Expression Levels of Flowering Time Genes (CONZ1, GIGZ1A, GIGZ1B, FKF1A, and FKF1B) in Seedlings under Long-Day Conditions Differentiates Early and Late Zea mays L. Lines.

Phenophase durations, including the timing of flowering and ripeness, were characterized in 20inbred lines of the maize Zea mays L. Expression of key flowering initiation genes (CONZ1, GIGZ1a, GIGZ1b, ZmFKF1a, and ZmFKF1b) under long-photoperiod conditions was studied in seedlings of six maize lines that differed in ripeness time. Significantly lower transcription levels of all of the five genes were found in early-ripening lines compared with late-ripening lines. Similar expression patterns were observed for the GIGZ1a and GIGZ1b paralogous genes, while ZmFKF1a significantly predominated in expression over its paralog ZmFKF1b.

Genetic variation in four maturity genes and photoperiod insensitivity effects on the yield components and on the growth duration periods of soybean

Soybean (Glycine max (L.) Merr.) is a typical short-day and thermophilic crop. Absence of or low sensitivity to photoperiod is necessary for short-day crops to adapt to high latitudes. Photoperiod insensitivity in soybeans is controlled by two genetic systems and involves three important maturity genes: E1, a repressor for two soybean orthologs of Arabidopsis FLOWERING LOCUS T, and E3 and E4, which are phytochrome A genes. The aim of this work was to investigate the role of four maturity genes (E1 through E4) on the yield components, seed quality and on phasic development of near isogenic by E genes lines of soybean: short-day (SD) lines with genotype e1E2E3E4e5E7, e1E2E3e4e5E7, E1e2e3E4e5E7 and photoperiodic insensitive (PPI) lines with genotype e1e2E3E4e5E7, e1e2e3E4e5E7 under a long photoperiod (the natural day length of 50 latitude) conditions and short day conditions. The results of the study showed that soybean development processes under conditions of different day lengths depend on the dominant/recessive state of the main maturity genes. In addition, the response to the photoperiod depends on certain combinations of genes. SD lines began flowering on average 16.9% later under the conditions of a natural long photoperiod. Dominant alleles of genes E1 and E3 extended the pre- and post-flowering phases under conditions of exposure to long and short photoperiods. The dominant allele of the E1 gene delayed the onset of flowering by an average of 26.9%, and the period of full maturity by 39.8% compared to the recessive e1. The dominant allele of the E3 gene, compared to the recessive e3, lengthened the transition to flowering by an average of 16.1%, and the period of full ripeness by 27.1%. The dominant allele of the E2 gene lengthened the duration of the vegetative phase by 20% under the conditions of a long photoperiod. No significant influence of the dominant E4 allele on the duration of the vegetative and generative phases of soybean development was found in our study. PPI lines begin flowering under the conditions of a long and short photoperiod at the same time, but the phases of flowering and full seed maturity in the line with genotype e1e2e3E4e5E7 occurred earlier, due to the loss of the photoperiod sensitivity of the E3 gene. PPI line with genotype e1e2e3E4e5E7 proved to be the most insensitive line to the effect of different photoperiod durations among the studied lines. It was shown that the dominant alleles of E1–E4 maturity genes reduced the parameters of seed weight per plant and the weight of 1000 seeds under the conditions of a natural long photoperiod in comparison with recessive alleles of these genes. The maximum weight of seeds per plant and the weight of 1000 seeds were recorded in the PPI line with genotype e1e2e3E4e5E7. It should be noted that the dominant alleles E1 and E3 increased yield under conditions of a short photoperiod. Maturity genes had different effects on the biochemical composition of seeds. It was shown that soybean lines with dominant E1, E2 and E4 genes showed a higher content of starch and a lower content of total nitrogen and oil in seeds under natural photoperiod conditions compared to lines with recessive alleles of these genes. The dominant E3 allele reduced the oil content and did not affect the starch and total nitrogen content of seeds under long day conditions compared to the recessive e3 allele. The analysis of the effect of photoperiod on the timing of phenophases, yield structure indicators and biochemical composition of seeds in soybean plants with different sensitivity to photoperiod showed that the PPI line with the genotype e1e2e3E4e5E7 was the most adapted to the natural conditions of 50 degrees latitude. The PPI line with the genotype e1e2e3E4e5E7 was characterized by the shortest phases of days from sowing to flowering and full maturity. As a result, this line had the shortest growing season without reducing the yield and seed quality. Clearly, photoperiod had strong effects on all stages of plant reproduction and often acted indirectly, as shown by delayed responses expressed in later phases of development. The obtained results can be useful for the selection of soybean cultivars adapted to the climatic conditions of cultivation of Kharkiv region.

Growth analysis of the everbearing strawberry ‘Delizzimo’ under controlled temperature and photoperiod conditions

BackgroundThere is limited information on the effect of environment on vegetative growth in everbearing (EB) strawberry (Fragaria x ananassa Duch.) and its comparison with the situation in seasonal flowering types.MethodsWe investigated the effects of photoperiod (daylengths of 10 and 20 h) and temperature (12, 19 and 26 ℃) on leaf growth, dry matter production and partitioning, concentrations of soluble sugars, starch, and chlorophyll in the F1 hybrid ‘Delizzimo’ grown in a single experiment in daylight phytotron compartments in Norway.ResultsPlants grown in the long photoperiod (LD) and higher temperatures had greater leaf growth and higher dry matter production than those under short day (SD) and low temperature conditions. Growth decreased over the 39 days of the experiment. The changes in growth in the different environments were associated with changes in relative growth rate (RGR) and these were driven by changes in net assimilation rate (NAR) and leaf area ratio (LAR). The plants directed more dry matter to the leaves and crowns under LD and high temperature conditions and less dry matter to the roots, thus increasing the plant’s shoot to root ratio. Long days decreased the concentrations of sugars and starch in most of the tissues, while the effect of temperature was more complex. Higher temperatures increased the concentrations of sugars in the leaves in LD, while starch accumulated in the roots under SD and low temperature conditions. Sucrose accumulated temporarily in the crowns at the time of flower bud formation in LD and higher temperatures.ConclusionsThe results of the experiment demonstrate that the effects of photoperiod and temperature on the vegetative growth of everbearing strawberry are similar to those reported for seasonal-flowering strawberry. Increases in temperature and photoperiod and the resulting enhancement of the RGR was associated with accumulation of soluble sugars (sucrose, glucose and fructose) in the above-ground parts of the plant, whereas low temperature and SD resulted in accumulation of starch in the roots.

Annual patterns of ocular melatonin level in the female grass puffer, Takifugu alboplumbeus: possible involvement in seasonal reproductive response.

The aim of this study was to investigate the expression patterns of ocular melatonin in the annual reproductive cycle of the female grass puffer. Spawning season of the female grass puffer is from June to July in Jeju, South Korea. Time-resolved fluoroimmunoassay revealed that levels of ocular melatonin, which show an annual change, peaked in May (spawning season). Additionally, expression of reproductive-related genes also showed annual patterns: GnRH1 peaked in August, GnRH2 peaked in February, GnRH3, Kiss2, and LPXRFa peaked in November. These results suggest that ocular melatonin may be related to the annual reproductive cycle in the grass puffer. To better understand the photic regulation of AANAT1a mRNA in the retina, we observed the nocturnal pattern of ocular melatonin levels daily, which shows a nocturnal pattern in both short photoperiod (SD) and long photoperiod (LD) conditions. In the brain, AANAT2 mRNA also shows a nocturnal pattern in both SD and LD; however, the time of peak expression of AANAT2 mRNA was unchanged in both conditions. Following intraperitoneal injection of melatonin for 2weeks, expression of GnRH2 and LPXRFa mRNA in the brain significantly increased, while that of Kiss2 mRNA was decreased, suggesting that melatonin has a reproduction-related effect. Furthermore, under SD and LD conditions for 14weeks, the gonadosomatic index more increased and the maturity of the ovary progressed under LD compared with those under SD, suggesting that the SD photoperiodic signal inactivated ovarian development. These results indicate that the ocular melatonin may have a possible role in the reproductive endocrinology of the grass puffer.

Integrating transcriptomic and proteomic analyses of photoperiod-sensitive in near isogenic maize line under long-day conditions

As a short-day plant species, maize requires an optimal photoperiod for inducing reproductive growth. However, there is a lack of information regarding photoperiod-induced changes in maize mRNA and protein levels. In this study, a photoperiod-insensitive maize inbred line and its near isogenic photoperiod-sensitive line were used. By integrating RNA-based transcriptomic and iTRAQ LC-MS/MS-based proteomic approaches, we generated a comprehensive inventory of the transcripts and proteins with altered abundances in response to a long photoperiod (LP) during growth stage transitions. We detected 22 000 transcripts in RNA-sequence runs and 5 259 proteins from an iTRAQ-based analysis. A weak correlation between mRNA- and protein-level changes was observed, suggesting the LP-induced transition between maize growth stages is largely regulated post-transcriptionally. Differentially expressed genes influenced by LP conditions were associated with several regulatory processes in both maize inbred lines, especially phosphate ion transport and the circadian rhythm. Additionally, 31 transcripts and six proteins related to photoperiodic flowering in maize were identified by comparing transcriptomic and proteomic data. This transcriptomic and proteomic analysis represents the first comprehensive and comparative study of gene/protein-level changes occurring in photoperiod-sensitive and -insensitive maize inbred lines during growth stage transitions under LP conditions.

Age-related and photoperiodic variation of the DAZ gene family in the testis of the Syrian hamster (Mesocricetus auratus).

SummaryThe Deleted in AZoospermia (DAZ) gene family regulates the development, maturation and maintenance of germ cells and spermatogenesis in mammals. The DAZ family consists of two autosomal genes, Boule and Dazl (Daz-like), and the Daz gene on chromosome Y. The aim of this study was to analyze the localization of DAZL and BOULE during testicular ontogeny of the seasonal-breeding Syrian hamster, Mesocricetus auratus. We also evaluated the testicular expression of DAZ family genes under short- or long-photoperiod conditions. In the pre-pubertal and adult testis, DAZL protein was found mainly in spermatogonia. BOULE was found in the spermatogonia from 20 days of age and during the pre-pubertal and adult period it was also detected in spermatocytes and round spermatids. DAZL and BOULE expression in spermatogonia was strictly nuclear only in 20-day-old hamsters. We also detected the novel mRNA and protein expression of BOULE in Leydig cells. In adult hamsters, Dazl expression was increased in regressed testis compared with non-regressed testis and DAZL protein expression was restricted to primary spermatocytes in regressed testis. These results show that DAZL and BOULE are expressed in spermatogonia at early stages in the Syrian hamster, then both proteins translocate to the cytoplasm when meiosis starts. In the adult regressed testis, the absence of DAZL in spermatogonia might be related to the decrease in germ cell number, suggesting that DAZ gene family expression is involved in changes in seminiferous epithelium during photoregression.

Diurnal and photoperiodic changes in thyrotrophin-stimulating hormone β expression and associated regulation of deiodinase enzymes (DIO2, DIO3) in the female juvenile chicken hypothalamus.

Increased thyrotrophin‐stimulating hormone β (TSHβ) expression in the pars tuberalis is assumed to be an early step in the neuroendocrine mechanism transducing photoperiodic information. The present study aimed to determine the relationship between long‐photoperiod (LP) and diurnal TSHβ gene expression in the juvenile chicken by comparing LP‐photostimulated birds with groups kept on a short photoperiod (SP) for 1 or 12 days. TSHβ expression increased by 3‐ and 23‐fold after 1 and 12 days of LP‐photostimulation both during the day and at night. Under both SP and LP conditions, TSHβ expression was between 3‐ and 14‐fold higher at night than in the day, suggesting that TSHβ expression cycles in a diurnal pattern irrespective of photoperiod. The ratio of DIO2/3 was decreased on LPs, consequent to changes in DIO3 expression, although there was no evidence of any diurnal effect on DIO2 or DIO3 expression. Plasma prolactin concentrations revealed both an effect of LPs and time‐of‐day. Thus, TSHβ expression changes in a dynamic fashion both diurnally and in response to photoperiod.

The Pattern of Entrainment of the Human Sleep-Wake Rhythm by the Natural Photoperiod in the North

Recently, it was shown that the sleep-wake rhythm of the inhabitants of the temperate zone is entrained to sun time. In the North, significant seasonal changes in the photoperiod may interfere with entrainment of the circadian system to sunlight. This investigation assessed the influence of photoperiod characteristics on the sleep length and sleep-wake rhythm of residents of high latitude. The study was conducted in four towns and six villages located between 59.5°N and 67.6°N latitude between the months of October and May from 2009 through 2011 and included 2822 subjects aged 10 to 97 yrs, 1621 of whom were females and 1201 males. The chronotype and sleep length of the subjects were assessed using the Munich Chronotype Questionnaire. The instructions for the questionnaire stressed the need to specify the sleep-wake schedule during the week preceding the date of completing the questionnaire. The study found that the length of sleep and the chronotype of the inhabitants surveyed in Northern European Russia depend on age, sex, type of settlement, and place of residence. The time of sunrise was a stronger predictor of sleep length and chronotype than the time of sunset and day length. A later chronotype and shorter sleep length were found for the subjects during the equinox (sunrise at 06:00 h) than under long-photoperiod conditions (sunrise at 04:00–05:00 h). During short-photoperiod conditions (sunrise at 07:00–10:00 h), no significant changes in the self-reported sleep-wake rhythm were found. The time of sunrise had the strongest impact on the sleep-wake rhythm of 30- to 97-yr-old persons. Sunrise had a stronger influence on chronotype and sleep length in January to May, when the days become longer, than in October to December, when the days become shorter. Reorganization of the Suprachiasmatic Nucleus Coding for Day Length

In mammals, the suprachiasmatic nucleus (SCN), the circadian pacemaker, receives light information via the retina and functions in the entrainment of circadian rhythms and in phasing the seasonal responses of behavioral and physiological functions. To better understand photoperiod-related alterations in the SCN physiology, we analyzed the clock gene expression in the mouse SCN by performing in situ hybridization and real-time monitoring of the mPer1::luc bioluminescence. Under long photoperiod (LP) conditions, the expression rhythms of mPer1 and Bmal1 in the caudal SCN phase-led those in the rostral SCN; further, within the middle SCN, the rhythms in the ventrolateral (VL)-like subdivision advanced compared with those in the dorsomedial (DM)-like subdivision. The mPer1::luc rhythms in the entire coronal slice obtained from the middle SCN exhibited 2 peaks with a wide peak width under LP conditions. Imaging analysis of the mPer1::luc rhythms in several subdivisions of the rostral, middle, caudal, and horizontal SCN revealed wide regional variations in the peak time in the rostral half of the SCN under LP conditions. These variations were not due to alterations in the waveform of a single SCN neuronal rhythm. Our results indicate that LP conditions induce phase changes in the rhythms in multiple regions in the rostral half of the SCN; this leads to different circadian waveforms in the entire SCN, coding for day length.

Reorganization of the suprachiasmatic nucleus by photic and non-photic environments

To better understand alterations in suprachiasmatic nucleus (SCN) physiology by photic and non-photic environments, we analyzed clock gene expression in the mouse SCN by in situ hybridization and real-time monitoring of mPer1::luc bioluminescence. An artificial light: dark: light: dark 7:5:7:5 condition induced antiphase oscillation of the clock gene in the dorsomedial-like (DM-like) and the ventrolateral-like (VL-like) subdivisions of the SCN, in parallel with maintaining synchronization between the two sides of the SCN. This antiphase oscillation seems to be responsible for the bimodal rhythms of mPer1 bioluminescence observed in the entire SCN and the components of splitting locomotor activity. A long photoperiod (LP) induced the phase advance of the rhythms of clock gene expression in the caudal SCN relative to those in the rostral SCN. In addition, in the middle SCN, the rhythms of the VL-like subdivision phase-led those in the DM-like subdivision. The mPer1::luc rhythms in the entire coronal slice obtained from the middle SCN exhibited two peaks with a wide peak width under LP conditions. Imaging analysis of the mPer1::luc rhythms revealed wide regional variations in the peak time in the rostral half of the SCN. These variations were not due to alterations in the waveform of a single SCN neuronal rhythm. In CS mice which easily entrain to daily restricted feeding (RF) cycles, the peak width for mPer1::luc bioluminescence rhythms in the SCN markedly increased during the RF schedule under LD conditions. These data in CS mice suggest non-photic cues alter SCN organization and provided a new method to reveal subdivisional changes of the SCN by feeding cues.

Distribution and ecology of Pseudo-nitzschia species (Bacillariophyceae) in surface waters of the Weddell Sea (Antarctica)

The distribution of six Pseudo-nitzschia species and their relationship with environmental conditions were studied for the first time in a vast zone of the Weddell Sea (∼61–77°S, Antarctica). Both qualitative and quantitative phytoplankton samples, collected during summer 2004, were examined using light and scanning electron microscopy. Phytoplankton abundance and composition showed great variability along our study area. Diatoms were the most conspicuous phytoplankton group in the northern area while small flagellates were generally dominant in the southern stations. The genus Pseudo-nitzschia was broadly distributed and significantly contributed to total diatom densities. A marked contrast in Pseudo-nitzschia species distribution was observed in three main zones divided by the Weddell Front (WF) and the Antarctic Slope Front (ASF). P. subcurvata and P. turgiduloides were the most abundant species in the neritic Weddell Sea zone, south of the ASF, mainly near the ice-edge in shallower waters and in conditions of long photoperiod. In contrast, P. prolongatoides and P. lineola dominated north of the ASF; the first was associated with deeper and nutrient-rich waters whereas the latter showed a weak relation with environmental variables examined. Finally, P. turgidula and P. heimii were mostly observed in the Weddell–Scotia Confluence Zone in the warmest and far from ice covered waters, north of the WF. A brief morphological Pseudo-nitzschia species description is given in the Appendix, including morphometrics and pictures.

Are the annual reproductive and body weight rhythms in the male European hamster (Cricetus cricetus) dependent upon a photoperiodically entrained circannual clock?

Most of the data obtained so far on the European hamster (Cricetus cricetus) suggest direct photoperiodically driven seasonal changes in sexual activity and body weight. The results of the present long-term study support the hypothesis that these annual changes are the expression of photoperiodically driven endogenous circannual rhythms. When subjected following capture (April-May) to constant conditions of long photoperiod (LP) and constant temperature a large number of the European hamsters present, in September-December, complete gonadal atrophy associated with a decrease in body weight. A sexual reactivation as well as an increase in body weight are observed in the same animals between January and April. Of the six animals that survived long enough, two only presented partial gonadal atrophy during the second year. These observations clearly demonstrate that the decline in sexual activity in subjective autumn does not require a decrease in photoperiod, at least in the first year. Theoretically, the observed rhythms, if circannual in nature, would be generated by a self-sustained annual oscillator (circannual clock) able to function in the absence of a photoperiodic input. Pinealectomy makes animals unable to detect or measure photoperiodic information. Of the six European hamsters tested (pinealectomized in June and then kept continuously under LP), five showed clear annual rhythms in body weight and reproductive capacities for two consecutive years. Clearly endogenous annual rhythms were expressed in these conditions. To be entrained to a 1-year period, such a circannual clock should, however, be able to react to either LP and/or to short-photoperiod (SP), at least at certain periods of the annual cycle. In animals exposed to LP in August or October, after gonadal atrophy had been established by exposure to natural SP, gonadal regrowth started in December or January, about 2 to 3 months earlier than in animals kept outside or in experimental SP. With the same experimental conditions, exactly the same results were obtained in pinealectomized animals; thus stimulatory effect of LP or LP-induced phase advance of the circannual clock can be excluded. The absence of the SP information would then induce such reaction. In animals kept under constant LP and temperature following capture, however, pinealectomy in January--when all animals are sexually active--induces gonadal atrophy within--weeks. This clearly demonstrates that LP is stimulatory at this time of the subjective year.

Photoperiodic regulation of body mass, food intake, hibernation, and reproduction in intact and castrated male European hamsters, Cricetus cricetus.

A group of sexually active male European hamsters were raised either in short-photoperiod conditions (SP; LD 8:16) or in long-photoperiod conditions (LP; LD 16:8) from their capture at the end of the hibernation period. Another group of hamsters was castrated in April and gonadectomized animals were maintained in SP and cold (7 degrees C) or in a succession of SP and LP plus cold. Another group, castrated in May or in September and raised in LP conditions, was transferred in September to SP conditions and cold. 1. Normal hamsters raised in continuous SP or LP apparently did not show signs of rhythmic behavior, except possibly in gonadal activity. 2. Body weight increased continuously, plasma testosterone levels oscillated between 1.5 and 2.5 ng/ml, and animals raised in SP and in cold did not enter hibernation. 3. Similar results were also found in castrated animals kept in SP conditions and cold. 4. The sequence LP-SP induced a decrease in food intake and body weight and a decrease in plasma testosterone levels and triggered entry into hibernation in both intact and castrated animals. 5. After 6 months continuously in SP and with exposure to cold spontaneous recrudescence in food intake and body weight occurred and hibernation ended in both intact and castrated animals. 6. In normal animals a spontaneous increase in plasma testosterone levels was observed. 7. In both normal and gonadectomized animals the phase of refractoriness could be broken by exposure to LP conditions. 8. The critical photoperiod lies between 15 and 15.5 h. Variations in the photoperiodic reaction controlling diapause induction in Lucilia caesar L. (Diptera: Calliphoridae).

Two strains of the blowfly Lucilia caesar L. were established in the laboratory, one producing a low incidence and the other a high incidence of diapause under the normally diapause-preventing conditions of long photoperiod. Crossbreeding was carried out between the strains, with the resulting hybrids having diapause performances intermediate between those of the parents. Little or no filial segregation occurred in two hybrid generations, indicating the multiple-gene condition of diapause inheritance. The genetic influence on diapause control is apparently enhanced by sexual characteristics since the male enters diapause more readily than the female in this species.

EXPERIMENTS ON PHOTOPERIOD IN RELATION TO THE VEGETATIVE GROWTH OF PLANTS

It has long been known that relative length of day and night effects the vegetative as well as the reproductive activity of plants. Since the appearance of the paper of Garner and Allard (7) which first dealt with this subject, abundant factual material has accumulated relating to the influence of photoperiod on such varied processes as shoot elongation, leaf size, accumulation of dry weight and root growth (6, 10). It has been frequently, although not invariably, found that vegetative growth as measured by one or more of these criteria, is greater in long days than in short days irrespective of the reproductive behavior of the species in question. The promotive effect of long daily photoperiods on vegetative growth does not appear to be directly attributable to increased duration of C02 assimilation (11). Plants grown under conditions of long day made up of a short photoperiod of natural light supplemented by light of exceedingly low intensity exhibit the vegetative vigor typical of plants on a long day rather than that of plants on a short day regime (13). It is known that the reproductive response of photoperiod sensitive plants is to be attributed to the production of a specific hormone (or hormones) in the leaves under conditions of suitable day length (8, 9). The production of this particular hormone or hormones would appear to depend on the length of day to which the leaves are subjected. It seems logical to inquire in how far the vegetative responses of plants to varying photoperiods are also attributable to the influence of day length on the production of the several hormones which regulate vegetative growth. In the present paper evidence will be presented to show that the production of vitamin Bi, a hormone for the growth of roots (1), is affected by the length of day to which the plant is subjected. It is known that length of day exercises a profound effect on the extent of the root system in many species of plants. Thus Weaver and Himmel (12) found that the root system is invariably more extensive under conditions of long photoperiod irrespective of whether the species of plant is long day, short day, or indeterminate with regard to flowering behavior. Crist and Stout (5) have also shown that the root system of plants grown under short photoperiod may be several times smaller than the root systems of similar plants grown under conditions of long photoperiod. Photoperiodism in Relation to Hormones as Factors in Floral Initiation and Development

1. A description is given of a simple method whereby one portion of a plant may be subjected to one photoperiod while another portion of the same plant is being subjected to another photoperiod. 2.
