Concentrations of neonicotinoid insecticides and their metabolites in multiple urine samples collected from pregnant women in Japan

Exposure to low doses of neonicotinoid (NEO) insecticides during pregnancy may have a negative impact on the neurodevelopment of children. Biomarkers are frequently used to investigate the exposure and its health effects in environmental epidemiology. In the present study, we aimed to evaluate the utility of urinary biomarkers of NEO exposure by calculating intra-class correlation coefficients (ICCs). Urine samples were collected from 30 pregnant women who were non-smokers and living in a central part of Japan, between 12 and 22 weeks of gestation. We collected first morning voided (FMV) urine and afternoon spot (PM) urine samples on the same day on five occasions within a 2-week period. The urinary concentrations of NEO and their metabolites (NEO/m) were measured by liquid chromatography–tandem mass spectrometry. Normalisation to the specific gravity and creatinine concentration was used in the calculation of ICC. The number of urine samples required to obtain a reliable biomarker estimate of NEO exposure was calculated. The most frequently detected NEO/m was N-desmethyl-acetamiprid (N-dm-ACE), followed by clothianidin (CLO), thiamethoxam (THX) and dinotefuran (DIN), in both FMV and PM urine samples. The ICC calculations showed poor reproducibility for the measurement of CLO and THX concentrations (ICCs ≤0.25) in both FMV and PM urine samples and for DIN (ICC ≤0.29) in the FMV urine samples, and moderate reproducibility was identified for N-dm-ACE (ICC >0.60) in both FMV and PM urine samples and for DIN (ICC >0.40) in the PM urine samples. A minimum of 1–19 spot urine samples per individual was required to provide a reliable biomarker estimate over a 2 week period. These results suggest that pregnant Japanese women are frequently exposed to NEOs, and that, with the exception of urinary N-dm-ACE, a single measurement of urinary NEO concentrations may not be an appropriate method of characterising overall exposure.

Honey bees and bumble bees may be exposed to pesticides differently when foraging on agricultural areas

In an agricultural environment, where crops are treated with pesticides, bees are likely to be exposed to a range of chemical compounds in a variety of ways. The extent to which different bee species are affected by these chemicals, largely depends on the concentrations and type of exposure. We quantified the presence of selected pesticide compounds in the pollen of two different entomophilous crops; oilseed rape (Brassica napus) and broad bean (Vicia faba). Sampling was performed in 12 sites in Ireland and our results were compared with the pollen loads of honey bees and bumble bees actively foraging on those crops in those same sites. Detections were compound specific, and the timing of pesticide application in relation to sampling likely influenced the final residue contamination levels. Most detections originated from compounds that were not recently applied on the fields, and samples from B. napus fields were more contaminated compared to those from V. faba fields. Crop pollen was contaminated only with fungicides, honey bee pollen loads contained mainly fungicides, while more insecticides were detected in bumble bee pollen loads. The highest number of compounds and most detections were observed in bumble bee pollen loads, where notably, all five neonicotinoids assessed (acetamiprid, clothianidin, imidacloprid, thiacloprid, and thiamethoxam) were detected despite the no recent application of these compounds on the fields where samples were collected. The concentrations of neonicotinoid insecticides were positively correlated with the number of wild plant species present in the bumble bee-collected pollen samples, but this relationship could not be verified for honey bees. The compounds azoxystrobin, boscalid and thiamethoxam formed the most common pesticide combination in pollen. Our results raise concerns about potential long-term bee exposure to multiple residues and question whether honey bees are suitable surrogates for pesticide risk assessments for all bee species.

A pilot nationwide baseline survey on the concentrations of Neonicotinoid insecticides in tap water from China: Implication for human exposure

Neonicotinoids (NEOs) have become the most widely used insecticides in the world. However, information on the NEO concentrations in tap water based on nationwide surveys is limited in China. In this study, the levels of six NEOs were measured in tap water samples collected from 38 cities in China. Across all sampling locations, the overall frequency of detection for at least one NEO was 100%, indicating that NEOs are ubiquitous in tap water from China. Imidacloprid was the most abundant NEO (median: 7.59 ng/L), followed by (in decreasing order) clothianidin (5.46 ng/L), dinotefuran (4.55 ng/L), thiamethoxam (4.50 ng/L), acetamiprid (2.72 ng/L), and thiacloprid (0.38 ng/L). Significantly positive correlations (r = 0.655–0.902, p < 0.001) among all pairs of NEOs were observed, which showed that the sources of NEOs in tap water were common or related. Across all sampling locations, regional differences in concentrations (ΣNEOs; sum of six NEOs varied from 0.79 ng/L to 415 ng/L) were observed within China, showing an increasing trend from northern to southern China. Although the estimated daily intake of NEOs were much lower than the reference dose, the potential health risk of NEOs via tap water consumption should raise more public concern considering the high detection rates of NEOs in tap water.

The sublethal effects of neonicotinoids on spiders are independent of their nutritional status

Spiders were recently shown to be adversely affected by field-realistic concentrations of a broad scale of neonicotinoid insecticides. Among the reported effects of neonicotinoids on invertebrates were declines in lipid biosynthesis and upregulation of β-oxidation, while vertebrate models suggest increased adipogenesis following treatment with neonicotinoids. Therefore, we hypothesized that there exists synergy between the effects of diet and concurrent exposure to field-realistic concentrations of neonicotinoid insecticides. To address this hypothesis, we fed first instars of the large wolf spider Hogna antelucana with two types of diets and exposed them to field-realistic concentrations of three formulations of neonicotinoids (thiamethoxam, thiacloprid and acetamiprid). We then measured the growth of the tested spiders; the lipid and protein content of their bodies; and their behavior, including ballooning, rappelling, and locomotor parameters. The two tested diets consisted of casein-treated and sucrose-treated Drosophila melanogaster. The dietary treatments affected the lipid and protein content of the spiders, their body weight and carapace length but did not affect any of the measured behavioral parameters. Surprisingly, we did not find any effects of acute exposure to neonicotinoid insecticides on the lipid or protein reserves of spiders. Exposure to neonicotinoids altered the behavior of the spiders as reported previously in other spider species; however, these effects were not affected by dietary treatments. Overall, the dietary treatments did not have any major synergy with acute exposure to field-realistic concentrations of neonicotinoid insecticides.

Serum concentrations of neonicotinoids, and their associations with lipid molecules of the general residents in Wuxi City, Eastern China

Exposure to neonicotinoid insecticides (NNIs) was proven to be harmful to organisms, however, there is a dearth of information regarding their occurrence and adverse effects on the general residents. Here, n = 120 human serum samples were collected from the same area of Wuxi city, Eastern China, and these serum samples were further analyzed for nine NNIs and nine target lipid molecules by use of ultrahigh performance liquid chromatography-quadrupole orbitrap high-resolution mass spectrometer. We observed that four out of nine NNIs exhibited relatively high detection frequencies (DF), and these NNIs were imidacloprid (IMI; DF = 28.3%), clothianidin (CLO; 16.7%), thiacloprid (THI; 14.2%), and acetamiprid (ACE; 12.5%), respectively, with 95th concentrations ranging from 32.0 to 427 pg/mL. Median concentrations of imidacloprid-equivalent total neonicotinoids (IMIeq) and ∑7NNI were 46.6 pg/mL and 26 pg/mL, respectively. Five out of nine lipid molecules exhibited higher levels, that were docosahexaenoic acid [FA(22:6)], 18:0 phosphocholine [LysoPC(18:0)], 18:0 phosphoethanolamine [LysoPE(18:0)], D18:1–18:0 sphingomyelin [SM(d18:1/18:0)], and 18:1–18:1 diglycerol [DG(18:1/18:1)], respectively. More interestingly, we observed statistically significant correlations (student’s t-test, one-way ANOVA, or Mann-Whitney test; p < 0.05) between NNI levels and population characteristics (i.e. age, smoking, and health status). Beyond that, we also observed statistically significant correlations between levels of selected NNIs (CLO, ACE, or THI) and lipid molecules [LysoPE(18:0), SM(d18:1/18:0), and DG(18:1/18:1)]. Collectively, for the first time, we provided the information on contamination levels of NNIs in serum samples of general residents in China and demonstrated the associations between concentrations of NNIs and levels of lipid molecular species.

No impact of neonicotinoids on male solitary bees Osmia cornuta under semi‐field conditions

AbstractThe ubiquitous use of agrochemicals is one driver for the ongoing loss of insect biomass and diversity. Data show that field‐realistic concentrations of neonicotinoid insecticides can negatively affect both population density and the fitness of solitary bees. However, the underlying mechanisms for these effects remain poorly understood. Here, using an established semi‐field experimental set‐up and Osmia cornuta as a solitary bee model, we examined the effects of field‐realistic concentrations of a common neonicotinoid insecticide (clothianidin) on male larvae and adults. Besides measuring lethal (i.e., overwintering success and adult survival) and sub‐lethal endpoints (i.e., emergence mass and emergence duration), we examined, for the first time, potential effects on the male reproductive physiology of a solitary bee (i.e., sperm quantity and viability). The data revealed no significant effects on any of the measured response variables. This may be due to the low degree of clothianidin exposure (0.56 ng g−1) and/or the apparent low susceptibility of solitary bee larvae to neonicotinoids. Furthermore, it is conceivable that ideal foraging conditions, combined with optimal weather and lack of other environmental stressors, may have improved the ability of bees to cope with the insecticide. To reliably assess and understand the environmental hazards of agrochemicals, a holistic approach, including laboratory, semi‐field and field data is required. Knowledge of underlying mechanisms will help to mitigate the current global declines of insect populations.

Neonicotinoid insecticides hinder the pupation and metamorphosis into adults in a crabronid wasp

Neonicotinoid insecticides are associated with a decline in the diversity and distribution of bees and wasps (Hymenoptera: Aculeata). The effects of neonicotinoids on the metamorphosis of aculeates have never been addressed in detail; however, recent evidence suggests that neonicotinoids induce wing abnormalities. We hypothesized that the metamorphosis success of bees and wasps differs in response to contact exposure to field-realistic concentrations of neonicotinoid insecticides or in response to combined exposure to neonicotinoid insecticides and benzimidazole fungicides. We treated prepupae of the model crabronid wasp Pemphredon fabricii with field-realistic concentrations of four neonicotinoids, acetamiprid, imidacloprid, thiacloprid and thiamethoxam, and/or with the benzimidazole fungicide thiabendazole. Treatment with acetamiprid or imidacloprid decreased the pupation rates to only 39% and 32%, respectively. Treatment with thiacloprid or thiamethoxam did not affect the pupation rate when applied alone, but the subsequent treatment of thiacloprid- or thiamethoxam-treated prepupae with thiabendazole led to significant decreases in pupation rates. A high concentration of acetamiprid, which severely affected the pupation rates, had moderate effects on metamorphosis into adults, resulting in 53% metamorphosis success (as opposed to 95% metamorphosis success in the water-treated group). However, imidacloprid or thiamethoxam treatment resulted in only 5%-10% metamorphosis success into adults. Overall survival decreased in response to treatment with any of the neonicotinoids or benzimidazoles or their combinations, with extremely low survival (<2%) following combined treatment with imidacloprid and thiabendazole or thiamethoxam and thiabendazole. In conclusion, neonicotinoids alter insect metamorphosis success, which can be further potentiated by their combination with other agrochemicals, such as benzimidazoles.

Distribution, Dissipation, and Metabolism of Neonicotinoid Insecticides in the Cotton Ecosystem under Foliar Spray and Root Irrigation

The uptake, distribution, metabolism, and degradation of three neonicotinoid insecticides (NIs)-imidacloprid (IMI), acetamiprid (ACE), and thiamethoxam (THI) in different parts of cotton plants were investigated under field conditions. Insecticides were either applied by foliar spraying or root irrigation. Foliar application resulted in high tissue concentration (average tissue concentration ratio, TCR: 46.78-68.61% for leaves and 12.2-31.40% for flowers). The flowers showed high NI residual. The metabolism and trends of NIs in different parts of cotton were reported here for the first time. Metabolites, toxic to bees, were detected in the flowers. The translocation factor was around 0.004 for the spray treatment and 0.2-0.7 for the root irrigation treatment. The average root concentration factors of IMI, ACE, and THI were 0.838, 8.027, and 1.014, respectively, indicating that the three NIs can be transported from the soil to the plant. The high concentrations of NIs and their metabolites in flowers indicate exposure risk to pollinators, such as bees.

Ecological risk assessment of the increasing use of the neonicotinoid insecticides along the east coast of China

In the past two decades, neonicotinoid insecticides have been widely used in agricultural activities in China. Many previous studies have investigated the neonicotinoid pollution in aquatic ecosystems, but the status of water safety of neonicotinoid uses in China is very scarce. The present study aims to reveal the spatial and temporal distribution of neonicotinoids in rivers, and then evaluate the ecological risks to aquatic animals. Water samples were collected from all sixteen rivers along the east coast of China during the dry and wet seasons in 2016, and nine individual commercialized neonicotinoids were quantified. Higher concentrations were found during the dry season (343 ± 210 ng/L) compared to those during the wet season (174 ± 162 ng/L). The concentration of neonicotinoid insecticides in river water is mainly affected by the intensity of agricultural activities. The spatial and temporal pollution patterns we discovered suggested the use of neonicotinoids has shifted from old types (i.e., imidacloprid and acetamiprid) to new types (i.e., dinotefuran and nitenpyram) in some areas. The estimated annual quantity of neonicotinoids released into the adjacent seas totaled 1256 ± 780 tons, most of which (95%) ran into the East China Sea due to heavy agricultural use in the Yangtze River Basin. Using the species sensitive distribution (SSD) method, the thresholds for aquatic animals were determined (acute: 362 ng/L; chronic: 58 ng/L). Under current agricultural practices, 27% and 84% of the river water samples exceeded the thresholds for acute and chronic ecological risks, suggesting intervention programs are in urgent need to ensure river water safety for aquatic life in China.

Distribution and Concentration of Neonicotinoid Insecticides on Waterfowl Production Areas in West Central Minnesota

Neonicotinoid insecticides have been reported to occur widely in surface waters, including those of wetlands within the Prairie Pothole Region (PPR). In the US portion of the PPR, the US Fish and Wildlife Service has established Waterfowl Production Areas (WPAs) in an effort to enhance waterfowl production. Most WPAs have an area of protected upland surrounding wetlands that can act as a buffer to reduce the transport of contaminants, including pesticides. We assessed the extent that neonicotinoid insecticides occurred in the ponded water of wetlands within WPAs located along a gradient of agricultural influence throughout west-central Minnesota. Of the five neonicotinoids we tested for, two were not detected. However, at least one of the other three, imidacloprid, clothianidin and thiamethoxam, were detected in 29% of our wetland water samples. Additionally, both the occurrence and total concentrations of neonicotinoids were higher in sites with higher surrounding crop use. Neonicotinoid insecticides, if persistent for long periods of time, have the potential to affect aquatic-invertebrate communities within PPR wetlands. Our research indicates that areas often perceived as protected may still be at risk to neonicotinoid contamination, emphasizing the importance of maintaining effective grassland buffers around wetlands.

Neonicotinoid insecticides in pollen, honey and adult bees in colonies of the European honey bee (Apis mellifera L.) in Egypt.

Honeybee losses have been attributed to multiple stressors and factors including the neonicotinoid insecticides (NIs). Much of the study of hive contamination has been focused upon temperate regions such as Europe, Canada and the United States. This study looks for the first time at honey, pollen and bees collected from across the Nile Delta in Egypt in both the spring and summer planting season of 2013. There is limited information upon the frequency of use of NIs in Egypt but the ratio of positive identification and concentrations of NIs are comparable to other regions. Metabolites of NIs were also monitored but given the low detection frequency, no link between matrices was possible in the study. Using a simple hazard assessment based upon published LD50 values for individual neonicotinoids upon the foraging and brood workers it was found that there was a potential risk to brood workers if the lowest reported LD50 was compared to the sum of the maximum NI concentrations. For non-lethal exposure there was significant risk at the worst case to brood bees but actual exposure effects are dependant upon the genetics and conditions of the Egyptian honeybee subspecies that remain to be determined.

Monitoring of neonicotinoid pesticides in beekeeping

The decline of pollinating species is correlated to the extensive use of neonicotinoids against pest insects for crop protection. In this study, the concentrations of neonicotinoid insecticides were determined in honeybees, honeycomb and honey samples, collected in Spring 2015 (blooming period) from different areas in Sicily (IT), to carry out an evaluation of bees products’ safety and an overview of neonicotinoid contamination in beekeeping. The results obtained showed only the presence of clothianidin in bee samples and these concentrations don’t represent a risk for bees’ vitality and safety. The absence of residue in all honey samples, instead, showed the quality of bee products.

Concentrations of neonicotinoid insecticides in honey, pollen and honey bees (Apis mellifera L.) in central Saskatchewan, Canada.

Neonicotinoid insecticides (NIs) and their transformation products were detected in honey, pollen and honey bees, (Apis mellifera) from hives located within 30 km of the City of Saskatoon, Saskatchewan, Canada. Clothianidin and thiamethoxam were the most frequently detected NIs, found in 68 and 75% of honey samples at mean concentrations of 8.2 and 17.2 ng g(-1) wet mass, (wm), respectively. Clothianidin was also found in >50% of samples of bees and pollen. Concentrations of clothianidin in bees exceed the LD50 in 2 of 28 samples, while for other NIs concentrations were typically 10-100-fold less than the oral LD50. Imidaclorpid was detected in ∼30% of samples of honey, but only 5% of pollen and concentrations were <LOD in bees. Transformation products of Imidaclorpid, imidaclorpid-Olefin and imidacloprid-5-Hydroxy were detected with greater frequency and at greater mean concentrations indicating a need for more focus on potential effects of these transformation products than the untransformed, active ingredient NIs. Results of an assessment of the potential dietary uptake of NIs from honey and pollen by bees over winter, during which worker bees live longer than in summer, suggested that, in some hives, consumption of honey and pollen during over-wintering might have adverse effects on bees.

Movement of Soil-Applied Imidacloprid and Thiamethoxam into Nectar and Pollen of Squash (Cucurbita pepo)

There has been recent interest in the threat to bees posed by the use of systemic insecticides. One concern is that systemic insecticides may translocate from the soil into pollen and nectar of plants, where they would be ingested by pollinators. This paper reports on the movement of two such systemic neonicotinoid insecticides, imidacloprid and thiamethoxam, into the pollen and nectar of flowers of squash (Cucurbita pepo cultivars “Multipik,” “Sunray” and “Bush Delicata”) when applied to soil by two methods: (1) sprayed into soil before seeding, or (2) applied through drip irrigation in a single treatment after transplant. All insecticide treatments were within labeled rates for these compounds. Pollen and nectar samples were analyzed using a standard extraction method widely used for pesticides (QuEChERS) and liquid chromatography mass spectrometric analysis. The concentrations found in nectar, 10±3 ppb (mean ± s.d) for imidacloprid and 11±6 ppb for thiamethoxam, are higher than concentrations of neonicotinoid insecticides in nectar of canola and sunflower grown from treated seed, and similar to those found in a recent study of neonicotinoids applied to pumpkins at transplant and through drip irrigation. The concentrations in pollen, 14±8 ppb for imidacloprid and 12±9 ppb for thiamethoxam, are higher than those found for seed treatments in most studies, but at the low end of the range found in the pumpkin study. Our concentrations fall into the range being investigated for sublethal effects on honey bees and bumble bees.
