Comparative analysis of physicochemical properties, bioequivalence, safety and tolerability of the first domestic semaglutide

Semaglutide is a representative of analogues of the incretin hormone human glucagon-like peptide-1 (GLP-1) and is currently used in Russia for the treatment of type 2 diabetes mellitus (T2DM; in monotherapy and in combination therapy), including patients with obesity and overweight.The aim of the work was to conduct a comparative assessment of the physicochemical properties, a biological activity, bioequivalence and safety, including tolerability and immunogenicity, of the drug Quincent® (semaglutide, 1.34 mg/ml, a solution for a subcutaneous administration, Promomed Rus LLC, Russia) and the drug Ozempic® (semaglutide, 1.34 mg/ml, a solution for a subcutaneous administration, Novo Nordisk A/S, Denmark) when administered to healthy volunteers.Materials and methods. To assess the degree of similarity of the study drug Quincenta® (semaglutide, 1.34 mg/ml, a solution for a subcutaneous administration, Promomed Rus LLC, Russia) with a chemically synthesized active substance to the original (reference) drug Ozempic® (semaglutide, 1.34 mg/ml, a solution for a subcutaneous administration, Novo Nordisk A/S, Denmark), a comparative study of physicochemical properties and a biological activity was carried out. To assess the bioequivalence of the study drug and the reference drug, an open randomized parallel comparative study with the participation of healthy volunteers (n=54), 54 participants of which had been included in the population, was conducted. The volunteers were randomized into 2 groups in a 1:1 ratio, and received a single dose subcutaneously either of the study drug (domestic semaglutide at a dose of 0.5 mg) or the reference drug (foreign semaglutide at a dose of 0.5 mg). The mode of administration was in the morning on an empty stomach. A semaglutide concentration was determined in serum samples using a previously validated enzyme-linked immunosorbent assay (ELISA) method. A quantitative determination of antibodies to semaglutide in the human serum by ELISA was carried out with a microplate photometer using ready-made kits pre-validated by the manufacturer. The conclusion about the bioequivalence of the compared drugs was made using an approach based on the assessment of 90% confidence intervals for the ratios of the geometric mean values of the parameters Cmax, AUC(0–t) of semaglutide in the measurement original units.Results. The results of the comparative analysis of the study drug and the reference drug demonstrate the comparability of their physicochemical properties and biological activity. The results of the clinical study demonstrated the bioequivalence of the test drug and the reference drug. Thus, the pharmacokinetic parameters of the drugs were comparable to each other: the Cmax value for the study drug was 42.088±8.827 ng/ml, for the reference drug Ozempic® it was 42.2556±7.84. Herewith, the half-life for the study drug and the reference drug was 168.39±39.47 and 157.99±28.57 hours, respectively. The resulting 90% confidence intervals for the ratio of the Cmax and AUC0–t values of the study drug and the reference drug were 90.89–109.15 and 91.66–111.27%, respectively. The tolerability of the drugs in the volunteers was notified as good. No adverse events were recorded during the study. No serious adverse events were reported throughout the study. According to the results of the immunogenicity analysis, no antibodies to Russian-made semaglutide were detected in the blood serum of the volunteers, which indicated the lack of Results. The bioequivalence and a similar safety profile, including the immunogenicity and tolerability of the Russian drug Quincenta® (semaglutide 1.34 mg/ml, Promomed Rus LLC, Russia) were shown in comparison with the foreign drug Ozempic® (semaglutide 1.34 mg/ml, Novo Nordisk A/C, Denmark).

Comparative study of physicochemical properties of geopolymers prepared by four Moroccan natural clays

The present work concerns the development of geopolymer materials based on natural clays rich in kaolinite, muscovite and pyrophyllite. The aim was to study the effect of the nature and fraction of the clay minerals composing the raw sample on the physico-chemical properties of the prepared geopolymers, viz., the matrix geopolymer phase fraction, particle size distribution, density, compressive strength and the adsorption capacity of methylene blue (MB) as a cationic dye model. The raw starting clays, their meta-clay phases calcined at 800 °C, and the prepared geopolymers were characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), particle size distribution (PSD) via laser diffraction method, and scanning electron microscopy (SEM). The matrix geopolymer fraction phase in the samples was qualitatively determined by the analysis of the spectra obtained by 27Al and 29Si solid-state NMR. The results showed that kaolinite-rich clay produced more geopolymer than clays with high phase fractions of muscovite or pyrophyllite and low kaolinite. The highest compressive strength was given by the clay that contained 26.7 % of calcite due to formation of calcium silicate hydrate phase. The maximum adsorption capacity of MB was obtained by the samples have a more heterogeneous surface morphology due to presence of unreacted muscovite and/or pyrophyllite in raw starting clay.

Comparative Study of Physicochemical Properties of Finely Dispersed Powders and Ceramics in the Systems CeO2–Sm2O3 and CeO2–Nd2O3 as Electrolyte Materials for Medium Temperature Fuel Cells

Finely dispersed (CeO2)1−x(Sm2O3)x (x = 0.05, 0.10, 0.20) and (CeO2)1−x(Nd2O3)x (x = 0.05, 0.10, 0.15, 0.20, 0.25) powders were synthesized via liquid-phase techniques based on the co-precipitation of hydroxides and were used to obtain ceramic materials comprising fluorite-like solid solutions with CSR in the range 69–88 nm (upon annealing at 1300 °C) and open porosity in the range 0.6–6.2%. The physicochemical properties of the synthesized materials were comparatively characterized. In general, the prepared materials were found to possess a mixed type of electrical conductivity, but in the medium-temperature range, the ionic component was predominant (ion transfer numbers ti = 0.93–0.73 at 300–700 °C). The highest ionic conductivity was observed for CeO2-based samples containing 20 mol.% Sm2O3 (σ700°C = 3.3 × 10−2 S/cm) and 15 mol.% Nd2O3 (σ700°C = 0.48 × 10−2 S/cm) was in the temperature range 500–700 °C. The physicochemical properties (density, open porosity, type and mechanism of electrical conductivity) of the obtained ceramic materials make them promising as solid oxide electrolytes for medium temperature fuel cells.

Comparative Study of Physicochemical Properties of Alginate Composite Hydrogels Prepared by the Physical Blending and Electrostatic Assembly Methods.

Alginate hydrogel commonly suffers from defects, such as weak mechanical properties, the shortage of long-term stability in physiological medium and the lack of mammalian cell adhesivity due to its strong hydrophilicity in biomedical application. For this reason, the homogeneous alginate hydrogels (Alg Gel) were successfully prepared by the D-glucono-δ-lactone/hydroxyapatite (HAP/GDL) cross-linking system, and then, the physical blending and alternating electrostatic assembly technology were proposed to fabricate alginate composite hydrogels (Alg-GT, Alg-CS-GT and ALG/GT-CS). The feasibility of the design methods was verified through the comparative analysis of their physicochemical properties and biological activity. In particular, the effects of physical blending and alternating electrostatic assembly technology on the pore structure, mechanical properties, swelling, degradation, cell adhesion and proliferation of composite hydrogels were also investigated. Experimental results showed that the formation of polyelectrolyte complexes by electrostatic assembly between biological macromolecules and the covalent cross-linking of EDC/NHS to GT improved the vulnerability of ion cross-linking, enhanced the mechanical properties and swelling stability of the composite hydrogels, and regulated their pore structure and in vitro biodegradability properties. Furthermore, MC3T3-E1 cells could exhibit good cell adhesion, cell viability and cell proliferation on the alginate composite hydrogels. Among them, Alg-CS-GT showed the best cell proliferation ability and differentiation effect due to its good cell adhesion. In view of the excellent physicochemical properties and biological activity of Alg-CS-GT, it exhibited great potential in biomedical application for tissue engineering.

Comparative study of physico-chemical properties of soil according to the age of aquaculture pond of Bangladesh

Soil quality is an important factor in fish pond productivity as it controls pond bottom stability, pH of overlying water and concentrations of plant nutrients required for the growth of phytoplankton. A satisfactory site for constructing fishponds is that where the soil is very deep with neutral pH, weather infiltration is very low; mineralization of organic matter takes place rapidly. The present study was carried out to assess different soil parameters in different aged ponds and to find out the relationship between different aged ponds with different bottom soil parameters. Three groups of ponds (1-5 years, 6-10 years and above 10 years) were analyzed. There was no significant change of soil pH among different aged ponds. Significant variations were observed in textural compositions of different aged group pond’s soil. Mean percentages of clay and silt were found to be increasing with the increase of pond’s age. Amount of organic matter and organic carbon also increased sharply with the increasing of pond’s age. No significant variation were found from different aged group ponds for soil pH but the amount of organic matter, organic carbon, silt and clay were significantly increased with the increasing of pond age that may deteriorate the quality of pond’s bottom soil.

Comparative Study of Physicochemical Properties and Enzymatic Status of Imidacloprid Treated Soils from Various Zones of Haryana, India

Imidacloprid is the most commonly used insecticide in Haryana that play animportant role in pest management and hence improved crop yield production. This study was done to investigate the consequences of Imidacloprid on the activity of dehydrogenase and urease enzymes in soils of selected zones of Haryana. A 28 days experiment was carried out where sampling was done after 1st, 7th, 14th, 21st and 28th day of Imidacloprid application at different concentrations like T1 (1 mg/kg), T2 (2 mg/kg) and T3 (10 mg/kg). After analysing the physicochemical properties of soils of various zones the enzymatic activities were measured using colorimetric assays. Dehydrogenase activity was decreased initially after imidacloprid exposure but after 14 days a stimulation was observed in dehydrogenase activity at lower doses. The decrease was directly proportional to concentration of imidacloprid and indirectly proportional to time of incubation. Urease activity was inhibited by highest dose of imidacloprid after first day exposure. However, a stimulation in urease activity was observed after 14 days of imidacloprid treatment at all doses. Obtained results showed that the activity of dehydrogenase and urease were affected by concentration and treatment time of Imidacloprid significantly in soils of each selected zone of Haryana. The results provide a clue about the possible ecological effects of Imidacloprid.

Comparative Study of Physico-Chemical Properties of Sediments from Two Sectors Close to Bandama in Northern Côte D’Ivoire (Sinematiali and Niakaramadougou)

This study aims to show the impact of successive floods on the spatial variation of the physico-chemical properties of sediments along the Bandama River in the localities of Sinématiali and Niakaramadougou because of their importance in the functioning of this ecosystem. Several samples were taken from both stations based on flood recurrence areas. The particle size analysis was done using the Robinson’s Pipette method. Traditional methods of sediment analysis have been used to measure organic carbon (O.C.), nitrogen (N), and other chemical properties including pH, organic matter (MO), and C/N ratio. Statistical analyzes were carried out to assess the differences between the physico-chemical parameters of the different sampling zones. In the area of niakamadougou, the lower values in MO were recorded in areas subject to more frequent flooding, thus close to the watercourse. In the Sinématiali area, lower M.O. values were recorded in sediments far from the stream. Total organic matter levels are higher in surface sediments with the lowest proportions of clay. The results show that the physico-chemical properties of the sediments vary at the level of the vertical distribution and according to their spatial distribution. Successive floods have a direct effect on the dynamics of the physico-chemical properties of sediments along the shore.

A comparative study of physicochemical properties, antioxidant and enzyme inhibition activities of oils extracted from seeds of seven new sunflower (Helianthus annuus L.) lines

This study was aimed to evaluate the physicochemical properties and chemical profile of seeds oil obtained from new seven sunflower (Helianthus annuus L.) lines as well as their antioxidant and enzyme inhibition activity. The seeds powder was extracted by maceration in n-hexane. The oil content of the seven lines was ranged from 20.04% to 36.65% and was either yellow or pale yellow in color. No significant variations were observed on the refractive index (1.46 unit) of the oil. Oils of the seven lines were significantly (p ? 0.05) different in their saponification values (32.13-282.66 mg KOH/g oil), peroxide values (1.76-13.26 mg KOH/g-oil), acid values (0.016?1.766 mg KOH/g oil) and free fatty acids content (6.26-72.23 mg KOH/g-oil). The chemical profile of the oil revealed that line APO42 contained the highest amount of monounsaturated fatty acids (55.9%). All the seven lines contained a considerable amount of linoleic acid (27.5%- 42.5%), and it represented the major compound in lines BOH3 (42.5%) and H1733 (39.8%). The variation was remarkable in their oleic acid content where the highest amount was observed in lines APO42 (55.4%) and APO43 (42.2%), respectively. Lines BOH3, APO43, APO41, and H1733 exerted the best total antioxidant activity in addition to their capacity to reduce Cu2+ and Fe3+, while lines H1733 and APO43 had a metal chelating activity as well. All oils showed weak acetylcholinesterase, butyrylcholinesterase, α-glucosidase and α-amylase inhibitory activities. Only four lines showed considerable enzyme inhibitory activity against tyrosinase enzymes. Multivariance analysis suggested that linoleic acid participated in the observed biological activity of the oils. In conclusion, these new lines might contribute to the nutritional and phytotherapeutic properties of sunflower oil in addition to other industrial applications.

Comparative Study of Physicochemical Properties of Nanoemulsions Fabricated with Natural and Synthetic Surfactants

This work aims to evaluate the effect of two natural (whey protein isolate, WPI, and soy lecithin) and a synthetic (Tween 20) emulsifier on physicochemical properties and physical stability of food grade nanoemulsions. Emulsions stabilized by these three surfactants and different sunflower oil contents (30% and 50% w/w), as the dispersed phase, were fabricated at two levels of homogenization pressure (500 and 1000 bar). Nanoemulsions were characterized for droplet size distribution, Zeta-potential, rheological properties, and physical stability. Dynamic light scattering showed that droplet size distributions and D50 values were strongly affected by the surfactant used and the oil content. WPI gave similar droplet diameters to Tween 20 and soy lecithin gave the larger diameters. The rheology of emulsions presented a Newtonian behavior, except for WPI-stabilized emulsions at 50% of oil, presenting a shear-thinning behavior. The physical stability of the emulsions depended on the surfactant used, with increasing order of stability as follows: soy lecithin < Tween 20 < WPI. From our results, we conclude that WPI is an effective natural replacement of synthetic surfactant (Tween 20) for the fabrication of food-grade nanoemulsions.

Comparative study of physicochemical properties of dextran and reuteran synthesised by two glucansucrases that are highly similar in amino acid sequence

AbstractRecently, lactic acid bacteria (LAB)‐derived α‐glucans have drawn strong attention owing to their various structural organisation and wide (potential) applications in the medicine, food, cosmetic, and biotechnology industries. However, the physicochemical properties of diverse α‐glucans, particularly reuteran, remain largely unknown, which significantly impedes their wider applications. This study comparatively evaluated the physicochemical properties of two different α‐glucans, Gtf‐DSM dextran, and GtfO reuteran produced by the dextransucrase Gtf‐DSM from Lactobacillus ingluviei DSM 14792 and the reuteransucrase GtfO from Lactobacillus reuteri ATCC 55730, which are two glucansucrases highly similar in amino acid sequence. The surface morphology analysis demonstrated that Gtf‐DSM dextran had a holey and fissured microstructure; however, GtfO reuteran had an irregular dendritic microstructure, forming a fibrous network. Compared to GtfO reuteran, Gtf‐DSM dextran showed a larger average particle diameter (162.4 versus 90.3 nm). Additionally, Gtf‐DSM dextran displayed some prominent properties, such as better thermostability, higher apparent viscosity, and stronger gel‐forming capability, than GtfO reuteran. These results provide a theoretical basis for stimulating the applications of LAB‐derived α‐glucans in various fields.

Comparative Study of Physicochemical Properties and Starch Granule Structure in Seven Ginkgo Kernel Flours.

Ginkgo biloba L. is an important economic tree species in China, and its kernels have been used as a popular food in Asian countries. Herein, the morphology, basic chemical components, starch granule structures, and physicochemical properties of kernel flours from seven ginkgo cultivars were investigated, and their relationships were analyzed. The kernels were oval or spherical in shape, with variable sizes. The starch granules exhibited both regular and irregular Maltese cross patterns. Amylose was mainly distributed in amorphous growth rings. A spatial variation in the 865/942 cm−1 ratio was observed within individual starch granules. Variations in total starch content, apparent amylose content (AAC), crude protein content (CPC), total amino acid content (TAAC), starch fine structure, and thermal and pasting properties were observed among the seven kernel flours. Pearson correlation coefficients and principle component analyses showed that the thermal properties were affected by kernel CPC, TAAC, AAC, and starch fine structure, while the pasting properties were affected by AAC and starch fine structure. Furthermore, experiments showed that the seed protein structure and α-amylase activity affected the pasting properties of ginkgo kernel flours.

Comparative Study of Physicochemical Properties and Biocompatibility (L929 and MG63 Cells) of TiN Coatings Obtained by Plasma Nitriding and Thin Film Deposition.

Ti6Al4V is one of the most lightweight, mechanically resistant, and appropriate for biologically induced corrosion alloys. However, surface properties often must be tuned for fitting into biomedical applications, and therefore, surface modification is of paramount importance to carry on its use. This work compares the interaction between two different cell lines (L929 fibroblasts and osteoblast-like MG63) and medical grade Ti6Al4V after surface modification by plasma nitriding or thin film deposition. We studied the adhesion of these two cell lines, exploring which trends are consistent for cell behavior, correlating with osseointegration and in vivo conditions. Modified surfaces were analyzed through several physicochemical characterization techniques. Plasma nitriding led to a more pronounced increase in surface roughness, a thicker aluminum-free layer, made up of diverse titanium nitride phases, whereas thin film deposition resulted in a single-phase pure titanium nitride layer that leveled the ridged topography. The selective adhesion of osteoblast-like cells over fibroblasts was observed in nitrided samples but not in thin film deposited films, indicating that the competitive cellular behavior is more pronounced in plasma nitrided surfaces. The obtained coatings presented an appropriate performance for its use in biomedical-aimed applications, including the possibility of a higher success rate in osseointegration of implants.

Comparative study of physicochemical properties, fatty acid composition, antioxidant and toxicological potential of Citrullus lanatus and Citrullus colocynthis seeds oils

Citrullus colocynthis (C. colocynthis) seed is a common and expensive cuisine in West African diet unlike the seeds from Citrullus lanatus (C. lanatus) that are agro waste. The oils from these seeds were comparatively profiled using their physicochemical properties, fatty acids and phenolic contents, in-vitro antioxidant capacity (DPPH, TEAC, FRAP, ORAC), inhibitions of lipid peroxidation and tyrosinase. Toxicities of the oils in mice were also assessed using plasma AST, ALT, cholesterol, HDL-C, liver, heart and kidney (GSH, GPx and SOD) and clastogenic effects in mouse bone marrow. Citrullus lanatus oil contains higher peroxides, saponification, saturated fatty acids and stearic acid (6-fold) levels, with a lower acid value when compared to C. colocynthis. Strong antioxidant capacity, inhibition of lipid peroxidation and tyrosinase by C. lanatus oil extract is proposed to be related to the presence of catechins that are absent in C. colocynthis. The in-vivo study indicated that C. lanatus induced significant increase in ALT while a significant increase of AST was observed in the C. colocynthis treated group. Citrullus lanatus induced a significantly (p<0.05) higher heart GPx activity when compared with C. colocynthis and the control groups. Both oils did not affect SOD activity and showed no clastogenic effect in mouse bone marrow (p>0.05). This work reports for the first time the presence of stearic acid and catechin in C. lanatus oil which is important for its compelling in vitro activity. High doses of both oils could evidently induce toxicological indices in mice.

Comparative studies of physicochemical properties of sweet potato (Ipomoea batatas) cookies from different variations of sweet potato

Sweet potato possesses superb nutritional values and it grows easily in a hot and humid climate. In Malaysia, sweet potatoes are commonly used in making traditional snacks and sweet desserts despite that sweet potato has greater potential for utilization in other new food products. This study was conducted to formulate cookies with partial substitution of wheat flour with orange-flesh (VitAto) or purple-fleshed (Anggun) sweet potatoes. The cookies were prepared from the formulation blend of sweet potato and wheat flour in the substitution levels of 20% and 40%, respectively. The proximate composition and physicochemical properties of the formulated cookies were studied. The proximate analysis results depicted that, compared to the control cookies, sweet potato incorporated cookies had higher moisture, ash, and carbohydrates contents but lower crude protein and crude fat. The cookies incorporated with sweet potato presented a significantly greater spread ratio and diameter than the control. The results of texture profile analysis indicated that partial substitution of sweet potato for wheat flour significantly decreased the hardness of cookies though it had zero effect on the attribute of springiness. These findings revealed that the sweet potato has positive potential uses in the development of cookies or other bakery products with enhanced nutritional quality.

Sliced raw jackfruit was dried using Tray drier (TD) and Freeze drier (FD).The comparison of effect of these drying methods on the physicochemical properties were investigated. Different properties like proximate, color, pH, water absorption capacity, oil absorption capacity, water activity, bulk density were analysed. The result outcomes of the flour dried under two different driers were compared and discussed. The moisture content of TD and FD raw jackfruit flour were not significantly different (p>0.05).The proximate content of the FD flour was higher than TD flour as much nutrients are retained in freeze drying process. The color of FD flour was lighter whereas TD flour had slight darker colour which affected the acceptability of the flour. The pH, water activity, water absorption capacity as well as oil absorption capacity values of TD and FD flour had slight variations. Freeze dried raw jackfruit flour shown better quality and nutritional retention than TD flour.

Comparative study of physicochemical properties of bio-hydroxyapatite with commercial samples

This article focused on the comparative study of the physicochemical properties of bovine and human bio hydroxyapatites with commercial samples used in dental implants. Raw and defatted bovine bone was calcined at 560 °C, while commercial samples were analyzed without any processing (RTR, Novabone®, and Nukbone® granules). Transmission Electron Microscopy and Scanning Electron Microscopy were used to study their crystal size and morphology. Thermogravimetric analysis was used to study the decomposition and transformation of the organic and inorganic phases. The crystalline phases and Full Width at Half Maximum were determined by X-ray. Infrared and Raman spectroscopies were used to determine the presence of organic materials. Results showed the presence of organic material in some commercial samples, which means that these biografts could have immunological reactions with the patient.
