Deciphering Acclimation to Sublethal Combined and Sequential Abiotic Stresses in Arabidopsis thaliana.

Plants are frequently exposed to environmental challenges. Responses to sub-lethal abiotic stress combinations are complex and often distinct from responses to individual stresses and remain poorly understood. Investigating traits and molecular factors mediating acclimation to stress combinations is essential for the development of climate change-resilient field crops. Here, we studied the morphological, physiological, and molecular responses of Arabidopsis thaliana to i) co-occurring high temperature and drought and ii) flooding followed by drought, both of which have increased in frequency due to climate change, and the individual component stresses: high temperature, drought and flooding. A set of 15 physiological and morphological traits were assessed during single and combined stresses. By combining these comprehensive trait analyses with transcriptome characterization, we established the generally additive negative effects of simultaneous or sequential stresses on plant morphology and physiology compared to the corresponding individual stresses. Although drought had a mild effect on various growth, morphological and physiological traits in both stress combinations, a unique transcriptome signature emerged upon combination with high temperature simultaneously or flooding sequentially. Molecular processes identified as important for multi-stress resilience included plastid-nucleus communication, ABA signaling and photo-acclimation. Based on the RNA-seq data, a set of 39 genes was identified as potential multi-stress response regulators. Mutants were tested to validate the contribution of these genes to plant survival and phenotypic acclimation under combined stress. We confirmed the involvement of several genes in regulating phenotypic acclimation traits. Among the identified factors were EARLY FLOWERING 6 (ELF6) and ARABIDOPSIS TÓXICOS EN LEVADURA 80 (ATL80), with substantial effects on plant growth, leaf development and plant survival (wilting) during high-temperature drought and post-submergence drought, respectively.

Maize Abiotic Stress Treatments in Controlled Environments.

Maize (Zea mays) is one of the world's most important crops, providing food for humans and livestock and serving as a bioenergy source. Climate change and the resulting abiotic stressors in the field reduce crop yields, threatening food security and the global economy. Water deficit (i.e., drought), heat, and insufficient nutrients (e.g., nitrogen and phosphorus) are major environmental stressors that affect maize yields, and impact growth and development at all stages of the plant life cycle. Understanding the biological processes underlying these responses in maize has the potential to increase yields in the face of abiotic stress. Optimizing individual or combined abiotic stress treatments in controlled environments reduces potential noise in data collection that can be present under less controlled growth conditions. Here, we describe methods and conditions for controlled abiotic stress treatments and associated controls during early vegetative growth of maize, conducted in greenhouses or growth chambers. This includes the environmental conditions, equipment, soil preparation, and intensity and duration of heat, drought, nitrogen deficiency, and phosphorous deficiency. Controlled experiments at early growth stages are informative for future in-field studies that require greater labor and inputs, saving researchers time and growing space, and thus research funds, before testing plants across later stages of development. We suggest that stress treatments be severe enough to result in a measurable phenotype, but not so severe that all plants die prior to sample collection. This protocol is designed to set important standards for replicable research in maize.

A review on abiotic stress vulnerability of wheat and its management in Terai, Nepal

Wheat being one of the most important crops of Nepal after rice and maize is prone to abiotic stresses like heat and drought stress. Climate change, increasing temperature and decreasing precipitation is found to be the major cause of drought and heat stress, especially making developing countries like Nepal to be most vulnerable to the negative impacts on productivity and sustainability. Abiotic stress imposes devastating changes on morphology, biochemistry and physiological processes of wheat. Loss in seed vigor, decrease in germination rate, poor crop stand and abnormal embryo are common impact seen due to combined Abiotic stress. Though many trial has been performed on effect of heat and drought, there is a need for trial imposing wheat to combined stress environment. This review especially deals with the impacts of Abiotic stress along with management including tolerant varieties such as Bhrikuti, NL1420, BL4669, NL1350 and NL1368 and agronomic techniques like mulching, use of early varieties, early sowing and incorporation of Biochar.

Unveiling the role of epigenetic mechanisms and redox signaling in alleviating multiple abiotic stress in plants.

Anthropogenic activities and subsequent global climate change instigate drastic crop productivity and yield changes. These changes comprise a rise in the number and severity of plant stress factors, which can arise simultaneously or sequentially. When abiotic stress factors are combined, their impact on plants is more substantial than that of a singleton stress factor. One such impact is the alteration of redox cellular homeostasis, which, in turn, can regulate downstream stress-responsive gene expression and resistance response. The epigenetic regulation of gene expression in response to varied stress factors is an interesting phenomenon, which, conversely, can be stable and heritable. The epigenetic control in plants in response to abiotic stress combinations and their interactions with cellular redox alteration is an emerging field to commemorate crop yield management under climate change. The article highlights the integration of the redox signaling pathways and epigenetic regulations as pivotal components in the complex network of plant responses against multi-combinatorial stresses across time and space. This review aims to lay the foundation for developing novel approaches to mitigate the impact of environmental stresses on crop productivity, bridging the gap between theoretical understanding and practical solutions in the face of a changing climate and anthropogenic disturbances.

Multi-Omics Exploration of ABA Involvement in Identifying Unique Molecular Markers for Single and Combined Stresses in tomato plants.

Over the past decade, our research group has found that plant responses to combined abiotic stresses are unique and cannot be inferred from studying plants exposed to individual stresses. Understanding how adaptative plant mechanisms integrate from stress perception to biochemical and physiological adjustments is a major challenge in abiotic stress signaling studies. Considering abscisic acid (ABA) as a key regulator in plant abiotic stress responses, in our study, ABA-deficient plants (flc) exposed to single or combined salinity and heat stresses were evaluated and different -omics analyses were conducted. Significant changes in biomass, photosynthesis, ions, transcripts, and metabolites occurred in mutant plants under single or combined stresses. Exogenous ABA application in flc mutants did not fully recover plant phenotypes or metabolic levels but induced cellular reprogramming with changes in specific markers. Multi-omics analysis aimed to identify ABA-dependent, ABA-independent, or stress-dependent markers in plant responses to single or combined stresses. We demonstrated that studying different -omics as a whole led to the identification of specific markers for each stress condition that were not detectable when each -omic was studied individually. This resource article provides an important and novel reference for scientists working in the field of plant abiotic stress. Future exploration of the transcriptomic, ionomic and metabolomic data presented in this study could lead to the identification of new pathways and genes associated with ABA signaling processes. These findings may be utilized to enhance crop resilience to heat waves, salinity, and their combination, contributing to addressing food security challenges in a climate change scenario.

Hypoxia represses pattern-triggered immune responses in Arabidopsis.

Biotic and abiotic stresses frequently co-occur in nature, yet relatively little is known about how plants co-ordinate the response to combined stresses. Protein degradation by the ubiquitin/proteasome system is central to the regulation of multiple independent stress response pathways in plants. The Arg/N-degron pathway is a subset of the ubiquitin/proteasome system that targets proteins based on their N termini and has been specifically implicated in the responses to biotic and abiotic stresses, including hypoxia, via accumulation of group VII ETHYLENE RESPONSE FACTOR (ERF-VII) transcription factors that orchestrate the onset of the hypoxia response program. Here, we investigated the role of the Arabidopsis (Arabidopsis thaliana) Arg/N-degron pathway in mediating the crosstalk between combined abiotic and biotic stresses using hypoxia treatments and the flg22 elicitor of pattern-triggered immunity (PTI), respectively. We uncovered a link between the plant transcriptional responses to hypoxia and flg22. Combined hypoxia and flg22 treatments showed that hypoxia represses the flg22 transcriptional program, as well as the expression of pattern recognition receptors, mitogen-activated protein kinase (MAPK) signalling and callose deposition during PTI through mechanisms that are mostly independent from the ERF-VIIs. These findings improve our understanding of the trade-offs between plant responses to combined abiotic and biotic stresses in the context of our efforts to increase crop resilience to global climate change. Our results also show that the well-known repressive effect of hypoxia on innate immunity in animals also applies to plants.

Impact of high temperature and drought stress on the microbial community in wolf spiders

High temperatures and drought present significant abiotic challenges that can limit the survival of many arthropods, including wolf spiders, which are ectothermic and play a crucial role in controlling pest populations. However, the impact of these stress factors on the microbiota of spiders remains poorly understood. In this study, we utilized 16 S rRNA gene sequencing to explore the diversity and composition of bacterial communities within Pardosa pseudoannulata under conditions of high temperature and drought stress. We found that Firmicutes, Bacteroidetes, and Proteobacteria were the predominant bacterial phyla present. Analyses of alpha diversity indicated an increase in bacterial diversity under combined stress conditions, as reflected by various diversity indices such as Ace, Chao1, Shannon, and Simpson. Furthermore, co-occurrence network analysis highlighted intricate interactions among the microbial taxa (e.g., Enterobacter, Chitinophaga, and Eubacterium), revealing the adaptive complexity of the spider's microbiome to environmental stress. Functional prediction analysis suggested that combined stress conditions might enhance key metabolic pathways, particularly those related to oxidative phosphorylation and amino acid metabolism. Using Random Forest analysis, we determined that changes in three heat shock proteins were largely attributed to variations in bacterial communities, with Firmicutes being notably influential. Collectively, this in-depth analysis offers novel insights into the responses of microbial communities within spider microbiomes to combined abiotic stresses, providing valuable information for understanding extreme climate impacts and informing ecological management strategies.

UV-B Radiation Exhibited Tissue-Specific Regulation of Isoflavone Biosynthesis in Soybean Cell Suspension Cultures.

Isoflavones, a class of substances with high biological activity, are abundant in soybeans. This study investigated isoflavone biosynthesis in soybean cell suspension cultures under UV-B radiation. UV-B radiation enhanced the transcription level and activity of key enzymes involved in isoflavone synthesis in cell suspension cultures. As a result, the isoflavone contents significantly increased by 19.80% and 91.21% in hypocotyl and cotyledon suspension cultures compared with the control, respectively. Meanwhile, a significant difference was observed in the composition of isoflavones between soybean hypocotyl and cotyledon suspension cultures. Genistin was only detected in hypocotyl suspension cultures, whereas glycitin, daidzein, and genistein accumulated in cotyledon suspension cultures. Therefore, UV-B radiation exhibited tissue-specific regulation of isoflavone biosynthesis in soybean cell suspension cultures. The combination of suspension cultures and abiotic stress provides a novel technological approach to isoflavone accumulation.

Solid humic substance enhanced spinach abiotic stress tolerance under combined drought, salinity, and daily heat stress

Spinach is a crop of economic importance in the Wintergarden region of Texas, where increasing soil salinity, irrigation water restrictions, and heat stress challenge its production. Improving abiotic stress tolerance and resource use efficiency are important goals for sustainable agricultural production. Although the effect of humic substances (HS) on abiotic stress tolerance in spinach has been evaluated under single stress, such as heat, drought, or salinity, their impact under combined abiotic stresses is unknown. We hypothesized that solid humic substances would be effective in alleviating multifactorial abiotic stress in spinach, which we examined in our first experiment, and further hypothesized that salinity stress in spinach would occur after a threshold intensity but that humic substance applications would ameliorate stress intensity past this threshold, which we examined in our second experiment. Three-week-old spinach seedlings (Spinacia oleracea L. cv. ‘Seaside’) were grown using an automatic gravimetric watering system in a climate-controlled high tunnel where daily maximum temperatures exceeded 32 °C. In a factorial experiment, we applied HS (control and 1 % v/v HS media amendment), drought (100 % and 50 % field capacity), and salinity (25 mL of RO water or 100 mM NaCl every other day) treatments. HS and HS×Salinity treatments more than doubled shoot and root FW and DW, leaf area, root length, and root surface area compared to other stresses, indicating HS improved growth and stress tolerance. Improved growth was matched with improved resource use efficiency, as HS and HS×Salinity treated plants exhibited the greatest WUE. Drought stress also increased WUE relative to the control but lowered transpiration rate responses to VPD in contrasts to HS and HS×Salinity treated plants, which increased their transpiration rates in response to increasing VPD. Salinity treatments increased sodium content (Na+) in both shoots and roots. No HS effect was observed in shoot Na+ accumulation, but HS reduced Na+ in roots under the Drought×Salinity treatment. Strong interactions between drought and salinity were observed for shoot and root Na+, Na+/K+, and shoot K+, showing that spinach accumulates Na+ and reduces K+ when drought and salinity stress are combined. HS had no apparent effect on leaf metabolites. Drought and salinity differentially impacted MDA, proline, and antioxidant content and activities, while HS showed no effect. Drought stress significantly increased MDA and proline while reducing TPC, TFC, ABTS, and FRAP. Salinity stress significantly decreased MDA, TPC, ABTS, FICA, and FRAP. In conclusion, HS improved overall spinach growth and WUE while decreasing root Na accumulation, and subsequently enhancing spinach stress tolerance.

Drought stress enhances plastid-mediated RNA interference for efficient the willow leaf beetle management

Plastid-mediated RNA interference has emerged as a promising and effective approach for pest management. By expressing high levels of double-stranded RNAs (dsRNAs) in plastid that target essential pest genes, it has been demonstrated to effectively control certain herbivorous beetles and spider mites. However, as plants are sessile organisms, they frequently experience a combination of biotic and abiotic stresses. It remains unclear whether abiotic stress, such as drought stress, influences the accumulation of dsRNAs produced in plastids and its effectiveness in controlling pests. In this study, we aimed to investigate the effects of drought stress on dsACT expression in transplastomic poplar plants and its control efficiency against the willow leaf beetle (Plagiodera versicolora). Our findings revealed that drought stress did not significantly affect the dsRNA contents in transplastomic poplar plants, but it did lead to higher mortality of insect larvae. This increased mortality may be attributed to increased levels of jasmonic acid and cysteine proteinase inhibitor induced by water deficit. These results contribute to understanding of the mechanisms linking water deficit in plants to insect performance and provide valuable insights for implementing appropriate pest control strategies under drought stress conditions.

Drought aggravates plant stress by favouring aphids and weakening indirect defense in a sugar beet tritrophic system

AbstractClimate change leads to more frequent droughts that may alter multitrophic networks in agroecosystems by changing bottom-up and top-down effects on herbivorous insects. Yet, how bottom-up effects of drought alter tritrophic interactions remains poorly understood. This study investigated two intensities of drought stress in the tritrophic system consisting of sugar beet (Beta vulgaris), an aphid (Aphis fabae), and its parasitoid (Aphidius colemani). We thoroughly investigated each trophic level, examining the performance of plants, pest insects, and parasitoids, as well as the attraction of parasitoids to herbivore-induced plant volatiles (HIPVs). Drought stress negatively affected plant growth but benefited A. fabae, leading to faster development and a higher reproduction rate. Drought-stressed plants also emitted less plant volatiles, which resulted in reduced attraction of A. colemani to aphid-infested plants. Drought indirectly affected parasitoid performance, as evidenced by lower emergence rates and production of fewer females, although mummification rates were higher on drought-stressed plants. Reduced parasitoid attraction and performance on drought-stressed plants may exert lower top-down pressure on aphid populations. Combined with increased aphid performance, this may facilitate aphid outbreaks, which could further weaken drought-stressed plants. Our findings highlight the need to study multiple trophic levels and emphasize the importance of incorporating HIPVs and parasitoid attraction when assessing combined abiotic and biotic stresses in crops.

Concordant Gene Expression and Alternative Splicing Regulation under Abiotic Stresses in Arabidopsis.

The current investigation endeavors to identify differentially expressed alternatively spliced (DAS) genes that exhibit concordant expression with splicing factors (SFs) under diverse multifactorial abiotic stress combinations in Arabidopsis seedlings. SFs serve as the post-transcriptional mechanism governing the spatiotemporal dynamics of gene expression. The different stresses encompass variations in salt concentration, heat, intensive light, and their combinations. Clusters demonstrating consistent expression profiles were surveyed to pinpoint DAS/SF gene pairs exhibiting concordant expression. Through rigorous selection criteria, which incorporate alignment with documented gene functionalities and expression patterns observed in this study, four members of the serine/arginine-rich (SR) gene family were delineated as SFs concordantly expressed with six DAS genes. These regulated SF genes encompass cactin, SR1-like, SR30, and SC35-like. The identified concordantly expressed DAS genes encode diverse proteins such as the 26.5 kDa heat shock protein, chaperone protein DnaJ, potassium channel GORK, calcium-binding EF hand family protein, DEAD-box RNA helicase, and 1-aminocyclopropane-1-carboxylate synthase 6. Among the concordantly expressed DAS/SF gene pairs, SR30/DEAD-box RNA helicase, and SC35-like/1-aminocyclopropane-1-carboxylate synthase 6 emerge as promising candidates, necessitating further examinations to ascertain whether these SFs orchestrate splicing of the respective DAS genes. This study contributes to a deeper comprehension of the varied responses of the splicing machinery to abiotic stresses. Leveraging these DAS/SF associations shows promise for elucidating avenues for augmenting breeding programs aimed at fortifying cultivated plants against heat and intensive light stresses.

A Role of Biosynthesized Zinc Oxide Nanoparticles (ZnO NPs) for Enhancing Seed Quality: A Review

Abiotic and biotic stresses impact seed growth, resulting in economic losses. Seeds are being subjected to an increasing number of biotic and abiotic stress combinations as a result of global warming and climate change, which has adverse effects on their growth and production. Drought, flood, salinity, heavy mineral contamination, cold, and heat were all found to have a negative impact on seed germination. Because of their massive surface area-to-volume ratio, nanoparticles—microscopic pieces with a nanoscale dimension ranging from 1-100 nm—have exceptional thermal conductivity, catalytic reactivity, nonlinear optical performance, and chemical durability. There are various methods for creating nanoparticles, such as chemical, physical, and biological ones. However, because dangerous chemical compounds are used as reducing agents, the chemical and physical procedures are expensive, complicated, and might be hazardous to the environment. The synthesis of nanoparticles using green approaches may be easily scaled up, and they are also cost-effective. Because of their superior qualities, greenly coordinated nanoparticles are currently preferred over traditionally delivered NPs. Green synthesis approaches are particularly appealing due to their ability to reduce nanoparticle toxicity. The use of vitamins, amino acids, and plant extracts has increased as a result. While dangerous and extremely hazardous substances are used in the chemical and physical processes that may cause environmental issues, capping and reducing agents are essential to the synthesis of nanoparticles. The capping or reducing agent used in physical and chemical processes is expensive. When used as a seed treatment, nanoparticles can enhance germination as well as the length, vigor, viability, and quality of the seedlings. This paper aims to provide an overview and evaluation of zinc oxide nanoparticles (ZnO NPs) as a potential substitute for biosynthesised nanoparticles in seed quality improvement.

Effects of arbuscular mycorrhizal fungi on carbon assimilation and ecological stoichiometry of maize under combined abiotic stresses

Abstract Arbuscular mycorrhizal fungi (AMF) enhance plant tolerance to abiotic stresses like salinity and improve crop yield. However, their effects are variable, and the underlying cause of such variation remains largely unknown. This study aimed to assess how drought modified the effect of AMF on plant resistance to high calcium-saline stress. A pot experiment was performed to examine how AMF inoculation affects the growth, photosynthetic activity, nutrient uptake and carbon (C), nitrogen (N) and phosphorus (P) stoichiometric ratio (C:N:P) of maize under high calcium stress and contrasting water conditions. The results showed that high calcium stress significantly reduced mycorrhizal colonization, biomass accumulation, C assimilation rate and C:N stoichiometric ratio in plant tissues. Besides, the adverse effects of calcium stress on photosynthesis were exacerbated under drought. AMF inoculation profoundly alleviated such reductions under drought and saline stress. However, it barely affected maize performance when subjected to calcium stress under well-watered conditions. Moreover, watering changed AMF impact on nutrient allocation in plant tissues. Under well-watered conditions, AMF stimulated P accumulation in roots and plant growth, but did not induce leaf P accumulation proportional to C and N, resulting in increased leaf C:P and N:P ratios under high calcium stress. In contrast, AMF decreased N content and the N:P ratio in leaves under drought. Overall, AMF inoculation improved maize resistance to calcium-salt stress through enhanced photosynthesis and modulation of nutrient stoichiometry, particularly under water deficit conditions. These results highlighted the regulatory role of AMF in carbon assimilation and nutrient homeostasis under compound stresses, and provide significant guidance on the improvement of crop yield in saline and arid regions.

Enhancing stress resilience in rice (Oryza sativa L.) through profiling early-stage morpho-physiological and molecular responses to multiple abiotic stress tolerance.

Under changing climatic conditions, crop plants are more adversely affected by a combination of various abiotic stresses than by a single abiotic stress. Therefore, it is essential to identify potential donors to multiple abiotic stresses for developing climate-resilient crop varieties. Hence, the present study was undertaken with 41 germplasm accessions comprising native landraces of Tamil Nadu, Prerelease lines and cultivars were screened independently for drought, salinity, and submergence at the seedling stage during Kharif and Rabi 2022-2023. Stress was imposed separately for these three abiotic stresses on 21-day-old seedlings and was maintained for 10 days. The studied genotypes showed a significant reduction in plant biomass (PB), Relative Growth Index (RGI), relative water content (RWC), leaf photosynthesis, chlorophyll fluorescence, and Chlorophyll Concentration Index (CCI) under drought followed by salinity and submergence. Stress-tolerant indices for drought, salinity, and submergence revealed significant variation for plant biomass. Furthermore, a set of 30 SSR markers linked to drought, salinity, and submergence QTLs has been used to characterize 41 rice germplasm accessions. Our analysis suggests a significantly high polymorphism, with 28 polymorphic markers having a 93.40% in 76 loci. The mean values of polymorphic information content (PIC), heterozygosity index (HI), marker index (MI), and resolving power (RP) were 0.369, 0.433, 1.140, and 2.877, respectively. Jaccard clustering grouped all the genotypes into two major and six subclusters. According to STRUCTURE analysis, all genotypes were grouped into two major clusters, which are concurrent with a very broad genetic base (K = 2). Statistically significant marker-trait associations for biomass were observed for five polymorphic markers, viz., RM211, RM212 (drought), RM10694 (salinity), RM219, and RM21 (submergence). Similarly, significant markers for relative shoot length were observed for RM551 (drought), RM10694 (salinity), and ART5 (submergence). Notably, the genotypes Mattaikar, Varigarudan samba, Arupatham samba, and APD19002 were identified as potential donors for multiple abiotic stress tolerance. Thus, identifying the genetic potential of germplasm could be useful for enhancing stress resilience in rice.

Specific ABA-independent tomato transcriptome reprogramming under abiotic stress combination.

Crops often have to face several abiotic stresses simultaneously, and under these conditions, the plant's response significantly differs from that observed under a single stress. However, up to the present, most of the molecular markers identified for increasing plant stress tolerance have been characterized under single abiotic stresses, which explains the unexpected results found when plants are tested under real field conditions. One important regulator of the plant's responses to abiotic stresses is abscisic acid (ABA). The ABA signaling system engages many stress-responsive genes, but many others do not respond to ABA treatments. Thus, the ABA-independent pathway, which is still largely unknown, involves multiple signaling pathways and important molecular components necessary for the plant's adaptation to climate change. In the present study, ABA-deficient tomato mutants (flacca, flc) were subjected to salinity, heat, or their combination. An in-depth RNA-seq analysis revealed that the combination of salinity and heat led to a strong reprogramming of the tomato transcriptome. Thus, of the 685 genes that were specifically regulated under this combination in our flc mutants, 463 genes were regulated by ABA-independent systems. Among these genes, we identified six transcription factors (TFs) that were significantly regulated, belonging to the R2R3-MYB family. A protein-protein interaction network showed that the TFs SlMYB50 and SlMYB86 were directly involved in the upregulation of the flavonol biosynthetic pathway-related genes. One of the most novel findings of the study is the identification of the involvement of some important ABA-independent TFs in the specific plant response to abiotic stress combination. Considering that ABA levels dramatically change in response to environmental factors, the study of ABA-independent genes that are specifically regulated under stress combination may provide a remarkable tool for increasing plant resilience to climate change.

'Against all floods': plant adaptation to flooding stress and combined abiotic stresses.

Current climate change brings with it a higher frequency of environmental stresses, which occur in combination rather than individually leading to massive crop losses worldwide. In addition to, for example, drought stress (low water availability), also flooding (excessive water) can threaten the plant, causing, among others, an energy crisis due to hypoxia, which is responded to by extensive transcriptional, metabolic and growth-related adaptations. While signalling during flooding is relatively well understood, at least in model plants, the molecular mechanisms of combinatorial flooding stress responses, for example, flooding simultaneously with salinity, temperature stress and heavy metal stress or sequentially with drought stress, remain elusive. This represents a significant gap in knowledge due to the fact that dually stressed plants often show unique responses at multiple levels not observed under single stress. In this review, we (i) consider possible effects of stress combinations from a theoretical point of view, (ii) summarize the current state of knowledge on signal transduction under single flooding stress, (iii) describe plant adaptation responses to flooding stress combined with four other abiotic stresses and (iv) propose molecular components of combinatorial flooding (hypoxia) stress adaptation based on their reported dual roles in multiple stresses. This way, more future emphasis may be placed on deciphering molecular mechanisms of combinatorial flooding stress adaptation, thereby potentially stimulating development of molecular tools to improve plant resilience towards multi-stress scenarios.

Identification of transcriptional modules linked to the drought response of Brassica napus during seed development and their mitigation by early biotic stress.

In order to capture the drought impacts on seed quality acquisition in Brassica napus and its potential interaction with early biotic stress, seeds of the 'Express' genotype of oilseed rape were characterized from late embryogenesis to full maturity from plants submitted to reduced watering (WS) with or without pre-occurring inoculation by the telluric pathogen Plasmodiophora brassicae (Pb + WS or Pb, respectively), and compared to control conditions (C). Drought as a single constraint led to significantly lower accumulation of lipids, higher protein content and reduced longevity of the WS-treated seeds. In contrast, when water shortage was preceded by clubroot infection, these phenotypic differences were completely abolished despite the upregulation of the drought sensor RD20. A weighted gene co-expression network of seed development in oilseed rape was generated using 72 transcriptomes from developing seeds from the four treatments and identified 33 modules. Module 29 was highly enriched in heat shock proteins and chaperones that showed a stronger upregulation in Pb + WS compared to the WS condition, pointing to a possible priming effect by the early P. brassicae infection on seed quality acquisition. Module 13 was enriched with genes encoding 12S and 2S seed storage proteins, with the latter being strongly upregulated under WS conditions. Cis-element promotor enrichment identified PEI1/TZF6, FUS3 and bZIP68 as putative regulators significantly upregulated upon WS compared to Pb + WS. Our results provide a temporal co-expression atlas of seed development in oilseed rape and will serve as a resource to characterize the plant response towards combinations of biotic and abiotic stresses.

Eco-physiology of maize crops under combined stresses.

The yield of maize (Zea mays L.) crops depends on their ability to intercept sunlight throughout the growing cycle, transform this energy into biomass and allocate it to the kernels. Abiotic stresses affect these eco-physiological determinants, reducing crop grain yield below the potential of each environment. Here we analyse the impact of combined abiotic stresses, such as water restriction and nitrogen deficiency or water restriction and elevated temperatures. Crop yield depends on the product of kernel yield per plant and the number of plants per unit soil area, but increasing plant population density imposes a crowding stress that reduces yield per plant, even within the range that maximises crop yield per unit soil area. Therefore, we also analyse the impact of abiotic stresses under different plant densities. We show that the magnitude of the detrimental effects of two combined stresses on field-grown plants can be lower, similar or higher than the sum of the individual stresses. These patterns depend on the timing and intensity of each one of the combined stresses and on the effects of one of the stresses on the status of the resource whose limitation causes the other. The analysis of the eco-physiological determinants of crop yield is useful to guide and prioritise the rapidly progressing studies aimed at understanding the molecular mechanisms underlying plant responses to combined stresses.
