Dynamic cell photo-manipulation technology for the molecular and mechanical regulation analyses of collective cell migration

Collective cell migration is an essential biological process. Migration behaviors of multiple cellular units depend on the mechanical and chemical properties of their scaffolds and the geometry of the cells. However, the mechanisms by which these properties synergistically regulate the collective cell characteristics remain unknown. A robust method is required to analyze collective cell migration. Therefore, in this study, we developed a new method for collective cell migration analysis using defined chemical, mechanical, and geometrical properties. Our method is based on a poly(acrylic acid) hydrogel, whose surface is functionalized with photocleavable poly(ethylene glycol) and a cell-adhesive peptide. By controlling the UV irradiation of the photoactivatable hydrogel, we created geometrically controlled cellular clusters and induced collective migration. Furthermore, chemical and mechanical cues exposed to cell clusters were manipulated depending on the surface density of the cell-adhesive peptide and crosslinking density of the hydrogel. As a proof of concept, we also demonstrated that the collective migration of epithelial cells was synergistically regulated by the chemical and mechanical properties of the scaffold. Our results suggest the new photoactivatable substrate as a promising tool for advanced molecular and mechanobiological analyses of collective cell migration.

Epithelial monolayers develop density and effective temperature differentials to migrate across confined matrices

Collective migration of epithelial cells across heterogeneous environments regulates vital physiological and pathological processes, such as embryogenesis, wound healing, tissue regeneration, and cancer metastasis. In response to matrix stiffness and topography, epithelial monolayers undergo jamming-unjamming transitions. Although previous studies have shown different migration phenotypes separately in confined and unconfined environment, physical transitions required for collective cell populations to negotiate confined regions remain unknown. To address this question, we fabricated PDMS substrates with contiguous wide (200 μm) and narrow (50 μm) microchannels, and studied collective migration of MCF10A (WT) epithelial cells. We found that cells develop a density differential in wide regions, relative to narrow channels, which could generate enough pressure for their eventual migration into confinement. To understand whether cell-cell cohesivity alters this requirement of density differential, we used cells with constitutively-active RhoA (+RhoA) for higher cohesivity and overexpressed ErbB2 (+ErbB2) for cancer-like reduced cohesion. We found that more cohesive cells (+RhoA) developed higher density differential relative to less cohesive cells for migration across confinement. While confined channels allowed for faster migration in WT cells, this confinement sensitivity was diminished for both alternations in cell cohesivity. With migration into confinement, cells deformed their shapes and increased their effective temperature, calculated by treating cells as a granular thermodynamic system. Further, more cohesive cells (+RhoA) and those in confined regions were effectively warmer. Overall, our results show that collective cell populations build a pressure differential between narrow and wide matrices to enter confined environments and these requirements vary for cell types of varying cohesivity. These findings show that collective cell migration across confinements can be interpreted in terms of basic physical principle of pressure and temperature of granular matter, which could inform our understanding of more complex living systems.

Actin-driven Golgi apparatus dispersal during collective migration of epithelial cells

As a sedentary epithelium turns motile during wound healing, morphogenesis, and metastasis, the Golgi apparatus moves from an apical position, above the nucleus, to a basal position. This apical-to-basal repositioning of Golgi is critical for epithelial cell migration. Yet the molecular mechanism underlying it remains elusive, although microtubules are believed to play a role. Using live-cell and super-resolution imaging, we show that at the onset of collective migration of epithelial cells, Golgi stacks get dispersed to create an unpolarized transitional structure, and surprisingly, this dispersal process depends not on microtubules but on actin cytoskeleton. Golgi-actin interaction involves Arp2/3-driven actin projections emanating from the actin cortex, and a Golgi-localized actin elongation factor, MENA. While in sedentary epithelial cells, actin projections intermittently interact with the apically located Golgi, and the frequency of this event increases before the dispersion of Golgi stacks, at the onset of cell migration. Preventing Golgi-actin interaction with MENA-mutants eliminates Golgi dispersion and reduces the persistence of cell migration. Taken together, we show a process of actin-driven Golgi dispersion that is mechanistically different from the well-known Golgi apparatus fragmentation during mitosis and is essential for collective migration of epithelial cells.

Photothermal Agarose Microfabrication Technology for Collective Cell Migration Analysis.

Agarose photothermal microfabrication technology is one of the micropatterning techniques that has the advantage of simple and flexible real-time fabrication even during the cultivation of cells. To examine the ability and limitation of the agarose microstructures, we investigated the collective epithelial cell migration behavior in two-dimensional agarose confined structures. Agarose microchannels from 10 to 211 micrometer width were fabricated with a spot heating of a focused 1480 nm wavelength infrared laser to the thin agarose layer coated on the cultivation dish after the cells occupied the reservoir. The collective cell migration velocity maintained constant regardless of their extension distance, whereas the width dependency of those velocities was maximized around 30 micrometer width and decreased both in the narrower and wider microchannels. The single-cell tracking revealed that the decrease of velocity in the narrower width was caused by the apparent increase of aspect ratio of cell shape (up to 8.9). In contrast, the decrease in the wider channels was mainly caused by the increase of the random walk-like behavior of component cells. The results confirmed the advantages of this method: (1) flexible fabrication without any pre-designing, (2) modification even during cultivation, and (3) the cells were confined in the agarose geometry.

Weakening of resistance force by cell\u2013ECM interactions regulate cell migration directionality and pattern formation

Collective migration of epithelial cells is a fundamental process in multicellular pattern formation. As they expand their territory, cells are exposed to various physical forces generated by cell–cell interactions and the surrounding microenvironment. While the physical stress applied by neighbouring cells has been well studied, little is known about how the niches that surround cells are spatio-temporally remodelled to regulate collective cell migration and pattern formation. Here, we analysed how the spatio-temporally remodelled extracellular matrix (ECM) alters the resistance force exerted on cells so that the cells can expand their territory. Multiple microfabrication techniques, optical tweezers, as well as mathematical models were employed to prove the simultaneous construction and breakage of ECM during cellular movement, and to show that this modification of the surrounding environment can guide cellular movement. Furthermore, by artificially remodelling the microenvironment, we showed that the directionality of collective cell migration, as well as the three-dimensional branch pattern formation of lung epithelial cells, can be controlled. Our results thus confirm that active remodelling of cellular microenvironment modulates the physical forces exerted on cells by the ECM, which contributes to the directionality of collective cell migration and consequently, pattern formation.

Nanotopography-Modulated Epithelial Cell Collective Migration.

Matrix nanotopography plays an essential role in regulating cell behaviors including cell proliferation, differentiation, and migration. While studies on isolated single cell migration along the nanostructural orientation have been reported for various cell types, there remains a lack of understanding of how nanotopography regulates the behavior of collectively migrating cells during processes such as epithelial wound healing. We demonstrated that collective migration of epithelial cells was promoted on nanogratings perpendicular to, but not on those parallel to, the wound-healing axis. We further discovered that nanograting-modulated epithelial migration was dominated by the adhesion turnover process, which was Rho-associated protein kinase activity-dependent, and the lamellipodia protrusion at the cell leading edge, which was Rac1-GTPase activity-dependent. This work provides explanations to the distinct migration behavior of epithelial cells on nanogratings, and indicates that the effect of nanotopographic modulations on cell migration is cell-type dependent and involves complex mechanisms.

UNC‐45A is a novel regulator of the intestinal epithelial cell migration

Intestinal epithelial cell migration is a fundamental homeostatic process that regulates establishment of the epithelial architecture in healthy gut and drives mucosal wound healing during intestinal inflammation. Epithelial cell migration depends on remodeling of the cytoskeleton, most notably actin filament turnover and activity of nonmuscle myosin II (NM II) motors. The mechanisms that regulate remodeling of the actomyosin cytoskeleton in migrating intestinal epithelial cells remain poorly understood. UNC-45A is a highly conserved member of the UNC45/CRO1/She4p family proteins and it acts as a chaperone facilitating NM II folding and self-assembly. While previous studies implicated this molecular chaperon in controlling motility of fibroblastic and some tumor cells, its roles in regulating intestinal epithelial cell migration remain unexplored. In the present study, CRISPR/Cas9 mediated gene editing was used to downregulate UNC-45A expression in HT29 and SK-CO15 human colonic epithelial cells. Loss of UNC-45A significantly decreased cell contractility in the collagen contraction assay, thereby indicating impaired activity of NM II motors. Importantly, knockout of UNC-45A significantly decreased both collective epithelial cell migration in a wound heling assay and individual cell migration in the Boyden chamber. The attenuated migration of UNC-45A-depleted epithelial cells was accompanied by their increased adhesion to the collagen matrix, disassembly of basal actin stress fibers and inhibited membrane protrusion at the migrating cell edge. Interestingly, UNC-45A expression was significantly downregulated in isolated intestinal epithelial cells and tissue sections obtained from the colonic mucosa of ulcerative colitis patients. Our results signify UNC-45A as a novel cytoskeletal regulator of intestinal epithelial cell migration, which functions could be inhibited in chronic inflammatory disorders.

Cellular mechanisms and developmental compensation during tissue fusion in the secondary palate

Cleft lip with or without cleft palate (CL/P) is the most common congenital craniofacial anomaly with considerable morbidity in affected children. Though extensive studies have established the basic histological and genetic mechanisms of palatal development, the cellular mechanisms that regulate secondary palate development are still not completely understood. Palate fusion is the crucial final step of palate development, in which the medial edge epithelial cells (MEE) make contact to form a midline epithelial seam (MES), which is subsequently removed to allow mesenchymal continuity. Though much progress has been made on understanding the cellular mechanisms of secondary palate fusion, major questions remain in part due to limitations in imaging technologies. Here we utilized new processing and imaging methods to visualize dynamic cell behaviors underlying secondary palate fusion temporally and spatially. We apply these methods to the study of mouse mutants in which apoptosis, or actomyosin contractility have been conditionally disrupted in the secondary palate epithelium. Our results indicate that apoptosis and cell extrusion contribute to MES removal, but that cellular compensation can overcome their loss. Our studies also reveal a unique form of collective epithelial cell migration that is critical for proper completion of secondary palate fusion and suggesting that secondary palate fusion may be a valuable model for understanding how collective epithelial migration occurs in other developmental and disease contexts. Finally, our observations show that genetically ablating actomyosin contractility disrupts MES removal by disrupting collective epithelial cell migration. Overall, our studies suggest a model of compensatory cellular mechanisms in epithelium removal during palate fusion.

Azobenzene-based sinusoidal surface topography drives focal adhesion confinement and guides collective migration of epithelial cells

Surface topography is a key parameter in regulating the morphology and behavior of single cells. At multicellular level, coordinated cell displacements drive many biological events such as embryonic morphogenesis. However, the effect of surface topography on collective migration of epithelium has not been studied in detail. Mastering the connection between surface features and collective cellular behaviour is highly important for novel approaches in tissue engineering and repair. Herein, we used photopatterned microtopographies on azobenzene-containing materials and showed that smooth topographical cues with proper period and orientation can efficiently orchestrate cell alignment in growing epithelium. Furthermore, the experimental system allowed us to investigate how the orientation of the topographical features can alter the speed of wound closure in vitro. Our findings indicate that the extracellular microenvironment topography coordinates their focal adhesion distribution and alignment. These topographic cues are able to guide the collective migration of multicellular systems, even when cell–cell junctions are disrupted.

Adherens junction regulates cryptic lamellipodia formation for epithelial cell migration.

Collective migration of epithelial cells plays crucial roles in various biological processes such as cancer invasion. In migrating epithelial sheets, leader cells form lamellipodia to advance, and follower cells also form similar motile apparatus at cell-cell boundaries, which are called cryptic lamellipodia (c-lamellipodia). Using adenocarcinoma-derived epithelial cells, we investigated how c-lamellipodia form and found that they sporadically grew from around E-cadherin-based adherens junctions (AJs). WAVE and Arp2/3 complexes were localized along the AJs, and silencing them not only interfered with c-lamellipodia formation but also prevented follower cells from trailing the leaders. Disruption of AJs by removing αE-catenin resulted in uncontrolled c-lamellipodia growth, and this was brought about by myosin II activation and the resultant contraction of AJ-associated actomyosin cables. Additional observations indicated that c-lamellipodia tended to grow at mechanically weak sites of the junction. We conclude that AJs not only tie cells together but also support c-lamellipodia formation by recruiting actin regulators, enabling epithelial cells to undergo ordered collective migration.

Understanding the Principles of Pattern Formation Driven by Notch Signaling by Integrating Experiments and Theoretical Models.

Notch signaling is an evolutionary conserved cell-cell communication pathway. Besides regulating cell-fate decisions at an individual cell level, Notch signaling coordinates the emergent spatiotemporal patterning in a tissue through ligand-receptor interactions among transmembrane molecules of neighboring cells, as seen in embryonic development, angiogenesis, or wound healing. Due to its ubiquitous nature, Notch signaling is also implicated in several aspects of cancer progression, including tumor angiogenesis, stemness of cancer cells and cellular invasion. Here, we review experimental and computational models that help understand the operating principles of cell patterning driven by Notch signaling. First, we discuss the basic mechanisms of spatial patterning via canonical lateral inhibition and lateral induction mechanisms, including examples from angiogenesis, inner ear development and cancer metastasis. Next, we analyze additional layers of complexity in the Notch pathway, including the effect of varying cell sizes and shapes, ligand-receptor binding within the same cell, variable binding affinity of different ligand/receptor subtypes, and filopodia. Finally, we discuss some recent evidence of mechanosensitivity in the Notch pathway in driving collective epithelial cell migration and cardiovascular morphogenesis.

ERK-Mediated Mechanochemical Waves Direct Collective Cell Polarization

During collective migration of epithelial cells, the migration direction is aligned over a tissue-scale expanse. Although the collective cell migration is known to be directed by mechanical forces transmitted via cell-cell junctions, it remains elusive how the intercellular force transmission is coordinated with intracellular biochemical signaling to achieve collective movements. Here, we show that intercellular coupling of extracellular signal-regulated kinase (ERK)-mediated mechanochemical feedback yields long-distance transmission of guidance cues. Mechanical stretch activates ERK through epidermal growth factor receptor (EGFR) activation, and ERK activation triggers cell contraction. The contraction of the activated cell pulls neighboring cells, evoking another round of ERK activation and contraction in the neighbors. Furthermore, anisotropic contraction based on front-rear polarization guarantees unidirectional propagation of ERK activation, and in turn, the ERK activation waves direct multicellular alignment of the polarity, leading to long-range ordered migration. Our findings reveal thatmechanical forces mediate intercellular signaling underlying sustained transmission of guidance cues for collective cell migration.

Planar-Polarized Semaphorin-5c and Plexin A Promote the Collective Migration of Epithelial Cells in Drosophila

Collective migration of epithelial cells is essential for morphogenesis, wound repair, and the spread of many cancers, yet how individual cells signal to one another to coordinate their movements is largely unknown. Here, we introduce a tissue-autonomous paradigm for semaphorin-based regulation of collective cell migration. Semaphorins typically regulate the motility of neuronal growth cones and other migrating cell types by acting as repulsive cues within the migratory environment. Studying the follicular epithelial cells of Drosophila, we discovered that the transmembrane semaphorin, Sema-5c, promotes collective cell migration by acting within the migrating cells themselves, not the surrounding environment. Sema-5c is planar polarized at the basal epithelial surface such that it is enriched at the leading edge of each cell. This location places it in a prime position to send a repulsive signal to the trailing edge of the cell ahead to communicate directional information between neighboring cells. Our data show that Sema-5c can signal across cell-cell boundaries to suppress protrusions in neighboring cells and that Plexin A is the receptor that transduces this signal. Finally, we present evidence that Sema-5c antagonizes the activity of Lar, another transmembrane guidance cue that operates along leading-trailing cell-cell interfaces in this tissue, via a mechanism that appears to be independent of Plexin A. Together, our results suggest that multiple transmembrane guidance cues can be deployed in a planar-polarized manner across an epithelium and work in concert to coordinate individual cell movements for collective migration.

The Na+ /H+ exchanger 1 (NHE1, SLC9A1) is an important regulator of motility in many cells and has been widely studied for its roles in cancer, although its possible role in collective migration of normal epithelial cells has remained unresolved. In the present study, we show that NHE1 expression in MDCK-II kidney epithelial cells accelerated collective cell migration. NHE1 localized to the leading edges of leader cells, as well as to cryptic lamellipodia in submarginal cell rows. Epidermal growth factor, a kidney morphogen, increased displacement of the front row of collectively migrating cells and reduced the number of migration fingers. NHE1 expression increased the number of migration fingers and increased displacement of submarginal cell rows, resulting in additive effects of NHE1 and epidermal growth factor. Finally, NHE1 expression resulted in disorganized development of MDCK-II cell cysts. Thus, NHE1 contributes to collective migration and epithelial morphogenesis, suggesting roles for the transporter in embryonic and early postnatal development.

Coordination of cell migration mediated by site-dependent cell–cell contact

Contact inhibition of locomotion (CIL), the repulsive response of cells upon cell-cell contact, has been the predominant paradigm for contact-mediated responses. However, it is difficult for CIL alone to account for the complex behavior of cells within a multicellular environment, where cells often migrate in cohorts such as sheets, clusters, and streams. Although cell-cell adhesion and mechanical interactions play a role, how individual cells coordinate their migration within a multicellular environment remains unclear. Using micropatterned substrates to guide cell migration and manipulate cell-cell contact, we show that contacts between different regions of cells elicit different responses. Repulsive responses were limited to interaction with the head of a migrating cell, while contact with the tail of a neighboring cell promoted migration toward the tail. The latter behavior, termed contact following of locomotion (CFL), required the Wnt signaling pathway. Inhibition of the Wnt pathway disrupted not only CFL but also collective migration of epithelial cells, without affecting the migration of individual cells. In contrast, inhibition of myosin II with blebbistatin disrupted the migration of both individual epithelial cells and collectives. We propose that CFL, in conjunction with CIL, plays a major role in guiding and coordinating cell migration within a multicellular environment.

Numerical investigation of the role of intercellular interactions on collective epithelial cell migration.

During collective cell migration, the intercellular forces will significantly affect the collective migratory behaviors. However, the measurement of mechanical stresses exerted at cell-cell junctions is very challenging. A recent experimental observation indicated that the intercellular adhesion sites within a migrating monolayer are subjected to both normal stress exerted perpendicular to cell-cell junction surface and shear stress exerted tangent to cell-cell junction surface. In this study, an interfacial interaction model was proposed to model the intercellular interactions for the first time. The intercellular interaction model-based study of collective epithelial migration revealed that the direction of cell migration velocity has better alignment with the orientation of local principal stress at higher maximum shear stress locations in an epithelial monolayer sheet. Parametric study of the effects of adhesion strength indicated that normal adhesion strength at the cell-cell junction surface has dominated effect on local alignment between the direction of cell velocity vector and the principal stress orientation, while the shear adhesion strength has little effect, which provides compelling evidence to help explain the force transmission via cell-cell junctions between adjacent cells in collective cell motion and provides new insights into "adhesive belt" effects at cell-cell junction.

Orientational Coupling Locally Orchestrates a Cell Migration Pattern for Re-Epithelialization.

Re-epithelialization by collective migration of epithelial cells over a heterogeneous environment to restore tissue integrity and functions is critical for development and regeneration. Here, it is reported that the spatial organization of adjacent adherent paths within sparsely distributed extracellular matrix (ECM) has a significant impact on the orientational coupling between cell polarization and collective cell migration. This coupling effect determines the migration pattern for human keratinocytes to regain their cohesion, which impacts the occupancy of epithelial bridge and the migration velocity in wound repair. Statistical studies suggest the converging organization of ECM, in which adjacent paths become closer to each other and finally converge to a junctional point, facilitating collective cell migration mostly within variable ECM organization, as the polarization of the advancing cell sheet is remodeled to align along the direction of cell migration. The findings may help to design implantable ECM to optimize efficient skin regeneration.
