Rapid Detection of Getah Virus Antibodies in Horses Using a Recombinant E2 Protein-Based Immunochromatographic Strip.

The prevalence and impact of Getah virus (GETV) are significant concerns in China. GETV can infect a wide range of animals, including horses, pigs, sheep, cattle, birds, and humans, resulting in substantial losses in the livestock and agricultural industries. GETV infection can cause the development of ulcers and inflammation in the mouth and gums of horses, which result in pain and discomfort and lead to symptoms such as reduced appetite, drooling, and difficulty chewing. As a result, there is a pressing need for efficient and rapid disease diagnosis methods. However, the currently available diagnostic methods have limitations in terms of operational time, equipment, and the experience of the individuals using them. In this study, a rapid, specific, and sensitive detection method was developed using a colloidal gold-based immunochromatographic strip (ICS) for the detection of antibodies against GETV in horses. To prepare the ICS, the antigen domain of the E2 glycoprotein of GETV was expressed using the Escherichia coli expression system after analysis with DNAstar v7.1 software. The nitrocellulose membrane was coated with rE2 protein or SPA to form the test line and control line, respectively. After optimizing the reaction conditions, the sensitivity, specificity, and repeatability of the strip were verified. The results showed that the test strip had a detection limit of up to 1:320 dilutions for GETV-positive serum, with no cross-reactivity observed with other equine-susceptible pathogens such as equine arteritis virus (EAV), equine herpesvirus-1 (EHV-I), equine infectious anemia virus (EIAV), equine influenza virus (EIV), African horse sickness virus (AHSV), and Japanese encephalitis virus (JEV). Furthermore, the ICS exhibited a concordance rate of 94.0% when testing 182 clinical serum samples compared to the virus neutralization test. Overall, this ICS diagnosis method will be an effective tool for the rapid detection of GETV in the field.

Immunochromatographic strip for rapid and sensitive detection of bupirimate residues in peach, orange, and carrot

Bupirimate (BPM) is a high-efficiency and low-toxicity fungicide used to combat powdery mildew in crops. To mitigate potential health risks to consumers resulting from improper BPM usage, we prepared a monoclonal antibody against BPM based on novel hapten synthesis, which has high sensitivity and strong specificity, and then successfully designed a colloidal gold-based immunochromatographic (ICG) strip. The newly designed ICG strip was then employed for detecting BPM residues in peach, orange, and carrot. The results show that for the peach, orange, and carrot samples, the calculated detection limits of the ICG strip are 9.36, 0.79, and 0.57 ng/g, respectively, and that it is resistant to the matrix effect and meets the maximum residue limit requirements of European Commission for BPM. Therefore, this developed ICG strip is expected to enable swift detection of BPM residues on the spot.

An immunochromatographic sensor for the detection of 5-fluorouracil in human plasma

This study prepared a colloidal gold-based immunochromatographic strip (CG-ICS) for the detection of 5-fluorouracil (5-FU) in human plasma, for therapeutic drug monitoring. Three haptens were synthesized with different carboxyl introductory positions and with different carbon chain lengths for the spacer arm, and were used to screen the coating antigens and immunogens. A highly sensitive and specific monoclonal antibody (mAb) 3C12 with an IC50 value of 4.359 ng/mL detected by ic-ELISA was produced for the construction of the strip. The visual limit of detection (vLOD) for semi-quantitative analysis of the CG-ICS was 50 ng/mL and the cut-off value was 2000 ng/mL. Furthermore, the calculated LOD (cLOD) value and linear range for quantitative detection were 1.464 ng/mL and 5.432–367.627 ng/mL, respectively. This detection required at most 10 min. In addition, the recovery rate of the spiked 5-FU in human plasma detected by the CG-ICS was 91.77 % to 94.04 %, which matched very well the results from LC-MS/MS. For real positive samples, the test results of the CG-ICS were consistent with those of LC-MS/MS. These findings suggest that the CG-ICS detection method is suitable for the rapid detection of 5-FU.

Development of a colloidal gold-based immunochromatographic assay for rapid detection of nasal mucosal secretory IgA against SARS-CoV-2.

Infection with SARS-CoV-2 begins in the upper respiratory tract and can trigger the production of mucosal spike-specific secretory IgA (sIgA), which provides protection against reinfection. It has been recognized that individuals with high level of nasal spike-specific IgA have a lower risk of reinfection. However, mucosal spike-specific sIgA wanes over time, and different individuals may have various level of spike-specific sIgA and descending kinetics, leading to individual differences in susceptibility to reinfection. A method for detecting spike-specific sIgA in the nasal passage would be valuable for predicting the risk of reinfection so that people at risk can have better preparedness. In this study, we describe the development of a colloidal gold-based immunochromatographic (ICT) strip for detecting SARS-CoV-2 Omicron spike-specific sIgA in nasal mucosal lining fluids (NMLFs). The ICT strip was designed to detect 0.125 μg or more spike-specific sIgA in 80 μL of NMLFs collected using a nasal swab. Purified nasal sIgA samples from individuals who recently recovered from an Omicron BA.5 infection were used to demonstrate that this ICT strip can specifically detect spike-specific sIgA. The signal levels positively correlated with neutralizing activities against XBB. Subsequent analysis revealed that people with low or undetectable levels of spike-specific sIgA in the nasal passage were more susceptible to SARS-CoV-2 reinfection. This nasal spike-specific sIgA ICT strip provides a non-invasive, rapid, and convenient method to assess the risk of reinfection for achieving precision preparedness.

Rapid detection of varicella-zoster virus based on an immunochromatographic strip

Varicella-zoster virus (VZV) is a highly infectious DNA virus that can cause varicella (chickenpox) and herpes zoster (HZ). A simple, sensitive and specific detection method is desirable for the VZV infection. In this study, VZV gE protein, expressed in CHO cells, was used to immunize BALB/c mice for the generation of monoclonal antibodies (mAbs). For the first time, we developed a colloidal gold-based immunochromatographic strip for rapid detection of VZV using a pair of mAbs against gE protein. The limit of detection (LOD) of the strip was 30 ng mL−1 of purified VZV gE antigen, and it could specifically test VZV without cross-reactivity with Enterovirus 71 (EV-71), Herpes simplex virus 1 (HSV-1) and Herpes simplex virus 2 (HSV-2). The coincidence rate between the strip and commercial real-time PCR diagnostic kit was 100% using vesicle as the clinical sample. Our strip provided a technical support for rapid and specific detection of VZV.

A gold-based immunochromatographic strip for the specific detection of tacrolimus in whole blood.

Tacrolimus is a macrolide immunosuppressant widely used in organ transplantation. Due to the narrow treatment window, therapeutic drug monitoring of the clinical application of tacrolimus is necessary. In this study, a carboxyl group introduced at the hydroxyl or carbon positions of tacrolimus was used to couple with carrier protein to synthesize complete antigens. After screening different immunogens and coating antigens, a highly-sensitive and specific monoclonal antibody (mAb) 4C5 was obtained, with a half inhibitory concentration (IC50) value of 0.26 ng mL-1 determined by indirect competitive enzyme-linked immunosorbent assay (ic-ELISA). A colloidal gold-based immunochromatographic strip (CG-ICS) was established to monitor tacrolimus in human whole blood based on the mAb 4C5. Through visual observation, it was found that the visual limit of detection (vLOD) and cut-off value of qualitative detection were 1.0 and 20.0 ng mL-1, respectively. The calculated LOD (cLOD) value of quantitative detection was 0.16 ng mL-1, and the linear range was 0.48-7.57 ng mL-1. In addition, the results of the CG-ICS for analyzing real positive samples of human whole blood were basically consistent with those of LC-MS/MS. Therefore, the CG-ICS was suitable for rapid and accurate clinical monitoring of tacrolimus.

Development of a colloidal gold immunochromatographic strip for the rapid detection of antibodies against Fasciola gigantica in buffalo

BackgroundFasciola gigantica, a tropical liver fluke, infects buffalo in Asian and African countries, causing significant economic losses and posing public health threats. The diagnostic of buffalo fascioliasis caused by F. gigantica is vital in fascioliasis control and preventation. The 22nd gel filtration chromatography fraction of F. gigantica Excretory-Secretory Products (FgESP), namely Fasciola 22 (F22), which was used as a diagnostic antigen in indirect ELISA, has demonstrated great potential for fascioliasis diagnosing. In the absence of rapid diagnostic methods, the use of a colloidal gold immunochromatographic strip based on F22 was applied to detect F. gigantica infection in buffalo.MethodsIn the present study, the 22nd gel filtration chromatography fraction of FgESP (F22) was used as an antigen to establish the colloidal gold-based immunochromatographic strip (ICS). The nitrocellulose membrane was incubated with F22 at the test line (T line) and goat anti-mouse secondary antibody at the control line (C line). The mouse anti-buffalo secondary antibody 2G7 conjugated to colloidal gold particles was used as the detection system for line visualization. The strip was assembled and developed by optimizing reaction conditions. The sensitivity, specificity, stability, and early diagnostic value of the strip were evaluated employing buffalo-derived sera.ResultsAn immunochromatographic strip for the rapid detection of antibodies against F. gigantica-FgICS was developed. The strip demonstrated high sensitivity and specificity. Sensitivity tests confirmed positive results even when the positive reference serum was diluted 4,096 times. Except for one Schistosoma japonicum-positive serum that tested positive via FgICS, specificity tests confirmed no cross-reactivity with other positive sera of Schistosoma japonicum and Babesia bovis. The strip remained stable after storage at 4°C for up to 3 months. In infected buffalo, antibodies could be detected as early as 14–21 days post-infection. The detection of 17 positive sera yielded an 82.4% positive rate via FgICS vs. a 100.0% positive rate via ELISA based on FgESP. For FgICS, the 95% confidence interval of sensitivity was 84.8–95.4%, while specificity was 4.2–14.7%.ConclusionThe immunochromatographic strip FgICS developed in this study provides a simple and rapid method of F. gigantica antibody detection and infected buffalo monitoring in the field.

An immunochromatographic test strip for on-site detection of Ralstonia solanacearum in tobacco leaves and soil

Tobacco bacterial wilt is one of the most devastating soil-borne diseases in tobacco-producing regions worldwide. It is often responsible for significant economic losses during tobacco production. A rapid, specific, and high-throughput on-site detection method is important for plant disease management. In this study, monoclonal antibody 3H3 and polyclonal antibody 0344 specific for Ralstonia solanacearum were used to prepare a colloidal gold-based immunochromatographic test strip (ITS). Under optimal conditions, the detection limit of the ITS was 105 CFU/mL. The ITS was able to detect different R. solanacearum strains collected from Shandong, Yunnan, Guizhou, and Sichuan provinces in China. Moreover, the ITS was highly specific for R. solanacearum, with no cross-reactivity with Alternaria alternata (Fries) Keissler, Pseudomonas syringae pv. angulata, and P. syringae pv. tabaci. Furthermore, R. solanacearum-spiked tobacco leaves and soil were used to evaluate the matrix interference of the developed ITS, which indicated the test strip was unaffected by leaf size or soil abundance.

A gold-based immunochromatographic strip for the detection of sirolimus in human whole blood.

Sirolimus is a potent macrolide immunosuppressant, with a long half-life, a narrow therapeutic window, and large individual variations in pharmacokinetics. In this study, an indirect enzyme-linked immunosorbent assay (ic-ELISA) and a colloidal gold-based immunochromatographic strip were developed to establish a rapid and efficient clinical screening of sirolimus. The hapten for sirolimus was synthesized by the oximation reaction and then coupled to keyhole limpet hemocyanin (KLH) and bovine serum albumin (BSA) to prepare the immunogen and coating antigen. Based on highly specific monoclonal antibodies (mAbs), the 50% inhibitory concentration (IC50) of the optimized ic-ELISA was 1.54 ng mL-1 and the limit of detection (LOD) was 0.19 ng mL-1 in whole blood samples. Good specificity was revealed by its low cross-reactivity (CR) with several analogs of sirolimus. The visual limit of detection (vLOD) and cut-off values of the strip were 20 and 100 ng mL-1 in whole blood samples, as assessed by the naked eye. Therefore, both methods could be used as potentially rapid monitoring approaches for the detection of sirolimus in human whole blood.

A colloidal gold-based immunochromatographic strip for rapid detection of SARS-CoV-2 antibodies after vaccination

Vaccination interventions is consideredan important preventive measure to block the transmission of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and protect the organism from pathogen infection effectively. However, a quick and accurate technique to evaluate the immune efficacy of the SARS-CoV-2 inactivated vaccine remains scarce. In this paper, an IgM-IgG antibody combined detection colloidal gold immunochromatography assay kit was optimized and developed, which can assess the efficacy of the inactivated SARS-CoV-2 vaccine. We collected fingertip blood samples from 3 vaccinees and 1 unvaccinated sample. The results showed that the proportion of antibody was high after the second shots immunization. The colloidal gold-based immunochromatographic strip is rapid, convenient and easy to operate. It can be used as an auxiliary method for preliminary evaluation of the antibody effect of vaccine recipients, and provide a reference index for the potential clinical application value of the vaccine.

Development of an antigen-ELISA and a colloidal gold-based immunochromatographic strip based on monoclonal antibodies for detection of avian influenza A(H5) viruses.

Avian influenza A(H5) viruses (avian IAVs) pose a major threat to the economy and public health. We developed an antigen-ELISA (ag-ELISA) and a colloidal gold-based immunochromatographic strip for the rapid detection of avian A(H5) viruses. Both detection methods displayed no cross-reactivity with other viruses (e.g., other avian IAVs, infectious bursal disease virus, Newcastle disease virus, infectious bronchitis virus, avian paramyxovirus). The ag-ELISA was sensitive down to 0.5 hemagglutinin (HA) units/100 µL of avian A(H5) viruses and 7.5 ng/mL of purified H5 HA proteins. The immunochromatographic strip was sensitive down to 1 HA unit/100 µL of avian A(H5) viruses. Both detection methods exhibited good reproducibility with CVs < 10%. For 200 random poultry samples, the sensitivity and specificity of the ag-ELISA were 92.6% and 98.8%, respectively, and for test strips were 88.9% and 98.3%, respectively. Both detection methods displayed high specificity, sensitivity, and stability, making them suitable for rapid detection and field investigation of avian A(H5) viruses.

Health Issues and Immunological Assessment Related to Wuhan's COVID-19 Survivors: A Multicenter Follow-Up Study.

Background: Currently, a large number of hospitalized coronavirus infectious disease-2019 (COVID-19) patients have met the clinical discharge criteria and have been discharged. Little is known about the sequelae and herd immunity, two important factors influencing the life quality and safety of COVID-19 survivors.Methods: Discharged COVID-19 patients from four medical facilities in Wuhan, China, were followed in order to record and investigate possible post-COVID-19 sequelae and herd immunity. After hospital discharge, patients reported to Fangcang shelter hospitals for an initial 14-day period of mandatory clinical monitoring. After release from these shelter hospitals, patients returned home for self-quarantine. Real-time quantitative PCR (RT-qPCR) was used for severe acute respiratory syndrome-related coronavirus 2 (SARS-CoV-2) detection. Colloidal gold-based immunochromatographic strip assay (ICGSA) was used for anti-SARS-CoV-2 immunoglobulin G (IgG) and immunoglobulin M (IgM) antibody testing. The data for this study are derived from case reports, medical records, and self-reports.Results: A total of 3,677 COVID-19 survivors [median age = 59 years, interquartile range (IQR) = 47–68, range = 10–98; 55.5% female] who were released from four hospitals in Wuhan, China, between January 18 and March 29, 2020 were followed for a median of 144 days (IQR = 135–157). During follow-up, 976 (26.5%) patients had at least one post-COVID-19 sequela. The incidence of post-COVID-19 sequelae among elderly COVID-19 survivors (age ≥60 years) was slightly increased compared to that of young COVID-19 survivors (age <60 years; relative risk = 1.05, 95% CI = 1.02–1.10, p = 0.007). During follow-up, a dramatic reduction of anti-SARS-CoV-2 IgG (88.0%, 95% CI = 84.2–90.4) and IgM (93.2%, 95% CI = 88.5–96.4) antibodies was observed. Among these COVID-19 survivors, 1.2% (n = 45) retested positive for SARS-CoV-2 and 1.0% (n = 37) died during follow-up. Of those who died during follow-up, 70.3% were male and all were negative for both IgG and IgM, except for one person who was IgG-positive.Conclusions: Our study documents significant post-COVID-19 sequelae that impair functions of multiple organ systems in COVID-19 survivors, suggesting that the long-term effects of this disease will negatively impact survivors' quality of life, continue to strain health care systems, and result in extended periods of lost productivity. Furthermore, female gender and anti-SARS-CoV-2 immunity may play an essential role in the survival after COVID-19 infection.

False-positive colloidal gold-based immunochromatographic strip assay reactions for antibodies to SARS-CoV-2 in patients with autoimmune diseases.

The outbreak of the novel 2019 coronavirus disease (COVID-19) was declared a global pandemic by the World Health Organization (WHO) on March 11, 2020. The diagnosis of COVID-19 is frequently based on a positive serological test. We noted the occurrence of false-positive results for COVID-19 in the colloidal gold-based immunochromatographic strip (ICS) assay in sera from patients with autoimmune diseases (ADs). This study aimed to evaluate the possible reasons for the false-positive results in two ICS assays (Wondfo ICS and Innovita ICS) and to investigate the effect of urea dissociation in reducing false-positive results. The sera of 135 patients with ADs, 13 confirmed COVID-19 patients, 95 disease controls, and 120 healthy controls were tested for immunoglobin M (IgM) and IgG against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) using Wondfo and Innovita ICS kits. The distributions of auto-antibodies in antibody-positive and antibody-negative groups were also compared, and bivariable logistic regression was used to assess auto-antibodies associated with false-positive results. A urea dissociation test of ICS was performed for the SARS-CoV-2 antibody-positive samples. Specificity of Wondfo ICS for the 95 disease controls was 94.74% compared to 98.95% and 96.84% for Innovita SARS-CoV-2 IgM and IgG, respectively. Specificity of Wondfo ICS for the 120 healthy controls was 97.5% compared to 100% and 99.17% for Innovita SARS-CoV-2 IgM and IgG, respectively. Specificity of Wondfo ICS for AD patients was 73.33% compared to 97.78% and 96.30% for Innovita SARS-CoV-2 IgM and IgG, respectively. Sensitivity was 74.07% for Wondfo compared to 70.37% for Innovita IgM and 66.67% for Innovita IgG. Using the Wondfo ICS, the percentage of elevated rheumatoid factor (RF) level (>20 IU/mL) was higher in the SARS-CoV-2 antibody-positive group compared with the antibody-negative group [27/36 (75.0%) vs. 34/99 (34.34%), P=0.001]. The elevated RF was associated with antibody positivity, with an odds ratio of 4.671 [95% confidence interval (CI), 1.88-11.69]. The specificity of the Wondfo ICS assay for the AD patients was increased from 73.33% to 94.07% after the urea dissociation assay. An elevated serum RF level could lead to false-positive results when detecting SARS-CoV-2 antibodies using the Wondfo ICS kit, and the urea dissociation assay would be helpful in reducing the incidence of false-positive results.

Development of a Colloidal Gold-Based Immunochromatographic Strip for Rapid Detection of Severe Acute Respiratory Syndrome Coronavirus 2 Spike Protein.

The outbreak and worldwide pandemic of the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) have a significant impact on global economy and human health. In order to reduce the disease spread, 16 monoclonal antibodies (McAbs) again SARS-CoV-2 were generated by immunized mice with the spike protein receptor binding domain (RBD), which was expressed in Chinese hamster ovary cell (CHO). A colloidal gold-based immunochromatographic strip was developed with two McAbs to detect SARS-CoV-2 spike protein, which can play a potential role in monitoring vaccine quality. The strip is highly specific, detecting only SARS-CoV-2 spike protein, and does not show any non-specific reactions with syndrome coronavirus (SARS-CoV), Middle East respiratory syndrome coronavirus (MERS-CoV) and other coronavirus and influenza viruses. The strip detected subunit vaccine in our laboratory with a detection limit of spike protein of 62.5 ng/mL. This strip provides an effective method in monitoring vaccine quality by detecting the antigen content of spike protein.

Rapid and sensitive pathogen detection platform based on a lanthanide-labeled immunochromatographic strip test combined with immunomagnetic separation

Sensitive point-of-care pathogen detection methods are urgently required as pathogens pose a significant threat to food safety and human health. Here, we reported a platform based on a lanthanide-labeled immunochromatographic strip test combined with immunomagnetic separation for the detection of pathogens in food and clinical samples. First, immunomagnetic separation technology was employed as a pre-treatment to eliminate the effect of background in the samples. Then, the lanthanide chelate-loaded carboxyl-modified polystyrene nanoparticles, exhibiting bright luminescence, wide Stokes shift, and excellent stability, were prepared and used to label pathogens with antibody. Finally, an immunochromatographic strip test, based on a sandwich format and supported by our custom-designed portable reader, was used to rapidly quantify the result. Using Salmonella typhimurium as a model pathogen, under optimal conditions, we achieved a detection limit of 103 CFU/mL and a linear range of 104–106 CFU/mL in milk and human serum with good recovery, reproducibility, and specificity. Compared with the existing colloidal gold-based immunochromatographic strip test, our platform showed high sensitivity (100-fold improvement) and provided quantitative determination. Our detection platform was therefore demonstrated to be highly sensitive, rapid (within 1.5 h), and cost-effective, providing a promising platform for point-of-care pathogen detection in food and clinical fields.

Novel, Highly Sensitive, and Specific Assay to Monitor Acute Myocardial Infarction (AMI) by the Determination of Cardiac Troponin I (cTnI) and Heart-Type Fatty Acid Binding Protein (H-FABP) by a Colloidal Gold-Based Immunochromatographic Test Strip

Acute myocardial infarction (AMI) is considered to be one of the predominant causes of human death; Therefore an early and accurate diagnostic method of AMI is urgently required. In this work, a novel colloidal gold-based immunochromatographic test strip was designed and fabricated for the combined determination of cardiac troponin I (cTnI) and heart-type fatty acid binding protein (H-FABP) for the diagnosis of AMI with high sensitivity and specificity. Colloidal gold particles (AuNPs) were synthesized through the reduction of gold chloride with sodium citrate, followed by conjugation with coating antibodies of cTnI or H-FABP by electrostatic interaction. The resultant conjugates were used as the instant visual signals and capture antibodies of cTnI and H-FABP were used as the test lines. Using a small sample volume (70 µL) and short detection time (10 min), the fabricated test strips exhibited excellent detection performance including a wide linear range and good accuracy and precision with no cross-reactivity. Moreover, the test strips were applied for different sample types such as serum and plasma, offering great convenience for clinical use. Additionally, the clinical samples were used to demonstrate the excellent validation of the test strips. The successfully combined determination of cTnI and H-FABP afforded the test strips with early diagnosis and enlarged the diagnosis window for AMI with other advantages such as low cost and user-friendliness. Hence, we believe that the test strips fabricated in this work have great potential to be applied for the early and accurate diagnosis of AMI in practical clinics.

Colloidal gold-based immunochromatographic strip assay for the rapid detection of diminazene in milk

ABSTRACT Diminazene (DIM) is widely used to treat trypanosomiasis in livestock, but its residues in animal products can cause food safety problems. Thus, a rapid colloidal gold-based immunochromatographic strip assay was established to analyse DIM residue in milk samples. A highly sensitive and specific monoclonal antibody (mAb) against DIM was produced with a new synthetic immunogen by the active ester method. The titre of the prepared antibody was up to 1:1.0 × 106, the affinity constant was 2.2 × 108 L mol−1 and the half-maximal inhibitory concentration (IC50) was 0.4 ng mL−1 based on enzyme-linked immunosorbent assay (ELISA). Then, the colloidal gold-labelled mAb probe was successfully prepared and used for preparing the immunochromatographic strip. The strip showed high sensitivity and specificity, the IC50 for DIM was 5.2 ng mL−1, the limit of detection was 1.2 ng mL−1 and the linear range of detection was 1.8–15.4 ng mL−1. The average recoveries ranged from 89.5% to 91.7%, and coefficients of variation were 6.6–7.2%. The immunochromatographic strip assay can be employed in the rapid detection of DIM residue in milk samples.

The Strategic Alliance between Clinical and Molecular Science in the War against SARS-CoV-2, with the Rapid-Diagnostics Test as an Indispensable Weapon for Front Line Doctors.

Our work concerns the actual problem of spread of SARS- CoV-2 outbreak which requires fast and correct as possible answer. In current scenario, the need of rapid answer put away the imperative of proper methodology. We focus on the serogical immunoassay for diagnosis of Covid-19 as an important weapon not only for diagnostic purpose, but also for epidemiologic one. The right equilibrium between high speed, low cost and accuracy is obtained with easy-to-use decentralized point-of-care test as the colloidal gold-based immunochromatographic strip assay which detects IgM and IgG antibodies directed against SARS-CoV-2. As our aim is to evaluate the efficacy of Covid-19 rapid tests and of serological assays in real-life settings, we designed a research protocol aimed to establish how to use correctly these diagnostics, taking into account the different possible clinical and epidemiological scenarios.
