Comprehensive two-dimensional gas chromatographic platforms comparison for exhaled breath metabolites analysis.

The high potential of exhaled breath for disease diagnosis has been highlighted in numerous studies. However, exhaled breath analysis is suffering from a lack of standardized sampling and analysis procedures, impacting the robustness of inter-laboratory results, and thus hampering proper external validation. The aim of this work was to verify compliance and validate the performance of two different comprehensive two-dimensional gas chromatography coupled to mass spectrometry platforms in different laboratories by monitoring probe metabolites in exhaled breath following the Peppermint Initiative guidelines. An initial assessment of the exhaled breath sampling conditions was performed, selecting the most suitable sampling bag material and volume. Then, a single sampling was performed using Tedlar bags, followed by the trapping of the volatile organic compounds into thermal desorption tubes for the subsequent analysis using two different analytical platforms. The thermal desorption tubes were first analyzed by a (cryogenically modulated) comprehensive two-dimensional gas chromatography system coupled to high-resolution time-of-flight mass spectrometry. The desorption was performed in split mode and the split part was recollected in the same tube and further analyzed by a different (flow modulated) comprehensive two-dimensional gas chromatography system with a parallel detection, specifically using a quadrupole mass spectrometer and a vacuum ultraviolet detector. Both the comprehensive two-dimensional gas chromatography platforms enabled the longitudinal tracking of the peppermint oil metabolites in exhaled breath. The increased sensitivity of comprehensive two-dimensional gas chromatography enabled to successfully monitor over a 6.5h period a total of 10 target compounds, namely α-pinene, camphene, β-pinene, limonene, cymene, eucalyptol, menthofuran, menthone, isomenthone, and neomenthol.

Impact of the adsorbent material on volatile metabolites during in vitro and in vivo bio-sampling

The increased attraction of biological volatile compounds has opened the route to a wide variety of sampling techniques, amongst which trap tubes packed with adsorbent materials are commonly used. Many types of adsorbent materials are available and the choice of the adsorbent can impact the obtained results in untargeted analysis. Therefore, a proper combination of the adsorbent material and the sample is necessary to increase the robustness and reproducibility of biological studies. In this study, the sampling performance of thermal desorption tubes with six common adsorbent material combinations, i.e., Tenax® TA, Tenax® TA/Carbopack™ B, Tenax® TA/Sulficarb, Tenax® TA/Carbograph™ 5TD, Tenax® TA/Carbograph™ 1TD/Carboxen® 1003, and Carboxen® 1016/Carbograph™ 5TD, was evaluated in two different setups: in vitro and in vivo sampling. The in vitro setup consisted of the headspace dynamic extraction of spiked serum, and a mixture of 19 standards was evaluated in terms of response and reproducibility. The in vivo setup consisted into two parts: the first one was based the evaluation of the standard mixture, which was flash-vaporised into Tedlar® bags containing exhaled breath; the second part was based on the longitudinal monitoring of breath metabolites originating from a beverage intake (i.e., brewed coffee), over a 90 min time period. The tubes were all desorbed and analysed in a comprehensive two-dimensional gas chromatography system coupled to a high-resolution time-of-flight mass spectrometer (GC × GC-HR ToF MS). In both sampling setups, the widest analytes coverage and the overall best extraction yield on the selected compounds were obtained using Tenax® TA, followed by Tenax® TA/Carbopack™ B. Tenax® TA provided the highest sampling reproducibility with 12 %RSD, 10 %RSD and <5 %RSD of the response during the experiments using the in vitro setup, the in vivo setup, and during the longitudinal tracking, respectively.

The sample preparation step involved purge-and-trap, with analyte pre-concentration in adsorption tubes. These were thermally desorbed in a (low-)flow modulated comprehensive two-dimensional gas chromatography system (GC×GC) coupled to time-of-flight mass spectrometry (ToF MS). Because of the high-density information, multivariate tools (correlation analysis, HCA, PCA) were exploited for the final untargeted and unsupervised data analysis. The chemical structures represents some of the unique compounds related to the fruit beer type.

Evaluation of different adsorbent materials for the untargeted and targeted bacterial VOC analysis using GC×GC-MS

The analysis of bacterial volatile organic compounds has gained attraction as a non-invasive way to identify disease-causing organisms, given that bacteria have unique metabolisms and volatile metabolic byproducts. In the present research, different adsorbent materials (Carbopack Y, X, B, Carboxen 1000 and Tenax TA), packed singularly or in combination, were compared in terms of sampling performance (sensitivity, repeatability and selectivity) for the extraction of standards and bacterial volatile metabolites in vitro (from Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli). After extraction, bacterial volatile organic compounds were desorbed and analyzed in a comprehensive two-dimensional gas chromatography system coupled to a time-of-flight mass spectrometer (GC × GC-ToF MS).The results show that Tenax has the greater ability to extract the standard mix as well as volatile organic compounds with better repeatability (4–26 RSD%), higher sensitivity (on average ∼24 fold) compared to Carbopack Y, X and Carboxen 1000 tube, which followed in terms of performance. In addition, Tenax confirmed the best sensitivity and discriminatory power with no misclassification in the untargeted and unsupervised analysis for the differentiation of the bacterial species.

Development of a Combined Heart-Cut and Comprehensive Two-Dimensional Gas Chromatography System to Extend the Carbon Range of Volatile Organic Compounds Analysis in a Single Instrument

The majority of atmospheric measurements of volatile organic compounds (VOCs) are usually limited to a small range, either in volatility or time resolution. A combined heart-cut gas chromatography (GC) with comprehensive two-dimensional GC (GC×GC) instrument was developed, specifically to increase the number of VOCs analysed using a single instrument. The system uses valve based modulation and was fully automated, making it suitable for use in the field. A laboratory comparison to an existing dual-channel GC (DC-GC) instrument demonstrated that this new GC-GC×GC can accurately measure atmospheric mixing ratios of C 5 -C 13 VOC species with a wide range of functionalities. Approximately hourly field measurements were conducted at a remote marine atmospheric research station in Bachok, Malaysia. This region was shown to be influenced by clean marine air masses, local anthropogenic and biogenic emission sources and aged emissions transported from highly polluted South East Asian regions. A dramatic shift in air mass direction was observed each day associated with the development of a sea breeze, which influenced the diurnal profiles of species measured at the Bachok site. A proton-transfer-reaction mass spectrometer (PTR-MS) was also deployed at Bachok and compared to the new GC-GC×GC instrument. Overall, the GC-GC×GC instrument has been shown to perform well in lab comparisons and during field observations. This represents a good compromise between volatility and high complexity o n l i n e measurements of VOCs.

Evaluation of a Rapid-Scanning Quadrupole Mass Spectrometer in an Apolar × Ionic-Liquid Comprehensive Two-Dimensional Gas Chromatography System

Comprehensive two-dimensional gas chromatography (GC×GC) is a powerful technique which can enable a great increase in GC peak capacities. However, since secondary-column separations are very rapid, detectors with a fast acquisition rate are mandatory. Such a requirement has certainly limited the use of the quadrupole mass spectrometer in the GC×GC field. The present research is focused on the evaluation of a novel rapid-scanning quadrupole mass spectrometry (qMS) detector, characterized by a 20,000 amu/s scan speed and a 50 Hz scan frequency, using a 290 amu mass range (40-330 m/z). The performance of the MS system was assessed by analyzing mixtures of 24 allergens, as well as a perfume sample, through GC×GC/qMS. The MS parameters evaluated at different acquisition rates (50, 33, and 25 Hz), as well as in the (simultaneous) scan/selected ion monitoring (SIM) mode, were the number of data points per peak, mass spectrum quality, peak skewing, and sensitivity. Two GC×GC/qMS methods, using the 50 Hz acquisition rate and the scan/SIM mode, were validated. Both methods provided similar results in terms of repeatability, accuracy, and linearity, while a great increase in sensitivity was observed (ca. a factor of 10) under scan/SIM conditions. The validated method proved to be suitable for the analysis of perfume allergens, according to the requirements of Directive 2003/15/EC.

Optimized Use of a 50 μm Internal Diameter Secondary Column in a Comprehensive Two-Dimensional Gas Chromatography System

The focus of the present research is directed toward the development of a comprehensive two-dimensional gas chromatography (GC x GC) method, characterized by a greatly increased separation power, if compared with GC x GC approaches using classical column combinations. The analytical objective was achieved by using a 0.05 mm internal diameter (i.d.) capillary as second dimension, a split-flow approach reported in previous research (Tranchida, P. Q., Casilli, A., Dugo, P., Dugo, G. and Mondello, L. Anal. Chem. 2007, 79, 2266-2275), and a twin-oven GC x GC instrument. The column combination employed was an orthogonal one: an apolar 30 m x 0.25 mm i.d. column was linked, by means of a Y-union, to a flame ionization detector (FID)-connected high-resolution 1 m x 0.05 mm i.d. polar one and to a 0.20 m x 0.05 mm i.d. uncoated capillary segment; the latter was connected to a manually operated split valve, located on top of the second GC. As previously shown, the generation of optimum gas linear velocities in both dimensions can be attained by splitting gas flows at the outlet of the first dimension (Tranchida, P. Q., Casilli, A., Dugo, P., Dugo, G. and Mondello, L. Anal. Chem. 2007, 79, 2266-2275). An optimized GC x GC method was developed and exploited for the analysis of a complex petrochemical sample. The satisfactory results attained were directly compared with those observed using the same instrumentation, equipped with what can be defined as a classical GC x GC split-flow column set: the same primary column was connected to an FID-linked 1 m x 0.10 mm i.d. polar one and to a 0.30 m x 0.10 mm i.d. uncoated capillary. It will be herein illustrated that there is still room for significant progress in the GC x GC field.

Generation of Improved Gas Linear Velocities in a Comprehensive Two-Dimensional Gas Chromatography System

A comprehensive two-dimensional gas chromatography (GC x GC) system (for convenience defined as "split flow" GC x GC), which may be operated at improved gas linear velocities in both dimensions, has been developed. The setup is formed of an apolar 30 m x 0.25 mm i.d. column connected, by means of a Y press fit, to a detector-linked 1 m x 0.1 mm i.d. polar analytical column, which passes through the (cryogenic) modulator, and to a 0.3 m x 0.1 mm i.d. retention gap, which is connected to a manually operated split valve. The latter enables the regulation of gas flows through the second analytical column [e.g., 60:40 (FID) ratio, 50:50 ratio, 40:60 (FID) ratio, etc.], in order to generate the most appropriate gas linear velocity, which is related to each specific analysis. In the pre-sent investigation, two sets of traditional and split flow GC x GC analyses were carried out on a cod liver oil fatty acid methyl ester sample by using the same temperature programs [180-250 degrees C at (a) 3 degrees C/min and at (b) 1.3 degrees C/min] and at an average first-dimension linear velocity of approximately 35.0 cm/s; thus, primary column retention times (and therefore elution temperatures) were essentially maintained. The second-dimension linear velocity was calculated to be approximately 333 cm/s in the traditional applications, while it was split valve-regulated until the most appropriate values [(a) approximately 213 cm/s; (b) approximately 264 cm/s] were attained in the alternative applications. Substantial improvements were observed and measured in the chromatography along the y-axis, while the contour plot chemical class structure was maintained.
