The impact of working conditions on the health of workers in contact with chrysotile-containing dust

The aim was to summarize the information in the literature on working conditions and professionally caused diseases in workers in contact with chrysotile-containing dust.During a 10-year-old literary search in Russian, Kazakh and English using the keywords «chrysotile asbestos», «employee health», «production environment», «risk factor», «professionally caused disease», «working conditions» from PubMed (Medline), eLibrary, Google Scholar databases.Assessing the health of the working population in the conditions of modern industry is becoming an urgent task that requires in-depth research of working conditions, lifestyle and health status of workers. According to the International Labour Organization, 160 million new cases of work-related diseases are registered annually in the world due to the effects of harmful industrial factors.Working conditions in the mining industry are associated with the release of dust, unfavorable microclimate, noise and vibration, which to one degree or another have a negative impact, it is the working conditions of workers that are the main risk factor for the formation of professional and production-related pathology.The problem of professionally caused diseases among workers of chrysotile production remains relevant and indicates the need for scientific epidemiological studies.

Spectroscopic Analysis of Chrysotile Asbestos and its Environmental Resistance in Asbestos Cement Waste Products

Most asbestos-related studies have focused on asbestos exposure risks, their associated health implications, and waste management issues. Our research introduced a unique perspective that has rarely been explored: the impact of environmental factors on asbestos cement products. The novelty of the study is that, in contrast to previous research, in addition to determining the material quality of asbestos, it analyses the trace materials, additives and the emissive nature of chrysotile fibers. This study aims to identify the chrysotile-asbestos content in three common asbestos cement products found in Hungary, with regard to the release of their fibers upon exposure to the environment and to identify trace elements that could be used to identify the origin and function of each of these products. Our analyses revealed the presence of chrysotile in each tested sample, with spectral matches ranging from 59.6% to 86.7%. Asbestos cement products exposed to various environmental influences for long periods showed a greater chrysotile emission capacity than those unexposed or hermetically sealed ones. Additionally, we established that all asbestos cement products contained glass fibers, with an average spectral match of 62.1%. We further identified polysilicate in the materials with an average spectral match of 66.0%, as it was included in asbestos cement products to enhance their heat resistance. Our results pave the way for a new methodology for assessing asbestos cement products with regard to the implementation of their trace element level assessments.

Ferritin adsorption onto chrysotile asbestos fibers influences the protein secondary structure

Asbestos fiber exposure triggers chronic inflammation and cancer. Asbestos fibers can adsorb different types of proteins. The mechanism of this adsorption, not yet completely understood, has been studied in detail mainly with serum albumin and was shown to induce structural changes in the bound protein. The findings of these works regarded mainly the changes of the protein structure, independently of any relation with asbestos-related diseases. For the first time, we have focused our attention to the consequences of the interaction between asbestos fibers and ferritin, a protein involved in iron metabolism, which is strongly modified in asbestos-related diseases. Even if it is known that ferritin can be adsorbed by asbestos fibers, the results of this interaction for the ferritin secondary structure has not previously been studied. One consequence of asbestos-ferritin interaction, is the formation of the so-called ferruginous/asbestos bodies (ABs). In the AB-coating material, the secondary structure of ferritin is modified, and at present, it is unclear whether or not this modification is a direct consequence of the asbestos interaction. In the present study, chrysotile asbestos, more than other asbestos fiber types tested, was found to rapidly bind holo-ferritin, and the presence of iron seemed to play a key role in this process, since iron-free apo-ferritin was adsorbed at a lower level, and iron-saturated chrysotile lost its ferritin-adsorbing capacity. To directly study the details of ferritin adsorption on asbestos fibers, High Resolution Transmission Electron Microscopy (HR-TEM) was employed together with FTIR microspectroscopy and Infrared nanoscopy, which to the best of our knowledge, have not previously been used for this purpose. Chrysotile-bound apo-ferritin underwent a significant change in secondary structure, showing a shift from a prevalent α-helix to a β-sheet conformation. Conversely, the adsorbed holo-ferritin structure appeared to be only weakly modified. These findings add a new potential mechanism to the toxic activities of asbestos: the fibers can modify the structure, and very likely, the function of adsorbed proteins. This, in relation to ferritin, could be a key mechanism in cell iron homeostasis alteration, typically reported in asbestos-related diseases.

INFLUENCE OF STRUCTURAL FEATURES OF CHRYSOTYLE CRYSTALS ON THE ACTION OF THE SYSTEM OF CHRYSOTYLE AND SULFURIC ACID

The nature of the interactions between chrysotile and stoichiometrically necessary amounts (SNA) of sulfuric acid calculated in relation to the amount (molar) of magnesium contained in chrysotile – Mg6[Si4O10](OH) has been studied. It has been shown that when using solutions containing sulfuric acid HCl (0-0.3), the amount of magnesium found in the solution corresponds to a proportional amount of the acid used. It was found that the output of magnesium into the solution slows down in the range of SNA (0.3-0.5). Proportionally, the dissolution of magnesium is restored in the range of SNA (0.5-0.7). However, the yield of magnesium in solution does not exceed 76% of its content in chrysotile due to the formation of polysilicon acid (SiO2·nN2O) in an acidic medium. The layer formed on the surface consisting of polysilicon acids is the main factor inhibiting the transition of magnesium into solution.It was revealed that the amount of magnesium passing into solution depends not only on the concentration of acid, but also on the density of the layered structure of chrysotile. The conclusions are based on the results of chemical and X-ray phase studies of reaction products occurring during the dissolution of chrysotile asbestos in solutions of sulfuric acid H2SO4.Based on results of the study, it was shown that the dissolution of magnesium silicates, the composition of which of minerals (chrysotile, lizardite, antigorite) of the serpentinite group with the general formula Mg6[Si4O10](OH)8 in acids depends not only on the patterns of acid-base interactions occurring under these conditions, but also on the structural features of the serpentinite crystal lattice.

Evaluation of airborne asbestos concentrations associated with the maintenance of brakes on an industrial overhead crane

Objectives To evaluate potential airborne asbestos exposures during brake maintenance and repair activities on a P&H overhead crane, and during subsequent handling of the mechanic’s clothing. Methods Personal (n = 27) and area (n = 61) airborne fiber concentrations were measured during brake tests, removal, hand sanding, compressed air use, removal and reattachment of chrysotile-containing brake linings, and reinstallation of the brake linings. The mechanic’s clothing was used to measure potential exposure during clothes handling. Results All brake linings contained between 19.9% to 52.4% chrysotile asbestos. No amphibole fibers were detected in any bulk or airborne samples. The average full-shift airborne chrysotile concentration was 0.035 f/cc (PCM-equivalent asbestos-specific fibers, or PCME). Average task-based personal air samples collected during brake maintenance, sanding, compressed air use, and brake lining removal tasks ranged from 0 to 0.48 f/cc (PCME). The calculated 30-minute time-weighted average (TWA) airborne chrysotile concentration associated with 5-15 minutes of clothes handling was 0–0.035 f/cc PCME. Conclusion The results indicated that personal and area TWA fiber concentrations measured during all crane brake maintenance and clothes handling tasks were below the current OSHA 8-h TWA Permissible Exposure Limit for asbestos of 0.1 f/cc. Further, no airborne asbestos fibers were measured during routine brake maintenance tasks following the manufacturer’s maintenance manual procedures. All short-term airborne chrysotile concentrations measured during non-routine tasks were below the current 30-minute OSHA excursion limit for asbestos of 1 f/cc. This study adds to the available data regarding chrysotile exposure potential during maintenance on overhead cranes.

Asbestos exposures associated with the use and handling of drilling mud additives.

This paper summarizes historical asbestos exposure data collected during the handling of short-fiber chrysotile asbestos that was used as an additive to drilling fluid in oil and gas exploration. A total of 1171 industrial hygiene (IH) personal and area air samples were collected and analyzed from more than 20 drilling rigs between 1972 and 1985. The dataset consists of 1097 short-term samples (<240min) with more than 80% having sample durations less than 30min. Average airborne fiber concentrations measured during asbestos handling activities ranged from 0.62 f/cc to 3.39 f/cc using phase-contrast microscopy (PCM). An additional 14 samples were considered long-term samples (>240min) and there were 60 samples with no reported sample duration. Eight-hour time-weighted average (8-h TWA) results, calculated using short-term samples, along with long-term samples greater than 240min, did not exceed contemporaneous Occupational Safety and Health Administration (OSHA) permissible exposure limits (PELs). This analysis fills a data gap in the evaluation of asbestos exposures from the use of drilling mud additives (DMAs) that contained chrysotile asbestos.

Burden of malignant mesothelioma in China during 1990–2019 and the projections through 2029

ObjectiveTo provide the most up-to-date data on the burden of malignant mesothelioma (MM) and the projections through 2029 in China. MethodsData on patients diagnosed with MM from China during 1990–2019 were obtained from the Global Burden of Disease (GBD) 2019 database, including annual cases and deaths data and age-standardized rates of incidence, mortality, and disability-adjusted life-years (DALYs) associated with MM among different age groups. Temporal trends during 1990–2019 were analyzed by the Joinpoint regression models using 95% confidence interval (CI), while the projections through 2029 were calculated by the Bayesian age-period-cohort model. Data on the production and consumption of asbestos in China were obtained from the United States Geological Survey on Mineral Commodity Summaries during 1996–2023. ResultsWe observed a significant elevation in incident new cases and deaths over the last 3 decades, increasing from 1193 in 1990 to 2815 in 2019 for incident cases and from 1134 in 1990 to 2773 in 2019 for death cases. We found a roughly 6% increase in the proportion of incident cases for those aged >70 years (30% in 2019 versus 24% in 1990), while for the proportion of deaths similar elevation for those aged >70 years was found. Additionally, men had significantly higher DALYs due to MM across age groups compared with women. Asbestos consumption in China dramatically dropped since 2012 and reached the bottom in 2017 with 230 kilotons. By 2029, the projected age-standardized rate for incidence and mortality is expected to reach 1.2 per million for both. ConclusionWe found, for the first time using GBD data on the Chinese population, that the burden of MM has been significantly increasing in China over the last three decades and will continue to increase in the upcoming decade, suggesting an urgent need for a complete ban on chrysotile asbestos in China.

Toxicity and inflammatory potential of mineral fibres: The contribute of released soluble metals versus cell contact direct effects.

Asbestos fibres have been considered an environmental hazard for decades. However, little is known about the attempts of circulating immune cells to counteract their toxicity. We addressed the early effects of fibre-released soluble factors (i.e. heavy metals) in naïve immune cells, circulating immediately below the alveolar/endothelial cell layer. By comparison, the direct fibre effects on endotheliocytes were also studied since these cells are known to sustain inflammatory processes. The three mineral fibres analysed showed that mainly chrysotile (CHR) and erionite (ERI) were able to release toxic metals in extracellular media respect to crocidolite (CRO), during the first 24 h. Nevertheless, all three fibres were able to induce oxidative stress and genotoxic damage in indirectly challenged naïve THP-1 monocytes (separated by a membrane). Conversely, only CHR-released metal ions induced apoptosis, NF-κB activation, cytokines and CD163 gene overexpression, indicating a differentiation towards the M0 macrophage phenotype. On the other hand, all three mineral fibres in direct contact with HECV endothelial cells showed cytotoxic, genotoxic and apoptotic effects, cytokines and ICAM-I overexpression, indicating the ability of these cells to promote an inflammatory environment in the lung independently from the type of inhaled fibre. Our study highlights the different cellular responses to mineral fibres resulting from both the nature of the cells and their function, but also from the chemical-physical characteristics of the fibres. In conclusion, CHR represented the main pro-inflammatory trigger, able to recruit and activate circulating naïve monocytes, through its released metals, already in the first 24 h after inhalation.

Low Exposures to Amphibole or Serpentine Asbestos in Germline Bap1-mutant Mice Induce Mesothelioma Characterized by an Immunosuppressive Tumor Microenvironment.

We show that germline Bap1-mutant mice have enhanced susceptibility to MM upon minimal exposure to chrysotile asbestos, not only amphibole fibers. Chrysotile induced a more profound immune tumor response than crocidolite in Bap1-mutant mice by upregulating CD39/CD73-adenosine and Ccl2/Ccr2 pathways and recruiting more M2 macrophages, which together contributed to an immunosuppressive tumor microenvironment. Interrogation of human MM RNA-seq data revealed interconnected immunosuppressive pathways consistent with our mouse findings.

Abstract 1468: Low exposures to amphibole or serpentine asbestos in germline Bap1-mutant mice induce mesotheliomacharacterized by an immunosuppressive tumor microenvironment

Abstract The purpose of this study was to determine if minimal exposures to amphibole (crocidolite) and serpentine (chrysotile) asbestos fibers significantly increase the incidence and rate of malignant mesothelioma (MM) onset in mice with a germline heterozygous Bap1 mutation, and to assess the effect of the resulting gene × environment interactions on the tumor immune microenvironment (TIME). The studies were carried out by chronically injecting intraperitoneally Bap1+/- and Bap1+/+ (wild-type, WT) mice with minimal doses of asbestos (in some experiments, as low as 0.0125 mg x 8 injections; total, 0.1 mg/mouse) and then monitoring the mice for tumor formation for up to 90 weeks. Pro-inflammatory and pro-tumorigenic factors in the TIME were evaluated by immunohistochemistry (IHC) and immunofluorescence (IF) for immune cell markers. With either type of asbestos and at each dose tested, MMs occurred at a significantly higher rate and earlier onset time in Bap1-mutant mice than in WT littermates. MMs from mice injected with chrysotile, but less so with crocidolite, had widespread adhesions in the peritoneal cavity owing to an overwhelming immune response, particularly at the higher dose used. IHC staining demonstrated an increased number of immune cells in granulomas and MMs. In particular, pro-inflammatory M1 macrophages were found mainly in granulomas, whereas pro-tumorigenic M2 macrophages were predominant in MMs. IF staining revealed that the relative number of M2 macrophages was strikingly greater in chrysotile-induced than crocidolite-induced MMs, suggesting that chrysotile induces a more profound immune response. MMs from Bap1-mutant mice showed upregulation of both CD39/CD73-adenosine and Ccl2/Ccr2 pathways, which collectively are known to mechanistically create favorable conditions for neoplastic transformation, invasion, and the development of an immunosuppressive TIME, in part by attracting M2 macrophages. This implies that chemoprevention and immunotherapy strategies targeting these pro-tumorigenic immune pathways could be beneficial in BAP1 mutation carriers. In conclusion, we provide evidence that low-level exposures to either chrysotile or crocidolite in germline Bap1 heterozygous mice result in a markedly increased risk of MM characterized by an immunosuppressive tumor microenvironment. Drawing parallels to humans, these experimental findings suggest that exposure of BAP1 mutation carriers to even minimal doses of different forms of asbestos may be associated with significantly enhanced susceptibility to MM. Iron nuclearity in mineral fibres: Unravelling the catalytic activity for predictive modelling of toxicity

Chronic inflammation induced in vivo by mineral fibres, such as asbestos, is sustained by the cyclic formation of cytotoxic/genotoxic oxidant species that are catalysed by iron. High catalytic activity is observed when iron atoms are isolated in the crystal lattice (nuclearity=1), whereas the catalytic activity is expected to be reduced or null when iron forms clusters of higher nuclearity.This study presents a novel approach for systematically measuring iron nuclearity across a large range of iron-containing standards and mineral fibres of social and economic importance, and for quantitatively assessing the relation between nuclearity and toxicity. The multivariate curve resolution (MCR) empirical approach and density functional theory (DFT) calculations were applied to the analysis of UV-Vis spectra to obtain information on the nature of iron and nuclearity.This approach led to the determination of the nuclearity of selected mineral fibres which was subsequently used to calculate a toxicity-related index. High nuclearity-related toxicity was estimated for chrysotile samples, fibrous glaucophane, asbestos tremolite, and fibrous wollastonite. Intermediate values of toxicity, corresponding to a mean nuclearity of 2, were assigned to actinolite asbestos, amosite, and crocidolite. Finally, a low nuclearity-related toxicity parameter, corresponding to an iron-cluster with a lower catalytic power to produce oxidants, was assigned to asbestos anthophyllite.

Measurement and removal of asbestos in residential dwellings to be demolished-urban transformation experience in Izmir, Turkey.

Asbestos has been used extensively in the construction industry for its superior insulation properties before its health hazards were discovered and its use eventually banned. It is likely that many residential buildings built before the 2000s in Turkey contain asbestos. Therefore, it is important to raise awareness of the potential danger of asbestos exposure during demolition work and to identify asbestos-containing materials and ensure their safe removal and disposal. This study is executed to determine the residential dwellings containing asbestos in Izmir, Turkey. The research included field studies to determine asbestos presence in the buildings that were damaged during the 2020 earthquake. Air measurements and bulk samples were taken from 50 buildings that would go through the demolition process. Eleven buildings were found to contain asbestos which corresponds to 22%. The detected asbestos type was 60% chrysotile (white asbestos). Results could be helpful for future demolition work, which are conducted in the same region that includes buildings with similar properties. Also, it is expected that the database created for this study could be useful in other studies in Turkey, where accurate statistical data related with asbestos measurements is essentially non-existent.

Construction with Slate – Quick Solution to the Country’s Housing Problems

General information about the state of the chrysotile cement industry in Russia and in the world since the beginning of the XX century and changes in recent decades is presented. The most intensive development of the industry is noted in Russia and the countries of the former USSR due to the largest reserves of chrysotile asbestos in these territories. The physical and technical properties of chrysotile cement products, due to which they have many advantages both in terms of operational reliability and economic attractiveness, are given. The high potential of chrysotile-cement materials in construction is justified. It is shown that the frame technology of SOVBI can provide a significant increase in demand for flat chrysotile cement sheets.

Design of a prognostic model for assessing the risk of developing asbestosis based on the results of molecular genetic studies

Introduction. Preventive measures, including the development of a system for diagnosing early signs of changes in the health status of employees and predicting individual risks of developing the disease, aimed at preserving labor resources, reducing disability and extending working life expectancy, are an absolute priority in the field of occupational safety.&#x0D; The aim of the study to develop a prognostic model of the risk of developing asbestosis based on molecular genetic studies in employees of enterprises for the extraction and enrichment of chrysotile asbestos.&#x0D; Materials and methods. Based on studies previously conducted in the Laboratory of Biomedical Research at the Izmerov Research Institute of Occupational Health, scientists have identified a set of informative and significant single-nucleotide polymorphic gene variants for use as predictors in the construction of a prognostic model in the development of asbestos in workers of enterprises for the extraction and enrichment of chrysotile asbestos. We examined 136 people who worked at the enterprise for the extraction and enrichment of chrysotile asbestos. The researchers formed two groups comparable in terms of work experience, dustiness, age and gender: 68 former employees of the main production specialties with an established diagnosis of asbestos and 68 workers without bronchopulmonary pathology.&#x0D; The authors calculated the exposure dose of chrysotile-containing dust and calculated it taking into account the percentage of time spent at the workplace, and also performed a molecular genetic study of single-nucleotide polymorphic variants of the genes IL-1β rs16944, IL-4 rs2243250, TGF-β1 rs1800471, SOD-2 rs4880 and MMP-9 rs17576.&#x0D; Results. A predictive model has been developed to determine the probability of developing asbestosis in workers of an enterprise for the extraction and enrichment of chrysotile asbestos based on the determination of single nucleotide polymorphisms IL-1β rs16944, SOD-2 rs4880 and MMP-9 rs17576. The results of genetic tests are entered into the formula and if the resulting probability of developing asbestosis is equal to or higher than the threshold (0.582), the employee is included in the high-risk group. The sensitivity and specificity of the developed model were 70.1% and 70.6%, respectively.&#x0D; Conclusion. Molecular genetic indicators can act as predictors in the development of asbestosis in workers of the chrysotile extraction and enrichment industry and can be used to form high-risk groups during preventive measures.&#x0D; Ethics. The study was conducted in compliance with ethical standards that guarantee respect for all research subjects and the protection of their health and rights in accordance with the requirements of the Helsinki Declaration of the World Medical Association. All persons included in the study gave informed voluntary consent to the examination in accordance with Federal Law No. 323-FZ dated 11/21/2011 "On the Basics of Public Health Protection in the Russian Federation". The study was approved by the conclusion of the local Ethics committee of Izmerov Research Institute of Occupational Health (minutes of the meeting of the Ethics committee of Izmerov Research Institute of Occupational Health No. 9 dated 11/29/2016).

INVESTIGATING CHRYSOTILE ASBESTOS WASTE AS A POTENTIAL MATERIAL FOR SYNTHETIC ADSORBENTS

Technological methods have been developed to acquire an active component with a high SiO2 content, which serves as a matrix foundation for synthetic adsorbents made from chrysotile asbestos production waste composite materials. Comparative analyses were conducted between the obtained adsorbent and a zeolite adsorbent of the NaX model. The physico-chemical analyses unveiled that the synthetic adsorbent obtained is not inferior in any aspect, and in certain instances, even outperforms in the demetallization process of heavy petroleum raw materials. The demetallization process exhibits an optimal temperature range of 380-410 °C, with a contact time of 60 minutes and a pressure range of 8-10 atm. Concurrently, the concentration of vanadium in heavy crude oil decreases significantly from 540 to 28 micrograms per kilogram, while nickel content diminishes from 50 to 26 micrograms per kilogram. Moreover, iron content experiences a reduction from 50 to 12 micrograms per kilogram, and sulfur content decreases from 3.50 % to 2.39 %.

Ammonia-induced corrosion and modification of chrysocolla surface for enhanced adsorption of sulfide species

The copper content of chrysocolla is an oxidized ore, which has high industrial value in terms of development and utilization. Flotation is generally used to recover such minerals. However, few studies have been conducted on the mechanism of ammonium enhanced flotation in the flotation process of chrysotile asbestos at the atomic level, including the enhanced sulfidation process in solution and mineral surface. This study demonstrated that the presence of NH3 only enhances the reaction between Na2S⋅9H2O and Cu ions. The formation of copper-ammonia complexes facilitated the sulfide reaction and led to the generation of more Cu-S species, which can adsorb onto the chrysocolla surface. The NH3 destroys the hydration film of the Cu6[Si6O18]6H2O (1 1 0) surface, creating favorable conditions for sulfidation. The CuS formed in catalytic sulfidation has higher stability compared with CuS formed through direct sulfidation. The NH3 molecule corrosion of the Cu6[Si6O18]6H2O (1 1 0) surface was advantageous in terms of energy, and the formation of Cu(NH3)42+ was − 539.32 kJ·mol−1. After the Cu atom ultimately lost 0.37 e electrons, the Cu-O bond broke, and Cu(NH3)42+ relaxed towards the vacuum layer, which formed a defect at the Cu6[Si6O18]6H2O (1 1 0) surface. The catalytic sulfidation path in the solution was Cu(NH3)4(OH)2 + HS− + 17 H2O + Na+ → CuS + 4NH3 + OH− + 17 H2O + Na+. The total energy of the reaction was − 0.88 kJ·mol−1, which indicated that the catalytic sulfidation reaction could spontaneously occur. The CuS generated by catalytic sulfidation was stably adsorbed to the Cu6[Si6O18]6H2O surface of defects through the formation of stable chemical bond between Cu and O. The obtained results revealed the complex interaction between ammonium and minerals, which paves the way for its efficient sulfide flotation.

Photonic and phononic properties of oriented 5 nm diameter tellurium nanowires

We study polarized optical absorption and Raman spectra (OAS and RS) of ∼5 nm diameter tellurium (Te) nanowires (NWs) confined in chrysotile asbestos nanotubes packed parallel to each other into nearly-perfect-hexagonal lattice. Te was encapsulated into the nanotubes from molt using ∼20 kbar pressure. Asbestos with Te NWs (asb-Te) shows a high OAS/RS anisotropy due to a strong NW-light interaction for the light polarized papallel to NWs (E//c) and a weak one for the light polarized perpendicular to NWs. Importantly, asb-Te RS display bands of confined acoustic phonons at ∼9 cm−1 and ∼23 cm−1, specific signatures of 5 nm diameter Te NWs. Density-functional-theory calculation shows agreement with the observed frequencies and flattening of NW acoustic phonon dispersions useful for thermo-electricity. Asb-Te optical phonon A1-mode Raman band displays frequency upshift compared to the bulk-Te band due to enhancement of the intrachain bonds on expense of interchain ones. Asb-Te 2nd-order RS confirm this. Surprisingly, E-modes display LO-phonon RS-activity in asb-Te instead of TO-phonon-activity in bulk-Te. We observe enhancement of the NW Raman signal compared to that of bulk Te, which is associated with the Mie-resonance-induced NW-light interaction enhancement for E//c, which is important for photonic and photo-electric applications.
