Occurrence and fate of five representative neonicotinoid insecticides across different wastewater treatment plants and the impact on receiving water bodies

Neonicotinoids (NEOs), despite their widespread use as insecticides, exhibit a notable knowledge deficit in regards to their presence in wastewater treatment plants (WWTPs) and their surrounding environments. This study delves into the presence and disposition of 5 NEOs: Thiamethoxam (THM), Clothianidin (CLO), Imidacloprid (IMD), Acetamiprid (ACE), and Thiacloprid (THA) across 3 domestic WWTPs and their receiving waters. Notably, THM, CLO, and ACE were consistently detected in all water and sludge samples, with THM emerging as the most abundant compound in both influent and effluent. Among the 3 WWTPs, WWTP 2, employing a fine bubble oxidation process, achieved the highest removal efficiency, surpassing 68%, in contrast to WWTP 1 (CAST) at 37% and WWTP 3 (A/A/O) at 7%. Biodegradation played a pivotal role in NEO removal, accounting for 36.7% and 68.2% of the total removal in WWTP 1 and WWTP 2, respectively. Surprisingly, in WWTP 3, biotransformation process inadvertently increased ACE and CLO concentrations by approximately 4.1% and 4.5%, respectively. The total NEO concentration in the receiving surface waters ranged from 72.7 to 155.5 ng/L, while sediment concentrations were significantly lower, spanning between 0.10 and 1.53 ng/g. WWTPs serve as both a removal and concentration point for NEOs, thereby significantly influencing their transportation. Additionally, the concentration of most NEOs in the receiving waters progressively increased from upstream to downstream, highlighting the substantial impact of WWTP discharges on natural water environments. This research offers valuable insights into NEO pollution surrounding WWTPs in the Pearl River Delta, ultimately aiding in pollution control and environmental protection decisions.

Low-Level Dietary Clothianidin Exposure Preferentially Causes Prepupal Mortality of Monarch Butterflies (Danaus plexippus).

Data from prior research indicate the prepupal stage of the monarch butterfly life cycle is more sensitive to clothianidin exposure than the larval stage. A set of experiments was conducted to determine if the dietary clothianidin exposures that cause prepupal mortality are environmentally relevant. Monarch larvae were raised from egg to pupae on clothianidin-contaminated swamp milkweed plants (Asclepias incarnata). Larval growth as well as larval and prepupal survival were monitored throughout the experiments, in which the exposures ranged from 1.4 to 2793.1 ng/g leaf. Exposures of 5.4 to 46.9 ng/g leaf resulted primarily in prepupal mortality, whereas higher exposures of 1042.4 to 2793.1 ng/g leaf resulted exclusively in larval mortality, indicating the prepupal stage is more sensitive to clothianidin exposure than the larval stage. A median lethal concentration and a 10% lethal concentration of 37 and 6 ng/g leaf, respectively, were estimated for prepupal mortality. Both effect concentrations are within the range of clothianidin concentrations reported in leaves collected from wild milkweed plants, indicating prepupal mortality is an environmentally relevant effect. Milkweed in agricultural field margins - A neonicotinoid exposure route for pollinators at multiple life stages

Neonicotinoid insecticides move from targeted crops to wildflowers located in adjacent field margins, acting as a potential exposure source for wild pollinators and insect species of conservation concern, including monarch butterflies. Monarchs rely on milkweed over multiple life stages, including as a host plant for eggs and a food source for both larvae (leaves) and adults (flowers). Milkweeds, which are closely associated with field margins, can contain neonicotinoid residues, but previous assessments are constrained to a single plant tissue type. In 2017 and 2018, we sampled milkweeds from 95 field margins adjacent to crop fields (corn, soybean, hay, wheat, and barley) in agricultural landscapes of eastern Ontario, Canada. Milkweeds were sampled during the flower blooming period and leaves and flower tissues were analysed. The neonicotinoids acetamiprid, clothianidin, thiamethoxam, and thiacloprid were detected. Maximum concentrations in leaf samples included 10.30 ng/g of clothianidin in 2017, and 24.4 ng/g of thiamethoxam in 2018. Clothianidin and thiamethoxam percent detections in flowers (72 % and 61 %, respectively) were significantly higher than detections in leaves (24 % and 31 %, respectively). Thiamethoxam concentrations were significantly higher in paired flower samples than leaf samples (median 0.33 ng/g vs <0.07 ng/g) while clothianidin concentrations also trended higher in flowers (median 0.18–0.55 ng/g vs <0.18 ng/g). Only thiamethoxam showed significant differences between years, and we found no effect of crop type, with hay, soybean and corn fields all yielding 50–56 % detections in leaves. We found significantly higher concentrations in older milkweed flowers than young flowers or leaves (medians 0.87 ng/g vs <0.18 ng/g and 0.45 ng/g vs <0.07 ng/g for clothianidin and thiamethoxam, respectively). Our results highlight the importance of considering variation in milkweed tissue type and age of flowers in neonicotinoid exposure risk assessments. Efforts to increase milkweed availability in agricultural landscapes should consider how exposure to neonicotinoids can be mitigated.

Eliminating explanations for Maladera formosae (Coleoptera: Scarabaeidae) preponderance in sandy soil.

Most field corn in the United States receives a neonicotinoid seed treatment for the management of early-season, soil-dwelling insect pests. Grubs of Maladera formosae (Brenske) (Coleoptera: Scarabaeidae) have been reported feeding on young field corn with both low and high rates of clothianidin seed treatments in Indiana, Michigan, and Ohio. Anecdotally, these infestations are restricted to sandy soils in the region. The purpose of this study was to (1) evaluate whether grub populations in corn are restricted to sandy soils, (2) assess whether soil type influences M. formosae survival, and (3) determine whether soil type affects clothianidin uptake by the plant, possibly explaining the observed differences in M. formosae abundance by soil type. We observed nearly 10-times more grubs in sand (>80% sand content) than loam (<80% sand content) soil within a single corn field. Grub survival to adult was not influenced by soil type. We then compared the concentrations of clothianidin seed treatment in the roots and shoots of corn seedlings grown in either sand or loam soil over time. Similar amounts of the active ingredient were found in the roots and shoots of corn grown in both soil types. Within 2 week, the clothianidin concentrations in both soil types had significantly declined in roots and shoots and were no different from the no-insecticide control. These findings suggest that factors other than insecticide exposure contribute to the higher abundance of M. formosae larvae in sand relative to loam soils, even within the same field.

Neonicotinoid retention and transport in a maize cropping system with contour prairie strips

Conservation areas established in agricultural fields can provide habitat for native organisms, but they also have the potential to accumulate and expose organisms to insecticides. Prairie strips are zones of cropland that have been converted to native prairie vegetation. Prairie strips increase biodiversity and reduce nutrient runoff, but they may also accumulate insecticides that endanger visiting organisms or facilitate the movement of insecticides across the landscape. In a study of paired catchments with ongoing neonicotinoid inputs, we measured the impact of prairie strips (10 % of cropland) on the accumulation and movement of the neonicotinoid insecticide clothianidin (CLO) into surface soil, deep soil, plant tissue, and groundwater across multiple slope positions during three phases of a maize growing season. While CLO accumulates in maize leaf tissue midseason following neonicotinoid application, we did not find evidence that prairie plant species in prairie strips accumulate CLO at concentrations lethal to pollinator insects. We also found that downslope soils contained the highest CLO concentrations in both catchments, showing that prairie strips did not eliminate downslope insecticide runoff. Our study adds to the existing literature examining prairie strip effects on downslope agrochemical transport, showing that when prairie strips are planted in cropland with ongoing neonicotinoid inputs, they can provide safe, low-insecticide habitat for visiting organisms amidst their other services, but may not reduce offsite insecticide runoff.

Following Regulation, Imidacloprid Persists and Flupyradifurone Increases in Nontarget Wildlife.

After regulation of pesticides, determination of their persistence in the environment is an important indicator of effectiveness of these measures. We quantified concentrations of two types of systemic insecticides, neonicotinoids (imidacloprid, acetamiprid, clothianidin, thiacloprid, and thiamethoxam) and butenolides (flupyradifurone), in off-crop nontarget media of hummingbird cloacal fluid, honey bee (Apis mellifera) nectar and honey, and wildflowers before and after regulation of imidacloprid on highbush blueberries in Canada in April 2021. We found that mean total pesticide load increased in hummingbird cloacal fluid, nectar, and flower samples following imidacloprid regulation. On average, we did not find evidence of a decrease in imidacloprid concentrations after regulation. However, there were some decreases, some increases, and other cases with no changes in imidacloprid levels depending on the specific media, time point of sampling, and site type. At the same time, we found an overall increase in flupyradifurone, acetamiprid, thiamethoxam, and thiacloprid but no change in clothianidin concentrations. In particular, flupyradifurone concentrations observed in biota sampled near agricultural areas increased twofold in honey bee nectar, sevenfold in hummingbird cloacal fluid, and eightfold in flowers after the 2021 imidacloprid regulation. The highest residue detected was flupyradifurone at 665 ng/mL (parts per billion [ppb]) in honey bee nectar. Mean total pesticide loads were highest in honey samples (84 ± 10 ppb), followed by nectar (56 ± 7 ppb), then hummingbird cloacal fluid (1.8 ± 0.5 ppb), and least, flowers (0.51 ± 0.06 ppb). Our results highlight that limited regulation of imidacloprid does not immediately reduce residue concentrations, while other systemic insecticides, possibly replacement compounds, concurrently increase in wildlife. Developmental toxicity of the neonicotinoid pesticide clothianidin to the larvae of the crustacean Decapoda, Penaeus vannamei

The developmental toxicity effects of neonicotinoid pesticides such as clothianidin have not been fully explored in agricultural applications. This is particularly noteworthy because such pesticides significantly impact the survival rates of invertebrates, with arthropod larvae being particularly vulnerable. This study aimed to address this research gap by specifically investigating the toxicological effects of clothianidin on the developmental stages of the larvae of the economically important aquaculture species Penaeus vannamei. In these experiments, shrimp eggs were exposed to seawater containing different concentrations of clothianidin beginning at N1, and each phase was observed and analyzed to determine its toxic impact on larval development. These results revealed that clothianidin induces an increase in deformity rates and triggers abnormal cell apoptosis. It also significantly reduced survival rates and markedly decreased body length and heart rate in the later stages of larval development (P3). Transcriptomic analysis revealed disruptions in larval DNA integrity, protein synthesis, and signal transduction caused by clothianidin. To survive prolonged exposure, larvae may attempt to maintain their viability by repairing cell structures and enhancing signal transduction mechanisms. This study offers the first empirical evidence of the toxicity of clothianidin to arthropod larvae, underscoring the impact of environmental pollution on aquatic health.

Risk analysis for groundwater intakes based on the example of neonicotinoids

Neonicotinoids are a class of broad-spectrum insecticides that are dominant in the world market. They are widely distributed in the environment. Understanding the sources, distribution, and fate of these contaminants is critical to mitigating their effects and maintaining the health of aquatic ecosystems. Contamination of surface and groundwater by neonicotinoids has become a widespread problem worldwide, requiring comprehensive action to accurately determine the mechanisms behind the migration of these pesticides, their properties, and their adverse effects on the environment. A new approach to risk analysis for groundwater intake contamination with emerging contaminants was proposed. It was conducted on the example of four neonicotinoids (acetamiprid, clothianidin, thiamethoxam, and imidacloprid) in relation to groundwater accessed by a hypothetical groundwater intake, based on data obtained in laboratory tests using a dynamic method (column experiments). The results of the risk analysis conducted have shown that in this case study the use of acetamiprid and thiamethoxam for agricultural purposes poses an acceptable risk, and does not pose a risk to the quality of groundwater extracted from the intake for food purposes. Consequently, it does not pose a risk to the health and life of humans and other organisms depending on that water. The opposite situation is observed for clothianidin and imidacloprid, which pose a higher risk of groundwater contamination. For higher maximum concentration of neonicotinoids used in the risk analysis, the concentration of clothianidin and imidacloprid in the groundwater intake significantly (from several to several hundred thousand times) exceeds the maximum permissible levels for drinking water (<0.1 μg/L). This risk exists even if the insecticides containing these pesticides are used according to the information sheet provided by the manufacturer (lower maximum concentration), which results in exceeding the maximum permissible levels for drinking water from several to several hundred times.

Effects of Clothianidin on Biochemical Parameters of Adult Zebrafish

The ubiquitous use of insecticides leads to detrimental effects on non-target organisms due to accidental exposure. Neonicotinoid insecticides are popularly used worldwide for their high affinity for arthropod nicotinic receptors which effectively kill insect pests. Low affinity towards vertebrate nicotinic receptors, make them safer as compared to traditional insecticides. Recent studies demonstrated that neonicotinoid exposure can cause some toxicity in vertebrates including humans. Zebrafish is one of the popular model organisms to study ecotoxicity. This is the first study on adult zebrafish to report the effect of novel neonicotinoid, Clothianidin on mortality, liver antioxidant stress profile, liver function profile and brain acetylcholinesterase (AChE). Observations were made over two treatment periods, 96 hours, and 21 days, in five groups exposed to varying concentrations of Clothianidin viz. 30mg/L, 50mg/L, 70mg/L, 90mg/L, 110mg/L and a control. Although no mortality was observed, Clothianidin exposure led to increased activity of superoxide dismutase (SOD) enzyme, lipid peroxidation and decreased catalase (CAT), glutathione-S-transferase (GST). Treated groups also showed increased concentrations of liver enzyme alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP) and acid phosphatase (ACP). These results indicate that Clothianidin causes disturbances in the hepatic function. A prominent decreasing trend observed in brain AChE shows that Clothianidin inhibits AChE in adult zebrafish. Further investigations on DNA damage and gene expression studies could be conducted to understand the exact mechanism of action of Clothianidin

Anopheles gambiae larvae’s ability to grow and emerge in water containing lethal concentrations of clothianidin, acetamiprid, or imidacloprid is consistent with cross-resistance to neonicotinoids

BackgroundFor decades, various agrochemicals have been successfully repurposed for mosquito control. However, preexisting resistance caused in larval and adult populations by unintentional pesticide exposure or other cross-resistance mechanisms poses a challenge to the efficacy of this strategy. A better understanding of larval adaptation to the lethal and sublethal effects of residual pesticides in aquatic habitats would provide vital information for assessing the efficacy of repurposed agrochemicals against mosquitoes.MethodsWe reared field-collected mosquito larvae in water containing a concentration of agrochemical causing 100% mortality in susceptible mosquitoes after 24 h (lethal concentration). Using this experimental setup, we tested the effect of lethal concentrations of a pyrrole (chlorfenapyr, 0.10 mg/l), a pyrethroid (deltamethrin, 1.5 mg/l), and three neonicotinoids including imidacloprid (0.075 mg/l), acetamiprid (0.15 mg/l), and clothianidin (0.035 mg/l) on mortality rates, growth, and survival in third-instar larvae of the two sibling species Anopheles gambiae and Anopheles coluzzii collected from Yaoundé, Cameroon.ResultsWe found that An. gambiae and An. coluzzii larvae were susceptible to chlorfenapyr and were killed within 24 h by a nominal concentration of 0.10 mg/l. Consistent with strong resistance, deltamethrin induced low mortality in both species. Lethal concentrations of acetamiprid, imidacloprid, and clothianidin strongly inhibited survival, growth, and emergence in An. coluzzii larvae. By contrast, depending on the active ingredient and the population tested, 5–60% of immature stages of An. gambiae were able to grow and emerge in water containing a lethal concentration of neonicotinoids, suggesting cross-resistance to this class of insecticides.ConclusionsThese findings corroborate susceptibility profiles observed in adults and suggest that unintentional pesticide exposure or other cross-resistance processes could contribute to the development of resistance to neonicotinoids in some Anopheles populations.Graphical

Clothianidin-resistant Anopheles gambiae adult mosquitoes from Yaoundé, Cameroon, display reduced susceptibility to SumiShield® 50WG, a neonicotinoid formulation for indoor residual spraying

BackgroundChronic exposure of mosquito larvae to pesticide residues and cross-resistance mechanisms are major drivers of tolerance to insecticides used for vector control. This presents a concern for the efficacy of clothianidin, an agricultural neonicotinoid prequalified for Indoor Residual Spraying (IRS).MethodsUsing standard bioassays, we tested if reduced susceptibility to clothianidin can affect the efficacy of SumiShield® 50WG, one of four new IRS formulations containing clothianidin. We simultaneously monitored susceptibility to clothianidin and to SumiShield 50WG, testing adults of Anopheles gambiae, An. coluzzii and Culex sp sampled from urban, suburban and agricultural areas of Yaoundé, Cameroon.ResultsWe found that in this geographic area, the level of susceptibility to the active ingredient predicted the efficacy of SumiShield 50WG. This formulation was very potent against populations that reached 100% mortality within 72 h of exposure to a discriminating concentration of clothianidin. By contrast, mortality leveled off at 75.4 ± 3.5% within 7 days of exposure to SumiShield 50WG in An. gambiae adults collected from a farm where the spraying of the two neonicotinoids acetamiprid and imidacloprid for crop protection is likely driving resistance to clothianidin.ConclusionsDespite the relatively small geographic extend of the study, the findings suggest that cross-resistance may impact the efficacy of some new IRS formulations and that alternative compounds could be prioritized in areas where neonicotinoid resistance is emerging.

Is Anopheles gambiae (sensu stricto), the principal malaria vector in Africa prone to resistance development against new insecticides? Outcomes from laboratory exposure of An. gambiae (s.s.) to sub-lethal concentrations of chlorfenapyr and clothianidin

Indiscriminate use of pesticides in the public health and agriculture sectors has contributed to the development of resistance in malaria vectors following exposure to sub-lethal concentrations. To preserve the efficacy of vector control tools and prevent resistance from spreading, early resistance detection is urgently needed to inform management strategies. The introduction of new insecticides for controlling malaria vectors such as clothianidin and chlorfenapyr requires research to identify early markers of resistance which could be used in routine surveillance. This study investigated phenotypic resistance of Anopheles gambiae (sensu stricto) Muleba-Kis strain using both WHO bottle and tube assays following chlorfenapyr, clothianidin, and alpha-cypermethrin selection against larvae and adults under laboratory conditions. High mortality rates were recorded for both chlorfenapyr-selected mosquitoes that were consistently maintained for 10 generations (24-h mortality of 92–100% and 72-h mortality of 98–100% for selected larvae; and 24-h mortality of 95–100% and 72-h mortality of 98–100% for selected adults). Selection with clothianidin at larval and adult stages showed a wide range of mortality (18–91%) compared to unselected progeny where mortality was approximately 99%. On the contrary, mosquitoes selected with alpha-cypermethrin from the adult selection maintained low mortality (28% at Generation 2 and 23% at Generation 4) against discrimination concentration compared to unselected progeny where average mortality was 51%. The observed resistance in the clothianidin-selected mosquitoes needs further investigation to determine the underlying resistance mechanism against this insecticide class. Additionally, further investigation is recommended to develop molecular markers for observed clothianidin phenotypic resistance.

Comparing the effects of three neonicotinoids on embryogenesis of the South African clawed frog Xenopus laevis

Neonicotinoids (NEOs) are widely used insecticides that are ubiquitous in agricultural use. Since NEOs are found in natural waters as well as in tap water and human urine in regions where NEOs are widely used, NEOs pose a potential hazard to non-target organisms such as animals and humans. Some of the commonly detected NEOs are imidacloprid (IMD), thiamethoxam (TMX), and its metabolite clothianidin (CLO). Although previously published scientific information, including an assessment of the environmental risks, particularly for bees, had resulted in a ban on the outdoor use of these three NEOs in the EU – their use is now only permitted in closed greenhouses – these NEOs continue to be used in agriculture in many other parts of the world. Therefore, a detailed study and comparison of the effects of NEOs on the embryonic development of non-target organisms is needed to further define the risk profiles.Embryos of the South African clawed frog Xenopus laevis, a well-established aquatic model, were exposed to different concentrations of IMD, TMX, or CLO (0.1–100 mg/L) to study and compare the possible effects of a single contaminant in natural water bodies on early embryogenesis. The results included a reduced body length, a smaller orbital space, impaired cranial cartilage and nerves, and an altered heart structure and function. At the molecular level, NEO exposure partially resulted in an altered expression of tissue-specific factors, which are involved in eye, cranial placode, and heart development.Our results suggest that the NEOs studied negatively affect the embryonic development of the non-target organism X. laevis. Since pesticides, especially NEOs, pollute the environment worldwide, it is suggested that they are strictly controlled and monitored in the areas where they are used. In addition, the question arises as to whether pesticide metabolites also pose a risk to the environment and need to be investigated further so that they can be taken into account when registering ingredients.

Bees are important pollinators of agricultural crops, but their populations are at risk when pesticides are used. One of the largest risks bees face is poisoning of floral nectar and pollen by insecticides. Studies of bee detection of neonicotinoids have reported contradictory evidence about whether bees can taste these pesticides in sucrose solutions and hence avoid them. Here, we use an assay for the detection of food aversion combined with single-sensillum electrophysiology to test whether the mouthparts of the buff-tailed bumblebee (Bombus terrestris) detect the presence of pesticides in a solution that mimicked the nectar of oilseed rape (Brassica napus). Bees did not avoid consuming solutions containing concentrations of imidacloprid, thiamethoxam, clothianidin, or sulfoxaflor spanning six orders of magnitude, even when these solutions contained lethal doses. Only extremely high concentrations of the pesticides altered spiking in gustatory neurons through a slight reduction in firing rate or change in the rate of adaptation. These data provide strong evidence that bumblebees cannot detect or avoid field-relevant concentrations of pesticides using information from their mouthparts. As bees rarely contact floral nectar with other body parts, we predict that they are at high risk of unwittingly consuming pesticides in the nectar of pesticide-treated crops.

Bioefficacy and Phytotoxicity of Clothianindin 50 WDG against Thrips and Mealybugs in Grapevine

Two separate experiments were conducted in the field to examine the effectiveness of clothianidin 50 WDG against thrips and mealybugs that were infesting grapevines. The findings highlighted that applying clothianidin 50 WDG @ 1000 g ha-1 proved highly efficient in curbing the population of the sucking pests, outperforming lower doses of the compound. Following two rounds of soil drenching spaced 14 days apart, there was an average reduction of 76.63% for thrips and 78.09% for mealybugs. Other concentrations of clothianidin 50 WDG (600, 500 and 400 g ha-1) demonstrated significant reductions in the populations of thrips and mealybugs, exhibiting a similar level of efficacy as the standard check (Methomyl 40 SP and Buprofezin 25 SC) in all field experiments. Notably, no signs of harm to the grapevine were observed at any of the tested doses. Clothianidin 50 WDG @ 1000 g ha-1 resulted in the highest grape yield, producing 14.25 and 8.94 kg vine-1 at Madhampatti and Kalampalayam, respectively. Throughout the clothianidin 50 WDG treatment plots, the presence of natural enemies was noted and their activity remained unaffected, indicating no adverse impact on the natural enemy population.

Concentrations of neonicotinoid insecticides and their metabolites in multiple urine samples collected from pregnant women in Japan

Exposure to low doses of neonicotinoid (NEO) insecticides during pregnancy may have a negative impact on the neurodevelopment of children. Biomarkers are frequently used to investigate the exposure and its health effects in environmental epidemiology. In the present study, we aimed to evaluate the utility of urinary biomarkers of NEO exposure by calculating intra-class correlation coefficients (ICCs). Urine samples were collected from 30 pregnant women who were non-smokers and living in a central part of Japan, between 12 and 22 weeks of gestation. We collected first morning voided (FMV) urine and afternoon spot (PM) urine samples on the same day on five occasions within a 2-week period. The urinary concentrations of NEO and their metabolites (NEO/m) were measured by liquid chromatography–tandem mass spectrometry. Normalisation to the specific gravity and creatinine concentration was used in the calculation of ICC. The number of urine samples required to obtain a reliable biomarker estimate of NEO exposure was calculated. The most frequently detected NEO/m was N-desmethyl-acetamiprid (N-dm-ACE), followed by clothianidin (CLO), thiamethoxam (THX) and dinotefuran (DIN), in both FMV and PM urine samples. The ICC calculations showed poor reproducibility for the measurement of CLO and THX concentrations (ICCs ≤0.25) in both FMV and PM urine samples and for DIN (ICC ≤0.29) in the FMV urine samples, and moderate reproducibility was identified for N-dm-ACE (ICC >0.60) in both FMV and PM urine samples and for DIN (ICC >0.40) in the PM urine samples. A minimum of 1–19 spot urine samples per individual was required to provide a reliable biomarker estimate over a 2 week period. These results suggest that pregnant Japanese women are frequently exposed to NEOs, and that, with the exception of urinary N-dm-ACE, a single measurement of urinary NEO concentrations may not be an appropriate method of characterising overall exposure.

Association between maternal urinary neonicotinoid concentrations and child development in the Japan Environment and Children’s Study

BackgroundRecent studies have reported the effect of neonicotinoid (NEO) exposures on development in human. However, information about the risk of childhood development delays due to NEO exposures is limited. ObjectivesThe study aimed to examine the association between NEO exposure and child development up to 4 years of age using data of the Japan Environment and Children’s Study. MethodsThe study employed urinary NEO and metabolite concentrations in the first and second or third trimesters; the Japanese translation of the Ages and Stages Questionnaire (third edition; J-ASQ-3) scores on developmental delay in five domains, namely communication, gross motor skills, fine motor skills, problem solving, and personal-social characteristics from 6 months to 4 years of age; and self-reported questionnaire data. The associations between urinary NEO concentrations and J-ASQ-3 results were analysed using the treed distributed lag mixture model. A total of 8538 participants were included in statistical analyses. ResultsThe determination rates of urinary acetamiprid-N-desmethyl (dm-ACE), clothianidin (CLO), dinotefuran (DIN) and thiamethoxam (THX) were greater than 50%. Median urinary dm-ACE, CLO, DIN and THX concentrations were 0.34, 0.14, 0.22 and 0.05 ng/ml, respectively, in samples collected during gestational weeks < 23, and 0.28, 0.12, 0.18 and 0.04 ng/ml, respectively, in those collected during gestational weeks ≥ 23. The binomial scores divided by the cut-off values of the J-ASQ were used in the treed distributed lag mixture model. The highest percentage for a domain with a value less than the cut-off value was ‘problem solving’ at 6 months of age among all the J-ASQ-3 scores (10.5%). There was no statistically significant association between maternal urinary dm-ACE, CLO, DIN and THX concentrations during pregnancy and the J-ASQ-3 results up to 4 years of age. Objective assessment of child development in different populations may be warranted to confirm our findings.

Higher temperature and daily fluctuations aggravate clothianidin toxicity towards Limnodrilus hoffmeisteri

In nature, aquatic organisms may suffer from chemical pollution, together with thermal stress resulted from global warming. However, limited information is available on the combined effects of pesticide with climate change on aquatic organisms. In this study, the acute toxicity of clothianidin to Limnodrilus hoffmeisteri as well as its effect on the induction of oxidative stress under both constant temperature and daily temperature fluctuation (DTF) regimes was investigated. Results showed that clothianidin exhibited the minimal toxicity to L. hoffmeisteri at 25 °C, which was magnified by both increased or decreased temperatures and 10 °C DTF. At different temperatures (15 °C, 25 °C and 35 °C), clothianidin exposure led to the elevated reactive oxygen species (ROS) levels and activated the antioxidant enzymes to resist against the oxidative stress. However, the antioxidant response induced by clothianidin was overwhelmed at high temperature as evidenced by decreased glutathione (GSH) content. Significant elevation of catalase (CAT) and peroxidase (POD) activities but depletion of GSH was also observed in worms treated with clothianidin under DTF after 24 h. The results indicated that high temperature and DTF could aggravate the clothianidin-induced oxidative stress. Moreover, the critical thermal maximum (CTmax) of the worms decreased with the increasing clothianidin concentrations, suggesting that exposure to clothianidin could reduce the heat tolerance of L. hoffmeisteri. Our work highlights the crucial importance to integrate temperature changes into risk assessment of pesticides under global warming.

Exposure to multiple neonicotinoid insecticides, oxidative stress, and gestational diabetes mellitus: Association and potential mediation analyses

As the most extensively used insecticides worldwide, neonicotinoid insecticides (NNIs) have received a growing global concern over their adverse health effects. This study aimed to assess the associations of urinary concentrations of NNIs in early pregnancy with gestational diabetes mellitus (GDM) and the mediation roles of oxidative DNA damage, RNA damage, and lipid peroxidation in the associations. With a prospective nested case-control study, 519 GDM cases and 519 controls were matched on the infant’s sex and maternal age. Urinary biomarkers of NNIs exposure and oxidative stress were measured in early pregnancy. We estimated the associations of single and the mixture of NNIs and their metabolites with GDM by conditional logistic regression and quantile g-computation models, respectively. The mediating roles of oxidative stress were evaluated by the structural equation model. The odds of GDM significantly increased by 15 %, 18 %, 26 %, 42 %, 49 %, and 13 % in each unit increment of ln-transformed concentrations of urinary imidacloprid (IMI), imidacloprid-olefin (IMI-olefin), desnitro-imidacloprid (DN-IMI), thiamethoxam (THM), clothianidin, and desmethyl-clothianidin, respectively. Exposure to the mixture of NNIs was associated with increased odds of GDM (adjusted OR: 1.76; 95 %CI: 1.45, 2.13). Advanced maternal age enhanced the associations of 5-hydroxy-IMI, DN-IMI, and IMI-olefin with GDM (P < 0.05), and being overweight/obese before pregnancy strengthened the effects of IMI, IMI-olefin, and THM on GDM (P < 0.05). In the association of NNIs exposure and GDM, the proportions mediated by oxidative DNA damage, RNA damage, and overall oxidative stress were 9.8 %, 11.8 %, and 14.5 %, respectively (P < 0.05). Exposure to individual NNIs and a mixture of NNIs were associated with GDM, and maternal age and pre-pregnancy BMI may modify the association. The possible mechanism underlying the association between NNIs and GDM may involve oxidative damage to nucleic acids.
