Antimicrobial Topical Polymeric Films Loaded with Acetyl-11-keto-β-boswellic acid (AKBA) and Silver Nanoparticles: Optimization, Characterization, and Biological Activity

The study examined the use of pure Acetyl-11-keto-β-boswellic acid (AKBA) and its derivatives as antimicrobial topical polymeric films. The concentrations of pure AKBA, boswellic acid (BA), and Silver Nanoparticles (AgNPs) of AKBA used were optimized at 0.05% (w/v) to assess the biopolymer characteristics. Carboxymethyl cellulose (CMC), sodium alginate (SA), and gelatin (Ge) composite were prepared in this study. The polymeric films were evaluated for their biological (antioxidant and antimicrobial activities) and mechanical characteristics such as tensile strength (TS) and elongation (%). Moreover, other parameters including water barrier properties and color attributes of the film were also evaluated. Furthermore, assessments were conducted using analytical techniques like FTIR, XRD, and SEM. Surface analysis revealed that AgNP precipitation led to a few particles in the film structure. Overall, the results indicate a relatively consistent microstructure. Moreover, due to the addition of AKBA and bioactive components, a significant decrease in TS, moisture content, water solubility, and water vapor permeation was observed. The films transparency also showed a decreasing trend, and the color analysis revealed decreasing yellowness (b*) of the films. Importantly, a significant increase in antioxidant activity against DPPH free radicals and ABTS cations was observed in the CSG films. Additionally, the AgNP-AKBA loaded films displayed significant antifungal activity against C. albicans. Moreover, the molecular docking analysis revealed the inter-molecular interactions between the AKBA, AgNPs, and composite films. The docking results indicate good binding of AKBA and silver nanoparticles with gelatin and cellulose molecules. In conclusion, these polymeric films have potential as novel materials with significant antioxidant and antifungal activities.

Silver Nanoparticles: Multifunctional Tool in Environmental Water Remediation.

Water pollution is a worldwide environmental and health problem that requires the development of sustainable, efficient, and accessible technologies. Nanotechnology is a very attractive alternative in environmental remediation processes due to the multiple properties that are conferred on a material when it is at the nanometric scale. This present review focuses on the understanding of the structure-physicochemical properties-performance relationships of silver nanoparticles, with the objective of guiding the selection of physicochemical properties that promote greater performance and are key factors in their use as antibacterial agents, surface modifiers, colorimetric sensors, signal amplifiers, and plasmonic photocatalysts. Silver nanoparticles with a size of less than 10 nm, morphology with a high percentage of reactive facets {111}, and positive surface charge improve the interaction of the nanoparticles with bacterial cells and induce a greater antibacterial effect. Adsorbent materials functionalized with an optimal concentration of silver nanoparticles increase their contact area and enhance adsorbent capacity. The use of stabilizing agents in silver nanoparticles promotes selective adsorption of contaminants by modifying the surface charge and type of active sites in an adsorbent material, in addition to inducing selective complexation and providing stability in their use as colorimetric sensors. Silver nanoparticles with complex morphologies allow the formation of hot spots or chemical or electromagnetic bonds between substrate and analyte, promoting a greater amplification factor. Controlled doping with nanoparticles in photocatalytic materials produces improvements in their electronic structural properties, promotes changes in charge transfer and bandgap, and improves and expands their photocatalytic properties. Silver nanoparticles have potential use as a tool in water remediation, where by selecting appropriate physicochemical properties for each application, their performance and efficiency are improved.

Electrical/thermal conductivities of low-temperature sintered/Ag-decorated epoxy microspheres

The last decades composite production has benefited from additive manufacturing rendering production of special parts feasible and economical. Energy and time-consuming processes have improved significantly since the implementation of low temperature sintering methods. In this work, we prepare sub-micron powders in an attempt to produce conductive composites via compression sintering at a low overall production cost. Epoxy microspheres of glassy and rubbery type are synthesized in a controlled manner to inherit certain properties, and then decorated via a controlled deposition process of silver nanoparticles to form fine powders. The resulting fine powders are compressed at room temperature conditions to consolidate and hopefully sinter at post-cure. We study the structure-property relationship of the powder structure and the effect in the conductive properties that arise after consolidation. Remarkably, the compressed powders achieved DC conductivity with extremely low silver nanoparticle content (7.7 wt %, approx. 0.9 vol %) as compared with other literature reports and a promising increase in thermal diffusivity of about 18 % with just 4.2 wt % of silver nanoparticles.

Antibacterial activity and biocompatibility of titanium nickelide augments with the addition of silver nanoparticles for bone grafting: an experimental study

BACKGROUND: The relevance of this study was supported by the increasing number of infectious complications of bone augmentation in children and adults. Currently, porous titanium nickelide alloys are among the most preferred materials used in bone plasty. Despite the observable advantages of porous nickelide titanium alloys in terms of biochemical and biomechanical compatibility with the body, research on the antibacterial activity of alloys is ongoing to counter the development of infections at the implant–biological tissue border.
 AIM: To perform an experimental study of the biocompatible antibacterial surface in porous titanium nickelide alloys with the addition of silver nanoparticles.
 MATERIALS AND METHODS: Titanium nickelide alloys with 62% porosity were obtained using the self-propagating high-temperature synthesis method from nickel, titanium, and nanosilver powders at concentrations of 0.2 at.% Ag, 0.5 at.% Ag, and 1.0 at.% Ag, respectively. The experiment was conducted on nine sexually mature female white laboratory rats. They were divided into three groups, with three rats each. All animals were implanted with titanium nickelide along with porous granules of silver additives. The first group was the control, the second received 0.2 at.% silver, and the third received 0.5% silver. The standard method of incubating Staphylococcus epidermidis in liquid broth in the presence of the studied images was used to determine bactericidal activity, followed by seeding on solid media and counting colonies.
 RESULTS: The antibacterial effect of the samples on S. epidermidis gradually increased with increasing silver concentration. The significance of the differences between the experiment and control was confirmed by Student’s criterion p 0.005, whereas the sample without silver nanoparticles and the control do not differ significantly. Thus, these alloys may have bioactive properties because they contain silver nanoparticles. An alloy with a silver concentration of 0.5 at.% Ag showed the best antibacterial activity to S. epidermidis. In the clinical evaluation of the results of the experimental study, purulent inflammatory complications were not observed in all animals at all times. On day 75, the animals underwent computed tomography, which showed good occupancy of the bone defect and absence of a dystrophic effect on the area where the bone and soft tissue are in contact with the material.
 CONCLUSIONS: If the concentration of silver nanoparticles is increased up to 0.5 at%, the antibacterial activity and cytocompatibility of the implant also increase. Clinical experimental evaluation in all groups of animals showed that osteointegration of alloys with 0.5 at.% Ag begins immediately after implantation and is completed 2 weeks earlier than that in the remaining groups.

XMEA: A New Hybrid Diamond Multielectrode Array for the In Situ Assessment of the Radiation Dose Enhancement by Nanoparticles.

This work presents a novel multielectrode array (MEA) to quantitatively assess the dose enhancement factor (DEF) produced in a medium by embedded nanoparticles. The MEA has 16 nanocrystalline diamond electrodes (in a cell-culture well), and a single-crystal diamond divided into four quadrants for X-ray dosimetry. DEF was assessed in water solutions with up to a 1000 µg/mL concentration of silver, platinum, and gold nanoparticles. The X-ray detectors showed a linear response to radiation dose (r2 ≥ 0.9999). Overall, platinum and gold nanoparticles produced a dose enhancement in the medium (maximum of 1.9 and 3.1, respectively), while silver nanoparticles produced a shielding effect (maximum of 37%), lowering the dose in the medium. This work shows that the novel MEA can be a useful tool in the quantitative assessment of radiation dose enhancement due to nanoparticles. Together with its suitability for cells' exocytosis studies, it proves to be a highly versatile device for several applications.

Plackett–Burman screening of physico-chemical variables affecting Citrus peel-mediated synthesis of silver nanoparticles and their antimicrobial activity

With the growing resistance of pathogenic microbes to traditional drugs, biogenic silver nanoparticles (SNPs) have recently drawn attention as potent antimicrobial agents. In the present study, SNPs synthesized with the aid of orange (Citrus sinensis) peel were engineered by screening variables affecting their properties via Plackett–Burman design. Among the variables screened (temperature, pH, shaking speed, incubation time, peel extract concentration, AgNO3 concentration and extract/AgNO3 volume ratio), pH was the only variable with significant effect on SNPs synthesis. Therefore, SNPs properties could be enhanced to possess highly regular shape with zeta size of 11.44 nm and zeta potential of − 23.7 mV. SNPs purified, capped and stabilized by cloud point extraction technique were then checked for their antimicrobial activity against Bacillus cereus, Listeria innocua, Listeria monocytogenes, Staphylococcus aureus, Enterobacter cloacae, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Salmonella typhimurium and Candida albicans. The maximum antimicrobial activity of SNPs was recorded against E. coli, L. monocytogenes and C. albicans with clear zone diameter of 33.2, 31.8 and 31.7 mm, respectively. Based on minimum inhibition concentration and minimum bactericidal concentration of SNPs (300 mg/l) as well as their effect on respiratory chain dehydrogenases, cellular sugar leakage, protein leakage and lipid peroxidation of microbial cells, E. coli was the most affected. Scanning electron microscopy, protein banding and DNA fragmentation proved obvious ultrastructural and molecular alterations of E. coli treated with SNPs. Thus, biogenic SNPs with enhanced properties can be synthesized with the aid of Citrus peel; and such engineered nanoparticles can be used as potent antimicrobial drug against E. coli.

Evaluation of Anticancer Activity and Structural Analysis of Biosynthesized Silver Nanoparticles (AgNPs) from Centella Asiatica

Introduction: The synthesis of silver nanoparticles (AgNPs) from an ethanolic extract of Centella asiatica leaf, which exhibits anticancer characteristics, was investigated in this study. Green synthesis, a non-harmful procedure for the environment, aids in the production of nanoparticles from Centella asiatica extract. Centella asiatica has long been regarded to have beneficial properties for neurological function, anti-inflammation, and wound treatment. Objective: FT-IR, GC-MS, and thin-layer chromatography were used to find the phytochemicals present in the extract, and the anti-cancer abilities of the nanoparticles were tested against the PC3 cell line. Methods: Soxhlet extraction is used to extract the bioactive, and then nanoparticles are synthesised. Following that, they are subjected to preliminary phytochemical analysis and thin-layer chromatography. These can assess the presence or absence of phytochemicals. Soon after, GCMS is done to reveal the molecular weight of the bioactive compounds using the charge-to-mass ratio. The structural peaks of different functional groups were identified using FT-IR. Under different concentrations, the silver nanoparticles were portrayed against the PC3 cell line of prostate cancer. Result: The concentration of silver nanoparticles was shown to negatively affect cell viability. The IC50 value was 11.8 µg/ml, showing that the inhibition of growth increased with increasing doses of the AgNPs. Conclusion: Our study highlights the significance of green synthesis of AgNPs from Centella asiatica extract, with bioactive compounds identified using GC-MS showing potential as anticancer agents against PC3 cells, suggesting a promising avenue for cancer therapy.

Antibacterial effect and biocompatibility of silver nanoparticle-coated bone allograft substitutes.

Osteoinduction, and/or osteoconduction, and antibacterial characteristics are prerequisites for achieving successful bone grafting. This study aimed to coat bone allografts with silver nanoparticles and assess their antibacterial activity and biocompatibility compared to uncoated bone allografts. In this study, the bone allografts were coated with varying concentrations of silver nanoparticles (5 mg/l, 10 mg/l, and 50 mg/l) through a simple adsorption technique. Subsequently, the coated samples underwent characterization using SEM, FTIR, EDS, and XRD. The Minimal Inhibitory Concentration (MIC) of the silver nanoparticles was determined against Staphylococcus aureus and Streptococcus mutans. Bacterial growth inhibition was evaluated by measuring turbidity and performing a disk diffusion test. Moreover, qualitative investigation of biofilm formation on the coated bone allograft was conducted using SEM. Following this, MG-63 cell lines, resembling osteoblasts, were cultured on the bone allografts coated with 5 mg/l of silver nanoparticles, as well as on uncoated bone allografts, to assess biocompatibility. The MIC results demonstrated that silver nanoparticles exhibited antimicrobial effects on both microorganisms, inhibiting the growth of isolates at concentrations of 0.78 mg/L for Staphylococcus aureus and 0.39 mg/L for Streptococcus mutans. The bone allografts coated with varying concentrations of silver nanoparticles exhibited significant antibacterial activity against the tested bacteria, completely eradicating bacterial growth and preventing biofilm formation. The osteoblast-like MG-63 cells thrived on the bone allografts coated with 5 mg/l of silver nanoparticles, displaying no significant differences compared to both the uncoated bone allografts and the control group. Within the limit of this study, it can be concluded that silver nanoparticles have a positive role in controlling graft infection. In addition, simple adsorption technique showed an effective method of coating without overwhelming the healing of the graft.

Development of antibacterial collagen membranes with optimal silver nanoparticle content for periodontal regeneration

The effective control of pathogenic bacteria is crucial in the restoration of periodontal tissue affected by periodontitis. Guided tissue regeneration (GTR) membranes are commonly used to aid in the repair of periodontal defects. Therefore, there is a clear advantage in developing antibacterial periodontal membranes that can effectively eliminate infections and promote tissue regeneration. This study aimed to create a collagen membrane with optimal content of silver nanoparticles (AgNPs) for effective antibacterial properties and minimal toxicity to mammalian cells. Ascorbic acid-reduced AgNPs were incorporated into collagen at the ratio of 0.5%, 1%, 2%, and 3% (based on total dry weight). Collagen/AgNPs hydrogels were compressed and freeze-dried to form membranes and then were characterized. Antibacterial activity was tested against Fusobacterium nucleatum and Enterococcus faecalis, and membrane cytocompatibility was accomplished on human gingival fibroblasts. Membranes with 2% and 3% AgNPs exhibited significant antibacterial activity, while 1% showed minimal activity and 0.5% and 0% showed none. HGF cells on the 3% AgNPs membrane had poor viability, proliferation, and adhesion, but 0%, 0.5%, 1%, and 2% AgNPs membranes showed desirable cellular behavior. In conclusion, the collagen membrane with 2% AgNPs demonstrated both antibacterial capacity and excellent cytocompatibility, making it a promising choice for periodontal treatments, especially in GTR approaches.

Alginate Silver Nanoparticles and Their Effect on Sperm Parameters of the Domestic Rabbit

Silver nanoparticles possess valuable physical, chemical, and biological properties, rendering them widely applied as bioactive agents in the industry. Nonetheless, their influence on the natural environment and on living organisms remains unclear. Therefore, this study aims to investigate the impact of polymer composites containing silver nanoparticles on sperm cells. The nanosilver polymer composites were chemically synthesized, employing sodium alginate as the stabilizer. The reducing agents employed were solutions comprising sodium borohydride and xylose. The concentration of silver nanoparticles obtained after synthesis was 100 parts per million. The examined biological species were rabbit sperm cells. The impact of nanosilver on the sperm was assessed through the elucidation of the toxicity profile, comet test, and analysis of morphological characteristics of the animal cells. The results of the study demonstrate a twofold impact of polymer composites infused with silver nanoparticles on domestic rabbit sperm when obtained through chemical synthesis using two reducing agents (xylose and sodium borohydride) at a 10 ppm concentration. The comet test showed no harmful effect on the DNA integrity of rabbit sperm by the tested compounds. Twenty-four-hour exposure of rabbit spermatozoa to silver nanoparticles, obtained by reducing xylose and borohydride, induced significant secondary changes in the morphological structure of male reproductive cells. These findings indicate the potential reproductive toxicity of silver nanoparticles.

The Effect of Silver Nanoparticles on the Electrical Activity of PC12 Quasi-Neuronal Networks and the Comparison of Two Microelectrode Array-Based Neural Signal Analysis Methods

Our research group previously proposed the voltage thresholdmeasurement method (VTMM) based on the microelectrode array (MEA), and used VTMM to prove that PC12 cells, as a simpler cell culture, can be used as an alternative cell model to examine the effects of external factors on cytotoxicity and electrical excitability of neurons. However, there is no comparison between VTMM and the commonly used MEA neural signal analysis method, and no researchers have used the commonly used analysis method to study the spontaneous electrical signals of PC12 quasi-neuronal networks. This research aims to compare the characteristics and results of two MEA-based neural signal analysis methods. For the first time, the changes in spike frequency of the PC12 networks after exposure to various concentrations of silver nanoparticles for different durations was studied by means of MEA analysis method. It is found that due to the low intensity of spontaneous activity of PC12 networks, the commonly used analysis method is not suitable for the study of electrical activity of PC12 networks. Under the action of silver nanoparticles, the results of VTMM are consistent with those of the commonly used MEA analysis method, and VTMM has the advantages of being suitable for more kinds of neuron networks, short culture time, concise and rapid data analysis.

Metal nanoparticles to improve the heat resilience in wheat (Triticum aestivum L.)

This study evaluated the efficacy of phytogenic silver and zinc nanoparticles in improving heat resilience in various wheat varieties. The silver and zinc nanoparticles were synthesized using plant leaf extract and characterized using various techniques. Four wheat varieties (DBW187, Black Wheat, DBW 50, and PBW 621) were subjected to field trials. The random block design was used, and nanoparticles in different concentrations were applied at various growth stages and morphologically, and yield parameters were recorded. UV–vis spectroscopy spectral analysis showed peaks for Ag nanoparticles at 420 nm wavelength and Zn nanoparticles at 240 and 350 nm wavelength, depicting the preliminary confirmation of nanoparticle synthesis. Electron microscopic analysis (TEM and SEM) provided morphological insights and confirmed synthesis of fine-sized particle mostly in a range between 10 and 60 nm. Energy dispersive x-ray analysis confirmed the elemental composition of the synthesized nanoparticles, with Ag and Zn elements detected in their respective samples. It also confirmed the oxide nature of synthesized ZnNPs. Dynamic light scattering analysis provided size distribution profiles, indicating average sizes of approximately 61.8 nm for Ag nanoparticles and 46.5 nm for Zn nanoparticles. The concentrations of Ag and Zn nanoparticles in the samples were found to be 196.3 ppm and 115.14 ppm, respectively, through atomic absorption spectroscopic analysis. Fourier transform infrared spectroscopy analysis revealed characteristic functional groups present in the nanoparticles. The results of field experiments established that Ag nanoparticles at 75 ppm concentration exhibited the most significant enhancements in plant growth. Conversely, Zn nanoparticles at a 100 ppm concentration demonstrated the most substantial improvements in the growth and yield of heat-stressed wheat varieties. The study concludes that optimized concentrations of silver and zinc nanoparticles can effectively improve heat stress resilience in wheat. These findings are promising to enhance abiotic stress resilience in crops.

The biochemical and growth-associated traits of basil (Ocimum basilicum L.) affected by silver nanoparticles and silver

BackgroundThe biochemical and growth changes resulting from exposure of basil (Ocimum basilicum L.) seedlings to silver nanoparticles and silver were investigated. Over a two-week period, seedlings were exposed to different concentrations (0, 40, and 80 ppm) of silver nanoparticles and silver.ResultsOur findings revealed that at concentrations of 40 and 80 ppm, both silver nanoparticles and silver nitrate led to decreased weight, root and shoot length, as well as chlorophyll a and b content. Conversely, these treatments triggered an increase in key biochemical properties, such as total phenols, carotenoids and anthocyanins, with silver nanoparticles showing a more pronounced effect compared to silver nitrate. Moreover, the levels of malondialdehyde (MDA) and hydrogen peroxide (H2O2) rose proportionally with treatment concentration, with the nanoparticle treatment exhibiting a more substantial increase. Silver content showed a significant upswing in both roots and leaves as treatment concentrations increased.ConclusionsApplication of varying concentrations of silver nanoparticles and silver nitrate on basil plants resulted in reduced growth and lower chlorophyll content, while simultaneously boosting the production of antioxidant compounds. Notably, anthocyanin, carotenoid, and total phenol increased significantly. However, despite this increase in antioxidant activity, the plant remained unable to fully mitigate the oxidative stress induced by silver and silver nanoparticles.

Analysis of antibiofilm activity gold and silver nanoparticles against uropathogenic Escherichia coli using confocal laser scanning microscopy

Infectious diseases, including urinary tract infections (UTIs), significantly impact global health, diminishing the quality of life and increasing clinical and economic burdens. Concerns arise due to the potential development of antimicrobial resistance (AMR) facilitated by biofilm formation. Nanotechnology, specifically silver and silver nanoparticles (1–100 nm), proves effective against bacterial strains, making them valuable in the biomedical field. Advances in microscopic imaging techniques, such as Confocal Laser Scanning Microscopy (CLSM), enhance our understanding of biofilms. This study aims to assess the effectiveness and optimal concentration of gold and silver nanoparticles in combating biofilms formed by uropathogenic Escherichia coli (UPEC) through CLSM analysis. The research comprises the following stages: rejuvenation of bacterial isolates and inoculum preparation, testing the anti-biofilm activity of gold and silver nanoparticles at concentrations of 25, 50, 75, and 100 ppm against UPEC using CLSM analysis. The findings reveal that both gold and silver nanoparticles exhibit anti-biofilm activity against UPEC at concentrations of 25, 50, 75, and 100 ppm, as confirmed by CLSM analysis. The optimal concentration for anti-biofilm activity of gold and silver nanoparticles against UPEC is identified as 100 ppm.

Antimicrobial Activity of Silver Nanoparticles Synthesized Using Ocimum tenuiflorum and Ocimum gratissimum Herbal Formulations.

Background The exploration of green synthesis for silver nanoparticles using diverse plant sources has gained significant attention. This study specifically investigates the use of Ocimum tenuiflorum and Ocimum gratissimum, known for their antibacterial properties, in synthesizing silver nanoparticles. The primary aim of this study is to evaluate the antimicrobial efficacy of silver nanoparticles synthesized using a herbal formulation composed of Ocimum tenuiflorum and Ocimum gratissimumagainst different oral pathogens. Materials and methods The process involved the combination of herbal extracts from Ocimum tenuiflorumand Ocimum gratissimumwith a silver nitrate solution leading to the synthesis of silver nanoparticles. The formation of silver nanoparticles was confirmed by ultraviolet and visible absorption spectroscopy. The obtained silver nanoparticles were used to study their antimicrobial activity. Antimicrobial activity was assessed using the agar well diffusion method against pathogens including Streptococcusmutans, Enterococcus faecalis, C. albicans, Lactobacillus acidophilus, and S. aureus. The zone of inhibition quantified antimicrobial effectiveness. A time-kill curve assay evaluated bactericidal properties and the concentration-dependent relationship between silver nanoparticles and the net growth rate of oral pathogens. Results Statistical analysis was done to compare measures such as mean, standard deviation, and percentages. The antimicrobial assessment demonstrated that 100 μg/mL of silver nanoparticles exhibited the highest efficacy against S. mutans, S. aureus, E. faecalis, and Lactobacillus sp. For C. albicans, all concentrations of silver nanoparticles and the control plant extract displayed similar antimicrobial activity. The time-kill assay illustrated effective inhibition at 100 μg/mL against all tested pathogens, including S. mutans, S. aureus, E. faecalis, C. albicans, and Lactobacillus sp.The result showed positive inhibitory activity of silver nanoparticles against all tested bacterial strains. Conclusion The significant antimicrobial efficacy of green-synthesized silver nanoparticles positions them as promising candidates for dental applications. Their demonstrated bactericidal and fungicidal activities suggest potential use as effective dental antimicrobial agents, opening avenues for innovative solutions in oral healthcare.

Determining the cytotoxicity of the Minimum Inhibitory Concentration (MIC) of silver and zinc oxide nanoparticles in ESBL and carbapenemase producing Proteus mirabilis isolated from clinical samples in Shiraz, Southwest Iran

ObjectiveProteus mirabilis is related to serious infections. The present study was designed to investigate the minimum inhibitory concentration (MIC) of silver nanoparticles (AgNPs) and zinc oxide nanoparticles (ZnONPs) and cytotoxicity among P. mirabilis isolates recovered from clinical samples in Shiraz.ResultsA total of 100 P. mirabilis isolates were screened by biochemical tests and polymerase chain reaction (PCR). Also, 25 (25%) and 7 (7%) isolates were positive for extended-spectrum beta-lactamase (ESBLs) and carbapenemase, respectively. Synthesized nanoparticles were characterized by UV–vis spectrum, X-ray diffraction (XRD), and electron microscopy. The average size of AgNPs and ZnONPs in the present study is 48 and < 70 nm, respectively. The MIC and the MBC of the ZnONPs were in the range of 31.25 µg/ml and 62.5 µg/mL, respectively. Also, for AgNPs, the MIC and the MBC were in the range of 7.8 µg/mL and 15.6 µg/mL, respectively. MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay in a primary culture of fibroblast L929 cells for this MIC indicated biocompatibility and low cytotoxicity of Ag NPs and for ZnONPs indicated significant cytotoxicity. Also, a MIC of AgNPs can be used as a therapeutic concentration without the effect of cytotoxicity in human cells.

Antibacterial effect of red laser-activated silver nanoparticles synthesized with grape seed extract against Staphylococcus aureus and Escherichia coli.

Living organisms, particularly humans, frequently encounter microorganisms such as bacteria, fungi, and viruses in their surroundings. Silver nanoparticles are widely used in biomedical devices because of their antibacterial and antiviral properties. The study evaluates the efficacy of red laser and silver nanoparticles from grape seed extract (AgNPs-GSE) in reducing Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus bacteria, which cause infections. The sample comprised three groups: a control group without laser irradiation (T0), Escherichia coli samples (A1 and A2) irradiated with a 405-nm diode laser at different times and concentrations of silver nanoparticles, and Staphylococcus aureus samples (A3 and A4) illuminated with a 405-nm diode laser at different times and concentrations. Bacteria in groups A2 and A4 were treated with a photosensitizer (PS) made from grape seed extracts, incubated for 10min, and then irradiated for 90, 120, 150, and 180s. The samples were cultured on TSA media, set at 37°C, counted using a Quebec colony counter, and analyzed using ANOVA and Tukey tests with a significance level of p < 0.05. The study illustrated that the combination of 10µl of AgNPs-GSE, exposure to a red laser at 405nm, and an energy density of 3.44J/cm2 effectively photoinactivated both Escherichia coli and Staphylococcus aureus bacteria. For Escherichia coli bacteria irradiated for 180s with concentrations of 1mM, 1.5mM, and 2mM AgNPs-GSE, bacterial viability decreased by 64.50%, 70.74%, and 79.53%, respectively. Similarly, Staphylococcus aureus bacteria, subjected to irradiation for 180s with concentrations of 1mM, 1.5mM, and 2mM AgNPs-GSE, demonstrated reductions in bacterial viability by 70.23%, 73.47%, and 85.04%, respectively. The findings from the present study indicate that at an energy density of 3.44J/cm2, it was possible to inactivate Escherichia coli by 79.53% and Staphylococcus aureus by 85.04%.

Modulation of Staphylococcus aureus Biofilm Formation through Subinhibitory Concentrations of Biogenic Silver Nanoparticles and Simvastatin

Staphylococcus aureus is a causative agent of nosocomial infections and its antibiotic-resistant strains give cause for concern. Solutions are being explored to improve treatment for these infections, including repositioning drugs such as statins and using nanoparticles with antimicrobial properties. This study evaluated the antimicrobial effects of simvastatin (SIM) and biologically synthesized silver nanoparticles (bio-AgNPs) in isolate form and in combination using assays of minimum inhibitory concentration (MIC), an in vitro biofilm model, and the association of antimicrobials against clinical strains of S. aureus. Bio-AgNPs showed a 53.8 ± 1.23 nm mean diameter and standard deviation, a 0.23 polydispersity index, and a −25.66 ± 2.19 mV mean potential and standard deviation. Transmission electron microscopy confirmed the formation of nanoparticles, and the presence of Ag0 and AgCl. S. aureus strains were sensitive to bio-AgNPs and SIM, showing 31.88–187.5 and 74.66–149.32 μM concentrations, respectively. The association assay showed 2.0 fractional inhibitory concentration indices (i.e., indifferent for clinical strains) and 0.32 values for the standard ATCC 29213 strain (synergy). Biofilm inhibition assays with isolated SIM and bio-AgNPs showed decreased biofilm formation 4× to ⅛ MICs concentrations, showing no synergism in association. These findings evince that simvastatin and bio-AgNPs at subinhibitory concentrations can serve as antimicrobial agents against S. aureus biofilm.

Blood cell alterations in Colossoma macropomum juveniles caused by silver nanoparticles.

This study evaluated the median lethal concentration of silver nanoparticles and their effects in fish tambaqui Colossoma macropomum. Therefore, an acute toxicity assay was carried out in completely randomized design evaluating six different concentrations of silver nanoparticles on blood parameters of tambaqui. The silver nanoparticles were produced by chemical reduction with polyvinyl alcohol (AgNP-PVA). The lethal concentration 50% (LC50) was estimated using probit regression. The blood was collected, analyzed and the data were submitted to T-test (dying x surviving fish) and Tukey test (surviving fish). An increase in glucose, hematocrit, total plasma protein, hemoglobin, erythrocytes, leukocytes, monocytes, and neutrophils as well as reduced MCV (mean corpuscular volume) in dying fish compared to surviving fish were observed. Survived fish exposed to 187.5 µg/L showed an increase in hematocrit, MCV, and MCH and a reduction in erythrocytes, total numbers of leukocyte, thrombocyte, lymphocyte, and neutrophil. The fish exposed to concentrations below 125 µg/L, had returned the blood parameter to baselines compared to control. The estimated LC50 was 165.09 µg/L and was classified as highly toxic for the fish tambaqui. In higher concentrations, it causes an acute respiratory toxicity, but in concentrations below 125 µg/L, the fish can adapt to the stressing agent.

The Effect of Dose Enhancement in Tumor With Silver Nanoparticles on Surrounding Healthy Tissues: A Monte Carlo Study.

Objectives: Cancer-related death rates account for approximately one-third of all deaths, and this rate is increasing remarkably every year. In this study, we examined the dose enhancement factor (DEF) in the tumor and surrounding tissues by adding different concentrations of silver nanoparticles (AgNPs) to the brain tumor using the Monte Carlo (MC) technique. Methods: This study used MCNP6.2 simulation software. A Planning Target Volume (PTV) of 1 × 1 × 1 cm3 was placed in the center of a cubic cranial model with dimensions of 5 × 5 × 5 cm3. Five different simulations were initially generated using the simple method. These simulations included pure PTV and PTV consisting of 4 different silver concentrations (5, 10, 20, and 30 mg/g). Additionally, a model was created using the nanolattice method, considering the size, position, and distribution of the AgNPs. Irradiation was performed using a source with a 6 MV linac photon spectrum. Measurements were performed using the *f8 tally, and DEF values were calculated. Results: In the simulation study using the simple method, the DEF value of PTV increased linearly with concentration, whereas the DEF values were lower than the simulation results with the nanolattice model (1.9 vs 1.4 for 30 mg/g NP concentration). Performing the simple method, we observed no remarkable dose increase in lateral OARs surrounding PTV. While a remarkable dose decrease was observed in distal OARs, a dose increase in the proximal OAR was observed, which was consistent with that of PTV. However, according to the results obtained by performing the nanolattice method, the dose increase was observed in both the proximal OAR and the distal OAR and was similar to that of PTV. Conclusion: While enhancing the dose in the tumor by adding NPs into the tumor, it is essential to consider whether it also increases the OAR dose. In addition, simulation studies on NPs showed that the dose increase varied significantly with particle size, position, and distribution. Hence, these factors should be considered carefully.
