A highly selective Cd‐based metal‐organic framework fluorescent probe for rapid recognition of cephalothin in water

Nowadays, the problem of antibiotic pollution in the water environment is becoming more and more serious, which has caused great harm to human health and the circulatory system. Therefore, it is extremely crucial to design an effective and sensitive method for the recognition of antibiotics in water. In this research, 1‐(4‐carboxyphenyl)‐1H‐pyrazole‐3‐carboxylic acid (H2L) was used as a ligand to prepare a luminescent two‐dimensional (2D) metal–organic framework (MOF), CdL·H2O (1). 1 can specifically recognize antibiotic cephalothin (CET) in aqueous environments through a fluorescence‐enhanced response, which demonstrated a favorable linear correlation between the fluorescence intensities of 1 and the concentrations of CET (0–50 μM) with a linear fluorescence enhancement slope KEC value of 5.4750 × 104 M−1 and a limit of detection (LOD) of 165 nM. The fast response, good selectivity, anti‐interference ability, and reusability for CET recognition were also verified. 1 represents the inaugural MOF‐based fluorescent material designed for the recognition of CET. Additionally, the detection mechanism for CET was explored in detail with a view to better understanding the fluorescence sensing process.

Concentration of cephalothin in body fluids and tissue samples of Thoroughbred horses.

Cephalothin (CET) concentrations in body fluids (plasma, synovial fluid, pleural fluid, peritoneal fluid, and aqueous humor) and tissue samples (bone, lung, jejunum, hoof, and subcutaneous tissue) were investigated to consider the treatment of infectious diseases in horses. CET 22 mg/kg body weight was intravenously administered to 12 horses. Samples were collected from four different horses at 1, 3, and 5 hr after administration. The CET concentration in body fluids other than aqueous humor was maintained above the MIC90 values of Streptococcus zooepidemicus and Staphylococcus aureus until 5 hr, but it was not maintained above that of S. aureus in bone. CET (22 mg/kg twice a day) is effective for septic arthritis, pleuritis, and peritonitis caused by gram-positive bacteria but ineffective for osteomyelitis.

Cephalothin Stability in Normal Saline, Five Percent Dextrose in Water, and Dianeal® Solutions

ABSTRACT The stability of cephalothin sodium (1 g) in polyvinyl chloride minibags containing 50 mL of 0.9% normal saline, 5% dextrose in water (D5W) or 500 mg in 2L of Dianeal® (containing 1.5% or 4.5% dextrose) was evaluated using a validated, stability-indicating, liquid chromatographic method during 30 days storage at 4° C and room temperature (23°C). Physical inspections and pH determinations also completed on each of the 11 study days during the 30 day storage period. During the 30-day study period all solutions lost more than 10% of the initial cephalothin concentration. In D5W and saline solutions stored at 4°C, 10% of the initial concentration was lost within 17 days, and at room temperature 10% was lost within one day. In Dianeal®, regardless of the dextrose concentration, solutions stored at 4°C, lost 10% of the initial concentration within eight days, and at room temperature 10% was lost within one day. During the 30-day study period the pH decreased in every solution. However, the decrease was less than 1.0 of a pH unit. The colour of solutions stored at room temperature gradually changed during the study period, becoming a deeper yellow. Solutions stored at °C remained clear and colourless throughout the study period. We conclude that cephalothin solutions (1 g/50 mL of D5W or normal saline) stored at 4°C for seven days followed by 12 hours storage at room temperature will retain 90% of the initial cephalothin concentration, whereas, cephalothin concentrations stored in Dianeal® (500 mg/2L) at 4°C will lose approximately 8% of the initial concentration after only three days storage at 4°C followed by an additional 12 hours storage at room temperature.

Alginate Oligomers Inhibit Growth of Bacteria Causing Bovine Mastitis and Potentiate the Activity of Antibiotics Commonly Used For Treatment of the Disease

Alginate oligomers have been shown to disrupt microbial biofilms and reduce minimum inhibitory concentrations (MIC) for many clinically relevant antibiotics to a range of human pathogens. The antibiofilm and potentiating effects of alginate oligomers has previously been focussed on bacterial and fungal strains that play significant roles in chronic human infections. This study on the effects of an alginate oligomer (OligoG CF-5/20) on bacteria that cause bovine mastitis is the first report that investigates a significant disease in animal health. The effect of OligoG CF-5/20 on the inhibitory concentrations of ampicillin, erythromycin, cephalothin and lincomycin to a test panel of nine bacterial strains associated with bovine mastitis were investigated. Two different media were used for cultivation, the standard Mueller-Hinton normally used for bacterial MIC determination, and a skimmed milk medium to mimic the native growth conditions for mastitis bacteria. OligoG CF-5/20 was shown to inhibit growth of all strains tested, and demonstrated a 2 to 8 fold reduction in MICs for erythromycin, cephalothin and lincomycin. The data highlights a potential role for alginate oligomers in potentiating the efficacy of antibiotics, and thereby potentially reducing antibiotic use, in the treatment of mastitis in dairy cattle.

Tissue Accumulation of Cephalothin in Burns: a Comparative Study by Microdialysis of Subcutaneous Interstitial Fluid Cephalothin Concentrations in Burn Patients and Healthy Volunteers

Burn tissue sites are a potential source of bacteremia during debridement surgery. Burn injury is likely to affect the distribution of antibiotics to tissues, but direct evidence of this is lacking. The aim of this study was to directly evaluate the influence of burn trauma on the distribution of cephalothin to peripheral tissues. We used subcutaneous microdialysis techniques to monitor interstitial fluid concentrations of cephalothin in the burnt and nonburnt tissues of adult patients with severe burns following parenteral administration of 1 g cephalothin for surgical prophylaxis. Analogous simultaneous studies conducted with healthy adult volunteers provided reference tissue concentration data. Equivalent tissue exposures were seen for burn and nonburn sites, giving overall median interstitial cephalothin concentrations (from 0 to 240 min) of 2.84 mg/liter and 3.06 mg/liter, respectively. A lower overall median interstitial cephalothin concentration of 0.54 mg/liter was observed for healthy individuals, and the patient nonburnt tissue and volunteer control tissue cephalothin concentrations exhibited significantly different data distributions (P < 0.001; Kolmogorov-Smirnov nonparametric test). The duration of tissue residence for cephalothin was longer for burn patients than for healthy volunteers. The results demonstrate the potential fallibility of using healthy population models to extrapolate tissue pharmacodynamic predictions from plasma data for burn patients.

Microbial Production of 7-Aminocepahlosporanic Acid and New Generation Cephalosporins (Cephalothin) by Different Processing Strategies

The development of beta-lactam antibiotics has been a continuous battle of the design of new compounds to withstand inactivation by the ever-increasing diversity of beta-lactamases. Semisynthetic cephalosporins like cephalothin were synthesized from 7-amino-cephalosporanic acid (7-ACA), and thiophene-2-acetic acid using cephalosporin-C acylase enzyme was studied. The production of cephalosporin-C acylase by Pseudomonas diminuta was used and the growth kinetics studied. The optimum condition of enzyme activity was determined by using response surface methodology. A 2(3) full-factorial composite design was employed for experimental design and the result analyzed. The pH value and temperature for optimum activity were 6.5 and 32 degrees C, respectively. The structural analog compound similar to the side-chain of semisynthetic cephalosporins, e.g., thiophene-2-acetic acid, was added. HPLC data analysis indicate that the concentration of cephalothin was 1.6 mg/mL.

In vitro activity effects of combinations of cephalothin, dicloxacillin, imipenem, vancomycin and amikacin against methicillin-resistant Staphylococcus spp. strains

Backgroundcombinations of drugs has been proposed as an alternative for oxacillin-resistant staphylococci infections, however, limited information about in vitro combinations are available for multi-resistant strains. The objective of this study was to describe the interaction of beta-lactams in combination with vancomycin or amikacin against 26 oxacillin and amikacin-resistant nosocomial Staphylococcus spp. isolates.Methodsactivity of dicloxacillin plus amikacin, cephalothin plus amikacin, cephalothin plus vancomycin, imipenem plus vancomycin and vancomycin plus amikacin was evaluated by checkerboard synergy tests and the fractional inhibitory concentration index (FIC) was calculated. Results: dicloxacillin plus amikacin, and cephalothin plus amikacin were synergistic or partially synergistic in 84.6% and 100% respectively. For nearly half of the isolates the mean concentrations of dicloxacillin, cephalothin and amikacin at which FIC indexes were calculated were achievable therapeutically. Vancomycin plus amikacin had synergistic effect only against two isolates, and partially synergistic in 38.6%. For the combinations vancomycin plus cephalothin and vancomycin plus imipenem the effect was additive in 76.9% and 80.7% respectively.Conclusionin this study the checkerboard analysis showed that amikacin in combination with cephalothin or dicloxacillin was synergistic against most of the resistant strains of S. aureus and coagulase-negative Staphylococcus. Vancomycin in combination with a beta-lactam (cephalothin or imipenem) showed additivity. An indifferent effect predominated for the combination vancomycin plus amikacin. Even though a synergistic effect is expected when using a beta-lactam plus amikacin combination, it is possible that the effect cannot be clinically achievable. Careful selection of antimicrobial combinations and initial MICs are mandatory for future evaluations.

Correlations between milk production and kinetic variables in milk of cephalothin administered to lactating goats

The aim of the present study was to correlate the milk production and the kinetic variables in milk of cephalothin administered to goats. Twenty healthy creole goats in milk production were used. Cephalothin was administered by intravenous route (20 mg/kg b.w.). Milk samples were collected at 0.25, 0.5, 1.0, 2.0, 3.0, 4.0, 5.0, 6.0, 8.0 and 12.0 hours postadministration of the antibiotic. Cephalothin concentrations were measured in milk samples by high performance liquid chromatography. The values (mean &amp;plusmn; standard error) of milk production collected during 24 hours previous to the administration of the antibiotic were 761.5 &amp;plusmn; 111.1 ml. The results of the kinetic variables (mean &amp;plusmn; standard error) of cephalothin in milk were: AUC = 5.4 &amp;plusmn; 1.6 &amp;micro;g/ml/h; C&lt;sub&gt;max&lt;/sub&gt;= 1.1 &amp;plusmn; 0.3 &amp;micro;g/ml and t&lt;sub&gt;max &lt;/sub&gt;= 1.7 &amp;plusmn; 0.1 h. The correlation coefficients AUC-milk production, C&lt;sub&gt;max&lt;/sub&gt;-milk production and t&lt;sub&gt;max&lt;/sub&gt;-milk production were: 0.602 (P &amp;lt; 0.01), 0.596 (P &amp;lt; 0.01) and 0.398 (P &amp;lt; 0.1), respectively. In conclusion, the areas under the curve and the maximum concentrations and the time to reach them in milk are in fact related to the volume of milk produced by the goats

Induction of leafy galls in Acacia mearnsii De Wild seedlings infected by Rhodococcus fascians

Plantlets of blackwattle (Acacia mearnsii De Wild) were inoculated with the bacterium Rhodococcus fascians and cultured in vitro. Leafy galls appeared at the cotyledonary nodes in 75% of the infected plants. The galls were separated from the plants and cultured on a medium containing three-quarters-strength MS salts (Murashige and Skoog, 1962), MS vitamins, 2% sucrose and an antibiotic (cephalothin), supplemented with or without 0.2% activated charcoal. Histological studies conducted from the sixth to the twenty-second day after plant infection revealed the presence of newly formed meristematic centers, first in the axillary region, then on the petioles and lamina of the leaflets around the apical meristem. Approximately 37% of the galls developed one shoot with both concentrations of cephalothin.

Electrochemical determination of Cephalothin antibiotic by adsorptive stripping voltammetric technique

A sensitive and reliable stripping voltammetric method was developed to determine Cephalothin antibiotic drug. This method is based on the adsorptive accumulation of the drug at a hanging mercury drop electrode and then a negative sweep was initiated, which yield a well defined cathodic peak at −625 mV versus Ag/AgCl reference electrode. To achieve high sensitivity, various experimental and instrumental variables were investigated such as supporting electrolyte, pH, accumulation time and potential, drug concentration, scan rate, convection rate and working electrode area. The monitored adsorptive current was directly proportional to the concentration of Cephalothin and it shows a linear response in the range from 4×10 −7 to 1.2×10 −6 mol l −1 (correlation coefficient=0.9995) and the detection limit (S/N=3) is 3.3×10 −9 mol l −1 at an accumulation time of 3 min. The developed AdSV procedure shows a good reproducibility, the relative standard deviation R.S.D.% ( n=10) at a concentration level of 5×10 −7 mol l −1 was 0.94%. Possible interferences by other pharmaceutical drugs and surfactants have been also evaluated. The applicability of this approach was illustrated by the determination of Cephalothin in pharmaceutical preparation and biological fluids such as serum and urine.

Effect of Sub-MICs of Antibiotics on the Hydrophobicity and Production of Acidic Polysaccharide by Vibrio vulnificus

Background: Subinhibitory concentrations of antibiotics, which can occur in vivo, have been demonstrated to alter the production of bacterial virulence factors, including the capsule, or the interaction between microorganism and phagocyte by affecting surface hydrophobicity. Methods: Using a microtiter assay system, the effect of subinhibitory concentrations of amikacin, gentamicin, cephalothin and doxycycline on the surface hydrophobicity and production of acidic polysaccharide by Vibrio vulnificus (8 human isolates, 8 environmental isolates) was determined. Results: All four drugs, in a dose-dependent manner, caused alterations in adherence to polystyrene, a measure of surface hydrophobicity, and the production of acidic polysaccharides, as determined by Alcian blue staining. Conclusion: The changes in capsule production and surface hydrophobicity measured in response to sub-MICs of antibiotics appear to be independent variables.

Selective enrichment broth for the isolation of Aeromonas sp. from chicken meat

Six selective agents (ampicillin, novobiocin, cephalothin, bile salts, brilliant green and ethanol) were tested during the development of a selective enrichment broth for the isolation of Aeromonas sp. from food. Cephalothin at 10 mg/l was found to be the best selective agent owing to its greater selectivity and efficiency in recovering stressed and lower cell concentrations of Aeromonas sp. Higher concentrations (15–25 mg/l) of cephalothin were inhibitory to some strains of A. sobria. Cephalothin (10 mg/l) was incorporated in buffered dextrin broth (BCDB-10) and alkaline peptone water (CAPW-10) and employed for the isolation of Aeromonas sp. from chicken meat naturally and artificially inoculated (with 10 9 cells/ml of A. hydrophila). The highest isolation rate (22%) with naturally contaminated chicken was achieved with CAPW-10 in comparison to 16% with BCDB-10 and 8% with APW. Similarly, from artificially inoculated samples, 100% isolation was accomplished with CAPW-10, against 80% with BCDB-10 and 50% with APW.

Determination of unbound cephalothin in rat blood by on-line microdialysis and microbore liquid chromatography

A method of analysis for the determination of unbound cephalothin in rat blood has been developed. The method was fully automated using an on-line microdialysis procedure. A microdialysis probe was inserted into the jugular vein/right atrium of male Sprague–Dawley rats to examine the unbound cephalothin level in the rat blood after cephalothin administration (50 mg/kg, i.v.). Dialysates were directly input to a liquid chromatographic system using an on-line injector. Samples were eluted with a mobile phase containing methanol–acetonitrile–100 m M monosodium phosphate (pH 5.0) (20:20:60, v/v). The UV wavelength was set at 254 nm for monitoring the analyte. Using the retrograde method, at infusion concentrations of 1 and 5 μg/ml of cephalothin, the in vivo microdialysis recoveries were 48.4±4.5% and 52.9±4.7% for the rat blood ( n=6). Intra- and inter-assay accuracy and precision of the analyses were≤10% in the range of 0.01 through 10 μg/ml. Pharmacokinetic parameters were calculated from the recovery corrected dialysate concentrations of cephalothin versus time data. The results suggest that the pharmacokinetics of unbound cephalothin in blood fitted best to the two-compartmental model following cephalothin administration (50 mg/kg, i.v.).

Transferability of cephalothin to the alveolar cavity in thoroughbreds.

Five Thoroughbreds were classified into 4 groups according to the administration method used for saline solution (saline), ambroxol, and cephalothin sodium (cephalothin). In group A, cephalothin was injected intravenously after oral administration of ambroxol. In group B, cephalothin was injected intravenously after oral administration of saline. Groups C and D were used as control groups. The dose of cephalothin or ambroxol was clinically administrated. Venous blood and bronchoalveolar lavage fluid (BALF) were sampled from each group. In groups A and B, cephalothin concentrations in plasma reached their maximum level 5 min after cephalothin administration and then declined over time. In plasma obtained from groups A and B, there were no significant differences in pharmacokinetic parameters (T1/2, Kel, Vd). By contrast, cephalothin concentrations in BALF reached their peak at 180 min after cephalothin administration in both groups A and B and maintained a relatively high level even after 300 min. These findings indicate that cephalothin requires a relatively long period of time to move from the blood stream to the alveolar cavity, but once transferred to the alveolar cavity, it is preserved for a long time. In groups A and B, cephalothin concentrations in BALF were approximately at the same level. However, in group A, total protein in BALF was lower at 60, 180, and 300 min than the other groups. Then, cephalothin concentration was adjusted to total protein in BALF. After adjustment to total protein in BALF, group A showed a concentration level of cephalothin approximately 1.5-fold higher than that of group B. This suggests that the transferability of cephalothin to the alveolar cavity improves as a result of the oral administration of ambroxol.

Development of beta-lactamase-mediated resistance to penicillin in middle-ear isolates of Moraxella catarrhalis in Finnish children, 1978-1993.

The frequency of beta-lactamase production was analyzed in a study of 1,452 strains of Moraxella catarrhalis and 2,738 strains of Haemophilus influenzae isolated from middle-ear fluid of children < 6 years of age at Tampere University Hospital in Tampere, Finland, between 1978 and 1993. In addition, 401 isolates of M. catarrhalis from similar samples collected in different parts of Finland in 1988-1990 were tested for beta-lactamase production; minimal inhibitory concentrations of ampicillin, cefaclor, cephalothin, erythromycin, tetracycline, and trimethoprim-sulfamethoxazole for these strains were determined. These data were compared with figures for the annual consumption of beta-lactam antimicrobials in the community in 1978-1993. A bimodal increase in the proportion of strains of M. catarrhalis producing beta-lactamase was detected: from 0 to 60% in 1978-1983 and from 60% to 80% in 1988-1990. Concurrently with the second increase, the consumption of cephalosporins increased substantially in the community. The frequency of beta-lactamase-producing strains of H. influenzae did not increase between 1978 and 1993.

Whole-blood platelet aggregation, buccal mucosa bleeding time, and serum cephalothin concentration in dogs receiving a presurgical antibiotic protocol

Summary Whole-blood platelet aggregation (using the impedance method) and adenosine triphosphate (atp) release, buccal mucosal bleeding time (bt), and serum cephalothin concentration were measured in 21 adult female Beagles before (pre) and 1 hour (1hr) after iv administration of cephalothin (22 mg/kg). A second injection of cephalothin (22 mg/kg) was given 3 hours after the first, and blood samples were obtained 1 hour (4 hr, 4 hours after the first injection) and 3 hours (6 hr, 6 hours after the first injection) after the second injection. Samples of jugular blood were obtained from each dog, using citrate as an anticoagulant. A platelet count was obtained for each sample. Platelet aggregation and atp released from the aggregating platelets were measured within 1 hour of sample collection, using a whole-blood aggregometer. Adenosine diphosphate (ADP) and collagen were used as aggregating agents. Aggregation was measured over 6 minutes for each aggregating agent; atp release in response to collagen, but not to ADP, was measured over the same period. For 1 hr samples, there was a significant (P &lt; 0.01) reduction from pre values in the ability of platelets to aggregate in response to ADP. Bleeding time was determined, using a published procedure, with each dog as its own control. Bleeding time during the same period was found to be significantly increased over pre values for 1 hr (P &lt; 0.01) and 6 hr (P &lt; 0.02) samples. There was no significant difference between bt for 1 hr and 4 hr samples. Median serum cephalothin concentrations in pre, 1 hr,4hr, and 6 hr samples did not correlate significantly to platelet aggregation, atp release, platelet count, or bt at any time during the study. Because serum cephalothin concentration was so low, and because there are few reports of the kinetics of cephalothin after iv administration, serum cephalothin was measured in 2 dogs over 2 hours after single iv administration of cephalothin. Values reached a peak at 2 minutes after drug administration, then decreased rapidly over 10 to 15 minutes, and were low by 2 hours.

Maintenance requirements of Escherichia coli ATCC 25922 in the presence of sub-inhibitory concentrations of various antibiotics.

Escherichia coli ATCC 25922 was grown in minimal medium M63, with glucose 0.1 gL as limiting energy substrate, in the presence of sub-inhibitory concentrations of netilmicin, habekacin, tobramycin, dibekacin, amikacin, kanamycin, amoxycillin, ampicillin, cephalothin, cefoxitin and nalidixic acid. Maintenance requirements were determined with a simple relationship derived from batch growth curves. The apparent relationship between maintenance requirements and antibiotic concentration is an exponential increase, log m(c) = log mo + k.c, where m is maintenance, c antibiotics concentration, mo maintenance without antibiotics, and k a constant. Values for k were found to be in the range 0.5-2.0 mg-1.L.

Effect of Sublethal Concentration of Antibiotics on the Adherence of &lt;i&gt;Staphylococcus epidermidis &lt;/i&gt;to Eukaryotic Cells

Adherence of Staphylococcus epidermidis to pharyngeal epithelial cells as measured by [3H]-leucine-labelled bacteria was found to be a simple, objective and highly reproducible technique. Sublethal concentrations of cephalothin, rifampicin and vancomycin significantly reduced the bacterial adherence to epithelial cells, the effect being most marked with 1/2 MIC. Cloxacillin increased the adherence for one strain while it decreased it for the second. The effect was dose-related and the maximum reduction was observed when bacteria were grown with an antibiotic overnight.

An in vitro model for assessing muscle irritation due to parenteral antibiotics

A rat skeletal muscle cell line (L6) was evaluated for its potential to discriminate the muscle-irritating liability of several parenteral antibiotics. The cells were exposed to clinical as well as diluted concentrations of tetracycline, cefoxitin, cephalothin, carbenicillin, erythromycin, ceforanide, cefazolin, and cephaloridine for 1 hr. Control cells were similarly exposed to culture media for 1 hr. The cells were subsequently assayed for their content of the muscle-associated enzyme creatine kinase (CK). Depletion of CK relative to control cultures was utilized as the index of cellular damage. The results of these analyses revealed the following ranking of antibiotic toxicity to L6 muscle cells: tetracycline, erythromycin, cefoxitin > cephalothin, carbenicillin > ceforanide, cefazolin > cephaloridine. The relative order of toxicity of these antibiotics to L6 cells is in good agreement with their reported muscle-irritating liability in man. The correlation between the results obtained in vitro and the irritancy data in vivo suggests that this model may be a useful adjunct to in vivo testing of parenteral antibiotics for muscle-irritation liability.

The role of beta-lactamase and the permeability barrier on the activity of cephalosporins against members of the Bacteroides fragilis group.

The role of beta-lactamase and the permeability barrier on the activity of some beta-lactams against 53 strains of the Bacteroides fragilis group was investigated. Minimal inhibitory concentrations of cefamandole, cefoxitin, and cephalothin were determined with or without the addition of clavulanic acid and (or) ethylenediaminetetraacetate using an agar dilution technique. A significant increase of susceptibility with clavulanic acid indicated a role for beta-lactamase, and with ethylenediaminetetraacetate, a role for a permeability barrier. We found that both beta-lactamase and low permeability decreased the activity of the beta-lactams to some extent depending on the bacterial species and the antibiotic. The species-specific exception was B. distasonis which showed only a permeability barrier to all antibiotics tested.
