Progress of highly reproducible capillary electrophoresis

Following rapid developments in capillary electrophoresis (CE), this technology has become an established analytical technique owing to its microscale characteristics, high speed, high efficiency, and versatility. However, the challenges of poor peak stability and/or reproducibility have consistently hindered its wider applications. CE has long been used as a measurement tool for plotting signal intensities versus the migration time; however, the migration time is not an independent variable in CE, but is affected by many direct and indirect parameters, including capillary (length, diameter, and inner surface properties), electric field (or voltage, current, and/or power), temperature, and running buffer (electrolytes, additives, solvents, and their concentration, buffering pH, etc.). These intricacies render the acquisition of reproducible electropherograms difficult. Various studies ranging from those on the early stages of CE development to those on the exploration of three important strategies have been conducted to address this issue. In the first strategy, the CE conditions, especially those parameters that can maintain a stable electro-osmotic flow, are strictly controlled and stabilized to significantly improve peak repeatability. In the second strategy, either the peak position is corrected using internal standards or the peak time is converted into other variables, such as electrophoretic mobility, to offset or eliminate some unstable factors, thereby improving the repeatability and even reproducibility of the peaks; this strategy is useful when plotting signals versus the migration time ratio, correlated migration time, effective mobility, or temperature-correlated mobility. In the third strategy, a new methodology called highly reproducible CE (HRCE) is established using theoretical studies to explore better principles for real-time CE with the aim of the complete removal of the challenge from the root. This strategy includes the development of novel methods that plot electropherograms based on weighted mobility, migrated charge, charge density, or partial differential molar charge density. Similar to ordinary CE approaches, this strategy can also draw electropherograms based on the ratios of these properties. As theoretically predicted, these novel methods can offset or resist changes in critical CE conditions (mainly electric field strength, capillary length and diameter, and/or some buffer parameters such as concentration). Our experimental results demonstrate that given certain prerequisites, a new set of methods can produce highly reproducible electropherograms. This review focuses on the theoretical basis and advancements of HRCE, and elucidates the link between electrophoretic migration/peak expression theories and their impact on reproducibility. Studies on the transformation of time-scale electropherograms in the CE literature are summarized and analyzed in general. However, this review does not directly discuss research on and progress in improving CE repeatability or reproducibility through instrument upgrades, parameter optimization, or practical method refinements.

Affinity Capillary Electrophoresis as a Tool To Characterize Molecularly Imprinted Nanogels in Solution.

In this work, an innovative and accurate affinity capillary electrophoresis (ACE) method was set up to monitor the complexation of aqueous MIP nanogels (NGs) with model cancer-related antigens. Using α2,6'- and α2,3'-sialyllactose as oversimplified cancer biomarker-mimicking templates, NGs were synthesized and characterized in terms of size, polydispersity, and overall charge. A stability study was also carried out in order to select the best storage conditions and to ensure product quality. After optimization of capillary electrophoresis conditions, injection of MIP NGs resulted in a single, sharp, and efficient peak. The mobility shift approach was applied to quantitatively estimate binding affinity, in this case resulting in an association constant of K ≈ 106 M-1. The optimized polymers further displayed a pronounced discrimination between the two sialylated sugars. The newly developed ACE protocol has the potential to become a very effective method for nonconstrained affinity screening of NG in solution, especially during the NG development phase and/or for a final accurate quantitation of the observed binding.

Dual optical detection approach for capillary electrophoresis following two-step liquid-liquid extraction to determine ten phenols in water samples

In this research, the analytical method was developed and evaluated for determining phenol and its nine derivatives belong to the US EPA priority pollutant list in water samples by using dual-channeled capillary electrophoresis (CE) coupled with two types of optical detectors, namely LED-induced fluorescence (LEDIF) and ultraviolet (UV) detectors. The optimal background electrolytes for the first and second CE channels were 20 mM borate (pH 9.80) with 400 µM fluorescein and 55 mM borate (pH 11.75), respectively. The two-step liquid-liquid extraction (LLE) was used for sample preparation and enrichment, in which phenol and its derivatives were extracted from the aqueous phase using 10 mL of n-hexane/1-octanol (60/40, v/v) and then were back extracted into a 0.1 M NaOH as a final acceptor phase. Under the optimal CE and two-step LLE conditions, the enrichment factors of 10 phenols were 184 – 1120-fold, and the method detection limits were lowered to 0.02–0.60 µg/L. The obtained intra-day and inter-day precisions in terms of relative standard deviations (RSD) were between 4.0 and 7.3 % and 6.7 and 14 %, respectively. This approach was used to determine phenols in water samples, with recoveries ranging from 82.0 to 108.9 %. In combination with sample enrichment by two-step LLE extraction, this is the first CE study conducted to determine phenols in the EPA list using two detector approaches, specifically CE-LEDIF/CE-UV.

Carboxymethylated maltodextrin as a chiral selector for the separation of some basic drug enantiomers using capillary electrophoresis

In this work, carboxymethylated maltodextrin (Cm-MD) was successfully synthesized as an efficient anionic chiral selector and applied for the enantiomer separation of some basic drugs including tramadol, venlafaxine, verapamil, hydroxyzine, citalopram, fluoxetine, and amlodipine by capillary electrophoresis (CE). The synthesized chiral selector was characterized by the nuclear magnetic resonance and Fourier transform infrared spectrophotometry. Under the optimized Cm-MD modified CE conditions (background electrolyte: phosphate buffer (pH 5.0, 50 mM) containing 5% (w/v) Cm-MD; applied voltage: 20 kV; and capillary column temperature: 25 °C), successful enantiomer separation of all studied chiral drugs were observed. By comparison of Cm-MD and MD for enantiomer separation of the model drugs, it was revealed that Cm-MD exhibits a higher resolution in comparison to the MD modified CE. This enhanced resolution could be attributed to the electrostatic interactions between the cationic drugs and anionic Cm-MD and opposite direction mobility of the host-guest complex relative to the chiral analyte. The optimized Cm-MD modified CE method was successfully used for the assay of the enantiomers of citalopram and venlafaxine in commercial tablets. The proposed method showed the linear range of 5.0–150.0 mg/L and 10.0–150.0 mg/L for both enantiomers of citalopram and venlafaxine, respectively. The limits of quantification were 5.0 and 10.0 mg/L for the enantiomers of citalopram and venlafaxine, respectively. The limit of detection for all enantiomers was found to be < 3.0 mg/L. Intra- and inter-day RSDs (n = 4) were less than 9.7%. The relative errors were less than 9.4% for all enantiomers. The obtained results in this research show that Cm-MD as a new, efficient and inexpensive chiral selector can be used for enantiomer separation of basic drugs using the CE technique.

Multilayer Coatings Based On Citrate-Stabilized Gold Nanoparticles and Polydiallyldimethylammonium Chloride for the Electrophoretic Separation of Carboxylic Acids

The conditions for the formation of physically adsorbed three-layer coatings of quartz capillary walls in capillary electrophoresis (CE) with successively deposited oppositely charged layers of polydiallyldimethylammonium chloride (PDADMAC) modifiers and citrate-stabilized gold nanoparticles (GNPs) are proposed. It was shown that three-layer PDADMAC–GNP–PDADMAC coatings favorably differ from monolayer coatings with PDADMAC by greater stability in a wide range of pH (2–10). The formed coatings were characterized by scanning electron microscopy, and the presence of a uniform dense layer of nanoparticles on the capillary surface was confirmed. The applicability of the modified capillaries under CE conditions was demonstrated by the separation of a mixture of 16 carboxylic acids. An increase in the separation selectivity achieved with the use of three-layer coatings based on GNPs was explained by the reversible exchange of citrate anions on the GNP surface with negatively charged analytes in the course of electrophoretic analysis.

Offline Coupling of Asymmetrical Flow Field-Flow Fractionation and Capillary Electrophoresis for Separation of Extracellular Vesicles.

Extracellular vesicles (EVs) play important roles in cell-to-cell communications and carry high potential as markers targeted in disease diagnosis, prognosis, and therapeutic development. The main obstacles to EV study are their high heterogeneity; low amounts present in samples; and physical similarity to the abundant, interfering matrix components. Multiple rounds of separation and purification are often needed prior to EV characterization and function assessment. Herein, we report the offline coupling of asymmetrical flow field-flow fractionation (AF4) and capillary electrophoresis (CE) for EV analysis. While AF4 provides gentle and fast EV separation by size, CE resolves EVs from contaminants with similar sizes but different surface charges. Employing Western Blotting, ELISA, and SEM, we confirmed that intact EVs were eluted within a stable time window under the optimal AF4 and CE conditions. We also proved that EVs could be resolved from free proteins and high-density lipoproteins by AF4 and be further separated from the low-density lipoproteins co-eluted in AF4. The effectiveness of the coupled AF4-CE system in EV analysis was demonstrated by monitoring the changes in EV secretion from cells and by direct injection of human serum and detection of serum EVs. We believe that coupling AF4 and CE can provide rapid EV quantification in biological samples with much reduced matrix interference and be valuable for the study of total EVs and EV subpopulations produced by cells or present in clinical samples.

Multivariate optimization of large-volume sample stacking with polarity switching by capillary electrophoresis for determination of gold nanoparticle size

Gold nanoparticles (AuNPs) are widely studied and applied due to their unique optical characteristics, such as surface plasmon resonance and extinction coefficient. In this study, large sample stacking combined with polarity switching (LVSS-PS) by capillary electrophoresis was established to determine AuNP mixtures with different particle sizes simultaneously. The multivariate experimental design containing fractional factorial design (fFD) and central composite design (CCD) was combined with LVSS-PS to estimate the optimal condition. The influence of factors on the response was evaluated by the use of the chromatography resolution statistic function to simultaneously investigate the resolution and separation under twenty-eight sets of capillary electrophoresis conditions. The results showed that the optimal condition of the separation buffer was 100 mM CAPS (pH 9.5) with 20 mM SDBS and 35 mM SDS. The separation temperature was set at 20 °C. Under this condition, four different diameters of AuNPs (5, 20, 50, and 80 nm) were successfully separated in the same run. The linear relationship between the electrophoretic mobility of AuNPs and their diameters was established, and the correlation coefficient was 0.99. Sodium citrate and gallic acid (GA) were capping agents to synthesize AuNPs. The established method was applied to determine the size of gold nanoparticles synthesized by sodium citrate and gallic acid as capping agents, respectively. The diameters of the AuNPs obtained by LVSS-PS are consistent with transmission electron microscopy.

Quantitative Characterization of Partitioning in Selection of DNA Aptamers for Protein Targets by Capillary Electrophoresis.

Partitioning of protein-DNA complexes from protein-unbound DNA is a key step in selection of DNA aptamers. Conceptually, the partitioning step is characterized by two parameters: transmittance for protein-bound DNA (binders) and transmittance for unbound DNA (nonbinders). Here, we present the first study to reveal how these transmittances depend on experimental conditions; such studies are pivotal to the effective planning and control of selection. Our focus was capillary electrophoresis (CE), which is a partitioning approach of high efficiency. By combining a theoretical model and experimental data, we evaluated the dependence of transmittances of binders and nonbinders on the molecular weight of the protein target in two modes of CE-based partitioning: nonequilibrium capillary electrophoresis of equilibrium mixtures (NECEEM) and ideal-filter capillary electrophoresis (IFCE). Our data suggest that as the molecular weight of the protein target decreases: (i) the transmittance for binders remains close to unity in NECEEM but decreases drastically in IFCE and (ii) the transmittance for nonbinders increases orders of magnitude in NECEEM but remains relatively stable at a very low level in IFCE. To determine the optimal CE conditions for a given size of protein target, a balance between transmittances of binders and nonbinders must be reached; such a balance would ensure the collection of binders of sufficient purity and quantity. We conclude that, as a rule of thumb, IFCE is preferable for large-size protein targets while NECEEM should be the method of choice for small-size protein targets.

Capillary Electrophoresis-Mass Spectrometry (CE-MS) by Sheath-Flow Nanospray Interface and Its Use in Biopharmaceutical Applications.

Both capillary electrophoresis (CE) and mass spectrometry (MS) technologies are powerful analytical tools that have been used extensively in the characterization of biologics in the biopharmaceutical industry. The direct coupling of CE with MS is an attractive approach, in that the high separation capability of CE and the ultrasensitive detection and accurate identification performance of MS can be combined to provide a powerful system for the analysis of complex analytes. In this chapter, we discuss the detailed procedure of carrying out CE-MS analysis using a nano sheath-flow interface and its applications including intact mass analysis of monoclonal antibodies and fusion proteins, and a biotransformation study of two Fc-FGF21 molecules in a single-dose pharmacokinetic mice study. Optimization processes, including the finetuning of CE conditions and MS parameters, are illustrated in this chapter, with focuses on method robustness and assay reproducibility.

A capillary electrophoresis method for the determination of soluble monosaccharides in Ginkgo biloba leaves.

A method for the simultaneous determination of six monosaccharides by pre-column derivatization with 1-phenyl-3-methyl-5-pyrazolone and capillary electrophoresis was developed in this work. The derivatization (i.e., reaction temperature, capillary electrophoresis duration, and extraction number) and separation (i.e., pH and buffer concentration) conditions for capillary electrophoresis were optimized. Results showed that the limits of detection under optimal conditions were in the range of 0.036-0.35mg/L with a mean correlation coefficient >0.99. The recoveries were in the range of 87.3-108.49%, and the relative standard deviations of intra- and inter-day variations were in the ranges of 2.2-3.8 and 3.2-5.0%, respectively. The method was successfully applied to the analysis of six free monosaccharides in three types of Ginkgo biloba leaves.

An analytical study of lipid-oligonucleotide aggregation properties

Lipid-oligonucleotides (LON) attract great interest as supramolecular scaffolds to improve the intracellular delivery of nucleic acids. Analytical characterization of LON assemblies is critical to formulation development, understanding in-vivo performance, as well as quality control. For this study, we selected LONs featuring different modifications on both oligonucleotide (with or without a G4 prone sequence) and lipid (mono or bis-alkyl chain covalently attached to the oligonucleotide sequence). Size exclusion chromatography (SEC) and, for the first time, capillary electrophoresis (CE) were investigated to study LON supramolecular self-assemblies. Results were correlated to those obtained with conventional physico-chemical characterization techniques i.e. gel electrophoresis, dynamic light scattering, and circular dichroism. In SEC, a separation between LON monomers and micelles was achieved in 5min on a TSK-gel G3000PW column at 70°C with 100% water, as mobile phase. CE conditions were optimized using a fused-silica capillary length of 10.0cm effective length at 15°C. Different background electrolytes were tested by varying the nature and the concentration of salts added. A sodium tetraborate buffer with 75mM NaCl appeared suitable to promote LON assembly. CE offers benefits to LON micelle analysis in terms of speed of analysis, high resolution, and low quantity of sample injected. Moreover, CE provides an appropriate tool to assess the impact of media of biological relevance on LON self-assembly. In this work, the key role of lipophilic tails and the formation of tetramolecular G-quadruplexes on the stability of LON micelles was confirmed.

Influence of Sieving Polymer Concentration and Applied Voltage on Separation Accuracy of the Capillary Gel Electrophoresis for Small DNA Fragments

In this study, measurement conditions of Capillary gel electrophoresis (CGE) were optimized in order to increase the separation accuracy for small DNA fragments. We adopted a 20-bp DNA Ladder including 13 small double-stranded fragments (20, 40, 60, 80, 100, 120, 140, 160, 180, 200, 300, 400, and 500-bp). The applied voltage and sieving polymer concentration were assessed because these two factors significantly affect the separation accuracy of DNA fragments. As a result, we succeeded in optimizing CGE separation conditions for small DNA fragments with a sufficient degree of accuracy.

Separation of subcellular fluorescent microspheres by capillary electrophoresis.

Fluorescent microspheres (FMs) are widely employed in diagnostics and life sciences research; here, we investigated the effect of capillary coating, polymer concentration, electric field strength, and sample concentration on the separation performance of 1.0μm FMs in hydroxyethyl cellulose (HEC) by capillary electrophoresis (CE). Results showed that (1) capillary coating could enhance the fluorescence signal. (2) For HEC with the same molecular weight, the higher HEC concentration is, the later the first peak appears in the electropherogram. (3) When FMs are diluted, increasing the electric field strength can enhance the migration speed and reduce the aggregation of FMs. (4) The number of FMs calculated is close to the theoretical value when it is diluted 10,000 times. The optimum conditions for CE were as follows: 6cm/8cm of effective length and total length of the coated capillary, 0.3% HEC (1300k), and 300V/cm of electric field strength. Such a study is helpful for the development of a FM counting system. Graphical abstract.

Application of Composite NanoMaterial to Determine Phenols in Wastewater by Solid Phase Micro Membrane Tip Extraction and Capillary Electrophoresis

Composite nanoparticles were used in solid phase micro membrane tip extraction and capillary electrophoresis to determine phenol and p-amino-phenol in wastewater. The optimized conditions were 100 g/L concentration, 40 min contact time, 11 pH, 5 mg/mL nanoparticles amounts, 60 min desorption time, 9 desorption pH and 298 K temperature. Capillary electrophoresis conditions were phosphate buffer (15 mM, pH 7.0) background electrolyte, 18 kV applied voltage, 214 nm UV detection, 30 s sample loading at 23 ± 1 °C. The maximum percent uptakes of p-amino-phenol and phenol were 80.0 and 85.0%. High ratio recoveries of p-amino-phenol and phenol from nanomaterial were 99.0 and 98. Consequently, the actual extractions of p-amino-phenol and phenol from wastewater were 79.2 and 83.30 percent. The migration times of phenol and p-amino-phenol and were 9.0 and 12.0 min. The detection limits of phenol and p-amino-phenols were 0.1 and 0.2 µg/L after extraction and CE. Therefore, this combination of solid phase micro membrane tip extraction and capillary electrophoresis may be considered as the ideal one for monitoring of toxic phenol and p-amino-phenol in water sample.

Online preconcentration and determination of chondroitin sulfate, dermatan sulfate and hyaluronic acid in biological and cosmetic samples using capillary electrophoresis.

Capillary electrophoresis with large-volume sample stacking using an electroosmotic flow pump was developed for the determination of chondroitin sulfate, dermatan sulfate, and hyaluronic acid. Central composite design was used to simultaneously optimize the parameters for capillary electrophoresis separation. The optimized capillary electrophoresis conditions were 200mM sodium dihydrogen phosphate, 200mM butylamine, and 0.5% w/v polyethylene glycol as a background electrolyte, pH 4 and -16kV. Exploiting large-volume sample stacking using an electroosmotic flow pump, the sensitivity of the proposed capillary electrophoresis system coupled with UV detection was significantly improved with limits of detection of 3, 5, 1mg/L for chondroitin sulfate, dermatan sulfate, and hyaluronic acid, respectively. The developed method was applied to the determination of chondroitin sulfate and hyaluronic acid in cell culture media, cerebrospinal fluid, cosmetic products, and supplementary samples with highly acceptable accuracy and precision. Therefore, the proposed capillary electrophoresis approach was found to be simple, rapid, and reliable for the determination of chondroitin sulfate, dermatan sulfate, and hyaluronic acid in cell culture media, cerebrospinal fluid, cosmetic, and supplementary samples without sample pretreatment.

Determination of the Equilibrium Constant and Rate Constant of Protein-Oligonucleotide Complex Dissociation under the Conditions of Ideal-Filter Capillary Electrophoresis.

Ideal-filter capillary electrophoresis (IFCE) allows selection of protein binders from oligonucleotide libraries in a single step of partitioning in which protein-bound and unbound oligonucleotides move in the opposite directions. In IFCE, the unbound oligonucleotide does not reach the detector, imposing a problem for finding the equilibrium constant ( Kd) and rate constant ( koff) of protein-oligonucleotide complex dissociation. We report a double-passage approach that allows finding Kd and koff under the IFCE conditions, i.e. near-physiological pH and ionic strength. First, a plug of the protein-oligonucleotide equilibrium mixture passes to the detector in a pressure-driven flow, allowing for both the complex and free oligonucleotide to be detected as a single first peak. Second, the pressure is turned off and the voltage is applied to reverse the migration of only the complex which is detected as the second peak. The experiment is repeated with a lower voltage consequently resulting in longer travel time of the complex to the detector, greater extent of complex dissociation, and the decreased area of the second peak. Finally, the peak areas are used to calculate the values of Kd and koff. Here we explain theoretical and practical aspects of the double-passage approach, prove its validity quantitatively, and, demonstrate its application to determine Kd and koff for an affinity complex between a protein and its DNA aptamer. The double-passage approach for finding Kd and koff of protein-oligonucleotide complexes under the IFCE conditions is a perfect complement for IFCE-based selection of protein binders from oligonucleotide libraries.

Investigations into improving the standard DNA testing of trace samples at ESR

ABSTRACTSince 2007, the Institute of Environmental and Science Research (ESR) has been using the AmpF/STR Identifiler profiling kit as the standard DNA test. Whilst this approach works well for most samples with a DNA input range of 400 pg to 2 ng, samples containing less DNA often have limited to no profiling success. If required, such samples can undergo testing using a more sensitive technique, such as mini-STR analysis or Low Copy Number (LCN) DNA analysis. The introduction of a more sensitive standard DNA test into the laboratory would likely prove to be greatly beneficial. Profiling success should increase, particularly for samples containing trace DNA concentrations, and the number of samples requiring rework and further analysis using an ultra-sensitive technique would potentially reduce. A number of new generation multiplex kits with increased sensitivity are now commercially available. One such kit is the AmpF/STR Identifiler Plus profiling kit, which has enhanced buffer formulation and an optimized PCR cycling protocol. This study investigates the profiling results from trace DNA samples using Identifiler at 28 cycles and Identifiler Plus, at 28 and 29 cycles, analysed using the 3500xL Applied BioSystems Genetic Analyzer. Any further benefits from including a post-PCR clean-up step and enhanced capillary electrophoresis conditions are investigated and discussed.

Comparative Enantioseparation of Amlodipine by HPLC and Capillary Electrophoresis

Abstract Objective: The purpose of this study was to separate the enantiomers of amlodipine by High Performance Liquid Chromatography (HPLC) using ovomucoid (OVM) as chiral selector, respectively by Capillary Electrophoresis (CE) using cyclodextrines and to evaluate the analytical performance of the both proposed methods. Material and methods: HPLC enantioseparation of amlodipine was performed on an HPLC Agilent Technologies 1100 series using as chiral stationary phase an Ultron ES OVM, 150x4.6 mm column with ovomucoid as chiral selector. The stereoselective CE analysis of amlodipine was achieved on Agilent Technologies 7100 CE using uncoated fused-silica capillaries 48 cm x 50 mm and different type of cyclodextrins as chiral selectors. Results: A mobile phase consisting of 80% Na2HPO4 10 mM at a pH level of 5.0 and 20% ACN, isocratic elution at a flow of 1 ml/min turned to be the optimal experimental conditions for HPLC analysis (R=5.51; α=1.71) with retention times shorter than 10 minutes for the two isomers, tR (S-AML) = 4.63 (min); tR (R-AML) = 5.54 (min). The migration times for amlodipine enantiomers were tm (S-AML) = 8.15 (min) and tm (R-AML)= 8.45 (min) and the optimum CE conditions have proven to be a buffer solution containing 25 mM H3PO4 at pH 3.0 and 20 mM α-CD as chiral selector and a capillary temperature set at 15°C (R=1.51; α=1.03). Conclusion: The analytical performances of the chromatographic method using OVM as chiral selector are superior to the electrophoretic analysis method but the CE method is more economical and may represent an alternative to the HPLC chromatographic separation.

Micellar electrokinetic chromatography fingerprinting combined with chemometrics as an efficient strategy for evaluating the quality consistency and predicting the antioxidant activity of Lianqiao Baidu pills.

An approach combining micellar electrokinetic chromatography fingerprinting with chemometrics was developed to evaluate the quality consistency of Lianqiao Baidu pills, which are traditional Chinese patent medicines composed of 19 herbs used mainly to treat skin ulcers, common cold, rheumatism, herpes, and constipation. The triangle optimization method was employed to choose a satisfactory background electrolyte, with the information index, I, as an objective function for assessing the capillary electrophoresis conditions. Then, under the optimal conditions, the micellar electrokinetic chromatography fingerprints of 28 batches of samples were established, and five marker compounds were quantitatively determined simultaneously. A limited-ratio quantified fingerprint method was introduced to evaluate the chromatographic fingerprints both qualitatively and quantitatively. Principle component analysis revealed that the 28 batches of samples can be clustered according to different manufacturers. Moreover, the relationship between the fingerprint and the antioxidant activity was explored by orthogonal partial least-squares regression, which provided critical medicinal efficacy information for quality control.
