Updating of aversive memories after temporal error detection is differentially modulated by mTOR across development.

The updating of a memory is triggered whenever it is reactivated and a mismatch from what is expected (i.e., prediction error) is detected, a process that can be unraveled through the memory's sensitivity to protein synthesis inhibitors (i.e., reconsolidation). As noted in previous studies, in Pavlovian threat/aversive conditioning in adult rats, prediction error detection and its associated protein synthesis-dependent reconsolidation can be triggered by reactivating the memory with the conditioned stimulus (CS), but without the unconditioned stimulus (US), or by presenting a CS–US pairing with a different CS–US interval than during the initial learning. Whether similar mechanisms underlie memory updating in the young is not known. Using similar paradigms with rapamycin (an mTORC1 inhibitor), we show that preweaning rats (PN18–20) do form a long-term memory of the CS–US interval, and detect a 10-sec versus 30-sec temporal prediction error. However, the resulting updating/reconsolidation processes become adult-like after adolescence (PN30–40). Our results thus show that while temporal prediction error detection exists in preweaning rats, specific infant-type mechanisms are at play for associative learning and memory.

A Variable Oscillator Underlies the Measurement of Time Intervals in the Rostral Medial Prefrontal Cortex during Classical Eyeblink Conditioning in Rabbits.

The way in which brains generate working memories (those used for the transient processing and storage of newly acquired information) is still an intriguing question. Here, we report that the firing activities of neurons located in the rostromedial prefrontal cortex recorded in alert behaving rabbits are controlled by a dynamic oscillator. This oscillator generated firing frequencies in a variable band of 3-12 Hz depending on the conditioned stimulus-unconditioned stimulus intervals (1 s, 500 ms, 250 ms) selected for classical eyeblink conditioning of behaving rabbits. Shorter (50 ms) and longer (2 s) intervals failed to activate the oscillator and prevented the acquisition of conditioned eyelid responses. This is an unexpected mechanism to generate sustained firing activities in neural circuits generating working memories.

Bidirectional Plasticity of Purkinje Cells Matches Temporal Features of Learning

Many forms of learning require temporally ordered stimuli. In Pavlovian eyeblink conditioning, a conditioned stimulus (CS) must precede the unconditioned stimulus (US) by at least about 100 ms for learning to occur. Conditioned responses are learned and generated by the cerebellum. Recordings from the cerebellar cortex during conditioning have revealed CS-triggered pauses in the firing of Purkinje cells that likely drive the conditioned blinks. The predominant view of the learning mechanism in conditioning is that long-term depression (LTD) at parallel fiber (PF)-Purkinje cell synapses underlies the Purkinje cell pauses. This raises a serious conceptual challenge because LTD is most effectively induced at short CS-US intervals, which do not support acquisition of eyeblinks. To resolve this discrepancy, we recorded Purkinje cells during conditioning with short or long CS-US intervals. Decerebrated ferrets trained with CS-US intervals ≥150 ms reliably developed Purkinje cell pauses, but training with an interval of 50 ms unexpectedly induced increases in CS-evoked spiking. This bidirectional modulation of Purkinje cell activity offers a basis for the requirement of a minimum CS-US interval for conditioning, but we argue that it cannot be fully explained by LTD, even when previous in vitro studies of stimulus-timing-dependent LTD are taken into account.

Red Nucleus Neurons Actively Contribute to the Acquisition of Classically Conditioned Eyelid Responses in Rabbits

The red nucleus (RN) is a midbrain premotor center that has been suggested as being involved in the acquisition and/or performance of classically conditioned nictitating membrane/eyelid responses. We recorded in rabbits the activity of RN and pararubral neurons during classical eyeblink conditioning using a delay paradigm. Neurons were identified by their antidromic activation from contralateral facial and accessory abducens nuclei and by their synaptic activation from the ipsilateral motor cortex (MC) and the contralateral cerebellar interpositus (IP) nucleus. For conditioning, we used a tone as a conditioned stimulus (CS) followed 250 ms later by a 100 ms air puff as an unconditioned stimulus (US) coterminating with it. Conditioned responses (CRs) were determined from the evoked changes in the electromyographic activity of the orbicularis oculi (OO) muscle. Recorded neurons were classified by their antidromic activation and by their changes in firing rate during the CS-US interval. Identified neurons increased their firing rates in relation to the successive conditioning sessions, but their discharge rates were related more to the EMG activity of the OO muscle than to the learning curves. Reversible inactivation of the IP nucleus with lidocaine during conditioning evoked a complete disappearance of both conditioned and unconditioned eyelid responses, and a progressive decrease in CR-related activity of RN neurons. In contrast, MC inactivation evoked a decrease in the acquisition process and an initial disfacilitation of neuronal firing (which was later recovered), together with the late appearance of CRs. Thus, RN neurons presented learning-dependent changes in activity following MC inactivation.

Trace classical conditioning as an approach to the study of reward-related behaviour in laying hens: A methodological study

Positive affective states may be as important for animal welfare as the absence of suffering. Thus, there is a need for knowledge of the mechanisms underlying positive affective states, including how they can be induced and assessed. Studies of behaviour during the anticipation of a signalled palatable food reward have been suggested as a promising tool for answering these questions for the following three reasons: Anticipatory behaviour is hypothesized to reflect an appetitive type of positive affective state associated with dopaminergic activity in the brain. Inducing anticipation is useful for inducing pleasure and reducing stress. Finally, anticipatory behaviour can be used to test a subject's sensitivity to reward. However, to test various hypotheses regarding anticipatory behaviour there is a need to develop standardized research protocols. The aim of the present study was to develop and test an automated method for studying anticipatory and consummatory behaviour in laying hens in their home environment based on trace conditioning. Experimental hens (EX) were first trained to acquire the association between CS (conditioned stimulus; a green light) and US (unconditioned stimulus; meal worm) using a trace interval of 3.5 s. To test if EX hens had formed an association between CS and US, they were exposed to CS alone. Then, EX hens were trained to anticipate by gradually increasing the CS–US interval from the initial 3.5 s trace interval with a 1 s increment on every other CS + US presentation up to 22 s after 3 days and 32 s after 5 days of training. Finally, they were tested using a CS–US interval of 22 and 32 s. Control hens (CO) were exposed to CS and US in a random order, and tested as described for the EX hens. The results demonstrated that the EX hens responded to the CS with an increased level of anticipatory behaviour compared to the CO hens, and that this difference between the groups was significant at a CS–US interval of 22 s. No differences between treatments were found in any of the consummatory behaviours. In conclusion, trace classical conditioning can be used for inducing anticipatory behaviour in ad libitum fed laying hens freely moving in the home pen environment. This method will facilitate the testing of hypotheses about reward-related behaviour in laying hens.

Discriminative conditioning with different CS–US intervals produces temporally differentiated conditioned responses in the two eyes of the rabbit (Oryctolagus cuniculus).

The conditioned eyeblink response (CR) in rabbits is lateralized to the eye targeted by the unconditioned stimulus (US). However, a contralateral component has been reported during concurrent discriminative conditioning of the two eyes. The authors investigated CRs produced by both eyes during conditioning with 2 different interstimulus intervals (ISIs) in which a short conditioned stimulus (CS) was paired with a US to the left eye and a long CS was paired with a US to the right eye. Whether the 2 CSs were more or less similar (or identical), the short CS produced short-latency CRs in the left eye, whereas the long CS produced long-latency CRs in the right eye. The contralateral responses to a CS trained at one ISI were separable into temporal corollaries of the ipsilateral response (suggesting a bilaterality of the CR) versus those to a CS trained at another ISI (indicating generalization between the CSs). The results indicate that the neuronal substrates subserving CRs of the two eyes involve not only a dominant lateralization but also some avenue of bilaterality.

The influence of CS-US interval on several different indices of learning in appetitive conditioning.

Four experiments examined the effects of varying the conditioned stimulus-unconditioned stimulus (CS-US) interval (and US density) on learning in an appetitive magazine approach task with rats. Learning was assessed with conditioned response (CR) measures, as well as measures of sensory-specific stimulus-outcome associations (Pavlovian-instrumental transfer, potentiated feeding, and US devaluation). The results from these studies indicate that there exists an inverse relation between CS-US interval and magazine approach CRs, but that sensory-specific stimulus-outcome associations are established over a wide range of relatively long, but not short, CS-US intervals. These data suggest that simple CR measures provide different information about what is learned than measures of the specific stimulus-outcome association, and that time is a more critical variable for the former than latter component of learning.

Differential Effects of Long-Term Potentiation Evoked at the CA3–CA1 Synapse before, during, and after the Acquisition of Classical Eyeblink Conditioning in Behaving Mice

Experimentally induced long-term potentiation (LTP) is a persistent increase in synaptic strength that decays across time. In contrast, changes in synaptic strength during actual learning are gradual processes that increase with training. We have studied here the effects of LTP evoked before, during, and after the acquisition of a well known associative learning paradigm: the classical eyeblink conditioning. We used a trace paradigm, with a tone as the conditioned stimulus (CS) and an electric shock presented to the supraorbital nerve as the unconditioned stimulus (US). A single electrical pulse was presented to the Schaffer collateral-commissural pathway to evoke field EPSPs (fEPSPs) during the CS-US interval. LTP induced by high-frequency stimulation of the Schaffer collaterals lasted for 6-10 d. When LTP was evoked before conditioning, animals were unable to acquire conditioned eyeblinks if the training started 8 d after LTP disappearance, and no change was detected in fEPSP evoked at the CA3-CA1 synapse during conditioning. In contrast, LTP-induced animals learned as did controls when the conditioning test was presented 20 d after LTP had decayed to baseline, and presented a normal increase in fEPSP slopes across conditioning. When evoked during the first two conditioning sessions, LTP prevented both eyeblink conditioning and fEPSP increase. Finally, LTP did not disrupt the normal performance of a recall test of a previously acquired eyeblink conditioning. In this latter experiment, both the LTP-induced potentiation of fEPSPs and their physiological potentiation decayed across time with a similar time constant, with no apparent effect on memory recall.

Lock-and-Key Mechanisms of Cerebellar Memory Recall Based on Rebound Currents

A basic question for theories of learning and memory is whether neuronal plasticity suffices to guide proper memory recall. Alternatively, information processing that is additional to readout of stored memories might occur during recall. We formulate a "lock-and-key" hypothesis regarding cerebellum-dependent motor memory in which successful learning shapes neural activity to match a temporal filter that prevents expression of stored but inappropriate motor responses. Thus, neuronal plasticity by itself is necessary but not sufficient to modify motor behavior. We explored this idea through computational studies of two cerebellar behaviors and examined whether deep cerebellar and vestibular nuclei neurons can filter signals from Purkinje cells that would otherwise drive inappropriate motor responses. In eyeblink conditioning, reflex acquisition requires the conditioned stimulus (CS) to precede the unconditioned stimulus (US) by >100 ms. In our biophysical models of cerebellar nuclei neurons this requirement arises through the phenomenon of postinhibitory rebound depolarization and matches longstanding behavioral data on conditioned reflex timing and reliability. Although CS-US intervals<100 ms may induce Purkinje cell plasticity, cerebellar nuclei neurons drive conditioned responses only if the CS-US training interval was >100 ms. This bound reflects the minimum time for deinactivation of rebound currents such as T-type Ca2+. In vestibulo-ocular reflex adaptation, hyperpolarization-activated currents in vestibular nuclei neurons may underlie analogous dependence of adaptation magnitude on the timing of visual and vestibular stimuli. Thus, the proposed lock-and-key mechanisms link channel kinetics to recall performance and yield specific predictions of how perturbations to rebound depolarization affect motor expression.

The cerebellar interpositus nucleus and the dynamic control of learned motor responses.

The role played by the cerebellum in movement control requires knowledge of interdependent relationships between kinetic neural commands and the performance (kinematics) of learned motor responses. The eyelid motor system is an excellent model for studying how simple motor responses are elaborated and performed. Kinetic variables (n = 24) were determined here by recording the firing activities of orbicularis oculi motoneurons and cerebellar interpositus neurons in alert cats during classical conditioning, using a delay paradigm. Kinematic variables (n = 36) were selected from eyelid position, velocity, and acceleration traces recorded during the conditioned stimulus-unconditioned stimulus interval. Optimized experimental and analytical tools allowed us to determine the evolution of kinetic and kinematic variables, the dynamic correlation functions relating motoneuron and interpositus neuron firing to eyelid conditioning responses, the falling correlation property of the interpositus nucleus across the successive training sessions, the time and significance of the linear relationships between these variables, and finally, the phase-inversion property of interpositus neurons with respect to acquired conditioned responses. Whereas motoneurons encoded eyelid kinematics at every instant of the dynamic correlation range and generated the natural oscillatory properties of the neuromuscular elements involved in eyeblinks, interpositus neurons did not directly encode eyelid performance: namely, their contribution was only slightly significant in the dynamic correlation range, and this regularity caused the integrated neuronal activity to oscillate by progressively inverting phase information. Therefore, interpositus neurons seem to play a modulating role in the dynamic control of learned motor responses, i.e., they could be considered a neuronal phase-modulating device.

Dissociation between CA3–CA1 Synaptic Plasticity and Associative Learning in TgNTRK3 Transgenic Mice

Neurotrophins and their cognate receptors might serve as feedback regulators for the efficacy of synaptic transmission. We analyzed mice overexpressing TrkC (TgNTRK3) for synaptic plasticity and the expression of glutamate receptor subunits. Animals were conditioned using a trace [conditioned stimulus (CS), tone; unconditioned stimulus (US), shock] paradigm. A single electrical pulse presented to the Schaffer collateral-commissural pathway during the CS-US interval evoked a monosynaptic field EPSP (fEPSP) at ipsilateral CA1 pyramidal cells. In wild types, fEPSP slopes increased across conditioning sessions and decreased during extinction, being linearly related to learning evolution. In contrast, fEPSPs in TgNTRK3 animals reached extremely high values, not accompanied with a proportionate increase in their learning curves. Long-term potentiation evoked in conscious TgNTRK3 was also significantly longer lasting than in wild-type mice. These functional alterations were accompanied by significant changes in NR1 and NR2B NMDA receptor subunits, with no modification of NR1(Ser 896) or NR1(Ser 897) phosphorylation. No changes of AMPA and kainate subunits were detected. Results indicate that the NT-3/TrkC cascade could regulate synaptic transmission and plasticity through modulation of glutamatergic transmission at the CA3-CA1 synapse.

Cocaine-induced conditioned taste avoidance over extended conditioned stimulus–unconditioned stimulus intervals

Although long-delay learning has been demonstrated numerous times in the conditioned taste avoidance procedure, the empirical evidence showing this is almost exclusively limited to studies using emetics. Given that compounds outside the emetic class (e.g. drugs of abuse) are also effective in inducing conditioned taste avoidances, the present study assessed the ability of cocaine, a non-emetic psychoactive compound, to support long-delay conditioning as the unconditioned stimulus in conditioned taste avoidance preparation. Using saccharin as the conditioned stimulus, two taste-drug pairings were followed by six extinction trials during which saccharin was presented without subsequent injections of cocaine. During the two conditioning trials, animals were injected subcutaneously with cocaine (32 mg/kg) at different conditioned stimulus-unconditioned stimulus intervals, that is, 10, 60, 120, 180, 240, 300, 420 and 540 min. A control group of animals was given an equi-volume injection of the drug vehicle at the 10-min conditioned stimulus-unconditioned stimulus interval. After two conditioning trials, all treatment groups consumed significantly less saccharin than controls, with the magnitude of the effect decreasing as the conditioned stimulus-unconditioned stimulus interval increased. After six extinction trials, animals injected with cocaine at 10, 60 and 120 min conditioned stimulus-unconditioned stimulus intervals still consumed significantly less than controls. These results with cocaine suggest that taste avoidance learning over long delays is not limited to classical emetic compounds and may, in fact, be characteristic of taste avoidance learning in general.

Purkinje cell activity in the cerebellar anterior lobe after rabbit eyeblink conditioning

The cerebellar anterior lobe may play a critical role in the execution and proper timing of learned responses. The current study was designed to monitor Purkinje cell activity in the rabbit cerebellar anterior lobe after eyeblink conditioning, and to assess whether Purkinje cells in recording locations may project to the interpositus nucleus. Rabbits were trained in an interstimulus interval discrimination procedure in which one tone signaled a 250-msec conditioned stimulus-unconditioned stimulus (CS-US) interval and a second tone signaled a 750-msec CS-US interval. All rabbits showed conditioned responses to each CS with mean onset and peak latencies that coincided with the CS-US interval. Many anterior lobe Purkinje cells showed significant learning-related activity after eyeblink conditioning to one or both of the CSs. More Purkinje cells responded with inhibition than with excitation to CS presentation. In addition, when the firing patterns of all conditioning-related Purkinje cells were pooled, it appeared that the population showed a pattern of excitation followed by inhibition during the CS-US interval. Using cholera toxin-conjugated horseradish peroxidase, Purkinje cells in recording areas were found to project to the interpositus nucleus. These data support previous studies that have suggested a role for the anterior cerebellar cortex in eyeblink conditioning as well as models of cerebellar-mediated CR timing that postulate that Purkinje cell activity inhibits conditioned response (CR) generation during the early portion of a trial by inhibiting the deep cerebellar nuclei and permits CR generation during the later portion of a trial through disinhibition of the cerebellar nuclei.

Hypothermia modifies the effective CS–US interval in conditioned taste aversion in rats

Conditioned taste aversion can be acquired when rats experience an unconditioned stimulus (US) while anesthetized. In contrast to anesthetics, a hypothermia-induced comatose state immediately after presentation of a taste conditioned stimulus (CS) prevented a taste-illness association at relatively short CS–US intervals and potentiated the aversion at longer intervals. Results at shorter CS–US intervals were explained on the basis of hypothermia’s temporally graded amnesitc properties. Evidence for conditioning at the longer intervals was discussed in relation to slowing down metabolism allowing for associations to be formed at CS–US intervals that normally do not result in evidence of conditioning. Manipulating body temperature during the CS–US interval was demonstrated to alter rats’ ability to bridge temporal gaps in associative learning.

On the relationship between anticipatory behaviour in a Pavlovian paradigm and Pavlovian-to-Instrumental Transfer in rats ( Rattus norvegicus)

The present rat study assessed the relationship between, and the sensitivity of, two different tests for appetitive conditioned responding to differences in the contingency between a conditioned stimulus (CS) and an unconditioned stimulus (US), and to differences in US magnitude. The first test used a Pavlovian-to-Instrumental Transfer (PIT) paradigm, assessing the capacity of the CS to enhance instrumental responding for food. The second test employed a Pavlovian conditioning paradigm with an extended CS–US interval, and total number of behavioural elements in this interval as a dependent measure. The PIT test proved to be sensitive to contingency but not reward magnitude differences, whereas the reverse was true for the Pavlovian test. Although there was a significant correlation between tests in the magnitude of the CS-induced increase of food-magazine entries, the main dependent measure from PIT (number of lever presses) and that from the Pavlovian test (total number of behavioural elements) did not correlate. It is suggested that in the PIT procedure, the CS induces a chain of behavioural responses of which lever pressing is just a single element and that the Pavlovian test, in principle, is more sensitive.

Eyeblink classical conditioning and interpositus nucleus activity are disrupted in adult rats exposed to ethanol as neonates.

Neonatal exposure to ethanol in rats, during the period of brain development comparable to that of the human third trimester, produces significant, dose-dependent cell loss in the cerebellum and deficits in coordinated motor performance. These rats are also impaired in eyeblink conditioning as weanlings and as adults. The current study examined single-unit neural activity in the interpositus nucleus of the cerebellum in adults following neonatal binge ethanol exposure. Group Ethanol received alcohol doses of 5.25 g/kg/day on postnatal days 4-9. Group Sham Intubated underwent acute intragastric intubation on postnatal days 4-9 but did not receive any infusions. Group Unintubated Control (from separate litters) did not receive any intubations. When rats were 3-7 mo old, pairs of extracellular microelectrodes were implanted in the region of the interpositus nucleus. Beginning 1 wk later, the rats were given either 100 paired or 190 unpaired trials per day for 10 d followed by 4 d of 100 conditioned stimulus (CS)-alone trials per day. As in our previous study, conditioned response acquisition in Group Ethanol rats was impaired. In addition, by session 5 of paired acquisition, Group Sham Intubated and Group Unintubated Control showed significant increases in interpositus nucleus activity, relative to baseline, in the CS-unconditioned stimulus interval. In contrast, Group Ethanol failed to show significant changes in interpositus nucleus activity until later in training. These results indicate that the disruption in eyeblink conditioning after early exposure to ethanol is reflected in alterations in interpositus nucleus activity.

Lesions of the Intermediate Medial Hyperstriatum Ventrale Impair Sickness-Conditioned Learning in Day-Old Chicks

Lesion studies show that the intermediate medial hyperstriatum ventrale (IMHV), a forebrain visual association area in chicks, is involved in learning and memory for one-trial passive avoidance and imprinting. We examined the effects of IMHV lesions in a one-trial, nongustatory, sickness-conditioned learning task. This task is similar to passive avoidance and imprinting because all three tasks require the chick to remember visual cues in order to respond correctly. However, sickness-conditioned learning differs from imprinting and passive avoidance because it uses sickness as the aversive stimulus and there is a longer conditioned stimulus-unconditioned stimulus interval (30-min delay compared to seconds). Bilateral IMHV lesions given 24 h before training impaired the ability of the chicks to avoid a bead associated with sickness produced by lithium chloride injection, as did pretraining unilateral left or right IMHV lesions. Post-training IMHV lesions given 1 h after training did not impair avoidance of the test bead in the sickness-conditioned learning task. However, lesioned chicks showed generalized avoidance of all test beads. The pretraining lesion results are similar to those found in imprinting and passive avoidance learning; however, the effects of unilateral IMHV lesions differed. Post-training lesion effects are similar to those found in passive avoidance learning. We propose that both left and right IMHV are necessary for sickness-conditioned learning and that post-training IMHV lesions impair the ability of the chick to learn or remember the association between the color of the bead and the aversive consequences of LiCl injection.

Ketaset–Rompun Extends the Temporal Gradient for Hypothermia-Induced Retrograde Amnesia

In studies of experimentally induced retrograde amnesia (RA), as the interval between training and the amnestic treatment is lengthened, amnesia decreases (4). This temporal gradient for RA has been reported with a wide variety of amnestic agents, including RA produced by thermoregulatory disturbances (8). This temporal gradient for RA is not unlike certain characteristics of classical conditioning, where weaker conditioned responding occurs when the interval between the conditioned stimulus (CS) and unconditioned stimulus (US) is lengthened. Furthermore, there is evidence that administration of anesthetics can lengthen the “effective conditioning” interval between the CS and US, as demonstrated in a conditioned taste-aversion (CTA) procedure (10). In that study, little conditioning was observed when a 3-h delay (or more) was incorporated between presentations of the CS (flavor) and the US (toxin). However, if subjects were anesthetized immediately after the CS was delivered and remained anesthetized during the CS–US interval, strong conditioning was observed with CS–US intervals of up to 9 h. The aim of the present experiment was to determine if the temporal gradient for hypothermia-induced RA could also be lengthened. That is, we tested whether the interval between training and hypothermia treatment could be lengthened by anesthetizing subjects with Ketaset–Rompun. The results indicate that the training-to-amnestic agent interval could be lengthened within moderate limits. The implications for hypothermia-induced RA is further discussed.

Relationships among age, conditioned stimulus-unconditioned stimulus interval, and neuropsychological test performance.

To evaluate the effect of age at various conditioned stimulus (CS)-unconditioned stimulus (US) intervals, 144 young, middle-aged, and older adults were tested on eyeblink classical conditioning at CS-US intervals of 500, 1,000, or 1,500 ms. Reaction time, response timing, motor learning, declarative memory, and attention were assessed to identify correlates of conditioning at various CS-US intervals. Previously reported middle-aged and older adults were impaired at a 400-ms CS-US interval, but the addition of 100 ms to the CS-US interval in this study enabled equal conditioning in middle-aged and young adults. At a 1,000-ms CS-US interval, older adults remained significantly impaired. It was only at the 1,500-ms CS-US interval that conditioning was equal for the 3 age groups. Measures of reaction time, timing, and motor learning were not correlated systematically with conditioning. Whereas the results of age differences at various CS-US intervals were clear and striking, patterns of relationships among neuropsychological and conditioning variables were not consistent in indicating sources of age differences.

Metabotropic Glutamate Receptor Activation in Cerebellar Purkinje Cells as Substrate for Adaptive Timing of the Classically Conditioned Eye-Blink Response

To understand how the cerebellum adaptively times the classically conditioned nictitating membrane response (NMR), a model of the metabotropic glutamate receptor (mGluR) second messenger system in cerebellar Purkinje cells is constructed. In the model, slow responses, generated postsynaptically by mGluR-mediated phosphoinositide hydrolysis and calcium release from intracellular stores, bridge the interstimulus interval (ISI) between the onset of parallel fiber activity associated with the conditioned stimulus (CS) and climbing fiber activity associated with unconditioned stimulus (US) onset. Temporal correlation of metabotropic responses and climbing fiber signals produces persistent phosphorylation of both AMPA receptors and Ca(2+)-dependent K+ channels. This is responsible for long-term depression (LTD) of AMPA receptors. The phosphorylation of Ca(2+)-dependent K+ channels leads to a reduction in baseline membrane potential and a reduction of Purkinje cell population firing during the CS-US interval. The Purkinje cell firing decrease disinhibits cerebellar nuclear cells, which then produce an excitatory response corresponding to the learned movement. Purkinje cell learning times the response, whereas nuclear cell learning can calibrate it. The model reproduces key features of the conditioned rabbit NMR: Purkinje cell population response is timed properly; delay conditioning occurs for ISIs of up to 4 sec, whereas trace conditioning occurs only at shorter ISIs; mixed training at two different ISIs produces a double-peaked response; and ISIs of 200-400 msec produce maximal responding. Biochemical similarities between timed cerebellar learning and photoreceptor transduction, and circuit similarities between the timed cerebellar circuit and a timed dentate-CA3 hippocampal circuit, are noted.
