Characterisation of cell wall polysaccharides, arabinogalactans-proteins (AGPs) and phenolics of Colanitida, Colaacuminata and Garciniakola seeds

Many Cola plant species are endemic to West and Central Africa. Cola acuminata and Cola nitida are used as masticatory when fresh, while the dried nuts are used for beverages and pharmaceutical purposes in Europe and North America. Garcinia kola seeds, that serve as a substitute for the true kola nuts, are used in African traditional medicine for the treatment of various diseases, including colic, headache and liver cirrhosis. Seeds extracts of G. kola are also known for their anti-inflammatory, antimicrobial and antiviral properties. To gain information on the chemical properties of the kolas, we have isolated and analyzed cell wall polysaccharides, arabinogalactan-proteins and phenolic substances from the seeds of the three kola species. The sugar composition of cell wall material of C. acuminata, C. nitida and G. kola revealed that Gal (up to 30%), Ara, GalA and Glc as the predominant monosaccharides, representing approximately 90% by mol of the total hydrolysable sugar present in this material. In Ammonium oxalate cell wall fraction, GalA was found to be the major sugar present in all kola species. In the alkali-soluble fraction, there were significant differences in the level of Glc and Gal. The level of Glc was high in C. acuminata and C. nitida while the level of Gal and Xyl were high in C. nitida and G. cola. Isolation and quantification of arabinogalactan-proteins demonstrate that G. kola seeds contained four to eight times more of these proteoglycans than the seeds of the other two species. Finally, analysis of soluble phenolic substances shows that caffeine and catechin were largely represented in C. acumina and C. nitida seeds, with caffeine accounting for ∼50% of all soluble phenolics. These findings indicate that the three Kola seeds are highly enriched in pectins and proteoglycans and that C. acuminata and C. nitida can be used as a possible source of caffeine and catechin.

Chemical Composition of Cell Wall Material from Sweet Potato Starch Residue

AbstractCell wall material was isolated from the residue obtained after the extraction of sweet potato starch by treatment with the α‐amylase. The wall material was fractionated by successive extraction with various reagents. The pectic substance was mainly composed of uronic acid (47.1%) residues and contained galactose as the predominant neutral sugar residues. The hemicellulosic fraction was further fractionated by precipitation with iodine in the presence of calcium chloride to give two fractions, a iodine precipitatable‐iodine complex (HC‐IP) and a supernatant solution (HC‐IS). The HC‐IS fraction had a higher glucose content (53.4%), whereas the HC‐IP fraction had a high content of xylose residues (35.1%). The HC‐IP fraction consisted mainly of xylan, as judged by the degradation with purified xylanase.

Analysis of mannans of two relatively avirulent mutant strains of Candida albicans.

We previously reported the isolation of two cerulenin-resistant mutant strains of Candida albicans 4918 that differ in adherence properties and are less virulent than the parental strain. In addition, biochemical characterization demonstrated significant differences in both protein and polysaccharide composition of cell wall material between the mutant and wild-type strains. These observations prompted studies concerning the chemical structure of mannans in these strains. After extraction and subsequent purification by ion-exchange chromatography, mannan fractions were subjected to either mild acid hydrolysis, alkali hydrolysis, or acetylation followed by acetolysis. Acid- and alkali-modified mannans were studied by proton magnetic resonance spectroscopy, and released products were analyzed by high-performance liquid chromatography on an Aminex HPX-42A column. The results demonstrated quantitative and qualitative differences between mannooligosaccharides of the wild-type and mutant strains in the identity of released oligosaccharides as well as in linkage of the oligosaccharides to the protein backbone.

The monosaccharide composition of cell wall material in cassava tuber (Manihot utilissima).

The monosaccharide composition of cell wall material (CWM) in the cassava tuber and the contents of the other constituents were determined for more advanced industrial utilization. Starch, 80% ethanol–soluble sugar, uronic acid, lignin, ash, and CWM contents in the cassava tuber 86.1, 2.4, 3.4, 0.5, 0.9, and 4.5%, respectively. Rhamnose, fucose, arabinose, xylose, mannose, galactose, and glucose contents in CWM were 1.9, 1.2, 2.6, 4.2, 2.0, 12.8, and 52.7%, respectively. Then, the degradation pattern of CWM by enzymatic and sequential acid hydrolysis was studied. Aspergillus niger cellulase preparation was the most effective, and 57.1 % of CWM was degraded by the enzyme preparation. On the other hand, about 50% of the hemicellulose part was extracted from CWM by hot water only.
