The impact of repeated pyrethroid pulses on aquatic communities

Pesticides are considered to be one of the main causes of the decline in macroinvertebrate biodiversity in small streams. In particular, pyrethroids are detected in agricultural surface waters worldwide and pose a high risk to aquatic invertebrates. Due to their knock-down effect, even short pyrethroid exposure pulses can have significant short- and long-term effects on macroinvertebrate communities. Therefore, it is necessary to consider more realistic exposure scenarios for the environmental risk assessment of pyrethroids and, consequently, to obtain more realistic effect data by using multi-stressor test systems.In an experimental setup with artificial indoor streams (AIS), four pyrethroid pulses simulated the exposure scenario of heavy rainfall events. Effects of these 12 h-exposures at different concentrations of deltamethrin (0.64 ng/L, 4 ng/L, 16 ng/L, 64 ng/L) with intervening recovery periods of six days were assessed on an aquatic community consisting of Gammarus pulex, Ephemera danica, Lumbriculus variegatus and Potamopyrgus antipodarum with various lethal and sub-lethal endpoints.The mortality rate of G. pulex significantly increased with increasing deltamethrin concentrations, whereas the mean number of offspring significantly decreased (NOECoffspring: 16 ng/L, LOECoffspring: 64 ng/L). The biomass of L. variegatus decreased with increasing deltamethrin concentrations (NOECdry weight: 16 ng/L, LOECdry weight: 64 ng/L).The findings of this study clearly demonstrate that 12 h-deltamethrin pulses at environmentally relevant concentrations adversely affect an aquatic community. Based on the results of this study a RAC value of 5.33 ng/L is assumed, which is below the concentrations measured in rivers of up to 58.8 ng/L. Unacceptable effects on the entire freshwater environment can therefore not be ruled out. The experimental AIS approach is a useful tool for assessing the effects of repeated pulse exposures that occur during surface runoff events.

Life table analysis and RNA-Seq reveal hormesis and transgenerational effects of deltamethrin on Aphis gossypii.

Deltamethrin, as a highly effective and broad-spectrum insecticide, has been widely used for agricultural pest control such as Aphis gossypii worldwide. Increasing evidence has shown that despite great economic benefits brought by it, deltamethrin has also non-negligible side effects. However, the potential risks and related molecular mechanisms remain largely unclear. Herein, the life table parameters and transcriptome sequencing analyses of the four successive aphid generations were performed to investigate the hormesis and transgenerational effects of deltamethrin on A. gossypii. The life table analysis showed that although the exposure of G0 aphid to 30% lethal concentration (LC30) deltamethrin significantly reduced the net reproduction rate (R0), intrinsic rate of increase (r), and fecundity of G0, but it significantly enhanced the R0 and fecundity of subsequent two generations (G1 and G2) of A. gossypii. Moreover, transcriptomic analyses showed that the signaling pathways related to posttranscriptional regulation (spliceosome), protein processing, longevity regulating, and cell proliferation (DNA replication, homologous recombination and non-homologous end-joining) were significantly up-regulated in G1 or G2 under LC30 deltamethrin treatment. Additionally, we also found that the deltamethrin-sulfoxaflor rotation of G0 and G1 still induced reproductive stimulation, but the reproductive stimulation induced by insecticides rotation treatment was significantly lower than that in the deltamethrin exposure alone. Our study demonstrates that sublethal concentrations of deltamethrin significantly enhanced the offspring fecundity of cotton aphid. In addition, our study also reveals the transcriptional response mechanism of hormesis-induced fecundity increase, providing valuable reference for optimizing the application of deltamethrin in integrated pest management. © 2024 Society of Chemical Industry.

Deltamethrin increased susceptibility to Aeromonas hydrophila in crucian carp through compromising gill barrier

Pyrethroids serve as a significant method for managing and preventing parasitic diseases in fish. Among these, deltamethrin (DEL) is used extensively in aquatic environments. Our previous work has been confirmed that DEL exposure can induce oxidative stress and immunosuppression on the gill mucosal barrier of crucian carp (Carassius auratus). However, it is not clear whether DEL affects the susceptibility of farmed fish to bacterial infection. In this study, fish was pre-exposed to different DEL concentration (0, 0.3 and 0.6 μg L−1) and then challenged by immersion with Aeromonas hydrophila (1.0 × 10^8 CFU mL−1). After immersion challenge, fish pre-exposed to DEL developed prominent lipopolysaccharides level in gill and serum and had a significantly lower survival rate compared to the control group. In DEL pre-exposure fish after immersion, the gill apoptosis levels were significantly higher and disrupted the tight junction barrier by downregulating the zo1 and claudin12. Furthermore, fish pre-exposed to DEL exhibited increased activities of superoxide dismutase (SOD), total antioxidant capacity (T-AOC), and malonaldehyde (MDA) levels in the early stage after immersion but experiencing decreased activities of glutathione peroxidase (GPx) and lysozyme (LZM) in the later stage after immersion. And this process was regulated by the NRF2 pathway. Additionally, fish pre-exposed to DEL after immersion had significantly lower mRNA levels of immune-related genes tlr4, myd88, tnfα, and il-1β. Overall, these findings indicate that DEL damaged the gill barrier, weakened the immune response, raised LPS levels, and heightened vulnerability to A. hydrophila infection in crucian carp, resulting in mortality. Thus, this work will help social groups and aquaculture workers to understand the potential risk of DEL exposure for bacterial secondary infection in cultured fish.

RaTexT®: a novel rapid tick exposure test for detecting acaricide resistance in Rhipicephalus microplus ticks in Brazil

BackgroundAcaricide resistance in cattle ticks is a significant concern in (sub)tropical regions, particularly Brazil. The Larval Packet Test (LPT) is the standard laboratory bioassay for resistance diagnosis, which requires triplicates of seven acaricidal dilutions plus controls to cover larval mortalities ranging between 0 and 100%. The value of the LPT lies in providing resistance ratios based on the ratio between the LC50 calculated with potentially resistant and susceptible ticks. However, LC50 ratios are difficult to translate into practical advice for farmers. Moreover, LPT requires laboratory facilities to maintain susceptible tick colonies, and it takes 6 weeks to obtain the larvae to be tested by LPT derived from engorged female ticks collected from cattle in the field. Our novel approach was twofold: first, we upgraded the LPT to the Resistance Intensity Test (RIT) by adopting the latest WHO guidelines for resistance detection in mosquitoes, which combines a 1 × recommended dose with 5 × and 10 × concentrated doses to reveal low, moderate and high resistance intensity, respectively. This reduced the number of test papers and tick larvae and, more importantly, provided relevant information on the resistance level. Our second innovative step was to abolish testing larvae entirely and expose partly engorged adult ticks to the same acaricidal doses immediately after removing them from cattle in the field. This resulted in the Rapid Tick exposure Test (RaTexT®), wherein partly engorged adult ticks were exposed to an acaricide-impregnated, specially designed matrix providing test results within 24 h. This approach directly compared resistance detection in tick larvae in the RIT with resistance in adult ticks in RaTexT®.MethodsLaboratory validation was conducted in Brazil with resistant and susceptible colonies of Rhipicephalus microplus ticks. For field validation, adult R. microplus ticks collected from different cattle farms in Brazil were evaluated for resistance to RaTexT®, and the results regarding their larval progenies were compared with those for the RIT. Partly engorged adult ticks derived from cattle infested with laboratory and field strains of R. microplus were exposed to deltamethrin in RaTexT® containers, which contained six rows of four interconnected compartments, accommodating five to eight semi-engorged female ticks with a preferred size ranging between 5 and 8 mm. The corresponding larvae of each strain were exposed in the RIT to the same deltamethrin concentrations in filter papers.ResultsIn RaTexT®, mortality in adult ticks from a resistant strain of R. microplus from Seropédica in Brazil was 38.4%, 54.2% and 75.0% at the 1 ×, 5 × and 10 × doses of deltamethrin, respectively. In RIT, mortality of larvae from the same resistant strain was 2.0%, 4.9% and 19.5% at 1 ×, 5 × and 10 × doses, respectively. The results of RaTexT® and RIT agreed since both tests identified a high level of resistance based on a cut-off of 90% mortality.In RaTexT®, mortality of adult ticks from a susceptible strain originating from Porto Alegre was 73.8%, 92.9% and 97.6% at the 1 ×, 5 × and 10 × doses, respectively. In RIT, mortality of larvae from the susceptible strain was 95.2%, 95.2% and 96.8% at the 1 ×, 5 × and 10 × doses, respectively. Interestingly, both tests identified a low number of unexpected resistant individuals in the susceptible strain since the mortality of neither larvae nor adults reached 100%. This effect remained unnoticed in the LPT, wherein a resistance ratio of 159.5 was found based on the LC50 of the resistant strain divided by the LC50 of the susceptible strain. Next, RaTexT® was compared with RIT using adult and larval ticks derived from three field strains of R. microplus in Brazil. RaTexT® detected high levels of resistance to deltamethrin in adult ticks in all strains, which was confirmed in larvae tested by the RIT. Both tests agreed on the same resistance level with significantly lower mortality rates in larvae than in adult ticks.ConclusionsRaTexT® is a novel rapid pen-site test for detecting acaricide resistance in adult livestock ticks. It potentially replaces laborious tests using larval ticks and provides results within 24 h relevant to acaricide resistance management of livestock ticks.Graphical

Relationship between deltamethrin resistance and gut symbiotic bacteria of Aedes albopictus by 16S rDNA sequencing

BackgroundAedes albopictus is an important vector for pathogens such as dengue, Zika, and chikungunya viruses. While insecticides is the mainstay for mosquito control, their widespread and excessive use has led to the increased resistance in Ae. albopictus globally. Gut symbiotic bacteria are believed to play a potential role in insect physiology, potentially linking to mosquitoes’ metabolic resistance against insecticides.MethodsWe investigated the role of symbiotic bacteria in the development of resistance in Ae. albopictus by comparing gut symbiotic bacteria between deltamethrin-sensitive and deltamethrin-resistant populations. Adults were reared from field-collected larvae. Sensitive and resistant mosquitoes were screened using 0.03% and 0.09% deltamethrin, respectively, on the basis of the World Health Organization (WHO) tube bioassay. Sensitive and resistant field-collected larvae were screened using 5 × LC50 (lethal concentration at 50% mortality) and 20 × LC50 concentration of deltamethrin, respectively. Laboratory strain deltamethrin-sensitive adults and larvae were used as controls. The DNA of gut samples from these mosquitoes were extracted using the magnetic bead method. Bacterial 16S rDNA was sequenced using BGISEQ method. We isolated and cultured gut microorganisms from adult and larvae mosquitoes using four different media: Luria Bertani (LB), brain heart infusion (BHI), nutrient agar (NA), and salmonella shigella (SS).ResultsSequencing revealed significantly higher gut microbial diversity in field-resistant larvae compared with field-sensitive and laboratory-sensitive larvae (P < 0.01). Conversely, gut microorganism diversity in field-resistant and field-sensitive adults was significantly lower compared with laboratory-sensitive adults (P < 0.01). At the species level, 25 and 12 bacterial species were isolated from the gut of field resistant larvae and adults, respectively. The abundance of Flavobacterium spp., Gemmobacter spp., and Dysgonomonas spp. was significantly higher in the gut of field-resistant larvae compared with sensitive larvae (all P < 0.05). Furthermore, the abundance of Flavobacterium spp., Pantoea spp., and Aeromonas spp. was significantly higher in the gut of field-resistant adults compared with sensitive adults (all P < 0.05). The dominant and differentially occurring microorganisms were also different between resistant larval and adult mosquitoes. These findings suggest that the gut commensal bacteria of Ae. albopictus adults and larvae may play distinct roles in their deltamethrin resistance.ConclusionsThis study provides an empirical basis for further exploration of the mechanisms underlying the role of gut microbial in insecticide resistance, potentially opening a new prospect for mosquito control strategies.Graphical

Long-lasting residual efficacy of Actellic®300CS and Icon®10CS on different surfaces against Anopheles stephensi, an invasive malaria vector.

Anopheles stephensi, a malaria-transmitting mosquito species, has developed resistance to various insecticides such as DDT, Dieldrin, Malathion, and synthetic pyrethroids. To combat this issue, the World Health Organization (WHO) suggests using Actellic®300CS and Icon®10CS for Indoor Residual Spraying to tackle pyrethroid-resistant mosquitoes. The aim of this research project was to evaluate the susceptibility of An. stephensi to certain insecticides at the diagnostic concentration + intensity 5x diagnostic concentration (5XDC) assays in Iran and to study the lasting effectiveness of Actellic®300CS and Icon®10CS against this particular malaria vector. This study assessed the susceptibility of An. stephensi populations in southern Iran to various insecticides, including deltamethrin 0.05%, DDT 4%, malathion 5%, bendiocarb 0.1%, a synergist assay with PBO 4% combined with deltamethrin 0.05%, and an intensity assay using 5x the diagnostic concentration of deltamethrin (0.25%) and bendiocarb 0.5%. Laboratory cone bioassay tests were conducted to determine the residual effectiveness of Actellic®300 and Icon®10CS insecticides on different surfaces commonly found in households, such as cement, mud, plaster, and wood. The tests were carried out following the WHO test kits and standard testing protocols. The An. stephensi populations in Bandar Abbas were found to be susceptible to malathion 5% and deltamethrin 0.25% (5XDC), but exhibited resistance to DDT, standard concentration of deltamethrin, and both standard and intensity concentrations of bendiocarb. In laboratory cone bioassay tests, An. stephensi mortality rates when exposed to Actellic®300CS and Icon®10CS on different surfaces remained consistently more than 80%. Actellic®300CS achieved more than 80% mortality on all substrates for the entire 300-day post-spraying period. Conversely, Icon®10CS maintained mortality rates more than 80% on plaster and wood surfaces for 165 days and on mud and cement surfaces for 270 days post-spraying. Both Actellic®300CS and Icon®10CS demonstrated 100% mortality within 72 h of each test on all surfaces throughout the entire 300-day post-spraying period. The study shows the varying levels of resistance of An. stephensi Bandar Abbas population to different insecticides and demonstrates the consistent performance of Actellic®300CS in controlling these mosquitoes on various surfaces. The findings suggest that long-lasting CS formulations may be more effective for malaria vector control compared to the current options. Further research is needed to validate these findings in field settings and assess the impact of these insecticides on malaria transmission.

Transgenerational effects of sublethal deltamethrin exposure on development and repellency behaviour in Callosobruchuschinensis

Callosobruchus chinensis, belongs to the Chrysomelidae family, is a major pest of pulse grains. It causes 60% weight loss of the pulses, making them inappropriate for planting or human consumption. C. chinensis is commonly controlled with insecticides; however, these insecticides have negative effects on the environment and non-target organisms. Moreover, repeated exposure to the insecticide causes the resistance in the C. chinensis. Initially the lethal concentration of deltamethrin (LC50) was obtained by the probit analysis. Later the study aimed to examine the impact of a sublethal concentration of deltamethrin on various developmental characteristics and repellency behaviour throughout six successive generations of the C. chinensis. For that, two concentration were selected viz sublethal low concentration (1/20th) and sublethal high concentration (1/5th) of the LC50 compared to control. Both concentrations significantly reduce the life table parameters like egg count, total hatching, total development duration, total emergence and adult longevity in the first generation but an increasing trend in all the parameters was observed in subsequent generations (progeny). Additionally, the repellency was maximum in 1st generation but it decreases with the exposure time and successive generations which shows that it was time and generation dependent. The findings indicate that although there are predominantly adverse effects within the generation, exposure to deltamethrin at sublethal levels over the generations can lead to beneficial effects in the offspring, enhancing their ability to withstand insecticides.

Effect of silver nanoparticles synthesized by Lecanicillium lecanii and their combinations with insecticides (Sivanto and Deltamethrin) on larvae and adults of Tribolium confusum Duv. (Tenebrionidae : Coleoptera)

This research focused on developing a nanopesticide derived from silver nanoparticles (AgNPs) synthesized through the metabolic activities of the fungus Lecanicillium lecanii Gams &amp; Zare 2001). The study aimed to assess its efficacy against the flour beetle Tribolium confusum and explore its synergistic effects with two chemical pesticides, Sivanto 20% and deltamethrin 2.5%. Biotinylated AgNPs were successfully produced using the biomass of the fungus L. lecanii. Characterization of silver nanoparticles was achieved through the observable color change of silver nitrate solution (AgNO₃) from a clear or watery state to red, brown, or dark red, confirmed by UVV is spectroscopy, with the highest absorption peak at 420 nm, indicative of silver element absorption. Transmission electron microscope images illustrated that the biologically prepared AgNPs with L. lecanii exhibited spherical shapes with sizes ranging from 12 to 40 nanometers. Various concentrations of nanopesticide, Sivanto, and deltamethrin were tested for the treatment of grains. A concentration of 1 ml/L of deltamethrin showed superior performance, with the mortality ratio increasing with prolonged exposure. Both Sivanto and deltamethrin (0.5 ml/L and 0.4 ml/L) caused 100% mortality, followed by the nanopesticide (30% concentration), reaching a corrected mortality rate of 39.3% against adults of T. confusum. In the larval experiment, the mortality rate was high when applying AgNPs at a concentration of 100%, comparable to Sivanto 0.5 ml/L under similar conditions. Sub-lethal effects of deltamethrin and permethrin resulted in ultrastructural alterations in scales and oxygen uptake changes of Anabas testudineus (Bloch, 1792)

The present study used scanning electron microscopy (SEM) to assess the toxicity of sub-lethal concentrations of deltamethrin (0.035, 0.007 and 0.0007 mg L−1) and permethrin (0.93, 0.093 and 0.0093 mg L−1) on the ultrastructure of the scales of Anabas testudineus (Bloch, 1792) during a 21 day exposure. The oxygen uptake of the fish during deltamethrin (0.007 and 0.0007 mg L−1) and permethrin (0.093 and 0.0093 mg L−1) exposure was also investigated. The SEM studies revealed abnormal morphological alterations and modifications of fish scales, which were concentration-dependent. Deltamethrin-exposed fish showed severe deformation and fusion of two circuli in different rows, thereby disrupting the normal radii pattern. This fusion was probably caused by the cyanide moiety of the pesticide. On the other hand, permethrin characteristically produced a thick mucus layer over the scale surface. The different concentrations of both the pesticides affected circuli pattern with severe breakage of circuli and loss of lepidonts present over their ridges. Erythrocyte extrusions were also seen at several places over the scale surface. There was significant reduction of oxygen uptake in fish exposed to permethrin at both the concentrations, but in deltamethrin treatments significant reduction occurred only at the higher concentration of 0.007 mg L−1, though this was less than the lower permethrin concentration of 0.0093 mg L−1, and moreover effects of both the pyrethroids on oxygen consumption increased at higher concentrations.

Contribution of Immune Responses to the Cardiotoxicity and Hepatotoxicity of Deltamethrin in Early Life Stage Zebrafish (Danio rerio).

Deltamethrin (DM) is a widely used insecticide that has demonstrated developmental toxicity in the early life stages of fish. To better characterize the underlying mechanisms, embryos from Tg(cmlc2:RFP), Tg(apo14:GFP), and Tg(mpx:GFP) transgenic strains of zebrafish were exposed to nominal DM concentrations of 0.1, 1, 10, 25, and 50 μg/L until 120 h post-fertilization (hpf). Heart size increased 56.7%, and liver size was reduced by 17.1% in zebrafish exposed to 22.7 and 24.2 μg/L DM, respectively. RNA sequencing and bioinformatic analyses predicted that key biological processes affected by DM exposure were related to inflammatory responses. Expression of IL-1 protein was increased by 69.0% in the 24.4 μg/L DM treatment, and aggregation of neutrophils in cardiac and hepatic histologic sections was also observed. Coexposure to resatorvid, an anti-inflammatory agent, mitigated inflammatory responses and cardiac toxicity induced by DM and also restored liver biomass. Our data indicated a complex proinflammatory mechanism underlying DM-induced cardiotoxicity and hepatotoxicity which may be important for key events of adverse outcomes and associated risks of DM to early life stages of fish.

Testing the Sensitivity of the Tropical Bed Bug Cimex hemipterus (Hemiptera: Cimicidae) to Deltamethrin, Phoxim and Propetamphos in Eastern Iran.

Bedbugs are nocturnal ectoparasites that have made a comeback after 20 years and have become one of the main challenges for pest control methods worldwide. Monitoring chemical pesticide resistance is crucial for identifying the best bed bugs management strategies to effectively manage arthropods. This study aims to assess the susceptibility of Cimex hemipterus (Hemiptera: Cimicidae) collected from different parts of Khorasan-Razavi Province, (northeast of Iran) to deltamethrin (pyrethroid), phoxim (organothiophosphate) and propetamphos (phosphoramidate). This study was conducted from Dec 2020 to May 2021. The efficacy of three insecticides (deltamethrin, phoxim, and propetamphos) on adult C.hemipterus was assessed using a bioassay method recommended by the World Health Organization (WHO),. Concentrations of deltamethrin used were 10, 20, 40, 80, 160, 320, 640, 1280, and 2560 ppm, while concentrations of phoxim and propetamphos were 10, 40, 160, and 320 ppm. The bed bugs were continuously exposed to the insecticide for 24 hours, and mortality was assessed at regular intervals during the observation period. The concentration-response data were subjected to POLO-PC software and data were analyzed by the one-way and two-way ANOVA procedures. The lethal concentration fifty values of deltamethrin, phoxim and propetamphos on the examined bed bugs were 0.551, 0.148 and 0.237 ppm, respectively. Insecticide effects of phoxim were significantly higher (P≤ 0.05) compared to each of either deltamethrin and propetamphos agents. The insecticide effects of phoxim against bed bugs were significantly higher compared to each of either deltamethrin or propetamphosinsecticides.

Development of Deltamethrin-Laced Attractive Toxic Sugar Bait to Control Aedes aegypti (Linnaeus) Population.

The attractive toxic sugar bait (ATSB) is a promising strategy for controlling mosquitoes at the adult stage. The strategy is based on the use of a combination of fruit juice, sugar, and a toxin in order to attract and kill the adult mosquitoes. The selection of the components and optimization of their concentrations is significant for the formulation of an effective ATSB. The present study formulated nine ATSBs and evaluated their efficacy against two laboratory strains (AND-Aedes aegypti and AND-Aedes aegypti-DL10) and two wildcaught colonized strains of Aedes aegypti (GVD-Delhi and SHD-Delhi). Initially, nine attractive sugar baits (ASBs) were prepared using a mixture of 100% fermented guava juice (attractant) with 10% sucrose solution (w/v) in 1 : 1 ratio. ATSBs were formulated by mixing each ASB with different concentrations of deltamethrin in the ratio of 9 : 1 to obtain final deltamethrin concentration of 0.003125-0.8 mg/10 mL ATSB. Cage bioassays were conducted with 50 mosquitoes for 24 h in order to evaluate the efficacy of each ATSB against the four strains of Ae. aegypti. The data were statistically analyzed using PASW software 19.0 program and 2-way ANOVA. The ATSB formulations registered 8.33-97.44% mortality against AND-Aedes aegypti and 5.15-96.91% mortality against AND-Aedes aegypti-DL10 strains of Ae. aegypti, while GVD-Delhi strain registered 2.04-95.83% mortality and SHD-Delhi strain showed 5.10-97.96% mortality. The administration of 0.8 mg of deltamethrin within 10 mL of attractive toxic sugar bait (ATSB) has led to the maximum mortality rate in adult mosquitoes. The ATSBs formulated with guava juice-ASB and deltamethrin (9 : 1) showed toxin dose-dependent toxicity by all the four strains of Ae. aegypti. Most effective dosage was found as 0.8 mg deltamethrin/10 mL ATSB which imparted 96% to 98% mortality in adult mosquitoes. The investigations demonstrated the efficacy of deltamethrin-laced ATSB formulations against Ae. aegypti and highlighted the need for conduct of structured field trials and investigating the impact on disease vectors and nontarget organisms.

Nanoencapsulated deltamethrin combined with indoxacarb: An effective synergistic association against aphids.

Widespread pesticide use for decades has caused environmental damage, biodiversity loss, serious human and animal health problems, and resistance to insecticides. Innovative strategies are needed to reduce treatment doses in pest management and to overcome insecticide resistance. In the present study, combinations of indoxacarb, an oxadiazine insecticide, with sublethal concentrations of deltamethrin encapsulated in lipid nanocapsules, have been tested on the crop pest Acyrthosiphon pisum. In vivo toxicological tests on A. pisum larvae have shown a synergistic effect of nanoencapsulated deltamethrin with a low dose of indoxacarb. Furthermore, the stability of deltamethrin nanoparticles has been demonstrated in vitro under different mimicking environmental conditions. In parallel, the integrity and stability of lipid nanoparticles in the digestive system of aphid larvae over time have been observed by Förster Resonance Energy Transfer (FRET) imaging. Thus, the deltamethrin nanocapsules/indoxacarb synergistic association is promising for the development of future formulations against pest insects to reduce insecticide doses.

Olfactory Learning Behavior and Mortality of the Honey Bee Apis mellifera jemenitica in Response to Pyrethroid Insecticide (Deltamethrin).

Honey bees are constantly threatened due to the wide use of pesticides. This study presents the effects of deltamethrin on the mortality, olfactory learning, and memory formation of the native Saudi bee Apis mellifera jemenitica. Topical and oral application of realistic field and serial dilutions of deltamethrin (250, 125, 62.5, and 25 ppm) caused significant mortality at 4, 12, 24, and 48 h posttreatment. Bee mortality increased with the increasing concentration of insecticide at all tested posttreatment times. Highest mortality was observed at 24 h and 48 h after both exposure routes. Food consumption gradually decreased with increasing concentration of deltamethrin during oral exposure. The LC50 of deltamethrin was determined at 12, 24, and 48 h for topical (86.28 ppm, 36.16 ppm, and 29.19 ppm, respectively) and oral (35.77 ppm, 32.53 ppm, and 30.78 ppm, respectively) exposure. Oral exposure led to significantly higher bee mortality than topical exposure of deltamethrin at 4 h and 12 h, but both exposure routes were equally toxic to bees at 24 h and 48 h. The sublethal concentrations (LC10, LC20, and LC30) of deltamethrin significantly impaired the learning during conditioning trials, as well as the memory formation of bees at 2, 12, and 24 h after topical and oral exposure. Thus, deltamethrin inhibits learning, and bees were unable to memorize the learned task.

Residues of Deltamethrin in Pine Needles and Pine Nuts of Catalonia (Spain).

In recent years, recurrent droughts have weakened stone pine (Pinus pinea) forests and facilitated the emergence of harmful pests and diseases, including the Leptoglossus occidentalis. The production of stone pine nuts has declined over the past five years. To control this hemipteran pest, a synthetic pyrethroid insecticide called deltamethrin is being tested. However, it is necessary to estimate the residue left by these treatments in forest stands. Therefore, a fast and robust analytical procedure was developed based on QuEChERS clean-up extraction, followed by gas chromatography coupled with an electron capture detector. This optimized method can detect residual concentrations of deltamethrin in pine nuts and pine needles up to 0.1 and 6 μg kg-1, respectively, with a limit of quantification of 0.4 and 20 μg kg-1. Great recoveries (between 84 and 102%) were obtained for both matrices, and no matrix effect was observed. The results showed that two weeks after spraying, the deltamethrin content in the needles of stone pines decreased by up to 75%, and after nine months, its presence was like that of nontreated trees.

CYP4FB1 and CYP301B1 mediate the cross‐resistance of Laodelphax striatellus (Hemiptera: Delphacidae) to three pyrethroids

AbstractThe small brown planthopper (SBPH), Laodelphax striatellus (Fallén), is an important pest of rice that has developed resistance and even cross‐resistance to insecticides. In a previous study, we found that the resistance mechanism of a deltamethrin‐resistant strain of SBPH (JH‐del) involved the enhancement of detoxification enzyme activity and was unrelated to target resistance. The JH‐del strain exhibited cross‐resistance to fenvalerate. To explore the detoxification enzymes responsible for cross‐resistance to pyrethroids in the SBPH, quantitative real‐time PCR, transgenic Drosophila technology, the GAL4/UAS system and inhibition assays of recombinantly expressed P450s were carried out. Transgenic Drosophila strains da &gt; CYP4FB1 and da &gt; CYP301B1 overexpressing SBPH CYP4FB1 and CYP301B1 showed significantly increased resistance to deltamethrin, fenvalerate and etofenprox. CYP4FB1 and CYP301B1 were overexpressed by 3.41‐ and 4.99‐fold, respectively, in the JH‐del strain. Recombinant CYP4FB1 and CYP301B1 showed good O‐demethylation and O‐deethylation activities on p‐nitroanisole (PNA) and 7‐ethoxycoumarin (EC), respectively. The half‐maximal inhibitory concentrations (IC50) of deltamethrin, fenvalerate and etofenprox on recombinant CYP4FB1 and CYP301B1 were 0.15–1.35 μM, while those of chlorpyrifos and imidacloprid against recombinant CYP4FB1 and CYP301B1 were 6.99–15.91 μM. Therefore, recombinant CYP4FB1 and CYP301B1 have a strong affinity for pyrethroids and a weak binding ability to other types of insecticides. In summary, CYP4FB1 and CYP301B1 are the detoxification enzymes that mediate the cross‐resistance of the JH‐del strain to three pyrethroids. The results revealed the cross‐resistance mechanism of SBPH to pyrethroids and provided a theoretical basis for the resistance management of SBPH.

Distribution and Elimination of Deltamethrin Toxicity in Laying Hens.

Deltamethrin, an important pyrethroid insecticide, is frequently detected in human samples. This study aims to assess the potential effects of deltamethrin on human health and investigate the patterns of residue enrichment and elimination in 112 healthy laying hens. These hens were administered 20 mg·kg-1 deltamethrin based on their body weight. Gas chromatography-mass spectrometry (GC-MS) was used to investigate the residue enrichment pattern and elimination pattern of deltamethrin in the hens. The results indicated a significant increase in the concentration of deltamethrin in chicken manure during the treatment period. By the 14th day of administration, the concentration of deltamethrin in the stool reached 13,510.9 ± 172.24 μg·kg-1, with a fecal excretion rate of 67.56%. The pulmonary deltamethrin concentration was the second highest at 3844.98 ± 297.14 μg·kg-1. These findings suggest that chicken feces contain substantial amounts of deltamethrin after 14 days of continuous administration, and that it can easily transfer to the lungs. After 21 days of drug withdrawal, the residual concentration of deltamethrin in the fat of laying hens was 904.25 ± 295.32 μg·kg-1, with a half-life of 17 days and a slow elimination rate. In contrast, the lungs showed relatively low elimination half-lives of 0.2083 days, indicating faster elimination of deltamethrin in this tissue. These results highlight differences in the rate of deltamethrin elimination in different tissues during drug withdrawal. The fat of laying hens exhibited the highest residue of deltamethrin and the slowest elimination rate, while the lungs showed the fastest elimination rate. Moreover, deltamethrin was found to accumulate in the edible tissues of eggs and laying hens, suggesting that humans may be exposed to deltamethrin through food.

Resistance to deltamethrin and its association with mutation sites in the sodium iron channel domain III gene in Rhipicephalus microplus in Huaihua City of Hunan Province

To investigate the level of deltamethrin resistance and mutation sites in the sodium iron channel gene in Rhipicephalus microplus in Huaihua City, Hunan Province, and to examine the correlation between deltamethrin resistance and mutation sites in the sodium iron channel gene in Rh. microplus. Rh. microplus was sampled from multiple yellow cattle farms in Huaihua City, Hunan Province from June to September 2022, and the level of resistance to deltamethrin was determined in ticks using the adult immersion test. The sodium iron channel domain III gene was amplified in deltamethrin-resistant and wild-type Rh. microplus using PCR assay. Following sequencing and sequence alignment, mutation sites were detected in bases. The sodium iron channel domain III gene in Rh. microplus was translated, and the signal peptide, transmembrane domain, and phosphorylation and glycosylation sites were detected in amino acid sequences. The tertiary structures of the sodium iron channel domain III protein of deltamethrin-resistant and wild-type Rh. microplus were deduced and compared, and the association be tween mutation sites in bases and resistance to deltamethrin was examined in Rh. microplus according the level of deltamethrin resistance, sequence alignment and protein tertiary structure. The median (LC50) and 95% lethal concentrations (LC95) of deltamethrin were 121.39 mg/L and 952.61 mg/L against Rh. microplus, with a resistance factor of 9.24 and level II resistance. The sequence of the sodium ion channel domain III gene was 1 010 bp in size, and mutation sites were detected in two neighboring bases in the sequence of the sodium ion channel domain III gene in deltamethrin-resistant Rh. microplus. Although no signal peptides were found in the sodium iron channel domain III protein of deltamethrin-resistant or wild-type Rh. microplus, 6 trans-membrane domains, 42 phosphorylation sites and 8 glycosylation sites were identified, with a significant difference in the tertiary structure of the sodium iron channel domain III protein between deltamethrin-resistant and wild-type Rh. microplus. Level II resistance to deltamethrin is detected in Rh. microplus in Huaihua City, Hunan Province, and two mutation sites that correlate with the emergence of deltamethrin resistance are identified in the sequence of the sodium iron channel domain III gene in deltamethrin-resistant Rh. microplus.

Low concentrations of acetamiprid, deltamethrin, and sulfoxaflor, three commonly used insecticides, adversely affect ant queen survival and egg laying

Ants are key ecosystem service providers and can serve as important biological control agents in pest management. However, the effects of insecticides on common farmland ant species are poorly understood. We tested the effects of three commonly used insecticides on ants (Hymenoptera, Formicidae). The tested insecticides were acetamiprid (neonicotinoid; formulated as Mospilan 20 SP), deltamethrin (pyrethroid; formulated as Sanium Ultra), and sulfoxaflor (sulfilimine; formulated as Gondola). We tested two ant (Hymenoptera: Formicidae) species with different colony founding strategies, Lasius niger (Linnaeus, 1758) and Myrmica rubra (Linnaeus, 1758). We sprayed their queens with insecticides at concentrations recommended for use in foliar applications in agriculture, i.e., at 1.25 g L−1 (acetamiprid), 0.6 g L−1 (sulfoxaflor), and 0.875 g L−1 (deltamethrin). Further, we diluted the compounds in distilled water and tested them at 10%, 1%, and 0.1% of the field-recommended concentrations, and used distilled water as a control. We monitored the survival of the queens and the number of eggs laid. All three tested insecticides caused severe lethal and sublethal concentration-dependent effects. Even at concentrations three orders of magnitudes lower than recommended for field applications, significantly lower numbers of eggs were found in the queens’ nests. The extent of the sublethal effects of acetamiprid and sulfoxaflor was concentration-dependent and differed between the two ant species. Besides bees and bumblebees, ants represent an important group of hymenopterans that are severely affected even by low concentrations of the tested compounds and therefore should be included in risk assessment schemes.

UDP-glucuronosyltransferase is involved in susceptibility of Chironomus kiiensis Tokunaga, 1936 (Diptera: Chironomidae) to insecticides

UDP-glucuronosyltransferases (UGTs) could transform various exogenous and endogenous compounds, which help detoxification of pesticides in insects. To investigate the role of UGTs in the detoxification metabolism of insecticides in Chironomus kiiensis, CkUGT302M1, CkUGT302N1, CkUGT308N1 and CkUGT36J1 genes were identified with 1449–1599 bp encoding 482–532 amino acids. Four UGT genes shared 40.86∼53.36% identity with other homologous insect species, and expressed in all developmental stages, notably in the larval and adult stages. Expression of CkUGTs was higher in the gastric caecum, midgut and head. Moreover, CkUGTs expression and activity were significantly increased in C. kiiensis larvae in exposure to sublethal concentrations of carbaryl, deltamethrin and phoxim, respectively. To further explore the functions of UGT genes, the CkUGT308N1 was effectively silenced in 4th instar C. kiiensis larvae by RNA interference, which resulted in the mortality of dsCkUGT308N1 treated larvae increased by 71.43%, 111.11% and 62.50% under sublethal doses of carbaryl, deltamethrin and phoxim at the 24-h time point, respectively. The study revealed that the CkUGT308N1 gene in C. kiiensis could contribute to the metabolism of pesticides and provide a scientific basis for evaluating the water pollution of pesticides.
