Exogenous Application Effect of Indole 3-Butyric Acid and Myo-inositol on Improving Growth, Productivity and Bulb Quality of Garlic

Two successive field trials were performed at the Agricultural Research Farm of the Faculty of Agriculture, Suez Canal University during the two successive of 2018/2019 and 2019/2020. to investigate the influence of 0, 0.5 and 1 mM of indole 3-butyric acid (IBA) and 0, 1, 1.5 and 2 mM myo-inositol (MI) and their interactive as folair application on vegetative growth, bulb yield, biochemical constituents and mineral contents of garlic plants cv. Seds 40. The treatments were laid out in a split plot order with three replicates. The obtained results reported that garlic plants positively responded to the exogenous treatments of IBA and MI individually or interactively. It showed generally that application of IBA at 0.5 mM recorded the highest values of vegetative growth, yiled, quality parmemters as well as the mineral content (N, P and K). In the same regard, application of MI at 1.5 and 2 mM significantly promoted moast of studied traits compared to control and the rest of MI concentration, in both growing seasons. Furthermore, the two-factor interaction was found to be significant for all measured traits in both seasons, except for K content. Whereas, exogenous treatment of IBA at 0.5 mM along with exogenous treatment of MI at 1.5 and/or 2 mM resulted in the highest means of most of the above mentioned traits. Thus, it could be suggested that applying a combination of IBA and MI was found to be effective for enhancing vegetative growth, bulb yield and quality as well as macro-elements content.

Spray with plant growth regulators at full bloom may improve quality for storage of 'Superior Seedless' table grapes by modifying the vascular system of the bunch

The quality traits of table grape most appreciated by the consumers are berry size, bunch architecture, and freshness appearance (especially after post-harvest cold storage). Loss of bunch freshness reduces the value and selling potential for the market. Application of plant growth regulators (PGRs) may improve quality, particularly in seedless cultivars that naturally set compact bunches with small berries. PGRs as auxins and gibberellins are involved in fruit set, growth, development and ripening processes, including vascular tissues differentiation. This study aimed to determine if the combination of auxin indole-3-butyric acid (IBA) and gibberellic acid (GA3) improve quality attributes of 'Superior Seedless' bunches. Field grown vines were sprayed at full bloom, with 20 mg L−1 IBA and 20 mg L−1 IBA +10 mg L−1 GA3, to evaluate their effects on bunch quality at harvest and post-harvest (after 60 d of cold storage; 0 °C and 95 % RH). IBA + GA3 treatment produced looser bunches with larger berries as compared to Control. Total vascular bundle area (xylem and phloem) and rachis hydraulic conductivity were increased by IBA + GA3. As well, IBA + GA3 improved rachis and berries firmness, also freshness appearance after 60 d cold storage. In conclusion, a single spray of IBA + GA3 at full bloom improved quality and cold storage capacity of ‘Superior Seedless’ table grape, but it is not clear if GA3 or the combination of IBA and GA3 contributed to the observed effects.

Histomorphology and proteomics during rooting of in vitro shoots in Cariniana legalis (Lecythidaceae), a difficult-to-root endangered species from the Brazilian Atlantic Forest

Rooting is an important step of in vitro propagation that involves auxin signaling and the growth-promoting properties of phenolic compounds. We evaluated the effects of indole-3-butyric acid (IBA) and phloroglucinol (PG) on the in vitro rooting of shoots, histomorphology and proteomic profile during the root development in Cariniana legalis. Micropropagated shoots were incubated in 1/4 Murashige and Skoog culture medium supplemented with IBA (0, 50 and 100 μM) and PG (0 and 30 μM). The root development, histomorphology and proteomic profile were analyzed. IBA was necessary for root induction and the combination of IBA (50 or 100 μM) with 30 μM PG significantly increased the number of roots. The histomorphology of shoots treated with 50 μM IBA + 30 μM PG showed that rooting started at 12 days of induction, with the presence of meristematic-type cells from the callus induced on the base of shoots. The 50 μM IBA + 30 μM PG (IBA + PG) treatment affected the abundance of proteins during the rooting of shoots. The up-accumulation of proteins related to carbohydrate and amino acid metabolic processes in 12-day IBA + PG-treated shoots compared with 0-day shoots (before root induction) was associated with the rooting promoted by IBA + PG. The down-accumulation of proteins involved in secondary metabolic processes in 12-day IBA + PG-treated shoots compared with 12-day with no plant growth regulators (PGR) shoots was related to the lack of rooting in with no PGR shoots, while the increased accumulation of antioxidant proteins was related to rooting in 12-day IBA + PG-treated shoots compared with no PGR shoots. This is the first work to demonstrate the effect of IBA + PG on the proteomic profile and histomorphology during the rooting of an economically and ecologically important endangered species, which is difficult to root in vitro. Indole-3-butyric acid improved the in vitro rooting changing the proteomic profile and histomorphology in micropropagated shoot of Cariniana legalis, an endangered species from Atlantic Forest, which is difficult-to-root.

Effects of vegetation indices, auxin, and biostimulator treatments on the rooting of Rhododendron pulchrum cuttings

The objective of our study was to develop a protocol enabling the use of vegetation indices to evaluate the rooting of Rhododendron pulchrum cuttings . Six root growth parameters were recorded after those cuttings were exposed to rooting media for 47 days. Among plants with different of soil-plant analysis development (SPAD) and normalized difference vegetation index (NDVI) values, those with higher values exhibited significantly higher values for the root number, root length, and root dry weight, suggesting that reflectance indices were useful in measuring the root growth parameters of R. pulchrum cuttings. Another aim of this work was to study the effects of a plant growth regulator (PGR) and biostimulant on rooting for cutting propagation. Treatments with indole-3-butyric acid (IBA) of 2000 mg×L -1 alone and combined with 1-naphthaleneacetic acid (NAA; 2000 mg×L -1 ), salicylic acid (SA; 10 -4 M), and thiamine (TA; 800 mg L - 1 ) were applied to cuttings of R. pulchrum . The same observations on six different rooting parameters were recorded and analyzed after cuttings were exposed to rooting media for 99 days for the auxin test, and 62 days for the SA and TA tests. Compared to NAA, IBA enhanced root growth and development as determined by increases in all parameters, and therefore it was used thereafter. Successful results for the root number and root dry weight were achieved using cuttings of R. pulchrum with a combination of IBA and SA. In addition, the mix of IBA and TA resulted in higher root numbers. These combined treatments are recommended for establishing stem cuttings for raising R. pulchrum plants in the nursery.

Adverse Effects of Salt Stress on Rootability of Rosmarinus officinalis Cuttings and their Alleviation by Indole-3-Butyric Acid (IBA) and Bacillus subtilis

This study was conducted at the Horticulture Farm, Faculty of Veterinary and Agricultural Science, El-Zawia University, Libya during 2014 and 2015 seasons, to investigate the effect of indole-3-butyric acid (IBA) concentrations (0, 50 and 100 ppm) alone or in combination with Bacillus subtilis on the rootability, root and shoot growth of Rosmarinus officinalis L. cuttings under salt stress conditions (control, 1000, 2000 and 4000 ppm NaCl). The obtained data revealed that irrigation of cuttings with NaCl at 2000 and 4000 ppm resulted in a significant reduction in rooting percentage, root and vegetative growth characteristics, contents of N, P and K as well as C/N ratio in cutting tissues compared to control and the lowest salinity level used, whereas Na % and Na/K ratio were high, especially at high salinity levels. Treatment of cuttings with IBA alone or combined with B. subtilis modified and alleviated most of the harmful effects of salt stress, especially at the lower salinity level. Generally, the combination of IBA at 50 ppm with B. subtilis had a profound effect in increasing rootability, root and vegetative growth characteristics of plants compared to the control and all other treatments. The same treatment increased contents of N, P and K as well as C/N ratio, whereas reduced Na % and Na/K ratio in the rooted cutting tissues. So, it could be recommended to use this treatment for alleviation of adverse effects of irrigation water salinity on rooting and growth of R. officinalis cuttings.

High frequency plant regeneration from cotyledonary node explants of Cucumis sativus L. cultivar ‘Green Long’ via adventitious shoot organogenesis and assessment of genetic fidelity by RAPD-PCR technology

Influence of cytokinins, silver nitrate (AgNO3) and auxins on plant regeneration from cucumber was investigated. The cotyledonary node explants were cultured on MS medium augmented with various concentrations (0.5-2.5mg l-1) of 6-benzyl amino purine (BAP) and kinetin (KIN) for shoot bud induction. BAP at 1.5mg l-1 was found to be the best concentration for induction of high frequency of multiple shoots (98.4%). Interestingly, maximum percent of multiple shoot regeneration (100%) as well as number of shoot buds (54.6 shoots/culture) was recorded on MS medium containing the combination of 4.5mg l-1 AgNO3 and 1.5mg l-1 BAP. Multiple shoot bud regeneration frequency as well as the number of shoots was positively correlated with the concentrations of AgNO3. Addition of silver nitrate in the medium not only enhanced the rate of multiple shoot bud regeneration but also elongation of shoot buds was observed. The highest percent of rooting (96.2%) was noticed on a medium containing the combination of indole 3-butyric acid (IBA), 1.5mg l-1 and KIN 0.5mg l-1. Acclimatized plantlets were successfully established in the field where the survival rate observed was 72%. The RAPD profiles of in vitro regenerated plants were found to be highly monomorphic and identical banding pattern with mother plant. DNA fingerprinting results confirmed that the tissue culture plantlets were found to be true-to-type. The present study describes efficient protocol for high frequency plant regeneration via adventitious shoot organogenesis in cucumber.

Vegetative propagation of argan tree (Argania spinosa (L.) Skeels) using in vitro germinated seeds and stem cuttings

In vitro culture and rooting of stem cuttings can be used for vegetative propagation of plant material. Argan trees (Argania spinosa (L.) Skeels) are endemic to semi-desert areas of South-western Morocco, where they cover more than 8000km2 and have important socio-economic and environmental impacts. Seed germination was improved by culturing in vitro isolated kernels on Murashige and Skoog (MS) axenic medium with 1.0mg/L gibberellic acid (GA3). Newly in vitro-produced seedlings were then transferred to MS medium containing 0.5mg/L thidiazuron (TDZ) and 1.0mg/L GA3 to promote shoot development. The microcuttings produced were rooted using MS medium supplemented with two synthetic auxins, 1-naphthaleneacetic acid (NAA) and indole-3-butyric acid (IBA), at 1.0mg/L each. The rooted microcuttings were then transplanted to soil and acclimated in a greenhouse environment. In addition, hardwood stem cuttings collected from adult argan trees were rooted by using a combination of IBA and NAA.

Studies on Hormonal Effects on Rooting of Marcotting and Stem-Cuttings of Akee Apple (<i>Blighia sapida</i> K. D. Koenig)

The effect of hormone on the rooting of stem-cuttings and marcotting of akee apple was studied using a combination between Indole -3- Butyric Acid (IBA) and 1- Naphthalene Acetic Acid (NAA). Stem-cuttings from mature akee trees from Challenge, Jalala and Ganmo in Ilorin, were treated with different dilutions of the liquid hormone in the combination of 1.0% Indole-3- butyric acid + 0.5% 1-Naphthaleneacetic acid before propagating them in a non-mist propagator. Marcotting was also carried-out on trees, using the hormonal combination of different dilutions. Observations and the results obtained revealed that the hormonal combination had significant effect on the rooting of stem-cuttings and marcotting. Both marcotting and stem cuttings did not produce at the end of the experiment roots in the absence of the hormonal treatment; a particular aspect was marcotting that initials produced roots. On the other hand, both marcotting and stem cuttings produced roots with the hormonal treatments; more roots were produced using the combination of 2,000 ppm of IBA and 1,000 ppm of NAA, compared with lower concentrations of the hormone mixtures. After callus formation, 2,000 ppm of IBA and 1,000 ppm of NAA combination gave the best results within stem-cuttings. Based on the results obtained, it was concluded that the combination of IBA and NAA in appropriate concentration promoted rooting in Akee apple and therefore are highly valuable for the vegetative propagation of this species through stem cutting and marcotting.

A simple but unique protocol was developed for root production and clonal propagation of Piper longum L. (pippali), an important medicinal plant of India. Roots and shoots were induced in apical and basal petiolar halves of leaf using auxins. Average root number, root length and survival of rooted leaf cuttings were not significantly affected by type of auxin and leaf cuttings. Highest number of roots (13.40), root length (9.65 cm), rooting behaviour (91.69%) and survival of rooted cuttings (83.33%) were recorded in petiolar leaf cuttings treated with indole 3-butyric acid (IBA)/naphthalene acetic acid (NAA) (1000 ppm each). The petiolar leaf cuttings treated with IBA/NAA showed significantly higher percentage of shooting (83.33). The shoot number (2.0 per cutting) was also significantly highest in IBA/NAA treated leaf cuttings. Pippali can be regenerated via leaf-cuttings, either without hormone treatment, or for better results by using low concentrations of IBA or combination of IBA and NAA hormones. Production of planting material using leaf can substitute traditional propagules such as sucker, vine cutting, etc. Key words: Auxins, indole 3-butyric acid (IBA), medicinal plants, naphthalene acetic acid (NAA), vegetative propagation.

Thidiazuron (TDZ) induced plant regeneration from cotyledonary petiole explants of elite genotypes of Jatropha curcas: A candidate biodiesel plant

Jatropha curcas, the energy plant has attained great attention in recent years because of its biodiesel production potential and medicinal value. This makes it imperative to search for techniques for its rapid propagation. In the present investigation an efficient and reproducible method for plant regeneration through direct shoot bud induction from cotyledonary petiole explants of elite genotypes (CSMCRI-JC-1, CSMCRI-JC-2 and CSMCRI-JC-3) of J. curcas was developed. The best shoot buds induction (51.19%) and number of shoot buds (9.75) per explant was observed when in vitro petiole explants were placed horizontally on MS medium supplemented with 2.27μM TDZ after 6 weeks. The induced shoot buds were transferred to MS medium containing 10μM kinetin (Kn), 4.5μM 6-benzyl aminopurine (BAP) and 5.5μM α-naphthaleneacetic acid (NAA) for shoot proliferation. The proliferated shoots could be elongated on MS medium supplemented with different concentrations and combinations of BAP, indole-3-acetic acid (IAA), NAA and indole-3-butyric acid (IBA). MS medium with 2.25μM BAP and 8.5μM IAA was found to be best combination for shoot elongation and 3.01–3.61cm elongation was achieved after 6 weeks. However, significant differences in plant regeneration and shoot elongation were observed among the genotypes studied. Orientation (horizontal or vertical) and source (in vitro or in vivo) of explants also significantly influenced plant regeneration. The elongated shoots could be rooted on half-strength MS medium supplemented with 0.25mg/L activated charcoal and different concentrations and combinations of IBA, IAA and NAA. Half strength of MS medium with 5μM IBA, 5.7μM IAA and 11μM NAA was found to be best for rooting. The rooted plants could be established in soil with more than 90% survival rate.

Plant regeneration of non-toxic Jatropha curcas—impacts of plant growth regulators, source and type of explants

Jatropha curcas is an oil bearing species with multiple uses and considerable economic potential as a biofuel plant, however, oil and deoiled cake are toxic. A non-toxic variety of J. curcas is reported from Mexico. The present investigation explores the effects of different plant growth regulators (PGRs) viz. 6-benzyl aminopurine (BAP) or thidiazuron (TDZ) individually and in combination with indole-3-butyric acid (IBA), on regeneration from in vitro and field-grown mature leaf explants, in vitro and glasshouse-grown seedlings cotyledonary leaf explants of non-toxic J. curcas. In all the tested parameters maximum regeneration efficiency (81.07%) and the number of shoot buds per explants (20.17) was observed on 9.08 μM TDZ containing Murashige and Skoog’s (MS) medium from in vitro cotyledonary leaf explants. The regenerated shoot buds were transferred to MS medium containing 10 μM kinetin (Kn), 4.5 μM BAP and 5.5 μM α-naphthaleneacetic acid (NAA) for shoot proliferation. The proliferated shoots could be elongated on MS medium supplemented with 2.25 μM BAP and 8.5 μM IAA. Rooting was achieved when the basal cut end of elongated shoots were dipped in half strength MS liquid medium containing different concentrations and combinations of IBA, IAA and NAA for four days followed by transfer to growth regulators free half strength MS medium supplemented 0.25 mg/l activated charcoal. The rooted plants could be established in soil with more than 90% survival rate.

Plant regeneration through the direct induction of shoot buds from petiole explants of Jatropha curcas: a biofuel plant

An efficient and reproducible method for the regeneration of Jatropha curcas plants has been developed. The method employed direct induction of shoot buds from petiole explants, without the formation of an intervening callus using a Murashige and Skoog (MS) medium supplemented with different concentrations of thidiazuron (TDZ). The best induction of shoot buds (58.35%) and the number of shoot buds per explant (10.10) were observed when in vitro petiole explants were placed horizontally on MS medium supplemented with 2.27 µM TDZ after 6 weeks. The induced shoot buds were transferred to MS medium containing 10 µM kinetin (Kn), 4.5 µM 6‐benzyl aminopurine (BAP) and 5.5 µM α‐naphthaleneacetic acid (NAA) for shoot proliferation. The proliferated shoots could be elongated on MS medium supplemented with different concentrations and combinations of BAP, indole‐3‐acetic acid (IAA), NAA and indole‐3‐butyric acid (IBA). MS medium supplemented with 2.25 µM BAP and 8.5 µM IAA was found to be the best combination for shoot elongation and 3.01–3.91 cm elongation was achieved after 6 weeks. However, significant differences in plant regeneration and shoot elongation were observed among the genotypes studied. Shoot regeneration from cotyledonary leaf explants of Jatropha curcas: a biodiesel plant

The proliferated shoots could be elongated on MS medium supplemented with different concentrations and combinations of BAP, indole-3-acetic acid (IAA), NAA, and indole-3-butyric acid (IBA). MS medium with 2.25 mu M BAP and 8.5 mu M IAA was found to be the best combination for shoot elongation. However, significant differences in plant regeneration and shoot elongation were observed among the genotypes studied. Elongated shoot treated with 15 mu M IBA, 5.7 mu M IAA, and 11 mu M NAA resulted in highest percent rooting. The rooted plants could be established in soil with more than 90% survival rate. The method developed may be useful in improvement of J. curcas through genetic modification.

Effect of IBA on the Rooting Ability of Hardwood and Semi-Hardwood Cuttings of Ulmus glabra (Huds.)

Ulmus glabra (Huds.) is a hardwood species valuated for it wood in Hyrcanian forest. Information on the vegetative propagation on this species is currently few. This study was conducted to evaluate the effects of a range of Indole-3butyric acid (IBA) concentration (1000, 2000, 4000, 6000 and 8000 p.p.m) in combination with two types of media (Perlite and Perlite + peat + sand). On the rooting capacity of Ulmus glabra (Huds.) hardwood and semi-hardwood cuttings. No rooting was observed on the cuttings of Ulmus glabra (Huds.) with control treatment (no IBA) in both types of cuttings. Whereas different rooting percentage were observed on the cuttings treated with IBA combined with two; types of media. The highest rooting percentage were 21.1% for hardwood and 71% for semi-hardwood cuttings when they were treated with IBA (8000 p.p.m) + mixed media. Among the hormones does the best rooting percentage was found at the treatment of 8000 p.p.m IBA. The results indicated that the combination of IBA with mixed media is highly effective in increasing rooting capacity when compare to the perlite. There were no significantly different in root count of cuttings between 8000 and 6000 p.p.m. IBA treatment.

Enhancement of IBA stimulatory effect on rooting of olive cultivar stem cuttings

Three groups of olive cultivars were characterized as showing low, moderate or high rooting percentage after application of indole-3-butyric acid (IBA) treatment. To improve the rooting of olive cuttings, urea-phosphate (UP) and paclobutrazol (PB) were tested in combination with IBA. UP alone did not stimulate rooting of olive cuttings; however, when applied together with IBA it significantly enhanced the rooting of cultivar ‘Manzanillo’ cuttings. PB alone had a weak effect on rooting of cuttings but in combination with IBA it improved the rooting of cultivars ‘Manzanillo’ and ‘Souri’ cuttings. A triple combination of IBA, UP and PB provided the most effective treatment for the improvement of rooting percentage. IBA treatments increased the number of roots per cutting in comparison with the control, but decreased the length of the roots of cultivar ‘Barnea’. IBA plus UP or PB further increased the number of newly formed roots. However, IBA plus UP markedly increased root length. The three compound treatment did not differ from IBA plus PB regarding root number per cutting, but the roots were longer than in IBA treated and control cuttings. The survival of rooted cuttings treated with IBA was relatively low, for all cultivars tested. IBA plus UP plus PB improved the survival of the rooted olive plants compared with IBA alone. Thus UP and PB were shown to enhance the effect of IBA in stimulation of rooting and survival of olive cuttings.

Regeneration of Peach [Prunus persica (L.) Batsch] Rootstock Cultivars from Cotyledons of Mature Stored Seed

Shoots were regenerated from cotyledons of mature stored seed of three peach rootstock cultivars (`Flordaguard', `Nemared', and `Medaguard'). Shoot regeneration rates were highest when cotyledons were cultured for 3 weeks in darkness on Murashige and Skoog (MS) medium with 2.5% sucrose and a combination of IBA (1.25 or 2.5 μm) and TDZ (6.25 or 12.5 μm). Regeneration rates for `Flordaguard', `Nemared', and `Nemaguard' were as high as 60%, 33%, and 6%, respectively. Length of seed storage (1 to 3 years) did not affect regeneration rates. Seventy percent of regenerated shoots produced rooted plants. This regeneration method is rapid and simple, and stored seed can be used year-round. It may be a useful regeneration system for gene transfer in seed-propagated peach rootstocks. Chemical names used: 5 indole-3-butyric acid (IBA); thidiazuron (TDZ).

Rapid In Vitro Micropropagation of Two Different Cultivars of Phaseolus calcaratus RBL-1 and RBH-35.

A successful protocol for rapid proliferation of adventitious shoot from hypocotyl and leaf explants of two cultivars of Phaseolus calcaratus cv., RBL-1 and RBH-35 is described. A high frequency of multiple shoot buds was obtained directly from cut ends of the explants on MS medium supplemented with different (1-10 mgl-1) concentrations of auxins viz. 2, 4-dichlorophenoxyacetic acid (2, 4-D), indole-3-butyric acid (IBA) and naphthaleneacetic acid (NAA) alone and in combination with cytokinin, 6-benzyladenine (BA). Lower concentrations of auxins (0.1 and 0.5mgl-1) alone and in combination with different concentrations of BA were in general found to be the best for multiple shoot differentiation within 3-4 weeks of culture. Rooting of in vitro regenerated shoots was induced by NAA (0.1mgl-1) alone or a combination of IBA and BA (0.1+0.1mgl-1). Regeneration was much higher from hypocotyl than from leaf.
