Time Is Ripe for Targeting Per- and Polyfluoroalkyl Substances-Induced Hormesis: Global Aquatic Hotspots and Implications for Ecological Risk Assessment.

Globally implemented ecological risk assessment (ERA) guidelines marginalize hormesis, a biphasic dose-response relationship characterized by low-dose stimulation and high-dose inhibition. The present study illuminated the promise of hormesis as a scientific dose-response model for ERA of per- and polyfluoroalkyl substances (PFAS) represented by perfluorooctanoic acid (PFOA) and perfluorooctanesulfonic acid (PFOS). A total of 266 hormetic dose-response relationships were recompiled from 1237 observations, covering 30 species from nine representative taxonomic groups. The standardized hormetic amplitudes followed the log-normal probability distribution, being subject to the limits of biological plasticity but independent of stress inducers. The SHapley Additive exPlanations algorithm revealed that the target endpoint was the most important variable explaining the hormetic amplitudes. Subsequently, quantitative frameworks were established to incorporate hormesis into the predicted no-effect concentration levels, with a lower induction dose and a zero-equivalent point but a broader hormetic zone for PFOS. Realistically, 10,117 observed concentrations of PFOA and PFOS were gathered worldwide, 4% of which fell within hormetic zones, highlighting the environmental relevance of hormesis. Additionally, the hormesis induction potential was identified in other legacy and emerging PFAS as well as their alternatives and mixtures. Collectively, it is time to incorporate the hormesis concept into PFAS studies to facilitate more realistic risk characterizations.

Separation of Perfluorooctanoic Acid from Water Using Meso- and Macroporous Syndiotactic Polystyrene Gels.

Per- and polyfluoroalkyl substances are an emerging class of contaminants that are environmentally persistent, bioaccumulative, and noxious to human health. Among these, perfluorooctanoic acid (PFOA) molecules are widely found in ground and surface water sources. A novel high surface area, meso- and macroporous syndiotactic polystyrene (sPS) wet gel is used in this work as the adsorbent of PFOA molecules from water at environmentally relevant PFOA concentrations (≤1 μg/L) and cleanse water to below the U.S. EPA's 2023 health advisory limit of 4 parts per trillion (ppt). The sigmoidal shape of the PFOA adsorption isotherm indicates a two-step adsorption mechanism attributed to the strong affinity of PFOA molecules for the sPS surface and molecular aggregation at solid-liquid interfaces or within the pores of the sPS wet gel. The adsorption kinetics and the effects of sPS wet gel porosity, pore size, and pore volume on the removal efficiency are reported. The adsorption kinetics is seen to be strongly dependent on pore size and pore volume.

Perfluorooctanoic acid-contaminated wastewater treatment by forward osmosis: Performance analysis

Perfluorooctanoic acid (PFOA) is a persistent compound, raising considerable global apprehension due to its resistance to breakdown and detrimental impacts on human health and aquatic environments. Pressure-driven membrane technologies treating PFAS-contaminated water are expensive and prone to fouling. This study presented a parametric investigation of the effectiveness of cellulose triacetate membrane in the forward osmosis (FO) membrane for removing PFOA from an aqueous solution. The study examined the influence of membrane orientation modes, feed pH, draw solution composition and concentration, and PFOA concentration on the performance of FO. The experimental results demonstrated that PFOA rejection was 99 % with MgCl2 and slightly >98 % with NaCl draw solutions due to the mechanism of PFOA binding to the membrane surface through Mg2+ ions. This finding highlights the crucial role of the draw solution's composition in PFOA treatment. Laboratory results revealed that membrane rejection of PFOA was 99 % at neutral and acidic pH levels but decreased to 95 % in an alkaline solution at pH 9. The decrease in membrane rejection is attributed to the dissociation of the membrane's functional groups, consequently causing pore swelling. The results were confirmed by calculating the average pore radius of the CTA membrane, which increased from 27.94 nm at pH 5 to 30.70 nm at pH 9. Also, variations in the PFOA concentration from 5 to 100 mg/L did not significantly impact the membrane rejection, indicating the process's capability to handle a wide range of PFOA concentrations. When seawater was the draw solution, the FO membrane rejected 99 % of PFOA concentrations ranging from 5 mg/L to 100 mg/L. The CTA FO treating PFOA-contaminated wastewater from soil remediation achieved a 90 % recovery rate and water flux recovery of 96.5 % after cleaning with DI water at 40 °C, followed by osmotic backwash. The results suggest the potential of using abundant and cost-effective natural solutions in the FO process, all without evident membrane fouling.

Early-life exposure to per- and polyfluoroalkyl substances: Analysis of levels, health risk and binding abilities to transport proteins

Per- and polyfluoroalkyl substances (PFAS) can pass through the placenta and adversely affect fetal development. However, there is a lack of comparison of legacy and emerging PFAS levels among different biosamples in pregnant women and their offspring. This study, based on the Shanghai Maternal-Child Pairs Cohort, analyzed the concentrations of 16 PFAS in the maternal serum, cord serum, and breast milk samples from 1,076 mother-child pairs. The placental and breastfeeding transfer efficiencies of PFAS were determined in maternal-cord and maternal-milk pairs, respectively. The binding affinities of PFAS to five transporters were simulated using molecular docking. The results suggested that PFAS were frequently detected in different biosamples. The median concentration of perfluorooctane sulfonate (PFOS) was the highest at 8.85 ng/mL, followed by perfluorooctanoic acid (PFOA) at 7.13 ng/mL and 6:2 chlorinated polyfluorinated ether sulfonate at 5.59 ng/mL in maternal serum. The median concentrations of PFOA were highest in cord serum (4.23 ng/mL) and breast milk (1.08 ng/mL). PFAS demonstrated higher placental than breastfeeding transfer efficiencies. The transfer efficiencies and the binding affinities of most PFAS to proteins exhibited alkyl chain length-dependent patterns. Furthermore, we comprehensively assessed the estimated daily intakes (EDIs) of PFAS in breastfeeding infants of different age groups and used the hazard quotient (HQ) to characterize the potential health risk. EDIs decreased with infant age, and PFOS had higher HQs than PFOA. These findings highlight the significance of considering PFAS exposure, transfer mechanism, and health risks resulting from breast milk intake in early life.

Temporal Trend of Serum Perfluorooctanoic Acid and Perfluorooctane Sulfonic Acid among U.S. Adults with or without Comorbidities in NHANES 1999–2018

Perfluoroalkyl and polyfluoroalkyl substances (PFASs) are associated with adverse health effects. This study examined the trend of perfluorooctanoic acid (PFOA) and perfluorooctane sulfonic acid (PFOS) levels in individuals with and without pre-existing comorbidities. We analyzed the characteristics of 13,887 participants across nine U.S. NHANES cycles (1999–2000 to 2017–2018) and calculated the geometric mean (GM) of PFOA and PFOS levels, standardized by sex and age. A joinpoint regression model was used to analyze the temporal trends of serum PFOA and PFOS levels. We observed declining PFOA and PFOS serum levels among adults in NHANES from 1999–2000 to 2017–2018. Serum PFOA and PFOS concentrations were higher in men, smokers, and individuals with pre-existing diabetes, CKD, hyperlipidemia, CVD, and cancer. We observed faster decline rates in PFOA levels among individuals with diabetes and CKD and faster decline rates in PFOS levels among individuals with diabetes and those without CKD. This study provided evidence of varying levels and changing trends of PFOA and PFOS between groups with and without established chronic disease, highlighting the role of environmental chemicals in the onset and development of chronic diseases.

Evaluation and prediction of anthropogenic impacts on long-term multimedia fate and health risks of PFOS and PFOA in the Elbe River Basin

Perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) have aroused great concern owing to their widespread occurrence and toxic effects. However, their long-term trends and multimedia fate remain largely unknown. Here, we investigate the spatiotemporal characteristics and periodic oscillations of PFOS and PFOA in the Elbe River between 2010 and 2021. Anthropogenic emission inventories and multimedia fugacity model were developed to analyse their historical and future transport fates and quantify related human risks in each medium for the three age groups. The results show that average PFOS and PFOA concentrations in the Elbe River were 4.08 and 3.41 ng/L, declining at the annual rate of 7.36% and 4.98% during the study period, respectively. Periodic oscillations of their concentrations and mass fluxes were most pronounced at 40–60 and 20–40 months. The multimedia fugacity model revealed that higher concentrations occurred in fish (PFOS: 14.29, PFOA: 0.40 ng/g), while the soil was their dominant sink (PFOS: 179, PFOA: 95 tons). The exchange flux between water and sediment was the dominant pathway in multimedia transportation (397 kg/year). Although PFOS and PFOA concentrations are projected to decrease by 22.41% and 50.08%, respectively, from 2021 to 2050, the hazard quotient of PFOS in fish is a low hazard. This study provides information for the assessment of PFOS and PFOA pollution in global watersheds and the development of related mitigation policies, such as banning fish predation in polluted rivers, to mitigate their risks.

Lanthanide metal-organic framework-based surface molecularly imprinted polymers ratiometric fluorescence probe for visual detection of perfluorooctanoic acid with a smartphone-assisted portable device

Perfluorooctanoic acid (PFOA) poses a threat to the environment and human health due to its persistence, bioaccumulation, and reproductive toxicity. Herein, a lanthanide metal-organic framework (Ln-MOF)-based surface molecularly imprinted polymers (SMIPs) ratiometric fluorescence probe (Eu/Tb-MOF@MIPs) and a smartphone-assisted portable device were developed for the detection of PFOA with high selectivity in real water samples. The integration of Eu/Tb MOFs as carriers not only had highly stable multiple emission signals but also prevented deformation of the imprinting cavity of MIPs. Meanwhile, the MIPs layer preserved the fluorescence of Ln-MOF and provided selective cavities for improved specificity. Molecular dynamics (MD) was employed to simulate the polymerization process of MIPs, revealing that the formation of multiple recognition sites was attributed to the establishment of hydrogen bonds between functional monomers and templates. The probe showed a good linear relationship with PFOA concentration in the range of 0.02–2.8 μM, by giving the limit of detection (LOD) of 0.98 nM. Additionally, The red-green-blue (RGB) values analysis based on the smartphone-assisted portable device demonstrated a linear relationship of 0.1–2.8 μM PFOA with the LOD of 3.26 nM. The developed probe and portable device sensing platform exhibit substantial potential for on-site detecting PFOA in practical applications and provide a reliable strategy for the intelligent identification of important targets in water environmental samples.

Nano-titanium dioxide exacerbates the harmful effects of perfluorooctanoic acid on the health of mussels

Exposing marine organisms to contemporary contaminants, such as perfluorooctanoic acid (PFOA) and nano-titanium dioxide (nano-TiO2), can induce multifaceted physiological consequences. Our investigation centered on the responses of the mussel, Mytilus coruscus, to these agents. We discerned pronounced disruptions in gill filament connections, pivotal structures for aquatic respiration, suggesting compromised oxygen uptake capabilities. Concurrently, the respiratory rate exhibited a marked decline, indicating a respiratory distress. Furthermore, the mussels' clearance rate, a metric of their filtration efficacy, diminished, suggesting the potential for bioaccumulation of deleterious substances. Notably, the co-exposure of PFOA and nano-TiO2 exhibits interactive effects on the physiological performance of the mussels. The mussels' digestive performance waned in the face of heightened PFOA and nano-TiO2 concentrations, possibly hampering nutrient assimilation and energy accrual. This was mirrored in the noticeable contraction of their energy budget, suggesting long-term growth repercussions. Additionally, the dysregulation of the gut microbiota and the reduction in its diversity further confirm alterations in intestinal homeostasis, subsequently impacting its physiological functions and health. Collectively, these findings underscore the perils posed by escalated PFOA and nano-TiO2 levels to marine mussels, accentuating the need for a deeper understanding of nanoparticle-pollutant synergies in marine ecosystems.

In-Vitro Effects of Perfluorooctanoic Acid on Human Sperm Function: What Are the Clinical Consequences?

Background: Lifestyle and environmental pollution harm male fertility. Perfluoroalkyl substances (PFAS) are bio-accumulates in the environment as well as in several human tissues, and one of the most common PFAS is perfluorooctanoic acid (PFOA). Therefore, this study aimed to evaluate the in vitro effects of PFOA with hydrophobic and waterproofing properties on motility and bio-functional sperm parameters. Methods: To accomplish this, 50 healthy men with normozoospermia and not exposed to high doses of PFAS were enrolled. Their spermatozoa were incubated for 3 h with increasing concentrations of PFOA (0, 0.01, 0.1, and 1 mM) to evaluate its effects. In particular, we evaluated the effects of PFOA on total and progressive sperm motility and, by flow cytometry, on the following bio-functional sperm parameters: degree of chromatin compactness, viability, early and late apoptosis, mitochondrial membrane potential, the degree of lipoperoxidation, and concentrations of mitochondrial superoxide anion. Results: The results showed that PFOA decreased both total and progressive sperm motility, impaired chromatin compactness, and increased sperm lipid peroxidation and mitochondrial superoxide anion levels. Conclusions: This study showed that PFOA alters several sperm parameters and thus it may play a negative role in male fertility.

A novel method to derive a human safety limit for PFOA by gene expression profiling and modelling.

Perfluorooctanoic acid (PFOA) is a persistent environmental contaminant that can accumulate in the human body due to its long half-life. This substance has been associated with liver, pancreatic, testicular and breast cancers, liver steatosis and endocrine disruption. PFOA is a member of a large group of substances also known as "forever chemicals" and the vast majority of substances of this group lack toxicological data that would enable their effective risk assessment in terms of human health hazards. This study aimed to derive a health-based guidance value for PFOA intake (ng/kg BW/day) from in vitro transcriptomics data. To this end, we developed an in silico workflow comprising five components: (i) sourcing in vitro hepatic transcriptomics concentration-response data; (ii) deriving molecular points of departure using BMDExpress3 and performing pathway analysis using gene set enrichment analysis (GSEA) to identify the most sensitive molecular pathways to PFOA exposure; (iii) estimating freely-dissolved PFOA concentrations in vitro using a mass balance model; (iv) estimating in vivo doses by reverse dosimetry using a PBK model for PFOA as part of a quantitative in vitro to in vivo extrapolation (QIVIVE) algorithm; and (v) calculating a tolerable daily intake (TDI) for PFOA. Fourteen percent of interrogated genes exhibited in vitro concentration-response relationships. GSEA pathway enrichment analysis revealed that "fatty acid metabolism" was the most sensitive pathway to PFOA exposure. In vitro free PFOA concentrations were calculated to be 2.9% of the nominal applied concentrations, and these free concentrations were input into the QIVIVE workflow. Exposure doses for a virtual population of 3,000 individuals were estimated, from which a TDI of 0.15ng/kg BW/day for PFOA was calculated using the benchmark dose modelling software, PROAST. This TDI is comparable to previously published values of 1.16, 0.69, and 0.86ng/kg BW/day by the European Food Safety Authority. In conclusion, this study demonstrates the combined utility of an "omics"-derived molecular point of departure and in silico QIVIVE workflow for setting health-based guidance values in anticipation of the acceptance of in vitro concentration-response molecular measurements in chemical risk assessment.

Effects of Temperature and DC Electric Fields on Perfluorooctanoic Acid Sorption Kinetics to Activated Carbon.

Sorption to activated carbon is a common approach to reducing environmental risks of waterborne perfluorooctanoic acid (PFOA), while effective and flexible approaches to PFOA sorption are needed. Variations in temperature or the use of electrokinetic phenomena (electroosmosis and electromigration) in the presence of external DC electric fields have been shown to alter the contaminant sorption of contaminants. Their role in PFOA sorption, however, remains unclear. Here, we investigated the joint effects of DC electric fields and the temperature on the sorption of PFOA on activated carbon. Temperature-dependent batch and column sorption experiments were performed in the presence and absence of DC fields, and the results were evaluated by using different kinetic sorption models. We found an emerging interplay of DC and temperature on PFOA sorption, which was linked via the liquid viscosity (η) of the electrolyte. For instance, the combined presence of a DC field and low temperature increased the PFOA loading up to 38% in 48 h relative to DC-free controls. We further developed a model that allowed us to predict temperature- and DC field strength-dependent electrokinetic benefits on the drivers of PFOA sorption kinetics (i.e., intraparticle diffusivity and the film mass transfer coefficient). Our insights may give rise to future DC- and temperature-driven applications for PFOA sorption, for instance, in response to fluctuating PFOA concentrations in contaminated water streams.

Trend of PFAS concentrations and prediction of potential risks in Taihu Lake of China by AQUATOX

Per- and polyfluoroalkyl substances (PFAS) are recognized as emerging environmental pollutants due to their high persistence and toxicities to humans and animals. Understanding the temporal trend of PFAS in the environment is important for their pollution control and making appropriate policies. Many studies have reported the PFAS concentrations in Taihu Lake, the third largest lake in China, while their temporal trend during the years was seldom investigated. This study summarizes the PFAS concentrations in the water, sediment and organisms in Taihu Lake from 2009 to 2020 to depict their temporal trends. Meanwhile, the ecological model of AQUATOX was applied to evaluate and predict the potential risks of PFAS from 2012 to 2030. The results showed that the total PFAS concentrations varied but without distinct increase or decrease in both water and sediment during the years, while PFAS concentrations in organisms significantly decreased. The yearly mean concentrations of perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) in the water were 21.7–25.4 ng/L and 9.7–26.5 ng/L respectively, lower than the Standards for Drinking Water Quality of China and the suggested water quality criteria to protect the aquatic organisms. In sediment, PFOA and PFOS concentrations were 0.16–0.69 ng/g and 0.15–0.82 ng/g respectively, much lower than the recommended sediment quality guideline values. Based on the AQUATOX prediction, there will be no major threats caused by PFAS to the growth of biota in Taihu Lake in the near future, while the biomass of some species (e.g. carp) will be affected under the perturbation of PFAS. Both field investigation and AQUATOX simulation showed that PFOS concentrations in invertebrates and fish descend steadily, while no remarkable decrease in PFOA concentrations was expected. This study suggests a decreasing ecological risk of PFAS in Taihu Lake, while highlights the necessity of continuous monitoring of PFAS contamination.

PFAS alters placental arterial vasculature in term human placentae: A prospective pregnancy cohort study

IntroductionPerfluoroalkyl substances (PFAS) are synthetic chemicals used in industrial and consumer goods that are widely detected in human populations and are associated with adverse health outcomes, including perinatal health risks and child health. One mechanism of influence may be the impact of PFAS exposure on placental structure and function. ObjectivesThe objective of this study is to investigate the relationship between maternal prenatal exposure to PFAS and measures of placental vascularization, and to assess whether changes in vascularization play a role in mediating the impact of PFAS on birth outcomes. MethodsUsing data from a prospective cohort study, we examined associations between second trimester PFAS (individually and as mixtures using Bayesian kernel machine regression) and placental arterial vasculature in term placentae (N = 158); secondarily we evaluated the degree to which alterations in placental arterial vasculature explained associations between PFAS exposure and birth outcomes. Placental arterial vasculature features were collected from arterial tracings of each placental image. ResultsIn both linear regression and mixture models, natural log-transformed perfluorooctanoic acid concentrations were negatively associated with surface vasculature, indexed by the mean distance from arterial end point to perimeter (β = −0.23, 95% CI: −0.41, −0.041); additionally, maximum arterial tortuosity was negatively associated with placental weight (β = −0.19, 95% CI: −0.34, −0.051). There were no reliable differences in effect by fetal sex. DiscussionThe findings provide some of the first evidence of PFAS exposure shaping a key measure of placental vascular function, which may underlie the impact of PFAS on perinatal and child health risks.

Ecotoxicity of perfluorooctanoic acid and perfluorooctane sulfonate on aquatic plant Vallisneria natans.

Perfluorinated compounds (PFCs) are persistent organic contaminants that are highly toxic to the environment and bioaccumulate, but their ecotoxic effects on aquatic plants remain unclear. In this study, the submerged plant Vallisneria natans was treated with short-term (7 days) and long-term (21 days) exposures to perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) at concentrations of 0, 0.01, 0.1, 1.0, 5.0, and 10 mg/L, respectively. The results showed that both high concentrations of PFOA and PFOS inhibited the growth of V. natans and triggered the increase in photosynthetic pigment content in leaves. The oxidative damage occurred mainly in leaves, but both leaves and roots gradually built up tolerance during the stress process without serious membrane damage. Both leaves and roots replied to short-term stress by activating superoxide dismutase (SOD), catalase (CAT) and polyphenol oxidase (PPO), while peroxidase (POD) was involved under high concentration stress with increasing exposure time. Leaves showed a dose-effect relationship in integrated biomarker response (IBR) values under short-term exposure, and the sensitivity of roots and leaves to PFOS was higher than that of PFOA. Our findings help to increase knowledge of the toxic effects of PFCs and have important reference value for risk assessment and environmental remediation of PFCs in the aquatic ecosystem.

Fluorine mass balance analysis in wild boar organs from the Bohemian Forest National Park

Wild boars have been reported as bioindicators for per- and polyfluoroalkyl substances (PFAS) in a variety of studies. However, data about PFAS levels in wild boars from sites with limited industrial and general human activity is scarce. In this study, wild boar (Sus scrofa) organs from the Bohemian Forest National Park (Czech Republic) were used as bioindicators for PFAS pollution. In this work, 29 livers and 24 kidneys from 30 wild boars (0.5–5 years) were investigated using a fluorine mass balance approach. For this, the samples were measured using high performance liquid chromatography with electrospray ionisation tandem mass spectrometry (HPLC-ESI-MS/MS), targeting 30 PFAS, including legacy and replacement PFAS, direct total oxidisable precursor assay (dTOPA) and combustion ion chromatography (CIC). Perfluorocarboxylic acids (PFCAs) from C7 to C14 and perfluorooctanesulfonic acid (PFOS) were detected in >50 % of samples. In the livers, PFCAs dominated the profile with median concentrations of 230 μg/kg for perfluorononanoic acid (PFNA) and 75 μg/kg perfluorooctanoic acid (PFOA). PFOA and PFNA concentrations in the livers were one order of magnitude higher than in livers from wild boars caught in rural NE Germany considered as background concentration. PFOS in liver contributed only 30 % to the Σc(PFASTarget) with a median concentration of 170 μg/kg. Kidneys and livers contain an average of 2460 μg F/kg and 6800 μg F/kg extractable organic fluorine (EOF) respectively. Σc(PFASTarget) add up to a maximum of 10 % of the extractable organic fluorine. After oxidisation of the samples, PFOA, PFNA and Σc(PFASdTOPA) increased in livers, but could not explain the EOF. The elevated concentration of PFOA and PFNA may indicate differences in biomagnification for different habitats or an unidentified PFAS source in proximity to the national park.

Texture Analysis as a Discriminating Tool: Unmasking Rodlet Cell Degranulation in Response to a Contaminant of Emerging Concern.

Contaminants of emerging concern (CECs) have garnered significant attention due to their potential impacts on ecology, wildlife, and human health. The interest in these contaminants arises from their inadequate regulation or lack of routine monitoring in natural environments. Among them, per- and polyfluoroalkyl substances (PFAS) are of particular concern due to their notable propensity to accumulate within the kidney, significantly influencing the excretion of these pollutants. Rodlet cells (RCs) have emerged as promising indicators of immunotoxicity in response to chemical stressors. A prior comprehensive study extensively detailed the effects of sub-chronic exposure to perfluorooctanoic acid (PFOA), a well-known PFAS compound, on RCs located in the hematopoietic tissue of the common carp kidney. Even at concentrations commonly found in the environment, PFOA exhibited a significant impact on the distribution patterns of RCs, concurrently enhancing exocytosis activity. The assessment of PFOA-induced RC degranulation employed texture analysis combined with linear discriminant analysis (LDA) to differentiate between various experimental exposure groups. The investigation encompassed three fish groups: an unexposed group, a group exposed to an environmentally relevant PFOA concentration (200 ng L-1), and a group exposed to a higher PFOA concentration (2 mg L-1). Texture analysis was conducted on high-resolution color (RGB) images obtained from light microscopy of ultrathin sections from five fish per experimental group, stained with toluidine blue. This analysis facilitated the quantification of potential cytoplasmic alterations associated with degranulation, encompassing all three RGB channels. The data subjected to LDA enabled the identification of the most distinctive texture characteristics, providing a reliable, objective, and reproducible method to differentiate between experimental groups. Remarkably, 98.0% of both the original and cross-validated cases were correctly classified. However, only one unexposed case was misclassified as a fish exposed to a 200 ng L-1 PFOA concentration, constituting the single false positive in the analysis. Utilizing texture analysis and LDA to quantify RC degranulation offers a dependable approach for assessing immunotoxicity within experimental models of toxicological and environmental pathology. This underscores the scientific significance of employing a morphological approach in such investigations.

Fluorinated Nonporous Adaptive Cages for the Efficient Removal of Perfluorooctanoic Acid from Aqueous Source Phases.

Per- and polyfluoroalkyl substances (PFAS) accumulate in water resources and pose serious environmental and health threats due to their nonbiodegradable nature and long environmental persistence times. Strategies for the efficient removal of PFAS from contaminated water are needed to address this concern. Here, we report a fluorinated nonporous adaptive crystalline cage (F-Cage 2) that exploits electrostatic interaction, hydrogen bonding, and F-F interactions to achieve the efficient removal of perfluorooctanoic acid (PFOA) from aqueous source phases. F-Cage 2 exhibits a high second-order kobs value of approximately 441,000 g mg-1 h-1 for PFOA and a maximum PFOA adsorption capacity of 45 mg g-1. F-Cage 2 can decrease PFOA concentrations from 1500 to 6 ng L-1 through three rounds of flow-through purification, conducted at a flow rate of 40 mL h-1. Elimination of PFOA from PFOA-loaded F-Cage 2 is readily achieved by rinsing with a mixture of MeOH and saturated NaCl. Heating at 80 °C under vacuum then makes F-Cage 2 ready for reuse, as demonstrated across five successive uptake and release cycles. This work thus highlights the potential utility of suitably designed nonporous adaptive crystals as platforms for PFAS remediation.

A Deep Insight into Perfluorooctanoic Acid Photodegradation Using Metal Ion-Exchanged Zeolites.

Treating perfluorooctanoic acid (PFOA) in an aqueous environment is problematic due to its low concentration and its high resistance to biological and chemical degradation. To tackle this challenge, combinations of pre-enrichment and photodegradation processes are promising solutions. In this work, we investigated metal ion-exchanged zeolites as adsorbents and photocatalysts for PFOA treatment. Among various transition metal ion-exchanged BEA zeolites, Fe-exchanged BEA (Fe-BEA) zeolites showed significant activity for the photodegradation of PFOA. The isolated iron species in Fe-BEA zeolite are responsible for PFOA photodegradation, whereas other iron species present from excess iron loading in the zeolite will lower its photocatalytic activity. Furthermore, it was proved via size exclusion tests using branched PFOA isomers that the photodegradation of PFOA took place on the internal surface rather than the external surface of Fe-BEA zeolite. Photodegradation of PFOA was also tested to be effective with Fe-exchanged BEA-type zeolites having various SiO2/Al2O3 ratios, but ineffective with FAU-type zeolites. The optimal Fe-BEA zeolite showed a sorption coefficient Kd of 6.0 × 105 L kg-1 at an aqueous phase PFOA concentration of 0.7 μg L-1 and a PFOA half-life of 1.8 h under UV-A irradiation. The presented study offers a deeper understanding of the use of metal ion-exchanged zeolites for photodegradation of PFOA.

Substitution and Orientation Effects on the Crystallinity and PFAS Adsorption of Olefin-Linked 2D COFs.

Solid phase adsorbents with high removal affinity for per- and polyfluoroalkyl substances (PFAS) in aqueous environments are sought. We report the synthesis and investigation of COF-I, a new covalent organic framework (COF) with a good affinity for PFAS adsorption. COF-I was synthesized by the condensation reaction between 2,4,6-trimethyl-1,3,5-triazine and 2,3-dimethoxyterephthaldehyde and fully characterized. In addition to the high crystallinity and surface area, COF-I showed high hydrolytic and thermal stability. Further, we converted its hydrophobic surface to a hydrophilic surface by converting the ortho-methoxy groups to hydroxyl derivatives and produced a new hydrophilic olefin-linked two-dimensional (2D) COF. We experimentally measured the crystallinity of both COFs by X-ray diffraction and used atomistic simulations coupled with cross-polarization/magic angle spinning solid-state nuclear magnetic resonance (CP/MAS ssNMR) to determine the relative amounts of AA-stacking and AB-stacking present. COF-I, with its hydrophobic surface and methoxy groups in the ortho positions, showed the best PFAS adsorption. COF-I reduced the concentration of perfluorooctanoic acid from 20 to 0.069 μg L-1 and to 0.052 μg L-1 for perfluorooctanesulfonic acid. These amounts are lower than the U.S. Environmental Protection Agency advisory level (0.070 μg L-1). High efficiency, fast kinetic adsorption, and reusability of COF-I are advantages of COF-I for PFAS removal from water.

Determinants of PFOA Serum Half-Life after End of Exposure: A Longitudinal Study on Highly Exposed Subjects in the Veneto Region.

Perfluoroalkyl substances (PFAS) are widely used, ubiquitous, and highly persistent man-made chemicals. Groundwater of a vast area of the Veneto Region (northeastern Italy) was found to be contaminated by PFAS from a manufacturing plant active since the late 1960s. As a result, residents were overexposed to PFAS through drinking water until 2013, mainly to perfluorooctanoic acid (PFOA). The aim of the present study was to estimate the rates of decline in serum PFOA and their corresponding serum half-lives, while characterizing their determinants. We investigated 5,860 subjects more than 14 years of age who enrolled in the second surveillance round of the regional health surveillance program. Two blood samples were collected between 2017 and 2022 (average time between measurements: 4 years). Serum PFOA excretion rates and half-lives were estimated based on linear mixed effect models, modeling subject-specific serum PFOA concentrations over time and correcting for background concentrations. For modeling determinants of half-life [age, sex, body mass index (BMI), smoking-habit, alcohol consumption, and estimated glomerular filtration rate (eGFR)], we added interaction terms between each covariate and the elapsed time between measurements. Perfluorooctanesulfonate (PFOS) and perfluorohexanesulfonic acid (PFHxS) apparent half-lives were also estimated. A separate analysis was conducted in children (). All analyses were stratified by sex. Median initial serum concentrations of PFOA was (range: 0.5-1,090), with a median reduction of 62.45%. The mean estimated PFOA half-life was 2.36 years [95% confidence interval (CI): 2.33, 2.40], shorter in women (2.04; 95% CI: 2.00, 2.08) compared to men (2.83; 95% CI: 2.78, 2.89). Half-lives varied when stratified by some contributing factors, with faster excretion rates in nonsmokers and nonalcohol drinkers (especially in males). This study, to our knowledge the largest on PFOA half-life, provides precise estimates in young adults whose exposure via drinking water has largely ceased.
