Allelochemicals Identified From Sweet Potato (Ipomoea batatas) and Their Allelopathic Effects on Invasive Alien Plants.

Sweet potato [Ipomoea batatas (L.) Lam] is grown as important cash and food crop worldwide and has been shown to exhibit allelopathic effects on other plants. However, its metabolome has not been studied extensively, particularly with respect to the production of phytotoxic bioactive secondary products. In this study, the chemical composition of petroleum ether extract of sweet potato was characterized, and the morphological and physiological effects of some individual components against four invasive alien weeds Bidens pilosa L., Galinsoga parviflora Cav., Lolium multiflorum Lam., and Phalaris minor Retz. were determined. Twenty-one components were identified by GS-MS, constituting 96.08% of petroleum ether extract in sweet potato. The major components were palmitic acid (PA) (17.48%), ethyl linoleate (EL) (13.19%), linoleic acid (LA) (12.55%), ethyl palmitate (EP) (11.77%), ethyl linolenate (ELL) (8.29%) oleic acid (5.82%), ethyl stearate (4.19%), and 3-methylphenol acetate (3.19%). The five most abundant compounds exhibited strong inhibition activity against the four invasive weeds tested. The highest inhibition rates were seen for LA, followed by PA and EP, respectively. Catalase (CAT), malondialdehyde (MDA), and peroxidase (POD) content of L. multiflorum were increased by the three allelochemicals, i.e., LA, PA and EP, but superoxide dismutase (SOD), chlorophyll-a and chlorophyll-b levels declined. Overall, the combined impact of all five compounds could be quite effective in suppressing the invasive weeds of concern.

CYTOTOXIC EFFECT AND PHYTOCHEMICAL STUDY OF PETROLEUM ETHER EXTRACT OF TILIA CORDATA MILL

The aim of this research was to investigate the chemical composition of petroleum ether extract of Tilia cordata aerial parts as well as to evaluate its cytotoxic activity. Gas chromatography and gas chromatography–mass spectrometry (GC-MS) were used to analyze the unsaponifiable matter and fatty acid methyl esters. Moreover, the cytotoxicity was examined against human hepatoma HepG2 cell line and breast adenocarcinoma MCF cell line. The result showed that thirteen compounds were identified in the fatty acid methyl esters fraction representing 93.71% of the total identified peak area. The major compounds were Octadecanoic acid methyl ester (36.26%) and eicosanoic acid methyl ester (29.42%), whereas nineteen compounds in the unsaponifiable fraction were identified representing 90.56 % of the total beak area. The major compounds were 1- Nonene (30.44%), 1-Hexadecene (24.83%) and phytol (10.40%). Moreover, petroleum ether extract showed a potent cytotoxic effect against human hepatoma HepG2 cell line and a moderate cytotoxic effect on breast adenocarcinoma MCF7 human tumor cell line. So the current research aims to be the first step toward the use of petroleum ether extract of Tilia cordata aerial parts as a potent cytotoxic drug.

Coriandrum sativum seeds extract mitigate progression of diabetic nephropathy in experimental rats via AGEs inhibition.

Inthe present study, we have demonstrated the phytochemical composition of petroleum ether extract of C. sativum (CPE) seeds by using chromatographic, spectroscopic as well spectrometric analysis. CPE was evaluated for its possible role in mitigation of diabetic nephropathy (DN) in Streptozotocin (STZ)-nicotinamide (NAD) induced type 2 diabetes model. Administration of CPE at doses of 100, 200, and 400 mg/kg for 45 days has produced significant attenuation of elevated biochemical parameters including serum glucose, lipid and creatinine levels. CPE has also reserved albuminuria and elevated creatinine clearance in treated diabetic rats. Advanced glycation end products (AGEs) formation in kidneyswas also considerably reduced along with noteworthy increase in level of superoxide dismutase (SOD), glutathione (GSH), and decrease in lipid peroxidation in terms of thiobarbituric acid reactive species (TBARS). Molecular docking studies were also employed to reveal out the possible mechanism. In conclusion, using STZ-NAD model, we have successfully predicted that by assets of bioactive constituents CPE might inhibit the progression of DN. C. sativum may act as potential adjuvant for antidiabetic therapy and needs to be investigated further.

In Vitro and In Vivo Enhancement of Adipogenesis by Italian Ryegrass (Lolium multiflorum) in 3T3-L1 Cells and Mice

Adipogenesis is very much important in improving the quality of meat in animals. The aim of the present study was to investigate the in vitro and in vivo adipogenesis regulation properties of Lolium multiflorum on 3T3-L1 pre-adipocytes and mice. Chemical composition of petroleum ether extract of L. multiflorum (PET-LM) confirmed the presence of fatty acids, such as α-linolenic acid, docosahexaenoic acid, oleic acid, docosatetraenoic acid, and caprylic acid, as the major compounds. PET-LM treatment increased viability, lipid accumulation, lipolysis, cell cycle progression, and DNA synthesis in the cells. PET-LM treatment also augmented peroxysome proliferator activated receptor (PPAR)-γ2, CCAAT/enhancer binding protein-α, adiponectin, adipocyte binding protein, glucose transporter-4, fatty acid synthase, and sterol regulatory element binding protein-1 expression at mRNA and protein levels in differentiated adipocytes. In addition, mice administered with 200 mg/kg body weight PET-LM for 8 weeks showed greater body weight than control mice. These findings suggest that PET-LM facilitates adipogenesis by stimulating PPARγ-mediated signaling cascades in adipocytes which could be useful for quality meat development in animals.

Chemical Composition and Cytotoxic Activities of Petroleum Ether Fruit Extract of Fruits of <i>Brucea javanica</i> (Simarubaceae)

Purpose: To investigate the chemical composition and antitumor activity of the petroleum ether extract of the dried ripe fruits of Brucea javanica. Methods: The composition of petroleum ether extract was analyzed by gas chromatography/mass spectrometric (GC/MS) and their antitumor activities were determined by MTT assay. Results: GC/MS spectrometry results indicate that the petroleum ether extract was a mixture of esters, fatty acids, sterides, pregnanones, terpenes, alkaloids, alkenes, alcohols, ketones, aldehydes and other compounds. The results also revealed the significant antitumor activity of the extract with IC50 of 9.14, 12.45, 15.15, 16.13, 22.26, and 27.97 μg/mL against A549, CNE, MCF-7, NCI-H460, HepG2, and KB-3-1 cell lines, respectively. Conclusion: The study establishes the chemical composition and cytotoxic activity of the petroleum ether extract of the plant fruits. There is need for further investigations to isolate more potent compounds and structurally modify the known compounds to retain activity and lower toxicity and thus lead to the possible development of Brucea javanica oil.
