Comparison of the Chemical and Microbial Composition and Aerobic Stability of High-Moisture Barley Grain Ensiled with Either Chemical or Viable Lactic Acid Bacteria Application

This experiment was aimed at comparing how a chemical additive or an inoculant would affect the dry matter (DM) losses, fermentation pattern, yeast and mold counts, and aerobic stability (AS) of the ensiled high-moisture barley grain (675 g kg−1 DM). Crimped barley grain was ensiled with or without chemical additive AIV Ässä Na and an inoculant SiloSolve FC, totaling three treatments (1 × 3 factorial scheme) for the fermentation periods lasting 7, 14, 28, 60, and 90 days. The application of a chemical additive showed higher pH levels, retained water-soluble carbohydrates (WSCs) better, lowered DM loss, and reduced concentrations of fermentation products. Barley grain treated with chemicals initially showed a marked reduction in yeast and fungal growth and a higher AS up to day 60 of storage but became less effective in later stages of storage, leading to decreased AS. Barley grain inoculated with homo- and heterofermentative strains had decreased silage pH; its WSC content was similar to control and had higher content of weak acids and 1,2 propanediol, reducing mold and yeast counts. With fermentation duration from 28 days and beyond (i.e., 90 days), the inoculant treatment achieved the longest AS and the lowest increase in pH and weight loss during the period of air exposure.

Molecular identification of anaerobic fungi isolated from buffalo rumen with their growth rate, cellulase enzyme activity, and fermentation products characteristics

Anaerobic fungi are essential microbes in the degradation process of feed fiber in the rumen with the ability to produce fibrolytic enzymes and rhizoids that can penetrate feed particles. The activity of cellulase enzymes in fungi is influenced by several factors such as the type of feed and livestock used as a source of isolate. Therefore, this research aimed to analyze the types of anaerobic fungi isolated from buffalo based on their DNA nitrogenous bases composition and also to evaluate the growth rate, cellulase enzyme activity, and fermentation product concentration of anaerobic fungi. The growth rate of fungi was examined by measuring the biomass formed, while cellulase enzyme activity was carried out using CMC, Avicel, and Filter Paper as substrates. The fungal fermentation products were analyzed using the HPLC and GC methods. The results showed that the fungi isolated from buffalo rumen were closely related to anaerobic fungi type Piromyces sp., Caecomyces sp., and Neocallimastix frontalis, with different growth rate, cellulase enzyme activity, and the fermentation products concentration in each type of anaerobic fungi. Therefore, it can be concluded that anaerobic fungi isolated from buffalo rumen can degrade cellulose. It was also discovered that fungi Neocallimastix frontalis had a higher growth rate, cellulase enzyme activity (CMCase, avicelase, and FPase), and fermentation products than Piromyces sp. and Caecomyces sp.

Effect of pH dynamic control on ethanol-lactic type fermentation (ELTF) performance of glucose

ABSTRACT This study proposed a new ethanol-lactic type fermentation (ELTF) and explored the optimal control strategy. Using batch experiments, the effects of pH, temperature and organic loading (OL) on ELTF were investigated. The sum of ethanol and lactic acid yield was highest at whole-control pH value of 4.0, 35°C temperature and OL of 33 gCOD/L. To improve ELTF, the dynamic pH control in the long-term CSTR was adjusted at 4.0 (1–28 days), 5.0 (29–44 days) and 4.0 (46–62 days) successively. The high concentration of ethanol and lactic acid was 8190.5 mg/L at 16th day of pH 4.0. At pH of 5.0, the average acidogenesis rate and total concentration of fermentation products increased 111.0% and 128.0%, respectively. Organisms of Lactobacillus and Bifidobacterium were the predominant bacteria in reactor. It can achieve the directional regulation of ELTF and provides parameter support for the application of two-phase anaerobic digestion.

Saccharomyces cerevisiae Fermentation of 28 Barley and 12 Oat Cultivars

As barley and oat production have recently increased in Canada, it has become prudent to investigate these cereal crops as potential feedstocks for alcoholic fermentation. Ethanol and other coproduct yields can vary substantially among fermented feedstocks, which currently consist primarily of wheat and corn. In this study, the liquified mash of milled grains from 28 barley (hulled and hull-less) and 12 oat cultivars were fermented with Saccharomyces cerevisiae to determine concentrations of fermentation products (ethanol, isopropanol, acetic acid, lactic acid, succinic acid, α-glycerylphosphorylcholine (α-GPC), and glycerol). On average, the fermentation of barley produced significantly higher amounts of ethanol, isopropanol, acetic acid, succinic acid, α-GPC, and glycerol than that of oats. The best performing barley cultivars were able to produce up to 78.48 g/L (CDC Clear) ethanol and 1.81 g/L α-GPC (CDC Cowboy). Furthermore, the presence of milled hulls did not impact ethanol yield amongst barley cultivars. Due to its superior ethanol yield compared to oats, barley is a suitable feedstock for ethanol production. In addition, the accumulation of α-GPC could add considerable value to the fermentation of these cereal crops.

Effects of plant extracts from Stryphnodendron adstringens (mart.) coville, Lafoensia pacari a. st.-hil, copaifera spp., and Pterodon emarginatus Vogel on in vitro rumen fermentation.

The effects of doses CTL (0mg), 30, 300 and 3000mg/L of extracts from Stryphnodendron adstringens (Mart.) Coville (SA), Lafoensia pacari A. St.-Hil (LP), Copaifera spp. (CO) and Pterodon emarginatus Vogel (PE) on ruminal fermentation were investigated in eight experiments conducted in randomized complete block designs. The in vitro system contained four fermentation vessels. Each treatment was allocated in one vessel in each run. Incubation was run four and five times with diets 50:50 and 10:90 (roughage to concentrate ratio) respectively. Incubation vessel was the experimental unit, and each incubation run was a block. All plant extracts negatively affected DM degradation at 3000mg/L. In diet 50:50, SA-3000 increased the molar proportion of propionate (p<0.05), yet it decreased the concentration of fermentation products (p<0.05); LP-300 reduced ruminal ammonia nitrogen (NH3 -N; 98 vs. 120mg/dl for LP-300 and average of CTL and LP-30, respectively) and isobutyrate (p<0.05); CO-300 reduced (p<0.05) NH3 -N (98 vs. 123mg/dl for CO-300 and average of CTL and CO-30, respectively); and P. emarginatus had no positive effects on rumen fermentation (p>0.05). In diet 10:90, SA-300 reduced (p<0.001) NH3 -N and total volatile fatty acids (VFA); LP-30 increased (p<0.05) total VFA (85 vs. 63mM for LP-30 and CTL, respectively), molar proportions of acetate and propionate, and had lower C2 :C3 than CTL (3.6 vs. 4.3, respectively); CO-300 decreased acetate and increased propionate, reducing C2 :C3 (p<0.001; 2.8 vs. 3.6 for CO-300 and average of other doses, respectively); PE-30 and PE-300 reduced NH3 -N by 14% and increased total VFA by 29% compared with CTL (p<0.05). Further in vivo investigations may consider L. pacari (LP-30), Copaifera spp. (CO-300) oleoresin and P. emarginatus oleoresin (PE-30 and PE-300) in diets with high inclusion of concentrate.

Ecogenomics-Based Mass Balance Model Reveals the Effects of Fermentation Conditions on Microbial Activity.

Fermentation of waste activated sludge (WAS) is an alternative approach to reduce solid wastes while providing valuable soluble products, such as volatile fatty acids and alcohols. This study systematically identified optimal fermentation conditions and key microbial populations by conducting two sets of experiments under different combinations of biochemical and physical parameters. Based on fermentation product concentrations, methane production, and solid removal, fermentation performance was enhanced under the combined treatments of inoculum heat shock (>60°C), pH 5, 55°C, and short solid retention time (<10 days). An ecogenomics-based mass balance (EGMB) approach was used to determine the net growth rates of individual microbial populations, and classified them into four microbial groups: known syntrophs, known methanogens, fermenters, and WAS-associated populations. Their growth rates were observed to be affected by the treatment conditions. The growth rates of syntrophs and fermenters, such as Syntrophomonas and Parabacteroides increased with a decrease in SRT. In contrast, treatment conditions, such as inoculum heat shock and high incubation temperature inhibited the growth of WAS-associated populations, such as Terrimonas and Bryobacter. There were also populations insensitive to the treatment conditions, such as those related to Microbacter and Rikenellaceae. Overall, the EGMB approach clearly revealed the ecological roles of important microbial guilds in the WAS fermentation system, and guided the selection of optimal conditions for WAS fermentation in future pilot-scale operation.

Rumen bacteria influence milk protein yield of yak grazing on the Qinghai-Tibet plateau.

ObjectiveRuminants are completely dependent on their microbiota for rumen fermentation, feed digestion, and consequently, their metabolism for productivity. This study aimed to evaluate the rumen bacteria of lactating yaks with different milk protein yields, using high-throughput sequencing technology, in order to understand the influence of these bacteria on milk production.MethodsYaks with similar high milk protein yield (high milk yield and high milk protein content, HH; n = 12) and low milk protein yield (low milk yield and low milk protein content, LL; n = 12) were randomly selected from 57 mid-lactation yaks. Ruminal contents were collected using an oral stomach tube from the 24 yaks selected. High-throughput sequencing of bacterial 16S rRNA gene was used.ResultsRuminal ammonia N, total volatile fatty acids, acetate, propionate, and isobutyrate concentrations were found to be higher in HH than LL yaks. Community richness (Chao 1 index) and diversity indices (Shannon index) of rumen microbiota were higher in LL than HH yaks. Relative abundances of the Bacteroidetes and Tenericutes phyla in the rumen fluid were significantly increased in HH than LL yaks, but significantly decreased for Firmicutes. Relative abundances of the Succiniclasticum, Butyrivibrio 2, Prevotella 1, and Prevotellaceae UCG-001 genera in the rumen fluid of HH yaks was significantly increased, but significantly decreased for Christensenellaceae R-7 group and Coprococcus 1. Principal coordinates analysis on unweighted UniFrac distances revealed that the bacterial community structure of rumen differed between yaks with high and low milk protein yields. Furthermore, rumen microbiota were functionally enriched in relation to transporters, ABC transporters, ribosome, and urine metabolism, and also significantly altered in HH and LL yaks.ConclusionWe observed significant differences in the composition, diversity, fermentation product concentrations, and function of ruminal microorganisms between yaks with high and low milk protein yields, suggesting the potential influence of rumen microbiota on milk protein yield in yaks. A deeper understanding of this process may allow future modulation of the rumen microbiome for improved agricultural yield through bacterial community design.

Effect of controlled atmosphere storage on the quality attributes and volatile organic compounds profile of dragon fruit (Hylocereus undatus)

Abstract The short shelf-life of dragon fruit is one of the inhibiting factors for the export of Vietnamese dragon fruit to distant markets. This study aimed at finding an optimal controlled atmosphere (CA) storage condition for dragon fruit by studying the effect of CA storage on fruit quality and its volatile organic compounds profile. Dragon fruit were stored at 4 different CA conditions and at ambient air as a control treatment. At harvest and at day 10, 20, 30, 40 and 50, different fruit quality attributes were measured, including scale color, firmness, electrolyte leakage, soluble solids content, total acidity, respiration rate, and disease score. Sensory evaluation and aroma analysis were also performed. The results revealed that CA storage significantly reduced the yellowing of the scales and maintained fruit acidity. The CA condition of 2 kPa O2 + 5 kPa CO2 at 6 °C seemed to be optimal for dragon fruit storage. Normal senescence caused the loss of many volatiles, while low O2 and high CO2 level had an opposite impact on the concentration of fermentation products. However, these volatile changes did not significantly impact on the sensory quality of the fruit during storage.

Recombinant bacteriophage LysKB317 endolysin mitigates Lactobacillus infection of corn mash fermentations

BackgroundCommercial ethanol fermentation facilities traditionally rely on antibiotics for bacterial contamination control. Here we demonstrate an alternative approach to treat contamination using a novel peptidoglycan hydrolase (LysKB317) isolated from a bacteriophage, EcoSau. This endolysin was specially selected against Lactobacillus strains that were isolated as contaminants from a fuel ethanol plant. The LysKB317 gene was recombinantly expressed in Escherichia coli as a 33 kDa purified enzyme.ResultsIn turbidity reduction assays, the recombinant enzyme was subjected to a panel of 32 bacterial strains and was active against 28 bacterial strains representing 1 species of Acetobacter, 8 species of Lactobacillus, 1 species of Pediococcus, 3 species of Streptococcus, and 1 species of Weissella. The activity of LysKB317 was optimal around pH 6, but it has broad activity and stability from pH 4.5–7.5 up to at least 48 h. Maximum activity was observed at 50 °C up to at least 72 h. In addition, LysKB317 was stable in 30% ethanol up to at least 72 h. In experimentally infected corn mash fermentations, 1 µM endolysin reduced bacterial load by 3-log fold change, while 0.01 µM reduced bacteria by 2-log fold change. Concentration of fermentation products (ethanol, residual glucose, lactic acid, and acetic acids) for infected cultures treated with ≥ 0.01 µM LysKB317 was similar to uncontaminated controls.ConclusionExogenously added LysKB317 endolysin is functional in conditions typically found in fuel ethanol fermentations tanks and may be developed as an alternative to antibiotics for contamination control during fuel ethanol fermentations.

An iterative approach to improve xylose consumption by Clostridium autoethanogenum: From substrate concentration to pH adjustment

Clostridium autoethanogenum is an autotrophic acetogen also able to utilize xylose as a carbon source. Here, we evaluated this organism's industrially relevant capability since the transformation of sugars derived from lignocellulosic materials to biofuels is of great interest. Initially, the organism was challenged with different initial xylose levels to determine xylose utilization and maximum inhibitory concentrations for C. autoethanogenum when grown in batch cultures. A critical factor that could limit xylose utilization is culture pH, and this factor was separately examined as a means to provide C. autoethanogenum with more suitable environmental conditions. Finally, optimum pH conditions for growth were tested to assess how efficiently initial xylose concentrations could be fully utilized. C. autoethanogenum specific growth rate was not impacted when cells were exposed to initial xylose concentrations up to 30 g L−1, while cultures amended with concentrations as high as 45 g L−1 showed a delay in growth and in formation of final fermentation product concentrations. Intermittent adjustment of the culture pH to 5.8 allowed for complete substrate utilization when step-wise fed-batch xylose additions were performed. Under these conditions the cultures showed a unique biphasic growth which enhanced biomass and end product formation. In C. autoethanogenum cultures exposed to an initial xylose concentration as high as 30 g L−1, pH adjustment to 5.8 supported full utilization of all available xylose and generation of up to 2.4 g L−1 ethanol and 9.5 g L−1 acetate. This work demonstrated that C. autoethanogenum can effectively biotransform xylose at concentrations typically present in lignocellulosic hydrolysate streams.

Comparison of Different Categories of Slovak Tokaj Wines in Terms of Profiles of Volatile Organic Compounds.

The present work deals with the characterization of volatile organic compounds (VOCs) in wines from the Slovak Tokaj wine region. Studied wine samples were divided into three groups—varietal wines from registered Tokaj vine varieties, film wines Tokajské samorodné dry, and naturally sweet botrytized wines Tokaj selections. The VOCs from wines were extracted using optimized solid phase microextraction (SPME) and analyzed by comprehensive two-dimensional gas chromatography (GC×GC) coupled to high-resolution time-of-flight mass spectrometry (HRTOF-MS). In total, 176 VOCs were identified in all 46 studied samples. It was found that the total number of VOCs in varietal wines was generally higher than in botrytized wines. All three studied categories showed characteristic VOC profiles with significant differences. Varietal wines were characterized by higher concentrations of esters and terpenoids originating from grapes. The presence of γ-octalactone, (E)-6-methylhept-2-en-4-one, and lack of benzaldehyde were typical for Tokajské samorodné dry. Tokaj selections expressed the highest concentration of diethyl malate, benzaldehyde, and furfurals. Several interesting trends were also observed. The concentration of fermentation products was highest in varietal wines, while long-term matured Tokaj special wines were typified by the presence of compounds related to noble-rotten raisins (2-phenylacetaldehyde, ethyl 2-phenylacetate, and 2-phenylethanol), wood (cis-whisky lactone), and aging (1,1,6-trimethyl-2H-naphthalene, furfural, and 5-methylfurfural).

Efficacy of different additives in ensiling faba bean and field pea based whole crop silages

Mixed stands of faba bean + wheat (FBW; faba bean 0.84), and pea + wheat (PW; field pea 0.89 on dry matter [DM] basis) were harvested in Siikajoki, Finland. The crops were ensiled in triplicate in laboratory silos using four additive treatments: control without additive, two commercial lactic acid bacteria stains (LAB) and ACID (formic and propionic acids). Ensiling time was 105 days. The crops contained 173 and 181 g DM kg-1 and the water soluble carbohydrate (WSC) concentration was 156 and 176 g kg-1 DM in FBW and PW, respectively. ACID restricted silage fermentation in FBW (WSC 146; fermentation products 43 g kg-1 DM). The other silages had high concentration of fermentation products (180–204) and low WSC concentration (&lt; 22 g kg-1 DM). The volatile fatty acid concentration was below 30 g kg-1 DM in all treatments. PW-ACID treatment resulted in higher ethanol concentration and better aerobic stability (&gt;235 hours) compared to control or LAB silages. When ensiling wet legume whole crops effluent losses, extensive fermentation and poor aerobic stability may cause problems.

Performance, Fermentation Characteristics and Composition of the Microbiome in the Digest of Piglets Kept on a Feed With Humic Acid-Rich Peat.

The transition from breast milk to solid feed is a dramatic change in the nutrition of piglets, frequently necessitating antibiotic treatment. In efforts to reduce the use of antibiotics, dietetic concepts based on natural feed additives are becoming more and more important. In the present study, experiments were carried out with 15 rearing piglets (days 28–56) divided into three groups that were offered different diets (Ctr [0% peat]; H1.5 [1.5% peat]; and H3.0 [3.0% peat] based on a commercial weaner recipe; all ~178 g CP, 13.7 MJ ME, 13.3 g Lys, as-fed). The contents of cecal and colon digesta were removed at necropsy. The gas formation (4 h) in colon digesta was measured using in vitro batch fermenters. For microbiome studies, 16S rRNA amplification was performed within the hypervariable region V 4 and sequenced with Illumina MiSeq platform. DNA read mapping and statistical analysis were performed using QIIME (version 1.8.0), MicrobiomeAnalyst, RStudio, and SAS Enterprise Guide. The mean body weight of the animals at the end of the trial did not show statistical differences (in kg: Ctr: 26.1 ± 4.85, H1.5: 28.5 ± 3.41, H3.0: 26.2 ± 4.92). The daily weight gains were high for this age (in g/day; Ctr: 607 ± 157; H1.5: 692 ± 101; H3.0: 615 ± 113) and the feed to gain ratio low (in kg/kg; Ctr: 1.538; H1.5: 1.462; H3.0: 1.462). Concentrations of short-chain fatty acids in the cecal content were significantly lower when peat was used (mmol/kg wet weight; Ctr: 173 ± 30.0; H1.5:134 ± 15.0; H3.0:133 ± 17.3). Numerical differences were found in the gas formation (in mL gas per 10 mL batch in 4 h; Ctr: 7.9 ± 2.2; H1.5: 7.4 ± 2.4; H3.0: 6.6 ± 1.1). The microbiome analyses in the cecal content showed significantly higher values for alpha diversity Chao 1 index for samples from the control group. Significant differences were found for bacterial relative abundance for Tenericutes at phylum level and Mollicutes at class level (p < 0.05) in cecal microbiota. Therefore, there was initial evidence that peat influences intestinal microflora causing a shift in the overall concentration of fermentation products in both, the cecal and the colon content.

Effect of oxygen exposure during fermentation on volatile sulfur compounds in Shiraz wine and a comparison of strategies for remediation of reductive character

Background and Aims Oxygen (O2) plays a fundamental role in the establishment of wine style and aroma. The effect of O2 treatment during fermentation on the formation of volatile sulfur compounds (VSCs) and the subsequent impact on the sensory profile of wine was investigated. Traditional VSC remediation strategies were also evaluated. Methods and Results Shiraz wines were treated with O2 at several levels during fermentation by sparging rotary fermenters with three types of gas mixtures (40% O2, 21% O2 and N2). The controls were not sparged with any gas and were subsequently subjected to three remedial treatments (aerative racking, early- and late-Cu2+ addition). Wines were analysed for VSCs, fermentation products, concentration of residual metals, as well as their sensory profile. Non-oxygenated ferments and wines contained a higher concentration of VSCs and a lower concentration of fermentation products and differences in the concentration of metals. Volatile sulfur compounds responsive to O2 treatment were identified and a relationship between O2 dose and concentration on VSC formation was demonstrated. The ‘early-Cu2+ addition’ remediation strategy proved the most effective in reducing the impact of ‘reductive’ aromas in non-oxygenated wines. Conclusions The risk of producing a wine with high VSC concentration that negatively impacts wine aroma as well as with a low concentration of fermentation products associated with positive red fruit aromas was decreased by treating wines with O2 during fermentation. Significance of the Study This work specifically examines important aspects of O2 treatment during fermentation in relation to ‘reductive’ aroma characters and the overall impact on the wine's sensory attributes.

Effect of the particle size of maize, rice, and sorghum in extruded diets for dogs on starch gelatinization, digestibility, and the fecal concentration of fermentation products.

The influence of rice, maize, and sorghum raw material particle size in extruded dry dog food on the digestibility of nutrients and energy and the fecal concentration of fermentation products was investigated. Three diets with similar nutrient compositions were formulated, each with 1 starch source. Before incorporation into diets, the cereals were ground into 3 different particle sizes (approximately 300, 450, and 600 µm); therefore, a total of 9 diets were in a 3 × 3 factorial arrangement (3 cereals and 3 particle sizes). Fifty-four beagle dogs (12.0 ± 0.1 kg BW) were randomly assigned to the diets, with 6 dogs per diet. The digestibility was measured with the chromium oxide method. The data were evaluated with ANOVA considering the carbohydrate source, grinding effect, and interactions. The means were compared with the Tukey test and polynomial contrasts (P < 0.05). With the same grinding procedure, rice was reduced to smaller particles than other cereals. The cereal mean geometric diameter (MGD) was directly related to starch gelatinization (SG) during extrusion. For rice diets, the MGD and SG did not change nutrient digestibility (P > 0.05); only GE digestibility was reduced at the largest MGD (P < 0.01). For maize and sorghum diets, the total tract apparent nutrient digestibility was reduced for foods with greater MGD and less SG (P < 0.01). A linear reduction in nutrient digestibility according to cereal particle size was observed for sorghum (r2 < 0.72; P < 0.01). Higher concentrations of fecal total short-chain fatty acids (SCFA) were observed for sorghum diets (P < 0.05) than for other diets. The rice diets led to the production of feces with less lactate (P < 0.05). The increase in raw material MGD did not influence fecal SCFA for rice diets, but for the dogs fed maize and sorghum foods, an increase in propionate and butyrate concentrations were observed as MGD increased (P < 0.05). In conclusion, for dogs fed different particle sizes of the cereal starches in the extruded diets, the digestibility and fecal characteristics were affected, and this effect was ingredient dependent.

Lactic acid production from acidogenic fermentation of fruit and vegetable wastes

This work focused on the lactic acid production from acidogenic fermentation of fruit and vegetable wastes treatment. A long term completely stirred tank reactor (CSTR) lasting for 50days was operated at organic loading rate (OLR) of 11gVS/(Ld) and sludge retention time (SRT) of 3days with pH controlled at 4.0 (1–24day) and 5.0 (25–50day). The results indicated that high amount of approximately 10–20g/L lactic acid was produced at pH of 4.0 and the fermentation type converted from coexistence of homofermentation and heterofermentation into heterofermentation. At pH of 5.0, the hydrolysis reaction was improved and the total concentration of fermentation products increased up to 29.5gCOD/L. The heterofermentation was maintained, however, bifidus pathway by Bifidobacterium played an important role.

This experiment quantified food intake, body weight and body composition responses to three different soluble fermentable dietary fibres in an animal model and explored underlying mechanisms of satiety signalling and hindgut fermentation.MethodsYoung adult male rats were fed ad libitum purified control diet (CONT) containing 5% w/w cellulose (insoluble fibre), or diet containing 10% w/w cellulose (CELL), fructo-oligosaccharide (FOS), oat beta-glucan (GLUC) or apple pectin (PECT) (4 weeks; n = 10/group). Food intake, body weight, and body composition (MRI) were recorded, final blood samples analysed for gut satiety hormones, hindgut contents for fermentation products (including short-chain fatty acids, SCFA) and intestinal tissues for SCFA receptor gene expression.ResultsGLUC, FOS and PECT groups had, respectively, 10% (P < 0.05), 17% (P < 0.001) and 19% (P < 0.001) lower food intake and 37% (P < 0.01), 37% (P < 0.01) and 45% (P < 0.001) lower body weight gain than CONT during the four-week experiment. At the end they had 26% (P < 0.05), 35% (P < 0.01) and 42% (P < 0.001) less total body fat, respectively, while plasma total glucagon-like peptide-1 (GLP-1) was 2.2-, 3.2- and 2.6-fold higher (P < 0.001) and peptide tyrosine tyrosine (PYY) was 2.3-, 3.1- and 3.0-fold higher (P < 0.001). There were no differences in these parameters between CONT and CELL. Compared with CONT and CELL, caecal concentrations of fermentation products increased 1.4- to 2.2-fold in GLUC, FOS and PECT (P < 0.05) and colonic concentrations increased 1.9- to 2.5-fold in GLUC and FOS (P < 0.05), with no consistent changes in SCFA receptor gene expression detected.ConclusionsThis provides animal model evidence that sustained intake of three different soluble dietary fibres decreases food intake, weight gain and adiposity, increases circulating satiety hormones GLP-1 and PYY, and increases hindgut fermentation. The presence of soluble fermentable fibre appears to be more important than its source. The results suggest that dietary fibre-induced satiety is worthy of further investigation towards natural body weight regulation in humans.

In vivo indices for predicting acidosis risk of grains in cattle: Comparison with in vitro methods1

Our objective was to evaluate a near-infrared reflectance spectroscopy (NIRS) used in the feed industry to estimate the potential for grains to increase the risk of ruminal acidosis. The existing NIRS calibration was developed from in sacco and in vitro measures in cattle and grain chemical composition measurements. To evaluate the existing model, 20 cultivars of 5 grain types were fed to 40 Holstein heifers using a grain challenge protocol and changes in rumen VFA, ammonia, lactic acids, and pH that are associated with acidosis were measured. A method development study was performed to determine a grain feeding rate sufficient to induce non-life threatening but substantial ruminal changes during grain challenge. Feeding grain at a rate of 1.2% of BW met these criteria, lowering rumen pH (P = 0.01) and increasing valerate (P < 0.01) and propionate concentrations (P = 0.01). Valerate was the most discriminatory measure indicating ruminal change during challenge. Heifers were assigned using a row by column design in an in vivo study to 1 of 20 grain cultivars and were reassigned after a 9 d period (n = 4 cattle/treatment). The test grains were dry rolled oats (n = 3), wheat (n = 6), barley (n = 4), triticale (n = 4), and sorghum (n = 3) cultivars. Cattle were adapted to the test grain and had ad libitum access to grass silage 11 d before the challenge. Feed was withheld for 14 h before challenge feeding with 0.3 kg DM of silage followed by the respective test grain fed at 1.2% of BW. A rumen sample was taken by stomach tube 5, 65, 110, 155, and 200 min after grain consumption. The rumen is not homogenous and samples of rumen fluid obtained by stomach tube will differ from those gained by other methods. Rumen pH was measured immediately; individual VFA, ammonia, and D- and L-lactate concentrations were analyzed later. Rumen pH (P = 0.002) and all concentrations of fermentation products differed among grains (P = 0.001). A previously defined discriminant score calculated at 200 min after challenge was used to rank grains for acidosis risk. A significant correlation between the discriminant score and the NIRS ranking (r = 0.731, P = 0.003) demonstrated the potential for using NIRS calibrations for predicting acidosis risk of grains in cattle. The overall rankings of grains for acidosis risk were wheat > triticale > barley > oats > sorghum.

An arabinoxylan-rich fraction from wheat enhances caecal fermentation and protects colonocyte DNA against diet-induced damage in pigs

Population studies show that greater red and processed meat consumption increases colorectal cancer risk, whereas dietary fibre is protective. In rats, resistant starches (a dietary fibre component) oppose colonocyte DNA strand breaks induced by high red meat diets, consistent with epidemiological data. Protection appears to be through SCFA, particularly butyrate, produced by large bowel carbohydrate fermentation. Arabinoxylans are important wheat fibre components and stimulate large bowel carbohydrate SCFA production. The present study aimed to determine whether an arabinoxylan-rich fraction (AXRF) from wheat protected colonocytes from DNA damage and changed colonic microbial composition in pigs fed with a diet high (30 %) in cooked red meat for 4 weeks. AXRF was primarily fermented in the caecum, as indicated by higher tissue and digesta weights and higher caecal (but not colonic) acetate, propionate and total SCFA concentrations. Protein fermentation product concentrations (caecal p-cresol and mid- and distal colonic phenol) were lower in pigs fed with AXRF. Colonocyte DNA damage was lower in pigs fed with AXRF. The microbial profiles of mid-colonic mucosa and adjacent digesta showed that bacteria affiliating with Prevotella spp. and Clostridial cluster IV were more abundant in both the mucosa and digesta fractions of pigs fed with AXRF. These data suggest that, although AXRF was primarily fermented in the caecum, DNA damage was reduced in the large bowel, occurring in conjunction with lower phenol concentrations and altered microbial populations. Further studies to determine the relationships between these changes and the lowering of colonocyte DNA damage are warranted.

Metabolic pathways of the colonic metabolism of flavonoids (flavonols, flavones and flavanones) and phenolic acids

Flavonoids are metabolized by the gut microflora giving rise to a wide range of phenolic acids that may exert systemic effects in the body. Nevertheless, the colonic metabolism pathways and the function of the metabolites formed are poorly studied. In the present study, the individual colonic metabolism of three subclasses of flavonoids (flavonols, flavones and flavanones) and phenolic acids was evaluated. For this, seven standards of flavonoids (quercetin, quercetin–rhamnoside, quercetin–rutinoside, myricetin, luteolin, naringenin and kaempferol–rutinoside) and two phenolic acids (protocatechuic acid and gallic acid) were submitted to an in vitro fermentation model using rat colonic microflora. Simultaneously, a nuts–cocoa cream enriched with these standards of flavonoids was incubated and the colonic metabolism of these compounds was evaluated. The results showed that the greatest number of colonic metabolites came from the fermentation of quercetin and quercetin–rhamnoside, and the maximum concentration of fermentation products was observed after 48 h of fermentation. On the other hand, a smaller number of fermentation products were observed after the colonic fermentation of kaempferol–rutinoside, naringenin, luteolin and myricetin. The phenolic acids were slightly metabolized by the colonic microflora.
