented in control cheese, can be made with using Lb. plantarum or Lb. acidophilus and 1.5% inulin in the presence of lipolysed milk with safflower oil as a source of free linolenic acid.

Effect of Lactiplantibacillus plantarum on the Conversion of Linoleic Acid of Vegetable Oil to Conjugated Linoleic Acid, Lipolysis, and Sensory Properties of Cheddar Cheese.

Conjugated linoleic acid (CLA) is perceived to protect the body from metabolic diseases. This study was conducted to determine the effect of Lactiplantibacillus plantarum (Lp. plantarum) on CLA production and sensory characteristics of cheddar cheese. Lp. plantarum can convert linoleic acid (LA) to CLA. To increase CLA in cheddar cheese and monitor the conversion of LA to CLA by Lp. plantarum, the LA content of cheese milk (3.4% fat) was increased by partially replacing fat with safflower oil (85% LA of oil) at 0, 3, 6, and 9% concentrations (T1, T2, T3, and T4). Furthermore, Lp. plantarum 108 colony-forming units (CFU)/mL (8 log CFU mL-1) was added in all treatments along with traditional cheddar cheese culture (Lactococcus lactis ssp. lactis and L. lactis ssp. cremoris). After 30 days of ripening, Lp. plantarum in T1, T2, T3, and T4 was 6.75, 6.72, 6.65, and 6.55 log CFU g-1. After 60 days of ripening, Lp. plantarum in T1, T2, T3, and T4 was 6.35, 6.27, 6.19, and 6.32 log CFU g-1. After 60 days of ripening, Lp. plantarum in T1, T2, T3, and T4 was 6.41, 6.25, 6.69, and 6.65 log CFU g-1. GC-MS analysis showed that concentrations of CLA in the 90 days' control, T1, T2, T3, and T4 were 1.18, 2.73, 4.44, 6.24, and 9.57 mg/100 g, respectively. HPLC analysis revealed that treatments containing Lp. plantarum and LA presented higher concentrations of organic acids than the control sample. The addition of safflower oil at all concentrations did not affect cheese composition, free fatty acids (FFA), and the peroxide value (POV) of cheddar cheese. Color flavor and texture scores of experimental cheeses were not different from the control cheese. It was concluded that Lp. plantarum and safflower oil can be used to increase CLA production in cheddar cheese.

Lithium Chloride Promotes Endogenous Synthesis of CLA in Bovine Mammary Epithelial Cells.

Although conjugated linoleic acid (CLA) can promote human health, its content in milk is insufficient to have a significant impact. The majority of the CLA in milk is produced endogenously by the mammary gland. However, research on improving its content through nutrient-induced endogenous synthesis is relatively scarce. Previous research found that the key enzyme, stearoyl-CoA desaturase (SCD) for the synthesis of CLA, can be expressed more actively in bovine mammary epithelial cells (MAC-T) when lithium chloride (LiCl) is present. This study investigated whether LiCl can encourage CLA synthesis in MAC-T cells. The results showed that LiCl effectively increased SCD and proteasome α5 subunit (PSMA5) protein expression in MAC-T cells as well as the content of CLA and its endogenous synthesis index. LiCl enhanced the expression of proliferator-activated receptor-γ (PPARγ), sterol regulatory element-binding protein 1 (SREBP1), and its downstream enzymes acetyl CoA carboxylase (ACC), fatty acid synthase (FASN), lipoprotein lipase (LPL), and Perilipin 2 (PLIN2). The addition of LiCl significantly enhanced p-GSK-3β, β-catenin, p-β-catenin protein expression, hypoxia-inducible factor-1α (HIF-1α), and downregulation factor genes for mRNA expression (P < 0.05). These findings highlight that LiCl can increase the expression of SCD and PSMA5 by activating the transcription of HIF-1α, Wnt/β-catenin, and the SREBP1 signaling pathways to promote the conversion of trans-vaccenic acid (TVA) to the endogenous synthesis of CLA. This data suggests that the exogenous addition of nutrients can increase CLA content in milk through pertinent signaling pathways.

Conjugated Linoleic Acids Have Anti-Inflammatory Effects in Cultured Endothelial Cells.

Conjugated linoleic acid (CLA) isomers may have a role in preventing atherosclerosis through the modulation of inflammation, particularly of the endothelium. However, whether low concentrations of CLAs are able to affect basal unstimulated endothelial cell (EC) responses is not clear. The aim of this study was to evaluate the effects of two CLAs (cis-9, trans-11 (CLA9,11) and trans-10, cis-12 (CLA10,12)) on the basal inflammatory responses by ECs. EA.hy926 cells (HUVEC lineage) were cultured under standard conditions and exposed to individual CLAs for 48 h. Both CLAs were incorporated into ECs in a dose-dependent manner. CLA9,11 (1 μM) significantly decreased concentrations of MCP-1 (p < 0.05), IL-6 (p < 0.05), IL-8 (p < 0.01) and RANTES (p < 0.05) in the culture medium. CLA10,12 (10 μM) decreased the concentrations of MCP-1 (p < 0.05) and RANTES (p < 0.05) but increased the concentration of IL-6 (p < 0.001). At 10 μM both CLAs increased the relative expression of the NFκβ subunit 1 gene (p < 0.01 and p < 0.05, respectively), while decreasing the relative expression of PPARα (p < 0.0001), COX-2 (p < 0.0001) and IL-6 (p < 0.0001) genes. CLA10,12 increased the relative expression of the gene encoding IκK-β at 10 μM compared with CLA9,11 (p < 0.05) and increased the relative expression of the gene encoding IκBα at 1 and 10 μM compared with linoleic acid (both p < 0.05). Neither CLA affected the adhesion of monocytes to ECs. These results suggest that low concentrations of both CLA9,11 and CLA10,12 have modest anti-inflammatory effects in ECs. Thus, CLAs may influence endothelial function and the risk of vascular disease. Nevertheless, at these low CLA concentrations some pro-inflammatory genes are upregulated while others are downregulated, suggesting complex effects of CLAs on inflammatory pathways.

Development of a whey-based beverage with enhanced levels of conjugated linoleic acid (CLA) as facilitated by endogenous walnut lipase

In this study, optimization of fermentation conditions, and applying endogenous walnut lipase were investigated for the manufacture of a fermented, whey-based beverage containing conjugated linoleic acid (CLA). Among different commercial starter and probiotic cultures, the culture containing Lactobacillus delbrueckii subsp. bulgaricus and Streptococcus thermophilus showed high potency for CLA synthesis. The fermentation time and the type of walnut oil (lipolyzed or non-lipolyzed) had significant effects on CLA production, as the highest CLA content (36 mg/g fat) was synthesized in the sample containing 1 % lipolyzed walnut oil fermented at 42 °C for 24 h. Moreover, fermentation time had the highest contribution on viable cell counts, proteolysis, DPPH scavenging activity, and final pH. A significant and positive correlation between cell counts and CLA content was also observed (r = +0.823, p < 0.05). This study establishes a cost effective approach for converting cheese whey to a value added beverage enriched with CLA.

Yerba mate (Ilex paraguariensis) as a source of antioxidants with soybean grain in supplementation of lactating ewes reared in tropical pastures.

The hypothesis tested is that the association of soybean grain and yerba mate as an antioxidant source can increase the concentration of polyunsaturated fatty acids (PUFA) with lower oxidation of milk from lactating ewes reared in tropical pastures. Sixteen ewes were randomly distributed in a 2x2 factorial scheme (with or without yerba mate at 110 g / kg of dry matter (DM); with or without soybean grain at 210 g/kg of DM). Yerba mate intake reduced DM intake, and concentrations of ether extract (EE), protein, lactose, defatted dry extract, density, omega-3 concentrations, and the ratio between saturated/monounsaturated fatty acids (FA), but increased the concentration of monounsaturated FA and the sequestering power of free radicals by DPPH without altering the concentrations of conjugated dienes and the concentrations of substances reactive to thiobarbituric acid in milk. Supplementation with soybean grain decreased the birth weight of the lambs and increased the concentrations of conjugated linoleic acid (CLA), PUFA, omega - 6, omega ratio - 6/omega - 3, and total blood cholesterol. The addition of yerba mate did not influence the oxidative profile of milk. Supplementation with soybean grain improved the FA profile of milk, increasing the concentrations of CLA and PUFA. The results suggest that the association of yerba mate and soybean grain helps to improve the fat quality of milk from ewes raised in tropical pastures, as shown by the enhanced antioxidant activity, although it does not help prevent oxidation of milk rich in PUFA.

Supplementation with Sunflower/Fish Oil-Containing Concentrates in a Grass-Based Beef Production System: Influence on Fatty Acid Composition, Gene Expression, Lipid and Colour Stability and Sensory Characteristics of Longissimus Muscle.

Beef contains an array of conjugated linoleic acid (CLA) isomers for which positive effects have been reported in animal models of human disease. The objectives were to develop a CLA-enriched beef production system and to assess its quality. Sixty Spring-born heifers were housed in Autumn and offered unwilted grass silage and a barley/soyabean concentrate or wilted grass silage and a concentrate containing sunflower oil and fish oil. In May, both groups were offered either pasture for 22 weeks, restricted pasture and sunflower oil and fish oil for 22 weeks, or pasture for 11 weeks and restricted pasture and sunflower oil and fish oil for the final 11 weeks. The predominant CLA isomer in beef was cis9, trans11 representing on average, 80% total CLA. The modified winter diet followed by supplementation for 22 weeks resulted in beef that had a CLA concentration that was higher, at a comparable intramuscular fatty acid concentration, than previously reported. The lipid and colour stability (over 10 days in modified atmosphere packaging) and sensory characteristics were generally not negatively affected. There were minor effects on the expression of candidate genes involved in lipid metabolism. Consumption of this beef would make a substantial contribution to the quantity of CLA suggested to have a positive effect on consumer health.

Fatty acid and conjugated linoleic acid content of Anatolian buffaloes at different muscle types and slaughter weight.

This research was performed to detect tissue fatty acid (FA) composition and conjugated linoleic acid (CLA) content in Anatolian buffaloes at different muscle types (MTs) and slaughter weights (SWs). The research was carried out on a private commercial livestock farm in Tokat. The research's animal material comprised 20 Anatolian buffalo calves with approximately 100kg body weights, weaned at about 150days of age.Before the experiment started, the calves were randomly dividedinto two slaughter groups (SW) as low weight (LW=225 kg) and high weight (HW=325 kg). Ten calves from each of the two experimental groups, which were intensively fed, were slaughtered in two different weights. After the slaughtering, FA composition and CLA content of the Semimembranosus (SM), Semitendinosus (ST), and Triceps brachii (TB) muscle tissues of the animals were examined. The study determined that palmitoleic acid (C16:1) and stearic acid (C18:0) were affected by MTs and oleic acid (C18:1) and α-linolenic acid (C18:3 n-3) were affected by SW (P < 0.05). The highest CLA was in the ST muscle type (0.298) and the LW group (0.289) of the SW groups (P > 0.05). With the increase of SW (in LW and HW groups), n-6/n3 (SM: 7.783 and 6.533; ST: 8.115 and 7.859; TB: 8.416 and 8.215) (P > 0.05) and PUFA ratio decreased (P < 0.05). The SW increase raised the SFA ratio in the SM muscle (P < 0.05) while lowering it in the TB muscle (P > 0.05). Again, with the increase in SW, AI and TI values increased in SM and ST muscles, while the same index values decreased in TB muscle (P > 0.05). In conclusion,when considering the PUFA/SFA ratio and the beneficial effects of CLAs on human health, ST in the MT and LW groups in SW, and thus ST and LW in MT and SW were prominent in Anatolian buffaloes.

Producing natural functional and low-carbon milk by regulating the diet of the cattle-The fatty acid associated rumen fermentation, biohydrogenation, and microorganism response.

Conjugated linoleic acid (CLA) has drawn significant attention in the last two decades for its various potent beneficial effects on human health, such as anticarcinogenic and antidiabetic properties. CLA could be generally found in ruminant products, such as milk. The amount of CLA in ruminant products mainly depends on the diet of the animals. In general, the fat content in the ruminant diet is low, and dietary fat supplementation can be provided to improve rumen activity and the fatty acid (FA) profile of meat and milk. Especially, dietary 18-carbon polyunsaturated FA (C18 PUFA), the dominant fat source for ruminants, can modify the milk FA profile and other components by regulating the ruminal microbial ecosystem. In particular, it can improve the CLA in milk, intensify the competition for metabolic hydrogen for propionate producing pathways and decrease methane formation in the rumen. Therefore, lipid supplementation appears to be a promising strategy to naturally increase the additional nutritional value of milk and contribute to lower methane emissions. Meanwhile, it is equally important to reveal the effects of dietary fat supplementation on rumen fermentation, biohydrogenation (BH) process, feed digestion, and microorganisms. Moreover, several bacterial species and strains have been considered to be affected by C18 PUFA or being involved in the process of lipolysis, BH, CLA, or methane emissions. However, no review so far has thoroughly summarized the effects of C18 PUFA supplementation on milk CLA concentration and methane emission from dairy cows and meanwhile taken into consideration the processes such as the microorganisms, digestibility, rumen fermentation, and BH of dairy cattle. Therefore, this review aims to provide an overview of existing knowledge of how dietary fat affects rumen microbiota and several metabolic processes, such as fermentation and BH, and therefore contributes to functional and low-carbon milk production.

Impact of safflower oil derived conjugated linoleic acid supplementation on fatty acids profile, lipolysis and sensory properties of cheddar cheese

ABSTRACT Nutritionists recommend to consume 3 g Conjugated linoleic acid (CLA) to prevent cancers, cardiovascular diseases, diabetes, obesity and other lifestyle-related disorders. To fulfill the daily requirements of CLA, large proportions of dairy and meat products have to be included in the dietary plan that may have negative impacts such as excessive intake of fat, cholesterol, and lactose. Current investigation was aimed to determine the effect of safflower oil derived CLA on fatty acids composition, lipolysis and sensory properties of cheddar cheese. Safflower oil-derived conjugated linoleic acid (CLA) was added to cheddar cheese at four levels i.e. 1%, 2%, 3% and 4% concentrations (T1, T2, T3 & T4), cheese without CLA supplement was used as control, vacuum packaged samples were ripened at 6–8°C for 90 days. Analysis of CLA supplement revealed that concentrations of ∆9c,11 t-18:2, ∆10 t,12c-18:2, ∆9c,11c-18:2, ∆9 t, 11c-18:2 were 52.16, 44.37, 0.27 and 0.55 mg/100 g. In control, concentrations of ∆9c,11 t-18:2 and ∆10 t,12c-18:2 was 0.24% and 0.07% with a total CLA content of 0.31%. In T3, concentrations of ∆9c,11 t-18:2, ∆10 t,12c-18:2, ∆9c,11c-18:2 and ∆9 t, 11c-18:2 were 1.12, 3.82, 1.63, 2.77 (total CLA 8.34mg100g). In T4, concentrations of ∆9c,11 t-18:2, ∆10 t,12c-18:2, ∆9c,11c-18:2 and ∆9 t, 11c-18:2, were 1.47, 3.50, 3.25 and 3.44% (total CLA 11.67 mg/100 g). Supplementation of cheddar cheese at all four levels significantly increased the concentrations of lactic acid, citric acid, propionic and acetic acids. After 90 days of ripening, free fatty acids in control, T1, T2, T3 and T4 were 0.14, 0.19, 0.23, 0.29 and 0.37% with no difference in composition and sensory properties. These results suggested that CLA content of cheddar cheese can be successfully increased with safflower oil-derived CLA supplement.

Effects of sucrose and sodium chloride pretreatments on cassava and plantain chips fried in CLA-enriched soybean oil: an analysis of acrylamide content, microstructure, and other physical properties

Abstract The purpose of this research was to evaluate the effect of soaking plantain and cassava chips in sucrose or sodium chloride (NaCl) on their physical properties and acrylamide content after frying in Conjugated Linoleic Acid (CLA)-enriched soybean oil. Plantain and cassava were cut into 1 mm thick slices and soaked for 20 min in a solution of 30% sucrose or 3% NaCl at 40 °C, separately. Soaking in sucrose and NaCl had no significant effect (p &lt; 0.05) on water loss, porosity, hardness, or microstructure. The acrylamide content ranged from 634 to 3177 µg/kg. The use of CLA-enriched oil had no significant effect (p &lt; 0.05) on the physical characteristics evaluated in this study. Frying reduced the CLA content in the chips from an initial 31.8% in the oil to 22.25 and 21.69% in plantain and cassava chips, respectively.

Physicochemical property optimization and nutrient redistribution in the muscle of sub-adult grass carp (Ctenopharyngodon idella) by conjugated linoleic acid.

We studied the effects of conjugated linoleic acid (CLA) on the amount of nutrients, flavour substances, and healthcare fatty acids, the physicochemical properties, and the potential molecular mechanisms in the muscles of sub-adult grass carp (Ctenopharyngodon idella). Fish were fed graded levels of CLA (0.0, 3.1, 6.4, 9.6, 12.7, and 15.9g/kg diets) for 60days. Protein, glutamic acid, alanine, inosine monophosphate (IMP), eicosapentaenoic acid (EPA; 20:5n-3), docosahexaenoic acid (DHA; 22:6n-3), and total CLA contents (p<0.05) increased in CLA 3.1∼12.7, 6.4∼9.6, 6.4∼9.6, 6.4∼15.9, 3.1∼9.6, 3.1∼9.6, and 3.1∼15.9g/kg diet, respectively (p<0.05). In addition, optimal CLA significantly increased pH24, shear force, collagen content, and myofibre density in the muscle (P<0.05); however, it decreased myofibre diameter (p<0.05). We concluded that 6-9g/kg CLA in the diet could improve the flesh quality of sub-adult grass carp.

Oilseed Supplementation Improves Milk Composition and Fatty Acid Profile of Cow Milk: A Meta-Analysis and Meta-Regression.

Lipids from oilseeds can be transferred to milk from cows or converted to other biomolecules with nutraceutical effects, resulting in healthier milk. However, there is a great uncertainty with regard to the effect of some variables related to the animal, the seed, the level of inclusion and the characteristics of the diet. The objective of this review was to show the effect of the inclusion of oilseeds in the diet of dairy cows on milk yield, milk components and the fatty acid profile in milk. A systematized search was carried out of published articles with high scientific rigor where the feeding strategy in dairy cows was the inclusion of oilseeds in the diet. Milk from oilseed-fed cows contained a higher amount of unsaturated 18-carbon fatty acids, including omega-3 series, rumenic and vaccenic fatty acids. Overall, supplementation with oilseeds in the cow’s diets increases the concentration of biomolecules in milk with potential positive effects on human health.Oilseed supplementation is a strategy to improve milk production and milk composition in dairy cows; however, the response to this approach is inconsistent. Thus, the aim of this study was to evaluate the effect of oilseed supplementation on milk production and milk composition in dairy cows via a meta-analysis and meta-regression. A comprehensive and structured search was performed using the following electronic databases: Google Scholar, Primo-UAEH and PubMed. The response variables were: milk yield (MY), atherogenic index (AI), Σ omega-3 PUFA, Σ omega-6 PUFA, fat, protein, lactose, linoleic acid (LA), linolenic acid (LNA), oleic acid (OA), vaccenic acid (VA), conjugated linoleic acid (CLA), unsaturated fatty acid (UFA) and saturated fatty acid (SFA) contents. The explanatory variables were breed, lactation stage (first, second, and third), oilseed type (linseed, soybean, rapeseed, cottonseed, and sunflower), way (whole, extruded, ground, and roasted), dietary inclusion level, difference of the LA, LNA, OA, forage and NDF of supplemented and control rations, washout period and experimental design. A meta-analysis was performed with the “meta” package of the statistical program R. A meta-regression analysis was applied to explore the sources of heretogeneity. The inclusion of oilseeds in dairy cow rations had a positive effect on CLA (+0.27 g 100 g−1 fatty acids (FA); p < 0.0001), VA (+1.03 g 100 g−1 FA; p < 0.0001), OA (+3.44 g 100 g−1 FA; p < 0.0001), LNA (+0.28 g 100 g−1 FA; p < 0.0001) and UFA (+8.32 g 100 g−1 FA; p < 0.0001), and negative effects on AI (−1.01; p < 0.0001), SFA (−6.51; p < 0.0001), fat milk (−0.11%; p < 0.001) and protein milk (−0.04%; p < 0.007). Fat content was affected by animal breed, lactation stage, type and processing of oilseed and dietary NDF and LA contents. CLA, LA, OA and UFA, desirable FA milk components, were affected by type, processing, and the intake of oilseed; additionally, the concentrations of CLA and VA are affected by washout and design. Oilseed supplementation in dairy cow rations has a positive effect on desirable milk components for human consumption. However, animal response to oilseed supplementation depends on explanatory variables related to experimental design, animal characteristics and the type of oilseed.

Effects of Grass-Fed Beef Suet and Liver Supplementation on Fatty Acid and Nutrient Profile of Chicken Eggs

ObjectivesThe nutrient composition of chicken eggs can be manipulated by altering the composition of layer hen feeds and adding supplements to chicken diets. Current nutrition guidelines recommend consuming diets with less saturated fatty acids (SFAs) and a lower omega-6: omega-3 (n-6: n-3) ratio to mitigate the risk of cardiovascular disease. There has been interest in utilizing the byproducts of grass-fed systems to reduce the SFA content and n-6: n-3 ratio of eggs in a sustainable manner. The objective of this study was to quantify and compare the FA and nutrient profile of pasture-raised chicken eggs supplemented with grass-fed beef suet and liver (n = 6) to pasture-raised chicken eggs fed a standard non-GMO layer hen feed (n = 6) and cage-free chicken eggs from a local grocery store (n = 6). MethodsEgg samples were collected and lyophilized prior to analysis. FA profiles were determined by microwave-assisted lipid extraction, acid-base methylation, and gas chromatography-mass spectrometry analysis. Total carotenoids were measured using UV-Vis spectrophotometry and total phenolic content was measured using a microplate reader and compared to a gallic acid standard. Vitamin and mineral content were assessed by a commercial laboratory. FA profiles of the layer hen feeds and the grass-fed beef suet and liver were also quantified. ResultsThere was no significant difference in total SFA among the three groups of eggs (P = 0.787). Conjugated linoleic acid (CLA) was significantly greater (P < 0.001) and the n-6: n-3 ratio was significantly lower (P < 0.001) in the chicken eggs supplemented with grass-fed beef suet and liver. Total carotenoid content was significantly greater in eggs supplemented with grass-fed beef suet and liver compared to the eggs from the grocery store (P < 0.001), but total phenolic content was significantly lower (P = 0.012). Vitamin B1, B3, B5, B12, and K1 content were significantly different among groups (P < 0.05). ConclusionsThese results indicate that supplementing chicken diets with grass-fed beef suet and liver increases the CLA content and decreases the n-6: n-3 ratio in eggs without influencing the SFA content. Effect of genotype, lactation and climatic factors on fatty acid profile of bovine milk

Milk fat from bovine milk contains fatty acids that may have favourable properties for human health, for example, conjugated linoleic acid (CLA) has nutraceutical activity. This research aimed to know the effects of genotype, days of lactation and climatic factors on the fatty acids (FA) profile of milk and particularly the content of CLA in milk fat. Seventeen first-calving milking cows in early lactation were used for the assessment of milk; 12 were Gyr and five were F1 (Holstein/Gyr) crosses. Sampling was carried out every 15 days, from the beginning to the end of lactation (300 days). Fatty acids were analyzed employing gas chromatography. The genotype did not influence the content of the fatty acid groups: saturated fatty acids (SFA), monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA) and CLA. The highest MUFA and PUFA contents were recorded at 100 days of lactation (32.334 and 3.553 g/100 g of FA, respectively), while SFA and CLA had their highest production at 200 days of lactation (63.238 and 1.378 g/100 g of FA, respectively). Regarding the climate, the highest temperature caused a decrease in the CLA content, because temperatures above 30 °C caused a decrease in the grazing time.

Antagonistic Effects of Conjugated Linoleic Acids of Lactobacillus casei against Foodborne Enterohemorrhagic Escherichia coli

Probiotics in fermented foods or commercially available supplements benefit the host by providing metabolites and peptides. The production of these metabolites varies with the available substrates or prebiotics present in the system and their concentration. In this study, 0.5% peanut flour (PF) was used to stimulate the growth and production of metabolites of wild-type Lactobacillus casei (LCwt) and compare with an engineered L. casei (LCCLA) capable of converting a higher amount of conjugated linoleic acid (CLA). The total extracellular metabolites present in the cell-free cultural supernatant (CFCS) of LCwt (without peanut), LCwt+PF (with peanut), and LCCLA were collected after 24 and 48 h of incubation, and their antagonistic activities against enterohemorrhagic Escherichia coli (EHEC EDL933) growth and pathogenesis were evaluated. All collected metabolites exhibited varying efficiency in restraining EHEC EDL933 growth, whereas supplementing a low concentration of CLA to the 48-h CFCS from LCwt showed augmented antagonism toward EHEC EDL933. A downregulation of key virulence genes was observed from metabolites collected at the 48-h time point. These observations indicate that the presence of metabolites in CFCSs-including CLA, which is produced by Lactobacillus and was identified by gas chromatography-mass spectrometry-plays a critical role. This study demonstrates the potential applicability of Lactobacillus-originated CLA in the prevention of EHEC EDL933-mediated illnesses.

Supplementation of Enriched Polyunsaturated Fatty Acids and CLA Cheese on High Fat Diet: Effects on Lipid Metabolism and Fat Profile.

Epidemiological studies have demonstrated a positive relationship between dietary fat intake and the onset of several metabolic diseases. This association is particularly evident in a diet rich in saturated fatty acids, typical of animal foods, such as dairy products. However, these foods are the main source of fatty acids with a proven nutraceutical effect, such as the ω-3 fatty acid α-linolenic acid (ALA) and the conjugated linoleic acid (CLA), which have demonstrated important roles in the prevention of various diseases. In the present study, the effect of a supplementation with cheese enriched with ω-3 fatty acids and CLA on the metabolism and lipid profiles of C57bl/6 mice was evaluated. In particular, the analyses were conducted on different tissues, such as liver, muscle, adipose tissue and brain, known for their susceptibility to the effects of dietary fats. Supplementing cheese enriched in CLA and ω-3 fats reduced the level of saturated fat and increased the content of CLA and ALA in all tissues considered, except for the brain. Furthermore, the consumption of this cheese resulted in a tissue-specific response in the expression levels of genes involved in lipid and mitochondrial metabolism. As regards genes involved in the inflammatory response, the consumption of enriched cheese resulted in a reduction in the expression of inflammatory genes in all tissues analyzed. Considering the effects that chronic inflammation associated with a high-calorie and high-fat diet (meta-inflammation) or aging (inflammaging) has on the onset of chronic degenerative diseases, these data could be of great interest as they indicate the feasibility of modulating inflammation (thus avoiding/delaying these pathologies) with a nutritional and non-pharmacological intervention.

Catalyst activation by cold plasma technology and its effect on isomerization of safflower seed oil

High voltage atmospheric cold plasma (HVACP) is a novel catalyst modification technology. In this study, a ruthenium/multi-walled carbon nanotubes (Ru/MWCNTs) catalyst was activated by HVACP instead of the thermal reduction method. The results showed that HVACP activation under the conditions of 180 s discharge time, 150 W discharge power, and 2.5 cm electrode spacing at 25 °C could effectively decrease the activation temperature and time. At this time, the Ru dispersion was 20.1% and the conjugated linoleic acid (CLA) yield was 43.0%. Catalyst characterization proved that the HVACP-treated catalyst did not aggregate and had a good activation effect. The catalyst was applied to the isomerization reaction of safflower seed oil to obtain CLA-rich safflower seed oil. The total CLA content was 43.02%, the trans-oleic acid content was 1.34%, the iodine value (IV) was 133.50 gI2/100 g, and after reusing five times, the relative activity of the catalyst remained at 77%, which provided the conditions for the preparation of functional oil.
