Evaluation of angiogenesis and microvascular remodeling in the subventricular zone of the brain of mice with experimental Alzheimer’s disease

BACKGROUND: Under the influence of external factors (learning), processes of microvasculature remodeling occur in the neurogenic niche to meet the metabolic needs of activated cells. However, it remains unclear how these mechanisms of brain plasticity are disrupted during neurodegeneration.
 AIM: To study the expression features of markers of angiogenesis and microvascular remodeling in the subventricular zone of the brain during training of animals, including against the background of the development of Alzheimer’s-type neurodegeneration in them.
 MATERIAL AND METHODS: The studies were carried out on C57BL/6 mice at the age of 8 months. Modeling of Alzheimer’s disease was carried out by intrahippocampal injection of 2 μl of a 1 mM solution of β-amyloid Aβ25-35. To assess cognitive deficits, a conditioned passive avoidance test using an aversive stimulus was used. The expression of LC3B, ZO1, VEGFR2, VEGFR3, CD146, ICAM2, Dll4, Tie2 in the subventricular zone per 100 DAPI-positive cells was assessed. The test results were processed using one-way ANOVA and Fisher's test, the Mann–Whitney U test, the results were considered significant at p 0.05.
 RESULTS: On the 9th day after the administration of β-amyloid, before the application of the aversive stimulus, an increase in the expression level of LC3 (7.95±5.83%, p=0.045), CD146 (18.35±0.01%, p=0.045) was recorded, as well as VEGFR3 (17.13±5.05%, p=0.045), which continued to increase after the presentation of the stimulus (26.61±0.01%, p=0.045). By the beginning of registration of cognitive impairment (38th day of the experiment), the expression level of VEGFR2 (20.61±2.8%, p=0.045) and ICAM2 (126.61±41.28%, p=0.045) increased, the content of Dll4 (29.66±8.72%, p=0.045) and Tie2 (36.39±7.8%, p=0.045) decreased in animals with experimental Alzheimer’s disease.
 CONCLUSION: An aversive stimulus stimulates microvascular remodeling mechanisms in the subventricular zone of the animal’s brain, but when exposed to β-amyloid, these processes are significantly disrupted.

Sex-specific differences and similarities of olanzapine and risperidone on avoidance suppression in rats in the conditioned avoidance response test

It is well known that antipsychotic drugs (APDs) are more effective in reducing symptoms in women than in men, and that women are more sensitive to the side effects of APDs. Therefore, it is of great importance that sex differences in drug responses are considered already in the early stages of drug development. In this study, we investigated whether sex-specific differences could be observed in response to the commonly prescribed APDs olanzapine and risperidone using the conditioned avoidance response (CAR) test. To this end we tested the effect of 1.25 and 2.5 mg/kg olanzapine and 0.25 and 0.4 mg/kg risperidone using female and male Wistar rats in the CAR test. Whereas there were no significant differences between the female and male rats in response to either dose of olanzapine administration, an injection of 0.4 mg/kg risperidone significantly suppressed avoidance more in female rats than in male rats. In addition, we found that the estrous cycle of the female rats did not have a significant effect on the avoidance response. In conclusion, we show that there are sex-specific differences as well as similarities between female and male rats in the CAR test and novel APDs should be tested on female and male rats in the future.

Lumateperone-mediated effects on prefrontal glutamatergic receptor-mediated neurotransmission: A dopamine D1 receptor dependent mechanism

Lumateperone is a novel drug approved for the treatment of schizophrenia in adults and depressive episodes associated with bipolar depression in adults, as monotherapy and as adjunctive therapy with lithium or valproate treatment in the United States. Lumateperone simultaneously modulates key neurotransmitters, such as serotonin, dopamine, and glutamate, implicated in serious mental illness. In patients with schizophrenia, lumateperone was shown to improve positive symptoms along with negative and depressive symptoms, while also enhancing prosocial behavior. Moreover, in patients with bipolar I or II disorder, lumateperone improved depressive symptoms as well. To further understand the mechanisms related to lumateperone's clinical response, the aim of this study was to investigate the effect of lumateperone on dopaminergic- and glutamatergic signaling in the rat medial prefrontal cortex (mPFC). We used the conditioned avoidance response (CAR) test to determine the antipsychotic-like effect of lumateperone, electrophysiology in vitro to study lumateperone's effects on NMDA- and AMPA-induced currents in the mPFC, and the neurochemical techniques microdialysis and amperometry to measure dopamine- and glutamate release in the rat mPFC. Our results demonstrate that lumateperone; i) significantly suppressed CAR in rats, indicating an antipsychotic-like effect, ii) facilitated NMDA and AMPA receptor-mediated currents in the mPFC, in a dopamine D1-dependent manner, and iii) significantly increased dopamine and glutamate release in the rat mPFC. To the extent that these findings can be translated to humans, the ability of lumateperone to activate these pathways may contribute to its demonstrated effectiveness in safely improving symptoms related to neuropsychiatric disorder including mood alterations.

Sodium nitroprusside enhances the antipsychotic-like effect of olanzapine but not clozapine in the conditioned avoidance response test in rats

The nitric oxide (NO)-donor, sodium nitroprusside (SNP) has been proposed as an adjunct treatment to enhance the effect of antipsychotic drugs (APDs). As NO constitutes an important downstream signaling molecule of N-methyl-D-aspartate receptors, SNP may alleviate symptoms of schizophrenia by modulating glutamatergic signaling. We previously showed that SNP enhances the antipsychotic-like effect of a sub-effective dose of risperidone in the conditioned avoidance response (CAR) test, indicating that adjunct SNP may be used to lower the dose of risperidone and in this way reduce the risk of side effects. By using the CAR test, we here investigated if SNP also enhances the antipsychotic-like effect of olanzapine or clozapine. Importantly, SNP (1.5 mg/kg) significantly enhanced the antipsychotic-like effect of olanzapine (1.25 and 2.5mg/kg) to a clinically relevant level, supporting the potential clinical use of SNP as an adjunct treatment to improve the effect of APDs. However, SNP (1.5 mg/kg) did not increase the antipsychotic-like effect of clozapine (5 and 6 mg/kg). Moreover, we found that the rats developed tolerance towards clozapine after repeated administrations. Thus, our study motivates further investigation using different preclinical models to assess the effect of adjunct treatment of SNP to APDs, also targeting the negative symptoms and cognitive deficits seen in schizophrenia.

Behavioral and neurochemical changes induced by post-weaning female rats isolation

Rodents post-weaning isolation could be considered a significant early-life model of chronic stress. The aim of our study was to investigate the effects of female rats post-weaning isolation on different behavioral tests and neurochemical parameters. On postnatal day 21, female pups were housed either isolated or socially in groups (4/cage) for 6 weeks. After the 6th week adult female rats were behaviorally examined using open field test, three chamber social interaction test, hole board test and condition avoidance test. In addition, hippocampal content of neurotransmitters, oxidative stress and inflammatory markers were measured. Isolated female rats exhibited behavioral changes, female rats shows a significant increase in anxiety and a significant decrease in locomotor activity in comparison to the control group using open field test . Moreover, female rats isolation promoted a significant decrease in sociality index as compared to control groups. Finally, female rats post-weaning isolation of invoked a significant impairment in long-term and short-term memory evaluated by both conditioned avoidance and hole board tests, in comparison to the control group. Furthermore, female rats post- weaning isolation induced oxidative stress, inflammation, and neurotransmitter alterations. Collectively female rats post weaning isolation had significantly prompted both behavioral and neurochemical changes which could represent the essential important symptoms of schizophrenia.

Discovery of a new class of multi-target heterocycle piperidine derivatives as potential antipsychotics with pro-cognitive effect

A series of benzoisoxazoleylpiperidine derivatives were synthesized by using the multi-target strategies and their potent affinities for dopamine (DA), serotonin (5-HT) and human histamine H3 receptors have been evaluated. Of these compounds, the promising candidate 4w displayed high affinities for D2, D3, 5-HT1A, 5-HT2A and H3, a moderate affinity for 5-HT6, negligible effects on the human ether-a-go-go-related gene (hERG) channel, low affinities for off-target receptors (5-HT2C, adrenergic α1 and H1). In addition, the animal behavioral study revealed that, compared to risperidone, compound 4w significantly inhibited apomorphine-induced climbing and MK-801-induced movement behaviors with a high threshold for catalepsy and low liabilities for weight gain and hyperprolactinemia. Results from the conditioned avoidance response test and novel object recognition task demonstrated that 4w had pro-cognitive effects. Thus, the antipsychotic drug-like activities of 4w indicate that it may be a potential polypharmacological antipsychotic candidate drug.

Effect of coenzyme Q10 and/or epigallocatechin gallate on memantine-treated amnesia model in rats

Background Alzheimer’s dementia is a progressive, fatal neurodegenerative disease that manifests as a disintegration of perception and memory. Objectives The current study evaluated the possible therapeutic effect of coenzyme Q10 (CoQ10) and/or epigallocatechin-3-gallate (EGCG) combined with memantine on scopolamine-induced amnesia in rats by evaluating the behavioral, biochemical, and histopathological changes. Materials and methods Rats were randomly allocated to 11 groups, each of which contained 16 rats. Six rats were used for biochemical tests, while ten rats were used for behavioral and histological examinations. Two behavioral assessments were conducted: an object-recognition test and a conditioned-avoidance test. The dopamine (DA) content of brain tissues was determined, as well as oxidative stress markers, such as superoxide dismutase, lipid peroxide end product malondialdehyde, and reduced glutathione. Besides, the activity of acetylcholine esterase (AchE), total antioxidant capacity, and inflammatory markers, such as tumor necrosis factor-alpha and interleukin-one beta, were determined in serum. Furthermore, histological examinations of whole-brain tissues were made. Results Scopolamine-treated rats were administered memantine at a dose of 20 mg/kg, coenzyme Q10 at a dose of 10 mg/kg, and EGCG at a dose of 10 mg/kg, individually or in combination, resulting in an enhancement of cognitive impairment in the condition-avoidance and object-recognition tests, as well as an improvement in all oxidative stress biomarkers, inflammatory biomarkers, and histological examination. Conclusion Rats were administered memantine and pretreated by the combination of CoQ10 and EGCG, resulting in potentiating the memantine action in scopolamine-induced amnesia in rats. The improvement in cognitive memory could be due to the synergistic effect of these drugs by decreasing AchE activity, DA level, anti-inflammatory, and antioxidant effects.

Enzyme-assisted Solvent Extraction of High-yield Paeonia suffruticosa Andr. Seed Oil and Fatty Acid Composition and Anti-Alzheimer's Disease Activity.

Enzyme-assisted solvent extraction (EASE) of Paeonia suffruticosa Andr. seed oil (PSO) was optimized by response surface methodology (RSM). The fatty acid composition and anti-Alzheimer's disease (AD) activity of PSO were analyzed. An enzyme mixture composed of cellulase and hemicellulase (1:1, w/w) was most effective in determining the extraction yield of PSO. The ideal extraction conditions were a pH value of 5.1, an enzymolysis time of 68 min, and a temperature of 50℃. The average extraction yield of PSO was 38.2 mL/100 g, 37.4% higher than that of untreated peony seed (27.8 mL/100 g). The fatty acid composition of PSO under optimal conditions for EASE was analyzed by gas chromatography-mass spectrometry (GC-MS). The predominant unsaturated fatty acids of PSO were determined to be more than 90.00%, including n-3 α-linolenic acid (43.33%), n-6 linoleic acid (23.40%) and oleic acid (23.59%). In this experiment, the anti-AD effect of PSO was also analyzed by performing learning and memory ability tests with Drosophila. PSO retarded the decrease in climbing ability in AD Drosophila. The 1% and 5% PSO groups were significantly different from the model group (b p < 0.05). The smell short-term memory ability test revealed the number of Drosophila in barrier and barrier-free centrifuge tubes in each group. PSO feeding improved learning and memory in AD Drosophila, with the highest number entering the barrierfree centrifuge tube. The performance index (PI) measured by the Pavlov olfactory avoidance conditioning test also demonstrated the effect of PSO on the learning and memory abilities of Drosophila. The PI of the PSO group was significantly increased compared to that of the model group. HE-stained brain tissue sections of AD Drosophila showed higher neurodegenerative changes, while PSO significantly reduced neurodegenerative damage. These results indicated that PSO can significantly improve the cognitive function of AD Drosophila and may help to prevent AD.

Study of aversive and p38 mapk-inhibitory properties of kappa-agonist with analgesic activity – compound RU-1205

Introduction: The clinical use of kappa-opioid agonists, despite their lack of significant drug potential, is limited by the development of severe sedation, dysphoria, depression, and anhedonia. To this date, there are kappa-opioid receptor agonists lacking these side effects due to the selective activation of intracellular signal transmission pathways without p38-MAPK-kinase activation.&#x0D; Materials and methods: We analyzed assessment of the docking energy of compound RU-1205 to the p38-MAPK ac­tive center by the method of similarity to SB203580. The study of possible aversive properties of RU-1205 (0.01–1 mg/kg s.c.) conducted in the tests of the intravenous self-administration and drug differentiation with butorphanol (0.01–0.3 mg/kg). The study of p38 MAPK-inhibitory activity was studied by the ability of RU-1205 to change the aversive prop­erties of U50488 (10 mg/kg i.p.) compared to MAPK-kinase inhibitor SB203580 in the conditioned place avoidance test.&#x0D; Results: The spatial similarity coefficient of the RU-1205 molecule with SB203580 by the molecular conformation method was 1.14 (high similarity), and the docking energy was -8.7 Kcal/mol. RU-1205 did not possess any properties similar to those of butorphanol and did not demonstrate any primary reinforcing aversive properties in the development of intravenous self-administration reaction. Compound RU-1205 did not demonstrate any aversive properties in the condi­tioned place avoidance test, and reduced the development of aversion caused by U-50488, when they were used together.&#x0D; Discussion: The in silico analysis suggested that, in addition to agonism towards the kappa-opioid receptor, RU-1205 com­pound exhibits the properties of a p38 MAPK kinase inhibitor, which means it may have a double pharmacological activity.&#x0D; Conclusion: Kappa agonist – compound RU-1205 – is not a trigger of the development of behavioral patterns in ani­mals corresponding to the development of addiction/dysphoria. The mechanism of such an activity may be associated with an inhibitory effect of compound RU-1205 on neuronal p38-MAPK-kinase.

Effects of quercetin on the neuromotor function and behavioral responses of Wistar and Zucker rats fed a high-fat and high-carbohydrate diet

Quercetin can affect some pathological manifestations in obesity. The mechanism underlying the presumed therapeutic effect of quercetin is probably related to the influence on the central processes regulating energy homeostasis. Thus, the purpose of this study was to examine the effect of quercetin on the neuromotor and behavioral functions in Zucker (Z) and Wistar (W) rats with genetically and/or diet-induced obesity. Rats of both strains received balanced or high fat and fructose diet (HFCD) in a 62-day experiment or the same diets supplemented with quercetin at the dose of 50 mg/kg body weight per day. The neuromotor function and behavioral responses were examined using the grip strength test, open field test, elevated plus maze test and conditioned passive avoidance response (CPAR) test. The quercetin potentiated a decrease in anxiety in W rats consumed HFCD and this effect was absent in Z rats with a defect in the leptin receptor gene. In contrast, quercetin increased locomotor activity and impaired short-term memory in the CPAR test only in Z rats with the absence of normal leptin reception. Against the background of the identified changes quercetin exerted significant effects on the lipid and nitrogen metabolism indices such as HDL cholesterol, AsAT/AlAT activities ratio, urea level as well as body and fat mass that were different in Z and W rats. The data obtained show that the effects of quercetin on behavior vary significantly between two strains of rat and consequently are mediated by processes of leptin reception.

Enhancement of the antipsychotic effect of risperidone by sodium nitroprusside in rats

Recently, a single injection of the nitric oxide donor sodium nitroprusside (SNP) was found to induce a rapid and sustained antipsychotic effect in treatment-resistant schizophrenia (TRS). Moreover, a single i.p. injection of SNP in rats was found to generate both rapid and persisting changes in brain synaptic plasticity, including enhanced excitatory postsynaptic current responses and spine morphology in layer V pyramidal cells in the medial prefrontal cortex (mPFC) brain slices. Here we used the conditioned avoidance response (CAR) test in rats to investigate the antipsychotic-like efficacy of SNP in combination with low-dose risperidone. In addition, we performed microdialysis experiments in freely moving rats to measure neurotransmitter efflux in the mPFC and the nucleus accumbens (NAc). Risperidone caused only 20% suppression of CAR, which is far below the degree of CAR suppression required to predict a significant clinical antipsychotic effect. Addition of a low dose of SNP to risperidone dramatically enhanced the antipsychotic-like effect to a clinically relevant level. SNP significantly enhanced the risperidone-induced dopamine output in the mPFC but not in the NAc. The increased prefrontal dopamine release induced by the drug combination may also improve cognition as indicated by previous preclinical and clinical studies and, furthermore, via enhanced synaptic spine function and morphology in mPFC generate a both rapid and prolonged antipsychotic and pro-cognitive effect. Our results delineate SNP as a promising new treatment option for schizophrenia, including TRS, when added to currently available antipsychotic medication in order to improve efficacy at maintained or even reduced dosage.

Discovery of a pyrazolo[1,5-a]pyrimidine derivative (MT-3014) as a highly selective PDE10A inhibitor via core structure transformation from the stilbene moiety

We have developed a new class of PDE10A inhibitor, a pyrazolo[1,5-a]pyrimidine derivative MT-3014 (1). A previous compound introduced was deprioritized due to concerns for E/Z-isomerization and glutathione-adduct formation at the core stilbene structure. We discovered pyrazolo [1,5-a] pyrimidine as a new lead scaffold by structure-based drug design utilizing a co-crystal structure with PDE10A. The lead compound was optimized for in vitro activity, solubility, and selectivity against human ether-á-go-go related gene cardiac channel binding. We observed that MT-3014 shows excellent efficacy in rat conditioned avoidance response test and suitable pharmacokinetic properties in rats, especially high brain penetration.

Comparative analysis of the influence of a high-fat/high-carbohydrate diet on the level of anxiety and neuromotor and cognitive functions in Wistar and DAT-KO rats.

We compared anxiety, neuromotor, and cognitive functions in mutant rats with different allelic variants of dopamine transporter DAT knockout receiving balanced or excess in fat and fructose diet. The experiments were performed in DAT−/− homozygotes, DAT+/− heterozygotes, and DAT+/+ wild type rats. The genotype of DAT‐KO rats was confirmed by restriction analysis of DAT gene compared to behavioral responses in the open field test (OF). Animals in the first groups of each strain were fed a balanced AIN93M diet; and those in the second groups with a high‐fat/high‐fructose diet. Neuromotor function was studied as grip strength, and behavioral responses were assessed in the elevated plus maze and conditioned passive avoidance response tests. The mass of the internal organs and white and brown fat, as well as selected lipid and nitrogen metabolism parameters in blood plasma were determined at the end of the experiment. DAT−/− had the highest specific grip strength, and showed an increase in initial exploratory activity in comparison with DAT+/− and DAT +/+. The exploratory activity was significantly reduced in the second test compared to the first one in DAT−/− and DAT+/− of first but not second group. Anxiety decreased with age in the second groups of DAT+/− and DAT+/+ (but not in DAT−/−) and was higher in DAT+/+ than in DAT+/− and DAT−/−. Excess fat and fructose resulted in the deterioration of short‐term memory in DAT+/+. Lipidomic indices of blood plasma were less responsive to diet in DAT−/− and DAT−/+ in comparison to DAT+/+. The increased AsAT/AlAT activity ratio in DAT−/− compared with those in DAT+/+ suggests the activation of catabolism activity in the mutants. The consumption of excess fat and fructose significantly modified the effects produced by DAT gene allelic variants presumably due to the influence on the processes of dopamine metabolism.

The role of MAPK signaling pathway in selenium amelioration of high fat/high cholesterol diet-induced tauopathy in rats

BackgroundMAP kinases, CREB, and Tau are signaling molecules among downstream synaptic targets involved in synaptic function, memory formation and cognition. PurposeHere we investigate the neuro-protective effect of selenium in HFHCD induced tauopathy and cognitive impairment in rats. The study was focused on the effects on synaptic plasticity related molecules in hippocampus, which in turn may be the mechanism responsible for underlying behavior alterations. MethodRats were divided into 2 main groups: one fed with normal rat chow diet and the other with HFHCD for 6 weeks. Every group was subdivided into three subgroups, non-treated, low dose Se (200 μg/kg) and high dose Se (400 μg/kg). The cognitive behaviors of the rats were tested using the Morris Water Maze test, hole board and conditioned avoidance tests. ResultsDaily administration of Se decreased the observed memory impairment induced by HFHCD as measured by behavioral tests. It significantly alleviated oxidative stress and restored protein expression of cyclic AMP response element protein (CREB) and brain derived neurotrophic factor (BDNF) and reduced p-Tau in the hippocampus. ConclusionSe reversed HFHCD-induced cognitive impairments via decrease expression of p38 MAPK that phosphorylate and aggregate Tau protein. Addition, It restored neuronal plasticity through increasing the expression of BDNF, and CREB in the hippocampus; thus, it can be considered as a possible beneficial therapeutic approach for prevention and treatment of HFHCD induced tauopathy and cognitive impairment, further studies are warranted in this field.

Interspecific Differences in Behavioral Responses and Neuromotorics between Laboratory Rodents Receiving Rations with Easily Digested Carbohydrates.

We assessed the effect of intake of easily digested carbohydrates for 133 days on quantitative parameters of neuromotorics and cognitive function in Wistar rats and C57Bl/6J mice. Neuromotorics (muscle tone) was assessed in rats and mice by the forelimb muscle force (grip strength) over 4 months. Anxiety was assessed in the elevated plus-maze test and cognitive function (short-term and long-term memory) was evaluated by conditioned passive avoidance response (CPAR) test over 3 months. The mice, in contrast to rats, receiving the diet with easily digested sugars demonstrated suppression of neuromotorics. Anxiety increased with age in female mice, but not in rats, irrespective of the diet. Cognitive function in rats receiving experimental rations did not change significantly in comparison with the control. In mice, consumption of equimolar mixture of fructose and glucose impared short-term, but not long-term memory, in comparison with the group receiving glucose alone. We revealed a small (by 14-17%), but statistically significant increase in the brain weight in mice receiving fructose and sucrose. The study demonstrates sufficient interspecies differences in the influence of carbohydrate rations on neuromotorics and behavioral responses in the in vivo metabolic syndrome model.

Discovery of 2-[(E)-2-(7-Fluoro-3-methylquinoxalin-2-yl)vinyl]-6-pyrrolidin-1-yl-N-(tetrahydro-2H-pyran-4-yl)pyrimidin-4-amine Hydrochloride as a Highly Selective PDE10A Inhibitor.

Phosphodiesterase (PDE) 10A is a dual hydrolase of cAMP and cGMP and highly expressed in striatal medium spiny neurons. Inhibition of PDE10A modulates the activity of medium spiny neurons (MSN) via the regulation of cAMP and cGMP. Signal control of MSN is considered associated with psychotic symptoms. Therefore PDE10A inhibitor is expected as a therapeutic method for psychosis disease such as schizophrenia. Avanafil (1) is a PDE5 inhibitor (treatment for erectile dysfunction) discovered by our company. We paid attention to the homology of PDE10A and PDE5 and took advantage of PDE5 inhibitor library to discover PDE10A inhibitors, and found a series of compounds that exhibit higher potency for PDE10A than PDE5. We transformed the afforded derivatives, which had weak inhibitory activity against PDE10A, and discovered stilbene as a PDE10A inhibitor. Brain penetration of this compound was improved by further conversion of N-containing heterocycles and their substituents. The afforded dimethylaminopyrimidine was effective for rat conditioned avoidance response (CAR) test; however, it did not exhibit good brain penetration. We performed in-depth optimization focusing on substituents of the quinoxaline ring, and produced 3-methyl-7-fluoro quinoxaline. This compound was the most effective in rat CAR test due to its strong PDE10A inhibitory activity and good pharmacokinetics.

Transplantation of Neuronal Precursors Derived from Induced Pluripotent Stem Cells into the Striatum of Rats with the Toxin-induced Model of Huntington’s Disease

Introduction. Huntington’s disease (HD) is a severe neurodegenerative disorder characterized by choreic hyperkinesis, cognitive decline, behavioral disorders, and progressive neuronal death, mostly in the striatum. Since HD is a fatal disorder, searching for efficient treatment methods, including those based on cell replacement therapy, is quite relevant. The experimental models of HD are used increasingly often. The objective of the study was to assess effectiveness and safety of transplantation of neuronal precursors differentiated from induced pluripotent stem cells (iPSCs) from a healthy donor into the striatum of rats with 3-NPAinduced HD model. Materials and methods. We studied the influence of neurotransplantation on the behavioral effects in rats with HD model induced by intrastriatal injection of 3-nitropropiotic acid (3-NPA). In the study group of animals (n = 11), human neuronal precursors derived from iPSCs of a healthy volunteer were transplanted into the caudate nuclei (5 × 105 per 5 μL of normal saline solution bilaterally); the control group of animals (n = 10) received normal saline solution. The animals were tested using the ANY-maze video tracking system; the parameters of the open-field test and the conditioned avoidance response test were evaluated. Results. An analysis of behavioral effects after transplantation demonstrated that introduction of neuronal iPSC derivatives into the caudate nuclei of rats with induced HD model was accompanied by recovery of locomotor activity of the animals (horizontal and vertical), as opposed to the control group. It was found when testing the reproducibility of the conditioned avoidance responses that the conditioned avoidance responses in control animals were weakened, whereas intrastriatal transplantation of neurons abruptly increased the latency of moving into the dark compartment of the chamber in the conditioned avoidance response test. Conclusions. The pilot experiment using the HD model showed that neurotransplantation using iPSC derivatives recovers the reduced locomotor activity in rats and improves memory trace keeping, which contributes to correction of locomotor and cognitive disorders induced by 3-NPA neurotoxin.

Significance of the nicotinic alpha7 receptor in cognition and antipsychotic-like behavior in the rat

Schizophrenic (SCH) patients show cognitive impairment in attentional performance. Positive allosteric modulators (PAMs) of α7 nicotinic acetylcholine receptors (nAChRs) such as the Alzheimer’s drug galantamine (GAL) and PAM-2 are documented to have pro-cognitive properties. However, it is not well established if these properties would be lost, or may hamper antipsychotic efficacy, when given as an adjunct to an antipsychotic which is needed for managing psychotic symptoms. Using adult male Wistar rats, we here investigated the effects of: a) GAL, alone or co-administered with the antipsychotic risperidone (RISP), on acute phencyclidine (PCP)-induced deficits in the attentional set-shifting (ASST) test; b) PAM-2, alone and co-administered with RISP, in the conditioned avoidance response (CAR) test for antipsychotic activity.Acute PCP produced selective and significant SCH-like impairment in extra dimensional shift (EDS) performance, which was completely reversed by GAL. The ability of GAL to reverse PCP-induced EDS impairment was not prevented when co-administered with RISP, suggesting that the combination of GAL and low doses of RISP may be used to improve the cognitive impairment in SCH. Pretreatment with methyllycaconitine (MLA), a selective α7 nAChR antagonist, completely prevented the reversal elicited by GAL, supporting the concept that α7 nAChRs are involved in this process. On the other hand, PAM-2 alone had no effects on CAR, but enhanced, although not significantly, the antipsychotic-like effect of RISP when administered together. In conclusion, α7 PAMs, in addition to alleviate the cognitive impairments observed in SCH patients, may enhance the antipsychotic efficacy of atypical antipsychotics.

Sex differences in aripiprazole sensitization from adolescence to adulthood

The present study investigated the potential sex differences in repeated aripiprazole (ARI) treatment-induced behavioral sensitization from adolescence to adulthood, and to determine whether ARI sensitization can be transferred to olanzapine (OLZ) and/or clozapine (CLZ) using the conditioned avoidance response (CAR) and phencyclidine-induced (PCP) hyperlocomotion tests of antipsychotic activity. Male and female Sprague-Dawley adolescence rats (P46) were first treated with ARI (10mg/kg) for 5 consecutive days (P46–50) and tested for avoidance response and ARI-induced inhibition of PCP-induced hyperlocomotion. After they became adults (>P68), rats were challenged with ARI (1.5mg/kg, sc) (P70), OLZ (0.5mg/kg, sc; P73), CLZ (5mg/kg, sc; P76) and again with ARI (1.5mg/kg, sc; P84) and tested for avoidance response and ARI-induced inhibition of PCP-induced hyperlocomotion again. During the drug treatment period in adolescence, repeated ARI treatment suppressed avoidance response, inhibited the PCP-induced hyperlocomotion, and these effects were progressively increased across the 5-day period in both males and females, confirming the induction of ARI sensitization. On the challenge days, rats previously treated with ARI in adolescence also had significantly lower avoidance and lower PCP-induced hyperlocomotion than the previous vehicle rats, confirming the expression of ARI sensitization and its persistence into adulthood. More importantly, female rats made significantly more avoidances than males in both ARI and vehicle groups, indicating higher sensitivity to the acute and long-term effects of ARI. Further, on the OLZ and CLZ challenge days, prior ARI treatment seemed to increase sensitivity to OLZ exposure, however, this increase was not significant. Similarly, rats also showed an ARI sensitization to OLZ and CLZ on challenge days. Collectively, results from this experiment demonstrated a sex difference in response to ARI and enhanced inhibition of PCP-induced hyperlocomotion in animals that were pretreated with ARI as compared to controls.
