Exploring Deep Eutectic Solvents as Pharmaceutical Excipients: Enhancing the Solubility of Ibuprofen and Mefenamic Acid

Objectives: The study explores the potential of various deep eutectic solvents (DESs) to serve as drug delivery systems and pharmaceutical excipients. The research focuses on two primary objectives: evaluating the ability of the selected DES systems to enhance the solubility of two poorly water-soluble model drugs (IBU and MFA), and evaluating their physicochemical properties, including density, viscosity, flow behavior, surface tension, thermal stability, and water dilution effects, to determine their suitability for pharmaceutical applications. Methods: A range of DES systems containing pharmaceutically acceptable constituents was explored, encompassing organic acid-based, sugar- and sugar alcohol-based, and hydrophobic systems, as well as menthol (MNT)-based DES systems with common pharmaceutical excipients. MNT-based DESs exhibited the most significant solubility enhancements. Results: IBU solubility reached 379.69 mg/g in MNT: PEG 400 (1:1) and 356.3 mg/g in MNT:oleic acid (1:1), while MFA solubility peaked at 17.07 mg/g in MNT:Miglyol 812®N (1:1). In contrast, solubility in hydrophilic DES systems was significantly lower, with choline chloride: glycerol (1:2) and arginine: glycolic acid (1:8) showing the best results. While demonstrating lower solubility compared to the MNT-based systems, sugar-based DESs exhibited increased tunability via water and glycerol addition both in terms of solubility and physicochemical properties, such as viscosity and surface tension. Conclusions: Our study introduces novel DES systems, expanding the repertoire of pharmaceutically acceptable DES formulations and opening new avenues for the rational design of tailored solvent systems to overcome solubility challenges and enhance drug delivery.

Material compatibility and processing challenges in droplet deposition modelling additive manufacturing: A study on pharmaceutical excipients Polyvinylpyrrolidone/vinyl acetate (PVP/VA) and Polycaprolactone (PCL)

Additive manufacturing (AM) enables the production of complex, lightweight, and customized components with superior quality. Selecting the right materials considering their thermal properties, printability, and layer adhesion is crucial in melting-based AM techniques. This study investigates Droplet Deposition Modelling (DDM), an innovative material extrusion process that utilizes thermoplastic granules. DDM is distinguished by its shorter manufacturing times and a wider range of materials, setting it apart from traditional material extrusion methods such as fused filament fabrication. We investigated the printability and part quality in DDM using two common pharmaceutical excipients: Polyvinylpyrrolidone/vinyl acetate 6:4 (PVP/VA), which is highly brittle, and Polycaprolactone (PCL), known for its low solubility and role in controlled drug release. Different ratios of PVP/VA and PCL were compounded via hot melt extrusion (HME) and used in DDM to study the impact of ingredient content on printability and part quality, employing geometrical models to assess material compatibility and printability. The study revealed that increasing PVP/VA content leads to higher viscosity, reduced flowability, and uneven deposition, with formulations of 80 % and 100 % PVP/VA showing poor processability. In contrast, formulations with 60 % and 40 % PVP/VA exhibited smooth processing and compatibility with DDM. We identified processing temperature and Drop Aspect Ratio (DAR) as key factors influencing material printability and part quality. Elevated processing temperatures and reduced DAR were found to increase interface temperatures, reduce diffusion, and potentially cause the 'elephant feet' issue. Additionally, smaller droplet sizes and material characteristics, such as higher interfacial tension in PCL, could lead to coalescence. Our findings highlight the complexities in optimizing DDM processing parameters and material blends, underscoring the need for careful formulation design to achieve high-quality 3D printed products.

The effect of glidant on the tabletting behavior of common pharmaceutical excipients

Glidant used for the purpose of powder flowability enhancement is well known within the pharmaceutical industry to improve tablet manufacturing. Despite the widespread use of glidant for this purpose, the effect of glidant on the effect of tableting behavior is not well studied. To address this deficiency, the effect on glidant on the tabletting behavior of seven common excipients was investigated. Tabletability, compressibility, compactability, and tablet expansion were studied. It was shown that glidant increased the tabletability for excipients known to exhibit plastic behavior. Based on compressibility and compactability, it was inferred that an increase in total bonding strength due to the presence of glidant was responsible for the improvement in tabletability. Results suggest this may be due to an increase in inter-particle bonding area. Conversely, glidants did not affect the tabletability of brittle excipients. The effect of glidant on die-fill bulk density was also studied to examine the impact associated with air entrapment and tablet expansion. While glidant significantly increased the die-fill powder density, this did not have an observable effect on tablet expansion or any other tabletting behavior examined.

Methods Development and Validation for the Estimation of Pioglitazone HCl in Bulk and Formulations by UV Spectroscopy and FTIR

The use of spectroscopic analysis, particularly UV spectrophotometer, is a simple and essential technique for bulk drug estimation, formulation studies, and compatibility assessments of drugs with various excipients. In the pharmaceutical industry, various analytical instruments, including Fourier transform infrared spectroscopy (FTIR), are employed for investigating drug-excipient interactions that can impact the stability of active pharmaceutical ingredients. This study aimed to develop a UV spectrophotometric method for the analysis of Pioglitazone hydrochloride in phosphate buffer (pH 7.4) and methanolic solution, assessing its linearity and compliance with Beer's Law. Furthermore, we aimed to use FTIR to characterize potential interactions between Pioglitazone and common pharmaceutical excipients, such as Guar Gum, Chitosan, and Sodium Alginate. Standard solutions of Pioglitazone were prepared in phosphate buffer (pH 7.4) and methanol. UV spectrophotometer was conducted to determine the maximum absorption wavelength. Calibration curves were constructed to evaluate linearity and adherence to Beer's Law. FTIR analyses were performed to investigate drug-excipient interactions by examining the functional groups. In phosphate buffer (pH 7.4), the maximum absorption wavelength for Pioglitazone hydrochloride was 268 nm. The calibration curve for Pioglitazone in phosphate buffer (pH 7.4) demonstrated linearity in the concentration range of 1–20 µg/ml, with a correlation coefficient of 0.998. In methanol, the maximum absorption wavelength for Pioglitazone hydrochloride was found to be 272 nm. The calibration curve in methanol exhibited linearity in the range of 1–20 µg/ml, with a correlation coefficient of 0.999. FTIR analysis revealed potential drug-excipient interactions, particularly in the case of Guar Gum, Chitosan, and Sodium Alginate, suggesting the formation of stable hydrogen bonds. The developed UV spectrophotometric method for Pioglitazone analysis is a reliable, cost-effective, and reproducible approach, making it a valuable tool for drug development and quality control. Additionally, the FTIR characterization confirmed interactions between Pioglitazone and common pharmaceutical excipients, enhancing our understanding of formulation compatibility.

Amino acids induced modulations in linear PEO‐PPO‐PEO triblock copolymer microstructures and its exploration in prototypical drug solubilization

AbstractThis manuscript explains the effect of biologically important additives on the micellar behavior of an ethylene oxide‐propylene oxide (PEO101‐PPO56‐PEO101) based linear block copolymer, Poloxamer 407.The effect of common pharmaceutical excipients and three vital amino acids viz. alanine, glycine, and lysine (Ala, Gly, and Lys) is examined with the help of cloud point (CP), dynamic light scattering (DLS), and small angle neutron scattering (SANS) techniques. The drug solubilization ability for the copolymer was checked for two model drugs Quercetin (QCT) and Ibuprofen (IBU) with the help of high‐performance liquid chromatography (HPLC) in the presence/absence of additives such as Ala, Gly, and Lys. The observed results reveal that higher amino acid concentrations, favor the solubilization of drug which can potentially benefit the use of these biocompatible copolymers in pharmaceutical and biopharma industries.

The validity of using bare graphite electrode for the voltammetric determination of paracetamol and caffeine

The validity of applying the readily available bare spectroscopic graphite electrode in the pharmaceutical analysis as a cost-effective, simple, and efficient alternative to the modified carbon-based electrodes was evaluated in this work using paracetamol and caffeine as test analytes. The cyclic voltammetry study on their electrochemical behavior revealed that paracetamol and caffeine display well-expressed oxidation peaks at potentials of typically 0.51 V and 1.36 V vs. Ag, AgCl/KClsat, correspondingly and that their oxidation is an adsorption-controlled process. Then the bare spectroscopic graphite electrode was applied for the separate and individual determination of the test analytes by differential pulse adsorptive stripping voltammetry. Under optimized experimental conditions (3 min accumulation time, pH 5.0) the electrode exhibited a linear response to paracetamol in the range from 5 μmol L−1 to 150 μmol L−1 and to caffeine in the range from 5 μmol L−1 to 60 μmol L−1. The detection limit achieved (0.2 μmol L−1 for each of the analytes) was found to be sufficient to carry out paracetamol and/or caffeine analysis in drugs. The results were validated by separate and individual paracetamol and caffeine determinations in commercial tablets with satisfactory recovery values (98.90–101.85 %). The presence of common pharmaceutical excipients did not affect the paracetamol and/or caffeine voltammetric response.

Spectrophotometric Methods for Determination of Clopidogrel in Bulk and in Pharmaceutical Formulations

The present article reports three simple, sensitive, and accurate spectrophotometric methods which have been progressed for monitoring of clopidogrel bisulphate (CG) in bulk and in pharmaceutical formulations. The three methods are depended on the production of yellow ion-pair complexes between the investigated drug and bromophenol blue (BPB), bromthymol blue (BTB), and bromocresol purple (BCP) as dyes in a universal buffer of pH range (2.0- 4.0). The chloroform was used for formed complexes extraction and measured at 421 nm, 414 nm, and 409 nm, respectively for the three systems. The best conditions of the reactions were tested and optimized. Beer’s law was obeyed in the concentration ranges 2.0 – 18.0 µg mL-1 with molar absorptivity of 4.4 × 104 , 6.4 × 104 and 6.9 × 104 L mol-1cm-1, for the BPB, BTB and BCP methods, respectively. Sandell’s sensitivity, correlation coefficient, quantification and detection limits are also studied. The investigated methods have been achieved successfully for the determination of the drug in pure form and in pharmaceutical formulations. Interference between common pharmaceutical excipients and additives was disappeared. Statistical comparison of the results with those observed by the HPLC method exhibits excellent agreement and shows no significant difference in terms of accuracy and precision.

A Zirconia‐Nanoparticles‐Modified Carbon Paste Electrode for Voltammetric Determination of Ibuprofen

AbstractZirconia‐modified carbon paste electrode (ZrO2/CPE) was prepared, characterized, and applied for efficient ibuprofen quantification by differential pulse adsorptive stripping voltammetry. The increased surface area of the modified electrode (up to 74 %), combined with the excellent zirconia adsorption performance allowed a 36 % signal increase to be achieved compared to bare CPE. Under optimized experimental conditions (69 : 1 w % graphite to ZrO2 ratio, 5 min accumulation time, pH 4.5) the ZrO2/CPE exhibited a linear response to ibuprofen in the range from 20 μmol L−1 to 250 μmol L−1. The LOD of 4 μmol L−1 (S/N=3) attained was found to be sufficient to perform ibuprofen analysis in pharmaceutical formulations. The results were validated by ibuprofen determination in commercial tablets with satisfactory recovery values (98.8–102 %). The presence of common pharmaceutical excipients did not affect the ibuprofen voltammetric response. The proposed sensor also demonstrated good reproducibility (RSD=2.84 %) and long term stability.

Relative Humidity Cycling: Implications on the Stability of Moisture-Sensitive Drugs in Solid Pharmaceutical Products

The stability of a moisture-sensitive drug in tablet formulations depends particularly on the environment's relative humidity (RH) and the products' prior exposure to moisture. This study was designed to understand drug stability in relation to the moisture interaction of the excipients, moisture history of the tablets, and RH of the environment. The stability study was performed on tablets containing acetylsalicylic acid (ASA), formulated with common pharmaceutical excipients like native maize starch, microcrystalline cellulose (MCC), partially pregelatinized maize starch (PGS), dicalcium phosphate dihydrate (DCP), lactose, and mannitol. The tablets were subjected to storage conditions with RH cycling alternating between 53% and 75%. Results were also compared to tablets stored at a constant RH of 53% or 75%. The excipients demonstrated marked differences in their interactions with moisture. They could be broadly grouped as excipients with RH-dependent moisture content (native maize starch, MCC, and PGS) and RH-independent moisture content (DCP, lactose, and mannitol). As each excipient interacted differently with moisture, degradation of ASA in the tablets depended on the excipients' ability to modulate the moisture availability for degradation. The lowest ASA degradation was observed in tablets formulated with low moisture content water-soluble excipients, such as lactose and mannitol. The impact of RH cycling on ASA stability was apparent in tablets containing native maize starch, MCC, PGS, or DCP. These findings suggested that the choice of excipients influences the effect of moisture history on drug stability. The results from studies investigating moisture interaction of excipients and drug stability are valuable to understanding the inter-relationship between excipients, moisture history, and drug stability.

A critical examination of three-point bending for determining Young’s modulus

The three point bending method is commonly used for determining Young’s modulus, E. However, the literature E values of some common pharmaceutical excipients, determined by three point bending, exhibit orders of magnitude variability that cannot be fully explained by normal variations in material properties. To understand the root cause for the large variations, we assessed several potential experimental factors, including instrument stiffness, data analysis method, test speed, alignment between loading beam and specimen surface, and ratio of sample thickness to distance between two supports. Our results show that E value is sensitive to 1) deformation of instrument under force, 2) force–displacement data analysis method, 3) misalignment between loading beam and specimen surface, 4) flashing at the edge of a specimen, 5) accuracy of the true density. Finally, strategies to improve the accuracy of E determination by the three point bending method were recommended.

Utility of 2,2׳- Bipyridyl Reagent in Spectrophotometric Assay of Acetylcysteine in Pure Form and in Its Pharmaceutical Preparations

This study includes development of a sensitive spectrophotometric method for the determination of acetylcysteine in aqueous solution. The method is based on the oxidation of acetylcysteine with a ferric ion, followed by reacting of produced ferrous ion with 2,2׳-bipyridyl to form a pink complex , which is stable and water-soluble and has the highest absorption at a wavelength of 524 nm. The limits of Beer's law were in the concentration range of (5-180) µg of acetylcysteine in a final volume of 20 milliliters (0.25-9.0 µg.ml-1) and the molar absorptivity of 8633.28 L. mol-1 .cm-1 , Sandell's sensitivity index of 0.0189 µg.cm-2. The relative error of -4.28% – 4.98 % and relative standard deviation of ± 0.56% - ± 3.75% depending on the concentration level. The limit of detection limit(LOD) and limit of quantitation (LOQ) were calculated and equal to 0.01837 and 0.06124 µg/ml respectively, no inference was observed in the common pharmaceutical excipients. The results obtain by proposed method were in good agreement with those obtain from official British pharmacopoeia using t-test (at 95% confidence limit) which indicates that there is no significant differences between them. The method was successfully applied to determine acetylcysteine in its pharmaceutical preparations.

Compressibility analysis as robust in-die compression analysis for describing tableting behaviour

Abstract Objectives Compressibility of materials is determining the tableting behaviour and tablet properties during and after manufacturing. The compressibility constant has not been investigated as an in-die method yet and was therefore compared with the Heckel analysis for describing the compressibility of materials during tableting in the present study. Methods Various parameters influencing tableting, such as compression pressure, compression speed and punch diameter, were used to analyse the robustness of both methods and to evaluate the informative value of the compression parameters. Twelve common pharmaceutical excipients were used to cover a wide range of material properties. Key findings The compressibility constant was successfully applied as an in-die method and proved to be more robust against the influencing parameters during tableting than the Heckel analysis. A good correlation between the out-of-die and in-die method was found for both methods but could not be observed between the compressibility constant and the yield pressure from Heckel analysis. The methods are both describing the volume reduction of materials under pressure but focus on different material properties. Conclusions For material characterisation in the field of tableting, the compressibility constant can be additionally applied in the future to be able to determine another compressibility parameter in combination with the yield pressure.

Development and validation of molnupiravir assessment in bulk powder and pharmaceutical formulation by the RP-HPLC-UV method.

An accurate, sensitive and selective RP-HPLC-UV method has been established for the estimation of Molnupiravir (MOL) in pure bulk powder and pharmaceutical formulation. Separation was achieved on an Inertsil C18 column (150.0 mm × 4.6 mm, 5.0 μm), using a mobile phase of 20 mM phosphate buffer pH 2.5 : acetonitrile (80 : 20, v/v%) in isocratic mode with a flow rate of 1.0 mL min-1. The λ max of MOL prepared in the chosen diluent (ethanol : water in equal proportions) was found to be 230.0 nm. The constructed calibration curve was found to be linear in the concentration range of 0.2-80.0 μg mL-1. The recovery% of MOL using the proposed method was 100.29%. The limit of detection (LOD) and limit of quantification (LOQ) were 0.04 μg mL-1 and 0.12 μg mL-1, respectively. No significant interference was detected in the presence of the common pharmaceutical formulation excipients. The method was validated following the ICH recommendations. All the obtained results were statistically compared with those using reported methods and there were no significant differences. The method developed in this work was successfully employed for the assessment of MOL in bulk powder and pharmaceutical formulation.

How can single particle compression and nanoindentation contribute to the understanding of pharmaceutical powder compression?

The deformation behaviour of a powder and, thus, of the individual particles is a crucial parameter in powder compaction and affects powder compressibility and compactibility. The classical approach for the characterization of the deformation behaviour is the performance of powder compression experiments combined with the application of mathematical models, such as the Heckel-Model, for the derivation of characteristic compression parameters. However, the correlation of these parameters with the deformation behaviour is physically often not well understood. Single particle compression and nanoindentation enables the in-depth investigation of the deformation behaviour of particulate materials.In this study, single particle compression experiments were performed for the characterization of the deformation behaviour of common pharmaceutical excipients and active pharmaceutical ingredients (APIs) with various, irregular particle morphologies of industrial relevance and the findings are compared with the results from powder compression. The technique was found useful for the characterization and clarification of the qualitative deformation behaviour. However, the derivation of a quantitative functional relationship between single particle deformation behavior and powder compression is limited. Nanoindentation was performed as complementary technique for the characterization of the micromechanical behavior of the APIs. A linear relationship between median indentation hardness and material densification strength as characteristic parameter derived by in-die powder compression analysis is found.

Subcutaneous Injection Site Pain of Formulation Matrices

The objective of this work was to systematically evaluate the effects of formulation composition on subcutaneous injection site pain (ISP) using matrices comprising of common pharmaceutical excipients. Two randomized, blinded, crossover studies in healthy subjects were conducted at a single site, where subjects received 1mL SC injections of the buffer matrices. ISP intensity was measured using a 100mm visual analogue scale (VAS), which was then analyzed via heatmap, categorical grouping, subgroup analysis, and paired delta analysis. Buffer type, buffer concentration and tonicity agent showed a substantial impact on ISP. Citrate buffer demonstrated a higher ISP than acetate buffer or saline). The 20mM citrate buffer was more painful than 10 or 5mM citrate buffers. NaCl and propylene glycol were significantly more painful than sugar alcohols (mannitol, sucrose, trehalose or glycerol). Histidine buffers exhibited ISP in the descending order of 150mM > 75mM > 25mM > 0mM NaCl, while histidine buffers containing Arginine-HCl at 0, 50, or 150mM all showed very low ISP. Histidine buffer at pH6.5 showed a lower ISP than pH5.7. This systematic study via orthogonal analyses demonstrated that subcutaneous ISP is significantly influenced by solution composition.

Sulbactam pivoxil powder attributes and compatibility study with excipients

BackgroundSulbactam pivoxil is an irreversible β-lactamase inhibitor that can be used with β-lactam antibiotics to improve antibacterial therapy by the oral route. Relevant properties of this drug for pharmaceutical manufacturing are not available in the open literature. In this work, a solid-state characterization of sulbactam pivoxil at the molecular, particle, and bulk levels was performed.ResultsParticles exhibited a mean diameter of about 350 μm, irregular shape crystals, and good flow properties. This work presents for the first time the crystal structure of this β-lactamase inhibitor obtained by X-ray diffraction analysis. Fourier-transform infrared results showed the characteristic bands of aliphatic hydrocarbons and ester groups. The differential scanning calorimetry curve exhibited a sharp endothermic peak at 109 °C corresponding to sulbactam pivoxil melting. The thermogravimetric curve revealed a mass loss at 184 °C associated with a decomposition process. This powder showed a moisture content of 0.34% and a water activity of 0.463. Potential interactions between sulbactam pivoxil and common pharmaceutical excipients were evaluated by thermal analysis. The endothermic peak and the enthalpies of melting were preserved in almost all the analyzed mixtures.ConclusionThe powder was constituted by micro-sized crystals of sulbactam pivoxil that had suitable physicochemical properties for processing in controlled humidity environments. Thermal analyses suggested that sulbactam pivoxil is compatible with most of the evaluated excipients. The information obtained in the present study is relevant for the development, manufacturing, and storage of formulations that include sulbactam pivoxil.

Experimental investigation of wet pharmaceutical granulation using in-situ synchrotron X-ray imaging

Observing and measuring wet granulation processes of pharmaceutical powders is challenging with conventional experimental methods due to their opacity and the complex interaction that happens in a short time. In this study, synchrotron in-situ imaging technique was employed to capture the dynamic granulation process with a single drop impacting method. Two common pharmaceutical excipients and an active pharmaceutical ingredient (API) were used as the powder beds. Three liquids were used as binders. A new parameter, overall movement ratio, rm, is introduced to quantitatively describe the movement of the liquid binders in the powder beds. The dynamic interactions between the liquid binder and solid powders were captured via high temporal and spatial resolution X-ray images. Results show that particles with smaller sizes have the tunneling mechanism, and the spreading mechanism occurs with larger particles, as expected. Liquid binder properties including viscosity and contact angle, and operation parameters including droplet release height and volume show significant impacts on the dynamic granulation process. This study demonstrates the potential of using in-situ synchrotron X-ray imaging to quantitatively study the dynamic wet granulation process.

Inner filter effect as a sensitive sensing platform for detection of nitrofurantoin using luminescent drug-based carbon nanodots.

In the present paper, a sensitive and selective inner filter effect sensing platform was designed to detect nitrofurantoin (NIT) in pharmaceutical dosage form. Nitrogen and phosphorus co-doped carbon nanodots (CNDs) prepared via solvothermal treatment of folic acid and phosphoric acid. The prepared CNDs exhibit greenish fluorescence at 470nm when excited at 340nm with fluorescence quantum yield up to 40%. The CNDs exhibit high stability in various pH, temperature, and ionic strength which adds valuable merits to its pharmaceutical applications. The emission is quenched in the presence of absorber (here NIT) while the fluorophores were not quench by the presence of common pharmaceutical excipients. A fluorometric assay was fabricated to determine NIT in capsules by quenching of the CNDs. The linear response for the proposed method was from 5.0μM to 90μM with the detection limit being 1.4μM. To validate the method, the recovery of NIT in spiked sample was measured which was 96.6% -103.3%. The method was applied to the determination of NIT in pharmaceutical capsule samples, with comparable results of a reference method stated by the British Pharmacopeia (BP). Furthermore, the sub-acute toxicity studies of CNDs were investigated using normal male Balb/c mice forcefully drunk with 3 different dosages of CNDs. Animals did not produce treatment related signs of toxicity or mortality in any of the animals tested during the 28days observation period. Additionally, no significant (P>0.05) changes in the body weight, haematological and biochemical parameters compared with the control group were not revealed. Similarly, histopathological examination of the internal vital organs did not show any morphological alterations.

Manipulation of Pharmaceutical Polymorphic Transformation Process Using Excipients.

In the pharmaceutical field, it is vital to ensure a consistent product containing a single solid-state form of the active pharmaceutical ingredient (API) in the drug product. However, some APIs are suffering from the risk of transformation of their target forms during processing, formulation and storage. The purpose of this review is to summarize the relevant category of excipients and demonstrate the availability and importance of using excipients as a key strategy to manipulate pharmaceutical polymorphic transformation. The excipient effects on solvent-mediated phase transformations, solid-state transitions and amorphous crystallization are significant. Common pharmaceutical excipients including amino acids and derivatives, surfactants, and various polymers and their different manipulation effects were summarized and discussed. Appropriate use of excipients plays a role in manipulating polymorphic transformation process of corresponding APIs, with a promising application of guaranteeing the stability and effectiveness of drug dosage forms.

Compatibility studies with pharmaceutical excipients for aripiprazole–heptakis (2,6-di-O-methyl)-β-cyclodextrin supramolecular adduct

Aripiprazole (ARP) is one of the newest antipsychotic drugs, exhibiting very low aqueous solubility and high lipophilicity. Considering the necessity of improvement of ARP physicochemical properties and its biopharmaceutical profile, cyclodextrin complexation of the drug substance was performed. As selected cyclodextrin, a functionalized β-cyclodextrin was used, namely heptakis(2,6-di-O-methyl)-β-cyclodextrin (DIMEB), and the supramolecular adduct ARP/DIMEB was prepared by kneading technique and characterized using thermoanalytical tools (TG—thermogravimetry/DTG—derivative thermogravimetry/HF—heat flow), powder X-ray diffractometry patterns (PXRD), universal-attenuated total reflectance Fourier transform infrared (UATR-FTIR) and UV (ultraviolet) spectroscopy and, as well, saturation solubility studies. Job’s method was used for the stoichiometry of APR/DIMEB inclusion complex determination, which was found to be 1:2. Molecular modeling studies were complementary realized as to get a view over the way that ARP is hosted inside the cyclodextrin. The compatibility between the inclusion complex and some common pharmaceutical excipients, namely starch, magnesium stearate, lactose monohydrate, microcrystalline cellulose and methylcellulose, has been evaluated by means of thermal methods of analysis (TG/DTG/HF), UATR-FTIR spectroscopy and PXRD pattern. The preformulation data regarding the compatibility of ARP/DIMEB complex with selected excipients suggested that under ambient conditions, chemical interactions are observed solely between ARP/DIMEB inclusion complex and magnesium stearate, as indicated by the UATR-FTIR spectroscopy. The incompatibility in the system ARP/DIMEB + MgS is also confirmed by the PXRD study, this second investigational technique revealing also the advanced amorphization of components during mixing of complex with starch, microcrystalline cellulose and methylcellulose, but without indicating interactions. Later on, under thermal stress, thermally induced interactions occur between the components in the systems containing magnesium stearate and methylcellulose, while starch, microcrystalline cellulose and lactose can be safely used as excipients in developing solid formulations containing ARP/DIMEB inclusion complex as active pharmaceutical ingredient.
