Controlling plasmonic suprastructures through self-assembly of gold nanoparticles with hybrid copolymer-lipid vesicles

Hybrid lipid membranes incorporating amphiphilic copolymers have gained significant attention due to their potential applications in various fields, including drug delivery and sensing. By combining the properties of copolymers and lipid membranes, such as enhanced chemical tunability and stability, environmental responsiveness, and multidomain nature, novel membrane architectures have been proposed. In this study, we investigated the potentialities of hybrid membranes made of two distinct components: the rigid fully saturated phospholipid 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) and the soft copolymer poly(butadiene-b-ethyleneoxide) (PBD-b-PEO). The objective was to explore the interaction of citrate-coated gold nanoparticles (AuNPs) and the hybrid membrane, aiming at constructing AuNPs-hybrid vesicles suprastructures with controlled and adjustable plasmonic properties. A series of experimental techniques were employed to investigate hybrid free-standing and supported membranes. The results revealed that the incorporation of the copolymer into the lipid membrane promotes AuNPs clustering, demonstrating a distinctive aggregative phenomenon of citrate-coated AuNPs on multidomain membranes. Importantly, we show that the size and morphology of AuNPs clusters can be precisely controlled in non-homogeneous membranes, enabling the formation of hybrid suprastructures with controlled patch properties. These results highlight the potential of lipid-copolymer hybrid membranes for designing functional materials with tailored plasmonic properties, with potential applications in nanomedicine and sensing.

PH-Mediated Size-Selective Adsorption of Gold Nanoparticles on Diblock Copolymer Brushes.

Precise control of nanoparticles at interfaces can be achieved by designing stimuli-responsive surfaces that have tunable interactions with nanoparticles. In this study, we demonstrate that a polymer brush can selectively adsorb nanoparticles according to size by tuning the pH of the buffer solution. Specifically, we developed a facile polymer brush preparation method using a symmetric polystyrene-b-poly(2-vinylpyridine) (PS-b-P2VP) block copolymer deposited on a grafted polystyrene layer. This method is based on the assembly of a PS-b-P2VP thin film oriented with parallel lamellae that remains after exfoliation of the top PS-b-P2VP layer. We characterized the P2VP brush using X-ray reflectivity and atomic force microscopy. The buffer pH is used to tailor interactions between citrate-coated gold nanoparticles (AuNPs) and the top P2VP block that behaves like a polymer brush. At low pH (∼4.0) the P2VP brushes are strongly stretched and display a high density of attractive sites, whereas at neutral pH (∼6.5) the P2VP brushes are only slightly stretched and have fewer attractive sites. A quartz crystal microbalance with dissipation monitored the adsorption thermodynamics as a function of AuNP diameter (11 and 21 nm) and pH of the buffer. Neutral pH provides limited penetration depth for nanoparticles and promotes size selectivity for 11 nm AuNP adsorption. As a proof of concept, the P2VP brushes were exposed to various mixtures of large and small AuNPs to demonstrate selective capture of the smaller AuNPs. This study shows the potential of creating devices for nanoparticle size separations using pH-sensitive polymer brushes.

Flash Colloidal Gold Nanoparticle Assembly in a Milli Flow System: Implications for Thermoplasmonic and for the Amplification of Optical Signals

The assembly and stabilization of a finite number of nanocrystals in contact in water could maximize the optical absorption per unit of material. Some local plasmonic properties exploited in applications, such as photothermia and optical signal amplification, would also be maximized which is important in the perspective of mass producing nanostructures at a lower cost. The main lock is that bringing charged particles in close contact requires the charges to be screened/suppressed, which leads to the rapid formation of micrometric aggregates. In this article, we show that aggregates containing less than 60 particles in contact can be obtained with a milli-flow system composed of turbulent mixers and flow reactors. This process allows to stop a fast non-equilibrium colloidal aggregation process at millisecond times after the initiation of the aggregation process which allows to control the aggregation number. As a case study, we considered the rapid mixing of citrate coated gold nanoparticles (NP) and AlCl3 in water to initiate a fast aggregation controlled by diffusion. Injecting a solution of polycation using a second mixer allowed us to arrest the aggregation process after a reaction time by formation of overcharged cationic aggregates. We obtained within seconds stable dispersions of a few milliliters composed of particle aggregates. Our main result is to show that it is possible to master the average aggregation number between 2 and 60 NP per aggregate by varying the a reaction time between 10 ms and 1 s.

Electrochemical Estimations of the Gold Nanoparticle Size Effect on Cysteine-Gold Oxidation

Gold nanoparticles are interesting for nanobiomedical applications, such as for drug delivery and as diagnostic imaging contrast agents. However, their stability and reactivity in-vivo are influenced by their surface properties and size. Here, we investigate the electrochemical oxidation of differently sized citrate-coated gold nanoparticles in the presence and absence of L-cysteine, a thiol-containing amino acid with high binding affinity to gold. We found that smaller sized (5, 10 nm) gold nanoparticles were significantly more susceptible to electrochemical L-cysteine interactions and/or L-cysteine-facilitated gold oxidation than larger (20, 50 nm) sized gold nanoparticles, both for the same mass and nominal surface area, under the conditions investigated (pH 7.4, room temperature, stagnant solutions, and scan rates of 0.5 to 450 mV s−1). The electrochemical measurements of drop-casted gold nanoparticle suspensions on paraffin-impregnated graphite electrodes were susceptible to the quality of the electrode. Increased cycling resulted in irreversible oxidation and detachment/oxidation of gold into solution. Our results suggest that L-cysteine-gold interactions are stronger for smaller nanoparticles.

Innovative and versatile nanoplasmonic approach for the full sensing of proteinogenic aminoacids in nutritional supplements

A current nourishment issue is the development of smart and reliable analytical strategies to control in a simple way main bioactive compounds of nutritional supplements whose increasing use is deemed a trend nowadays. With this aim a quick and highly sensitive plasmonic sensor using simple citrate coated gold nanoparticles (AuNPs) as optical probe, was developed for both qualitative and quantitative global assessment of all the proteinogenic amino acids in nutritional supplements. AuNPs of five different sizes (from 19 to 74 nm) were synthesized, characterized and evaluated as optimal transductor element for the sensing approach. Critical physic-chemical conditions controlling aggregation (pH, incubation time, AuNPs amount and ionic strength) were investigated on the main five types of aas, structurally different attending to their R-side chain and with expected distinctive behaviour on aggregation mechanisms, which are also discussed. All proteinogenic amino acids induced AuNPs aggregation at low pH (2.5) causing a change in the colour solution from red to blue, as well as a redshift in the plasmon band from 518 nm (disperse NPs) to 650 nm (aggregated NPs). Based on this sensing approach two different strategies are allowed, a preliminary qualitative/semi-quantitative screening just by the naked eye (simple spot test) and a second quantitative confirmation procedure using the analytical signal (A650/A518). Reliability of quantitative approach was assessed by an exhaustive validation procedure, where matrix effects and potential interferences usually present in commercial samples and affecting the analytical signal were mainly focussed. The results for the analysis of complex nutritional samples were validated by means of a statistical comparison with those ones of the official reference Kjeldahl method (paired Student test-t) at a 95% confidence level. This is the first sensing approach able to provide the global estimation of proteinogenic aas amount based on their simply AuNPs aggregation induction, irrespectively of their R-side chain structure.

A Framework of Paracellular Transport via Nanoparticles-Induced Endothelial Leakiness.

Nanomaterial‐induced endothelial leakiness (NanoEL) is an interfacial phenomenon denoting the paracellular transport of nanoparticles that is pertinent to nanotoxicology, nanomedicine and biomedical engineering. While the NanoEL phenomenon is complementary to the enhanced permeability and retention effect in terms of their common applicability to delineating the permeability and behavior of nanoparticles in tumoral environments, these two effects significantly differ in scope, origin, and manifestation. In the current study, the descriptors are fully examined of the NanoEL phenomenon elicited by generic citrate‐coated gold nanoparticles (AuNPs) of changing size and concentration, from microscopic gap formation and actin reorganization down to molecular signaling pathways and nanoscale interactions of AuNPs with VE‐cadherin and its intra/extracellular cofactors. Employing synergistic in silico methodologies, for the first time the molecular and statistical mechanics of cadherin pair disruption, especially in response to AuNPs of the smallest size and highest concentration are revealed. This study marks a major advancement toward establishing a comprehensive NanoEL framework for complementing the understanding of the transcytotic pathway and for guiding the design and application of future nanomedicines harnessing the myriad functions of the mammalian vasculature.

Development of MRI-Detectable Boron-Containing Gold Nanoparticle-Encapsulated Biodegradable Polymeric Matrix for Boron Neutron Capture Therapy (BNCT).

This study aimed to develop a novel magnetic resonance imaging (MRI)-detectable boron (B)-containing nanoassemblies and evaluate their potential for boron neutron capture therapy (BNCT). Starting from the citrate-coated gold nanoparticles (AuNPs) (23.9 ± 10.2 nm), the diameter of poly (D, L-lactide-co-glycolide) AuNPs (PLGA-AuNPs) increased approximately 110 nm after the encapsulation of the PLGA polymer. Among various B drugs, the self-produced B cages had the highest loading efficiency. The average diameter of gadolinium (Gd)- and B-loaded NPs (PLGA-Gd/B-AuNPs) was 160.6 ± 50.6 nm with a B encapsulation efficiency of 28.7 ± 2.3%. In vitro MR images showed that the signal intensity of PLGA-Gd/B-AuNPs in T1-weighted images was proportional to its Gd concentration, and there exists a significantly positive relationship between Gd and B concentrations (R2 = 0.74, p < 0.005). The hyperintensity of either 250 ± 50 mm3 (larger) or 100 ± 50 mm3 (smaller) N87 xenograft was clearly visualized at 1 h after intravenous injection of PLGA-Gd/B-AuNPs. However, PLGA-Gd/B-AuNPs stayed at the periphery of the larger xenograft while located near the center of the smaller one. The tumor-to-muscle ratios of B content, determined by inductively coupled plasma mass spectrometry, in smaller- and larger-sized tumors were 4.17 ± 1.42 and 1.99 ± 0.55, respectively. In summary, we successfully developed theranostic B- and Gd-containing AuNPs for BNCT in this study.

The effect of pH on the sorption of gold nanoparticles on illite

Sorption between nanoparticles (NPs) and minerals may critically affect the migration of associated elements as well as the environmental impact of NPs. Since illite is widely present in soil, sediment, and water, we have experimentally investigated the sorption behavior of citrate-coated gold nanoparticles (AuNPs) as model NPs on illite under different pH and mineral mass conditions. We demonstrated that sorption of these negatively charged AuNPs strongly depended on the suspension pH. At pH above 8, which coincided with the apparent point of zero charge (pH 7.9) of our illite sample, only marginal sorption of AuNPs was observed. At pH 3–8, significant sorption of AuNPs on illite was found, with almost complete sorption occurring at more acidic conditions (pH 3–4). TEM observations revealed that sorption took place through the attachment AuNPs on illite edges. At pH 2, AuNPs mostly formed chain-like fused structures and precipitated out of the suspension. Based upon the above pH dependence, residual organic ligand content after sorption, and complementary sorption results with positively charged AuNPs, we conclude that the sorption process is mainly driven by the electrostatic attraction between negatively charged AuNPs and positively charged illite edges, with possible competitive involvement of citrate molecules. We expect that our findings will improve our understanding of NP–mineral interaction and the environmental fate of NPs.

Effect of Phosphate, Sulfate, Arsenate, and Pyrite on Surface Transformations and Chemical Retention of Gold Nanoparticles (Au-NPs) in Partially Saturated Soil Columns.

The understanding of engineered nanoparticle (ENP) fate and transport in soil-water environments is important for the evaluation of potential risks of ENPs to the ecosystem and human health. The effects of pyrite grains and three types of oxyanions-sulfate, phosphate, and arsenate-on the retention of citrate-coated gold nanoparticles (citrate-Au-NPs) were studied in partially saturated soil column experiments. The mobility of Au-NP was found to be in the order: Au-NP-sulfide (originating from pyrite) > Au-NP-sulfate > citrate-Au-NP > Au-NP-arsenate > Au-NP-phosphate. Chemical retention mechanisms, including hydrogen bonding and calcium bridging, are proposed and discussed. The retention of Au-NPs in soil columns increases with the increased ability of transformed Au-NP surfaces to create strong hydrogen bonding through adsorbed oxyanions with soil surfaces. Oxyanions were also found to reduce aggregation and aggregate size of Au-NPs upon interaction with Ca2+ solution. While the effects of cationic substances on ENP transport and stability have been studied frequently, the results here demonstrate that anionic substances have a substantial effect on Au-NP transport and stability. Furthermore, this study highlights the importance of examining ENPs under environmentally relevant condition, and the significant effect of ENP transformations on their mobility in soils.

Nanoparticle Assisted Remodeling of Proteotoxic SOD1 Mutants Alters the Biointerface of the Functional Interaction of Microtubules and Kinesin Motors.

Transport deficits with motor neuron degeneration have been implicated in amyotrophic lateral sclerosis (ALS). We report a biomimetic system composed of microtubules/kinesin motor that mimics the dysregulated motor dynamics of ALS. Pathogenic ALS mutants A4V SOD1 completely shut off motility. Treatment with 5 nm citrate coated gold nanoparticles recovers the impaired motor stepping by remodeling the A4V SOD1 rather than stabilizing microtubules or protein folding. Instead, gold nanoparticles alter the protein by a mechanism that reforms protein elements of A4V SOD1, in turn fixing the aberrant interaction of kinesin with microtubules. Reinstating kinesin motility holds potential for managing debilitating ALS.

The mechanism of tuning the morphology of bio-conjugated ZnO nanoparticles with citrate coated gold nanoparticles for degradation of EBT: DFT and experimental study

The electronic structure and the structural properties of the citrate capped gold (Au/Cit) adsorbed on the amino acids (AA) [Histidine (His), Arginine (Arg) and Aspartic acid (Asp)] conjugated Zinc Oxide (ZnO) nanoparticles (NPs), ZnO/AA are studied using the Density Functional Theory (DFT) simulations. On the basis of first principles calculation, we find that there is an exceptional high interaction between Au/Cit and Asp conjugated ZnO NPs (ZnO/Asp). This attaching mode can substantially change the morphology of the ZnO/Asp. On the other hand, the morphology of the Arg and His conjugated ZnO NPs (ZnO/Arg & ZnO/His) do not undergo considerable change because of the comparatively less binding between Au/Cit and ZnO/AA (Arg and His). Our conclusions of theoretical modelling are supported by the experimental results. The experimental work highlights the importance of the morphology of ZnO/AA, coated with Au/Cit (ZnO/AA-Au/Cit) nanocomposites in increasing the photocatalytic activity, opening applications for the degradation of Eriochrome Black T (EBT) in aqueous solution. Based on photodegradation studies and first principles calculations, it is observed that ZnO/Arg-Au/Cit is more effective photocatalyst for the degradation of EBT in aqueous solution as compared to ZnO/Asp-Au/Cit and ZnO/His-Au/Cit.

DNA Binding to Gold Nanoparticles through the Prism of Molecular Selection: Sequence-Affinity Relation.

Native DNA strongly adsorbs to citrate-coated gold nanoparticles (AuNPs). The resulting composites (DNA/AuNPs) are valuable materials in many fields, especially in biomedicine. For this reason, the process of adsorption is a focus for intensive research. In this work, DNA adsorption to gold nanoparticles was studied using a molecular selection procedure followed by high-throughput DNA sequencing. The chemically synthesized DNA library containing a central N26 randomized fragment was sieved through four cycles of adsorption to AuNPs in a tree-like selection-amplification scheme (SELEX (Selective Evolution of Ligands by EXponential enrichment)). The frequencies of occurrence of specific oligomeric DNA motifs, k-mers ( k = 1-6), in the initial and selected pools were calculated. Distribution of secondary structures in the pools was analyzed. A large set of diverse A, T, and G enriched k-mers undergo a pronounced positive selection, and these sequences demonstrate faster and strong binding to the AuNPs. For facile binding, such structural motifs should be located in the loop regions of weak intramolecular complexes-hairpins with imperfect stem, or other portion of the structure, which is unpaired under selection conditions. Our data also show that, under the conditions employed in this study, cytosine is significantly depleted during the selection process, although guanine remains unchanged. These regularities were confirmed in a series of binding experiments with a set of synthetic DNA oligonucleotides. The detailed analysis of DNA binding to AuNPs shows that the sequence specificity of this interaction is low due to its nature, although the presence and the number of specific structural motifs in DNA affect both the rate of formation and the strength of the formed noncovalent associates with AuNPs.

The Aromatic Amine pKa Determines the Affinity for Citrate-Coated Gold Nanoparticles: In Situ Observation via Hot Spot-Normalized Surface-Enhanced Raman Spectroscopy

The in situ characterization of organic pollutant dynamics on engineered interfaces provides fundamental insights about pollutant fate, transport, and toxicity in aquatic systems. In this study, the association between aromatic amines and citrate-coated gold nanoparticles (cit-AuNPs) under different pH conditions was monitored in situ using hot spot-normalized surface-enhanced Raman spectroscopy (HSNSERS). HSNSERS minimizes the uncertainty of SERS measurements that arises from substrate heterogeneity, laser intensity fluctuation, and laser focusing, and its application enabled quantification of the influence of solution pH on the measured SERS signals. Our results demonstrate that the accumulation of chloroanilines on cit-AuNP surfaces is dictated by solution pH and analyte pKa. At circumneutral pH (≈5.6), only the chloroanilines with relatively high pKa values were readily detected by SERS, while those with low pKa values (<3) were undetectable. In contrast, at low pH, only the chloroanilines with low pKa values were detected. These results highlight the importance of electrostatic forces in modulating the accumulation of the pollutant on cit-AuNP surfaces and suggest the potential for selective pollutant detection by solution pH adjustment. HSNSERS, an innovative analytical platform, can be easily extended to in situ monitoring of the accumulation of pollutants onto gold, silver, and copper nanoparticles triggered by different environmental stimuli (e.g., pH).

The contribution of hydrogen peroxide to the radiosensitizing effect of gold nanoparticles.

Plasmid DNA in aerated aqueous solution is used as a probe to determine whose of the reactive oxygen species (ROS) generated after absorption of ultra-soft X-rays (USX) take part in biomolecule damage in the presence and in absence of Gold Nano-Particles (GNP) and specific scavengers. Citrate-coated GNPs with core sizes of 6, 10 and 25 nm are synthetized and characterized, especially in terms of plasmon band shift, ζ-potential and hydrodynamic radii (respectively 9, 21 and 30 nm). We confirm the radiosensitizing effect of GNP and show that the SSB number per plasmid increases when, for a same mass of gold element, the core size of the gold nanoparticles decreases. Hydroxyl radicals (OH) are scavenged using the positively-charged 2-amino-2-hydroxymethyl-1,3-propanediol (TRIS) and the neutral dimethyl sulfoxide (DMSO) molecules. Due to both negatively-charged environments of DNA and GNP, at identical scavenging capacity, TRIS is more effective at quenching OH than DMSO. The strong radiosensitizing effect of hydroxyl radicals is confirmed. Methanoate anions are then used to transform OH into hydrogen peroxide; the latter being known to be non-aggressive regarding DNA in the absence of easily oxidable metallic ions (Fenton reactions). Surprisingly, in the presence of GNP, high DNA damage yields are observed even though hydrogen peroxide might not be hold as responsible. Conversely, the radiosensitizing effect of GNP is not observed anymore when H2O2 is scavenged using pyruvate ions. We demonstrate that hydrogen peroxide constitutes quite unexpectedly a hidden stock of OH which are activated at the surface of the GNP by decomposition of H2O2 molecules.

Adsorption Dynamics and Structure of Polycations on Citrate-Coated Gold Nanoparticles

Despite the widespread application of engineered multilayered polyelectrolyte-coated gold nanoparticles (AuNPs), their interparticle interaction is not fully understood in part because of a lack of molecular scale observation of the layer-by-layer assembly of the polyelectrolyte coating. While top-down coarse-grained models of polyelectrolyte-coated AuNPs have focused on polyelectrolytes of short length, from 10 to a 100 monomers represented as one charged bead per monomer, here we use molecular dynamics and bottom-up coarse-grained approaches to access more typical polymer lengths on the order of 200 monomers. Specifically, we simulate the adsorption dynamics and structure of one or two such long polycations on negatively charged 4 nm citrate-coated AuNPs within implicit or explicit solvents. The first polycation coats approximately half of the AuNP surface regardless of solvent model and leaves a significant part of the anionic citrate layer exposed to absorption by a second polycation. Citrate stabilized gold nanoparticles interfere with amyloid fibril formation: D76N and ΔN6 β2-microglobulin variants.

Protein aggregation including the formation of dimers and multimers in solution, underlies an array of human diseases such as systemic amyloidosis which is a fatal disease caused by misfolding of native globular proteins damaging the structure and function of affected organs. Different kind of interactors can interfere with the formation of protein dimers and multimers in solution. A very special class of interactors are nanoparticles thanks to the extremely efficient extension of their interaction surface. In particular citrate-coated gold nanoparticles (cit-AuNPs) were recently investigated with amyloidogenic protein β2-microglobulin (β2m). Here we present the computational studies on two challenging models known for their enhanced amyloidogenic propensity, namely ΔN6 and D76N β2m naturally occurring variants, and disclose the role of cit-AuNPs on their fibrillogenesis. The proposed interaction mechanism lies in the interference of the cit-AuNPs with the protein dimers at the early stages of aggregation, that induces dimer disassembling. As a consequence, natural fibril formation can be inhibited. Relying on the comparison between atomistic simulations at multiple levels (enhanced sampling molecular dynamics and Brownian dynamics) and protein structural characterisation by NMR, we demonstrate that the cit-AuNPs interactors are able to inhibit protein dimer assembling. As a consequence, the natural fibril formation is also inhibited, as found in experiment.

Interference of citrate-stabilized gold nanoparticles with β2-microglobulin oligomeric association.

Protein fibrillation is involved in many serious diseases, and protein oligomers are proved to be precursors of amyloid fibrils. NMR and QCMD experiments allowed us to establish that the interaction between citrate-stabilized gold nanoparticles and a paradigmatic amyloidogenic protein, β2-microglobulin, is able to interfere with protein association into oligomers.

Fast and Strong Adsorption of Native Oligonucleotides on Citrate-Coated Gold Nanoparticles.

The adsorption of oligonucleotides on citrate-coated gold nanoparticles (AuNPs) is studied under conditions "right after the synthesis", i.e., in a weak citrate solution at a pH value close to neutral (5.8 ± 0.2). We found that short-term elevation of reaction temperature under these conditions provides fast and strong adsorption of oligonucleotides on the surface of AuNPs. The affinity of oligonucleotides to AuNPs depends on the length of the oligonucleotide and its nucleotide composition. The shortest oligonucleotide in this study, T6, is the most affine, having the equilibrium binding constant KD = 0.10 ± 0.04 nM and the highest surface density-up to 200 molecules per one particle. Olygothymidylates are at least as affine to AuNPs as oligoadenylates, while oligocytidilates show the lowest affinity. We also studied the interaction of resulting DNA/AuNPs with a series of low- and high-molecular thiols, which provide a variety of operations with adsorbed oligonucleotides: displacement (complete or partial) and encapsulation in a secondary shell. These experiments imitate someway the conditions in a living cell or serum, and show that DNA/AuNPs obtained by this method can be applied in a number of bionanotechnological applications, including delivery of nucleic acid therapeutics and theranostics.

Epidermal Penetration of Gold Nanoparticles and Its Underlying Mechanism Based on Human Reconstructed 3D Episkin Model.

Nanomaterials are widely used in diverse aspects, and their translocation behavior through the skin would be helpful in understanding the corresponding exposure risks. To reveal how surface functionalization of nanoparticles influences the skin penetration, three kinds of gold nanoparticles (GNPs) with negatively, neutrally, and positively charged surfaces, that is, cetyltrimethylammonium bromide-coated GNPs (CTAB@GNP), polyvinylpyrrolidone-coated GNPs (PVP@GNP), and citrate-coated GNPs (Citrate@GNP), were studied using human reconstructed 3D Episkin model. The measurement of Au distribution in diverse layers of the Episkin model indicated that all three GNPs could penetrate through the epidermis, wherein CTAB@GNP with positive surface charges exhibited the highest efficiency in skin penetration. The upward osmosis of the medium proteins confirmed the occurrence of skin permeation induced by GNP treatment, and the lipid network in the stratum corneum was also altered as the consequence of GNP exposure. When compared to Citrate@GNP and PVP@GNP, CTAB@GNP significantly compromised the tight junction of keratinocytes, causing paracellular penetration of nanoparticles. The existence of cytoplasmic gold showed the transcytosis pathway through endocytosis and exocytosis processes was the main epidermic penetration behavior of the tested GNPs. The study on GNP penetration process through the 3D Episkin model has, on one hand, offered a promising approach to evaluate the translocation process of nanoparticles across the skin, and, on the other hand, provided mechanism explanation for diverse penetration behaviors of GNPs with different surface charges. The findings herein would be of great help in nanotechnology improvement and nanosafety evaluation.

Salt-Induced Aggregation of Negatively Charged Gold Nanoparticles Confined in a Polymer Brush Matrix

We report on the salt-induced aggregation of citrate-coated gold nanoparticles (AuNPs) confined within poly(N-isopropylacrylamide) (PNIPAM) brushes grafted from flat substrates. Compared to highly dispersed AuNPs, a red-shift and broadening of the surface plasmon (SP) band is observed when the AuNPs are confined by the PNIPAM brush matrix due to their close vicinity. Additional red-shifting and broadening occur upon immersion in aqueous salt solutions (1 M NaF, NaCl, NaBr, and KCl). Nanoparticle assemblies are established due to salt-induced aggregation of AuNPs and are dependent on the type of salt. In the presence of KCl, nanoparticle assemblies are built up that result in the formation a second plasmon peak at ∼700 nm. The color change of PNIPAM/AuNP is associated with (1) the collapse of the PNIPAM brushes in the presence of salt and (2) nanoparticle aggregation due to electrostatic screening of the negative charges around the AuNPs by the salt ions.
